Physical constrains such as water discharge, suspended solids and turbidity act as dominant factors in driving the planktonic diatom assemblages of the River Adige (North-Eastern Italy). Two sampling stations, characterised by different hydromorphological features (Cortina all’Adige and Boara Pisani, with torrential and more potamal characteristics, respectively) were sampled fortnightly following an integrated approach encompassing physical, chemical and biological measurements and aiming at identifying the dominant factors controlling the temporal development of the community. A morpho-functional approach was used to classify the diatom assemblages where Morpho-Functional Diatom Groups (MFDG) were defined for diatom genera, according to their morphology, habitat selection and modality of adhesion to river substrate. In the two sampling points, algal growth was never limited by nutrients or zooplankton. The irregular development of MFDG was determined by the stochastic hydrological events and changes in variables related to water discharge (suspended solids and light attenuation). Tychoplanktonic, benthic and drifted taxa (such as Diatoma spp., Encyonema spp., Navicula spp. and Nitzschia spp.) were dominant in the torrential station (Cortina all’Adige), while the contribution of euplanktonic unicellular centric taxa (such as Cyclotella spp., and Stephanodiscus spp.) was higher in the potamal station (Boara Pisani).  相似文献

The levels of activity of 2-phosphoglycolate phosphatase in the green algae, Chlamydomonas reinhardtii and Chlorella vulgaris, were in the range of 37 to 60 micromoles per milligram chlorophyll per hour and in the blue-green algae, Anacystis nidulans and Anabaena variabilis were 204 to 310 micromoles per milligram chlorophyll per hour. The activity in each species was similar regardless of whether the algae were grown with air or 5% CO2 in air. The enzyme purified 530-fold from Chlamydomonas was stable, had a broad pH optimum between 6 and 8.5, and was specific for the hydrolysis of P-glycolate with a Km of 23 micromolar. The enzyme purified 18-fold from Anacystis was labile, had a sharp pH optimum at 6.3, and was also specific for P-glycolate with a Km of 94 micromolar. The molecular weight of the enzyme from Chlamydomonas was estimated to be 92,000 by gel filtration.

The phosphatase from both sources required a divalent cation for activity. The Chlamydomonas enzyme was most effectively activated by Co2+, but was also activated by Mg2+ (Ka = 30 micromolar), Mn2+, and Zn2+. The Anacystis enzyme was most effectively activated by Mg2+ (Ka = 140 micromolar), and was also activated by Co2+ and Mn2+, but not by Zn2+. Anions were also required for maximum activity of the enzyme from both sources. The Chlamydomonas enzyme was activated about 2- to 3-fold by chloride (Ka = 140 micromolar), bromide, nitrate, bicarbonate (Ka = 600 micromolar) and formate. The Anacystis enzyme was activated over 10-fold by chloride (Ka = 870 micromolar), bromide, iodide, and nitrate, but was not activated by bicarbonate or formate.

The properties of the algal enzymes were similar to those previously reported for higher plants. The levels and kinetic properties of the enzyme seemed sufficient to account for the flux through the glycolate pathway that occurs in these algae. The phosphatase was not associated with the ribulose 1,5-bisphosphate carboxylase/oxygenase responsible for P-glycolate formation in the carboxysomes of Anacystis.

Initial Cu++ sorption by Cyclotella meneghiniana Kütz. (Cu++-sensitive) and Chlamydomonas reinhardtii Dangeard (Cu++-resistant) was rapid in the first 5 min of Cu++ incubation with little sorption after 2 h. On a cell to cell basis, Cyclotella sorbed ca. five times more Cu++ from the medium than Chlamydomonas. In MBL medium with EDTA Cyclotella and Chlamydomonas cells sorbed 21.0 and 4.41 nM Cu++/106 cells respectively in 6 h with 0.3 mg Cu++/l in the medium. Proportionally similar quantities of Cu++ were sorbed when the cells were Cu++ incubated in MBL + citrate or filtered lake water. Cleaned cell walls of Cyclotella sorbed little Cu++ (1.7 nM/106 cells) as compared to living cells (17.5 nM Cu++/106 cells) in 3 h. Therefore, in living Cyclotella most of the Cu++ taken up must be absorbed by the protoplasm or perhaps by the organic layer surrounding the silica wall. Cleaned cell walls of Chlamydomonas sorbed 3.5 nM Cu++/106 cells and living Chlamydomonas cells sorbed 2.6 nM Cu++/106 cells. This indicates that most of the Cu++ sorbed by Chlamydomonas cells remained bound to the cell wall and probably did not readily enter into the protoplasm: When placed in Cu++ free medium after Cu++ incubation, Cyclotella and Chlamydomonas cells released 46 and 59% respectively of the Cu++ sorbed.  相似文献

Diatoms are a group of highly abundant and diverse aquatic algae species. They contain high lipid content along with many bioactive compounds that can be exploited for biotechnological applications. Despite these attractive attributes, diatoms are underrepresented in production projects due to difficulties in their cultivation. To optimize the growth of three freshwater diatom isolates, Cyclotella sp., Synedra sp. and Navicula sp., an orthogonal assay on N, P, Si and Fe, as well as temperature and pH, was designed using traditional single‐factor tests. We also studied the effect of using nanosilica as an alternate Si source on growth and found that the diatom isolates studied achieved their highest growth rates under different combinations of nutrient and environmental conditions. Silica had the greatest influence on growth, followed by phosphate and iron. The optimized growth conditions for Synedra sp. were N: 30 mg L?1, P: 3 mg L?1, Si: 14.8 mg L?1, Fe: 0.448 mg L?1, temperature 25°C and pH 8. For Navicula sp.: N: 20 mg L?1, P: 2.5 mg L?1, Si: 19.7 mg L?1, Fe: 0.112 mg L?1, temperature 30°C and pH 7.5–8. For Cyclotella sp.: N: 20 mg L?1, P: 2.5 mg L?1, Si: 19.7 mg L?1, Fe: 0.448 mg L?1, temperature 30°C and pH 7.5–8. Nano silica negatively affected growth in Navicula sp. and Cyclotella sp., but no such effect was observed in Synedra sp. Fatty acid profiling showed C16:0, C16:1(n ? 7), C18:0 and C20:5(n ? 3) as major fatty acids, with no significant differences in fatty acid methyl ester profiles between traditional and modified media. This work gives us a new insight into the growth requirements of freshwater diatom species, which are less studied than marine species.  相似文献

Growing algae to scrub nutrients from manure presents an alternative to the current practice of land application and provides utilizable algal biomass as an end product. The objective of this study was to assess algal growth, nutrient removal, and nitrification using higher light intensities and manure loading rates than in the previous experiments. Algal turfs, with periphyton mainly composed of green algal species, were grown under two light regimes (270 and 390 μmol photons·m?2· s?1) and anaerobically digested flushed dairy manure wastewater (ADFDMW) loading rates ranging from 0.8 to 3.7 g total N and 0.12 to 0.58 g total P·m?2·d?1. Filamentous cyanobacteria (Oscillatoria spp.) and diatoms (Navicula, Nitzschia, and Cyclotella sp.) partially replaced the filamentous green algae at relatively high ADFDMW loading rates and more prominently under low incident light. Mean algal production increased with loading rate and irradiance from 7.6±2.71 to 19.1±2.73 g dry weight· m?2·d?1. The N and P content of algal biomass generally increased with loading rate and ranged from 2.9%–7.3% and 0.5%–1.3% (by weight), respectively. Carbon content remained relatively constant at all loading rates (42%–47%). The maximum removal rates of N and P per unit algal biomass were 70 and 13 mg·g?1 dry weight·m?2·d?1, respectively. Recovery of nutrients in harvested algal biomass accounted for about 31%–52% for N and 30%–59% for P. Recovery of P appeared to be uncoupled with N at higher loading rates, suggesting that algal potential for accumulation of P may have already been saturated. It appears that higher irradiance level enhancing algal growth was the overriding factor in controlling nitrification in the algal turf scrubber units.  相似文献

Photorespiration by Chlamydomonas reinhardtii and Anacystis nidulans was measured as the oxygen inhibition of CO2 uptake and the CO2 compensation points. Net photosynthesis was oxygen dependent in Chlamydomonas grown in 5% CO2, but CO2 insensitive in cultures bubbled with air. Anacystis, even when cultured in 5% CO2, exhibited an CO2 insensitive net photosynthesis. The CO2 compensation point of Chlamydomonas grown in cultures bubbled with air and Anacystis grown in 5% CO2 enriched air, were reached shortly after the measurement was begun and the values were very low, less than 10 μl CO2 1?1; while Chlamydomonas grown in 5% CO2 enriched air for 4 days showed a high, but temporary CO2 compensation point (60 μl CO2 1?1). After a two hour adaptation in low CO2, a stable, low CO2 compensation point was reached. It seems that photorespiration can only be detected by the methods used in this study when the algae are cultured in high CO2, but a mechanism exists which blocks photorespiration when the green algae are adapted to low CO2 concentrations. When Chlamydomonas was treated with Diamox, an inhibitor of carbonic anhydrase, after cultivation in low CO2 (air), the cells behaved as if they had been grown in high CO2. They showed an oxygen sensitive net photosynthesis and a high CO2 compensation point. This indicates that carbonic anhydrase plays an important role in the regulation of a measurable photorespiration in Chlamydomonas. The results are discussed in relation to previous observations of photorespiration measured by enzyme assay, metabolic products and gas exchange properties.  相似文献

Two experimental series were run to evaluate the potential of algal development on dissolved organic nitrogen (DON) compounds as the sole source of nitrogen (N) nutrition. Monocultures of several common Lake Kinneret algae (Pediastrum duplex, Synechococcus sp., Microcystis aeruginosa, Aphanizomenon ovalisporum and Cyclotella sp.) were incubated for 3 weeks in the laboratory with different inorganic (NH4+, NO3-) or organic (hypoxanthine, urea, guanine, ornithine, glucosamine, lysine) nitrogen sources. Even though the cultures were not axenic, marked differences were observed in algal growth response. Pediastrum, Cyclotella and Aphanizomenon grew well on most N sources, and cyanobacterial growth and yield were consistently greatest when the urea was the only N source. We also followed algal growth and eventual species dominance in batch samples of GF/F-filtered lake water, supplemented with orthophosphate and different inorganic or organic N compounds and inoculated with concentrated lake phytoplankton. Although no clear impact on phytoplankton growth (as chlorophyll concentration) was observed, in seven out of 11 experiments we could discern changes in the algal species that became dominant in flasks with different organic and inorganic N sources. Our results are consistent with the proposition that components of the DON pool are not only an important potential, direct or indirect N source for phytoplankton, but also that different algal species can exploit these sources with varying capabilities so that different N substrates may selectively stimulate the development of dominant algal species.   相似文献

摘要:【目的】研究三峡水库神农溪库湾春季水华期间浮游藻类演替及其成因分析。【方法】2014年3–5月在神农溪库湾布置了6个断面(SN01–SN06),在神农溪汇入长江干流河口附近水域设置1个断面CJBD,对浮游藻类、相关环境因子及水动力因子进行了同步监测,据此分析了水体层化结构及水动力特性。【结果】神农溪在监测时段内共检测到浮游藻类6门38种(属);库湾浮游藻类生物量时间上差异显著(ANOVA,P<0.05)。春季浮游藻类群落结构具有明显的演替规律,3月份暴发大面积的硅藻水华(藻密度>100×105 cells/L),小环藻(Cyclotella spp.)为优势藻种;4月在SN02–SN06暴发以小球藻(Chlorella spp.)为主要优势种、衣藻(Chlamydomonas spp.)为次优势种的绿藻水华(藻密度>100×105 cells/L),5月份受水位大幅消落影响,浮游藻类生物量降低且无明显优势藻种。【结论】在具备充足的营养盐的水体中,水体层化结构与水动力特性对浮游藻类演替影响重大。三峡水库水位处于快速消落阶段时,流速成为抑制神农溪库湾藻类生长的主要因素。  相似文献

Eighteen long-term competition experiments were performed on two freshwater algae, a blue-green alga, Anabaena flos-aquae, and a diatom, Cyclotella sp., under controlled light and temperature conditions and various nutrient limitations. As predicted, Anabaena displaced Cyclotella when nitrate was in short supply to both species, whereas Cyclotella became dominant when both species were phosphate-limited. The two species stably coexisted when phosphate and silicate were limited. Anabaena either displaced or coexisted with Cyclotella when nitrate and phosphate or nitrate, phosphate and silicate were limited, depending on their initial density ratios. This study revealed strong effects of initial population densities on the outcomes of algal competition for limiting nutrients.  相似文献

比较河流浮游藻类和着生藻类群落的时空格局及其与环境因子关系的差异,有助于了解两类藻的区别与联系。然而,目前这方面的研究还不多。基于2019年秋季和2020年夏季金沙江上段干流17个样点藻类及水体理化指标的调查数据,分析了不同季节浮游藻类和着生藻类群落结构及其主要环境驱动因子,比较了两类藻的多样性格局及其与环境关系的异同。结果发现,调查河段的浮游藻类和着生藻类均以硅藻为主,其中浮游藻类以极小曲壳藻(Achnanthes minutissima)、钝脆杆藻(Fragilaria capucina)、适中舟形藻(Navicula accomoda)为主要优势种,着生藻类以极小曲壳藻(Achnanthes minutissima)、扁圆卵形藻(Cocconeis placentula)、橄榄绿色异极藻(Gomphonema olivaceum)为主要优势种。浮游藻类和着生藻类秋季平均密度分别为:2.41×10~5个/L、9.43×10~3个/cm~2,均明显高于夏季的平均密度(4.84×10~4个/L、4.84×10~3个/cm~2)。两类藻的群落格局表现出明显的季节变化,但只有着生藻分类单元...  相似文献

研究了北京市区沿城乡梯度带河流底栖硅藻的空间分布及其季节变化特征。通过在2014年的春季、夏季和秋季在山区对照溪流、城市上游河道和城市下游河道3个样区,共23个样点进行了底栖硅藻样品采集。分析表明,主要优势种的空间差异明显。在对照溪流,主要优势种为极小曲丝藻(Achnanthidium minutissimum);在城市上游河道,主要优势种分别为短文假十字脆杆藻(Pseudostaurosira brevistriata)和连结十字脆杆藻腹面变种(Staurosira construens var.venter);在城市下游河道,主要优势种为谷皮菱形藻(Nitzschia palea)。然而,3个样区内的主要优势种相对丰度季节变化较小(P0.05)。其中,A.minutissimum及P.brevistriata的季节变化特征均为春季秋季夏季,而S.construens var.venter及N.palea的季节变化特征均为春季夏季秋季。结果发现,北京市城乡梯度带的河流硅藻种类组成差异较大,但主要优势属种季节变化相对较小,表明硅藻种类能较好地反映城乡梯度带的河流环境变化状况,可用于北京市河流水质生物监测。  相似文献

The composition of the diatom community on the bryozoans Electra pilosa, Membranipora membranacea, Flustra foliacea, and Alcyonidium gelatinosum from the German Bight was studied by light and scanning electron microscopy. In total, members of 26 diatom genera were found, with Cocconeis, Tabularia, Licmophora, Amphora, and Navicula being the most abundant. The amount and the composition of the diatom covering seem to be typical for single bryozoan species. Electra pilosa and Membranipora membranacea showed a rather dense covering with 71–547 cells/mm2 and 77–110 cells/mm2, respectively. The most prominent genus on Electra pilosa was Cocconeis, reaching up to 58% of all diatoms in one sample, followed by Navicula, Tabularia and Amphora. The most abundant genera on Membranipora membranacea were Tabularia and Licmophora, making up almost 70% of all diatoms in one sample, followed by Navicula, Cocconeis and Amphora. The diatom composition was very stable on all Electra samples, but varied on Membranipora samples. With <1–27 cells/mm2, diatoms were much less abundant on Alcyonidium gelatinosum. Members of the genera Tabularia and Navicula were the most frequently found benthic diatoms, whereas the planktonic forms Coscinodiscus, Cyclotella, and Thalassiosira made up 35% of the diatoms. On Flustra foliacea, diatoms were virtually absent, with fewer than 5 cells/mm2. The low diatom numbers are probably due to toxic metabolites produced by the host. In the phytoplankton of northeastern West Siberian water bodies, Bacillariophyta were a dominant group of the 7 most abundant algal divisions, with the occurrence of the members of the class Centrophyceae varying from 27 to 100%. Scanning electron microscopy revealed complete species composition of the class Centrophyceae that dominates the phytoplankton of Lake Delingde, the Delingde and Bol’shaya Kheta rivers (Aulacoseira, 3 species; Cyclostephanos, 1; Cyclotella, 5; Discostella, 1; Melosira, 1; Pliocaenicus, 1; Puncticulata, 1; Stephanodiscus, 6). The phytoplankton of the class Centrophyceae in the Taz River now has a new member, Puncticulata radiosa. New species of the genus Cyclotella in West Siberian algal flora were revealed: C. arctica, C. comensis, and C. vorticosa. The findings helped more precisely define the ranges of the rare taxa and add new diagnostic features to the diagnoses of C. comensis and Stephanodiscus invisitatus.  相似文献

An algicidal bacterium Pseudomonas fluorescens HYK0210-SK09 (SK09) was applied to a natural bloom of Stephanodiscus hantzschii using a small-scale mesocosm (SM) and a large-scale mesocosm (LM) to clarify the algicidal effects and evaluate the response of the planktonic community and environments. When SK09 cells were inoculated at a final concentration of 5 × 106 cells mL−1, the abundance of S. hantzschii decreased significantly by 95% in SM and 85% in LM. The microcosm in the laboratory revealed that the abundance of Pseudomonas increased rapidly after inoculation with a corresponding decrease in the S. hantzschii population. Nutrient concentrations increased following the decline in the diatom cells. The abundances of nondominant species such as Chlamydomonas, Cryptomonas, and Navicula increased slightly with increased nutrient concentrations. The abundance of heterotrophic protists also increased significantly due to utilization of SK09 as food. The present study demonstrates that SK09 is an effective biocontrol agent for natural S. hantzschii bloom, and grazing pressure plays a crucial role in the successful application of algicidal bacteria to natural environments.  相似文献

The population dynamics of a planktonic rotifer (Polyarthra vulgaris) were examined in a brown water, acid lake in northern Michigan, U.S.A. Predation by Chaoborus punctipennis and low food (Navicula spp. and Cyclotella spp.) concentrations were the main factors limiting P. vulgaris populations of all factors examined. Cross-reconstitution of the extrinsic proteins and Photosystem II (PS II) from a green alga, Chlamydomonas reinhardtii, and a higher plant,Spinacia oleracea, was performed to clarify the differences of binding properties of the extrinsic proteins between these two species of organisms. (1) Chlamydomonas PsbP and PsbQ directly bound to Chlamydomonas PS II independent of the other extrinsic proteins but not to spinach PS II. (2) Chlamydomonas PsbP and PsbQ directly bound to the functional sites of Chlamydomonas PS II independent of the origins of PsbO, while spinach PsbP and PsbQ only bound to non-functional sites on Chlamydomonas PS II. (3) Both Chlamydomonas PsbP and spinach PsbP functionally bound to spinach PS II in the presence of spinach PsbO. (4) While Chlamydomonas PsbP functionally bound to spinach PS II in the presence of Chlamydomonas PsbO, spinach PsbP bound loosely to spinach PS II in the presence of Chlamydomonas PsbO with no concomitant restoration of oxygen evolution. (5) Chlamydomonas PsbQ bound to spinach PS II in the presence of Chlamydomonas PsbP and PsbO or spinach PsbO but not to spinach PS II in the presence of spinach PsbP and Chlamydomonas PsbO or spinach PsbO. (6) Spinach PsbQ did not bind to spinach PS II in the presence of Chlamydomonas PsbO and PsbP. On the basis of these results, we showed a simplified scheme for binding patterns of the green algal and higher plant extrinsic proteins with respective PS II.  相似文献

We constructed a complete physical map and a partial gene map of the chloroplast genome of Cyclotella meneghiniana Kützing clone 1020-1a (Bacillariophyceae). The 128-kb circular molecule contains a 17-kb inverted repeat, which divides the genome into single copy regions of65 kb and 29 kb. This is the largest genome and inverted repeat found in any diatom examined to date. In addition to the 16S and 23S ribosomal RNA genes, the inverted repeat contains both the ndhD gene (as yet unexamined in other diatoms) and the psbA gene (located similarly in one of two other examined diatoms). The Cyclotella chloroplast genome exists as two equimolar populations of inversion isomers that differ in the relative orientation of their single copy sequences. This inversion heterogeneity presumably results from intramolecular recombination within the inverted repeat. For the first time, we map the ndhD, psaC, rpofi, rpoCl, and rpoC2 genes to the chloroplast genome of a chlorophyll c-containing alga. While the Cyclotella chloroplast genome retains some prokaryotic and land plant gene clusters and operons, it contains a highly rearranged gene order in the large and small single copy regions compared to all other examined diatom, algal, and land plant chloroplast genomes.  相似文献

The diatom flora of the Salton Sea, California   总被引:1,自引:0,他引:1  
We report on diatom species of the Salton Sea, a highly saline (43 g l–1) inland lake in California. We identified and photographed all diatom taxa encountered in the phytoplankton and benthos of the Salton Sea and its immediate tributaries. Ninety-four taxa were distinguished based on their morphological features using light- and electron microscopy. In the Salton Sea, there are four general categories of diatom assemblages related to their habitats: (1) A planktonic assemblage composed of Chaetoceros muelleri var. subsalsum, Cyclotella choctawhatcheeana, Cyclotella sp., Cylindrotheca closterium, Pleurosigma ambrosianum, Thalassionema sp.; (2) a benthic assemblage with diatoms that live on the bottom (e.g. genera Caloneis, Diploneis, Entomoneis, Gyrosigma, Plagiotropis, Pleurosigma, Surirella and Tryblionella), or in algal mats (Proschkinia bulnheimii, several species of Navicula and Seminavis gracilenta); (3) an epiphytic community attached to the macroscopic green algae which grow on the rocks and other hard surfaces near shore (e.g. Achnanthes brevipes, Licmophora ehrenbergii, Tabularia parva); and (4) a freshwater assemblage composed of species that get washed in by the rivers and other inflows discharging into the Sea (e.g. Achnanthidium minutissimum, Cocconeis pediculus, Cyclotella atomus, C. scaldensis, Nitzschia elegantula, T. weissflogii). The most striking feature of the phytoplankton is the abundance of species formerly known only from marine environments; this is not surprising given the high salinity and the peculiar history of the lake.  相似文献

The relative concentration of biliproteins, phycobilisomes, chlorophyll a, and reaction centers I and II are reported for Neoagardhiella bailyei, a macrophytic red alga collected in the field and compared with Anacystis nidulans, a cyanobacterium cultured in the laboratory. The ratios of chlorophyll to reaction center I, of chlorophyll to reaction center II, and the mass of phycobiliprotein per reaction center II are quite similar in Neoagardhiella and Anacystis, supporting the concept that the red algal chloroplast is derived from a cyanobacterial progenitor. The ratios of reaction center I to reaction center II are about 2.3 in both species. The Anacystis phycobilisome has about 40% of the mass of the Neoagardhiella phycobilisome, 4.9 and 14.9 × 106 daltons, respectively. The reaction center II/phycobilisome ratio is about 1.7 in Anacystis and 4.1 in Neoagardhiella. Phycobilisome size and physical restrictions on phycobilisome packing may be a major constraint on the reaction center II-phycobilisome association and the assembly of the photosynthetic membrane in both the red algae and cyanobacteria.  相似文献
