A syntaxonomic revision of vegetation of anthropogenic metalliferous habitats (mine spoils of lead, zinc and copper mining) in the Eastern Alps (Austria, Germany, Italy and Slovenia) was made. The communities studied belong to theThlaspietea rotundifolii (Linaria alpina-Cerastium uniflorum comm.,Papaveri kerneri-Thlaspietum kerneri, Violetum dubyanae, Thlaspietum cepaeifolii, Thlaspietum rotundifolii, Scrophulario juratensis-Erysimetum sylvestris, Minuartia gerardii-Silene glareosa comm.,Epipactido atrorubentis-Silenetum glareosae andSileno alpestris-Moehringietum muscosae) and to theAsplenietea trichomanis (Sileno rupestris-Asplenietum). There is neither floristic support nor syntaxonomic justification for the concepts of theGalio anisophylli-Minuartion vernae, theVioletalia calaminariae and theVioletea calaminariae in the Eastern Alps. These units should be included within theThlaspion rotundifolii, Thlaspietalia rotundifolii andThlaspietea rotundifolii, respectively.  相似文献

AvaI andBsoBI restriction endonucleases are isoschizomers which recognize the symmetric sequence 5′CYCGRG3′ and cleave between the first C and second Y to generate a four-base 5′ extension. TheAvaI restriction endonuclease gene (avaIR) and methylase gene (avaIM) were cloned intoEscherichia coli by the methylase selection method. TheBsoBI restriction endonuclease gene (bsoBIR) and part of theBsoBI methylase gene (bsoBIM) were cloned by the “endo-blue” method (SOS induction assay), and the remainder ofbsoBIM was cloned by inverse PCR. The nucleotide sequences of the two restriction-modification (RM) systems were determined. Comparisons of the predicted amino acid sequences indicated thatAvaI andBsoBI endonucleases share 55% identity, whereas the two methylases share 41% identity. Although the two systems show similarity in protein sequence, their gene organization differs. TheavaIM gene precedesavaIR in theAvaI RM system, while thebsoBIR gene is located upstream ofbsoBIM in theBsoBI RM system. BothAvaI andBsoBI methylases contain motifs conserved among the N4 cytosine methylases.  相似文献

InNeurospora crassa thenit-2 andnmr-1 (ms-5) loci represent the major control genes encoding regulatory proteins that allow the coordinated expression of various systems involved with the utilization of a secondary nitrogen source. In this paper we examine the effect of thenit-2 andms-5 (nmr-1 locus) mutations on the regulation of the ammonium assimilation enzymes, glutamine synthetase and glutamate dehydrogenase, which are regulated by the products of these genes; however, glutamate synthase is not so regulated. Glutamine synthetase and glutamate dehydrogenase levels are also regulated by the amino nitrogen content. We present evidence that thems-5 andgln r strains, which behave very similarly in their resistance to glutamine repression, are different and map in different loci.  相似文献

In petunia, a mitochondrial (mt) locus,S-Pcf, has been found to be strongly associated with cytoplasmic male sterility (CMS). TheS-Pcf locus consists of three open reading frames (ORF) that are co-transcribed. The first ORF,Pcf, contains parts of theatp9 andcoxII genes and an unidentified reading frame,urf-s. The second and third ORFs contain NADH dehydrogenase subunit 3 (nad3) and ribosomal protein S12 (rps12) sequences, respectively. Thenad3 andrps12 sequences included in theS-Pcf locus are identical to the corresponding sequences on the mt genome of fertile petunia. In both CMS and fertile petunia, only a single copy ofnad3 andrps12 has been detected on the physical map of the main mt genome. The origin of theurf-s sequence and the molecular events leading to the formation of the chimericS-Pcf locus are not known. This paper presents evidence indicating that two different mt sequences, related tourf-s and found in fertile petunia lines (orf-h and Rf-1), might have been involved in the molecular evolution of theS-Pcf locus. Southern analysis of mtDNA derived from both fertile and sterile petunia plants suggests that one of theseurf-s related sequences (showing 100% homology tourf-s and termedorf-h) is located on a sublimon. An additional, low-homologyurf-s related sequence (Rf-1) is shown to be located on the main mt genome 5′ to thenad3 gene. It is, thus, suggested that the sequence of events leading to the generation of theS-Pcf locus might have involved introduction of theorf-h sequence, via homologous recombination, into the main mt genome 5′ tonad3 at the region where the Rf-1 sequence is located.  相似文献

Restriction endonuclease fragment length polymorphisms (RFLPs) were found using the cDNA probe Hox-2.1 for the homeo box-2.1 gene in the mouse. Polymorphism was detected in restriction patterns generated by fragments fromHindIII digestion. The great majority of laboratory strains of mice carries theHox-2.1 a allele. Only two laboratory strains carry theHox-2.1 b allele. Among strains of wild origin, the European subspecies (Mus m. domesticus, M. m. brevirostris, andM. m. musculus) and some Asian subspecies (M. m. castaneus) carry theHox-2.1 a allele. The subspecies from Far Eastern countries (M. m. molossinus, Chinese mice of wild origin, andM. m. yamashinai) carry theHox-2.1 ballele. Using the RFLP, theHox-2.1 gene was mapped on chromosome 11. Three-point cross test data showed that the recombination frequency is 29.6% between theHba and theHox-2.1 genes and 23.5% between theHox-2.1 and theEs-3 genes. The gene order ofHba-Hox-2.1-Es-3 has been confirmed.  相似文献

Casein kinase II is composed of two catalytic (a) and two regulatory (Β) subunits, the amino acid sequences of theα andΒ subunits are highly conserved between species. To examine whether heterologous casein kinase II could be formed, recombinantα andΒ subunits from human andDrosophila were reconstituted from inclusion bodies. Casein kinase II containing either humanα andDrosophila Β orDrosophila α and humanΒ subunits exhibited enzymatic properties similar to those of the homologous holoenzymes with regard to specific activity, salt optima, and autophosphorylation. However, renaturation and reconstitution of casein kinase II was dependent on the type ofΒ subunits and the redox conditions, with theDrosophila Β subunits requiring more reduced conditions. ChimericΒ subunits prepared from human andDrosophila cDNA revealed that the N-terminal region was responsible for the requirement for the reduced redox state during renaturation. TheN-terminal region also affected solubility and electrophoretic mobility of theΒ subunit.  相似文献

This study characterises the bacteria associated with a marine hatchery in Tunisian coastal marine waters. Presumptive vibrios (TCBS agar) and heterotrophic aerobic microflora (CFU) were studied at different stages within the hatchery: seawater, batches of algal cultures, rotifers andArtemia culture tanks. The bacterial strains were isolated on TCBS Agar plates and described using different bacteriological tests (standardised micromethods “API 20 E Strips”, exoenzymes production, growth at different temperatures, pH and salinity, vibriostatic agent O/129 and antibiotics susceptibility). Two dominant genera of bacteria were found (Vibrio andAeromonas) associated with some strains of thePseudomonadaceae family.Vibrio alginolyticus was the dominant bacteria (75% of total isolates) found in rotifers (Brachionus plicatilis) andArtemia cultures (Artemia salina). In larvae rearing tanks, an increase ofVibrionaceae was noted after larvae were fed withArtemia. Most of the studied bacteria used the skin mucus ofSparus aurata larvae as their sole source of carbon. All theV. alginolyticus strains were β-haemolytic, hydrolyse the DNA and were susceptible to several tested antibiotics.  相似文献

The sporoderm ultrastructure of some genera in theLauraceae, Hernandiaceae andGomortegaceae (Laurales) was re-investigated. This and a comparison with some Zingiberalean genera (Canna, Heliconia, Strelitzia) leads to a re-interpretation of the peculiar pollen wall structure of the Lauralean families recently described in this journal (Kubitzki 1981). Their pollen is now interpreted as being nearly exine-less: The (sporopollenin-containing) exine consists only of a thin, skin-like, coherent layer ornamented with spines and globules. The (cellulosic) intine is composed of an unstratified inner and a thick, channelled outer layer. The striking similarity of the pollen ultrastructure between theLaurales studied and some members of theZingiberales can be interpreted as an example of convergent evolution in angiosperm pollen walls.  相似文献

The organization of the phosphate-specific transport (pst) operon inPseudomonas aeruginosa has been determined. The gene order of thepst operon ispstC, pstA, pstB, phoU, and a well-conserved Pho box sequence (16/18 bases identical) exists in the promoter region. The most striking difference from the knownEscherichia coli pst operon is the lack of thepstS gene encoding a periplasmic phosphate (Pi)-binding protein. Even though the threepst genes were absolutely required for Pi-specific transport, expression of thepst operon at high levels did not increase Pi uptake inP. aeruginosa. DNA sequences for thepstB andphoU genes have been determined previously. The newly identifiedpstC andpstA genes encode possible integral membrane proteins of 677 amino acids (M r 73 844) and 513 amino acids (M r 56 394), respectively. The amino acid sequences of PstC and PstA predict that these proteins contain a long hydrophilic domain not seen in theirE. coli counterparts. A chromosomal deletion of the entirepst operon renderedP. aeruginosa unable to repress Pi taxis under conditions of Pi excess. ThephoU andpstB genes are essential for repressing Pi taxis. However, mutants lacking either PstC or PstA alone were able to repress Pi taxis under conditions of Pi excess.  相似文献

The sieve-element plastids of 126 species of theArales were investigated by transmission electron microscopy. With the exception ofPistia (with S-type plastids) all contained the monocotyledon specific subtype-P2 plastids characterized by cuneate protein crystals. While the species studied from bothAcoraceae andLemnaceae have form-P2c plastids (i.e., with cuneate crystals only), those of theAraceae belong to either form P2c (14 species), P2cs (the great majority) or P2cfs (Monstera deliciosa, only, with form-P2cs plastids in the otherMonstera species studied). The form-P2cs plastids of theAraceae are grouped into different categories according to the quantity and quality of their protein and starch contents. The subfamilyLasioideae is redefined to comprise all aroid P2c-taxa and those P2cs-genera that contain only one or very few starch grains. Only little starch is also recorded in the sieve-element plastids ofGymnostachys (Gymnostachydoideae), with the other plastid data denying a close relationship toAcorus. While equal amounts of starch and protein are generally present in sieve-element plastids of the subfamiliesPothoideae, Monsteroideae, Colocasioideae, Philodendroideae, andAroideae, maximum starch content and only very few protein crystals are found in form-P2cs plastids ofCalla (Calloideae),Ariopsis (Aroideae), andRemusatia (Colocasioideae?). In the latter, both morphology and size of sieve-element plastids are close to those ofPistia.—In theAraceae the diameters of the sieve-element plastids exhibit a great size range, but are consistent within a species and within a defined part of the plant body. Comparative data are mainly available for stem and petiole sieve-element plastids.—The accumulated data are used to suggest an affiliation of the species to subfamilies and to discuss the phylogeny of theArales. Forms and sizes of their plastids support a separation of bothAcoraceae andLemnaceae from theAraceae. The presence of S-type plastids inPistia does not favour direct and close relationships to the form-P2c genusLemna.—The prevailing form-P2cs plastids might support proposals that place theArales (together with also form-P2cs plastid containingDioscoreales) in the neighbourhood of basal dicotyledons. BesidesAsarum andSaruma (Aristolochiaceae), with monocotyledonous form-P2c plastids,Pistia (with dicotyledonous S-type plastids) gives another example for a link between the two angiosperm classes.  相似文献

One of the four ribosomal RNA operons (rrnA) from theAgrobacterium vitis vitopine strain S4 was sequenced.rrnA is most closely related to therrn operons ofBradyrhizobium japonicum andRhodobacter sphaeroides and carries an fMet-tRNA gene downstream of its 5S gene, as in the case ofR. sphaeroides. The 16S rRNA sequence of S4 differs from theA. vitis K309 type strain sequence by only one nucleotide, in spite of the fact that S4 and K309 have very different Ti plasmids. The predicted secondary structure of the S4 23S rRNA shows several features that are specific for the alpha proteobacteria, and an unusual branched structure in the universal B8 stem. The 3′ ends of the three otherrrn copies of S4 were also cloned and sequenced. Sequence comparison delimits the 3′ ends of the four repeats and defines two groups:rrnA/rrnB andrrnC/rrnD.  相似文献

The sieve-element plastids of members of several genera in theBuxaceae (Buxus, Pachysandra andSarcococca) were found to be of the specific subtype PVI, which contains a central globular protein crystal.Simmondsia (Simmondsiaceae) andDaphniphyllum (Daphniphyllaceae), on the other hand, were found to contain S-type sieve-element plastids. The occurrence of the highly restricted PVI plastids in theBuxaceae mitigates against a close relationship between theBuxaceae andSimmondsia, Daphniphyllum andEuphorbiaceae. Exine sculpturing of theBuxaceae andSimmondsiaceae also shows no close similarities. Both of these EM characters are discussed in connection with other available data and with respect to earlier systematic treatment of these families.  相似文献

Multiple sclerosis patients-111 of them-were typed for theHLA-D allelesw1, w2, andw3 and a new determinant, EI. Statistically significant increase was obtained for thew2 andw3 frequencies as compared to healthy controls. The distribution of HLA-D phenotypes among MS patients revealed a good fit according to the Hardy-Weinberg law. By calculating linkage disequilibrium parameters, theHLA-B7,Dw2 allele combination was found to be more closely associated than in normal controls, whereas forHLA-Bw35, Dw1, no linkage disequilibrium could be detected in the patients' group. From these data we conclude that in multiple sclerosis, the disturbance affects the frequency ofDw3 and the gametic association between alleles of theHLA-B andD loci, as well as the already known increase ofHLA-B7 andDw2.  相似文献
