在实验室控制条件下,以转Bt基因棉花GK12、33B、SGK321上的烟粉虱为食料饲养龟纹瓢虫,研究转Bt基因棉花上的烟粉虱对龟纹瓢虫生长发育及捕食的影响,同时利用Y型嗅觉仪,观察龟纹瓢虫对来自不同类型棉花的棉叶及其烟粉虱的嗅觉反应和视觉反应。结果表明,转Bt基因棉花和对应的常规棉亲本上的烟粉虱对龟纹瓢虫的发育历期和存活率没有明显的影响。龟纹瓢虫对棉花叶片、烟粉虱若虫、蜜露、蜕等4种物质的视觉反应之间没有明显的差异。对4种嗅源物质的嗅觉选择性的大小依次为:烟粉虱若虫>蜕>棉花>蜜露;在两类不同的棉花之间,转基因棉花和常规棉,龟纹瓢虫对GK12、33B、SGK321等三种转基因棉花上的烟粉虱若虫的嗅觉选择性明显较对应的常规棉亲本SM3、33、SY321上的烟粉虱若虫小;对烟粉虱若虫的蜕,两种转单价基因棉花GK12、33B较对应的常规棉亲本上的小,而转双价基因的棉花SGK321上与对应的常规棉之间没有明显的差异。烟粉虱密度大于200头.皿-1时,龟纹瓢虫捕食转基因棉花上烟粉虱的数量大于对应的常规棉花亲本,但烟粉虱密度小于200头.皿-1时,龟纹瓢虫捕食转基因棉花上烟粉虱的数量小于对应的常规棉花亲本。龟纹瓢虫对烟粉虱的捕食符合HollingⅡ型反应。龟纹瓢虫取食转基因棉花上的烟粉虱的理论极限值、瞬间攻击率均大于常规棉花。  相似文献

转Cry1Ac活性杀虫蛋白及慈菇蛋白酶抑制剂B基因的棉花   总被引:6,自引:0,他引:6  
根据植物基因的结构特征。合成了Cry1Ac活性杀虫蛋白的编码序列并与内质网定位肽编码序列组成嵌合杀虫蛋白基因Bt29K。构建了含Bt29K基因及慈菇蛋白酶抑制剂B(API-B)基因表达框的双抗虫基因植物表达载体。通过根癌土壤杆菌(Agrobacterium trmefaciens(Smith et TOwnsend)ConnLBA4404)介导转化了棉花(Gossypium hirsu-tunL.)的两个生产品种(系)。根据抗棉铃虫(Heliothis armigera)试验及农艺性状的观察调查结果。经6代筛选,获得了抗棉铃虫90.0%_99.7%且农艺性状优良的9个双价抗虫棉纯合品系。分子生物学分析结果表明,两个抗虫基因在棉花基因组中的插入拷贝数为1个或2个,活性Cry1Ac和API-B蛋白在转基因抗虫棉株系中的表达量分别约占总可溶性蛋白的0.17%和0.09%。对双抗纯合系植株及仅转Bt基因的棉花纯合系抗虫性检测结果表明前者的抗虫性明显高于后者,因此推断本研究采用的双抗虫基因表达载体构建策略是合理的。  相似文献

棉铃虫田间种群Bt毒素Cry1Ac抗性基因频率的估算   总被引:7,自引:0,他引:7  
采用改进的F1筛查法检测了2005年采自华北地区的棉铃虫Helicoverpa armigera (Hübner)田间种群对Bt毒素Cry1Ac的抗性基因频率。2005年从河南安阳和河北沧县转Bt基因抗虫棉上采集二代棉铃虫卵,在室内用人工饲料饲养至2龄幼虫,用1 μg/cm2 的Cry1Ac活化毒素进行初筛,将初筛存活成虫与室内筛选的GYBT抗性品系成虫进行单对杂交,并用区分剂量(2.5 μg/cm2)对F1代进行检测。经检测,2005年河南安阳棉铃虫种群和河北沧县棉 铃虫种群对Cry1Ac抗性基因频率基本一致,分别为1.4×10-3和1.5×10-3。用毒素涂表法测定了2004、2005年采自河南安阳、河北高阳、河北沧县、新疆阿克苏和新疆沙湾棉铃虫田间种群对Cry1Ac活化毒素的敏感性水平,结果表明华北棉区与新疆内陆棉区棉铃虫种群对Cry1Ac的敏感性存在一定的地区性差异(<8倍)。总体上,我国华北棉区棉铃虫种群对Cry1Ac还未产生明显抗性,抗性基因频率处于正常水平。棉铃虫对转Bt基因抗虫棉的抗性风险依然存在,需要尽快启动全国性的早期抗性检测和预警工作。  相似文献

转Cry1Ac+Cry2Ab基因棉对棉蚜生命表参数及种群动态的影响   总被引:1,自引:0,他引:1  
为研究新型转Cry1Ac+Cry2Ab基因棉对棉蚜Aphis gossypii Glover生命表参数及种群动态的影响。2010—2011年以常规棉中棉所49为对照,对新型转Cry1Ac+Cry2Ab基因棉在室内进行了生物测定和田间进行了系统的调查。结果表明,和常规棉相比,转Cry1Ac+Cry2Ab基因棉花上棉蚜的净增值率降低81.69%,差异达显著水平;内禀增长率和周限增长率分别降低65.00%和13.01%,但差异不显著;平均世代周期和种群加倍时间分别增加5.54%和154.19%,后者差异达显著水平。和常规棉相比,2010年转Cry1Ac+Cry2Ab基因棉花百株苗蚜、伏蚜和秋蚜的数量分别降低10.79%、37.18%和17.49%,差异均未达显著水平;2011年转Cry1Ac+Cry2Ab基因棉花百株苗蚜的数量增加2.03%,伏蚜和秋蚜的数量分别降低37.41%和64.03%,差异均未达显著水平。  相似文献

【目的】转基因作物对非靶标昆虫的影响是转基因作物环境安全评价的重要内容,研究Cry2Ab蛋白对龟纹瓢虫的影响,对转基因作物的环境安全评价具有重要意义。【方法】采用实验动物学、分子生物学等方法,研究Cry2Ab蛋白对龟纹瓢虫发育历期、成虫体重、雌雄比例及体内氨基酸种类和含量的影响。【结果】与蔗糖对照组相比,Cry2Ab蛋白对龟纹瓢虫不同龄期的发育历期、成虫体重和雌雄比例均无明显差异,对体内氨基酸种类和含量也没有显著差异。【结论】Cry2Ab蛋白对龟纹瓢虫的生长发育及代谢无显著影响。  相似文献

文章以转Cry1Ac基因棉(中棉所41)和常规棉(中棉所49)为对照,研究了转Cry1Ac+Cry2Ab基因棉(639020)在棉花生长的关键时期——蕾期(二代棉铃虫发生期)、花期(三代棉铃虫发生期)和花铃期(四代棉铃虫发生期)对棉铃虫的控制作用,同时研究了639020棉田主要捕食性天敌(中华草蛉幼虫、龟纹瓢虫、小花蝽和草间小黑蛛)对烟粉虱的捕食功能,明确了639020棉花在生长的关键时期对棉铃虫的控制效果及对棉田主要捕食性天敌捕食功能反应的影响。结果表明,639020棉花对二代和三代棉铃虫具有良好的控制作用,抗虫性分别比中棉所41提高了52.85%和16.22%,其中前者差异达显著水平,后者差异不显著。在棉花蕾期、花期和花铃期,639020棉田棉铃虫落卵量都比中棉所41棉田和中棉所49棉田低(除二代棉铃虫发生期);棉铃虫幼虫数量都极显著低于常规棉,且都低于防治指标,但与中棉所41棉田无显著差异。639020棉田中华草蛉、龟纹瓢虫、小花蝽和草间小黑蛛对烟粉虱的捕食功能与中棉所41棉田和常规棉田相比无显著变化。研究结果以期为新型转基因棉花环境安全性研究及其外源基因的抗虫遗传效应和生产应用前景进行安全性评价。  相似文献

室内研究了中华草蛉Chrysopa sinicaTjeder、龟纹瓢虫Propylaea japonica(Thunberg)和异色瓢虫Leis axyridis(Pallas)对B型烟粉虱Bemisiatabaci(Gennadius)若虫的捕食功能反应和寻找效应。结果表明,3种天敌成虫和幼虫的捕食量均随着猎物密度的增加而上升,当猎物增加到一定水平,捕食量趋向稳定,捕食功能反应曲线符合HollingⅡ型方程。异色瓢虫、中华草蛉和龟纹瓢虫成虫和幼虫对烟粉虱若虫的捕食量差异显著,理论最大日捕食量分别为417,263,156头和625,238,108头。3种天敌对烟粉虱若虫的寻找效应随着天敌密度的增加而下降,干扰作用逐渐增强,其寻找效应表现为龟纹瓢虫(0.5656)>异色瓢虫(0.4371)>中华草蛉(0.4029),对烟粉虱的控制能力表现为异色瓢虫>龟纹瓢虫>中华草蛉。  相似文献

【背景】转基因棉花在商业化种植之前,必须评价其环境安全性。其中新型棉花材料的生存竞争能力和对物种丰富度的影响是评价的重要内容。【方法】以转Cry1Ac+Cry2Ab基因棉为试验材料,转Cry1Ac棉花中棉所41和非转基因棉花中棉所49为对照品种,分别于2014年5~9月对棉花株高、主茎叶片数、叶绿素含量、比叶面积、果枝数、蕾铃数等生长参数进行比较,同时对二代、三代和四代棉铃虫发生期棉田物种丰富度进行系统调查。【结果】转Cry1Ac+Cry2Ab棉花的生长势与转Cry1Ac棉花和非转基因棉花基本相当,没有表现出明显的竞争优势;产量构成参数在成铃和脱落等方面比非转基因棉表现出良好的优势。对棉田节肢动物物种丰富度的影响表明,转Cry1Ac+Cry2Ab棉花对靶标害虫棉铃虫具有良好的控制效果,对主要刺吸性害虫棉蚜、棉蓟马、烟粉虱、绿盲蝽与天敌龟纹瓢虫、草间小黑蛛、草蛉和小花蝽等的种群丰富度在个别时期有所影响,但总体上与转Cry1Ac棉田和非转基因棉田没有显著性差异。【结论与意义】转Cry1Ac+Cry2Ab棉花无竞争优势,但目标性状优势较好;对棉田节肢动物物种丰富度无明显影响。研究结果为新型转Cry1Ac+Cry2Ab棉花对棉田环境安全方面的研究进一步补充了内容,为转基因棉花的环境安全评价提供科学数据。  相似文献

在室内对转基因水稻KMD1中的Cry1Ab毒蛋白经食物链在几种主要害虫及其捕食性天敌体内的积累进行了研究。结果表明: 无论是水稻孕穗期还是成熟期,二化螟Chilo suppressalis连续取食KMD1或取食KMD1.36 h后移至对照品种秀水11上取食不同时间后,幼虫体内的Cry1Ab含量均随取食时间延长逐渐下降。稻眼蝶 Mycalesis gotama幼虫连续取食KMD1或在KMD1上取食两天后移至秀水11上继续取食不同时间,体内的Cry1Ab含量也都随取食时间延长而下降,但下降速度较二化螟更快。取食KMD1的二化螟和稻眼蝶幼虫的粪便中均检测到较高浓度的Cry1Ab,对照组中均无Cry1Ab。取食KMD1的二化螟幼虫血淋巴中检测到Cry1Ab,含量为3.5 ng/g。取食KMD1的褐飞虱 Nilaparvata lugens、稻蚜Sitobion avenae以及饲喂取食过KMD1的二化螟或稻眼蝶幼虫的拟水狼蛛Pirata subpiraticus体内都含有一定浓度的Cry1Ab,其中,拟水狼蛛体内的CrylAb含量以饲喂取食KMD1稻眼蝶幼虫的含量最高,约为饲喂取食KMD1二化螟幼虫的60倍。这些结果表明Cry1Ab可以沿水稻害虫天敌食物链传递。  相似文献

本研究比较了转Bt Cry1Ac+CP_4EPSPS基因抗棉铃虫抗草甘膦棉花与常规棉花在新疆棉区对草甘膦的耐受性差异。两年的研究结果表明,转Bt Cry1Ac+CP_4EPSPS基因抗棉铃虫抗草甘膦棉花对草甘膦有较好的耐受性,而对照棉花中棉所49对草甘膦耐受性较差。苗期喷施草甘膦后转基因抗棉铃虫抗草甘膦棉花生长发育没有受到影响,而对照棉花中棉所49喷施草甘膦后生长发育受到了明显的影响,个别植株甚至死亡。转Bt Cry1Ac+CP_4EPSPS基因抗棉铃虫抗草甘膦棉花株高、真叶数、蕾数、产量等指标与对照相比差异显著。转Bt Cry1Ac+CP_4EPSPS基因棉花对草甘膦的耐受程度显著高于非转基因棉花。草甘膦对转基因抗草甘膦棉花无负面影响。  相似文献

With the cultivation of Bt cotton, the produced insecticidal Cry proteins are ingested by herbivores and potentially transferred along the food chain to natural enemies, such as predators. In laboratory experiments with Bollgard II cotton, concentrations of Cry1Ac and Cry2Ab were measured in Lepidoptera larvae (Spodoptera littoralis, Heliothis virescens), plant bugs (Euschistus heros), aphids (Aphis gossypii), whiteflies (Bemisia tabaci), thrips (Thrips tabaci, Frankliniella occidentalis), and spider mites (Tetranychus urticae). Tritrophic experiments were conducted with caterpillars of S. littoralis as prey and larvae of ladybird beetles (Harmonia axyridis, Adalia bipunctata) and lacewings (Chrysoperla carnea) as predators. Immunological measurements (ELISA) indicated that herbivores feeding on Bt cotton contained 5%–50% of the Bt protein concentrations in leaves except whiteflies and aphids, which contained no or only traces of Bt protein, and spider mites, which contained 7 times more Cry1Ac than leaves. Similarly, predators contained 1%–30% of the Cry protein concentration in prey. For the nontarget risk assessment, this indicates that Bt protein concentrations decrease considerably from one trophic level to the next in the food web, except for spider mites that contain Bt protein concentrations higher than those measured in the leaves. Exposure of phloem sucking hemipterans is negligible.  相似文献

The efficacy of Cry1Ac Bacillus thuringiensis (Bt) cotton plants against field populations of Helicoverpa armigera (Hübner) has been inconsistent over the growing season. Any reduction in efficacy (where efficacy is the capacity of the plant to affect the survival of the insect) increases the opportunities for H. armigera to evolve resistance to Bt toxin. Changes in efficacy could be due to changes at the level of gene expression and/or in the physiological makeup of the plant and may be induced by environmental conditions. Two environmental factors, temperature and insect damage, were investigated. Temperature was found to affect efficacy, whether plants were grown at different temperatures continuously or were exposed to a change in temperature for a short period. Damage caused by chewing insects (H. armigera larvae) produced a dramatic increase in the efficacy of presquare Bt cotton. In contrast, damage by sucking insects (aphids) did not induce changes in efficacy. Changes in efficacy seemed to be mediated through modification of the physiological background of the plant rather than changes in the level of Cry1Ac expression or in the concentration of the Bt toxin. The impact of the non-Bt responses of plants on strains of H. armigera should be evaluated. It is possible that by enhancing existing defensive mechanisms of plants, the rate of evolution of resistance to Bt toxins could be retarded by increasing the plants overall toxicity through the additive effects of the toxins and plant defenses.  相似文献

The movement of Bacillus thuringiensis (Berliner) (Bt) Cry1Ac endotoxin through high trophic levels was assessed to help elucidate the effects of Bt toxin on non‐target insects. The diamondback moth (Plutella xylostella L., Lepidoptera: Plutellidae), the parasitic wasp (Cotesia vestalis Haliday, Hymenoptera: Braconidae) and the predatory green lacewing Chrysoperla carnea (Stephen) (Neuroptera: Chrysopidae) were used as a model system in this laboratory study. Bt‐resistant P. xylostella larvae fed Cry1Ac‐expressing transgenic oilseed rape (OSR, Brassica napus L., Cruciferae), before and after parasitization by C. vestalis, consumed Cry1Ac with the ingested plant material but only a proportion of Cry1Ac consumed was recovered from the bodies and faeces of P. xylostella larvae. Cry1Ac was not detected in newly emerged parasitoid larvae. In contrast, Cry1Ac was detected in C. carnea larvae fed on resistant P. xylostella larvae reared on Bt OSR. However, no Cry1Ac could be detected in C. carnea larvae when the lacewings were transferred to P. xylostella larvae reared on conventional OSR and tested 24–48 h. The metabolizing ability of Cry1Ac is discussed for the larvae of P. xylostella and C. carnea.  相似文献

Bt毒蛋白在转Bt基因棉中的表达及其在害虫-天敌间的转移   总被引:16,自引:2,他引:14  
以常规棉泗棉3号作为对照,采用酶联免疫生测法(ELISA)和室内生物测定法,研究了转Bt基因棉新棉33B和GK-12不同组织器官中Cry1Ac或Cry1Ab毒蛋白的表达及其向靶标害虫(棉铃虫)、非靶标害虫(棉蚜)以及天敌(龟纹瓢虫)的传递和影响。研究结果表明,新棉33B各组织器官中Bt毒蛋白的表达量较高,为79.7~1 390.0 ng/g鲜重,GK-012较低为165~2640 ng/g鲜重。在花盛期,新棉33B各组织器官中Bt毒蛋白的表达量依次为:柱头、花 >子房、花瓣>群尖;而5~7叶期的初展嫩叶、现蕾初期的幼蕾及花铃期的幼铃表达量相当,而且与花盛期群尖的表达量没有明显区别。同样处于花盛期的GK-12,其各组织器官中Bt毒蛋白的表达量依次为:花药>柱头>花瓣>群尖>子房;而5~7叶期的初展嫩叶、现蕾初期的幼蕾及花铃期的幼铃表达量相当,而且与花盛期群尖的表达量没有明显差异。常规对照棉的幼铃、花药、柱头以及子房中痕量Bt毒蛋白的存在可能与传粉昆虫等的活动有关。在转Bt基因棉田采集的棉蚜和棉铃虫老龄幼虫,其体内均可检测到Bt毒蛋白;在新棉33B棉田采集的龟纹瓢虫幼虫和成虫体内也可检测出Bt毒素。当以Bt棉田的棉蚜饲喂龟纹瓢虫时,龟纹瓢虫的生长发育、存活以及繁殖等基本没有受到影响。  相似文献

Abstract Stylet penetration behaviors of Bemisia tabaci biotype B on two transgenic cotton lines “GK12” and “GK19” expressing Bt toxic protein Cry1A (Bt cotton) and a non-Bt conventional cotton line “Simian-3” (CK cotton) were recorded with the direct current electrical penetration graph (DC-EPG) technique. Our results suggested that EPG waveform patterns, types and characteristics [non-probe (NP), pathway (C), potential drops (pd) and phloem phase (E(pd))] of Bemisia tabaci biotype B were very similar on the three cotton lines. There were no obvious differences of pathway variables among whiteflies on the three cotton lines. Some phloem variables related to E(pd)1 differed. Duration of 1st E(pd)1 and mean duration of E(pd)1 on both GK12 and GK19 were significantly shorter than that on CK cotton (P < 0.05). Fewer whiteflies on GK have long E(pd)1. Other phloem variables including total duration of E(pd) summed, mean E(pd) duration and percentage of whiteflies reaching the phloem phase were similar among the three cotton lines.  相似文献
