Summary Unambiguous methodologies were developed for the accurate and reproducible determination of specific D-amino acids in the physiological fluids of common laboratory rodents. Depending on the strain of rodent and the type of amino acid examined, excreted D-amino acids ranged from the low percent levels to over 40 percent of the total specific amino acid level. Relative plasma levels tended to be considerably lower, typically an order of magnitude less. A number of factors were found to alter the relative amounts of excreted D-amino acids. This included: diet, age, pregnancy, advanced cancer, and antibiotics. The two factors that seemed to result in substantially lower levels of excreted D-amino acids were fasting and young age. Pregnancy was the only factor that consistently resulted in higher relative D-amino acid excretion. Much of the observed data are believed to be related to the efficiency with which the kidney reabsorbs L-amino acids. No claims are made as to the meaning and/or importance of free D-amino acids in regards to pathology, age, clinical usefulness and so forth. However, a knowledge of normal D-amino acid levels and dynamics is necessary before it is possible to identify perturbations caused by either natural or pathological conditions. Growth and nitrogenase activity (acetylene reduction) of Azotobacter vinelandii in chemically defined N-free media were studied in the presence of p-hydroxybenzoic, vanillic, p-coumaric, and ferulic acids at concentrations from 0.01 to 1% (w/v). Growth and nitrogenase activity were only detected when the microorganism was cultured on p-hydroxybenzoic acid either as sole carbon source or mixed with other phenolic acids, suggesting that p-hydroxybenzoic acid could be utilized as a carbon source by A. vinelandii for growing under certain environmental conditions. 相似文献

In the present study to search for the source of free D-amino acids, D- and L-enantiomers of the major non-essential amino acids, i.e., the free form of serine, alanine, proline, aspartate and glutamate were analyzed by HPLC in human saliva, submandibular glands and oral epithelial cells. The D-enantiomer ratios to total of free alanine or proline were 35% and 20%, respectively, in saliva. The ratios of the other D-amino acids were less than 11%. The effect of ingested food and oral bacteria on the saliva amino acid levels was suggested to be insignificant. D-Alanine and d-aspartate were also detected in the submandibular gland in ratios up to 5%, and D-alanine and d-proline were found in oral epithelial cells in ratios of 18% and 5%, respectively. The submandibular gland and oral epithelial cells are suggested to be possible sources of the saliva D-alanine and D-aspartate. 相似文献

Chromatographic determination of L- and D-amino acids in plants 总被引:1,自引:0,他引:1
Quantities of free L- and D-amino acids (L- and D-AAs) in plants (leaves of coniferous and decidious trees, fleshy fruits, leaf blades of fodder grasses, and seeds and seedlings of edible legumes) were determined. Amino acid (AA) enantiomers were converted into diastereomers using pre-column derivatization with o-phthaldialdehyde together with N-isobutyryl-L(or D)-cysteine followed by separation of the resulting fluorescent isoindol derivatives on an octadecylsilyl stationary phase using high-performance liquid chromatography. Relative amounts of D-AAs were also determined by enantioselective gas chromatography-mass spectrometry on Chirasil-L-Val. Free D-AAs acids in the range of about 0.2% up to 8% relative to the corresponding L-AAs acids were found in plants. D-Asp, D-Asn, D-Glu, D-Gln, D-Ser and D-Ala could be detected in most of the plants, and D-Pro, D-Val, D-Leu and D-Lys in certain plants. As D-AAs were detected in gymnosperms as well as mono- and dicotyledonous angiosperms of major plant families it is concluded that free D-AAs in the low percentage range are principle constituents of plants. 相似文献

Summary. The concentrations of free D- and L-amino acids were determined in the gastric juice from four groups: patients suffering
from early gastric carcinoma with or without Helicobacter pylori infection, and patients without carcinoma but with peptic ulcers, duodenal ulcers or chronic gastritis with or without H. pylori infection. H. pylori is a bacterium associated with gastric inflammation and peptic ulcers and is a risk factor for stomach cancer. The highest
D-amino acid ratios (free D-amino acid concentration to the total corresponding free D- and L-amino acid concentration) were
29%, 26%, 18%, 4% and 1% for proline, alanine, serine, aspartate and glutamate, respectively. The gastric juice levels of
L-alanine, L-serine, L-proline, L-glutamate and D-alanine in the samples obtained from subjects bearing early gastric carcinoma
and H. pylori were significantly higher than in the samples from the other three groups. Except for D-alanine, there was no correlation

Trigonopsis variabilis induced for D-amino acid oxidase and catalase was immobilized by entrapment in Polyacrylamide beads obtained by radiation
polymerisation. Permeabilization of the cells was found to be essential for optimal activity of the enzymes in free cells.
However, the process of entrapment itself was found to eliminate the permeability barrier of cells immobilized in Polyacrylamide.
The two enzymes exhibited a differential response on Polyacrylamide entrapment. Thus, D-amino acid oxidase activity was stabilized
to heat inactivation whereas catalase in the same cells showed a destabilization on entrapment in Polyacrylamide. The coimmobilized
enzyme preparation showed an operational half life of 7–9 days after which the D-amino acid oxidase activity remained stable
at a value 35–40% of that of the initial activity for a study period of 3 weeks. Coimmobilization of MnO2 was not effective in enhancing the operational life of the enzyme preparation. 相似文献

Summary Free L- and D-amino acids (L-AA, D-AA) were isolated from an Appenzeller cheese, from raw milk, and from an ethanolic extract as well as a total hydrolysate of cow's rumen microorganisms, and their relative amounts were determined by reversed-phase high-performance liquid chromatography after derivatization witho-phthaldialdehyde together withN-isobutyryl-L-(or D)-cysteine. D-Ala, D-Asp and D-Glu were found, among other D-AA in all cases and a microbial origin of free D-AA found in cheese and milk was rationalized. From the results, and taking other findings of the occurrence of D-AA in food and beverages into account, the highest intake of D-AA is to be expected from the consumption of ripened cheeses. Toxic and nontoxic peptides were isolated from the cyanobacterium Microcystis aeruginosa PCC 7806 by a procedure including extraction of cells with water-saturated 1-butanol, chromatography of the extract on silica
gel plates and high performance liquid chromatography (HPLC) on Partisil-5. The toxin was shown to be only a minor constituent,
being negatively charged and thus separable by electrophoresis, within the HPLC-purified fraction. It contained erythro-β-methyl-D-Asp, D-Glu, D-Ala, L-Leu, and L-Arg known to be part of the Microcystis peptide-toxin with Mr 994. The major part of the HPLC-purified fraction was assigned, however, to a nontoxic peptide with a Mr of 956. Partial hydrolysis studies of the nontoxic peptide(s) revealed amino acid sequences composed of D-Glu, N-methyl-Phe, and 3,4-dehydro-Pro, aside from the common L-amino acids. Cyclic linkage in the nontoxic peptide(s) appears likely. 相似文献

The gene encoding D-amino acid oxidase (DAAO) from Trigonopsis variabilis CBS 4095 has been cloned and expressed in Escherichia coli BL21 (DE3). Unfortunately, it was observed that the host cell was negatively affected by the expressed DAAO, resulting in
a remarkable decrease in cell growth. To overcome this problem, we investigated several factors that affect cell growth rate
and DAAO production such as addition time of inducer and dissolved oxygen (DO) concentration. The addition time of lactose,
which was used as an inducer, and DO concentration appeared to be critical for the cell growth of E. coli BL21 (DE3)/pET-DAAO. A two-stage DO control strategy was developed, in which the DO concentration was controlled above 50%
until specific stage of bacterial growth (OD600 30–40) and then downshifted to 30% by changing the agitation speed and aeration rate, and they remained at these rates until
the end of fermentation. With this strategy, the maximum DAAO activity and cell growth reached 18.5 U/mL and OD600 81, respectively. By reproducing these optimized conditions in a 12-m3 fermentor, we were able to produce DAAO at a productivity of 19 U/mL with a cell growth of OD600 80. 相似文献

Elimination of plasmids from Thermus flavus, T. thermophilus and three wild Thermus strains caused alterations in growth temperature range, pigmentation and membrane fatty acids without affecting viability. Following plasmid elimination all Thermus strains lost their ability to grow above 70°C. In addition, the minimum growth temperature was lowered by 5–10°C. Fatty acids were reduced by an average of approximately 35%. In addition, the contribution of iso- and anteisobranched fatty acids were altered in four of the five strains. The iso C15:0/iso C17:0 ratio approached 1.0 in all strains, whereas the anteiso C15:0/anteiso C17:0 was reduced to 0.2. The iso C16:0/normal-C16:0 ratio increased in all strains due to an increase in iso C16:0 in four strains and a reduction in normal-C16:0 relative to iso C16:0 in one strain. However, it was evident that the plasmid-free strains were able to compensate for these alterations in membrane fluidity to a certain extent by reducing the average chain length of isobranched acids. Altered fatty acid metabolism at the level of precursors may have influenced membrane composition and consequently growth temperature range. 相似文献

d-Amino acid N-acetyltransferase is a unique enzyme of Saccharomyces cerevisiae acting specifically on d-amino acids. The enzyme was found to be encoded by HPA3, a putative histone/protein acetyltransferase gene, and we purified its gene product, Hpa3p, from recombinant Escherichia coli cells. Hpa3p shares 49% sequence identity and 81% sequence similarity with a histone acetyltransferase, Hpa2p, of S. cerevisiae. Hpa3p acts on a wide range of d-amino acids but shows extremely low activity toward histone. However, Hpa2p does not act on any of the free amino acids except l-lysine and d-lysine. Kinetic analyses suggest that Hpa3p catalyzes the N-acetylation of d-amino acids through an ordered bi-bi mechanism, in which acetyl-CoA is the first substrate to be bound and CoA is the last product to be liberated. 相似文献

Heterocyst development in ammonia-grown cultures of Anabaena variabilis and Anabaena 7120 was fully induced by the amino-acid analog methionine sulfoximine (MSO) at concentrations of 0.5–1.0 M. Glutamine, glutamate, aspartate, and alanine at 0.5 mM blocked the induction of heterocysts by MSO in A. variabilis. With Anabaena 7120, glutamine and glutamate were fully effective and alanine partially effective in preventing MSO-induced heterocyst formation. In MSO-treated algae, glutamine synthetase activity was reduced to less than 15% of control values within 4–6 h. Inactivation of the enzyme was prevented by all four amino acids tested. 相似文献

Summary Amino acid, polyamine and protein concentrations in seeds and their evolution during seed germination of two dipterocarp species, Hopea odorata and Dipterocarpus alatus, were determined with the help of a multianalytical system. Glutamic acid and glutamine were the major amino compounds present. Hopea seeds also contain high levels of aspartic acid/asparagine, serine, threonine, arginine and alanine, while those of Dipterocarpus contain high levels of alanine, arginine and threonine. These species were quite different in their germination behavior and thus in their protein and amine metabolism rates. In Hopea, polyamines increased during the first 3 days of germination and reached a maximum by the 3rd day, 1 day before maximum germination rate. In Dipterocarpus polyamines reached their maximum at the 6th day while maximum germination rate is observed by the 7th day. This suggests that polyamine compounds could play a role in the early part of the germination process in Hopea and Dipterocarpus seeds. The possibility that control of polyamine biosynthesis could be used for the establishment of biochemical methods to improve seed storage and to control germination of these recalcitrant seeds is discussed. 相似文献

Patterns of morphological variation and reproductive isolation were examined for several North American populations of copepods in the Acanthocyclops vernalis Fischer A., 1853 (Copepoda, Cyclopinae) species complex. The copepods were collected from six sites in Wisconsin, U.S.A. Morphological analysis of 120 adult females revealed that a character used previously to distinguish species in this group was unreliable because of phenotypic plasticity. Most of the morphological variance was due to environment (Laboratory vs. field) and to field site. Relatively little of the variation was due to measurement error or asymmetry. Multivariate ordination analysis produced poorly-defined clusters of individuals, suggesting that different biological species are difficult or impossible to distinguish using a set of easily-measurable morphological characters. In our study, morphological similarity was independent of geographic distance among sites, between 0.05 and 300 km. Isofemale lines within sites showed little or no reproductive isolation, but nearly complete isolation among sites. Reproductive isolation was also independent of morphology. These results suggest that the Acanthocyclops population at each site could be considered a distinct cryptic biological species. These copepods expressed morphological stasis – persistence of morphological uniformity despite reproductive isolation. Because of the effect of site and environment on morphology, we recommend using much larger collections (many sites), common garden experiments, and a multi-disciplinary approach (morphological, reproductive, chromosomal, and molecular) as the basis for future taxonomic research on putative copepod species. 相似文献

The composition of fatty acids in 12 strains of the genera Thermus,
Meiothermus,

Summary. We studied the role of the D-amino acids (D-aa) D-serine, D-alanine, D-methionine, D-aspartate, D-tyrosine and D-arginine
on rat liver mitochondria. The stability of D-amino acids, mitochondrial swelling, transmembrane potential and oxygen consumption
were studied under oxidative stress conditions in rat liver mitochondria. In the presence of glutamate-malate all D-aas salts
increased mitochondrial swelling, while in the presence of succinate plus rotenone only D-ala, D-arg and D-ser, induced mitochondrial
swelling. The transmembrane potential (ΔΨ) was decreased in the presence of 1 μM Ca2+. The D-aas inhibited oxygen consumption in state 3. The D-aa studied exerted effects on mitochondria via an increase of free
radicals production.

Alkylsulfonic acids and some S-containing detergents as sulfur sources for growth of Chlorella fusca
Chlorella fusca can utilize the following substances as sole sulfur sources for growth: C1 to C8 n-alkane-1-sulfonates, linear alkylbenzenes sulfonates (LAS), -sulfonated fatty acid esters, polyethylene glycol sulfate and alkylsulfates. Good sulfur sources are alkylsulfonic acids, which are comparable to sulfate. Ethanesulfonic acid was used for comparison of the growth on sulfate and on a sulfonic acid, because best growth was achieved on this C2-sulfonic acid.Growth data of Chlorella on the enviromental important detergents linear alkylbenzene sulfonic acids, -sulfonated fatty acid methylester, Texapon and Sulfopon are presented. So far only microorganisms have been discussed as a source for degradation of sulfonic acids and detergents. It is suggested that green algae could be of similar importance for the biodegradation of these compounds.Abbreviations LAS

Detailed C-banded karyotypes of eight diploidArtemisia species from three different sections are reported together with preliminary observations on three additional related diploid species. In the majority, the overall amount of banding is relatively low. Bands are mostly confined to distal chromosome regions; intercalary banding is virtually absent and centromeric heterochromatin is also scarce. With the exception ofA. judaica there is in general great uniformity in karyotype structure but considerable interspecific variation in total karyotype length (and hence DNA content) ranging from 44 µm inA. capillaris (2n = 18) to 99 µm inA. atrata (2n = 18).A. judaica (2n = 16; total karyotype length 97 µm) was distinguished by its karyomorphology, with one large non-banded metacentric chromosome pair and 7 pairs of smaller terminally banded meta- or submetacentric chromosomes. 相似文献
