The sulfation of glycosaminoglycans by ascorbic acid 2-[3 5S]sulfate was studied in costal cartilage and chrndrocytes in vitro. Negligable (if any) sulfation of glycosaminoglycans was detected with immediately isolated ascorbic acid 2-[3 5S]sulfate. However, formation of [3 5S]glycosaminoglycans was readily detected with ascorbic acid 2-[3 5S]sulfate which had been stored at −20°C for several days. The [3 5S]glycosaminoglycans did not result from the direct transfer of 3 5S from ascorbic acid 2-sulfate but rather from a decomposition product of ascorbic acid 2-[3 5S]sulfate.Evidence is presented to show that the sulfation pathway with the decomposition product involves exchange with inorganic sulfate, and strongly suggests that sulfation proceeds via 3′-phosphoadenosine 5′-phosphosulfate. The decomposition product appears similar to inorganic sulfate in several test systems. In view of these observations, it is suggested that previous conclusions implicating ascorbic acid 2-sulfate as a biological sulfate donor, based on the use of ascorbic acid 2-[3 5S]sulfate be re-evaluated.  相似文献

The range of delta-selectivity of linear and cyclic analogues of enkephalin in rat brain was found to be: [D-Pen2, L-Pen5] enkephalin (DPLPE) greater than [D-Pen2, D-Pen5] enkephalin (DPDPE) greater than [D-Thr2, Leu5] enkephalyl-Thr6 (DTLET) greater than [D-Ser2, Leu5] enkephalyl-Thr6 (DSLET). Saturation experiments performed with [3H]DPDPE and [3H]DTLET in NG108-15 cells and rat brain showed similar binding capacities for both the ligands, but the delta-affinity of [3H]DTLET (KD approximately 1.2 nM) was much better than that of [3H]DPDPE (KD approximately 7.2 nM). The rather low delta-affinity of DPDPE induced high experimental errors cancelling the benefit of its better delta-selectivity. Binding experiments in rat or guinea-pig brains showed, in both cases, the better delta-selectivity of [3H]DTLET compared to [3H]DSLET. The former peptide remains at this time the most appropriate radioactive probe for binding studies of delta-receptor.  相似文献

Short (10 min) and long (24 hr) treatments with 10 μg/ml dextran sulfate produced a marked increase in the incorporation of [35S] sulfate into cell-associated and extracellular glycosaminoglycans of SV40-transformed 3T3 cells. Similar treatment of 3T3 cells did not affect glycosaminoglycan synthesis. DEAE-dextran treatment for 24 hr produced a large increase in the cell-associated glycosaminoglycans and a proportional decrease in the extracellular sulfated glycosaminoglycans. The types of glycosaminoglycans synthesized were not affected significantly by the treatments.  相似文献

We have designed the nitrophenylfurylacryl-substituted benzophenone 4f as a non-thiol farnesyltransferase inhibitor utilizing a novel aryl binding site of farnesyltransferase. Variation of the 2-acylamino substituent at the benzophenone core structure of our initial lead 4f yielded several non-thiol farnesyltransferase inhibitors with improved activity. These compounds display activity in the low nanomolar range.  相似文献

The transport of [3H]2-deoxyglucose by brain slices was studied. Cerebral cortex slices were incubated in vitro in the presence of [3H]2-deoxyglucose, orl-[3H] glucose as a marker for diffusion. Transport was defined as the difference between [3H]2DG uptake andl-[3H]glucose uptake. Half-maximal velocity was seen at 2.0 mM 2DG and [3H]2DG transport was not inhibited by 20-fold higher concentrations ofl-glucose. Net [3H]2DG transport was unchanged in media deficient in Na+, K+, Mg2+, Ca2+ or Cl. Uptake was significantly inhibited by 1.0 mM 2,4-DNP and a suggestion of inhibition by azide was seen. These data are consistent with a hypothesis that hexose transport in the brain depends to some extent upon mitochondrial energy.  相似文献

Cell-free extracts of Acremonium chrysogenum and Streptomyces clavuligerus oxidize the 3-methyl group of desacetoxycephalosporin C to a 3-hydroxymethyl group. The enzyme responsible for this reaction in these organisms was purified 20- and 30-fold respectively by chromatography on DEAE-cellulose. The enzymes, which were assayed with [3-methyl-3H]desacetoxycephalosporin C as substrate, have the properties expected of 2-oxoglutarate-linked dioxygenases. They require 2-oxoglutarate, Fe2+ cations and a mixture of reducing agents (dithiothreitol and ascorbate) for full activity. The enzyme from A. chrysogenum, but not that S. clavuligerus, is activated about 10-fold when it is preincubated with a reaction mixture from which either desacetoxycephalosporin C or 2-oxoglutarate is omitted. Fe2+ cations seem to play a key role in this activation. Both enzymes seem highly specific for cephalosporins with the natural 7beta-(5-D-aminoadipamido) side chain and are likely to be responsible for the oxidation of the 3-methylcephem nucleus in vivo.  相似文献

Deuterated oleates have been synthesized by semihydrogenation of acetylenic intermediates. [11-2H2]Oleate was prepared by two-carbon chain extension of the C16 alcohol obtained from [1-2H2]octyl bromide and 7-octyn-1-ol. [8-2H2] and [7-2H2]oleates were both prepared from dimethyl suberate, tetradeutero intermediate C16 alcohols were synthesized from [1,8-2H4] and [2,7-2H4]octane diols by monobromination, conversion to deuterated 9-decyn-1-ols and reaction with octyl bromide. Oxidation gave [8-2H2]-9-octadecynoate and [2,7-2H2]-9-octadecynoate, after semihydrogenation of the latter, deuterons at C-2 were removed by exchange with aqueous alkali. [6-2H2] and [5-2H2]oleates were obtained from methyl 5-tetradecynoate, semihydrogenation, deuterium exchange at C-2 and two malonate extensions gave [6-2H2]oleate; reduction with lithium aluminum deuteride, two malonate extensions and semihydrogenation gave the [5-2H2] ester. [4-2H2] and [3-2H2]oleates were both obtained from methyl 7-cis-hexadecenoate, exchange of the α protons and chain extension gave the [4-2H2] ester and reduction with lithium aluminum deuteride and chain extension gave the [3-2H2] ester.  相似文献

We have developed the [5-(4-nitrophenyl)-2-furyl]acrylic acid substituted benzophenone 4g as a novel lead for anti-malarial agents. Here, we demonstrated that the acyl residue at the 2-amino group of the benzophenone core structure has to be a phenylacetic acid substructure substituted in its para-position with methyl or other substituents of similar size. The trifluoromethyl substituted derivative displayed an IC(50) of 47 nM against the multi-drug resistant Plasmodium falciparum strain Dd2.  相似文献

5'-Phenyl-3'H-spiro[indoline-3,2'-[1,3,4]thiadiazol]-2-one inhibitors of ADAMTS-5 (Aggrecanase-2) have been prepared via commercially available starting materials. Selected compounds 23, 33-35 show sub-micromolar ADAMTS-5 potency and strong SAR trends with selectivity over the related metalloproteases ADAMTS-4 (Aggrecanase-1), MMP12, and MMP13. This series of compounds represents progress toward a selective ADAMTS-5 inhibitor as a disease modifying osteoarthritis drug.  相似文献

A number of N-substituted piperazinylquinolone derivatives were synthesized and evaluated for antibacterial activity against Gram-positive and Gram-negative bacteria. Preliminary results indicated that most compounds tested in this study demonstrated comparable or better activity against Staphylococcus aureus and Staphylococcus epidermidis than their parent piperazinylquinolones as reference drugs. Among these derivatives, ciprofloxacin derivative 5a, containing N-[2-[5-(methylthio)thiophen-2-yl]-2-oxoethyl] residue, showed significant improvement of potency against staphylococci, maintaining Gram-negative coverage.  相似文献

After incubation of 3beta-hydroxy-5-[17,21,21,21-2H]-pregnen-20-one with the microsomal fraction of boar testis, the metabolites were analyzed by gas chromatography and gas chromatography-mass spectrometry. The following metabolites were identified: 3beta,17alpha-dihydroxy-5-[21,21,21-3H]pregnen-20-one, 3beta-hydroxy-5-androsten-17-one, 5-androstene-3beta,17beta-diol, and 5-[17beta-2H]androstene-3beta,17alpha-diol. The presence of a 2H atom at the 17beta position of 5-androstene-3beta,17alpha-diol was confirmed by oxidizing the steroid with 3beta-hydroxy-steroid dehydrogenase of Pseudomonas testosteroni to obtain 17alpha-hydroxy-4-[2H]androsten-3-one and then by oxidizing the latter steroid with chromic acid to obtain nonlabeled 4-androstene-3,17-dione. Among these metabolites, the first three can be interpreted to be synthesized by a well documented pathway, including 17alpha-hydroxylation followed by side chain cleavage as follows: 3beta-hydroxy-5-[17,21,21,21-2H]pregnen-20-one leads to 3beta,17alpha-dihydroxy-2-[21,21,212H]-pregnen-20-one leads to 3beta-hydroxy-5-androsten-17-one leads to 5-androstene-3beta,17beta-diol. On the other hand, 5-androstene-3beta,17alpha-diol, which contained a 2H atom at the 17beta position, is not likely to be synthesized via above mentioned pathway in which nonlabeled 3beta-hydroxy-5-androsten-17-one is formed as the first C19-steroid. It seems that an alternate side chain cleavage mechanism leading from pregnenolone to 17alpha-hydroxy-C19-steroid exists in boar testis.  相似文献

We have shown previously that a highly sulfated sequence, GalNAc(4,6-SO(4))-GlcA(2SO(4))-GalNAc(6SO(4)), is present at the nonreducing terminal of chondroitin sulfate (CS), and this structure was synthesized from a unique sequence, GalNAc(4SO(4))-GlcA(2SO(4))-GalNAc(6SO(4)), by sulfation with N-acetylgalactosamine 4-sulfate 6-O-sulfotransferase. Uronosyl 2-O-sulfotrasferase (2OST), which transfers sulfate from 3'-phosphoadenosine 5'-phosphosulfate (PAPS) to position 2 of the GlcA residue of CS, is expected to be involved in synthesis of these structures; however, the specificity of 2OST concerning recognition of the sulfation pattern of the acceptor has largely remained unclear. In the present study, we examined the specificity of 2OST in terms of recognition of the sulfation pattern around the targeting GlcA residue. The recombinant 2OST could sulfate CS-A, CS-C, and desulfated dermatan sulfate. When [(35)S]glycosaminoglycans formed from CS-A after the reaction with the recombinant 2OST and [(35)S]PAPS were subjected to limited digestion with chondroitinase ACII, a radioactive tetrasaccharide (Tetra A) was obtained as a sole intermediate product. The sequence of Tetra A was found to be DeltaHexA-GalNAc(4SO(4))-GlcA(2SO(4))-GalNAc(6SO(4)) by enzymatic and chemical reactions. These observations indicate that 2OST transfers sulfate preferentially to the GlcA residue located in a unique sequence, -GalNAc(4SO(4))-GlcA-GalNAc(6SO(4))-. When oligosaccharides with different sulfation patterns were used as the acceptor, GalNAc(4SO(4))-GlcA-GalNAc(6SO(4)) and GlcA-GalNAc(4SO(4))-GlcA-GalNAc(6SO(4)) were the best acceptors for 2OST among trisaccharides and tetrasaccharides, respectively. These results suggest that 2OST may be involved in the synthesis of the highly sulfated structure found in CS-A.  相似文献

[2-14C]Tetrahydropteroylglutamic acid and 5-[G-3H]methyltetrahydropteroylglutamic acid were given intraperitoneally to fruit bats. Folate polyglutamates were formed in the liver from both substrates in different amounts and at different rates. The methylfolate pool appeared to remain separate from the tetrahydrofolate pool. More polyglutamate was formed from tetrahydropteroylglutamic acid than from 5-methyltetrahydropteroylglutamic acid. There was a fall in the folate content of the liver in the vitamin B-12-deficient bat and a more rapid incorporation of folates into polyglutamates but thereafter a more rapid loss of the labelled folate from liver.  相似文献

Grape leaves (Vitis labrusca L.) that are removed from the positionopposite the flower cluster either 28 or 14 days before anthesiscleave L-ascorbic acid (AA) at the C4-C5 bond into a C4 and,presumably, a C2 fragment. Leaves taken from this position 14days after anthesis fail to cleave AA. The C4 fragment is utilizedfor L(+)-tartaric acid (TA) biosynthesis while the C2 fragmentis recycled into hexose and products of the hexose metabolism.When [6-14C]AA is the source of the label, the sucrose-derivedglucose from labeled leaves has a distribution of 14C in thecarbon skeleton as follows: Cl, 35%; C2, 14%; C3, 4%; C(4+5),13% and C6, 34%.
