To resist terrestrial UV radiation, plants employ DNA-damage-repair/toleration (DRT) activities, as well as shielding mechanisms. Little is known about the structure and regulation of plant DRT genes. We isolated DRT cDNAs from Arabidopsis thaliana, by selecting for complementation of Escherichia coli mutants lacking all bacterial defenses against UV-light damage to DNA. These mutants are phenotypically deficient in recombinational and mutagenic toleration (RecA), excision repair (Uvr) and photoreactivation toreactivation (Phr). Among 840 survivors of heavily UV-irradiated (10–7 survival) mutants harboring plasmids derived from an Arabidopsis cDNA library in the vector YES, we identified four unique plant cDNAs, designated DRT100, DRT101, DRT102, and DRT103. Drt101 and Drt102 activity were specific for UV-light damage, and complemented both UvrB and UvrC phenotypes in the dark. Apparent Uvr correction efficiencies were 1 to 40% for Drt101, and 0.2 to 15% for Drt102, depending on the UV fluence. Drt101 and Drt102 showed no extensive amino-acid homology with any known DNA-repair proteins. Drt100 appeared to correct RecA, rather than Uvr, phenotypes. Although the light dependence of Drt103 activity was consistent with its identification as a photoreactivating enzyme, its predicted amino-acid sequence did not resemble known photolyase sequences. The N-terminal coding sequence of Drt101 suggests that it is targeted to chloroplasts, as reported for Drt100. These cDNAs afforded only modest increases in survival during the original selection procedure. The fact that they were readily isolated nevertheless suggests that selections may be made powerful enough to overcome barriers to expression and function in bacteria, at least for cDNAs of reasonable abundance.  相似文献

Photosystem II (PS II) chlorophyll (Chl) a fluorescence lifetimes were measured in thylakoids and leaves of barley wild-type and chlorina f104 and f2 mutants to determine the effects of the PS II Chl a+b antenna size on the deexcitation of absorbed light energy. These barley chlorina mutants have drastically reduced levels of PS II light-harvesting Chls and pigment-proteins when compared to wild-type plants. However, the mutant and wild-type PS II Chl a fluorescence lifetimes and intensity parameters were remarkably similar and thus independent of the PS II light-harvesting antenna size for both maximal (at minimum Chl fluorescence level, Fo) and minimal rates of PS II photochemistry (at maximum Chl fluorescence level, Fm). Further, the fluorescence lifetimes and intensity parameters, as affected by the trans-thylakoid membrane pH gradient (pH) and the carotenoid pigments of the xanthophyll cycle, were also similar and independent of the antenna size differences. In the presence of a pH, the xanthophyll cycle-dependent processes increased the fractional intensity of a Chl a fluorescence lifetime distribution centered around 0.4–0.5 ns, at the expense of a 1.6 ns lifetime distribution (see Gilmore et al. (1995) Proc Natl Acad Sci USA 92: 2273–2277). When the zeaxanthin and antheraxanthin concentrations were measured relative to the number of PS II reaction center units, the ratios of fluorescence quenching to [xanthophyll] were similar between the wild-type and chlorina f104. However, the chlorina f104, compared to the wild-type, required around 2.5 times higher concentrations of these xanthophylls relative to Chl a+b to obtain the same levels of xanthophyll cycle-dependent fluorescence quenching. We thus suggest that, at a constant pH, the fraction of the short lifetime distribution is determined by the concentration and thus binding frequency of the xanthophylls in the PS II inner antenna. The pH also affected both the widths and centers of the lifetime distributions independent of the xanthophyll cycle. We suggest that the combined effects of the xanthophyll cycle and pH cause major conformational changes in the pigment-protein complexes of the PS II inner or core antennae that switch a normal PS II unit to an increased rate constant of heat dissipation. Laurencia brongniartii is usually found at depths below 4 m, but can be found in shallow subtidal areas in crevices and on the walls of a coral reef in Amami Oshima Island, Kagoshima Prefecture, Japan, where irradiances were significantly lower than those at similar depths in open water. In preparation for the possible cultivation of this species for its antibiotic compounds, the effects of temperature and irradiance on photosynthesis and growth were measured. Photosynthesis and growth rates of L. brongniartii explants were highest at 26 and 28 °C, which closely corresponded to temperatures found during August to late December when it was most abundant. The estimated maximum photosynthesis rate (P max) was 4.41 mol photon m–2 s–1 at 26 °C and 4.07 mol photon m–2 s–1 at 28 °C. Saturating irradiance occurred at 95 mol photon m–2 s–1 at 26 °C and 65 mol photon m–2 s–1 at 28 °C. In contrast, growth experiments at 41.7 mol photon m–2 s–1 caused bleaching of explants and the maximum growth rate observed during the study was 3.02 ± 0.75% day–1 at 28 °C and 25 mol photon m–2 s–1. The difference in the saturating irradiance for photosynthesis and the irradiance that caused bleaching in growth experiments suggests that long-term exposure to high irradiance was detrimental and should be addressed before the initiation of large scale cultivation.  相似文献

Five cDNAs for genes differentially expressed during fruit development of kiwifruit (Actinidia deliciosa var.deliciosa cv. Hayward) were isolated from a library made from young fruit, 8–10 days after anthesis. One gene (pKIWI503) has low levels of expression in young fruit but is induced late in fruit development and during fruit ripening, and has some homology to plant metallothionein-like proteins. The other four genes are highly expressed in young fruit with reduced expression in the later stages of fruit development. pKIWI504 has strong homology to plant metallothionein-like proteins and pKIWI505 exhibits homology to the -subunit of the mitochondrial ATP synthase gene. The two other genes (pKIWI501 and 502) encode proteins with no significant homology to other known sequences.  相似文献

Photosynthetic adaptation of the unicellular green alga Scenedemus obliquus to different light conditions was investigated with respect to chlorophyll synthesis. Cultures were grown under white light (20 W · m–2) from fluorescent lamps and were then transferred and subjected to the actual adaptation regime which consisted of a 24-h irradiation by different fluence rates and wavelengths. Fluence rate-response curves for chlorophyll synthesis were measured between 4 · 10–2 and 1 · 102 W · m–2. In white light from incandescent lamps, in blue and red light the fluence rate-response curves for chlorophyll (Chl) a and also for Chl b were bell-shaped. In red light the threshold was about the same as under blue light. The maximal amounts of Chl a and b were about twofold increased under blue light relative to the values obtained with red light. Action spectra for the stimulation of chlorophyll synthesis (Chl a + Chl b) as well as those for the separate chlorophylls showed two maxima near 450 and 500 nm. However, the action spectrum for Chl b synthesis demonstrated a considerably higher value in the 450-nm peak. Experiments with the photosynthesis inhibitor 3-(3,4-dichlorphenyl)-1,1-dimethylurea (DCMU) indicated that photosynthetic energy supply supported the photostimulation of chlorophyll synthesis. The action spectra indicate the cooperation of two photoreceptors. The 460-nm peak is attributed to the typical blue-light receptor, being more active in Chl b formation. Adjustment in leaf area : mass ratio called leaf area ratio (LAR) is one of the strategies to optimize photon harvesting. LAR was recorded for 10 genotypes of Hevea brasiliensis under high irradiance and low temperature and the genotypes were categorized into two groups, i.e. high LAR and low LAR types. Simultaneously, the growth during summer as well as winter periods, photosynthetic characteristics, and in-vitro oxidative damage were studied. Low LAR (19.86±0.52 m2 kg–1) types, recorded an average of 18.0 % chlorophyll (Chl) degradation under high irradiance and 7.1 % Chl degradation under low temperature. These genotypes maintained significantly higher net photosynthetic rate (P N) of 10.4 mol(CO2) m–2 s–1 during winter season. On the contrary, the high LAR (24.33±0.27 m2 kg–1) types recorded significantly lower P N of 4 mol(CO2) m–2 s–1 and greater Chl degradation of 37.7 and 13.9 % under high irradiance and low temperature stress, respectively. Thus LAR may be one of the physiological traits, which are possibly involved in plant acclimation process under both stresses studied.  相似文献

Thylakoids isolated from cells of the red alga Porphyridium cruentum exhibit an increased PS I activity on a chlorophyll basis with increasing growth irradiance, even though the stoichiometry of Photosystems I and II in such cells shows little change (Cunningham et al. (1989) Plant Physiol 91: 1179–1187). PS I activity was 26% greater in thylakoids of cells acclimated at 280 mol photons · m–2 · s–1 (VHL) than in cells acclimated at 10 mol photons · m–2 · s–1 (LL), indicating a change in the light absorbance capacity of PS I. Upon isolating PS I holocomplexes from VHL cells it was found that they contained 132±9 Chl/P700 while those obtained from LL cells had 165±4 Chl/P700. Examination of the polypeptide composition of PS I holocomplexes on SDS-PAGE showed a notable decrease of three polypeptides (19.5, 21.0 and 22 kDa) in VHL-complexes relative to LL-complexes. These polypeptides belong to a novel LHC I complex, recently discovered in red algae (Wolfe et al. (1994a) Nature 367: 566–568), that lacks Chl b and includes at least six different polypeptides. We suggest that the decrease in PS I Chl antenna size observed with increasing irradiance is attributable to changes occurring in the LHC I-antenna complex. Evidence for a Chl-binding antenna complex associated with PS II core complexes is lacking at this point. LHC II-type polypeptides were not observed in functionally active PS II preparations (Wolfe et al. The Porphyridium cruentum light harvesting complex (LHC) binds Chl a, zeaxanthin and -carotene and comprises at least 6 polypeptides of a multigene family. We describe the first in vitro reconstitution of a red algal light-harvesting protein (LHCaR1) with Chl a/carotenoid extracts from P. cruentum. The reconstituted pigment complex (rLHCaR1) is spectrally similar to the native LHC I, with an absorption maximum at 670 nm, a 77 K fluorescence emission peak at 677 nm (ex. 440 nm), and similar circular dichroism spectra. Molar ratios of 4.0 zeaxanthin, 0.3 -carotene and 8.2 Chl a per polypeptide for rLHCaR1 are similar to those of the native LHC I complex (3.1 zeaxanthin, 0.5 -carotene, 8.5 Chl a). The binding of 8 Chl a molecules per apoprotein is consistent with 8 putative Chl-binding sites in the predicted transmembrane helices of LHCaR1. Two of the putative Chl a binding sites (helix 2) in LHCaR1 were assigned to Chl b in Chl a/b-binding (CAB) LHC II [Kühlbrandt et al. (1994) Nature 367: 614–21]. This suggests either that discrimination for binding of Chl a or Chl b is not very specific at these sites or that specificity of binding sites evolved separately in CAB proteins. LHCaR1 can be reconstituted with varying ratios of carotenoids, consistent with our previous observation that the carotenoid to Chl ratio is substantially higher in P. cruentum grown under high irradiance. Also notable is that zeaxanthin does not act as an accessory light-harvesting pigment, even though it is highly likely that it occupies the position assigned to lutein in the CAB LHCs.This revised version was published online in October 2005 with corrections to the Cover Date.  相似文献

Certain forms of oligocellulose and certainbacterially excreted peptides were identified asendogenous and exogenous elicitors, respectively, ofa tip bleaching response in Gracilaria conferta(Schousboe ex Montagne) J. et G. Feldmann. Thehalf-maximal tip bleaching response was observed when31.1 M cellobiose or 11.6 M cellotetraosewere present in the growth medium. In contrast, noresponse was detected after exposure to glucose,cellotriose, cellopentaose or maltooligosaccharides.The response was thus strongly dependent on themolecular size of the oligocellulose and onlysaccharides that consisted of an even number ofglucose residues were elicitor-active. Three bacterialspecies that had earlier been identified as potentialinducers of the tip bleaching symptom excretedelicitor-active compounds into the growth medium.These compounds were protease-sensitive and thuspeptides or proteins. The tip bleaching-inducingcompound that was excreted by one Cytophaga-likeorganism was partially purified. Physical, chemical and biological observations made in late July and August 1997 across the Azores Front (37° N, 32°W to 32° N, 29°W) are presented. The objectives of the study were: (1) to analyse horizontal and vertical profiles of temperature, salinity, density, nutrients and chlorophyll-a (Chl a) of the top 350 m; (2) to identify the main differences in the deep Chl a Maximum (DCM) and hydrographic structure between the water masses that pass north and south of the Azores Front; and (3) to estimate phytoplankton primary production in these water masses. Horizontal and vertical profiles of salinity, temperature, density, nutrients and phytoplankton pigments in the top 350 m were analysed. The Front separates two distinct water types: the 18°C Mode Water (18 MW) of sub-tropical origin, and the 15°C Mode Water (15 MW) of sub-polar origin. Differences in the DCM and hydrographic structure between 18 MW and 15 MW were observed in the contour plots of each section. The average Chl a concentration between 5 and 200 m depth decreased significantly from 15 MW to 18 MW. The same pattern was observed for the Chl a concentration at the DCM depth. A vertical one-dimensional model was used to estimate the phytoplankton primary production in the 15 MW and 18 MW and led to an estimated water column average gross primary productivity (GPP) between 1.08 and 2.71 mg C m–3 d–1 for the 15 MW and about half of these values for the 18 MW. These results indicate that the typical south–north positive slope on DCM depth parallels a latitudinal increase on GPP, suggesting that the location of the Azores Front may have a significant regional impact on GPP.  相似文献

We have isolated Chl a-Chl c-carotenoid binding proteins from the dinoflagellates Prorocentrum minimum and Heterocapsa pygmaea grown under high (500 mol m–2 s–1, HL) and low (35 mol m–2 s–1, LL) light conditions. We compared various isolation procedures of membrane bound light harvesting complexes (LHCs) and assayed the functionality of the solubilized proteins by determining the energy transfer efficiency from the accessory pigments to Chl a by means of fluorescence excitation spectra. The identity of the newly isolated protein-complexes were confirmed by immunological cross-reactions with antibodies raised against the previously described membrane bound Chl a-c proteins (Boczar et al. (1980) FEBS Lett 120: 243–247). Spectroscopic analysis demonstrated the relatedness of these proteins with the recently described Chl-a-c 2-peridinin (ACP) binding protein (Hiller et al. (1993) Photochem Photobiol 57: 125–131; Iglesias Prieto et al. (1993) Phil Trans R Soc London B 338: 381–392). The water-soluble peridinin-Chl a binding-protein (PCP) was not detectable in P. minimum. Two functional forms of ACP with different pigmentation were isolated. Temperature dependent changes in absorbance and fluorescence of chlorophyll a (Chl a) were analyzed in membrane fragments and in a Chl-protein complex reconstituted with lipids isolated from the cyanobacterium Anacystis nidulans. Absorbance versus temperature curves measured at 656 nm showed an inflection point at 23–24°C and at 14–16°C in the membrane fragments prepared from A. nidulans cells, grown at 39° and 25°C, respectively. Temperature-induced absorbance changes measured at 680 and 696 nm did not show clear break points. The presence of lipids was essential in order to see a clear maximum in the fluorescence versus temperature curve of Chl a in a Chl-protein complex. Gelidium sesquipedale is the most important raw material used for extraction of agar in Spain. Based on chemostats, a system of culture for macroalgae with a continuous flow of culture medium has been developed. A stressed morphotype from the South of Spain was cultured, and the effects of different rates of NO 3 flow on growth and internal constituents were investigated in the laboratory. Cultivation was successful after optimizing factors affecting growth, such as irradiance level, renewal rate and water movement. Mass production was dependent on N supply. With a flow of 35 mol NO3 g–1 DW d–1, optimal values of growth (2.1% d–1) and biomass yield were obtained. In these conditions, biomass yield resembled the values observed in natural populations (about 500 g DW m–2 y–1). When the flow of N was reduced to 15 mol NO 3 g–1 DW d–1, growth rate and biomass yield were reduced three-fold, and were null when N was supplied as 7 mol NO 3 g–1 DW d–1. C:N ratio was an index of the physiological status of the tissue, remaining low when N was sufficient and raised to critical values when N supply was limited. Phycobiliproteins, kept at a constant irradiance level, were affected by N supply, acting as an internal nitrogen reserve, unlike chlorophylla. An effective phycobiliprotein synthesis took place when the flow of N was sufficient. Agar yield, on dry weight basis, was similar as a function of N flow, whereas agar yield of the culture was higher when N was sufficient as a result of growth not being limited by N.This system of culture, commonly used in microalgal studies, may have an important use in macroalgae as a system to obtain biomass of high quality as well as a good tool for physiological studies in conditions of continuous and controlled flow of nutrients.  相似文献

Toxicological responses of the filamentous N2-fixing cyanobacteriumNostoc calcicola Bréb. towards Hg2+ were studied to enumerate the decisive lethal events. In low-dose, long-term experiments (0.05–0.25 m Hg2+, 10 days), photoautotrophic growth was severely inhibited with concurrent loss of photosynthetic pigments (phycocyanin>chlorophyll >carotenoids) and nucleic acids. The termination of growth after a day 4 exposure to 0.25 m Hg2+ has been attributed to the complete inhibition ofin vivo photosynthetic activity in the cyanobacterium (O2 evolution>14CO2 incorporation). The elevated Hg2+ concentrations irreversibly damaged the cell membrance as observed under light microscopy, and as indicated by the leakage of intracellular electrolytes and phycocyanin. In high-dose, short-term experiments (0.5–20.0 m Hg2+, up to 6 h), thein vivo activities of selected enzymes (glutamine synthetase > nitrate reductase > nitrogenase) were less inhibited by Hg2+ than the uptake of nutrient ions (NH 4 + >NO 3 >PO 4 3– ).  相似文献

Two methods of induced in vivo chlorophyll (Chl) fluorescence were used to investigate the effects of varying thallus temperature and hydration on the performance of photosynthetic apparatus of a foliar lichen Cetraria islandica: slow Chl fluorescence induction kinetic with the analysis of quenching mechanisms, and rapid irradiance response curves of photosynthesis derived from quantum yield of photochemical reactions of photosystem 2 (2) recorded at increasing irradiances. We compared responses of photosynthetic apparatus in populations of C. islandica growing in lower altitude (LAP: 1 350 m a.s.l.) and in higher altitude (HAP: 2 000 m a.s.l.). At each altitude, the samples were collected both in fully irradiated sites (HI) and in shade (LI). Temperature optimum of photosynthetic processes was the same for LAP and HAP thalli of LI populations (18 °C), while it was significantly lower for HI HAP (14 °C). Gradual dehydration of fully hydrated thalli led to initial increase (up to 20 % of water saturation deficit, WSD) in FV/FM and 2, no change at 20–50 % WSD, and a dramatic decrease of the parameters within 50–80 % of WSD. LI HAP of C. islandica was the best adapted population to low temperature having higher rates of photochemical processes of photosynthesis than HI HAP within temperature range of –5 to +5 °C. The differences between populations were apparent also in Chl content and thallus morphology.  相似文献

A eubacterium producing a blue pigment was isolated from a drinking water filter, and subsequently identified as Vogesella indigofera. This bacterium was further investigated for its morphological and biochemical characteristics after exposure to hexavalent chromium, Cr6+. The threshold Cr6+ concentration inhibiting the pigment production by V. indigofera was 200–300 g ml–1 in liquid cultures of nutrient broth and 100–150 g ml–1 on nutrient agar plates. The Cr6+ concentration preventing V. indigofera growth was 300–400 g ml–1 in liquid cultures, but greater than 150 g ml–1 on agar plates. Moreover, rugose colonies without the blue pigmentation were observed on agar plates amended with 150 (g Cr6+) ml–1. The biochemical utilization profiles of the colonies without pigmentation did not differ from the original pigment-producing ones, indicating phenotypic plasticity of this bacterium. The difference of phenotypic expression of V. indigofera under various Cr6+ concentrations might have potential application as a pollution bioindicator for heavy metals.  相似文献

Summary The effects of the new antibiotic netilmicin (NT) were studied onEuglena gracilis green cells. It was found that, in the presence of the drug, chlorophyll synthesis was strongly inhibited and plastid structure dramatically altered as revealed by fluorescence and electron microscopic observations. Importantly, NT at low concentrations (10–20 g/ml) and for short periods of time (36–72 hours) induced a marked bleaching effect that was permanent and accompanied by the persistence in the colorless cells of poorly differentiated plastids. Other cell components were not influenced as shown by their appearance and also by the kinetics of growth and the cell viability. On the basis of the results and of the literature on the bleaching agents it is suggested that NT is an untoxic antibiotic which specifically inhibits protein synthesis at prokaryotic level.This work was supported by a grant from Italian Research Council (CNR), Contract N. 82.02016.04.  相似文献

Nutrient limitation of primary production was experimentally assessed using an in situ bioassay technique in the Quebrada Salto, a third-order tropical stream draining the northern foothills of the Cordillera Central in Costa Rica. Bioassays employed artificial substrata enriched with nutrients that slowly diffuse through an agar-sand matrix (Pringle & Bowers, 1984). Multiple comparisons of regression coefficients, describing chlorophyll-a accrual through time for different nutrient treatments, revealed positive micronutrient effect(s). Micronutrient treatment combinations (Fe, B, Mn, Zn, Co, Mo, EDTA), supplemented with and without nitrate and phosphate, exhibited significantly greater chlorophyll-a accrual over all other treatments (P < 0.05), supporting over three times that of the control after 14-d of substratum colonization. Neither of the major nutrients (N or P) produced a significant stimulation, although the N treatment displayed 50% more chlorophyll-a than the control after 14-d. Similarly, Si, EDTA, and Si + N + P treatments did not exhibit chlorophyll-a response curves that were significantly different from the control. During the experiment, mean NH4-N and (NO2 + NO3)-N concentrations in the Salto were 2.0 µM (28.6 µg · l–1) and 7.2 µM (100.2 µg · l –1), respectively. High concentrations of PO4-P ( = 2.0 µM; 60.9 µg · l–1) and TP ( = 3.0 µM; 94.0 µg · l–1) were also found, and consequently low molar N:P ratios = 4.7). Despite the potential for N limitation in the system, both N and P appear to be at growth saturating levels. This may be due to micronutrient limitation and/or light limitation of periphyton growth in densely shaded upstream portions of the stream.  相似文献

From April 1994 to April 1996, we carried out monthly surveys on the physical and chemical variability of ebbing water at two intertidal stations near the emerged tidal flat and surface water at a nearby subtidal station, in a tidal estuary of the Seto Inland Sea, Japan. The fresh water runoff was a major source of new nitrogen (nitrate+nitrite = –2.1 × salinity + 74.3, r 2 = 0.76, p < 0.001, at the inner intertidal station). In contrast, the absence or weak correlation of ammonium, phosphate and silicate with salinity in ebbing water highlighted the importance of in situ biologically-mediated processes, including the excretory activity of intertidal dominant bivalves (Ruditapes philippinarum and Musculista senhousia) and primary producer nutrient uptake. Among suspended particulate matter (SPM), the negative correlation of Chl a with salinity (p< 0.001) suggested that an upper intertidal zone, where high microphytobenthic assemblages occur, may represent a considerable source of microalgal biomass. Whereas, a stronger correlation of POC with pheo-pigments (r 2 0.92) than with Chl a (r 2 0.44), a low fraction of living phyto-Carbon (ca. 5%) and a high pheo-pigment/Chl a ratio (ca. 3) indicated a high portion of refractory algal material and a close water–sediment coupling on the flat, in which abundant macrozoobenthos is likely to play an important role. In spite of the strong variability of this ecosystems, this study also demonstrated that highly significant seasonal patterns can be recognized in our study area. However, patterns varied depending on the different nutrient species, SPM and sites. In ebbing water, ammonium concentrations were significantly higher in the cold period (i.e. November–April: water temperature 10.0 ± 4.4 °C), when primary producer biomass decreases, while Chl a and pheo-pigment content were 4.3 and 4.8 higher in the warm period (May–October: water temperature 23.5 ± 4.2 °C), respectively. At the subtidal site, nutrients were 1.9 (silicate)–2.9 (phosphate) and SPM was 1.8 (POC)–2.1 (pheo-pigments) times higher in the warm period. On an annual basis, mean pheo-pigment and POC content was 12–25 times and 4–8 times higher on the intertidal zone than on the subtidal zone, respectively, while nutrient concentrations were within the same order of magnitude. Such a qualitative difference (i.e. nutrients vs. SPM) between sites suggests that particle deposition and/or removal is likely to occur along the estuary, resulting in a limited SPM content at the surface layer of the subtidal site, while nutrients are more directly transported into the surface layer of the subtidal zone by lower salinity water mass intrusion from the intertidal zone, most importantly during the warm period.  相似文献
