A disengagement of the cyanide-resistant, alternative respiratory pathway in soybean (Glycine max (L.) Merr.) callus tissue was observed prior to the start of deoxyisoflavone production stimulated by addition of the cytokinin benzyladenine. To test whether this loss of alternativepathway activity was part of the response to cytokinin, inhibitors of the alternative pathway were assayed for their ability to elicit cytokinin-like responses. Salicylhydroxamic acid (SHAM) was found to produce a deoxyisoflavone difference spectrum similar to that observed following treatment of the callus tissue with benzyladenine, while propyl gallate (PG) was without effect. Both SHAM and PG were further tested for cytokinin-like activity in other bioassays. In two anti-senescence bioassays using leaf tissue (of Avena sativa L. and Xanthium pensylvanicum Wallr.) and in the Cucumis sativus L. bioassay which measures stimulation of weight gain by excised cotyledons, both SHAM and PG were effective cytokinins at 1 mM and 0.1 mM, respectively. In two other bioassays (betacyanin formation in Amaranthus caudatus L. seedlings and the soybean-callus celldivision assay), SHAM appeared to be toxic. Abstract: Bacteria represent a substantial fraction of the microorganisms that inhabit leaf surfaces. We collected samples of the moss Funaria hygrometrica (L.) in the field and analysed the epiphytes on the gametophyte by the agar impression method and scanning electron/fluorescence microscopy. On the phylloid surface numerous bacteria were detected, notably in the grooves between adjacent lamina cells. Methanol-ammonium salts agar surfaces impressed with isolated phylloids of green gametophytes resulted in the growth of methylotrophic colonies. Two Methylobacterium strains ( M. mesophilicum and M. sp., isolated from the Funaria phylloids) were found to simulate the well-known effect of cytokinin application on bud formation in Funaria protonemata. In addition, Methylobacterium inoculation promoted the growth of protonemal filaments. The significance of this novel Methylobacterium -land plant interaction is discussed.  相似文献

Summary The activities of the cytokinins benzyladenine (BA), 6-(3-methylbut-2-enylamino)purine (IPA) and zeatin in promoting bud formation in protonemata of Funaria hygrometrica (L.) Sibth. were compared to the biological activities of the corresponding ribosides. Cytokinins were found to have considerably higher hormonal activity as free bases than as ribosides. This observation will be discussed in relation to the chemical identity of the celldivision factor (CDF) isolated from crown gall tumors of Vinca rosea L.  相似文献

The induction of shoot buds in the moss Funaria hygrometrica is a classic and quantitative bioassay for cytokinin. This cytokinin-stimulated response can be inhibited by the plant hormone abscisic acid, ABA; the inhibition is concentration dependent and was proposed for use as a bioassay for ABA. This paper characterizes the ABA inhibition of the cytokinin-stimulated formation of shoot buds. Experiments transferring protonema between cytokinin and cytokinin plus ABA show that ABA does not interfere with the initial perception of cytokinin. Other experiments compare the results of transferring protonema from cytokinin to cytokinin-free medium or to medium with cytokinin plus ABA and reveal that ABA acts by blocking the cytokinin-mediated stable commitment of nascent buds. Extension of the technique of double-reciprocal plots to this whole-organism bioassay finds that ABA is not a competitive inhibitor of cytokinin. Analysis of the ABA inhibition of bud formation identifies a new regulatory step in the developmental process of bud formation in mosses.  相似文献

Abstract

The recolonisation of bryophytes on soil during 3½ years following fire in a mixed forest in south-west Tasmania is characterised by the presence of Marchantia berteroana, Funaria hygrometrica, Ceratodon purpureus and Polytrichum juniperinum, while limited cover is provided by Campy lop us introjlexus, Tortella calycina and Bryum sauteri. It has been verified that the protonemal growth of the early colonising mosses is more rapid than non-colonising specles. The preference for spores of Funaria hygrometrica, Ceratodon purpureus and Marchantia berteroana to germinate on burnt soil is confirmed. However, it has been shown that burnt substrates do not inhibit the germination of non-colonising species. Therefore, the rapid development of protonemata and gametophytes in the colonising species may be a critical factor for their success.  相似文献

Protonemata of the moss Funaria hygrometrica Sibth. (L.) show a strong pH dependence for auxin accumulation. IAA uptake is enhanced when the pH of the incubation medium is lowered from 7.6 to 4. In low light intensity grown protonemata (0.56 W m−2) a component of IAA uptake could be saturated by IAA; efflux of IAA could be inhibited by 2,3,5-triiodobenzoic acid. This is explained by the presence of influx and efflux carriers for IAA. In protonemata grown at high light intensity (2.00-2.30 W m−2) these carriers could not be shown to be present. These results are discussed with regard to the different physiological behavior of moss protonemata grown under different light conditions.  相似文献

Plant growth regulators now include more than the classic examples auxin, cytokinin, ethylene, and gibberellin, but little is known about the activity of these additional classes of molecules in nonvascular plants. The formation of buds by protonema of the moss Funaria hygrometrica is perhaps the best known and most fully characterized developmental system in the nonvascular plants. Examination of the effects of exogenously supplied salicylic acid and acetylsalicylate on this bioassay system shows that salicylates can regulate growth and development in mosses, producing a dose-dependent inhibition of bud formation. Other experiments show that this action is distinct from any direct effect on the well-known cytokinin stimulation of bud formation, occurs late in the process of bud formation, occurs prior to the stable commitment of nascent buds, and is not an effect on the outgrowth of young shoots. Because mosses are the sister clade to the vascular plants, these results suggest that the ability to perceive and transduce salicylate signals is an ancient feature of land plant physiology.  相似文献

The relationship between benzyladenine and assimilate distributions in shoot tips of English Ivy (Hedera helix L.) was studied. Exogenous applications of benzyladenine resulted in small amounts of the hormone moving to the apical meristem. The large part of the recovered benzyladenine was found in the young leaves of the shoot tip. Benzyladenine increased the amount of 14C-labelled assimilates in the shoot tip, but the distribution pattern of those assimilates within the tip was like that of the untreated control.  相似文献

The induction of shoot buds from the filamentous protonema of moss is a classic bioassay for cytokinin. While a large literature documents this response in many species of moss and for a wide range of natural and synthetic cytokinins, to date only substituted adenine cytokinins have been examined in detail. This paper shows that at least some of the novel phenylurea cytokinins will induce bud formation in mosses. Funaria responds to thidiazuron much as it responds to benzyladenine. Exposure to either substance results in log-linear dose-dependent increases in bud number that reach similar maximal numbers of buds at the optimal concentration of compound. The related compound chloro-pyridyl-phenylurea (CPPU) is slightly less active, but induces buds over a wider range of concentration. Carbanilide (diphenylurea or DPU), an active cytokinin in other systems, induces very few buds in Funaria, but does so over a wide range of concentration. Bioassay of mixtures of benzyladenine and DPU finds no evidence of competition for cytokinin receptors. That result could support suggestions that the phenylurea cytokinins act indirectly, by altering endogenous cytokinin metabolism, but we favor another interpretation. Unlike other cytokinin-responsive systems, the induction of buds from moss protonema involves two cytokinin-mediated events. The number of buds is controlled by the second cytokinin-mediated event. If DPU has little or no affinity for the receptor triggering this second event, DPU treatments will produce few to no buds, and kinetic analysis using bud number would find no evidence for competition with benzyladenine. Our results would support the hypothesis that bud induction in Funaria involves two chemically distinct cytokinin receptors.  相似文献

The activity of the alternative path of O2 consumption in detached and intact roots of barley [ Hordeum distichum (L.) Lam. cv. Maris Mink] was determined by titration with salicylhydroxamic acid (SHAM) in the presence and absence of cyanide. In the absence of cyanide, only high concentrations were inhibititory (> 5 m M ). whilst in its presence low SHAM concentrations (2.5–5.0 m M ) gave maximum inhibition: the resulting ϱ Valt plots were non-linear. A SHAM-stimulated peroxidase could readily be washed from these roots, but non-linearity cannot be explained in terms of SHAM-stimulation of this peroxidase as it is not active in the absence of an exogenous supply of NADH. In detached roots the degree of inhibition of respiration with 25 m M SHAM was nearly double the capacity of the alternative path (measured as the degree of inhibition by SHAM in the presence of cyanide), suggesting non-specific inhibition. Effects of SHAM on cytochrome path activity in intact roots were examined by reverse titration with cyanide in the presence and absence of SHAM. At 5 m M SHAM had no effect on the cytochrome path, but at 25 m M it inhibited. We conclude that the only factor causing non-linearity of ϱValt plots in barley roots is non-specific inhibition of the cytochrome path by high concentrations of SHAM; consequently only low concentrations of SHAM (2.5–5.0 m M ) are suitable for estimating alternative path activity in barley roots.  相似文献

The content of endogenous cytokinins, especially zeatin and isopentenyladenine, as well as the dry mass yield, were dependent on the kind of synthetic cytokinin present in the culture medium and decreased in the following order: thidiazuron, kinetin, benzyladenine, no cytokinin. The apparent capacity of the alternative pathway, as measured after blocking of the cytochrome pathway by cyanide, was inversely proportional to the content of endogenous cytokinins. Some synthetic cytokinins (e.g., benzyladenine), when exogenously applied, are known to inhibit selectively the alternative pathway. However, this does not necessarily imply that the mechanism of action of endogenous cytokinins on the respiration pattern is limited to a single effect on the alternative pathway. Blissus occiduus Barber is an important pest of buffalograss, Buchlo? dactyloides (Nuttall) Engelmann, turf. No-choice studies documented the susceptibility of selected turfgrasses, crops, and weeds to B. occiduus feeding. Highly to moderately susceptible grasses included buffalograss; yellow Setaria glauca (L.) and green foxtail Setaria viridis (L.); Kentucky bluegrass, Poa pratensis L.; perennial ryegrass, Lolium perenne L.; brome, Bromus spp. Leyss.; zoysiagrass, Zoysia japonica Steudel; Bermuda grass, Cynodon dactylon (L.) Pers.; sorghum, Sorghum bicolor (L.) Moench; tall fescue, Festuca arundinacea Schreb.; and barley Hordeum vulgare (L.). Slightly to nonsusceptible grasses included fine fescue, Festuca ovina hirtula L.; rye, Secale cereale L.; crabgrass Digitaria sanguinalis (L.); bentgrass, Agrostis palustris Huds.; wheat, Tritium aestivun L.; corn, Zea mays L.; fall panicum Panicum dichotomiflorum Michx.; and St. Augustinegrass, Stenotaphrum secundatum (Walt.) Kuntze. The reproductive potential of B. occiduus was also investigated on these same grasses. B. occiduus produced offspring on 15 of the 18 turfgrass, crop, and weed species evaluated. No reproduction occurred on either Bermuda grass or St. Augustinegrass, and buffalograss plants were killed by B. occiduus feeding before offspring could be produced.  相似文献

Pollen of pigeonpea(Cajanus cajan (L.) Millsp.) cultivars H-77-216 and ICPL-151 were cultivatedin vitro at six different temperatures (12, 17, 22, 27, 32, 37 °C). Pollen of cv. H-77-216 started to germinate at 17 °C whereas the pollen of cv. ICPL-151 at 22 °C, the optimal temperatures were 22 and 27 °C, respectively. Pollen germination at different temperatures was found to be positively correlated with the tube length. Per cent pollen bursting increased with rising temperature. The indeterminate cv. H-77-216 showed a wide range of suitable temperatures (17 – 27 °C) for pollen germination while the determinate cv. ICPL-151 had optimum at 27 °C  相似文献

Chilling effects on respiration during the recovery period were studied in two maize (Zea mays L.) cultivars differing in their tolerance to chilling: Penjalinan, a chilling-sensitive cultivar, and Z7, a chilling-tolerant cultivar. Both cultivars were exposed to 5 degrees C for 5 d, after which measurements were taken at 25 degrees C. Chlorophyll fluorescence analysis in dark-adapted leaves showed less damage in cv Z7 than in cv Penjalinan during recovery from the chilling treatment. Studies of the electron partitioning between the cytochrome and the alternative respiratory pathways during chilling recovery using the oxygen isotope fractionation technique showed that, although total leaf respiration was not affected by the chilling treatment in either of the two cultivars, electron partitioning to the alternative pathway was significantly increased in the more stressed chilling-sensitive cv Penjalinan, suggesting that increased activity of the alternative pathway is not related to the plant tolerance to chilling. These results suggest a possible role of the alternative pathway in plants under stress rather than specifically contributing to plant resistance to chilling.  相似文献

Observations in the held suggest that moss shoots are seldom freely consumed by either vertebrate or invertebrate herbivores, but damage to immature capsules by slugs is abundant. Palatability of the mosses Mnium hornum, Brachythecium rutabulum and Funaria hygrometrica to the generalist herbivores, Arion subfuscus and A. rufus was investigated by offering the slugs four stages in the moss life cycle; protonema, leafy shoot, immature capsule and mature capsule. The slugs showed a preference for immature capsules and protonemata in all moss species tested. Consumption of shoots of M. hornum and B. rutabulum was negligible even after seven days starvation, but some shoots of F. hygrometrica were eaten in every overnight trial performed. Shoot extracts on starch-wafer substrate appeared to be more acceptable than intact shoots suggesting that the cell wall provides the barrier to free consumption. The form and location of a possible feeding deterrent is discussed.  相似文献

The effects of KCN (0.5mmol/L) and NaN3 (0.01 mmol/L) pretreatment on the operation of the alternative pathway in subcultured tobacco (Nicotiana rustica L. cv. Gansu yellow flower) callus were analyzed. After treatment with KCN and NaN3 for 12 h, the total respiration rate (Vt) decreased by 12% and 17%, whereas oxygen consuption by the cytochrome pathway decreased by 22% and 28% respectively. The capacity of the alternative pathway (Valt) remained constant, while the activity of the alternative pathway (ρ· Valt ) inreased slightly. This changing pattern led to a declined contribution of the cytochrome pathway to the total respiration rate and an increased activity of the alternative pathway. Treatment with KCN for 24 h brought about a slight rise of oxygen consumption by the cytochrome pathway as compared with that in callus treated for 12 h, but the oxygen consumption was still lower than that in the untreated callus. Treatment with NaN3 for 24 h resulted in a profound decrease of the cytochrome pathway operation and a continuing increase of the alternative pathway operation. These data indicated that the enhanced operation of the alternative pathway played a compensatory role to the total respiration when the cytochrome pathway was partially inhibited in tobacco callus.  相似文献
