The lipid content and composition of Nereis (Hediste) diversicolor O. F. Müller (Annelida, Polychaeta, Nereidae) a mud-dwelling, intertidal errant polychaete in the Tagus estuary (Portugal), were examined on the monthly basis by lipid extraction, TLC and capillary GC. In this estuary, N. diversicolor is by far the dominant species among polychaeta and the main food item in the natural diet of several flatfishes. The biochemical elucidation of its lipid structure and distribution throughout the year, described in this study, provides information not only about the physiological role of lipids in the animal under consideration but also about dietary fatty acid requirements of some flatfishes in the wild and under laboratory conditions.The total lipid content varied between a maximum of 19.3% lyophilized dry weight in February (4.4% fresh weight) and a minimum of 6.6% in August (1.9% fresh weight). The major lipid classes were triacylglycerol, phospholipid, free sterol, free fatty acid, sterol ester/wax ester and alkyldiacylglycerol.The fatty acid composition was rather unsaturated with a 1:2 mean ratio of n-3: n-6. The major fatty acids were C160:0, C18:1n-9, C18:2n-6, and C20:5n-3; there were smaller amounts of C180:0, C18:1n-11, C18:1n-7, C18:3n-3, C20:1, C20:2n-6, C20:4n-6, C22:2, C22:5n-3, and many other fatty acids were detected at trace levels. The unsaturation ranged from 36.9 mg/g dry weight in summer to 107.4 mg/g in winter. An accumulation of fatty acids from plant origin was evident, in particular linoleic acid (C18:2n-6), which was quantitatively one of the major fatty acids throughout the year.  相似文献

The root tuber peel of Flemingia vestita has been in use in local traditional medicine against intestinal worm infections in Meghalaya (North-East India). In order to evaluate and authenticate the anthelminitc efficacy of the isoflavones of F. vestita, the root peel extract of this putative plant was tested against several helminth parasites, extensively on Rallietina echinobothrida, with respect to different parameters of these parasites. In this paper, we describe various methods to evaluate the anthelmintic efficacy of this medicinal plant with respect to carbohydrate metabolism in R. echinobothrida at paralytic time caused by the isoflavones of F. vestita. To meet the high energy demand by the parasite due to the anthelmintic stress, glucose breakdown follows the PEPCK-malate pathway in the parasite.  相似文献

【目的】探究缺失编码丙酮酸脱氢酶蛋白的aceE基因对猪链球菌生长特性、三羧酸循环和丙酮酸代谢的影响。【方法】通过测量菌液的OD600值,绘制野生型菌株与aceE基因缺失突变株的生长曲线;利用试剂盒测定三羧酸循环和丙酮酸代谢旁路中乙酰CoA、琥珀酸CoA、延胡索酸、草酰乙酸、丙酮酸、乳酸和ATP的含量,通过荧光定量qRT-PCR确定柠檬酸合酶基因、苹果酸脱氢酶基因、琥珀酸脱氢酶基因、异柠檬酸脱氢酶基因、丙酮酸脱羧酶基因、乳酸脱氢酶基因、乙醇脱氢酶基因和乙醛脱氢酶基因的表达水平。【结果】与野生株相比,菌株ΔaceE在平台期OD600值下降;添加1g/L乙酸盐能够显著提升菌株ΔaceE平台期OD600值。菌株ΔaceE的丙酮酸含量上升,ATP含量下降;三羧酸循环代谢中乙酰CoA、琥珀酸CoA、延胡索酸含量降低;柠檬酸合酶基因和苹果酸脱氢酶基因表达水平上升,琥珀酸脱氢酶基因和异柠檬酸脱氢酶基因表达水平下调;在丙酮酸代谢旁路中丙酮酸脱羧酶基因、乳酸脱氢酶基因、乙醇脱氢酶基因和乙醛脱氢酶基因表达水平上升。【结论】结果显示,菌株ΔaceE三羧酸循环活性降低,虽然能够通过PDH旁路将部分丙酮酸分解为乙...  相似文献

The hippopotamus (Hippopotamus amphibius) spends more than 12 h a day in the water. Hippos are often submitted to water temperatures that vary with the seasons. We hypothesize that this difference between cool and warm water temperatures leads to variations in behavioural thermoregulation. We recorded the exposure of hippos to sunshine at the beginning and at the end of the dry season. Our results show that (1) sunshine exposure lasted much longer in cool water and (2) sun-bathing occurred during the hottest hours. It is therefore likely that in cool water hippos were cold and expressed an original behaviour of search for heat.  相似文献

We studied the seasonal variation on aerobic metabolism and the response of oxidative stress parameters in the digestive glands of the subpolar limpet Nacella (P.) magellanica. Sampling was carried out from July (winter) 2002 to July 2003 in Beagle Channel, Tierra del Fuego, Argentina. Whole animal respiration rates increased in early spring as the animals spawned and remained elevated throughout summer and fall (winter: 0.09 ± 0.02 μmol O2 h− 1 g− 1; summer: 0.31 ± 0.06 μmol O2 h− 1 g− 1). Oxidative stress was assessed at the hydrophilic level as the ascorbyl radical content / ascorbate content ratio (A / AH). The A / AH ratio showed minimum values in winter (3.7 ± 0.2 10− 5 AU) and increased in summer (18 ± 5 10− 5 AU). A similar pattern was observed for lipid radical content (122 ± 29 pmol mg− 1 fresh mass [FW] in winter and 314 ± 45 pmol mg− 1 FW in summer), iron content (0.99 ± 0.07 and 2.7 ± 0.6 nmol mg− 1 FW in winter and summer, respectively) and catalase activity (2.9 ± 0.2 and 7 ± 1 U mg− 1 FW in winter and summer, respectively). Since nitrogen derived radicals are thought to be critically involved in oxidative metabolism in cells, nitric oxide content was measured and a significant difference in the content of the Fe–MGD–NO adduct in digestive glands from winter and summer animals was observed. Together, the data indicate that both oxygen and nitrogen radical generation rates in N. (P.) magellanica are strongly dependent on season.  相似文献

Two fruit-feeding insects, a gall wasp, Allorhogas sp. (Hymenoptera: Braconidae), and a beetle, Apion sp. (Coleoptera: Curculionoidea), were evaluated in their native habitat in Brazil as potential biological control agents of Miconia calvescens DC (Melastomataceae). Allorhogas sp. occurred at two out of three field sites with native populations of M. calvescens, and Apion sp. occurred at all three sites. Both species exhibited aggregated distributions among M. calvescens trees sampled at each site. Allorhogas sp. infested 9.0% and 3.8% of fruits at each of two sites. The number of larvae and pupae of Allorhogas sp. and/or an unidentified parasitoid (Hymenopetera: Eulophidae: Tetrastichinae) ranged from one to five per infested fruit. Fruits infested with Allorhogas sp. were 20% larger and had 79% fewer seeds than healthy fruits. Although adults of Apion sp. were found on leaves and inflorescences of M. calvescens at all three sites, larvae and pupae were found in fruits at only one site, where a maximum of 1.4% of fruits were infested. Fruits infested by Apion sp. contained only one larva or pupa, and were 15% smaller and had 62% fewer seeds than healthy fruits. While a variety of apionids have been used for biological control in the past, this is the first time a braconid wasp has been considered for biological control of a weed.  相似文献

The seasonal growth and nutrient status of the temperate subtidal macroalga Adamsiella chauvinii was determined at three soft sediment sites with different ranges of tidally-driven current speeds. A. chauvinii thalli exhibited maximum growth rates during the late summer (February) and no evidence of nitrogen limited growth in summer as often exhibited by macroalgae growing on hard substrata in temperate environments. We suggest that growth was maintained during the summer period, when seawater nitrogen concentrations were low, by localized sources of ammonium probably produced by sediments and fauna within the canopy. Growth rates were up to 50% lower at the site with the slowest water velocity throughout most of the experimental period. However a greater tissue nitrogen content of A. chauvinii thalli at the slow flow site did not support evidence for mass-transfer limitation. It is suggested that as a consequence of slow flow, sedimentation on blade surfaces reduced the amount of light available for photosynthesis and growth. Lower growth rates at the slow flow site reduced metabolic demand for nutrients resulting in accumulation of tissue nitrogen and phosphate.  相似文献

Summary In a population of Little Owls (Athene noctua) at the Lower Rhine (FRG), the factors were studied that influence the size and shape of territories of this non-migrating, all-year territorial owl species. These studies were carried out between September 1984 and June 1987. The birds were individually marked with transmitter packages so that their locomotional movements could be monitored. Using a standard protable stimulus (a dummy with an attached loudspeaker) the territorial boundaries of 19 male Little Owls were surveilled during the different seasons of the year. Male Little Owls defended their territories in all seasons, but distinct seasonal changes of intraspecific aggressiveness and territory size were observed. The seasonal variations of territory size followed a common pattern in all studied Little Owls, in spite of individual differences in the size of the defended areas. The largest territories were defended in March/April during the courtship season . In May/June all male Little Owls reduced the size of their territories . This corresponds to the breeding and nestling period. The yearly minimum of territory size was reached in the summer months July and August when the fledglings were still fed in the parental territory. When the first-year birds started to disperse in autumn (September/October), the size of the defended areas of the adult tenants again increased . In winter (November to February), a further increase in territory size was observed for periods of warm weather . Male Little Owls, however, were less aggressive during winter than in the following courtship season. On cold winter days with a ground cover of snow territorial aggressiveness ceased. Pastures and meadows offered a continuous food supply almost throughout the year. In some seasons they were overproportionally represented in the territories of Little Owls compared to the general surroundings. In reaction to changing accessibility of food, the hunting ranges within the home ranges were shifted much faster than the boundaries of the defended territories. The significance of various factors contributing to the variability of territory size in Little Owls are discussed (e.g. availability of food, seasonal and individual differences in aggressiveness, experience in the occupied area, population density).  相似文献

Results of molecular studies regarding the phylogenetic placement of the order Ostropales and related taxa within Lecanoromycetes were thus far inconclusive. Some analyses placed the order as sister to the rest of Lecanoromycetes, while others inferred a position nested within Lecanoromycetes. We assembled a data set of 101 species including sequences from nuLSU rDNA, mtSSU rDNA, and the nuclear protein-coding RPB1 for each species to examine the cause of incongruencies in previously published phylogenies. MP, minimum evolution, and Bayesian analyses were performed using the combined three-region data set and the single-gene data sets. The position of Ostropales nested in Lecanoromycetes is confirmed in all single-gene and concatenated analyses, and a placement as sister to the rest of Lecanoromycetes is significantly rejected using two independent methods of alternative topology testing. Acarosporales and related taxa (Acarosporaceae group) are basal in Lecanoromycetes. However, if the these basal taxa are excluded from the analyses, Ostropales appear to be sister to the rest of Lecanoromycetes, suggesting different ingroup rooting as the cause for deviating topologies in previously published phylogenies.  相似文献

Leishmania spp are protozoans capable of carbohydrates degradation and as energy source they can use glucose, aminoacids or lipids from the environment. The products of the metabolic pathways such as organic acids may be used as an index of their energetic metabolic profile. Therefore, in this study a metabolic profile comparison was made between promastigotes from one reference strain (MHOM/BR/1975/M2903) and two different isolates of Leishmania (Viannia) braziliensis (MHOM/BR/2003/IMG3 and MHOM/BR/2005/RPL5). The parasites culture was performed in complete Grace’s culture media seeded in 24-well plates at 26 °C. During the growth curve performance samples were collected from the logarithmic and stationary phases of culture and therefore analyzed by high performance liquid chromatography (HPLC) and spectrophotometry assays to determine the concentrations of glucose, lactate, citrate, α-ketoglutarate, succinate, fumarate, malate, oxaloacetate and β-hydroxybutirate which are indicative of the energetic pathways. It was possible to detect an increase in the glucose from the stationary phase from the M2903 strain when compared to the logarithmic phase while in the IMG3 and RPL5 isolates there was a decrease (p < 0.05). The spectrophotometric and chromatographic results indicated that the logarithmic phase which presents higher energy consumption due to the intense replication rate have the energetic pathways intensified. It was also possible to note some metabolic differences between the analyzed parasites which may indicate possible adaptations of the parasite when facing different environmental and physiological conditions during its life cycle and that these differences may help in the understanding of the diversity of the host-parasite relationship from Leishmania parasites.  相似文献

A proteinaceous trypsin inhibitor was purified from Crotalaria pallida seeds by ammonium sulfate precipitation, affinity chromatography on immobilized trypsin-Sepharose and TCA precipitation. The trypsin inhibitor, named CpaTI, had M(r) of 32.5 kDa as determined by SDS-PAGE and was composed of two subunits with 27.7 and 5.6 kDa linked by disulfide bridges. CpaTI was stable at 50 degrees C and lost 40% of activity at 100 degrees C. CpaTI was also stable from pH 2 to 12 at 37 degrees C. CpaTI weakly inhibited chymotrypsin and elastase and its inhibition of papain, a cysteine proteinase, were indicative of its bi-functionality. CpaTI inhibited, in different degrees, digestive enzymes from Spodoptera frugiperda, Alabama argillacea, Plodiainterpunctella, Anthonomus grandis and Zabrotes subfasciatus guts. In vitro and in vivo susceptibility of Callosobruchus maculatus and Ceratitis capitata to CpaTI was evaluated. C. maculatus and C. capitata enzymes were strongly susceptible, 74.4+/-15.8% and 100.0+/-7.3%, respectively, to CpaTI. When CpaTI was added to artificial diets and offered to both insect larvae, the results showed that C. maculatus was more susceptible to CpaTI with an LD(50) of 3.0 and ED(50) of 2.17%. C. capitata larvae were more resistant to CpaTI, in disagreement with the in vitro effects. The larvae were more affected at lower concentrations, causing 27% mortality and 44.4% mass decrease. The action was constant at 2-4% (w/w) with 15% mortality and 38% mass decrease.  相似文献

铁离子是大多数细菌生存所必需的一种营养物质,但过多的铁离子会通过芬顿反应产生的活性氧对细菌造成损伤。因此,细菌通过摄取、调控、螯合、外排等机制维持体内铁离子的稳态。鸭疫里默氏杆菌(Riemerella anatipestifer)是一种最新被归类于威克斯菌科里氏杆菌属的革兰氏阴性菌。该菌主要感染禽类,参与该菌的铁离子代谢基因具有特别之处。本文对鸭疫里默氏杆菌铁离子代谢机制研究进展进行了系统总结和阐述,包括该菌的TonB系统、TonB依赖性受体、Fur蛋白及Dps蛋白等在铁离子转运、调控、螯合中的功能,以及以上蛋白在鸭疫里默氏杆菌致病中的作用,以期更全面地理解鸭疫里默氏杆菌铁代谢机制,并为进一步深入研究该菌铁离子代谢提供理论依据和参考。  相似文献

含水量是影响种子超低温保存效果的关键因素,而其作用机制尚不完全清楚。为探讨含水量对种子超低温保存生活力的影响途径,该研究以八棱海棠种子为材料,通过硅胶干燥法获得不同含水量的种子,测定超低温保存后种子生活力、糖含量及相关酶指标的变化并分析相关性。结果表明:(1)超低温保存15 d后,不同含水量种子生活力不同,随着种子含水量的降低,种子生活力呈现先升高后降低的趋势,含水量为9.02%的八棱海棠种子生活力最高,为53.33%;超低温保存120 d后,种子生活力随着含水量下降一直升高,含水量为6.40%生活力最高,为27.78%。这表明八棱海棠种子含水量对超低温保存后的生活力有明显影响,但受液氮保存时间影响,随着液氮保存时间的延长,最适含水量降低。(2)相关分析显示,超低温保存后种子含水量与生活力呈极显著负相关(r=-0.82);与果糖和蔗糖含量、酸性转化酶、果糖激酶呈显著负相关,而种子萌发率与这些指标呈显著正相关。这表明种子含水量通过影响酸性转化酶活性而影响蔗糖和果糖含量,进而影响蔗糖代谢,响应低温和脱水胁迫,最终导致生活力差异。种子生活力还受到介导果糖激酶的果糖代谢影响。此外,海藻糖也是种...  相似文献

采用实验生态学方法,在室内水槽条件下研究了金乌贼(Sepia esculenta Hoyle,1885)繁殖过程中社群等级的形成对其行为表型和能量代谢的影响,分析测定了不同优势等级雌雄个体腕部肌肉和性腺组织中己糖激酶(Hexokinase, HK)、丙酮酸激酶(Pyruvate kinase, PK)、乳酸脱氢酶(Lactate dehydrogenase, LDH)、苹果酸脱氢酶(Malate dehydrogenase, MDH)、柠檬酸合酶(Citrate synthetase, CS)活性以及乳酸(Lactic acid, LD)含量。结果显示:(1)金乌贼繁殖期不同优势等级雌雄个体之间行为表型具有显著差异,优势雄性个体游动悬浮、争斗时间显著高于劣势个体,而优势雌性个体静止伏底时间高于劣势个体,游动悬浮时间低于劣势雌性;(2)优势雄性个体在争斗过程中主要通过无氧代谢提供能量,而处于游动悬浮状态时通过有氧代谢提供能量。主要表现在优势雄性个体肌肉中无氧代谢酶(PK、HK、LDH)活性显著高于劣势个体(P<0.05),有氧代谢酶(MDH、CS)活性也显著高于劣势个体,雌性个体之...  相似文献

In 2002 and 2003, a study was conducted to determine the effect of bacterial strains, Burkholdria OSU 7, Bacillus OSU 142, and Pseudomonas BA 8, on biological control of brown rot disease (Monilinia laxa Ehr.) on apricot cv. Hacıhaliloğlu in Malatya province of Turkey. Apricot orchard at full blooming stage was inoculated with conidial suspension (1 × 106 spores/ml) of M. laxa Ehr. After inoculation, two apricot trees for each application were treated with each of the three biological control agents (Burkholdria gladii OSU 7, Bacillus subtilis OSU 142, and Pseudomonas putida BA 8) by spraying (1 × 109 cfu/ml) on inoculated branches. Disease incidence was evaluated for untreated (control 1) and four different treatment groups including commercial disease management (control 2, positive control: 3% Bourdox in fall, 50% Cupper at pink flower, 30 g/100 l Corus at first blooming, and 300 g/100 l Captan at last blooming stage) and treatments including each of the three bacterial strains (OSU 7, OSU 142, and BA 8). The results showed that disease incidence for negative control (control 1) was 9.94, which was significantly higher than disease incidence for commercial application (2.57%) or bacterial treatments (2.82–5.00%) in the first year. In 2003, the lowest disease incidence observed in OSU 7 treatment (6.80%), while disease incidence rate for positive control and negative control were 9.45% and 28.46%, respectively. This result may suggest that OSU 7 has potential to be used as biopesticide for effective management of brown rot disease on apricot.  相似文献
