To more completely assess the means by which alcohol impairs the female reproductive cycle in rats, we have measured hypothalamic luteinizing hormone-releasing hormone (LHRH), pituitary LHRH receptor content, and the serum levels of luteinizing hormone (LH), follicle-stimulating hormone (FSH), prolactin (Prl), and progesterone (P). After two successive cycles, the animals began receiving either an alcohol or a isocaloric control liquid diet regimen beginning on the first day of diestrus, with continued monitoring of the estrous cycle throughout the experiment. An additional set of controls consisted of animals maintained on lab chow and water provided ad libitum. Our results indicate that those animals receiving the control diets showed uninterrupted estrous patterns, whereas those animals receiving the alcohol diet remained in diestrus. Additionally, the alcohol-treated animals showed an increase (p less than 0.05) in LHRH content, with a concomitant decrease (p less than 0.01) in serum LH, and an increase (p less than 0.01) in serum Prl. No significant differences were detected in serum FSH levels or pituitary LHRH receptor content. No differences were detected in serum P levels. These results indicate that short-term alcohol administration disrupts the female reproductive cycle, causing persistent diestrus, and support our hypothesis that the alcohol-induced depression in serum LH levels is due to a diminished release rate of hypothalamic LHRH.  相似文献

The effect of experimentally-induced diabetes mellitus on reproductive organ weights, serum and pituitary gonadotropin levels and serum testosterone levels was studied in 3-month old rats. In experiment 1, intact rats were treated with alloxan monohydrate or streptozotocin. In experiments 2 and 3, intact and castrated rats were rendered diabetic with alloxan (experiment 2) or streptozotocin (experiment 3). The duration of each experiment was 3 weeks. In each experiment diabetes resulted in body weight losses or reduced body weight gain, elevated serum glucose concentrations and reduced assessory sex gland weights (intact rats). Serum levels of testosterone were depressed (P less than 0.05 or P less than 0.01) in diabetic rats. Serum levels of LH were significantly (P less than 0.05) lower in intact diabetics than in controls when pooled data from the three experiments were compared. Serum levels of FSH were not affected by diabetes. Pituitary concentrations of FSH were elevated (P less than 0.05) in diabetics in two of the three experiments, while LH concentrations were elevated (P less than 0.05 or P less than 0.01) in diabetics in all experiments. The hypersecretion of gonadotropins in castrated rats was not affected by diabetes.  相似文献

Timing of ovulation and changes in plasma progesterone, luteinizing hormone (LH), and prolactin (PRL) during periovulatory stages were determined in Holtzman rats exhibiting regular 4- or 5-day cycles under a daily artificial illumination from 0500 to 1900 h. The 5-day cycling rats ovulated between 0130 and 0930 h on estrus, whereas some of the 4-day cycling animals ovulated as early as about 0130 h and others as late as 1130 h on estrus. Onset time of preovulatory LH and progesterone surges was about 1500 h on proestrus in both the 4- and the 5-day cycling rats. Peak levels of plasma LH and progesterone were measured at 1700 to 1900 h on proestrus, while the first rises and peak values of plasma PRL were evident a few hours earlier than those of plasma LH in the rats with two cycle lengths. Plasma LH levels at 1900 h on proestrus as well as plasma progesterone levels at 1600 and 2300 h on proestrus and at 0130 and 0330 h on estrus were significantly lower in the 5-day cycling rats than in the 4-day cycling animals (p less than 0.05). In contrast, PRL levels from 1500 through 2300 h on proestrus remained consistently higher in 5-day cycling rats than in 4-day cycling rats, and significant differences in PRL levels between these rats were apparent at 1500, 1600, and 2100 h (p less than 0.05-0.01). Thus, these results demonstrate that the 5-day cycling rats exhibit the attenuated magnitude of LH surge accompanied by the augmented preovulatory PRL release, and that plasma progesterone levels reflect the magnitude of LH surge. A tentative working hypothesis concerning the etiology of the 5-day cycle has been proposed.  相似文献

Coccinia indica (Family: Cucurbitaceae, locally known as telakucha) leaves were extracted with 95% ethanol. Following evaporation of the solvents, the residue was suspended in distilled water. When this suspension was fed orally to male normal-fed and 48-hr starved rats, the blood glucose was lowered 21% (P less than 0.01) in normal-fed and 24% (P less than 0.001) in 48-hr starved animals respectively. Starvation had induced a 3-fold increase in the activity of glucose-6-phosphatase and this activity was depressed 19% (P less than 0.05) by extract feeding while basal activity of the enzyme in normal-fed rats remained unaffected. Consistent with the depression of glucose-6-phosphatase, urea cycle enzyme arginase was also depressed 21% (P less than 0.001) and 12% (P less than 0.01) in the liver of 48 hr-starved and normal-fed animals respectively. Unlike glucose-6-phosphatase, starvation induced levels of gluconeogenic enzymes alanine aminotransferase and aspartate aminotransferase were not affected by Coccinia extract. These results suggest that the hypoglycemic effect of C. indica is partly due to the repression of the key gluconeogenic enzyme glucose-6-phosphatase.  相似文献

In order to further investigate the previously reported hypogonadal state of chronically uremic rats, we examined the effects of in vivo pretreatment with human chorionic gonadotropin (hCG) on in vivo and in vitro Leydig cell function, comparing paired intact rats with rats made chronically uremic by 5/6 nephrectomy. The in vitro testosterone (T) secretory responses to varying concentrations of hCG or dibutyryl cAMP and the number of gonadotropin receptors were determined following hemicastration. The rats were then treated with hCG for 3 days and the remaining testes were removed and studied as before. Compared with intact rats, the uremic rats had higher serum concentrations of urea nitrogen (P less than 0.001); serum T concentrations were lower in uremic rats before (P less than 0.001), but not after (P greater than 0.6) treatment. Treatment produced increases in serum T only in uremic rats (P less than 0.001). Serum LH was lower in uremic rats before treatment (P less than 0.001) and was reduced (P less than 0.001) to similar levels (P greater than 0.8) in both groups after treatment. Baseline in vitro T secretion was lower (P less than 0.001) from Leydig cells of uremic than intact rats both before and after treatment. Analysis of variance of dose-response curves showed pre- and post-treatment T secretory responses to hCG or dibutyryl cAMP in vitro to be less from Leydig cells of uremic rats (P less than 0.01). Nonstress blood samples were obtained from intact and thyroidectomized (TE) male rats at 3-hr intervals over a 24-hr period via rapid decapitation. The animals were thyroidectomized when 40 days old and used 6 weeks later. Intact animals showed periodicity in serum LH (P less than 0.01) and prolactin (P less than 0.01). Both gonadotropins began increasing after 8 PM and peak levels occurred at 11 PM. In contrast, 24-hr periodicity was not observed in serum FSH. Corticosterone levels in these same serum samples showed the expected circadian periodicity. After TE, the 24-hr pattern in all gonadotropins was altered significantly. Serum LH increased (P less than 0.01) and circadian periodicity appeared to be absent. FSH and prolactin levels were increased and decreased, respectively (P less than 0.01), with serum prolactin showing a 9-hr phase shift. Prolactin began increasing at 2 AM and reached a peak at 8 AM. Corticosterone in TE animals showed a 24-hr rhythm similar to that of intact rats. These findings confirm our previous observations that nonstress serum LH and prolactin levels fluctuate with a 24-hr periodicity and suggest that the level of, and the phase angle betweeen, these rhythms is markedly influenced by pituitary-thyroid activity.  相似文献

The primary objectives of this study were to: 1) determine if removal of 1.5 ovaries from young rats would mimic reproductive characteristics that normally occur with advancing age and 2) determine if removal of 1.5 ovaries from aged rats would further advance the process of reproductive aging. Removal of 1.5 ovaries increased the number of young (P less than 0.05) and old (P less than 0.01) rats that exhibited abnormal estrous cycles. In addition, concentrations of follicle-stimulating hormone (FSH) were higher at both ages in the groups with half an ovary. The increased concentrations of FSH are consistent with a decrease in the number of growing follicles after removal of 1.5 ovaries. All groups had lower concentrations of estradiol (E2) than young controls. There was a significant increase in the number of abnormal embryos with age and removal of 1.5 ovaries when rats were mated during a 5-day estrous cycle, but there was no effect if they were mated during a 4-day estrous cycle. From the results of this study, we conclude that the reduction in ovarian tissue in young and aged rats mimicked several reproductive characteristics of advancing age. Also, an effect of aging on the hypothalamus was evident in this study.  相似文献

Administration of polyethylene glycol (PEG, intraperitoneal, 3 ml, 30% solution) to adult male rats (300 g) resulted in an approximately 20% increment in plasma volume (PV) 24 h after PEG injection. When these animals were exercised (9.14 m/min, level treadmill) in a warm (30 degrees C, 30-40% relative humidity) environment, their mean endurance was increased from 67.9 (saline-treated controls, CONT) to 93.6 min (P less than 0.01). Total water loss was increased from 12.2 (CONT) to 17.2 g (PEG, P less than 0.01). Atropine administration (ATR, 200 micrograms/kg, tail vein) significantly (P less than 0.05) reduced both the endurance and the salivary water loss of CONT and PEG-treated rats, whereas it increased the heating rate (P less than 0.01) of both groups. PEG treatment reduced (P less than 0.01) the hematocrit and circulating protein levels both before and subsequent to exercise in the warm environment. Clinical chemical indexes of heat/exercise injury were generally unaffected by pharmacological intervention, whereas clinical chemical responses to exercise were related to the endurance time of each group. We concluded that expansion of PV by PEG provided significant beneficial effects on performance and thermoregulation during exercise in a warm environment.  相似文献

To determine if deficits in sexual activity observed in pituitary-grafted male rats are due to elevated serum prolactin (PRL) levels found in these animals, the effects of whole pituitary grafts, pars distalis grafts, and ovine (o) PRL treatment on male copulatory behavior were compared. Adult sexually experienced CDF male rats were given four whole pituitary grafts, four pars distalis grafts, or were sham operated. Both groups of grafted animals exhibited suppressed copulatory behavior patterns when tested 18 days after pituitary transplantation. Animals given whole pituitary grafts had significantly longer latencies to mount (P less than 0.05) and to intromit (P less than 0.01) than did the sham-operated controls, while the animals given anterior pituitary grafts differed from the sham-operated controls in latencies to mount (P less than 0.05) and to intromit (P less than 0.01), as well as in the number of intromissions (P less than 0.05). Prolactin-injected animals had significantly reduced intromission rates (P less than 0.01) and significantly increased latencies to mount (P less than 0.05) and to intromit (P less than 0.01) when compared to vehicle-injected controls. Furthermore, the time course of behavioral suppression was similar in oPRL-treated animals to that observed in pars distalis-grafted males, with both groups showing the onset of deficits in sexual activity within 8 to 9 days from the induction of the hyperprolactinemic state. The similarity in pattern and time to onset of behavioral suppression in pituitary-grafted and oPRL-treated animals suggests that behavioral deficits observed in animals with pituitary grafts result from chronic elevation of serum PRL levels.  相似文献

A synthetic diet preparation supplemented with 10% by weight of either safflower oil, hydrogenated coconut oil containing 3% safflower oil, or 'max EPA' fish oil was fed to rats over a 8-week period. Serial measurements of serum fatty acids, serum thromboxane B2 and urinary prostaglandin excretion were taken during the treatment period to assess the rate of change in fatty acid composition and prostaglandin synthesis following dietary manipulation. There was no significant change in weight gain between the dietary groups during the treatment period. Significant changes in serum fatty acids occurred within 48 h of treatment, with the 'max EPA' oil group having arachidonic acid levels reduced by 23% (P less than 0.01) compared to the coconut oil group. Conversely, rats fed safflower oil had an 18% enhancement of arachidonic acid during the same time period. Whole blood synthesis of thromboxane B2 was significantly depressed (P less than 0.01) after 48 h in rats fed 'max EPA' oil compared to the safflower oil or coconut oil groups. This suppression reached a maximum of 65% (P less than 0.001) after 7 days of dietary 'max EPA' oil treatment. The safflower oil and coconut oil-fed groups showed the same levels of serum thromboxane B2 production over the treatment period. Urinary excretion of both 6-ketoprostaglandin F1 alpha and prostaglandin E2 varied significantly (P less than 0.01) between the groups after 7 days of dietary treatment. Rats fed 'max EPA' oil had depressed urinary prostanoid excretion compared to the safflower and coconut oil groups which remained very similar to each other. After the 8-week treatment period rats were killed and the phospholipid fatty acid composition and prostaglandin-generating capacity of platelets, aorta and renal tissue was examined. Prostanoid production by kidney cortex and medulla and segments of aorta was consistently suppressed in rats fed 'max EPA' oil. These observations correlated well with changes in the phospholipid fatty acid profiles in these tissues. This study shows rapid changes in serum fatty acids and thromboxane B2 generation following dietary manipulation, while changes in urinary excretion or prostanoid metabolites occur only after a longer time period.  相似文献

The effect of human chorionic gonadotropin (hCG) on in vitro progesterone (P) level in the uterus was investigated by using short-term incubations of uterine tissue taken from 4-day cyclic rats at different stages of the estrous cycle. Control incubations resulted in a decrease of endogenous P content in uteri removed from rats in proestrus (PRO), estrus (EST), and metestrus (MET): -25% (n = 6), -60% (n = 6), and -45% (n = 8), respectively. The amount of P found after incubation of MET tissue in the presence of hCG was significantly higher (p less than 0.001) than that found after control incubations. The hCG effect was dose-dependent and was not observed with PRO or EST tissue. Although the mean P level found in MET tissue after incubation with 10 IU/ml hCG was not significantly different from the mean level found in unincubated tissue (1562 +/- 341 vs. 1470 +/- 174 pg/mg protein, n = 8), an obvious synthesis was observed in two experiments. It thus seems likely that observed hCG effect would involve a de novo P synthesis rather than a decrease of P catabolism. Furthermore, hCG induced a dose-dependent increase of cyclic adenosine 3', 5'-monophosphate (cAMP) uterine levels in MET tissue. N,O'-dibutyryl cyclic AMP [Bu)2 cAMP) at 5 mM induced a significative increase (p less than 0.01) of P uterine level in EST and MET tissue compared to control incubations, but had no effect on PRO tissue. In most mammals, pituitary-testicular hormone secretion is very active during the perinatal period, but the physiological significance of this function for later pituitary-gonadal interactions and sexual maturation is largely unknown. Short-term neonatal treatment with gonadotropin-releasing hormone (GnRH) antagonist results in delayed sexual maturation and infertility in male rats. We have now extended our earlier findings and studied in more detail the pituitary-gonadal function in adult rats after such neonatal treatment. In this study, the pituitary-testicular activity of newborn male rats was temporarily blocked by treatment with a GnRH antagonist analogue (N-Ac-4-Cl-D-Phe1, 4-Cl-D-Phe2, D-Trp3, D-Phe6, des-Gly10-GnRH-D-alanylamide; Organon 30039; 2 mg/kg s.c. twice daily) on Days 1-5 of life. Timing of puberty was slightly delayed in the treated rats (average: 2 days, p less than 0.05), as determined by the age of the balano-preputial separation. In adult rats (90-110 days), only 3 of the 17 rats treated neonatally with GnRH antagonist were fertile (14 of 17 controls, p less than 0.01), despite normal circulating androgen levels. Pituitary and serum follicle-stimulating hormone (FSH) levels were slightly but consistently elevated (20-30%; p less than 0.05) in antagonist-treated animals, whereas luteinizing hormone (LH) levels (both immunoreactive and bioactive) were unaffected. The relationships of ovarian blood flow (OBF) to ovarian function, body weight, cyclic state, and age of the corpus luteum were studied in aging rats with acutely implanted electromagnetic blood flow probes. Ovarian function was monitored by estimation of serum 17-beta-estradiol (E2) and progesterone (P) levels during (i) different stages of body maturation as indexed by body weight, and (ii) various stages of the estrous and pseudopregnancy (PSP) cycles. A distinct elevation in basal OBF rates was observed to occur in relation to body weight/age. Basal OBF rates and body weight exhibited a near linear correlation (r = 0.985; P less than or equal to 0.001) between 200 and 260 g, with a rise in mean basal OBF rates from 1.1 to 3.2 ml/min occurring in Day-8 PSP rats (Day 0 = ovulation). In weight-regulated (greater than or equal to 250 g) cycle rats, OBF increased in parallel with serum E2 levels during the proestrus-estrus period in association with ovulation, and subsequently declined to basal levels on diestrus. In PSP rats, OBF remained elevated between Days 1 and 8, during the period of peak luteal function. Between Days 8 and 12, both OBF and luteal function declined in a parallel manner as the ovary prepared for the subsequent ovulatory period. These results indicate that during the estrous cycle, OBF rates and serum E2 levels rise in a parallel manner, whereas during PSP, serum P levels and OBF are positively correlated. The parallel decline in OBF and serum P during luteolysis suggest that both parameters are functionally related and may be regulated by either a systemic or intraovarian controlling mechanism.  相似文献

We measured uptake and metabolism of progesterone (P4) during the estrous cycle and Days 2-6 of pseudopregnancy (PSP) to determine uterine P4 dynamics during preimplantation. Rats were infused with [3H]P4 for 60 min, blood was obtained, the uterus was removed, and endometrium and myometrium were isolated. Tissue radiolabeled P4 and P4 metabolites (5 alpha-pregnane-3,20-dione, DHP; 20 alpha-hydroxy P4; 17 alpha-hydroxy P4, and hydroxylated DHP derivatives) were extracted and separated by thin-layer chromatography (TLC). Serum P4 was measured by radioimmunoassay (RIA) in another group of rats. Endometrial and myometrial concentrations of [3H]P4 were greater (p less than 0.05) than plasma values. In contrast, [3H]DHP levels in the endometrium were higher (p less than 0.01) than values in myometrium or plasma. Compared to values in the estrous cycle, endometrial ratios of [3H]DHP/[3H]P4 and [3H]metabolites/[3H]P4 decreased (p less than 0.02) on Days 3-5 of PSP. Serum P4 levels during the estrous cycle (13-25 ng/ml) increased (p less than 0.01) to 120 ng/ml on Days 3-5 of PSP. Estimated concentration of P4 in the endometrium during the estrous cycle (90 ng/g) increased (p less than 0.05) to 580 ng/g by Day 5 of PSP. Similar observations were noted for the estimated endometrial concentrations of DHP and all P4 metabolites. We suggest that both endometrium and myometrium take up and metabolize P4 during the estrous cycle and early PSP. However, endometrial P4 metabolism during PSP is greater than during the estrous cycle, in part because of increased ovarian secretion and endometrial concentration of P4.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Intact and hypophysectomized rats were treated with graded doses of testosterone via subcutaneous Silastic implants over a 13-week period. Serum inhibin concentrations fell 50% (P less than 0.001) after 2 weeks of hypophysectomy, remaining suppressed at this level for 13 weeks. The administration of testosterone to hypophysectomized rats (serum testosterone values 2-12 ng/ml; control values 5.5 ng/ml) was without effect on serum inhibin values. In contrast, administration of testosterone to intact animals for 7 weeks resulted in an initial fall (P less than 0.05) in inhibin levels to 50-70% of controls then increasing to reach control levels at higher doses. Serum FSH concentrations were similarly biphasic with increasing dose of testosterone and values for these two hormones were significantly correlated (r = 0.44, P less than 0.01). Segments of seminiferous tubules in culture from rats after various times of hypophysectomy showed a partly suppressed secretion of inhibin. The administration of testosterone did not modify inhibin production although inhibin production was sensitive to FSH. It is concluded that (1) serum inhibin concentrations are partly suppressed after hypophysectomy and testosterone has no effect on serum inhibin values; and (2) the suppression of serum inhibin in intact rats treated with increasing doses of testosterone is attributable to the concomitant fall in serum FSH concentrations.  相似文献

1. The effect of a low salt (LS, 0.3% NaC1) and control (HS, 1% NaC1) diet on in vitro aldosterone and corticosterone production, the serum corticosterone level, the serum sodium concentration and adrenal and body weight was studied in 30-day-old male rats, some of which were weaned prematurely at the age of 15 days (PW) and some left with the female up to the end of the experiment (NW). 2. Aldosterone production in the control (HS-NW) animals was 1.07+/-0.07 mug/100 mg adrenal/hour (mean +/-S.E.M.), in HS-PW animals 0.6+/-0.07 (P less than 0.01), while in LS-NW and LS-PW animals it rose to 1.59+/-0.1 and 1.81+/-0.14 respectively. The effect of the salt regimen was significant in both the NW group (P less than 0.01) and the PW group (P less than 0.01). Premature weaning did not inhibit aldosterone production in LS-PW animals. 3. Corticosterone production in animals fed on the control diet was 1.81+/-0.16 mug corticosterone/100 mg adrenal/hour in HS-NW rats and 0.91+/-0.09 in the HS-PW group (P less than 0.01). On the low salt diet it fell to 1.4+/-0.11 in LS-NW rats (HW-NW) vs LS-NW: P less than 0.01) and to 0.4+/-0.06 in LS-PW animals (HS-PW vs LS-PW: P less than 0.01). The difference between LS-NW and LS-PW was likewise statistically significant (P less than 0.01). Changes in production were not accompanied by parallel changes in the serum corticosterone level, where an analysis of variance showed no significant difference. 4. The low salt diet reduced the serum sodium concentration in both NW and PW animals (HS-NW 132.9+/-0.86 mEd HS-PW 132+/-0.86, LS-PW 128.5+/-1.16: P less than 0.01). The differences between NW and PW animals were not significant. 5. A low salt intake also reduced the body weight both of animals left with the female (HS-NW vs LS-NW: P less than 0.01) and of prematurely weaned animals (HS-PW vs LS-PW: P less than 0.01). Early weaning significantly affected body weight in LS animals only, the body weight of LS-PW animals being significantly lower than that of LS-NW animals (P less than 0.02). 6. The results show that infant rats are hypersensitive to a low salt intake at the end of the weaning period and that this phenomenon is not mediated by lower reactivity of the zona glomerulosa and of its regulation.  相似文献

The age at vaginal opening, estrous cyclicity, serum levels of luteinizing hormone (LH), follicle stimulating hormone (FSH) and prolactin (PRL) on the day of proestrus, and number of ova and ovarian weight as measured on the day of estrus in spontaneously hypertensive (SH) and genetically matched normotensive Wistar Kyoto (WKY) female rats were compared. In SH rats, there was a significant delay in the vaginal opening, but the regular 4-day estrous cycle followed afterwards. No significant changes were observed in the afternoon increase in serum LH, FSH and PRL on the day of proestrus in SH and WKY rats, although the basal levels of LH and PRL in the morning (11:00 h) were lower in SH rats than in WKY rats. The mean number of ova in SH rats was also less than in WKY rats, whereas the ovarian relative weight was similar in both species of rats. It can be said that SH rats undergo certain, but not critical, endocrine and/or neuroendocrine changes related to reproduction.  相似文献

Studies were carried out in rats to examine the role of calcitonin deficiency in the pathogenesis of ovariectomy-induced osteopenia. The parathyroid glands of 80 female Wistar rats were autotransplanted to their thigh muscle and the animals divided into 4 groups. Group 1 rats were sham ovariectomized, and thyroidectomized to make them calcitonin deficient; Group 2 rats were thyroidectomized, and ovariectomized to make them deficient in ovarian hormones as well; Group 3 rats were sham thyroidectomized and sham ovariectomized, and Group 4 rats were sham thyroidectomized and ovariectomized. A fifth group of rats were unoperated upon and served as controls. Thyroidectomized animals were maintained on thyroxine replacement and 11 months after ovariectomy all the animals were bled, killed and their femurs dissected out. In both the thyroid intact and thyroidectomized animals, ovariectomy decreased femur density significantly (P less than 0.01). Similarly, ovariectomy resulted in a decrease in femur calcium (P less than 0.01) in both groups of animals, and in a significant decrease in serum calcitonin (P less than 0.05) in the thyroid intact animals. We conclude from these findings that ovarian hormone deficiency can cause bone loss independently of lowering circulating calcitonin levels.  相似文献

The impact of maternal starvation during Days 17-20 of gestation was examined in 20-day fetal rat brain tissue cultured for 6 days in MEM and 10% adult rat serum. Acetylcholinesterase (AChE) activities were consistently greater in fetal brain cell cultures from starved mothers. When fetal tissues from starved mothers were continuously exposed to 72-h fasted serum, AChE activities increased from 1.03 +/- 0.14 to 1.59 +/- 0.21 mumol/h/mg protein (P less than 0.001). In fetal tissues from fed mothers, lower AChE activities were increased from 0.78 +/- 0.09 to 1.04 +/- 0.07 mumol/h/mg protein (P less than 0.05) when 72-h fasted serum was used to replace the fed serum during incubation. When fetal brain cell cultures from fed mothers were exposed for 6 days to graded concentrations of fed serum (2.5-15%), the activities of AChE fell reciprocally from 1.34 +/- 0.10 to 0.82 +/- 0.12 mumol/h/mg protein (P less than 0.05). The levels of AChE activity in tissues exposed to fasted serum were consistently greater, but fell similarly from 1.62 +/- 0.10 to 0.97 +/- 14 mumol/h/mg protein (P less than 0.01), when serum concentrations were increased from 2.5 to 15%. AChE activities were 30% higher in tissues incubated with cycloheximide 10(-3) M (P less than 0.02). Unlike AChE, fetal brain enolase activities were unaffected by maternal starvation. In fetal brain cell cultures from fed mothers, enolase fell from 1.85 +/- 0.10 to 1.37 +/- 0.12 mumol/min/mg protein following exposure to fasted instead of fed serum (P less than 0.02). In fetal cultures from starved mothers, enolase activities were depressed similarly from 1.76 +/- 0.08 to 1.41 +/- 0.09 mumol/min/mg protein when fasted replaced fed serum (P less than 0.02). Thus, the fetal brain cell cultures appear to maintain enzymatic realignments imposed by maternal starvation for at least 6 days. In addition, serum from fasted animals has significant growth inhibiting properties manifested by heightened activities of AChE and lower activities of enolase.  相似文献

In Phase I of this study to enhance ovulation rate and hence litter size, gilts received 0 (sham control), 0.625, 1.25, 2.5 or 5.0 mg epostane/kg body weight on Days 10, 11 and 12 of the oestrous cycle (5 gilts/group). After epostane treatment, plasma progesterone concentrations were reduced (P less than 0.01) in a dose-related manner, % progesterone decline = 21.30 x square root of (dose) + 10.45, R2 = 0.70, but recovered to pretreatment levels by 24 h. In Phase II the effects of epostane on ovulation rate and litter size were tested at two study centres. At each centre 108 gilts were treated with the same doses of epostane as used in Phase I and the doses were given for 7 days (Days 15-21) or 12 days (Days 10-21) during the first oestrous cycle. Gilts were inseminated twice during the oestrus after treatment and were slaughtered 30 days later. Mean (+/- s.d.) ovulation rate was 16 +/- 2.7 (N = 8) and 21 +/- 4.0 (N = 61) for control and epostane-treated gilts in Centre A and 12 +/- 2.4 (N = 5) and 17 +/- 3.8 (N = 55) respectively in Centre B (P less than 0.01 for both) and was dose related (ovulation rate = 3.38 x square root of (dose) + 16.17, R2 = 0.31). The effects of 7- or 12-day epostane treatment on ovulation rate were not different (P greater than 0.05), indicating that effects of treatment after Day 14 of the oestrous cycle are most important to subsequent ovulation frequency.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
