The extent of induction of aldrin epoxidation and glutathione S-transferase (GST) in the bulb mite Rhizoglyphus robini was dependent on the ingested plant material or on the type of inducer. GST was strongly induced in mites fed on garlic or exposed to trans-stilbene oxide as well as by the pesticides fenpropathrin and propoxur. Several phytochemicals such as 3-indoleacetonitrile, flavone, chalcone and pulegone were potent GST inducers stimulating the enzyme activity by 2–4-fold. Other toxicants, pesticides and allelochemicals yielded moderate to slight stimulation and a few were even inhibitory. In general, aldrin epoxidation was moderately stimulated by plant materials and xenobiotics. The epoxidation activity was increased in mites fed on garlic and carrot, or exposed to toxicants such as pentobarbital and -hexachlorocyclohexane, pesticides like chlordimeform, propoxur, carbaryl and lindane, as well as allelochemicals such as flavone, ß-carotene, indole-3-carbinol and safrole. The relevance of the study vis-à-vis adaptation of herbivorous arthropods to their habitats and acquirement of tolerance to chemical control agents, is discussed.  相似文献

For lindane degradation, a cell suspension of Clostridium rectum strain S-17 demands the addition of substrates such as leucine, alanine, pyruvate, a leucine-proline mixture, and molecular hydrogen. In the presence of leucine-proline mixture, lindane decomposed in parallel with isovaleric acid formation, and both lindane degradation and isovaleric acid formation were inhibited by monoiodoacetic acid, suggesting a close relation between lindane degradation and the Stickland reaction. Lindane was degraded by cell-free extracts of C. rectum in the presence of dithiothreitol (DTT). Radiogaschromatograms of n-hexane soluble metabolites from [14C] lindane showed the presence of monochlorobenzene and -3,4,5,6-tetrachlorocyclohexene, Leucine, NADH, and NADPH were somewhat less active than DTT for lindane degradation in cell-free extracts. Summary Clams and gastropods survived 96-hour immersion in concentrations up to 25 mg/l of four organophosphorus insecticides (DDVP, dioxathion, methyl parathion, Phosdrin(R)) or 10 mg/l of four organochloride compounds (p,p-DDT, lindane, endrin, dieldrin) at 24 salinity and 20°C. During a post-treatment observation period of 33 days for gastropods and 133 days for clams, the following was noted: a reduction in number of egg cases deposited by gastropods initially subjected to comparatively high levels of endrin, dieldrin, and lindane; an apparent stimulatory effect on gastropod fecundity by relatively low concentrations of p,p-DDT, endrin, dieldrin, and lindane; and a decrease in survival of clams and gastropods initially subjected to high levels of DDVP or methyl parathion.
Résumé Des groupes différents de mollusques bivalve, Mercenaria mercenaria, ou gastéropode, Nassa obsoleta, ont survécu des traitements de 25 mg/l d'insecticides organophosphoriques (DDVP, dioxathion, méthyl parathion, Phosdrin(R)) ou de 10 mg/l de chlorures d'hydrocarbures (p,p-DDT, lindane, endrine, dieldrine). Ces traitements de 96 heures pour chaque groupe exposé à une concentration d'insecticide furent accomplis à une salinité de 24 et à une température de 20°C. Après ce treaement initial les gasteropodes furent placés dans de nouveaux aquariums contenant de l'eau non-contaminée et gardés sous observation pendant 33 jours. Les bivalves, traités de façon identique, furent gardés pendant une periode de 133 jours. La période d'observation révéla les différences suivantes entre les animaux traités et les témoins : a/ réduction du nombre des plaques de ponte pour le gastéropode soumis à de fortes doses d'endrine, de dieldrine, et de lindane; b/ stimulation apparent de la fécundité comme le laisse supposer l'augmentation du nombre des plaques de pontes pour les gastéropodes traités avec de faibles concentrations de p,p-DDT, d'endrine, de dieldrine, et de lindane; c/ une augmentation du taux de mortalité chez les bivalves et les gastéropodes soumis à de fortes doses de DDVP ou de méthyl parathion.

Several insecticides have been used as protectants against lettuce root aphid, including, in 1955, lindane, schradan, aldrin and endrin; the last-named giving the most promising results. With a dressing of an equal weight of 50% endrin dust added to lettuce seed, 86% reduction in aphid numbers was obtained. In 1957, a seed dressing with endrin also gave a good control, which was superior to that obtained with thimet 44D. However, in 1956 and 1958 endrin seed dressing failed to control the aphid. When a seed dressing with endrin was supplemented by an endrin spray applied to the seedlings a consistently good control resulted.
A high level of aphid control was obtained in 1958 with two sprays of diazinon applied along the lettuce rows, the first applied after singling and the other at the early rosette stage. Diazinon or endrin, applied to the soil at 4 lb. actual insecticide/acre and rotavated into the top 3 in. before sowing, also gave high levels of control.
No recommendations for the use of endrin on lettuce can be made until levels and persistence of residues are known.  相似文献

Three lindane (-1,2,3,4,5,6-hexachlorocyclohexane) treated soils were studied under laboratory conditions to determine the interaction between lindane and the soil microorganisms. Microbial populations and respiration were monitored to study insecticide effects. Formation of lindane degradation products and chloride content were examined to determine effects of the microorganisms. Some populations in lindane treated soils showed temporary declines but all ultimately recovered to at least the level of the controls in 16 weeks. Respiration was stimulated over a 9-week period especially in the sandy and clay loams, suggesting the possibility of microbial degradation of the insecticide. Lindane degradation products separated and identified by TLC included -2,3,4,5,6-pentachloro-1-cyclohexene (-PCCH), -3,4,5,6-tetrachloro-1-cyclohexene (-TCCH), -3,4,5,6-tetrachloro-1-cyclohexene (-TCCH), and pentachlorobenzene (PCB). Summary Conditions are described for the RNA-directed cell-free synthesis of the three galactose enzymes of Escherichia coli. Together with the DNA-directed synthesis described previously, this system permits the measurement of the three gene-products encoded by the gal operon as active enzymes synthesized in vitro in response to either gal-DNA or gal-RNA. The yield of enzyme is proportional to the amount of RNA added. Thus, the RNA-directed enzyme synthesis can serve as an assay system for functional mRNA. This test has been employed to determine the kinetics of synthesis and degradation of functional gal mRNA under the conditions of cell-free enzyme synthesis. The functional half-life of gal mRNA in this system is 6–7 minutes and is higher than expected from in vivo measurements.In contrast to the DNA-directed cell-free synthesis, the RNA-directed synthesis of the galactose enzymes is neither stimulated by cyclic adenosine-3:5-monophosphate nor by inducer.  相似文献

Properties of -glucan synthetase from S. cerevisiae were studied. The enzyme exhibited optimal activity at pH 6.7 and 24 C. Km for UDP-glucose was 0.12mm. Addition of Mg++ or Mn++ stimulated its activity by 60% and 21% respectively. High concentrations of EDTA and hydroxyquinoline were inhibitory. Glucan synthetase was fully active in cell-free extracts. Small concentrations of trypsin or subtilopeptidase A from Bacillus subtilis, caused only a slight increase in glucosyl transferase activity, but larger concentrations destroyed -glucan synthetase. Acid proteases were neither stimulatory nordestructive. Thus it seemsunlikelythat -glucan synthetase exists in a zymogen form. Glucan synthetase was unstable. It was inactivated more rapidly at 28 C than at 0 C. The presence of substrate, -glucan or the protease inhibitors PMSF, Antipain or Pepstatin A did not protect -glucan synthetase from inactivation. Glucan synthetase was not stimulated by addition of cellobiose or -glucans. The synthesis of -glucans was competitively inhibited by UDP (Ki=0.45mm). DOPA synthesis from phenylalanine was studied in PC12 cells incubated with m-hydroxybenzylhydrazine, to inhibit aromatic L-amino acid decarboxylase. DOPA synthesis rose with increasing concentrations of either phenylalanine or tyrosine; maximal rates (~55 pmol/min/mg protein for tyrosine; ~40 pmol/min/mg protein for phenylalanine) occurred at a medium concentration of ~10 M for either amino acid. The Km for either amino acid was about 1 M (medium concentration). At tyrosine concentrations above 30 M, DOPA synthesis declined; inhibition was observed at higher concentrations for phenylalanine (300 M). These effects were most notable in the presence of 56 mM potassium. Measurements of intracellular phenylalanine and tyrosine suggested the Km for either amino acid is 20–30 M; maximal synthesis occurred at 120–140 M. In the presence of both phenylalanine and tyrosine, DOPA synthesis was inhibited by phenylalanine only at a high medium concentration (1000 M), regardless of medium tyrosine concentration. The inhibition of DOPA synthesis by high medium tyrosine concentrations was antagonized by high medium phenylalanine concentrations (100, 1000 M). Together, the findings indicate that for PC12 cells, phenylalanine can be a significant substrate for tyrosine hydroxylase, is a relatively weak inhibitor of the enzyme, and at high concentrations can antagonize substrate inhibition by tyrosine.  相似文献

The influence of lindane upon phosphatidylinositol hydrolysis in rat brain cortex slices has been investigated using anion-exchange chromatography in order to separate the water-soluble inositol metabolites. Acetylcholine, noradrenaline, and lindane induce the accumulation of myo-[2-3H]inositol as the water-soluble inositol metabolites. However, the cholinergic muscarinic antagonist atropine inhibited the stimulatory response of carbachol, but practically unmodified the effect that lindane has on inositol phosphate production. Also, prazosin anti-1 adrenoreceptors blocked noradrenaline-induced phosphoinositide hydrolysis, but had no effect on lindane-induced increase of inositol phosphate levels. The results suggest that lindane does not exert a general effect on the receptor-stimulated formation of inositol phosphates by both muscarinic and 1-adrenergic agonists.  相似文献

Summary The inductive effect of lactose, -methyl-thio-D-galactopyranoside, (TMG) and glucose on galactosidase synthesis in Kluyveromyces lactis has been studied. Whereas TMG gave a five fold stimulation of the rate of -galactosidase synthesis, lactose only gave a small stimulation. Glucose caused represssion at levels above 10-3M but stimulated -galactosidase synthesis when added at lower concentrations.  相似文献

The effects of a cyclodiene (endrin) and a cyclohexane (lindane) insecticide have been tested on gamma-aminobutyric acid (GABA) receptors in the central nervous system of the cockroach (Periplaneta americana), by using electrophysiological methods and an in vitro functional receptor assay. In electrophysiological experiments on an identified motor neuron (Df), endrin blocked the GABA response with a 50% inhibition concentration of 5.0 x 10(-7) M in a non-competitive manner. The actions of endrin were irreversible under the experimental conditions adopted. Increasing the intracellular chloride concentration reduced the effectiveness of endrin, whereas a change in the potassium concentration failed to influence the block by endrin of GABA responses. Lindane exhibited similar actions to endrin on insect GABA receptors, but was approximately an order of magnitude less effective. In a microsac preparation from cockroach nerve cords, endrin, at a concentration of 1.0 x 10(-5) M, completely blocked GABA-stimulated 36Cl- uptake, whereas the same concentration of lindane was less potent, only blocking about 40% of uptake under similar conditions. Neither insecticide had any effect on L-glutamate-activated chloride channels. The results demonstrate that endrin and lindane block functional insect neuronal GABA receptors.  相似文献

Sediment from 9 lakes and lagoons in 3 North African countries was analysed for a suite of organochlorine insecticides and polychlorinated biphenyls (PCBs). Residues of -HCH and pp-DDE were detected in most cores, and radiometric dating of the sediment enabled time profiles to be constructed which are indicative of recent use of the insecticide lindane and previous use of the insecticide DDT in the region. Absolute concentrations of these pesticides were relatively low compared to reported values for other world-wide locations, but exceeded recommended Canadian and Dutch environmental quality standards at several sites. Maximum fluxes of -HCH and pp-DDE (190 and 95 g m–2 yr–1, respectively) were relatively high and comparable to some sediments in North America and the United Kingdom. Other organochlorine pesticides including dieldrin were detected at low levels in some samples. Selected PCB congeners were detected at trace levels at 2 sites only, indicating low levels of industrial contamination at the sampling locations. Tropical and sub-tropical regions of the world have been suggested to be contemporary sources of globally distributed organochlorine contaminants. These data are discussed with respect to this hypothesis.  相似文献

Transitions in the growth limiting factor from light (I) to nitrogen (N) and vice versa caused changes in geosmin production, protein and carbohydrate content, and the synthesis of pigments such as chlorophyll a (Chl a), phycobiliproteins (PBPs), and -carotene of the cyanobacterium Oscillatoria brevis. Following IN transition the first 150h, the decrease in protein content was compensated for by an increase of carbohydrates, and thereby, a constant biomass level was maintained in this period. Thereafter, biimass dropped to 15% of its initial level. A decrease in geosmin and pigment content was observed during transition from IN-limited growth. However, geosmin increased relative to phytol (Chl a) and -carotene which may indicate that a lowered demand for phytol and -carotene during N-limited growth allows isoprenoid precursors to be directed to geosmin rather than to pigment synthesis. Synthesis of Chl a and -carotene at the expense of geosmin was suggested for the observed start of increase in geosmin production only at the time that Chl a and -carotene had reached their I-limited steady state. Transition from nitrogen to light limited growth caused an acceleration of metabolism shown by a rapid decrease in carbohydrate content accompanied by an increase in protein content. The growth rate of the organisms temporarily exceeded the dilution rate of the culture and the biomass level increased 6-fold. Summary Rat parenchymal hepatocytes isolated with collagenase were cultured as monolayers in Williams medium E supplemented with calf serum. Freshly isolated cells showed very low activities of various liver functions, and they had to be cultured for 6-24 h to allow recovery of these functions. Insulin and dexamethasone greatly increased cell viability in primary culture. After culture for 24 h, these cells showed various liver functions as seen in vivo and responded well to various added hormones and amino acids. The concentrations of amino acids in the medium regulated synthesis of serum proteins and insulin stimulated lipogenesis, which in turn regulated synthesis of lipoproteins. Insulin also stimulated glycogen synthesis and the stimulation was parallel with the number of insulin receptors. Glucagon stimulated glycogenolysis and its stimulation involved the function of the cytoskeleton. Glucagon and dexamethasone induced various enzymes of amino acid catabolism, such as tryptophan oxygenase, tyrosine aminotransferase and serine dehydratase. These inductions were inhibited by insulin or catecholamine. The effect of catecholamine was due to its -adrenergic action. The -action of isoproterenol was low in freshly isolated cells, but increased during culture of the cells. Acquirement of hormonal responses during neonatal development can be studied in this culture system. Mature hepatocytes in culture are usually quiescent, but when insulin and epidermal growth factor were added, DNA synthesis by the cells increased markedly and they showed density-dependent growth. In this culture system, serum could be omitted for 2 days when the dishes were coated with fibronectin without appreciable change of functions, but serum was needed for longer culture of the cells. A factor that increased cell survival was found in serum and in pituitary gland.These results show that hepatocytes in primary culture are a simple and useful system for studies of liver functions in vitro and related works were also reviewed.  相似文献

Summary Juvenile hormone (JH) treatment coordinately stimulated the dose-dependent synthesis of vitellogenin and endoplasmic reticulum (ER) membrane phospholipids in fat body cells from allatectomized adult females ofLeucophaea maderae. Animals were pulse-labeled in vivo with [32P] to simultaneously measure the rates of synthesis of the phosphorylated subunits of vitellogenin and the structural phospholipids of the ER membranes. Phospholipid synthesis in ER membranes from nontarget tissues for JH such as thoracic muscle, midgut, and larval fat body was unresponsive to hormone treatment. The proliferation of ER in response to JH treatment was thus restricted to tissue that was competent to synthesize vitellogenin.Primary and secondary vitellogenin induction was measured in allatectomized adult females treated 12 days apart with JH-III. The time-course of the primary response for vitellogenin and ER phospholipid synthesis was characterized by a 24 h latent period, a rapid increase to a maximum at 72 h, and then a gradual decline. During secondary induction, vitellogenin accumulated in the hemolymph nearly twice as fast as before and peaked at a concentration of 38 g/l. This vitellogenin titer was approximately two-fold higher than that found at the height of the primary response. During both primary and secondary stimulation with JH, ER phospholipid synthesis, as measured by [14C]choline incorporation into microsomal phosphatidylcholine, was stimulated five-fold over the untreated control animals. The amplified production of vitellogenin during the secondary response was associated with a 24 h-earlier peak of ER phospholipid synthesis in the fat body.  相似文献

Siderophore synthesis of Pseudomonas putida F1 was found to be regulated by quorum sensing since normalized siderophore production (per cell) increased 4.2-fold with cell density after the cells entered middle exponential phase; similarly, normalized siderophore concentrations in Pseudomonas aeruginosa JB2 increased 28-fold, and a 5.5-fold increase was seen for P. aeruginosa PAO1. Further evidence of the link between quorum sensing and siderophore synthesis of P. putida F1 was that the quorum-sensing-disrupter (5Z)-4-bromo-5-(bromomethylene)-3-butyl-2(5H)-furanone (furanone) from the marine red alga Delisea pulchra was found to inhibit the formation of the siderophore produced by P. putida F1 in a concentration-dependent manner, with 57% siderophore synthesis repressed by 100 g/ml furanone. In contrast, this furanone did not affect the siderophore synthesis of Burkholderia cepacia G4 at 20–40 g/ml, and stimulated siderophore synthesis of P. aeruginosa JB2 2.5- to 3.7-fold at 20–100 g/ml. Similarly, 100 g/ml furanone stimulated siderophore synthesis in P. aeruginosa PAO1 about 3.5-fold. The furanone appears to interact with the quorum-sensing machinery of P. aeruginosa PAO1 since it stimulates less siderophore synthesis in the P. aeruginosa qscR quorum-sensing mutant (QscR is a negative regulator of LasI, an acylated homoserine lactone synthase).  相似文献

Paramecium aurelia exconjugants contain new macronuclear anlagen and numerous fragments of the old pre-zygotic macronucleus. Macronuclear anlagen develop during the first two cell cycles after conjugation. During this time their volume increases from about 11 m3 to about 3700 m3 and more than 10 doublings of DNA content occur. The rate of DNA synthesis is between two and three times as great as in the vegetative macronucleus. — In macronuclear fragments, however, DNA synthesis is suppressed. The rate of DNA synthesis in macronuclear fragments during the extended first cell cycle after conjugation (11 1/2 hr. vs. 5 1/2 hr. for the vegetative cell cycle) is only about one-third of the rate in vegetative macronuclei and there is only a 65% increase in the mean DNA content of fragments. The rate of fragment DNA synthesis continues to decrease during each of the subsequent two cell cycles. — Unlike the rate of DNA synthesis, the rate of RNA synthesis per unit of DNA is similar in macronuclear anlagen, macronuclear fragments and fully developed macronuclei. Macronuclear fragments continue to synthesize RNA at the normal rate long after the new macronuclei are fully developed. Fragments contribute about 80% of all RNA synthesized during the first two cell cycles after conjugation. RNA synthesis begins very early in the development of macronuclear anlagen and nucleolar material appears during the first half-hour of anlage development. — Chromosome-like structures were never observed during anlage development and there was no evidence of two periods of DNA synthesis separated by a DNA poor stage as has been observed in several hypotrichous Ciliates.  相似文献

Summary Escherichia coli was depleted of ribosomes by a thermal shock at 47° C which quantitatively destroyed the 30S ribosomal subunits. During recovery in minimal medium at 30° C RNA is synthesized while protein synthesis resumes only after about 90 min. It is shown that lac mRNA is synthesized in the complete absence of ribosomal activity and hence RNA synthesis is not coupled to protein synthesis. Lac mRNA from a series of lac nonsense mutants was examined in both heated and untreated cells. It was found that the polar effect of nonsense mutation is relieved in the absence of ribosomes and that this relief is due to the synthesis of larger mRNA molecules. The intracellular concentration of cAMP in the green alga Chlorella fusca was in the range of 2 · 10-9 to 10-8 moles/g dry weight and was strongly dependent on the growth conditions. The cAMP level was high with high light intensity, low nitrate or glucose concentration. Intracellular cAMP increased only by factor of 2 when high amounts (up to 10-3 M) of cAMP were added to the medium. Most of the given cAMP was converted to 5-AMP.Addition of cAMP had little effect on the chlorophyll content of the cells, only at 10-6 M some enhancement in photoautotrophic cultures was observed. On the other hand high amounts of cAMP in the medium increased the growth rate. DBcAMP* showed a positive effect on chlorophyll synthesis and growth rate at much lower concentrations compared to cAMP.Stimulation effects of exogenous cAMP on the synthesis of chlorophyll were also observed in mixotrophic cultures with a high glucose/nitrate ratio, conditions where chlorophyll synthesis is repressed.
