Spores of Bacillus cereus were heated and recovered in order to investigate the effect of water activity of media on the estimated heat resistance (i.e., the D value) of spores. The water activity (ranging from 0.9 to 1) of the heating medium was first successively controlled with three solutes (glycerol, glucose, and sucrose), while the water activity of the recovery medium was kept near 1. Reciprocally, the water activity of the heating medium was then kept at 1, while the water activity of the recovery medium was controlled from 0.9 to 1 with the same depressors. Lastly, in a third set of experiments, the heating medium and the recovery medium were adjusted to the same activity. As expected, added depressors caused an increase of the heat resistance of spores with a greater efficiency of sucrose with respect to glycerol and glucose. In contrast, when solutes were added to the recovery medium, under an optimal water activity close to 0.98, a decrease of water activity caused a decrease in the estimated D values. This effect was more pronounced when sucrose was used as a depressor instead of glycerol or glucose. When the heating and the recovery media were adjusted to the same water activity, a balancing effect was observed between the protective influence of the solutes during heat treatment and their negative effect during the recovery of injured cells, so that the overall effect of water activity was reduced, with an optimal value near 0.96. The difference between the efficiency of depressors was also less pronounced. It may then be concluded that the overall protective effect of a decrease in water activity is generally overestimated.  相似文献

The heat resistance to hot air of spores of Bacillus cereus (ATCC 14579) attached to carriers of stainless steel or silicone rubber was investigated in a range from 1% to 100% relative humidity (RH). Apart from an initial stage, linear survivor curves were obtained for all relative humidities. Neither the attachment itself nor the material of the carrier had an influence on the resistance. A distinct maximum of heat resistance was found at 40% RH. At 122°C the rate constants at 40% RH were five orders of magnitude smaller than at 100% RH and two orders of magnitude smaller than at 1% RH. At relative humidities of more than 40% the rate constants were strongly temperature dependent, whereas at lower relative humidities they were less temperature dependent. No significant influence of the relative humidity on the Arrhenius activation energy was found within each humidity range. The mean values were 295 kJ mol-1 for relative humidities of 60% to 100% RH and 165 kJ mol-1 for 1% to 20% RH. The occurrence of a maximum is ascribed to the existence of two inactivation mechanisms, the first is retarded and the second is accelerated by a reduction of relative humidity. It is assumed that the first mechanism is a protein denaturation. The second mechanism may be an oxidative process.  相似文献

Spores of Bacillus subtilis SA22 harvested after 22 d incubation on nutrient agar at 30°C were more resistant to 0–04% peracetic acid at 20°C than spores harvested following 2 d incubation. Similarly, spores of B. subtilis globigii B17, harvested after 7 d incubation on a sporulation agar were up to 10 times less resistant to 0.04% peracetic acid at 20°C than spores harvested after 35 d incubation. An increase in resistance to heating at 100°C and to exposure to 17.7% hydrogen peroxide at 20°C occurred as the age of B. subtilis SA22 spores prior to harvesting increased, whereas differences in resistance were not observed with spores of B. subtilis globigii B17.  相似文献

The relative stability of spores of Bacillus cereus grown at three different temperatures was examined by using quasi-elastic light scattering (q.l.s.) in conjunction with turbidity and scanning electron microscopy (s.e.m.). Cultures grown at 20, 30 and 40 degrees C (BC20, BC30 and BC40 respectively) were compared in terms of (i) their effective hydrodynamic radius, rH, as determined from q.l.s. and (ii) their gross morphology, as determined from s.e.m. The effects of autoclaving at 121.1 degrees C on both these properties was also examined. We observed (1) that cultures BC20 and BC30 appeared to have similar values for rH, whereas that of BC40 appeared some 50% higher, and (2) BC40 had a correspondingly much lower heat resistance (its structural integrity was lost after about 20 min autoclaving, whereas that of BC20 and BC30 was retained even after 80 min autoclaving). These data were in good agreement with independent measurements of heat-resistance coefficients. Changes in the hydrodynamic radius, polydispersity (both using q.l.s.) and turbidity were monitored with time on addition of the disinfectants sodium hypochlorite and peracetic acid; again BC40 appeared to have a lower resistance.  相似文献

Microgermination of Bacillus cereus spores   总被引:5,自引:3,他引:2  
The biphasic nature of germination curves of individual Bacillus cereus T spores was further characterized by assessing the effects of temperature, concentration of germinants, and some inorganic cations on microgermination. Temperature was shown to affect both phases of microgermination as well as the microlag period, whereas the concentration of l-alanine and supplementation with adenosine exerted a significant effect only on the microlag period. The germination curves of individual spores induced by inosine were also biphasic and resembled those of spores induced by l-alanine. High concentrations (0.1 m or higher) of calcium and other inorganic cations prolonged both phases of microgermination, particularly the second phase, and had a less pronounced effect on the microlag period. The second phase of microgermination was completely inhibited when spores were germinated either in the presence of 0.3 m CaCl(2) or at a temperature of 43 C; this inhibition was reversible. Observations on the germination of spore suspensions (kinetics of the release of dipicolinic acid and mucopeptides, loss of heat resistance, increase in stainability, decrease in turbidity and refractility) were interpreted on the basis of the biphasic nature of microgermination. Dye uptake by individual spores during germination appeared also to be a biphasic process.  相似文献

Bacillus subtilis(B. subtilis) cells were placed in various environmental conditions to study the effects of aeration, water activity of the medium, temperature, pH, and calcium content on spore formation and the resulting properties. Modification of the sporulation conditions lengthened the growth period of B. subtilis and its sporulation. In some cases, it reduced the final spore concentration. The sporulation conditions significantly affected the spore properties, including germination capacity and resistance to heat treatment in water (30 min at 97°C) or to high pressure (60 min at 350 MPa and 40°C). The relationship between the modifications of these spore properties and the change in the spore structure induced by different sporulation conditions is also considered. According to this study, sporulation conditions must be carefully taken into account during settling sterilization processes applied in the food industry.  相似文献

Aims: To determine the mechanism of wet heat killing of spores of Bacillus cereus and Bacillus megaterium. Methods and Results: Bacillus cereus and B. megaterium spores wet heat‐killed 82–99% gave two bands on equilibrium density gradient centrifugation. The lighter band was absent from spores that were not heat‐treated and increased in intensity upon increased heating times. These spores lacked dipicolinic acid (DPA) were not viable, germinated minimally and had much denatured protein. The spores in the denser band had viabilities as low as 2% of starting spores but retained normal DPA levels and most germinated, albeit slowly. However, these largely dead spores outgrew poorly if at all and synthesized little or no ATP following germination. Conclusions: Wet heat treatment appears to kill spores of B. cereus and B. megaterium by denaturing one or more key proteins, as has been suggested for wet heat killing of Bacillus subtilis spores. Significance and Impact of the Study: This work provides further information on the mechanisms of killing of spores of Bacillus species by wet heat, the most common method for spore inactivation.  相似文献

Heat activation of bacterial spores at low pH was investigated in detail. Unlike activation of spores in distilled water at a neutral pH, activation at low pH involves two superimposed processes: enhanced activation and death. Low-pH-activated spores failed to germinate in d-alanine, in contrast to spores activated at neutral pH, owing to the abolition of alanine-racemase activity. Morphological and permeability changes such as release and partial disruption of spores were dipicolinic acid-observed during low-pH activation. The kinetics pattern of low-pH activation, as well as the change in properties of the spores thereafter, suggest that the mechanism of low-pH activation differs from that of other kinds of heat-activation.  相似文献

Aims: To compare physical properties of spores that were produced in broth sporulation media at greater than 108 spores ml−1. Methods and Results: Bacillus atrophaeus reproducibly sporulated in nutrient broth (NB) and sporulation salts. Microscopy measurements showed that the spores were 0·68 ± 0·11 μm wide and 1·21 ± 0·18 μm long. Coulter Multisizer (CM3) measurements revealed the spore volumes and volume-equivalent spherical diameters, which were 0·48 ± 0·38 μm3 and 0·97 ± 0·07 μm, respectively. Bacillus cereus reproducibly sporulated in NB, sporulation salts, 200 mmol l−1 glutamate and antifoam. Spores were 0·95 ± 0·11 μm wide and 1·31 ± 0·17 μm long. Spore volumes were 0·78 ± 0·61 μm3 and volume-equivalent spherical diameters were 1·14 ± 0·11 μm. Bacillus atrophaeus spores were hydrophilic and B. cereus spores were hydrophobic. However, spore hydrophobicity was significantly altered after treatment with pH-adjusted bleach. Conclusions: The utility of a CM3 for both quantifying Bacillus spores and measuring spore sizes was demonstrated, although the volume between spore exosporium and spore coat was not measured. This study showed fundamental differences between spores from a Bacillus subtilis- and B. cereus-group species. Significance and Impact of the Study: This is useful for developing standard methods for broth spore production and physical characterization of both living and decontaminated spores.  相似文献

The effect of ultrasonic treatment on the heat resistance of Bacillus spores differs widely both with the species and the strain tested. Ultrasonic waves do not affect the heat resistance of some strains, whereas they greatly reduce that of other strains of the same species. The heat-sensitizing effect of ultrasonic waves is more pronounced when short heating periods are used. Ultrasonic treatment does not seem to affect the "Z value".  相似文献
