Water movement to and from a root depends on the soil hydraulicconductivity coefficient (Lsoil), the distance across any root-soilair gap, and the hydraulic conductivity coefficient of the root(LP). After analytical equations for the effective conductanceof each part of the pathway are developed, the influences ofsoil drying on the soil water potential and Lsoil are describedduring a 30 d period for a loamy sand in the field. The influencesof soil drying on LP for three desert succulents, Agave deserti,Ferocactus acanthodes, and Opuntia ficus-indica, are also describedfor a 30 d period. To quantify the effects of soil drying onthe development of a root-soil air gap, diameters of 6-week-oldroots of the three species were determined at constant watervapour potentials of –1.0 MPa and –10 MPa as wellas with the water vapour potential decreasing at the same rateas soil drying during a 30 d period. The shrinkage observedfor roots initially 2·0 mm in diameter averaged 19% duringthe 30d period. The predominant limiting factor for water movementwas LP of the root for the first 7 d of soil drying, the root-soilair gap for the next 13 d, and Lsoil thereafter. Compared withthe ease of water uptake from a wet soil, the decrease in conductancesduring soil drying, especially the decrease in Lsoil causedthe overall conductance to decrease by 3 x 103-fold during the30 d period for the three species considered, so relativelylittle water was lost to the dry soil. Such rectifier-like behaviourof water movement in the soil-root system resulted primarilyfrom changes in Lsoil and, presumably, is a general phenomenonamong plants, preventing water loss during drought but facilitatingwater uptake after rainfall. Pacemaker potentials were recorded in situ from myenteric interstitial cells of Cajal (ICC-MY) in the murine small intestine. The nature of the two components of pacemaker potentials (upstroke and plateau) were investigated and compared with slow waves recorded from circular muscle cells. Pacemaker potentials and slow waves were not blocked by nifedipine (3 µM). In the presence of nifedipine, mibefradil, a voltage-dependent Ca2+ channel blocker, reduced the amplitude, frequency, and rate of rise of upstroke depolarization (dV/dtmax) of pacemaker potentials and slow waves in a dose-dependent manner (1–30 µM). Mibefradil (30 µM) changed the pattern of pacemaker potentials from rapidly rising, high-frequency events to slowly depolarizing, low-frequency events with considerable membrane noise (unitary potentials) between pacemaker potentials. Caffeine (3 mM) abolished pacemaker potentials in the presence of mibefradil. Pinacidil (10 µM), an ATP-sensitive K+ channel opener, hyperpolarized ICC-MY and increased the amplitude and dV/dtmax without affecting frequency. Pinacidil hyperpolarized smooth muscle cells and attenuated the amplitude and dV/dtmax of slow waves without affecting frequency. The effects of pinacidil were blocked by glibenclamide (10 µM). These data suggest that slow waves are electrotonic potentials driven by pacemaker potentials. The upstroke component of pacemaker potentials is due to activation of dihydropyridine-resistant Ca2+ channels, and this depolarization entrains pacemaker activity to create the plateau potential. The plateau potential may be due to summation of unitary potentials generated by individual or small groups of pacemaker units in ICC-MY. Phosphorus and nitrogen uptake capacities were assessed during36–58 d drying cycles to determine whether the abilityof sagebrush (Artemisia tridentata Nutt.) to absorb these nutrientschanged as the roots were subjected to increasing levels ofwater stress. Water was withheld from mature plants in large(6 I) containers and the uptake capacity of excised roots insolution was determined as soil water potentials decreased from–0.03 MPa to –5.0 MPa. Phosphorus uptake rates of excised roots at given substrateconcentrations increased as preharvest soil water potentialsdecreased to –5.0 MPa. Vmax and Km also increased as soilwater potentials declined. Declining soil water potentials depressednitrogen uptake at set substrate concentrations, but uptakecapacity, calculated as the sum Vmax for both NH+4+NO3,did not change significantly with drying. The sum Vmax correlatedwith root nitrogen concentration. Root uptake capacity for nitrogen and phosphorus was extremelystable under severe water stress in this aridland shrub. Maintenanceof uptake capacity, coupled with a previously demonstrated abilityto conduct hydraulic lift, may enable A. tridentata better tomaintain nitrogen and phosphorus uptake as soil water availabilitydeclines. These mechanisms may be important in the ability ofA. tridentata to maintain growth, complete reproduction, andgain an advantage against competitors late in the season whenthe soil layers with higher nutrient availability are dry. A thermocouple psychrometer method, previously described foruse in determining tissue and sap water potentials, has beenadapted for determining matric potentials. Matric values ranging from approximately zero to –10 x105 Pa were observed with wilting wheat leaves. Mean valuesof total tissue water potential and of its solute (osmotic),matric, and cell wall pressure components were –16.6,–18.3, –30 and +4.7 Pa x 105, respectively. Matric potentials are often ignored in investigations of plantwater relations. This practice is shown to result in underestimationof wall pressures (in this case by a mean of 64 per cent) andsometimes in spuriously negative values.  相似文献

We examined 1) whether the effects of lowered trans-sarcolemmal Na+ gradient on force differed between nonfatigued fast- and slow-twitch muscles of mice and 2) whether effects on action potentials could explain the decrease of force. The Na+ gradient was reduced by lowering the extracellular [Na+] ([Na+]o). The peak force-[Na+]o relationships for the twitch and tetanus were the same in nonfatigued extensor digitorum longus and soleus muscles: force was maintained over a large range of [Na+]o and then decreased abruptly over a much smaller range. However, fatigue was significantly exacerbated at a lowered [Na+]o that had little effect in nonfatigued soleus muscle. This finding suggests that substantial differences exist in the Na+ effect on force between nonfatigued and fatigued muscle. The reduced contractility in nonfatigued muscles at lowered [Na+]o was largely due to 1) an increased number of inexcitable fibers and threshold for action potentials, 2) a reduction of action potential amplitude, and 3) a reduced capacity to generate action potentials throughout trains. sodium gradient; muscle contraction; action potential train; extensor digitorum longus; soleus  相似文献

Myrothamnus flabellifoliusWelw. is a desiccation-tolerant (‘resurrection’)plant with a woody stem. Xylem vessels are narrow (14 µmmean diameter) and perforation plates are reticulate. This leadsto specific and leaf specific hydraulic conductivities thatare amongst the lowest recorded for angiosperms (ks0.87 kg m-1MPa-1s-1;kl3.28x10-5kg m-1MPa-1s-1, stem diameter 3 mm). Hydraulic conductivitiesdecrease with increasing pressure gradient. Transpiration ratesin well watered plants were moderate to low, generating xylemwater potentials of -1 to -2 MPa. Acoustic emissions indicatedextensive cavitation events that were initiated at xylem waterpotentials of -2 to -3 MPa. The desiccation-tolerant natureof the tissue permits this species to survive this interruptionof the water supply. On rewatering the roots pressures thatwere developed were low (2.4 kPa). However capillary forceswere demonstrated to be adequate to account for the refillingof xylem vessels and re-establishment of hydraulic continuityeven when water was under a tension of -8 kPa. During dehydrationand rehydration cycles stems showed considerable shrinking andswelling. Unusual knob-like structures of unknown chemical compositionwere observed on the outer surface of xylem vessels. Relationships between nitrate (NO-3) supply, uptake and assimilation,water uptake and the rate of mobilization of seed reserves wereexamined for the five main temperate cereals prior to emergencefrom the substrate. For all species, 21 d after sowing (DAS),residual seed dry weight (d.wt) decreased while shoot plus rootd.wt increased (15–30%) with increased applied NO-3concentrationfrom 0 to 5–20 mM . Nitrogen (N) uptake and assimilationwere as great with addition of 5 mM ammonium (NH+4) or 5 mMNO-3but NH+4did not affect the rate of mobilization of seedreserves. Chloride (Cl-) was similar to NO-3in its effect onmobilization of seed reserves of barley (Hordeum vulgare L.).Increased rate of mobilization of seed reserves with additionalNO-3or Cl-was associated with increases in shoot, root and residualseed anion content, total seedling water and residual seed watercontent (% water) 21 DAS. Addition of NH+4did not affect totalseedling water or residual seed water content. For barley suppliedwith different concentrations of NO-3or mannitol, the rate ofmobilization of seed reserves was positively correlated (r >0.95)with total seedling water and residual seed water content. Therate of mobilization of seed reserves of barley was greaterfor high N content seed than for low N content seed. Seed watercontent was greater for high N seed than for low N seed, 2 DAS.Additional NO-3did not affect total seedling water or residualseed water content until 10–14 DAS. The effects of seedN and NO-3on mobilization of seed reserves were detected 10and 14 DAS, respectively. It is proposed that the increasedrate of mobilization of seed reserves of temperate cereals withadditional NO-3is due to increased water uptake by the seedlingwhile the seed N effect is due to increased water uptake bythe seed directly. The effect of three different anion channel inhibitors, namely(5-nitro-2-3-phenylpropyl-amino)benzoic acid (NPPB), Zn2+ andanthracene-9-carboxylic acid (A-9-C) on the action potentialin the liverwort Conocephalum conicum were tested. All threecaused an increase of the excitability threshold and a decreaseof action potential amplitudes. This confirms the involvementof anion channels in the action potentials in Conocephalum.In plants treated with 1 or 2 mM A-9-C but not with NPPB (50or 100 µM) and Zn2+ (100 or 500 µM), a light-inducedtransient depolarization occurred. In contrast to action potentials,the amplitude of this voltage transient depended on the lightintensity and on the duration of preceding dark period. Alsoin contrast to action potentials, which are blocked by TEA,when applied together with A-9-C, TEA even increased the amplitudesof the light-induced voltage transients to up to 170 mV. Thedepolarization was obviously limited by the voltage-dependentopening of K+ channels in the absence of TEA. The amplitudeof the light-induced voltage transients (in the presence ofTEA) increased in elevated CaCl2 concentrations pointing toa Ca2+ permeability giving rise to the depolarization. However,none of the Ca2+ channel blockers tested, La3+, Gd3+, nifedipine,verapamil or diltiazem, had an effect. The light-induced voltagetransients in A-9-C treated plants are quite different fromlight- and electrically triggered action potentials but sharesome similarities with light-induced generator potentials. Soybean [Glycine max (L.) Merrill] seeds and cotyledons weregrown in an in vitro culture system to investigate the relationshipsbetween cell expansion (net water uptake by the seed) and drymatter accumulation. Seeds or cotyledons grown in a completenutrient medium containing 200 mol m–3 sucrose continueddry matter accumulation for up to 16 d after in planta seedsreached physiological maturity (maximum seed dry weight). Seedor cotyledon water content increased throughout the cultureperiod and the water concentration remained above 600 g kg–1fresh weight. These data indicate that the cessation of seeddry matter accumulation is controlled by the physiological environmentof the seed and is not a pre-determined seed characteristic.Adding 600 mol m–3 mannitol to the medium caused a decreasein seed water content and concentration. Seeds in this mediumstopped accumulating dry matter at a water concentration ofapproximately 550 g kg–1. The data suggest that dry matteraccumulation by soybean seeds can continue only as long as thereis a net uptake of water to drive cell expansion. In the absenceof a net water uptake, continued dry matter accumulation causesdesiccation which triggers maturation. Key words: Glycine max (L.) Measurements of the growth of sainfoin and lucerne were madein the field after cutting on 31 May 1977. Sainfoin reacheda total above-ground dry weight of 408 g m–2 over thegrowing period of 48 days compared with 598 g m–2 in lucerne.Final leaf area indices (LAIs) were 2.8 in sainfoin and 6.1in lucerne. The specific leaf areas (SLAs) for sainfoin wereapproximately half those of lucerne throughout the regrowthperiod. The maximum rates of leaf appearance were 0.12 leavesper day in sainfoin and 0.85 leaves per day in lucerne. Themaximum mean rate of plant extension growth for lucerne of 2.12mm h–1 occurred during the night, whereas, in sainfointhe maximum rate of 1.72 mm h–1 occurred during the day. Measurements of extinction coefficients for PAR ranged from0.45 to 0.89 in sainfoin and from 0 42 to 0.57 in lucerne. Asthe lucerne crop increased in size leaf water potentials andsolute potentials became more negative. In sainfoin leaf waterpotentials remained remarkably high throughout the growth period,solute potentials decreased and turgor potentials increased.The stomatal conductances of the two species were similar. The photosynthetic capacities and rates of dark respirationper unit leaf area in both species were similar. The rate ofcanopy ‘gross’ photosynthesis at 295 W m–2was always greater in lucerne than in sainfoin. This was largelya matter of differences between the species in LAI, althoughat higher LAIs the more erect structure of lucerne leads toa better utilization of photosynthetically active radiation. Hydrobiological studies in the Straits of Dover show a differencebetween the characteristics of the French coastal water andthe open sea water. A cruise between Boulogne-sur-mer and Dover(October 15, 1985) confirms the existence of a frontal area(4 miles off the French coast). This area is characterized byan increase of the salinity (1%) and decrease of the suspendedmatter content (from 27 to 20 mg l–1), of primary production(from 15 to 3 mg C m–3h–1) and of chlorophyll aconcentration (from 4.5 to 1.5 mg m–3). The presence ofdifferent zooplanktonic species in the two water masses alsocharacterizes this discontinuity. Oncaea sp., Corycaeus anglicus,Centropages typicus and Calanus helgolandicus are indicatorspecies of open sea water, while Cyciopina littoralis and ‘Saphirella’are only present in coastal water.  相似文献

Developing cladodes had lower water potentials and developingfruits had higher water potentials than the underlying cladodesof the widely cultivated prickly pear cactus, Opuntia ficus-indica.The 0.06 MPa lower value in 4-week-old daughter cladodes indicateda typical water potential gradient from the underlying clad-odealong the xylem of –0.2 MPa m–1; the 0.17 MPa highervalue in 4-week-old fruits, which decreased to 0.07 MPa by 10weeks, implicated the phloem as their supplier of water. Thephloem sap of the underlying cladodes had an osmotic pressureof only 0.90 to 0.98 MPa, so the phloem could supply a relativelydilute solution to the photosynthetically dependent fruits (daughtercladodes of O. ficus-indica are photosynthetically independentat 4 weeks). Although the water potentials were similar foradjacent tissues, the osmotic pressures were lower for the water-storagecompared with the photosynthetic tissue; the osmotic pressureswere higher for xylem sap from fruits, for which xylary flowapparently occurred toward the underlying cladodes, than fordaughter cladodes. The relative capacitance (change in relativewater content divided by change in tissue water potential) wasapproximately 0.71 MPa–1 for the water-storage tissueand the photosynthetic tissue of both daughter cladodes andfruits at 4 weeks of age. When these organs approached maturityat 10 weeks, the relative capacitance increased about 40% fortheir water-storage tissue, but decreased 30% for their photosynthetictissue. As the plant water content decreases during drought,about twice as much water will thus be lost per unit volumeof the water-storage tissue compared with the photosynthetictissue of maturing fruits and cladodes. Mucilage content in the stems of four sympatric cactus speciesvaried from none for Ferocactus acanthodes, 19% by dry weightfor Opuntia basilaris, 26% for Opuntia acanthocarpa, and 35%for Echinocereus engelmannii. Although the mucilage differedchemically among the species (the arabinose content ranged from17% to 51% of the sugar monomers), its relative capacitance(change in relative water content per unit change in water potential)remained about 15 Mpa–1. The relative capacitance of thewater-storage parenchyma averaged 1·04 Mpa–1 andwas consistent with the mucilage content, being lowest for F.acanthodes and highest for E. engelmannii. Mucilage isolatedfrom hydrated tissue was accompanied by solutes with an osmoticpressure of about 0·2 MPa. Such associated solutes influencethe water-release characteristics of mucilage and hence itsrole as an apoplastic capacitor. In particular, extracellularsolutes can facilitate the release of appreciable mucilage-boundwater to the cells at tissue water potentials occurring duringthe initial phases of drought. Evoked potentials recorded by implanted electrodes were usedto measure olfactory thresholds of unanesthetized laboratorymice (Mus musculus) to aliphatic carboxylic acids (formic acid,acetic acid, n-propionic acid, n-butyric acid, n-valeric acid,and n-caproic acid). The thresholds conformed to the typicalmammalian pattern, sensitivity to carboxylic acids increasingwith increasing chain length. The highest threshold was thatto formic acid, 7?1011 molecules/cm3 of air; that to caproicacid was lowest, 1.1?108 molecules/cm3.  相似文献

The ion contents and membrane potentials of the cells of young,hydroponically cultured seedlings of Atriplex hastata L. var.salina, Wallr. have been measured at several different NaClconcentrations. The total tissue concentrations of Na+ and Clincrease as external NaCl increases, but there is always a markedexcess of internal Na+ over Cl; this is balanced by endogenousorganic anion formation with a concomitant extrusion of H+ tothe bathing solution. Membrane potentials of the root cells remain essentially invariantwith changes in external NaCl at approx. –130 mV; thereis no evidence of a radial gradient of potential across theroot. The potential seems to contain a cyanide-sensitive electrogeniccomponent, also invariant with NaCl concentration, of about–70 mV, and a diffusion component. The electrogenic componentseems likely to be a H+ efflux, probably through a H+ uniportATPase.  相似文献

Diffusion potentials (concentration and bi- or multi-ionic potentials)in KC1, NaCl, or LiCl solutions have been measured across anisolated cell wall of Nitella, with or without the same concentrationof CaCl2 on either side of the cell wall. The absolute valueof the potentials decreases as the external Ca2+concentrationincreases and it may happen that an inversion of the sign ofthe concentration potentials results when the external Ca2+solution reaches 1 mM. Dosages of K+ and Ca2+ in the cell wallhave shown that Ca2+ easily displaces the monovalent ion fromthe exchange sites and tends to neutralize the cationic exchanger.However, in most cases, the measured potentials are still morenegative than the theoretical potentials which would be setup by a neutral-site membrane in the same conditions. Theseresults suggest that Ca2+ largely reduces the discriminationproperties of the cell wall between cations and anions.  相似文献

Movements of 11C and 14C in N. translucens occur at rates twoto five times greater than those of visually observed streaming.Like cytoplasmic streaming, longitudinal tracer movement isstopped reversibly by action potentials, irreversibly by N-ethylmaleimide(NEM) and is unaffected by colchicine (colch), suggesting thatboth processes share a common mechanism. Colchicine causes anincreased loading delay time. In both N. translucens and C. corallina the activity of 11Cand 14C at the nodes shows a sharp discontinuity: it is lowon the feed side of the node and high at the node itself. The dependence of membrane potentials on changes in the extra-cellularK+ concentration [K+]e was investigated in potato tuber sliceswith dripping perfusion, and in growing Vigna hypocotyl segmentswith pressurized intra-organ perfusion methods. Only under anoxiawere the membrane potential of potato tuber slices and the electricpotential difference between the parenchyma symplast and xylem(Vpx) of Vigna hypocotyl segments depolarized markedly (46 mVand 42 mV/log[K+]e unit, respectively) with increasing [K+]eabove the critical values. The electric potential differencebetween the parenchyma symplast and organ surface (Vps of thehypocotyl segments remained nearly unchanged up to 30 mEq [K+]e.Under highly aerobic conditions the membrane potentials wererelatively independent of [K+]e except at very high K+ concentrations.Vps showed even hyperpolarization with the increasing KCl concentrationin the perfusion solution that is not in direct contact withthe surface membrane of the parenchyma symplast. The respiration-dependentelectrogenic components of the membrane potentials regularlyincreased with the increasing [K+]e. A voltage-dependent homeostaticcontrol of membrane potential is discussed. The effects of increasing water stress on water relations, leafconductance, leaf extension and leaf rolling of Festuca arundinaceain sward (I m2) were investigated under wind tunnel conditions.The plants were grown in a container 60 cm deep and the experimentwas conducted over a 36 d period. Upon cessation of watering(day 11), leaf extension and conductance were affected. Within8 d, the onset of leaf rolling helped to reduce transpirationand to maintain leaf water potential. Nocturnal recovery of turgor potential helped in maintainingleaf extension at a moderate level and in the final 5 d waterand osmotic potentials dropped sharply as leaf rolling becamemore acute and leaf extension stopped. The grass combines various morphological and physiological mechanismsto prevent water losses and maintain growth. The effects of salinity on growth, water relations, glycinebetainecontent, and ion accumulation in the perennial halophyte Atriplexgriffithii var. stocksii were determined. The following questionswere addressed: (1) What effect does salinity have on growthresponses at different ages? (2) Is A. griffithii an ion accumulator?(3) Does A. griffithii accumulate glycinebetaine in responseto salinity? Atriplex griffithii plants were grown in pots at0, 90, 180 and 360  m M NaCl in sand culture in a plantgrowth chamber and plants were harvested after 30, 60 and 90d. Plant total dry weight was significantly inhibited at 360m M NaCl. Root growth showed a substantial promotion at 90 mM NaCl. The water potential and osmotic potential of shootsbecame more negative with increasing salinity and time of growth.The Na+and Cl-content in both shoots and roots increased withincreases in salinity. Increased treatment levels of NaCl induceddecreases in Ca+, K+and Mg2+in plants. Atriplex griffithii accumulateda large quantity of ions, with the ash content reaching 39%of the dry weight in leaves. Inorganic ion accumulation is significantin osmotic adjustment and facilitates water uptake along a soil-plantgradient. Glycinebetaine concentration was low in roots, andin stems it increased with increases in salinity.
