Gibberellic acid (GA3) applied at different times during the growth of wild carrot ( Daucus carota ssp. Carota ) cell suspension cultures inhibited anthocyanin accumulation. Application of 3 × 10–6 M GA3 to cultures on day 0 or day 4 gave, respectively, 10 or 35% of anthocyanin accumulation relative to levels occurring when GA3 was applied at the end of the growth period. Endogenous GAs were separated by high pressure liquid chromatography, and identified and quantified by gas chromatography-selected ion monitoring. Gibberellins GA1, GA3 and traces of GA8. GA19 and GA20 were identified in carrot cell suspension cultures of both high and low anthocyanin-accumulating clones. The concentrations of GA1. GA3 and GA8 in the two clones were similar and were not significantly different after the application of uniconazole which promoted anthocyanin accumulation. This suggests that these endogenous GAs are not the sole factors controlling the accumulation of anthocyanin in these different clones. Exogenous GA3 and uniconazole had no effect on 3'-nucleotidase and 5'-nucleotidase activity in the carrot cell suspension cultures. Thus 3'-nucleotidase does not appear to play a role in the inhibition of anthocyanin accumulation by exogenous GA3.  相似文献

The chelating agents, EDDHA, its iron salt, EDTA, and salicylic acid enhance bud formation in Bartramidula bartramioides (Griff.) Wijk & Marg. Salicylic acid elicits optimal response at 10–4 M , whereas the other substances do so at 10–7 M . Increased concentration of ferric citrate and cupric sulphate also stimulate bud induction. The accumulation of Fe3+ and Cu2+ is facilitated by chelators. The endogenous iron content is maximum at 10–7 M EDDHA or EDTA, which is also the concentration optimal for bud induction.  相似文献

Seeds of Kalanchoë blossfeldiana Poelln. cv. Feucrblüte, incubated on gibberellic acid, become very light-sensitive through a synergism between the far-red absorbing form of phytochrome and the growth substance, which results in high physiological activity of short far-red (FR) exposures. On 2 × 10-3 M gibberellic acid (GA3), one saturating FR pulse is somewhat more effective than one saturating red light (R) irradiation. Fluence-response curves for R and FR confirm this observation. At lower GA3 concentrations, this difference disappears and the effects of one saturating R and FR pulse decrease in an identical way with the GA3 concentration. When two saturating irradiations, separated by 24 h are given, the effect of FR falls off faster than that of R at low GA3 concentrations. Consequently, the second irradiation must have a different impact in comparison with the first one. Of the other growth substances tested, only a mixture of gibberellins A4 and A7 had an analogous, still more pronounced effect than GA3. Abscisic acid (ABA) inhibits the phytochrome-mediated germination of Kalanchoë , both in the absence and presence of GA3. An antagonism between ABA and GA3 was demonstrated.  相似文献

Abstract. A monoclonal antibody prepared against barley ( Hordeum vulgare L., cv. Himalaya) nuclease (EC 3.1.30.2) was characterized with solid-state enzyme-linked immunosorbent assays and immuno-blotting. The antibody was specific for intracellular and secreted nuclease. Hormonal regulation of the synthesis and secretion of nuclease in isolated aleurone layers was investigated by immunoprecipitation of biosynthetically-labelled nuclease using polyclonal antibodies and by immunoblot analyses using the monoclonal antibody, respectively. Gibberellic acid (GA3) induced the de novo synthesis and secretion of nuclease in a time-and concentration-dependent manner. Nuclease was detected in aleurone layers incubated in 1 mmol m−3 GA3, after 24 h. The maximum rates of nuclease synthesis and secretion occurred 36–48 h after hormone treatment. A minimum concentration of 10−6 mol m−3 GA3 was required for nuclease synthesis and secretion, whereas the maximum rate of nuclease secretion occurred at concentrations of 10−5 mol m−3 and higher. In the presence of abscisic acid, the synthesis and secretion of nuclease from GA3-treated aleurone layers was almost completely inhibited. Based on these findings, the authors conclude that all nuclease within and secreted from aleurone layers treated with GA3 is the result of its de novo synthesis.  相似文献

Antheridia were induced by exogenously applied GA3 at concentrations between 10−6 and 3 × 10−4 M in very young filamentous protonemata of Lygodium japonicum grown in darkness; the longer the dark preculture of protonemata, the lower was the sensitivity of the protonemata to GA3. Antheridial initials were discernible after 36 hr of GA3 treatment in the most sensitive protonemata, and the timing of antheridial initiation was delayed with increasing protonemal age.
This quantitative response of the protonemata provided the basis for a new method of assaying gibberellins in terms of the degree of antheridial formation. According to this method, all the gibberellins tested and one of their precursors were active in inducing antheridia in the protonemata, and the activity spectrum of the gibberellins was as follows: GA7>GA4>GA9>GA3>GA5>GA1>GA8.
The amounts of antheridiogen contained in conditioned media were measured by the present bioassay. A semi-logarithmic relation was shown between the percentage of antheridial formation and the concentration of conditioned medium within a certain dilution range. The amounts of antheridiogen secreted by the prothallia were quantitatively compared by transferring samples onto fresh media for a short period of time.  相似文献

Flowering in Poa pratensis L. cv. Holt and Bromus inermis Leyss. cv. Manchar requires exposure to short days (SD) for primary induction to occur, followed by long days (LD) to allow the inflorescence to develop. Weekly sprays with gibberellic acid (GA3) during primary induction inhibited flower initiation in both P. pratensis and B. inermis . With 10−4 M GA3 flowering of P. pratensis was suppressed even after an induction period of 10 weeks. Since both GA3 and non-inductive LD conditions greatly stimulate leaf elongation, the degree of primary induction was closely negatively correlated with plant height (leaf sheath and blade length) at the end of the induction period. GA3 application or the interpolation of LD during SD induction were most inhibitory during the later middle part of the SD period, whereas they were stimulatory near the beginning or immediately before the SD period. We suggest that changes in the portfolio or levels of endogenous gibberellins mediate photoperiodic control of growth and floral initiation in these plants. However, GA3 sprays could not substitute for LD in causing heading and culm elongation in SD induced plants of the two species. The results are discussed in the light of results with other plants with dual floral induction requirements.  相似文献

Effect of auxins on spermidine uptake into carrot protoplasts   总被引:1,自引:0,他引:1  
The effect of an auxin, indole-3-acetic-acid (IAA), on spermidine uptake into protoplasts of carrot ( Daucus carota L. cv. Ingrid) was studied. In the presence of 1 m M Ca2+, IAA (10−7 to 10−4 M ) enchances [14C]-spermidine uptake into carrot protoplasts, while no stimulation occurs in the absence of Ca2+. The time course of the uptake with and without IAA is very rapid and reaches saturation within 1 to 2 min. Preincubation of protoplasts with IAA inhibits the spermidine uptake. La3+, known not to penetrate the plasmalemma, exerts the same effect as Ca2+, but gives lower uptake values than Ca2+. The application of vanadate, an ATPase inhibitor, strongly inhibits IAA-stimulated spermidine uptake, suggesting that an energy-dependent mechanism may be involved in this transport. Neither spermidine nor Ca2+ alone stimulate IAA uptake. The synthetic auxin 2,4-dichlorophenoxyacetic acid, yields the same results as IAA with regard to time course of spermidine uptake with and without preincubation while, unlike IAA, no significant effect was observed on the Ca2+ -induced increase of spermidine uptake.  相似文献

Potassium promotes growth in several plant tissues. Elongation growth of the hypocotyls of Amaranthus caudatus L. ev. Lalsag is mainly controlled by gibberellins, but K+ also promotes growth. In the present study the interaction of K+ with gibberellin (GA3) and chlorocholine chloride (CCC) has been investigated. When K+ was applied externally in the dark, hypocotyl growth was promoted in the seedlings. External application of GA3 did not promote growth in the dark. GA3 was effective in the light and K+ was synergistic with GA3 in promoting elongation. Application of CCC in the dark makes the seedlings sensitive to GA3. The inhibition of growth by CCC was also reversed by K+. The results indicate a possible role of K+ in GA3 induced elongation of hypocotyls.  相似文献

The effects of the naturally occurring polyamines, spermine, putrescine, and spermidine were explored on mitochondrial state 3. state 4, and uncoupled respiration activities, ADP/O ratio, respiratory control ratio of pepper ( Capsicum annuum L. cv. Early Cal Wonder) and avocado ( Persea americana Mill. cv. Booth-8 or Simmonds) mitochondria oxidizing either succinate, external NADH, malate, α-ketoglutarate or tetramethyl- p -phenylenediamine. Abnormally high concentrations of spermine and spermidine such as might occur during chilling stress of these chilling-sensitive fruits were detrimental to several oxidase activities, especially to external NADH oxidase. State 3 respiration for NADH oxidase was inhibited more than 70% by 10 m M spermine. The spermine inhibition of uncoupled NADH oxidase was not reversed by the presence of divalent cations including Ca2+, Mg2+, Mn2+, and Sr2+ at concentrations up to 10 m M or by 100 m M KCl. The inhibition primarily affected the Vmax. Other possible sites of polyamine interactions are discussed.  相似文献

Raphanus sativus L. Chinese Radish Jumbo Scarlet has characteristics that make it an excellent plant model for vernalization studies. This study further characterizes flower induction of R. sativus Chinese Radish Jumbo Scarlet. Seed were imbibed in distilled water containing 0, 10−5 M or 10−3 M GA3 for 24 h and were then exposed to 6 ± 0.5°C (vernalized) for 0, 2, 4, 6, or 8 days. Seedlings were then grown under a short- (8 h) or long-day photoperiod (8 h with or without a 4-h night interruption; 2200–0200 h). Of unvernalized plants grown under long- and short-day conditions, 45 and 3% flowered, respectively. Saturation of the vernalization response occurred after a 4- or 8-day vernalization treatment when plants were placed under long- or short-days, respectively. Basal leaf number and days to anthesis decreased when seeds were cooled for 2 or 4 days and were imbibed with 10−3 M GA3 compared to distilled water only. These data indicate that R. sativus Chinese Jumbo Scarlet has principally an obligate vernalization requirement when grown under short-days. GA3 application only facilitated flowering when the length of the vernalization treatment was marginal. Taken together, these data support the use of this plant as a model plant for identifying vernalization responses under short-day conditions.  相似文献

Ascorbic acid is a strong inhibitor of indole-3-acetic oxidation catalyzed by commercial horse-radish peroxidase. In the presence of excess ascorbic acid, the indole-acetic acid oxidation catalysis is apparently blocked. The activity of peroxidase for indoleacetic acid at pH 3.7 and 33°C, in the presence of 2,4-dichlorophenol and MnCl2 as promotors was measured by polarographic technique. The Km was 0.27 m M and the maximum velocity was 1.02 mmol O2 (mg protein)−1 min−1. Dixon plots lead to an apparent Ki of 1.25 (μ M for ascorbic acid and the inhibition was apparently competitive. Ascorbic acid, besides appearing to be a strong inhibitor of the IAA oxidase activity of peroxidase, seemed to protect IAA from total degradation. Addition of more than 5 μ M ascorbic acid produced both an exponential increase in the lag time before the onset of reaction and, at the end, an oxidation protection of 26 μ M IAA when 111 μ M IAA was present at the stawrt. The possibility of ascorbic acid-IAA auxin from endogenous oxidation in plants, is proposed.  相似文献

The plant-growth-promoting rhizobacteria (PGPR), Bacillus pumilus and Bacillus licheniformis, isolated from the rhizosphere of alder ( Alnus glutinosa [L.] Gaertn.) have a strong growth-promoting activity. Bioassay data showed that the dwarf phenotype induced in alder seedlings by paclobutrazol (an inhibitor of gibberellin [GA] biosynthesis) was effectively reversed by applications of extracts from media incubated with both bacteria and also by exogenous GA3. Full-scan gas chromatography-mass spectrometry analyses on extracts of these media showed the presence of GA1, GA3, GA4and GA20, in addition to the isomers 3- epi -GA1 and iso -GA3. Isotope dilution analysis indicated that epi -GA1 was an artefact. Likewise, iso -GA3 is also probably an artifact spontaneously formed during extraction and/or analysis. In both culture media, GA1 was present in higher concentrations (130–150 ng ml−1) than GA3 (50–60 ng ml−1), GA4 (8–12 ng ml−1) and GA20 (2–3 ng ml−1). The data indicated that culture of both bacteria accumulate bioactive C19-gibberellins in relative high amounts and that these GAs appear to be physiologically active in the host plant. The evidence suggests that the promotion of stem elongation induced by the PGPR could be mediated by bacterial GAs.  相似文献

Abstract: The influence of putrescine, spermidine, spermine, and some aliphatic α,ω-diamines on the uptake of neurotransmitters by rat forebrain synaptosomes was investigated. Choline uptake was most effectively inhibited by spermine (IC50= 0.22 m M ), less so by spermidine (IC50= 4.0 m M ), but not by putrescine (IC50 > 100 m M ). At 10 m M, 1,3-diaminopropane, cadaverine, and 1,8-diaminooctane all inhibited choline uptake by 50% or more. Spermine and spermidine inhibited the uptake of dopamine with IC50 values of 2.7 and 2.2 m M , respectively. Putrescine was only slightly inhibitory (IC50= 17.3 m M ) and the other diamines were inactive. The uptake of γ-aminobutyrate (GABA) was only slightly inhibited (15–40%) by the polyamines at 10 m M . With the exception of inhibition of glycine uptake by 1,8-diaminooctane (60%) and of glutamate uptake by cadaverine (35%) none of the polyamines, tested at 10 m M , affected the uptake of adenosine, glutamate, and glycine significantly. A possible modulatory role for polyamines in synaptic transmission through interaction by negatively charged groups of the synaptic membrane with the polycationic compounds is discussed.  相似文献

1. Termite nitrogenase activity was highest in autumn and spring (≈ 3 μg N2 fixed termite fresh mass (g)–1 day–1) and lowest in winter and summer (≈ 0·8 μg N2 fixed termite fresh mass (g)–1 day–1).
2. The nitrogenase activity of worker termites was significantly higher than all other castes (1·58 ± 0·27 μg N2 fixed termite fresh mass (g)–1 day–1).
3. Worker termites constituted the largest proportion of all the castes throughout the study period (≈ 90%).
4. The localized input of fixed nitrogen by termites may reach 15·3 mg N log–1 day–1 and 5·6 g N log–1 year–1.  相似文献

The role of gibberellins (GAs) in the induction of parthenocarpic fruit-set and growth by the pat-3/pat-4 genetic system in tomato ( Lycopersicon esculentum Mill.) was investigated using wild type (WT; Cuarenteno) and a near-isogenic line derived from the German line RP75/59 (the source of pat-3/pat-4 parthenocarpy). Unpollinated WT ovaries degenerated but GA3 application induced parthenocarpic fruit growth. On the contrary, parthenocarpic growth of pat-3/pat-4 fruits, which occurs in the absence of pollination and hormone treatment, was not affected by applied GA3. Unpollinated pat-3/pat-4 fruit growth was negated by paclobutrazol, an inhibitor of ent -kaurene oxidase, and this inhibitory effect was negated by GA3. The quantification of the main GAs of the early 13-hydroxylation pathway (GA1, GA8, GA19, GA20, GA29 and GA44) in unpollinated ovaries at 3 developmental stages (flower bud, FB; pre-anthesis, PR; and anthesis, AN), by gas chromatography-selected ion monitoring, showed that the concentration of most of them was higher in pat-3/pat-4 than in WT ovaries at PR and AN stages. The concentration of GA1, suggested previously to be the active GA in tomate, was 2–4 times higher. Unpollinated pat-3/pat-4 ovaries at FB, PR and AN stages also contained relatively high amounts (5–12 ng g−1) of GA3, a GA found at less than 0.5 ng g−1 in WT ovaries. It is concluded that the mutations pat-3/pat-4 may induce natural facultative parthenocarpy capacity in tomato by increasing the concentration of GA1 and GA3 in the ovaries before pollination.  相似文献

Fifteen different gibberellins (GA's) were tested for their ability to induce elongation growth under short day conditions in seedlings of Salix pentandra L. GA's were applied either to the apex or they were injected into a mature leaf. GA3 was highly active and also GA4+7 and GA4 showed high activity. GA1, GA2, GA5, GA9, GA13, GA20, GA36 and GA47 showed moderate activity. GA16, GA17, GA27 and GA41 exhibited low or no activity in doses up to 10 μg per plant. In general, a better growth response was obtained with an application to the apex than with an injection into the leaf.  相似文献

When 1 m M spermidine or spermine was included in an absorption solution which contained 20 m M Na+ and 1 m M Rb+, Na+ influx into excised maize roots ( Zea mays L. cv. Golden Cross Bantam) was reduced. Rb+ influx was reduced in the presence of spermidine and uneffected in the presence of spermine when compared with control solutions. When 1 m M Ca2+ replaced the polyamines, Na+ influx was strongly reduced and Rb+ influx was promoted. Rb+ influx from 1 m M Rb+ solutions which did not contain Na+ was also promoted by 1 m M Ca2+, but was inhibited by 1 m M spermidine. This Ca2+ promotion of Rb+ influx could be reversed by 10 times greater concentration of spermidine in the absorption solution. H+ efflux from excised roots was inhibited by spermidine when compared with Ca2+ or control solutions, however, the plasma membrane ATPase was not inhibited by spermidine. It is concluded that external Ca2+ plays two separate roles in membrane function, only one of which can be substituted for by polyamines. The first role, maintenance of membrane integrity, can be substituted for by spermidine or spermine. The second function, maintenance of the Rb+ transport mechanism, is Ca2+ specific and cannot be substituted for by spermidine or spermine. The results of this study are discussed in terms of electrostatic interactions between the plasma membrane and the Ca2+ or polyamines.  相似文献

Plants of Poa pratensis cv. Holt initiate inflorescence primordia when exposed to short days (SD) and low temperature, but require a secondary induction by at least 4 long days (LD) for further inflorescence development and stem elongation. Single or double applications of 10 µg per plant of gibberellins A1, A3, A5 and 16,17‐dihydro A5 (DHGA5) induced inflorescence development in a high proportion of plants in SD, but only if the plants were detillered to a single stem. Exposure to 2 LD cycles did not cause heading and flowering alone but enhanced the effect of exogenous gibberellins (GAs), bringing flowering to 100%. GA5 and DHGA5 were less effective than GA1 and GA3 in SD, especially with double applications, but were more effective than GA1 and GA3 when given together with 2 LD. The GAs had differential effects on vegetative growth and flowering, GA5 and DHGA5 causing much less leaf and stem growth than the other two GAs. Marginal induction, whether by LD or GA application, resulted in a high proportion of spikelets with viviparous proliferation. Thus, whereas GAs are inhibitory to the primary induction by SD, they can replace secondary induction by LD when vegetative growth is limited.  相似文献

Four-week-old sunflower plants ( Helianthus annuus L. cv. Halcón), grown in different nutrient solutions, were used to study the effects of gibberellic acid (GA3) on K+ (Rb+) uptake by roots or transport to the shoot. Gibberellic acid application to the nutrient solution did not affect the exudation process of excised roots. When GA3 was sprayed on leaves 2 to 6 days before excising the roots, the rate of exudation and the K+ flux increased. When the exudation study was done keeping the roots in a nutrient solution in which Rb+ replaced K+, the GA3 effects were evident also on Rb+ uptake and transport. In intact plants, GA3 increased the Rb+ transported to the shoot but did not affect Rb+ accumulation in the root. It is suggested that these GA3 effects can be explained if it is assumed that GA3 acts on the transport of ions to the xylem vessels.  相似文献

In the temperate-zone woody species Salix pentandra elongation growth is regulated by the photoperiod. Long days sustain active growth, whereas short days induce cessation of apical growth, which is a prerequisite for winter hardening. It is shown that this is correlated to quantitative changes in levels of endogenous GA19 GA20, and GA1. Within two short days the amount of the active GA1 and its immediate precursor GA20, decreased markedly in young leaves us well as in stem tissue. Also, the amount of GA19, declined, but the decrease was delayed relative to that of GA1 and GA20. The ability of S. pentandra seedlings to respond to exogenous GA19, decreased with increasing numbers of short days. Observations that support the hypothesis that the level of GA1 in S. pentandra is regulated by the photoperiod in a quantitative mode with conversion of GA19, to GA20, being one target for control.
Different distribution of GAs in various plant parts was observed. The level of GA was higher in young leaves than in other plant parts, and the amount of GA19 was 5–10 times higher in stem tissue than in leaves and roots. The ratios of GA8 to GA1 and GA20, were higher in roots as compared with other parts, as rods contained very low levels of GA1 and GA20, but amounts of GA20 comparable with other parts.  相似文献
