将sos型H-ISFET与戊二醚交联的牛血清蛋白-青霉素酶膜组合成单管输出式青霉素酶-H+-ISFET传感器的探头(以下简称青霉素-酶FET),并用于测定溶液中的青霉素含量。青霉素-酶FET在0.005mol/L、O.Olmol/L、0.02mol/L磷酸缓冲液中的响应灵敏度分别为11—12mV/m mol/L、7.5—8.0mV/m mol/L以及3.7—4.OmV/m m01/L;响应时间为30s,在0.02mol/L磷酸缓冲液中的标定曲线线性范围为O.5—25mmo】/L’相关系数为O.9976。该青霉素·酶FET在青霉索浓度为10mmol/L的0.01mo1/L磷酸缓冲液中重复测定8次的标准偏差sD为1.67mv,变异系数cv为2.1%;贮存寿命大于4个月,使用寿命在1个月以上(每天测试一次)。  相似文献

从北京地区土样中分离到一株革兰氏阴性、无芽胞、稀周毛及运动的菌株S-1231。它能以糖类为底物产丁二酸型胞外多糖,不能利用淀粉和纤维素。该菌发酵葡萄糖产酸,生长12-24小时.细胞杆状O.7一O.8×1.3—1.5μm,圆端,单个或成对。在营养洋菜平板上菌落圆形、低凸、表面光滑、润泽、边缘整齐。该菌产生3-酮基乳糖,接种向日葵不致瘤,DNA中G十c含量62.8 -63.4免分子%。该菌氧化酶阳性,接触酶阳性,产生H2s,35℃生长,2%NaCl生长,及石蕊牛奶产碱,该菌定为放射形土壤杆菌生物变型Ⅰ。组分分析表明.该菌株产生的胞外多糖(简称Agran-s)由D-葡萄糖(69.1%),D-半乳糖(8.6%),丙酮酸(9.5%)和丁二酸(10.8%)组成。甲基化分析表明,多糖Agran-s含有下列主要结构单位(均为β-糖苷键):(1→3)一连接的D-葡萄糖(21 2%);(1→3)-连接的D-半乳糖(11.4%);(1-6)-连接的D葡萄糖(10.5%);(1→4)-连接的D-葡萄耱(30.4%)。(1→4,1→6)连接的D-葡萄糖(22.2%)和末端D-葡萄糖(4.3%)。  相似文献

用基因重组及定位突变技术成功地构建了t·PA的Kl区缺失突变体t—PAdelKl、PAI-1结合位点缺失突变体t—PA del(296—302)及两者的组合突变体t—PA del(Kl·296—302),并在COS-7细胞中实现三者的暂时性表达,在cHO细胞中实现了t·PA deI(K1.296—302)的稳定性表达.对表达产物的生物学特性分析表明,t—PA del(296—302)及t-PA del(K1,296—302)获得了Pal-1抗性.因时t-PA del(K1.296—302)在大鼠体内的半衰期延长约5倍,而与纤维蛋白的亲和力只略有下降。因此,此组合突变体有可能成为优于野生t—PA的新型溶栓剂候选株.  相似文献

带有枯草杆菌B一1,3—1,4葡聚糖酶基因(bglS)7.1kb的EcoRⅠ片段,经亚克隆,将1.5kb的EcoRⅠ—PstⅠ酶切片段插入pucl9的po1ylinker上,转化E.Coil JM101后合成出β-l,3-l,4葡聚糖酶,它能专一性地降解大麦β-1,3—1,4葡聚糖和地衣多糖,酶的大部分(>50%)留在胞内,其中25%分泌到胞外。根据酶特性分析,大肠杆菌中合成的β一1,3—1,4葡聚糖酶完全与出发菌株枯草杆菌相同。  相似文献

应用5L生物反应器悬浮培养乙肝重组DNA转化的CHO细胞B43株生产HBsAg。试验了细胞接种量、微载体的加入方式、培养方法(半流加培养和连续灌流名养)对细胞生长形态和HBsAg分泌的影响,初步建立了5L生物反应器的生产工艺,在5L生物反应器最适合连续灌流培养的条件是pH 7.30—7.40,Do 25~3;%,T 36.5℃,微载体6~8g/L,灌流速度5.5—7.5L,24h细胞连续培养60天,细胞密度维持在5.O×10 6~1.O×10 7/ml之间,收获细胞液的RPHA效价为l:512—1:1024.HlBsAg含量为3-5mg/I。  相似文献

研制了一种融微生物肥有机肥和无机肥于一体的新型复合肥料。测定结果,含微生物活菌总量为2.5亿个/g,其中固氮、解磷、解钾功能做生物活菌量达1.0亿个/g,有机干物质34.3-37.3%,含氮(N)7.8-8.9%,全磷(P2O5)5.2-5.5%,全钾(K2O)4.8—5.0%。毒性试验结果表明,该肥无毒无害,对环境不构成危害。生产过程中采取了有机物料发酵和功能微生物大量繁殖一次同步完成,添加除臭微生物、机械自动翻拌等工艺。田间试验表明,与等养分或等价格投入的化肥相比,增产9-18%,对多种经济作物品质有明显的改善作用,且有一定的改土培肥效应。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. We also discuss the development and practicality of various cell culture and animal model systems to study KSHV replication and pathogenesis.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
