AIMS: To express and identify the Salmonella enterica ser. Enteritidis outer membrane proteins that are involved in the attachment to human intestinal epithelial cells. METHODS AND RESULTS: Salmonella Enteritidis attachment proteins were expressed by infection of human intestinal epithelial cells (int-407) with Salm. Enteritidis strain SE28, followed by pulse labelling with [35S]-methionine. Outer membrane proteins were separated by TritonX-114 phase partitioning, and were detected by Salm. Enteritidis-specific polyclonal antibody. Outer membrane proteins of molecular weights 82.3, 75.6, 49.3, 35.5 and 19.3 kDa were newly synthesized. Expression of 31.2 and 16 kDa proteins was up-regulated, whereas the expression of 40.7 kDa protein was down-regulated. Polyclonal antibodies against the 82.3 and 75.6 kDa proteins significantly (P < 0.05) reduced the binding of Salm. Enteritidis to int-407 cells in vitro. CONCLUSIONS: Outer membrane proteins 82.3 and 75.6 kDa are potentially involved in the attachment of Salm. Enteritidis to the intestinal mucosa. SIGNIFICANCE AND IMPACT OF THE STUDY: Outer membrane proteins 82.3 and 75.6 kDa identified in this study could be used as potential vaccines to block or reduce Salm. This study was conducted to investigate the effects of increasing levels of alfalfa meal in the diet of laying quails on egg production, feed consumption, feed efficiency, egg quality, egg yolk cholesterol and selected serum parameters. In this study, 192, 10-week old quails (Coturnix coturnix japonica) were evenly distributed to four groups with four replicates of 12 quails each. The control group was fed a basal diet containing 0% alfalfa meal and the remaining groups received 3, 6 or 9% alfalfa meal for 12 weeks. Live weight, feed consumption, and egg production were recorded and feed efficiency were calculated. Eggs were examined for interior and exterior quality and egg yolk cholesterol content. At the end of the experiment, blood samples were collected and sera were analysed for serum Ca, inorganic P (Pi), Mg, triglycerides and total cholesterol. Any level of alfalfa meal had no effect on live weight, egg production, feed consumption, feed efficiency, egg weight, and egg yolk index. Six percent and 9% alfalfa meal increased specific gravity of whole egg and eggshell thickness as well as serum Pi levels. Nine percent alfalfa meal reduced serum triglycerides, total cholesterol levels and egg yolk cholesterol content. The results of this experiment indicated that addition of 9% alfalfa meal into the laying quail diet may improve eggshell quality and reduced serum triglycerides and serum and egg yolk cholesterol without any adverse effect on performance.  相似文献

A diet containing a high concentration of soy isoflavone was administered to laying hens and the contents of the isoflavones transferred to the plasma and egg yolk were measured. A method for quantitatively measuring the concentration of isoflavone in the yolk was first established, before a high concentration of soy isoflavone was administered to the laying hens over an 18-day period. The concentrations of isoflavone in the plasma and egg yolk reached their highest on the 12th day of the feeding period, the values being 3,167 nmol/l and 65.29 μg/100g, respectively. The concentration of cholesterol in the yolk was slightly affected during the early stages of the feeding period. These findings clearly demonstrate that soy isoflavone was transferred into the yolk from the feed and that the cholesterol concentration in the yolk was affected by administering the soy isoflavone-enriched feed.  相似文献

The biomass of the red alga Porphyridium sp.constitutes a unique combination of soluble sulfatedpolysaccharide that accounts for about 70% of thealgal dry weight, and various polyunsaturatedfatty acids (PUFA) such as arachidonic andeicosapentaenoic acid (AA, 20:4 6 and EPA,20:5 3). In view of earlier results in ourlaboratory showing a reduction in serum cholesteroland triglyceride levels in rodents fed with red algalbiomass, we set out to examine the influence of algalbiomass as a feed additive on the metabolism ofchickens, with an emphasis on blood and eggcholesterol levels. For that purpose, lyophilizedalgal biomass was fed to 12–13, 30-week-old, WhiteLeghorn chickens for 10 days at a proportion of 5% or10% of the standard chicken diet. Twelve chickensfed with unsupplemented diet served as the control. No differences in body weight, egg number, and eggweight were found between the algal-fed chickens (atboth concentrations) and the control. However,chickens fed with algal biomass consumed 10% lessfood for both groups, and their serum cholesterollevels were significantly lower (by 11% and 28% forthe groups fed with 5% and 10% supplement,respectively) as compared with the respective valuesof the control group. Egg yolk of chickens fed withalgae tended to have reduced cholesterol levels (by10%) and increased linoleic acid and arachidonic acidlevels (by 29% and 24%, respectively). In addition,the color of the egg yolk was darker as a result ofthe higher carotenoid levels (2.4 fold higher) forchickens that fed with 5% supplement. Theseresults encourage the development of an improvedchicken feed having dietary fibers and polyunsaturatedfatty acids.  相似文献

Growing and laying chickens were fed graded levels of selenium in the form of sodium selenite. One day old Norwegian bred broiler chickens and 20 weeks old Norwegian bred White Leghorn chickens were divided into 5 groups each and fed a basal diet supplemented with 0, 0.1, 1.0, 3.0 or 6.0 μg Se/g for 6 and 31 weeks, respectively. At the end of the experiments significantly higher concentrations of selenium were found in the groups fed 1.0, 3.0 and 6.0 μg Se/g diet compared to the control group. Correspondingly higher concentrations of selenium were found in egg samples. The increase in egg yolk selenium was much higher than in egg white. Significant correlations were found between the amounts of selenium added to the ration and the selenium concentrations in liver, kidney, breast muscle, egg white, yolk and homogenized egg. There were no differences in body weight gain and egg production between the groups. A possible positive contribution to animal and human health of selenium supplementation of animals’ diet above the required level is discussed.  相似文献

BACKGROUND AND AIM: Helicobacter pylori is known to be a major pathogenic factor in the development of gastritis, peptic ulcer disease and gastric cancer. Recently, chicken egg yolk immunoglobulin Y (IgY) has been recognized as an inexpensive antibody source for passive immunization against gastrointestinal infections. The present study was designed to investigate the effect of anti-urease IgY on H. pylori infection in Mongolian gerbils. METHODS: H. pylori-infected Mongolian gerbils were administered a diet containing anti-urease IgY, with or without famotidine (F). After 10 weeks, bacterial culture and measurement of the gastric mucosal myeloperoxidase (MPO) activity were performed. In a second experiment, another group of gerbils was started on a diet containing F + IgY a week prior to H. pylori inoculation. After 9 weeks, these animals were examined. RESULTS: In the H. pylori-infected gerbils, there were no significant differences in the level of H. pylori colonization among the different dietary and control groups. However, the MPO activity was significantly decreased in the H. pylori group administered the F + IgY diet compared with that in the H. pylori group administered the IgY, F, or control diet. Furthermore, in the gerbils administered the F + IgY diet prior to the bacterial inoculation, inhibition of H. pylori colonization and suppression of the elevated gastric mucosal MPO activity were observed. CONCLUSIONS: Oral administration of urease-specific IgY not only inhibited H. pylori disease activity in H. pylori-infected gerbils, but also prevented H. pylori colonization in those not yet infected. These encouraging results may pave the way for a novel therapeutic and prophylactic approach in the management of H. pylori-associated gastroduodenal disease.  相似文献

This experiment was carried out to determine the effects of the usage of dried brewing yeast in quail diets on laying performance, egg traits and blood parameters. A total of 240 Japanese quails (Coturnix coturnix japonica) aged 10 weeks were randomly allocated into one control group and three treatment groups. Each group was divided into five replicates as subgroups, comprising 12 quails each. Dried brewing yeast (Saccharomyces cerevisiae) was used at the levels of 1.5%, 3.0% and 4.5% in the diets of the first, second and third treatment groups, respectively. Soyabean meal was replaced with dried brewing yeast. The diets were formulated to be isocaloric and isonitrogenous. The experimental period lasted 18 weeks. Dietary treatments did not significantly affect body weight, daily feed intake, daily protein intake, egg production, egg weight, feed efficiency, mortality, egg shell thickness, egg albumen index, egg yolk index, egg Haugh unit, the percentages of egg shell, albumen and yolk, excreta moisture and small intestinal pH. Inclusion of 3% and 4.5% dried brewing yeast in diets reduced egg yolk cholesterol concentration as mg per yolk and mg per g yolk (P < 0.01). Blood serum cholesterol of groups fed diets with dried brewing yeast was significantly lower (P < 0.01) than that of the control group. Feeding diets containing 3.0% and 4.5% dried brewing yeast resulted in significant increases (P < 0.01) in blood serum levels of total protein, alanine aminotransferase at the end of the experiment. Blood serum levels of uric acid, triglyceride, aspartate aminotransferase and alkaline phosphatase were not affected by dietary dried brewing yeast. It is concluded that dried brewing yeast can be used up to 4.5% in the diets of laying quails without adverse effects on the measured parameters.  相似文献

The study investigated whether different dietary energy and protein sources affect laying performance, antioxidant status, fresh yolk fatty acid profile and quality of salted yolks in laying ducks. In all, 360 19-week-old Longyan ducks were randomly assigned to four diets in a factorial arrangement (2×2). The four diets consisted of two energy sources, corn (CO) or sorghum (SO) and two protein sources, soybean meal (SM) and rapeseed meal with corn distillers dried grains with solubles (RMD), and each treatment contained six replicates of 15 birds each. The experimental diets were isocaloric (metabolizable energy, 10.84 MJ/kg) and isonitrogenous (CP, 17%). The results showed that egg production, average egg weight, egg mass and feed conversion ratio were not affected by diets (P>0.05). Plasma contents of reduced glutathione (GSH), GSH/oxidized glutathione and total antioxidant capacity were lower (P<0.05) in ducks fed the RMD diets compared with those fed SM diets with a substantial increase (P=0.006) in plasma content of malondialdehyde (MDA). Egg yolks from ducks fed SO diets had higher proportions of polyunsaturated fatty acids (PUFA) and lower saturated and monounsaturated fatty acids compared with CO diets (P<0.001). Similarly, ducks fed RMD diets had a higher content of PUFA and n-6/n-3 ratio in fresh yolks (P<0.001), and increased salted yolk MDA, carbonylated proteins content and incidence of hard salted yolks (P<0.05) compared with SM diets. Scanning electron microscopy showed that salted yolks contained rougher polyhedral granules and fewer fat droplets, and were surrounded with a layer of bunchy fibers in ducks fed SO+RMD than those fed CO+SM diet. In conclusion, the current study showed that feeding laying ducks with diets containing SO or RMD reduced antioxidant capacity and increased egg yolk concentrations of PUFA. It appeared that egg yolks from ducks fed these diets were more sensitive to lipid peroxidation and protein oxidation during salting, and reduced the quality of salted yolks.  相似文献

Phenotypic measurements of chicken egg character and production traits are restricted to mature females only. Marker assisted selection of immature chickens using quantitative trait loci (QTL) has the potential to accelerate the genetic improvement of these traits in the chicken population. The QTL for 12 traits (i.e. body weight (BW), six for egg character, three for egg shell colour and two for egg production) of chickens were identified. An F2 population comprising 265 female chickens obtained by crossing White Leghorn and Rhode Island Red breeds and genotyped for 123 microsatellite markers was used for detecting QTL. Ninety-six markers were mapped on 25 autosomal linkage groups, and 13 markers were mapped on one Z chromosomal linkage group. Eight previous unmapped markers were assigned to their respective chromosomes in this study. Significant QTL were detected for BW on chromosomes 4 and 27, egg weight on chromosome 4, the short length of egg on chromosome 4, and redness of egg shell colour (using the L*a*b* colour system) on chromosome 11. A significant QTL on the Z chromosome was linked with age at first egg. Significant QTL could account for 6-19% of the phenotypic variance in the F2 population.  相似文献

In Australia, free-range egg production pullets are typically reared indoors, but adult layers get outdoor access. This new environment may be challenging to adapt to, which could impair egg production and/or egg quality. Adaptation might be enhanced through rearing enrichments. We reared 1386 Hy-Line Brown® chicks indoors with three treatments across 16 weeks: (1) a control group with standard litter housing conditions, (2) a novelty group providing novel objects that changed weekly, and (3) a structural enrichment group with custom-designed structures to partially impair visibility across the pen and allow for vertical movement. Pullets were transferred to a free-range system at 16 weeks of age with daily outdoor access provided from 25 until 64 weeks. Daily egg production at different laying locations (large nests, small nests and floor), weekly egg weights and egg abnormalities were recorded from 18 to 64 weeks old. External and internal egg quality parameters of egg weight, shell reflectivity, albumen height, haugh unit, yolk colour score, shell weight and shell thickness were measured at 44, 52, 60 and 64 weeks. There was a significant interaction between rearing treatment and nest box use on hen-day production from weeks 18 to 25 (P < 0.0001) with the novelty hens laying the most eggs and the control hens the fewest eggs in the nest box. Similarly, from 26 to 64 weeks, the novelty hens laid more eggs in the large nest boxes and fewer eggs on the floor than both the structural and control hens (P < 0.0001). Egg weight and abnormalities increased with age (P < 0.0001), but rearing treatment had no effect on either measure (both P ≥ 0.19). Rearing treatment affected shell reflectivity and yolk colour with the control hens showing paler colours across time relative to the changes observed in the eggs from enriched hens. The novelty hens may have established nest box laying patterns as they were more accustomed to exploring new environments. The differences in egg quality could be related to stress adaptability or ranging behaviour. This study shows that enriching environments during rearing can have some impacts on production parameters in free-range hens.  相似文献

The effects of dietary coenzyme Q10 (CoQ10) on cholesterol metabolism in laying hens were investigated. Dietary CoQ10 significantly reduced egg yolk cholesterol content and suppressed hepatic hydroxymethylglutaryl-CoA reductase (HMGR) activity. It is therefore likely that CoQ10 acts as an HMGR inhibitor in the livers of laying hens, which in turn results in a reduction in egg-yolk cholesterol.  相似文献

Salmonella Enteritidis (SE) has been the major cause of the food-borne salmonellosis pandemic in humans over the last 20 years, during which contaminated hen's eggs were the most important vehicle of the infection. Eggs can be contaminated on the outer shell surface and internally. Internal contamination can be the result of penetration through the eggshell or by direct contamination of egg contents before oviposition, originating from infection of the reproductive organs. Once inside the egg, the bacteria need to cope with antimicrobial factors in the albumen and vitelline membrane before migration to the yolk can occur. It would seem that serotype Enteritidis has intrinsic characteristics that allow an epidemiological association with hen eggs that are still undefined. There are indications that SE survives the attacks with the help of antimicrobial molecules during the formation of the egg in the hen's oviduct and inside the egg. This appears to require a unique combination of genes encoding for improved cell wall protection and repairing cellular and molecular damage, among others.  相似文献

AIMS: To develop an animal model to study dose-response relationships of enteropathogenic bacteria. METHODS AND RESULTS: Adult, male Wistar Unilever rats were exposed orally to different doses of Salmonella enterica serovar Enteritidis after overnight starvation and neutralization of gastric acid by sodium bicarbonate. The spleen was the most sensitive and reproducible organ for detection of dose-dependent systemic infection. Illness was only observed in animals exposed to doses of 10(8) cfu or more. At lower doses, histopathological changes in the gastro-intestinal tract were observed, but these were not accompanied by illness. Marked changes in numbers and types of white blood cells, as well as delayed-type hyperresponsiveness, indicated a strong, dose-dependent cellular immune response to Salm. Enteritidis. CONCLUSION: The rat model is a sensitive and reproducible tool for studying the effects of oral exposure to Salm. Enteritidis over a wide dose range. SIGNIFICANCE AND IMPACT OF THE STUDY: The rat model allows controlled quantification of different factors related to the host, pathogen and food matrix on initial stages of infection by food-borne bacterial pathogens.  相似文献

Avian communal breeding systems generate alternative behavioural strategies for females, resulting in differences in reproductive success. Identifying eggs of different females in such systems is problematic, however, due to egg destruction before incubation, difficulty of capturing adults, and/or inaccuracy of egg identification based on egg morphometry. Here, we describe a technique that uses electrophoresis of yolk proteins to determine egg ownership, which we applied to communally breeding guira cuckoos (Guira guira). Validation of the method included identical yolk protein banding patterns in all eggs of the same female, but different patterns in eggs of different females in budgerigars (Melopsittacus undulatus), and identical patterns in yolk follicles of the same females in guira cuckoos. We applied the protocol to 195 guira cuckoo eggs from 34 joint nests in 2 years. All multiple guira cuckoo eggs laid on the same day in single nests had distinct banding patterns of yolk proteins, practically eliminating the possibility of more than one female being represented by the same pattern. Some identical banding patterns were repeated in different days within a nesting bout, indicating that some females laid several eggs in shared nests. Identical patterns occasionally occurred in renestings of groups, indicating that some females lay eggs in consecutive nestings. Yolk protein electrophoresis is a useful tool to identify egg maternity in other circumstances, such as polygynous mating systems with joint nests and intraspecific parasitism. Additionally, it is an alternative method for species where electrophoresis of egg white proteins does not show sufficient polymorphism.  相似文献

An experiment was conducted to determine the effect of dietary vitamin E and C on serum metabolites, yolk cholesterol, egg quality, and performance of layer hens. One hundred sixty-eight commercial Hy-Line W-36 layer hens were randomly divided into seven groups and six replicates with four hens in each. Dietary treatments were introduced after the pre-experimental period (10 days) to adjust egg production. Treatments were levels of vitamin E or C (100, 200, and 400 mg/kg diet) supplementation to the basal diet for 4 weeks, whereas the control group received no supplementation. Egg production, egg weight, and feed consumption were recorded during the study. Shell thickness, Haugh unit score, yolk color, yolk weight, yolk cholesterol, and blood parameters were measured at the end of experiment. There was no significant effect of dietary vitamin E or C on hen performance. Egg yolk cholesterol concentrations decreased linearly by antioxidant vitamin supplementation (P?<?0.01). Egg yolk cholesterol reduction did not have any negative effect on egg production rate. Antioxidants, especially vitamin C, increased serum glucose concentration (P?<?0.05). Serum total cholesterol content did not change by vitamin supplementation but cholesterol in high-density lipoprotein (HDL-C) decreased and cholesterol in low-density lipoprotein (LDL-C) increased (P?<?0.05), as dietary vitamin E or C supplementation increased in diets. These results are in conflict with the previous hypothesis that antioxidants have a role in LDL-C removal from the blood or increasing HDL-C. Vitamin E was more effective than vitamin C in this case and if these results are confirmed by further studies, they may result to revision in researchers’ point of view about antioxidant especially in human medicine.  相似文献

The effects of vitamin E (dl-α-tocopheryl acetate) and selenium (Se; Na2-SeO3) on egg production, egg quality, and mineral content of egg yolk in Japanese quails reared under a low ambient temperature (6°C) were evaluated. Birds (n=300; 7 wk old) were randomly assigned to 12 treatment groups, 25 birds per group. The birds in a 3×2 factorial design received either three levels of vitamin E (125, 250, and 500 mg/kg diet) or two levels of selenium (0.1 or 0.2 mg/kg diet). After 2 wk on feed, six groups of the birds were maintained at 18°C (thermoneutral temperature [TN]), and the other half were acclimated over 3 d to a decreased environmental temperature of 6°C (cold stress [CS]). The performance, egg quality, and mineral content of egg yolk were not influenced by supplemental vitamin E and selenium in quails not exposed to cold stress (p≥0.09). Two hundred fifty and 500 mg vitamin E/kg diet compared with 125 mg/kg diet and higher dietary selenium inclusions (0.2 vs 0.1 mg/kg) resulted in a better body weight, egg production, and feed efficiency (p=0.01) in quails reared under CS. Similarly, egg weight, egg specific gravity, eggshell thickness, and Haugh unit were positively influenced with vitamin E (p=0.01) and selenium (p≤0.05) supplementation. Egg yolk concentrations of Zn, Fe, and Mn were higher with higher dietary vitamin E (p=0.01) and selenium (p=0.05). There was no interaction detected for parameters measured in the present study (p≥0.3). The results of the present study showed that a combination of 250 or 500 mg vitamin E and 0.2 mg selenium per kilogram of diet provides the greatest effects on performance and egg quality of Japanese quails reared under cold stress and suggest that such a supplementation can be considered as a protective management practice in Japanese quail diets to reduce the detrimental effects of cold stress.  相似文献

Liposomes from egg yolk lecithin and egg yolk lecithin/ganglioside are cytotoxic for Trypanosoma brucei and Trypanosoma congolense bloodstream forms in vitro. The trypanocidal effect is influenced by the liposome age and concentration. This effect is diminished in the presence of whole blood in vitro and could not be observed in vivo. Freeze-fractured parasite membrane showed an intramembranous particle aggregation after incubation with liposomes. Liposomes from egg yolk lecithin kill trypanosomes more rapidly than do liposomes from egg yolk lecithin/cholesterol.  相似文献

High resolution nuclear magnetic resonance spectra of native or protease-treated hen’s egg yolk plasma (very low density lipoproteins) were taken either in water or deuterated water; the protease-treated samples showed a sharpening of choline methyl proton signal of phospholipid, indicating the hindrance of the choline head-group rotation by the phospholipids in the native very low density lipoproteins. With both native and the protease-treated egg yolk plasma, elevated temperatue increased the signal intensity and produced line-sharpening of Q choline methyl protons and the — CH2-C-protons of the methylene group adjacent to the carboxyl group of esterified fatty acids, indicating prior restriction of mobility of these groups. Total extracted lipids of egg yolk plasma containing traces of chloroform, methanol and water (which keep the sample in one phase) also gave similar temperature dependence. Addition of water to the same sample and sonication resulted in the loss of temperature dependence. Frozen and thawed protease-treated egg yolk plasma also behaved in a similar manner. The absence of temperature dependence in these latter two samples is believed to be due to formation of bilayers of phospholipids following phase separation of triglycerides and phospholipids. The results support a model in which the lipoprotein particles of the egg yolk plasma have a lipid-core structure containing triglycerides in the centre with a monomolecular layer of lecithin at the surface, the polar heads of which are surrounded by proteins.
