Scanning electron microscopy was used to observe microbiological primary fouling of glass surfaces exposed in estuarine waters. Observations on clean glass, and glass treated with water-repellent coatings, showed that bacterial slimes adhered less strongly to the waterrepellent glass. An experiment using pure cultures of bacteria and latex particles showed that attached bacteria promoted the settlement of latex particles on the glass.  相似文献

Mild steel (MS), stainless steel (SS) and copper (Cu) test panels were immersed in the surface water of Dona Paula Bay over a period of 15 d. During the immersion period data on the hydrography, nutrients and suspended matter were also collected. The suspended matter and fouling products on the MS, SS and Cu panels were analysed for organic carbon (OC), organic nitrogen (ON), chlorophyll a (chl a), protein and carbohydrate concentration and composition, and the dry weight (DW) was recorded. Compared to suspended matter, the chemical and biochemical components of the fouling products showed strong temporal and substratum related differences. The microfouling biomass (as DW, OC, ON, chl a and protein) on all the test panels generally increased over the period of immersion. Carbohydrates were more abundant in the suspended matter whereas fouling products were enriched in proteins. The contribution of protein-carbon to the total carbon increased over the period of immersion for the microfouling products on MS and SS whilst it did not show a consistent trend on Cu. Whereas, the carbohydrate-carbon contribution to the total carbon increased for the fouling products on MS, it did not exhibit a particular pattern on SS or Cu over the period of immersion. Capillary gas chromatographic analysis showed the presence of glucose, galactose, mannose, arabinose, xylose fucose and ribose in both the fouling products and suspended matter. However, there were differences in the relative distribution of these monosaccharides in the suspended matter and the fouling products. Glucose was the most abundant monosaccharide, which showed strong temporal variations in suspended matter. In contrast, the wt % concentrations of individual monosaccharides showed large temporal differences for the fouling products, which were strongly influenced by the period of immersion and the type of test substratum. Glucose and fucose were relatively more abundant in the fouling products on SS and Cu, whilst glucose was the most abundant monosaccharide on MS. The monosaccharide and chemical composition data suggest strong temporal changes in the composition of the fouling products.  相似文献

Changes in microcirculation during low-intensity He-Ne laser irradiation were studied by measurement of blood vessels and lymphangions' diameter in anesthetized rat mesentery using method of videomicroscopy in vivo. We demonstrated that the red coherent light induced a significant increase in contractility of vessels' smooth muscle cells. The flow decreased in the mesenteric microvessels during H2O2-mediated oxidant stress was restored after He-Ne laser irradiation.  相似文献

AIMS: The efficacy of ultraviolet germicidal irradiation (UVGI) and the UVGI dose necessary to inactivate fungal spores on an agar surface for cultures of Aspergillus flavus and Aspergillus fumigatus were determined. METHODS AND RESULTS: A four-chambered UVGI testing unit with a 9-W, Phillips, low pressure, mercury UVGI lamp in each chamber was used in this study. An aperture was adjusted to provide 50, 100, 150, and 200 micro W/cm2 of uniform flux to the surfaces of the Petri dish, resulting in a total UVGI dose to the surface of the Petri dishes ranging from 12 to 96 mJ/cm2. The UVGI dose necessary to inactivate 90% of the A. flavus and A. fumigatus was 35 and 54 mJ/cm2, respectively. CONCLUSIONS: UVGI can be used to inactivate culturable fungal spores. Aspergillus flavus was more susceptible than A. fumigatus to UVGI. SIGNIFICANCE AND IMPACT OF THE STUDY: These results may not be directly correlated to the effect of UVGI on airborne fungal spores, but they indicate that current technology may not be efficacious as a supplement to ventilation unless it can provide higher doses of UVGI to kill spores traveling through the irradiated zone.  相似文献

The action of a low-intensity 632.8 nm-wavelength laser radiation on blood elements was studied. The degree of binding the fluorescence probes 1.8 ANS by blood proteins and cells was taken to be the index of the response to irradiation. The change of fluorescence of the oppositely charged probes indicates that a He-Ne laser radiation causes processes leading to a change of charge in proteins of a blood plasma with the negative charge decreased in most cases. The effect depends on individual characteristics of a biological object, on the exposure dose and the incubation time following the irradiation.  相似文献

An immunofluorescence technique was developed for the in situ identification of specific bacteria in marine microfouling films. Microorganisms adherent to glass plates after 30 days of immersion in a synthetic seawater system were cultured and classified by biochemical tests, flagellar arrangement, and the API 20E system. All isolates were gram-negative aerobic or facultative motile rods, predominantly Pseudomonas spp. Rabbit antisera to the five dominant organisms including Achromobacter spp., Comamonas terrigena, P. putrefaciens, a yellow-pigmented Pseudomonas sp., and Vibrio alginolyticus were prepared. These antisera were shown to be species specific in indirect immunofluorescence assays against a battery of 26 marine isolates from 14 bacterial species, with the exception of antisera to the Pseudomonas spp, which cross-reacted with each other but not with test bacteria of other genera. These immunofluorescent reagents enabled the in situ identification of all five bacterial species in microfouling films. Low-surface-energy test plates had smaller numbers of adherent bacteria in microfouling films than medium-surface-energy test plates, suggesting that the degree of microfouling may be influenced by the surface energy. In addition, the reagents could identify up to 39% of the attached bacteria in microfouling films spontaneously formed on steel plates in flow cells deployed in different areas of the Atlantic Ocean. The microbial composition of the ocean-formed films varied with the geographical area of their formation. The present results indicate that immunofluorescence techniques may provide a rapid and reliable means to identify, in situ, specific bacteria in marine microfouling films.  相似文献

Diatoms are a major component of the slime layers that form on artificial surfaces in marine environments. In this article, the role played by diatoms during the pioneering stages of colonization of three marine antifouling (AF) coatings, viz Intersmooth 360®, Super Yacht 800® and a fouling-release (FR) coating Intersleek 700®, was investigated. The study was conducted over three distinct seasons in two very different marine environments in Australia, ie temperate Williamstown, Victoria and tropical Cairns, Queensland. Diatom fouling occurred more rapidly on the FR coating Intersleek 700, compared to both biocidal AF paints. However, colonization by diatoms on all three coatings was generally slow during the 16-day study. Benthic diatoms do not subsist by floating around in the water column, rather they only gain the opportunity to colonize new surfaces when they either voluntarily release or are displaced from their benthic habitat, thereafter entering the water column where the opportunity to adhere to a new surface presents itself. However, once settled, fouling diatoms grow exponentially from the site of attachment, spreading out until they populate large areas of the surface. This mode of surface colonization correlates more with an ‘infection’ type, epidemiology model, a mechanism that accounts for the colonization of significant regions of the coating surface from a single fouling diatom cell, forming ‘clonal patches’. This is in comparison to the bacterial colonization of the surface, which exhibits far more rapid recruitment and growth of cells on the substratum surface. Therefore, it is hypothesized that fouling diatoms may be characterized more by their ability to adhere and grow on surfaces already modified by bacterial biofilms, rather than on their strength of adhesion. Cell morphology and the ability to avoid shear may also be an important factor.  相似文献

The ultrastructural organization of loach embryo cells (Misgurnus fossilis L.) at the stage of the first and the tenth embryo divisions was investigated under the influence of low intensity helium-neon laser irradiation of 5 min exposure. It was determined that the effect of laser irradiation led to ultrastructural changes: the shape of mitochondria and multivesicular bodies changes. The obtained results explain the possible mechanism of influence of low-intensity laser irradiation at the cellular level.  相似文献

The effects of ultraviolet light (UV) irradiation on the rate of DNA replication in synchronized Chinese hamster ovary (CHO) cells were investigated. A technique for measuring semiconservative DNA replication was employed that involved growing the cells in medium containing 5-bromodeoxyuridine and subsequently determining the amount of DNA that acquired hybrid buoyant density in CsCl density gradients. One of the advantages of this technique was that it allowed a characterization of the extent of DNA replication as well as rate after irradiation. It was found that while there was a dose-dependent reduction in the rate of DNA replication following UV-irradiation, doses of up to 10 J/m2 (which produce many dimers per replication) did not prevent the ultimate replication of the entire genome. Hence, we conclude that dimers cannot be absolute blocks to DNA replication. In order to account for the total genome replication observed, a mechanism must exist that allows genome replication between dimers. The degree of reduction in the rate of replication by UV was the same whether the cells were irradiated at the G1-S boundary or 1 h into S-phase. Previous work had shown that cells in early S-phase are considerably more sensitive to UV than cells at the G1-S boundary. Experiments specifically designed to test for reiterative replication showed that UV does not induce a second round of DNA replication within the same S-phase.  相似文献

The effect of ultraviolet irradiation on acid-soluble and neutral-salt-soluble calf-skin collagen was studied by chromatography, gel filtration, amino acid analysis and sedimentation of the sub-units, and the reaction kinetics of degradation were obtained from viscosity and optical rotation measurements. 2. It was demonstrated that, whereas the structure of neutral-salt-soluble calf-skin collagen may be represented by the formula (alpha(1))(2)alpha(2), the acid-soluble extract has the formula alpha(1).(alpha(2))(2). The acid-soluble collagen is also unusual in containing a large amount of a component that could be beta(22). 3. Ultraviolet irradiation causes the progressive degradation of the collagen molecule into smaller molecular fragments that subsequently lose their helical nature. The rate constants show that the denaturation of soluble collagens by ultraviolet irradiation is much slower, under the conditions used, than denaturation by heat or enzymes.  相似文献
