Rhizoctonia solani isolates used in this investigation were identified as anastomosis-4 (AG-40), collected from different localities from Assiut governorate in Egypt. Pathogenicity test of seven isolates of R. solani was evaluated on soybean Giza 111 cultivar under greenhouse conditions. All tested isolates were able to infect soybean plants causing root rot with different degrees of severities, isolate No. 1, 2 and 3 showed significantly highest root rot severity, while isolate No. 5 gave the lowest percentage of root rot rating. The sodium dodecyl sulphate polyacrylamide gel electrophoresis patterns were used to compare three isolates of R. solani. There are no variations among R. solani isolates except a few exceptions according to their protein patterns. DNA markers obtained from all isolates showed genetic similarity among different isolates obtained from different geographical regions barring few exceptions. Correlation between DNA patterns of R. solani isolates and their virulence was detected, but no correlation with anastomosis groups (AG). 相似文献

A study was carried out using simple laboratory techniques to examine the influence of the antagonistic isolates of Trichoderma harzianum, T. viride, Bacillius subtilis and Pseudomons flourescence and their culture filtrates on selected soil-borne root rot pathogens Rhizoctonia solani and Fusarium solani. Testing procedures were standardised using two different methods. The experiments were based on the principle of dual culture and agar diffusion techniques. The experiment involved the recording of the percentage of reduction in growth and inhibition zones formed by various filtrates of antagonistic culture growth. The results showed that the antagonists tested had the ability to reduce the linear growth of fungal pathogens. Also, the cultures filtrates of antagonists had antifungal activities by forming inhibition zones. Culture filtrates have shown a strong clear inhibition zone which increases in diameter as the incubation period of antagonists increases. This observation was related to the increase in the activity of chitinolytic enzymes as secondary metabolic compounds produced in growth media by prolonging the period of incubation. The study has proved that such enzymes can be effectively used for suppression of soil-borne pathogens and that it can evolve as a potential biocide. 相似文献

Abstract Field trials were conducted to determine the efficacy of the integration of treatments based on a bioagent (Trichoderma viride), botanical (leaf extract and cake of Pongamia glabra), fungicide (carboxin) and Rhizobium sp. in various modes of applications for the management of web blight (Rhizoctonia solani Kühn) and their influence on grain yield of urdbean (Vigna mungo [L.] Hepper). Modes of applications were soil treatment, seed treatment and foliar spray. Among 54 integrations evaluated, soil application of P. glabra cake (200 kg ha?1), seed treatment with integration of T. viride (2 g kg?1 seed), carboxin (1 g kg?1 seed) and Rhizobium sp. (25 g kg?1 seed), and foliar spray of P. glabra leaf extract (10%) were found superior as they increased seed germination by 31.6%, yield by 36.0% and reduced disease intensity by 94.8%, and also significantly enhanced plant height, root length and number of root nodules of urdbean. The same integration without corboxin was also effective and could be used in organic production of urdbean. The integration of any two modes of application was found superior over any one alone. 相似文献

Glomus mosseae and the two pod rot pathogens Fusarium solani and Rhizoctonia solani and subsequent effects on growth and yield of peanut (Arachis hypogaea L.) plants were investigated in a greenhouse over a 5-month period. At plant maturity, inoculation with F. solani and/or R. solani significantly reduced shoot and root dry weights, pegs and pod number and seed weight of peanut plants. In contrast, the
growth response and biomass of peanut plants inoculated with G. mosseae was significantly higher than that of non-mycorrhizal plants, both in the presence and absence of the pathogens. Plants inoculated
with G. mosseae had a lower incidence of root rot, decayed pods, and death than non-mycorrhizal ones. The pathogens either alone or in combination
reduced root colonization by the mycorrhizal fungus. Propagule numbers of each pathogen isolated from pod shell, seed, carpophore,
lower stem and root were significantly lower in mycorrhizal plants than in the non-mycorrhizal plants. Thus, G. mosseae protected peanut plants from infection by pod rot fungal pathogens.

Thirty-five strains ofTrichoderma viride andT. harzianum were screened for their antagonistic ability against the rice sheath blight pathogen,Rhizoctonia solani. The strains that inhibited/overgrew the phytopathogenic fungus were considered effective. Light microscopic studies showed the antagonism of the hyphae of effectiveTrichoderma strains towards their host hyphae. Chitinase activity ofTrichoderma culture filtrates was enhanced, when colloidal chitin was used as the sole carbon source, instead of glucose. Chitinase pattern differed among the four select strains. The chitinase isoforms are induced differentially by carbon sources. The effect of four bio-inoculants namely, Glomus aggregatum, Streptomyces sp., Bacillus subtilis and Trichoderma harzianum and a fungicide, Ridomil-mancozeb was evaluated on the biomass production and control of root rot and wilt disease under glasshouse conditions. Results showed that 21-day- prior inoculation with G. aggregatum was most effective where none of the treated plants produced disease symptoms and interestingly their growth was increased by 39.4%. The colonisation by G. aggregatum (>80%) also increased P concentration in shoot. While, similar treatment with Streptomyces sp., B. subtilis, T. harzianum and Ridomil-mancozeb individually failed to produce any significant effect over Rhizoctonia solani inoculated control, where all inoculated plant died, prematurely. The simultaneous and 3-day-post treatments of G. aggregatum were non-effective but simultaneous treatment with Streptomyces sp. produced 70% disease control, while B. subtilis and T. harzianum individually provided 50% control. Their effects were either better or at par when compared with the simultaneous treatment of Ridomil-mancozeb. In 3-day-post treatment, Streptomyces, B. subtilis and Ridomil-mancozeb individually provided 50% disease control, whereas T. harzianum was least effective as it could protect only 25% plants against infection. The results reveals that 15 days prior treatment of G. aggregatum can significantly controls the root rot and wilt disease of pyrethrum. Further, treatment of Streptomyces can also serve the next effective post infection control method. 相似文献

The efficacy of the newly developed seed dressing and soil application formulations of Trichoderma viride, T. virens and T. harzianum were evaluated individually and in combinations under pot and field experiments for the management of dry root rot (Rhizoctonia bataticola) of chickpea (Cicer arientinum). In pot experiments, T. harzianum based seed dressing formulation, Pusa 5SD, and soil application formulations, Pusa Biogranule 5 (PBG 5) and Pusa Biopellet 10G (PBP 10G), were found to be effective in reducing dry root rot incidence in chickpea and increasing the seed germination, shoot and root lengths of the crop. Under field experiments, a combination of soil application of T. harzianum based PBP 10G and seed treatment with Pusa 5SD+carboxin was found to be the best by providing the highest seed germination, shoot and root lengths and grain yield and the lowest dry root rot incidence in chickpea. 相似文献

Aims
The objective of this work was to design an amplified fragment length polymorphism (AFLP)‐derived specific primer for the detection of Fusarium solani aetiological agent of peanut brown root rot (PBRR) in plant material and soil.Methods and Results
Specific primers for the detection of the pathogen were designed based on an amplified region using AFLPs. The banding patterns by AFLPs showed that isolates from diseased roots were clearly distinguishable from others members of the F. solani species complex. Many bands were specific to F. solani PBRR, one of these fragments was selected and sequenced. Sequence obtained was used to develop specific PCR primers for the identification of pathogen in pure culture and in plant material and soil. Primer pair FS1/FS2 amplified a single DNA product of 175 bp. Other fungal isolates occurring in soil, included F. solani non‐PBRR, were not detected by these specific primers. The assay was effective for the detection of pathogen from diseased root and infected soils.Conclusions
The designed primers for F. solani causing PBRR can be used in a PCR diagnostic protocol to rapidly and reliably detect and identify this pathogen.Significance and Impact of the Study
These diagnostic PCR primers will aid the detection of F. solani causing PBRR in diseased root and natural infected soils. The method developed could be a helpful tool for epidemiological studies and to avoid the spread of this serious disease in new areas. 相似文献9.

Aim: To evaluate Ochrobactrum anthropi TRS‐2 isolated from tea rhizosphere and its talc based formulation for growth promotion and management of brown root rot disease of tea. Methods and Results: Ochrobactrum anthropi TRS‐2, isolated from tea rhizosphere could solubilize phosphate, produce siderophore and IAA in vitro and also exhibited antifungal activity against six test pathogens. Application of an aqueous suspension of O. anthropi to the rhizosphere of nursery grown tea seedlings of five varieties of tea (TV‐18, T‐17, HV‐39, S‐449, UP‐3 and) led to enhanced growth of the treated plants, as evidenced by increase in height, in the number of shoots and number of leaves per shoot. Treatment with O. anthropi also decreased brown root rot of tea, caused by Phellinus noxius. Multifold increase in activities of chitinase, β‐1,3‐glucanase, peroxidase and phenylalanine ammonia lyase in tea plants was observed on application of O. anthropi to soil followed by inoculation with P. noxius. A concomitant increase in accumulation of phenolics was also obtained. Further, talc based formulation of O. anthropi was prepared and its survival determined every month up to a period of 12 months. Ochrobactrum anthropi could survive in the formulation up to a period of 9 months with a concentration of 7·0 log10 CFU g?1, after which there was a decline. Talc formulation was as effective as aqueous suspensions in both plant growth promotion and disease suppression. Conclusion: Ochrobactrum anthropi, either in aqueous suspension or as talc formulation induced growth of tea plants and suppressed brown root rot disease. It induced defense responses in tea plants. Significance and Impact of the Study: Ochrobactrum anthropi and its talc based formulation can be considered as an addition to available plant growth promoting rhizobacteria (PGPR) currently being used for field application. The present study offers a scope of utilizing this bacterium for growth promotion and disease management which would help in reduction of the use of chemicals in tea plantations. 相似文献

Iris bulbs and Crocus corms were planted at two planting dates in sandy soil infested with Pythium spp. At monthly intervals during the growing season root rot infection was assessed over 3 consecutive years and disease development curves were predicted for both crops. The disease development was remarkably different for Iris and Crocus and the curve shape was determined by the crop rather than by the Pythium species. Planting date had a significant effect on disease development in both crops. No correlation was found between disease development and soil temperature. 相似文献

A 3-year study was conducted to evaluate the effectiveness of two disease-suppressive Streptomyces spp. to control sugar beet Rhizoctonia solani damping off under field conditions. Streptomyces seed treatments reduced seedling damping off in naturally (2005) and artificially (2006 and 2007) infested soils. All biocontrol agents provided better efficacy than Vitavax to control seedling damping-off. There were no significant differences among Streptomyces isolates. Isolate C increased plant stand by 19.5, 50.5 and 53.75% in 2005, 2006 and 2007, respectively. Evaluation of final harvest revealed that the root yield of the biocontrol agents increased compared to untreated control in these years. 相似文献

The spatial and temporal colonisation of tomato plants by Phytophthora cryptogea was studied in rockwool nutrient culture. Root growth and the distribution and progress of infection were measured on dissected root fragments obtained from a detailed destructive sampling of the substrate. Dissected fragments of root or roots and fibres were plated on BNPRA agar, a selective Phytophthora medium. Roots colonised all parts of the rockwool substrate 60 days after planting with the exception of surface marginal and central areas of the slab which had a lower solution content. Most root biomass occurred in and immediately beneath the original growing block. The distribution of P. cryptogea closely followed the pattern of root colonisation. An alternative, novel method for root analysis involved the dissolution of the mineral fibre and its formate bonding resin by digestion in 1 m H3PO4 for 45 min. Comparative recoveries of P. cryptogea from plated fragments of dissected root and fibre or comminuted samples of acid-released or dissected root showed that the acid treatment initially reduced the number of Phytophthora colonies in block and slab roots by 67% and 61% respectively. After 28 days, colony recovery from acid-released roots in the rockwool slab increased and was between 4% and 13% lower than from other plating methods. Since 1 m H3PO4 was lethal to zoospores and surface sporangia, the colonies recovered were interpreted as originating exclusively from root lesions. Root fresh weight of healthy and inoculated plants declined during the initial period of fruit formation. P. cryptogea infection led to a progressive reduction in root weight in the growing block and main slab and 28 days after inoculation, was approximately 50% of the controls. The acid digestion of rockwool fibre is proposed as a new approach to the problem of root and pathogen analysis in this substrate. 相似文献

The effect of root-zone temperature on Phytophthora cryptogea root rot was studied in tomato cv. Counter grown under winter and summer conditions in rockwool culture. A nutrient temperature of 25°C resulted in increased root initiation and growth, higher in winter-grown than in summer-grown plants. Rhizosphere zoospore populations were greatly reduced at 25°C and above. Growth of P. cryptogea in vitro was optimal between 20°C and 25°C and completely suppressed at 30°C. Encystment was enhanced by increased temperatures above 20°C. Zoospore release in vitro occurred in cultures maintained at constant temperatures in the absence of the normal chilling stimulus. Optimal release was at 10°C; no zoospores were released at 30°C. Inoculated, winter-grown tomato plants maintained at 15°C developed acute aerial symptoms and died after 21 days. Comparable plants grown at a root-zone temperature of 25°C remained symptomless for the 3-months duration of the experiment. Summer-grown infected plants at the higher root temperature wilted but did not die. Enhanced temperature was ineffective as a curative treatment in summer-grown plants with established infection. Aerial symptoms of Phytophthora infection are seen as a function of the net amount of available healthy root. With high root zone temperatures this is determined by new root production and decreased inoculum and infection. 相似文献

The effect of root temperature on growth and yield of rockwool-grown tomato plants infected with Phytophthora cryptogea was investigated. Measurements of shoot and root growth were taken at high (25oC) and low (15oC) root temperatures during the generative phase of growth. The growth of roots of healthy and P. cryptogea-infected tomato plants in rockwool blocks was higher in plants grown with roots at 25oC than at 15oC after 60 days and a similar effect was found in slabs after 98 days. Under sub-optimal conditions for growth the disease became severe when root temperatures were low. Growth of roots was greatest when roots were maintained at a high temperature in combination with an ambient air temperature of c. 15oC and the response was greater in cv. Counter than cvs Calypso and Marathon. Water-soluble carbohydrates of roots were higher in those produced in blocks than slabs and were reduced by infection compared to healthy plants with roots at 15oC and 25oC. Reduced transpiration rates were found 17 days after inoculation in symptomless plants grown at a root temperature of 25oC. Infection, regardless of the temperature of the roots or cultivar, led to reduced stem growth. The plants grown at 25oC were taller than those with a root temperature of 15oC. After 9 wk of harvest, the cumulative fruit yields in infected cvs Counter and Calypso grown at 25oC were comparable to that in healthy plants grown at either temperature and cumulative fruit numbers followed a similar pattern. High root temperatures led to delayed fruit ripening between weeks 3–10 and a larger number of unripe fruit. The weight of unripe fruit from infected plants grown at 25oC at the terminal harvest was higher than from healthy plants with roots maintained at 15oC. 相似文献

Eight fungicides (benodanil, carboxin, cyproconazole, fenpropimorph, fur-mecyclox, iprodione, pencycuron and tolclofos-methyl) were evaluated, under growth chamber conditions, as seed treatments against pre-emergence damping-off and post-emergence seedling root rot in six Brassica species. Five cultivars of B. rapa, four cultivars of B. juncea, four cultivars of B. napus and one cultivar/ strain from each of B. carinata, B. nigra and B. oleracea were grown in soilless mix infested with an isolate of Rhizoctonia solani AG-2-1. B. nigra and B. juncea were considerably less susceptible to R. solani than the four other species. Cyproconazole at 0.05-0.1 g a.i./kg seed and the other fungicides at 2–4 g a.i./ kg seed provided almost complete control of pre-emergence damping-off in most Brassica species and their cultivars. Their efficacy varied against the post-emergence seedling root rot. Furmecyclox, iprodione, tolclofos-methyl and pencycuron consistently gave good control of seedling root rot in all Brassica species and their cultivars. Benodanil and fenpropimorph provided moderate control, and carboxin and cyproconazole gave poor control against root rot. Efficacy of carboxin, cyproconazole, benodanil and fenopropimorph against seedling root rot varied significantly (P ≤ 0.05) among cultivars within a Brassica species. 相似文献

Assessment of laboratory methods for evaluating cassava genotypes for resistance to root rot disease
Field evaluation of six cassava genotypes for resistance to root rot disease was compared with three rapid laboratory methods (whole root inoculation, root slice inoculation, and stem inoculation) for resistance screening. Both the field evaluation and the three laboratory methods separated the varieties into resistant and susceptible groups. Genotypes 30572 and 91/02324 were resistant while 92/0247, 92/0057 and TME-1 were susceptible. One genotype (30001) was not consistent in its reaction between field evaluation and laboratory assays. In the laboratory assays with three fungal pathogens, different pathogens varied in their levels of virulence on host genotypes. With the most virulent pathogen (Botryodiplodia theobromae), the majority of the genotypes reacted in the same way across trials with the root slice and whole root assays. Due to the good correlation between the whole root assay and the field results, we recommend this for the routine assessment of cassava resistance to root rot disease and for the analysis of virulence of pathogen isolates. However, because of the advantages in terms of economy of labour, space, time, quantity of root and inoculum required, the root slice assay could be used for the preliminary screening of large cassava accessions. Abstract Although Rhizoctonia solani is a cosmopolitan soilborne pathogen, the genus includes isolates with different pathogenicity ranging from high virulence to avirulence. The biocontrol strain Pseudomonas fluorescens P190r and the arbuscular mycorrhizal (AM) fungus Glomus mosseae BEG12 were inoculated alone or in combination in tomato plants infested by the mildly virulent pathogen R. solani #235. Plant growth as well as root morphometric and topological parameters were evaluated. The infection of R. solani was significantly reduced by all the combinations of the beneficial microorganisms. Root systems of R. solani‐infected plants were weakly developed but highly branched with a herring‐bone pattern, while those inoculated with the AM fungus, alone or in combination with the bacterial strain, were longer and more developed, and displayed a dichotomous pattern. The interactions among these three microorganisms affected plant growth and root architecture of tomato plants. 相似文献

Abstract Helminthosporium tetramera isolates were isolated and identified from root of wheat and rice crops and their aggressiveness was studied using aggressiveness analysis in the greenhouse. Isolates of H. tetramera were genetically characterized using Random Amplified Polymorphism DNA (RAPD). The investigations were based on two surveys of wheat and one survey of rice. In root aggressiveness analysis, the overall number of aggressive isolates was higher on wheat compared to rice. In the foliar aggressiveness test, the number of non-aggressive isolates was almost the same on wheat and rice varieties. RAPD was used to study the polymorphism and genetic variation within the population of H. tetramera that established to study correlation between taxonomical, aggressiveness and genetical characters. The H. tetramera tree is constructed based on the pattern of bands. This study highlighted the correlation between morphological, aggressiveness and genetic variations of H. tetramera. 相似文献

Double-stranded (ds)RNA of various types was detected in 65 (21.8%) of 298 isolates from vegetative hyphae of Rosellinia necatrix by electrophoresis, but dsRNA was not detected from 39 ascosporic isolates. There were 45 distinct dsRNA profiles in the
65 isolates: they varied in the number of electrophoretic bands from 1 to 12 and in size from less than 1000 bp to more than
10 kbp. Each dsRNA profile was unique to each locality. dsRNAs having the same profiles were restricted to isolates of the
same mycelial compatibility groups (MCG) from the same trees, with an exception where different profiles were detected in
different isolates of the same MCGs.
