Electroblotting method employing a semidry blotting apparatus for the subsequent protein microsequence analysis (Hirano, 1987) was improved. This method is convenient and allows rapid and efficient transfer of the proteins from a polyacrylamide gel (1 mm thick) onto the Polybrene-coated glass-fiber sheet or polyvinylidene difluoride membrane filter in only 20 min. The electroblotted proteins could be sequenced directly with the gas-phase protein sequencer at a 20-pmole level. This method was applied to the sequence analysis of winged bean seed proteins. A portion of the crude extracts from only one-twentieth of a seed of the winged bean was separated by two-dimensional polyacrylamide gel electrophoresis and electroblotted, and the N-terminal amino acid sequences of the blotted proteins were analyzed. The sequences of about 60% of the blotted major proteins, including nine Kunitz trypsin inhibitor-like proteins with heterogeneity in the N-terminal sequences, a protein that has a homologous sequence to the leghaemoglobin, nitrogen-fixing root nodule-specific protein, and a soybean basic 7S globulin-like protein could be easily identified. 相似文献

The moisture, lipids and fatty acid composition of developing winged bean (Psophocarpus tetragonolobus) seeds were studied. The moisture content decreased steadily as the seeds matured. The lipid content increased gradually and reached a maximum ca 6 weeks after flowering (WAF). In the early stage (2 WAF) of the developing seeds there were more polar lipids (glycolipids and phospholipids) than neutral lipids but, as the seeds developed, neutral lipids gradually accumulated while the polar lipids decreased until 6 WAF. Thereafter, both the neutral lipid and polar lipid levels remained little changed. The amounts of palmitic and stearic acids decreased, but the level of behenic acid increased as the seeds matured. On the other hand, the oleic acid content increased while that of linolenic acid decreased rapidly as the seeds matured. The concentration of linoleic acid, however, fluctuated during the development of the seeds. 相似文献

Recent evidence concerning chromosome numbers of previously unavailable species in the genus and observational evidence of false rust (Synchytrium psophocarpi (Rac.) Gäumann) onP. grandiflorus Wilczek in Zaïre is discussed. The role of some Iranian strains of Pseudomonas spp. as biocontrol agents against Meloidogyne incognita and their ability to colonise pistachio roots was investigated. The results of in vitro experiments indicated that all tested bacteria produced significant suppression of M. incognita and showed that all strains were able to kill M. incognita juveniles with strain VUPf428 achieving about 99% mortality at 72 h. The results of in vivo treatments indicated that the best strains that could build high populations in soil infested with nematodes were VUPf5, VUPf52 and VUPf205. These isolates also caused highest reduction in galling and nematode multiplication in a greenhouse test although all strains native to Iran could colonise pistachio roots in pots. Some strains could produce secondary metabolites such as siderophores, proteases and volatile metabolites at high population levels. 相似文献

Examination under the light muscope of thin (2 μm) serial sections of syncytia embedded in epoxy resin reveals a close association between these structures and adjacent cells. Sometimes these cells protrude into the cytoplasm of a syncytium but at other times their walls and those of syncytia break down and permit cytoplasmic contact. These results support the hypothesis that syncytia are formed partly by incorporation of cells whose walls have dissolved rather than by expansion of a single cell. Onion is an important indispensable item in every kitchen as condiment and vegetable, hence commands an extensive internal market. The green leaves and immature and mature bulbs are eaten raw or used in preparation of vegetables. Among the various pests and diseases associated with onion, the root knot nematode (Meloidogyne incognita) had proved itself as an important limiting factor for successful cultivation of this crop. Recently root-knot nematode was observed to cause serious losses to green onion crop grown mostly in the field located near the Jamuna & Ganga river belt of Allahabad and the surrounding area in U.P. Differential host test was done to confirm the root knot disease in onion. of M. javanica larvae, indicating that active nematicidal compounds are intermediary in polarity. In field experiments, biocontrol fungus
and bacterium significantly suppressed soilborne root-infecting fungi including Macrophomina phaseolina, Fusarium oxysporum, Fusarium solani, Rhizoctonia solani and Meloidogyne javanica, the root-knot nematode. P. lilacinus parasitized eggs and female of M. javanica and this parasitism was not significantly influenced in the presence of P. aeruginosa. P. aeruginosa was reisolated from the inner root tissues of tomato, whereas P. lilacinusdid not colonize tomato roots.

Abundance, genetic diversity and parasitic ability in the facultative nematode parasite Pochonia chlamydosporia var. chlamydosporia were compared in soils from two sites in Portugal under long-term tomato cultivation where root-knot nematodes (Meloidogyne sp.) were present. Fungal abundance assessed by selective agar or real-time quantitative PCR with specific primers was similar in both soils. PCR fingerprinting of isolates with ERIC primers indicated that the dominant P. c. var. chlamydosporia biotypes (profiles A and B) in both soils were very closely related, although a second biotype (profile C) was detected in one soil. When tomato plants infected with M. incognita were grown in the two soils, only profiles A and B were recovered from eggs. Primers based on polymorphisms in vcp1 demonstrated that isolates with profiles A and B were likely to prefer root-knot nematodes, whereas profile C preferred cyst nematodes. In the soil containing profiles A, B and C, egg parasitism by P. chlamydosporia was estimated at 1% using water agar plates with antibiotics but fewer than 0.2% of M. incognita eggs were shown to be infected with P. c. var. chlamydosporia when using species-specific β-tubulin-PCR primers. In contrast, the soil containing only profile B showed 22% egg parasitism on water agar plates and more than 2.5% of eggs were confirmed as P. c. var. chlamydosporia by species-specific β-tubulin-PCR primers. The results, which reveal limited diversity within the fungus at the two sites, are discussed in relation to biological control of plant-parasitic nematodes. 相似文献

Thirty-five nematode species belonging to 19 genera were extracted from 432 soil and root samples collected in 19 districts in six provinces in the North Central Coast, Central Highlands and Phu Quoc Island. Based on our study, the predominant nematode species associated with black pepper plants in Vietnam is Meloidogyne incognita: almost every soil and root sample examined during our study was infected with this root-knot nematode species. No difference in frequency of occurrence of M. incognita among the three agro-ecological regions surveyed was observed. However, the root population density of M. incognita in black pepper plants was on average about five times higher in the North Central Coast and Central Highlands than in Phu Quoc Island. Large differences in root population density of M. incognita on black pepper plants were observed in different districts within the same agro-ecological region. Black pepper roots infected with M. incognita showed the same type of galls as described in the literature. The percentage of root galling averaged about 40% in all the three agro-ecological regions. The percentage of black pepper plants with yellow leaves was on average about 20–25% in the North Central Coast and Central Highlands, which was somewhat higher compared to Phu Quoc Island (16.9%). Large differences in percentage of black pepper plants with yellow leaves were observed in different districts within the same agro-ecological region. The same type of yellow leaves as described in the literature for black pepper plants infected with Meloidogyne species was observed in our study. In general, a positive relationship between the soil and root population densities of M. incognita on black pepper plants and percentage of plants with yellow leaves was observed. However, in one district (Quang Tri) a relatively low nematode population density was associated with a high percentage of plants with yellow leaves. Five nematode species were recorded for the first time on black pepper plants in Vietnam. Our study reconfirms previous reports that Radopholus similis does not occur in Vietnam. 相似文献

Effect of different level of inoculums of root knot nematode on predacity of Arthrobotrys oligospora
Arthrobotrys oligospora is one of the most important predaceous fungi occurring widely in different types of soil. This fungus produces the hyphal bails and network compound. This hyphal nets as trapping structure which may be one dimensional to two or three dimensional and are formed through repeated hyphal anastomosis, which capture nematodes either through the adhesive material present on the surface of hyphal nets or due to physical entanglement. When moving nematodes pass through such hyphal bails, they are captured by sticky substances present on the bails. Such nematodes struggle violently to escape the capture but they are usually entangled at several places of their body. They absorb the nutrients and finally kill the nematodes. Irrespective of the different levels of inoculums of root knot nematode, capturing of nematodes increased with increasing period of incubation. After 5 days of inoculation, 93–97.33% nematodes were captured irrespective of the nematode number. 相似文献

During a survey of nurseries and plantations of black pepper plants in Quang Tri province in Vietnam during the rainy season of 2007, nine fungal taxa were isolated from the roots of the black pepper plants. Fusarium solani was found in about one out of four black pepper root samples examined but not in the nurseries and also not from black pepper plants younger than five years growing in plantations. Since in these nurseries about one out of two black pepper plants examined had yellow leaves, this observation suggests that another pathogen must be the initial cause of the yellowing of the leaves. A likely pathogenic candidate is M. incognita which was extracted from every single black pepper plant examined in the nurseries. During the same survey, we also observed that F. solani was not isolated from the roots of black pepper plants that did not had yellow leaves and that the percentage of black pepper plants with yellow leaves increased with increased frequency of occurrence of F. solani. This observation indicates that F. solani plays a role in the yellowing of the leaves of black pepper plants in a later stage of the development of the plants. The results of a greenhouse experiment showed the negative effects inoculation with M. incognita alone or in combination with F. solani may have on the percentage of black pepper plants with yellow leaves and on plant growth. No effect of inoculation with F. solani before, at the same time, or two weeks after inoculation with M. incognita on root galling and nematode reproduction was observed. 相似文献

Root gravitropism may be an important element of plant response to phosphorus availability because it determines root foraging in fertile topsoil horizons, and thereby phosphorus acquisition. In this study we seek to test this hypothesis in both two dimensional paper growth pouch and three-dimensional solid media of sand and soil cultures. Five common bean (Phaseolus vulgaris L.) genotypes with contrasting adaptation to low phosphorus availability were evaluated in growth pouches over 6 days of growth, and in sand culture and soil culture over 4 weeks of growth. In all three media, phosphorus availability regulated the gravitropic response of basal roots in a genotype-dependent manner. In pouches, sand, and soil, the phosphorus-inefficient genotype DOR 364 had deeper roots with phosphorus stress, whereas the phosphorus-efficient genotype G19833 responded to phosphorus stress by producing shallower roots. Genotypes were most responsive to phosphorus stress in sand culture, where relative root allocation to the 0–3- and 3–6-cm horizons increased 50% with phosphorus stress, and varied 300% (3–6 cm) to 500% (0–3 cm) among genotypes. Our results indicate that (1) phosphorus availability regulates root gravitropic growth in both paper and solid media, (2) responses observed in young seedlings continue throughout vegetative growth, (3) the response of root gravitropism to phosphorus availability varies among genotypes, and (4) genotypic adaptation to low phosphorus availability is correlated with the ability to allocate roots to shallow soil horizons under phosphorus stress. 相似文献

Measuring the completeness of survey inventories created by citizen-science initiatives can identify the strengths and shortfalls in our knowledge of where species occur geographically. Here, we use occurrence information from eBird to measure the survey completeness of the world's birds in this database at three temporal resolutions and four spatial resolutions across the annual cycle during the period 2002 to 2018. Approximately 84% of the earth's terrestrial surface contained bird occurrence information with the greatest concentrations occurring in North America, Europe, India, Australia and New Zealand. The largest regions with low levels of survey completeness were located in central South America, northern and central Africa, and northern Asia. Across spatial and temporal resolutions, survey completeness in regions with occurrence information was 55–74% on average, with the highest values occurring at coarser temporal and coarser spatial resolutions and during spring migration within temperate and boreal regions. Across spatial and temporal resolutions, survey completeness exceeded 90% within ca 4–14% of the earth's terrestrial surface. Survey completeness increased globally from 2002 to 2018 across all months of the year at a rate of ca 3% yr–1. The slowest gains occurred in Africa and in montane regions, and the most rapid gains occurred in India and in tropical forests after 2012. Thus, occurrence information from a global citizen-science program for a charismatic and well-studied taxon was geographically broad but contained heterogeneous patterns of survey completeness that were strongly influenced by temporal and especially spatial resolution. Our results identify regions where the application of additional effort would address current knowledge shortfalls, and regions where the maintenance of existing effort would benefit long-term monitoring efforts. Our findings highlight the potential of citizen science initiatives to further our knowledge of where species occur across space and time, information whose applications under global change will likely increase. 相似文献

Meloidoyne incognita (root-knot nematode) and Fusarium solani (root-rot pathogen) were the common soil-borne pathogens and cause severe damage to bean plants in newly reclaimed sandy soil in Nubaryia district, Behera Governorate, Egypt. The antagonistic effects of Trichoderma album and Trichoderma viride as well as three commercial products namely Rhizo-N® (Bacillus subtilis), Bio-Arc® 6% (Bacillus megaterium) and Bio-Zeid® 2.5% (T. album) were tested against M. incognita and F. solani under naturally infected field conditions. T. album and T. viride highly reduced the frequency (%) population of pathogenic fungi such as Fusarium spp., F. solani and Rhizoctonia spp., than the commercial products. Results indicated that all the tested bio-control agents reduced, significantly, the nematode criteria as evidenced by the number of juvenile (J2) in soil and number of galls and egg masses on roots of common bean and Fusarium root-rot incidence (%). Rhizo-N® highly reduced the number of J2 in soil, while T. album was the best in reducing the number of galls and egg masses in roots. The bio-control agents also increased the plant growth parameters of common bean plants i.e. plant height, plant weight, branch no./plant, pods no./plant, pod weight/plant, pod weight, seeds no./plant, fresh seeds weight/pod, dry seeds weight/pod and dry weight of 100 seeds. 相似文献

The relationship between ethylene and adventitious root formation in mung bean hypocotyl cuttings was studied.Ethephon, an ethylene-releasing compound, at 5 x 10 -5 M increased root number and root dry weight on hypo-cotyl cuttings. When ethephon was applied to hypocotyl at different times after excision, there were two effectivetimes for root production i.e. between 06 h and 18-24 h. These two time periods correspond to the induction phase and the late initiation phase of root development, respectively. After excision, three peaks of ethylene productionwere observed. The first peak commencing at 6 h started the sequence of reactions leading root formation, the second peak appearing at 12 h coincided with the beginning of the increase of the IAA level during primordia initiation, and the third peak showing at 48 h played a role in root differentiation and growth. Ethylene stimulated rooting by enhancing the increase in auxins. Thus it appears that the IAA-induced ethylene production may be a factor involved in the stimulation of adventitious root formation. 相似文献

AbstractAn experiment was conducted to study the effects of interaction between Meloidogyne incognita and Fusarium solani on plant length, fresh and dry weights, number of pods, chlorophyll, carotenoid, nitrogen and phosphorus contents and nitrate reductase activity in lentil plants. The results reveal a maximum damage occurring in all the plant growth, biochemical and nutrient parameters, in plants inoculated with M. incognita 10 days prior to F. solani (Mi?→?Fs). This was followed by simultaneous (Mi?+?Fs) inoculations, fungus inoculation 10 days prior to nematode (Fs?→?Mi), M. incognita alone and F. solani alone treatments. BACKGROUND AND AIMS: Although Lythrum salicaria (purple loosestrife) was introduced to North America from Europe in the early 1800s, it did not become invasive until the 1930s. Whether hybridization with L. alatum (winged loosestrife) could have played a role in its ultimate spread was tested. METHODS: Six diagnostic morphological traits (flower number per axil, leaf placement, calyx pubescence, style type, plant height and leaf shape) were surveyed in 30 populations of Lythrum across eastern North America. Patterns of AFLP variation were also evaluated using five primer pairs in a 'global screen' of the same North American populations of L. salicaria and L. alatum described above, in L. salicaria from 11 European populations located in Germany, England, Ireland, Austria and Finland, and in six L. salicaria cultivars. KEY RESULTS: All of the North American L. salicaria populations had individuals with alternate leaf placement and 1-2 flowers per leaf axil, which have not been described in Eurasian L. salicaria but predominate in North American L. alatum. In addition, two L. salicaria populations were intermediate in height and leaf ratio between the typical L. salicaria and L. alatum populations in their native fields and when grown in a common greenhouse. In screens of variation patterns using 279 AFLPs, only two fragments were found that clearly supported introgression from L. alatum to L. salicaria. CONCLUSIONS: The evidence indicates that L. salicaria may have hybridized with L. alatum, but if so, only a small fraction of L. alatum genes have been retained in the genome of L. salicaria. This is unlikely to have led to a dramatic adaptive shift unless the introgression of a few key genes into L. salicaria stimulated a genomic reorganization. It is more likely that crossing among genotypes of L. salicaria from multiple introductions provided the necessary variability for new adaptations to arise. 相似文献
