Fusarium root rot in row crops is typically managed by cultural practices and fungicide seed treatments. Biological control using microbial agents is another option but needs further development for improved disease management. Screening to identify biocontrol agents are crucial. However, relationships among the steps and how to improve the screening process are unresolved questions. Strains of Burkholderia (4), Bacillus (5) and Trichoderma (26) were studied in vitro against six Fusarium pathogens. All the bacteria and five selected Trichoderma strains were tested in planta in the greenhouse against diseases of Fusarium graminearum and Fusarium oxysporum. Burkholderia ambifaria C628, Bacillus simplex R180, and all Trichoderma isolates showed high reduction in disease levels in corn, soybean and wheat, ranging from 16 to 63%. Responses of the biocontrol agents during in vitro and in planta screening did not always correlate. In vitro and in planta tests should be considered independently in selecting biocontrol candidates. 相似文献

内切葡聚糖酶的化学修饰研究表明:色氨酸残基可能位于活性位点,与底物结合有关.荧光光谱测定指出该酶的荧光几乎都来自色氨酸残基,酶分子中色氨酸微环境对pH变化非常敏感,降低pH导致了酶分子构象发生了较大变化,配基结合使酶分子色氨酸微环境产生了改变,引发了与pH诱导不同的构象变化. 相似文献

木霉菌(Trichoderma)作为一种重要的生防因子,可以产生几丁质酶降解植物的多种病原真菌细胞壁。利用低能离子束注入木霉菌使其产生变异,再通过初筛选和复筛选两个过程,获得T90_1木霉菌株,并用草莓灰霉病菌(Botrytis cinerea Pers.)来检验T90_1防治真菌病害的能力。发现该菌株通过侵染、缠绕等多种重寄生方式,并分泌降解病原真菌细胞壁的物质,使病原菌原生质外渗,改变细胞内有序的代谢状况,从而抑制或杀死病原菌。初步揭示该菌株抗真菌的相关机制。 相似文献

Relative effectiveness of seven different application methods of five native bacterial and fungal biocontrol agents (BCAs) and one fungicide (azoxystrobin) was evaluated against Rhizoctonia solani, under pot and field conditions on rice cultivar Pusa Sugandha-5. Plants grown in pots infected with R. solani suffered a 30%–49% decrease in plant growth and yield of grain. However, treatment with BCAs reduced the adverse effect of the pathogen but significantly varied with the treatment schemes. Amongst the treatments, soil application (SA) at 20 + 40 days after planting (DAP) followed by foliar application (FA) at 60 DAP was recorded as most efficacious, and reduced the severity of disease by 42–68%, resulting in a 21–36% plant growth promotion and yield enhancement. Treatment comprising SA 20 + FA 40 DAP was next in effectiveness but statistically equal to seed priming (SP) + SA 40 DAP of treatment. Amongst the BCAs, Pseudomonas putida was shown to be the most efficient, trailed by Trichoderma harzianum, P. fluorescens, T. viride and Bacillus subtilis. Field trials under naturally infested fields have also validated the effectiveness of P. putida. The SA 20 + 40 + FA 60 DAP with P. putida and T. harzianum were found quite effective and decreased the disease severity and incidence (40–81%), and improved the grain yield (42–72%). Relatively lower ShB control was recorded with SA 20 + FA 60 DAP, however, it was statistically at par with SA 20 DAP treatment and equal to SA 20 + FA 40 DAP. 相似文献

In the current study, 18 Trichoderma spp. isolates were obtained from different provinces in central and northern regions of Turkey. The ability of nine selected isolates to suppress the colonisation of roots by P. betae and the multiplication of BNYVV in sugar beet roots under controlled conditions were tested. Roots of seedlings growing in the P. betae-BNYVV-infested soil were analysed by enzyme-linked immunosorbent assay to test for the presence of BNYVV and checked microscopically for the density of cystosori of P. betae. The numbers of P. betae resting spores in cystosori for each treatment were counted using a light microscope. Except for isolates Tr-1 and Tr-5, the effect of selected Trichoderma isolates on suppressing multiplication of BNYVV varied between 4 and 53%. The total number of resting spores in the roots varied between 14.4 and 25.1 for the different Trichoderma spp. treatments. The lowest number of resting spores in clusters was recorded in T. harzianum Tr-8. In addition, the shapes of resting spores were not normal in the Tr-8 treatments. The cystosori from this treatment were also abnormally dark in colour and had deformed walls. 相似文献

A broad spectrum of fungal antagonists was evaluated as potential biocontrol agents (BCAs) against the soil-borne pathogen Rhizoctonia solani using a new combination of in vitro and in vivo assays. The in vitro characterisation of diverse parameters including the ability to parasitise mycelium and to inhibit the germination of Rhizoctonia sclerotia at different temperatures resulted in the selection of six potential fungal antagonists. These were genotypically characterised by their BOX-PCR fingerprints, and identified as Trichoderma reesei and T. viride by partial 18S rDNA sequencing. When potato sprouts were treated with Trichoderma, all isolates significantly reduced the incidence of Rhizoctonia symptoms. Evaluated under growth chamber conditions, the selected Trichoderma isolates either partly or completely controlled the dry mass loss of lettuce caused by R. solani. Furthermore, the antagonistic Trichoderma strains were active under field conditions. To analyse the effect of Trichoderma treatment on indigenous root-associated microbial communities, we performed a DNA-dependent SSCP (Single-Strand Conformation Polymorphism) analysis of 16S rDNA/ITS sequences. In this first assessment study for Trichoderma it was shown that the pathogen and the vegetation time had much more influence on the composition of the microbiota than the BCA treatment. Xanthomonas campestris pv. mori causes bacterial leaf spot (BLS) of mulberry (Morus spp.) worldwide. Twenty mulberry lines with contrasting response to BLS showed significant (p?<?0.01) variation between resistant and susceptible groups for stomata frequency (SF), trichome density (TD), total soluble sugar (TSS) and polyphenol (PP) content, while non-significant for chlorophyll content. Heritability (broad-sense) estimates obtained from the variance component analysis were high (≥0.72) for these traits. The disease severity index values of the lines were significantly negatively correlated with TD (r?=??0.66; p?<?0.01), TSS (r?=??0.73; p?<?0.01) and PP (r?=??0.61; p?<?0.01), while positively correlated with SF (r?=?0.65; p?<?0.01). The cluster analysis was based on highly significant four variables and DSI values, and classified the lines into two major clusters. The resistant lines that come under cluster-I had high TD, TSS and PP, while those in susceptible category had high SF in cluster-II. These findings suggest that direct selection for SF, TD, TSS and PP will be effective for development of host resistance against BLS in mulberry. 相似文献

从华南地区采集土样,采用愈创木酚平板筛选产漆酶菌株,获得了一株短周期产漆酶的小型丝状真菌。通过观察菌落特征、生长情况以及显微镜下菌丝和孢子的形态,初步鉴定该菌株为木霉属的一个种(Trichodermaspp、),命名为木霉LaTr01菌株。通过单因素方法研究该菌产漆酶的发酵条件,结果表明:LaTr01的产酶培养基以麦芽糖为最佳碳源;以酵母提取物为最适氮源;培养24h后加入Cu“比培养开始加入Cu ^2+LaTr01产酶活高出约1倍。采用麦芽糖、酵母提取物、Cu^2+浓度L9(3^3)的正交试验优化漆酶发酵条件,结果表明,氮源是影响该菌产漆酶的最重要因素,碳源次之,Cu^+浓度影响较小;LaTr01菌株产生漆酶的最佳条件为:5g/L酵母提取物、20g/L麦芽糖、1.5mmol/LCu^2+,Cu^2+加入时间为培养24h后。在优化的培养条件下,该菌酶活可达480.556U/L。 相似文献

Double auxotrophic and morphological mutants of Trichoderma pseudokoningii Rifai were fused by anastomosis and by protoplast fusion. The recovery of recombinants from heterokaryons on different selective media and from heterokaryotic colonies indicated the occurrence of parasexual events. Prototrophic colonies growing on minimal medium produced binucleate spores, green in colour, revealing a non-autonomous system for conidial pigmentation. Recombinants were obtained from these dikaryotic colonies suggesting the occurrence of a highly unstable diploid phase. 相似文献

Suppression of root rot diseases of ornamental plants is a potential benefit of formulating soilless container media with compost. A green compost (green nursery compost, GNC), obtained by a circular-economy approach from residues of pruning of woody plants and grass clippings during the nursery activities was analysed for its suppressiveness of root rot diseases using lavender. To this end, a bioassay was develop by formulating potting mixes containing GNC with two rates of peat substitution (25% and 50%) and infested with the root rot pathogens Sclerotinia sclerotiorum, Rhizoctonia solani and Phytophthora nicotianae. Contrasting results were obtained by using both substrates with a significant reduction of root rot by S. sclerotiorum, no effect on the containment of that by P. nicotianae, and an increase of symptoms caused by R. solani. The specific suppressiveness observed may be attributed to the colonisation of compost by specific groups of antagonistic microorganisms. This hypothesis was investigated by the analysis of culturable fungal community, which resulted in the isolation of Trichoderma harzianum and T. atroviride as preponderant fungal species. Trichoderma representative isolates exerted in vitro antagonistic activities against the target pathogens with varying efficiencies indicating the employment of multiple complementary mechanisms, which may have contributed to the observed specific suppressiveness. Both substrates containing GNC resulted suitable for nursery cultivation of lavender, showing a growth performance similar to that obtained with peat-based substrate. Present results indicate that on-farm compost is a suitable component of mixed-peat substrates capable to support plant growth and provide specific disease suppression. 相似文献

Bean anthracnose caused by Colletotrichum lindemuthianum is a serious seed borne disease. For devising an effective management strategy, the efficacy of different bioagents, viz. Trichoderma viride, Trichoderma harzianum, Trichoderma hamatum and Gliocladium virens conducted under in vitro and in vivo conditions revealed maximum inhibition of mycelial growth in dual culture (59.48%) and inverted plate (55.98%) with T. viride. All the bioagents overgrew the pathogen and the principal mechanism of mycoparisitism observed was coiling, brusting and disintegration of pathogen hyphae. Culture filtrate from T. viride was found best as it completely inhibited radial growth at 25 and 50% concentration and reduced the spore germination of test fungus significantly. However, lower concentrations of culture filtrate from all bioagents showed little effect on spore germination. Seed application of bioagents was found better as compared to soil application. A maximum increase in seed germination and inhibition of seed borne infection was observed with T. viride followed by T. harzianum under pot culture conditions. T. viride has the maximum potentiality to suppress the spore germination, mycelial growth, seed borne infection of C. lindemuthianum and increased seed germination when compared with the other biocontrol agents. 相似文献
