Root-rot and wilt caused by Fusarium chlamydosporum affects the cultivation of Coleus forskohlii, a medicinal plant grown for its roots, which contain a pharmaceutically important compound called forskolin. In this study, management of this disease under low and high inoculum levels was assessed with four arbuscular mycorrhizal (AM) fungi and a strain of Pseudomonas fluorescens. The AM fungus Glomus fasciculatum and P. fluorescens were the most effective treatments that reduced the severity of root-rot and wilt of C. forskohlii by 56–65% and 61–66%, respectively, under lower and higher levels of pathogen F. chlamydosporum. G. fasciculatum increased the dry shoot and root weight by 108–241% and 92–204%, respectively, while in plants treated with P. fluorescens, an increase of 97–223% and 97–172% in dry shoot and root weight, respectively, was observed. Although P. fluorescens was effective, it gave higher root yields only under lower inoculum level of the pathogen. G. fasciculatum performed equally well under both lower and higher inoculum levels. Increase in yields with both the biocontrol agents was accompanied by increase in P uptake (230–303%) and in K uptake (270–335%). The forskolin content of the roots was significantly increased (14–21%) by G. fasciculatum, P. fluorescens or G. mosseae under lower inoculum level of pathogen.  相似文献

The root-rot or wilt of Coleus forskohlii is a very serious soil-borne disease caused by Fusarium chlamydosporum. A field study was undertaken to study the possibility of controlling the disease using three biocontrol agents viz., Glomus mosseae, Pseudomonas fluorescens, Trichoderma viride, singly and in combination. Planting of coleus cutting was done in wilt sick soil. Inoculation with Trichoderma viride + Glomus mosseae gave the best result in controlling the disease. The same treatment also resulted in maximum growth, yield and root forskolin concentration of coleus. Plants treated with T. viride + G. mosseae showed a disease severity index of 33.28% compared to uninoculated control plants, which had a maximum disease severity index of 85.5%. The fungicide Emisan (0.2%) was not as effective as the biocontrol agents in controlling the pathogen.  相似文献

Abstract

Aqueous extracts of 46 plants belonging to 32 different families of the plant kingdom were screened for antifungal activity against eight important species of Fusarium viz., Fusarium equiseti, F. moniliforme, F. semitectum, F. graminearum, F. oxysporum, F. proliferatum, F. solani and F. lateritium. The test fungi were isolated from maize, paddy and sorghum seeds collected from Mysore district, Mysore, India. Among the several plants screened only 12 plants have recorded significant antifungal activity. The antifungal activity of aqueous extracts varied among the test pathogens and was compared with that of the synthetic fungicides Blitox, Captan, Dithane M-45 and Thiram. F. proliferatum, which showed high susceptibility for the aqueous extracts, was tested using different solvent extracts viz., petroleum ether, benzene, chloroform, methanol and ethanol extracts of all the 12 plants. The results revealed that these plants could be exploited for ecofriendly management of the diseases caused by the test fungal pathogens and seed biodeterioration during storage.  相似文献

Asthma is characterized by airway inflammatory infiltration, which leads to airway remodeling and airway hyperreactivity. Coleus forskohlii (CFK) has been used to treat asthma, however, the mechanism involved is not clear. To explore the antiasthma mechanism of extracts of Coleus forskohlii (ECFK), guinea pigs were administered with a spray of phosphoric acid histamine, and rats were sensitized with ovalbumin (OVA). Hematoxylin and eosin staining (H&E) were used to evaluate pathological changes in lung tissue. Enzyme-linked immunosorbent assay (ELISA) was used to determine cytokine levels in serum and bronchoalveolar lavage fluid (BALF). Immunohistochemistry and Western blot analysis were used to assess the expression of intercellular cell adhesion molecule-1 (ICAM-1), phosphorylation of p65 (p-p65), matrix metallopeptidase 9 (MMP-9), and tissue inhibitor of metalloproteinase 1 (TIMP-1). After ECFK treatment, the asthma incubation period of guinea pigs was significantly prolonged. The H&E results showed that the number of eosinophils in the 12.8 g/kg ECFK group was significantly lower when compared with the control group. Moreover, ELISA results demonstrated that interleukin (IL)-4, IL-5, and IL-17 in serum and BALF were significantly decreased, and interferon-γ (IFN-γ) and IL-10 were increased after ECFK treatment. In addition, ECFK treatment resulted in downregulation of ICAM-1, p-p65, MMP-9, and TIMP-1 in lung tissue after being sensitized by OVA. In conclusion, our findings indicated that ECFK significantly alleviated OVA-induced inflammatory infiltration and airway remodeling in asthma. This study laid a theoretical foundation for the clinical use of ECFK.  相似文献

Root-rotted samples of grapevine cv. superior were collected from Nobaria province, Beheira Governorate, Egypt. Fusarium oxysporum Schlech. was the most fungal causing root-rot syndrome of grapevine and directly affected the yield productivity. Seven isolates of Streptomyces were isolated from grapevine rhizospheric soil and screened for antagonistic activities against F. oxysporum on dual culture plate. All isolates showed antifungal activity, but isolate No. 1 exhibited the highest activity. It was identified as Streptomyces alni according to morphological, cultural, physiological and biochemical studies. The properties of the antagonism were revealed by scanning electron microscopy (SEM) examination of F. oxysporum and S. alni on PDA medium. The forms of antagonism found in this study according to the interaction between the S. alni and the pathogen indicated a hyperparasite, including inhibition of fungal growth and colonisation of S. alni over F. oxysporum hyphae. Also, malformation and lysis of F. oxysporum hyphae and conidiophores were observed. Conidia and normal branches of fungal hyphae were absent. Greenhouse and field studies were performed to evaluate the ability of S. alni and some commercial biofertilisers incorporated into the soil for root-rot control. Pot trails indicated that antagonistic S. alni isolate and biofertilisers i.e. blue green algae, phosphoren and rhizobacterin reduced the root-rot incidence of grapevine plants Cv. superior. Soil treatment before sowing with 50 ml of S. alni suspension (1 × 108 spore/ml) + 50 g of rhizobacterin for each pot was the best and significant treatment reduced root-rot of grapevine plants. Also, the total count of F. oxysporum in rhizosphere soil of grapevine treated plants was reduced compared with control. Under field conditions, drenching soil of diseased grape trees with a spore suspension of S. alni (1 × 108 spore/ml) 200 ml/tree + 250 g/tree of rhizobacterien caused a significant reduction in root rot of treated grapevine trees as well as high fruit yield/tree when compared with other treatments. The obtained results suggest that S. alni could be used successfully in combination with biofertilisers, as environmentally safe, for controlling root-rot of grapevine and other soil-borne plant pathogens especially with organic farming systems.  相似文献

The effects of salicylic acid (SA) at three concentrations i.e. 2.5, 5 and 7 mM and plant extracts from pick tooth (Ammi visnaga), liquorice (Glycyrrhiza glabra), artemisia (Artemisia judaica), mint (Mentha viridis), clove (Syzygium aromaticum) and blue gum (Eucalyptus globulus) on the infection of rice kernel smut disease caused by Tilletia barclayana were studied. Spraying of rice plants with different concentrations of SA at seven days before infection was the most effective treatment against pathogen infection. Among all plant extract treatments, M. viridis and S. aromaticum were the most effective treatments. Additionally, our results showed increased levels of peroxidase, polyphenol oxidase, phenylalanine ammonia lyase and chitinase as well as total protein contents in the treated plants compared with the control. In conclusion, accumulations of these oxidative enzymes in plants treated with SA and plant extracts provide their role in the activation of induced resistance against T. barclayana.  相似文献

Eleven ethanolic extracts from spices of Melissa officinalis, Mentha piperita, Laurus nobilis, Rhus coriaria, Dianthus coryophyllum, Piper nigrum, Capsicum annum, Juniperus oxycedrus, Erica arborea, Colutea arborescens, and Cuminum cyminum collected from various regions of Turkey and local markets were assayed for the in vitro antibacterial activity against 3 Gram-positive (Bacillus subtilis, Staphylococcus aureus and S. epidermidis) and 2 Gram-negative bacteria (Escherichia coli and Pseudomonas aeruginosa), using agar dilution methods. In addition, their possible toxicity to Candida albicans and Aspergillus niger was determined, using both agar dilution and disc-diffusion methods. The minimum inhibition concentration (MIC) of the M. piperita, L. nobilis and J. oxycedrus ethanolic extracts was 5 mg/mL for all the microorganisms tested. P. aeruginosa was the most sensitive bacterial strain to P. nigrum and E. arborea extracts among both Gram-positive and Gram-negative bacteria tested with MIC of 5 mg/mL. The extracts of L. nobilis, D. coryophyllum, J. oxycedrus and C. arborescens showed higher inhibitory activity against the yeast C. albicans and the fungus A. niger than the standard antifungal nystatin.  相似文献

The present study was designated to evaluate the antifungal activity and to root out the antifungal plant leaf extracts from this Indian folk-flore. The in vitro antifungal assay was performed by agar diffusion test and minimum inhibitory concentration (MIC) for hexane, ethyl acetate, methanol and distilled water plant leaf extracts. Extraction of 17 different plant leaves was carried out in different solvents such as hexane, ethyl acetate, methanol and distilled water. Among them extractive yield of methanol was maximum than the rest of the three solvents. These extracts were screened for their antifungal activity against nine different fungi. Among these ethyl acetate extracts of Adhatoda vasica, Ocimum sanctum and Holoptelea integrifolia exhibited maximum antifungal activity against Alternaria sp., Aspergillus parasi, Aspergillus nidulans, Trichoderma harzianum and Aspergillus flavus with MIC of 80, 40 and 20 ppm against Aspergillus nidulans and Alternaria sp. Ethyl acetate extracts showed promising antifungal activity against Adhatoda vasica, Ocimum sanctum and Holoptelea integrifolia against Aspergillus nidulans, and Alternaria sp. might be applicable as fungicide against fungal plants disease.  相似文献

The aim of the present study was to examine the efficacy of various seed extracts of Terminalia chebula as an antifungal potential against certain important plant pathogenic fungi. The organic extracts of methanol, ethyl acetate and chloroform at the used concentration of 1500 ppm/disc revealed remarkable antifungal effect as a fungal mycelial growth inhibitor against Fusarium oxysporum, Fusarium solani, Phytophthora capsici and Botrytis cinerea, in the range of 41.6–61.3%, along with MIC values ranging from 62.5 to 500 μg/ml. Also, the extracts had a strong detrimental effect on spore germination of all the tested plant pathogens along with concentration as well as time-dependent kinetic inhibition of B. cinerea. The results obtained from this study suggest that the natural products derived from Terminalia chebula could become an alternative to synthetic fungicides for controlling such important plant pathogenic fungi.  相似文献

Aims:  To evaluate the anti-oxidant properties of extracts from 20 medicinal herbs growing in western Siberia using microbial test systems and different in vitro methods.
Methods and results:  In vivo anti-oxidant activity of extracts was evaluated for their capacity to protect bacteria, Escherichia coli , against bacteriostatic and bactericidal effects of H2O2 and menadione, and action on anti-oxidant gene expression. In vitro anti-oxidant activity has been examined by a number of methods including: the 1,1-diphenyl-2-picrylhydrazyl radical (DPPH)-scavenging assay, chelating activity and capacity to protect plasmid DNA against oxidative damage. In addition, total polyphenol content was determined. The extracts of Fragaria vesca , Rosa majalis , Pentaphylloides fruticosa , Alchemilla vulgaris and Pulmonaria mollis possessed the highest levels of anti-oxidant activity in vivo and in vitro . The protective properties were more closely related to the DPPH radical-scavenging activity, tannin content and action on anti-oxidant gene expression than to other parameters.
Conclusion:  The extracts of medicinal plants may have anti-oxidant effects on bacteria simultaneously through several different pathways, including direct inhibition of reactive oxygen species, iron chelation and anti-oxidant genes induction.
Significance and Impact of the Study:  Using microbial test systems, we revealed herbs that may be used as potential sources of natural anti-oxidants.  相似文献

In Egypt, sesame cultivation is subject to attack by wilt and root-rot diseases caused by Fusarium oxysporum f.sp. sesami (Zap) Cast. and Macrophomina phaseolina (Maubl) Ashby causing losses in quality and quantity of sesame seed yield. Bacillus subtilis and Trichoderma viride isolates which were isolated from sesame rhizosphere were the most effective to antagonise fungal pathogens, causing high reduction of hyphal fungal growth. Trichoderma viride was found to be mycoparasitic on Fusarium oxysporum f.sp. sesami and M. phaseolina causing morphological atternation of fungal cells and sclerotial formation. In general, Bacillus subtilis, T. viride, avirulent Fusarium oxysporum isolate and Glomus spp. (Amycorrhizae) significantly reduced wilt and root-rot incidence of sesame plants at artificially infested potted soil by each one or two pathogens. Data obtained indicate that Glomus spp significantly reduced wilt and disease severity development on sesame plants followed by T. viride. Meanwhile, avirulent Fusarium oxysporum isolate followed by Glomus spp. were effective against root-rot disease incidence caused by M. phaseolina. Glomus spp. followed by B. subtilis significantly reduced wilt and root-rot disease of sesame plants. All biotic agents significantly reduced F. oxysporum f.sp. sesami and M. phaseolina counts in sesame rhizosphere at the lowest level. Glomus spp. and the avirulent isolate of F. oxysporum eliminated M. phaseolina in sesame rhizosphere. Meanwhile T. viride was the best agent at reducing F. oxysporum at a lower level than other treatments. Application of VA mycorrhizae (Glomus spp.) in fields naturally infested by pathogens significantly reduced wilt and root-rot incidence and it significantly colonised sesame root systems and rhizospheres compared to untreated sesame transplantings.  相似文献

Eleven plant species were collected from the Blyde River Canyon area, Mpumalanga, South Africa. Methanolic crude extracts of the plant samples were concentrated to dryness, dissolved in sterile distilled water and bioassayed in the laboratory for their fungicidal potential against seven plant fungal pathogens, namely Botrytis cinerea, Fusarium oxysporum, Sclerotium rolfsii, Rhizoctonia solani, Verticillium dahliae, Botryosphaeria dothidea and Pythium ultimum, at equal concentrations. A modified agar dilution method was used to determine the inhibitory effect of the plant extracts on the mycelial radial growth of the plant pathogens. Antifungal activity was confirmed in all the plants, but not in all plant parts and also not against all the test organisms. The crude extract of Eucomis autumnalis performed best of all the plant extracts tested, as it showed significant antifungal activity against all seven of the plant pathogenic test organisms and compared favourably to the inhibition of the mycelial growth by a broad spectrum synthetic fungicide (carbendazim/ difenoconazole). The crude extract of Schrebera alata came out second best, in the sense that at least one plant part inhibited the mycelial radial growth of four of the seven test organisms by at least 50%.  相似文献

Casein hydrolysate at 2.0 g l–1 significantly enhanced forskolin content (2.3 mg g–1 cell dry wt) in a rhizogenic tumourous line, GCO-RCH-2 of Coleus forskohlii. In rooty teratoma line, RC-ST-2/4, forskolin content enhanced to 1.7 mg g–1 cell dry wt in presence of 2.5 g l–1 casein hydrolysate. Unlike untransformed calli and rhizogenic/root cultures, all the forskolin yielding transformed cultures of C. forskohlii have been maintained for over 5 years.  相似文献

This study was undertaken to investigate the influence of plant probiotic fungus Piriformospora indica on the medicinal plant C. forskohlii. Interaction of the C. forskohlii with the root endophyte P. indica under field conditions, results in an overall increase in aerial biomass, chlorophyll contents and phosphorus acquisition. The fungus also promoted inflorescence development, consequently the amount of p-cymene in the inflorescence increased. Growth of the root thickness was reduced in P. indica treated plants as they became fibrous, but developed more lateral roots. Because of the smaller root biomass, the content of forskolin was decreased. The symbiotic interaction of C. forskohlii with P. indica under field conditions promoted biomass production of the aerial parts of the plant including flower development. The plant aerial parts are important source of metabolites for medicinal application. Therefore we suggest that the use of the root endophyte fungus P. indica in sustainable agriculture will enhance the medicinally important chemical production.  相似文献

根腐病是药用植物三七Panax notoginseng的多发性病害,而土壤真菌群落组成与根腐病的发生存在一定的联系。利用末端限制性酶切片段长度多态性分析和Biolog技术研究了云南文山三七种植园健康和发病三七植株根际土壤真菌群落结构和碳源利用特征,结果表明健康和发病植株的根际土壤真菌多样性和代谢活性差异不显著,但是真菌群落组成存在差异。健康和发病植株根际土壤都以小茎点霉Phoma exigua及镰孢菌属Fusarium真菌为主要种群,而健康植株根际土壤中火丝菌属Pyronema和被孢霉属Mortierella等真菌类群的相对丰度显著高于发病植株根际土壤,尖孢镰孢菌F.ox‐ysporum和刺座霉属的Volutella colletotrichoides的相对丰度则显著低于发病植株。  相似文献

Aims: To investigate the influence of polyphenols and plant extracts on the susceptibility of Escherichia coli to antibiotics. Methods and Results: Susceptibility of E. coli to antibiotics in the presence of extracts and polyphenols was estimated by the determination of the minimum inhibitory concentrations (MICs). To study gene expression, we used strains of E. coli carrying fusions between promoters of genes katG, sodA, iucC and structural β‐galactosidase gene. Treatment with polyphenols and some plant extracts significantly decreased the antibacterial effects of antibiotics, to a larger extent, ciprofloxacin. The most remarkable protective effect was observed for the extracts of Chamerion (Epilobium) angustifolium, Filipendula vulgaris, Tanacetum vulgare and Serratula coronata. These extracts increased the MICs of ciprofloxacin by four and more times. In case of kanamycin, extracts of Artemisia austriaca and Artemisia pontica increased MICs by four and eight times, respectively. Polyphenol quercetin also caused protective effect against ciprofloxacin, increasing the MIC by four times. A positive correlation was found between protective effects of polyphenols and extracts and their antioxidant activity. Conclusion: Medicinal plant extracts and polyphenols may protect cells of E. coli against antibiotic toxicity. Significance and Impact of the Study: The results of this study may be used to enhance the efficiency of antibacterial therapies.  相似文献

目的 探讨中药覆盆子提取物联合唑类药物的体外抗真菌作用.方法 采用CLSI公布的M27-A方案微量液基稀释法和棋盘式微量稀释法,测定覆盆子提取物单用及联合唑类药物对不同念珠菌的MIC值和FICI指数.结果 覆盆子不同溶液提取物与氟康唑均表现出协同关系,以覆盆子醇提物为例,单用对念珠菌的MIC80测定值范围主要集中在0.16~1.25 mg/mL,与氟康唑合用后表现出协同关系(FICI≤0.5),且MIC80测定值范围降至0.01 ~0.04 mg/mL;合用后的氟康唑抗真菌活性也明显增强.另外,覆盆子醇提物与不同唑类药物合用后均有协同关系,其MIC80测定值由单用时大于10 mg/mL降至0.04 mg/mL.结论 覆盆子醇提物和唑类药物单用时对耐药念珠菌的抑菌作用较弱,但二者合用后表现出明显的协同关系,对耐药念珠菌的抑菌作用明显增强.  相似文献

Ethanolic extracts of 20 plant species, selected from the ethnobotanical literature, were analysed for their pharmacological potential as antineoplastic agents against the HEp-2 cell line. Psoralea corylifolia and E. grandidens were the most efficacious species eliciting IC50 values of 22 microg/ml and 57 microg/ml respectively. Psoralea corylifolia, additionally tested against lung carcinoma (A549) cells gave an IC50 value of 68 microg/ml. Such data would justify a search for active compounds from this species.  相似文献
