L-Pipecolic acid is a chiral pharmaceutical intermediate. An enzymatic system for the synthesis of L-pipecolic acid from L-lysine by commercial L-lysine α-oxidase from Trichoderma viride and an extract of recombinant Escherichia coli cells coexpressing Δ1-piperideine-2-carboxylate reductase from Pseudomonas putida and glucose dehydrogenase from Bacillus subtilis is described. A laboratory-scale process provided 27 g/l of L-pipecolic acid in 99.7% e.e.  相似文献

A gene (aman6) encoding endo-1,6-α-D-mannanase, a yeast mannan backbone degrading enzyme from Bacillus circulans was cloned. The putative aman6 was 1767 base pairs long and encoded a mature 1,6-α-D-mannanase protein of 589 amino acids and a signal peptide of 36 amino acids. The purified mature 1,6-α-D-mannanase from the Escherichia coli transformant showed 61-kDa protein, and N-terminal amino acid sequence and other general properties of the recombinant enzyme were identical to those of 1,6-α-D-mannanase from Bacillus circulans TN-31.  相似文献

Abstract

In this article, we describe the synthesis of 5-nitro-1-(2-deoxy-α-D-erythro-pentofuranosyl)cytosine (), 5-nitro-1-(2-deoxy-β-D-erythro-pentofuranosyl)cytosine (), 5-amino-1-(2-deoxy-α-D-erythro-pentofuranosyl)cytosine (), 5-nitro-1- (2-deoxy-β-D-erythro-pentofuranosyl)cytosine (), 5-nitro-1-(2,3-dideoxy-β- D-ribofuranosyl)uracil (), 5-amino-1-(2,3-dideoxy-α,β-D-ribofuranosyl)uracil (7), 5-nitro-1-(2,3-dideoxy-α,β-D-ribofuranosyl)cytosine (8) and 5-amino-1-(2,3-dideoxy-β-D-ribofuranosyl)cytosine (). The prepared compounds were tested for their activity against HIV and HBV viruses, but they did not show significant activity.  相似文献

A peptidoglutaminase activity in microorganisms was detected using carbobenzoxy-l-glutamine or tertiary-amyloxycarbonyl-l-glutaminyl-l-proline as substrate. By screening, an organism which produces a relatively large amount of peptidoglutaminase was isolated from soil. The organism was identified as Bacillus circulans. The highest enzyme formation by the bacterium occurred during stationary growth phase in the basal medium containing lactose (0.5%) and polypepton (1%).  相似文献

Microorganisms producing D-β-acetylthioisobutyric acid from methyl D-β-acetylthioisobutyrate were screened from stock cultures. The D-β-acetylthioisobutyric acid-producing ability was found in 15 strains belonging to the genera Pseudomonas, Agrobacterium, Enterobacter, Cellulomonas, Rhodococcus, Brevibacterium, and Torulopsis. A strain of Pseudomonas fluorescens, IFO 3081, was selected as the best microorganism. The cells having activity (558 units/g of dry cells) could be easily prepared by cultivation at 25°C at pH 6.6 for 24 hr in a glucose-containing medium. The D-form of methyl DL-β-acetylthioisobutyrate was selectively hydrolyzed with the cells so that D-β-acetylthioisobutyric acid (97.2% enantiomeric excess) was produced in a high yield.  相似文献

The 7-keto-8-aminopelargonic acid (KAPA) synthetase activities of cell-free extracts from various bacteria were investigated. The experiments on the substrate specificity of KAPA synthetase, using crude cell-free extracts from bacteria having high enzyme activity, showed that l-serine and pyruvic acid could replace l-alanine, but that, when the enzyme was partially purified, these compounds were not effective. Many kinds of amino acids such as l-cysteine, l-serine, d-alanine, glycine, d-histidine, and l-histidine, inhibited the enzyme activity. This inhibition was found to be competitive with l-alanine. Pyridoxal 5′-phosphate, which is a cofactor of the enzyme, also inhibited the enzyme activity at high concentrations. The repression of KAPA synthetase by biotin occurred in Bacillus subtilis and B. sphaericus but not in Micrococcus roseus and Pseudomonas fluorescens, even at a concentration of 1000 mµg per ml of biotin.  相似文献

A succinimide-assimilating bacterium, Pseudomonas putida s52, was found to be a potent producer of pyruvate from fumarate. Using washed cells from P. putida s52 as catalyst, 400 mM pyruvate was produced from 500 mM fumarate in a 36-h reaction. Bromopyruvate, a malic enzyme inhibitor, was used for the selection of mutants with higher pyruvate productivity. A bromopyruvate-resistant mutant, P. putida 15160, was found to be an effective catalyst for pyruvate production. Moreover, under batch bioreactor conditions, 767 mM of pyruvate was successfully produced from 1,000 mM fumarate in a 72-h reaction with washed cells from P. putida 15160 as catalyst.  相似文献

A novel approach to the taxonomy of several free-living nitrogen-fixing bacteria is proposed on the basis of two DNA parameters. 1) DNA base composition, expressed as average molar (guanine + cytosine) content, determined by thermal denaturation and 2) DNA homology, determined by DNA hybridization with bothPseudomonas fluorescens andPseudomonas putida. The following taxonomic conclusions emerged:

Twenty endophytic bacteria were isolated from surface-sterilized stems and roots of cucumber plants. After removal of potential siblings and human pathogens, the remaining seven strains were identified based on their 16S rDNA as Pseudomonas fluorescens (2 strains) and P. putida (5 strains). Three strains, namely P. fluorescens CS1, P. fluorescens CR2 and P. putida CR3, were able to suppress tomato foot and root rot (TFRR). Special attention was paid to the characterization of the BIOLOG carbon oxidation profiles of the isolated pseudomonads in order to identify nutrients which might be important for their endophytic lifestyle. Comparative analysis of the profiles of these seven strains with those of seven rhizospheric Pseudomonas spp. revealed that endophytes were able to oxidize l-arabinose and 2,3-butanediol significantly more often than the rhizospheric group. An independent growth experiment performed in tubes using l-arabinose and 2,3-butanediol as sole carbon sources showed the same results as seen using BIOLOG for l-arabinose, but not for 2,3-butanediol. Since l-arabinose is one of the most abundant sugars in xylem of cucumber plants and was not detected in their rhizosphere, our data suggest that utilization of l-arabinose might be a trait contributing to the endophytic lifestyle of the isolated Pseudomonas endophytes.  相似文献

The identification of carotenoids in B. trispora during pigment production from deproteinized hydrolyzed whey supplemented with plant oils was studied. The carotenoid content in Blakeslea trispora were β-carotene, γ-carotene, and lycopene. The composition of carotenoids depends of the amount of oils added to the cheese whey. At the maximum concentration of carotenoids, the proportions of β-carotene, γ-carotene, and lycopene (as percent of total carotenoids) was 60.1%, 32.5%, and 7.4%, respectively.  相似文献

Follicle-stimulating hormone (FSH) and luteinizing hormone (LH) have a central role in follicle growth and maturation, but no clear differences between breeds with different ovulation rates have been found. Therefore, this study investigated mRNA expression of FSHβ, LHβ, FSH receptor (FSHR), LH receptor (LHR), and estrogen receptor-β (ERβ) genes in prolific Lezhi black (LB) goats and nonprolific Tibetan (TB) goats by real-time PCR. Follicles and pituitaries were recovered from goats at 12–24 h after onset of estrus. Real-time PCR analysis revealed that the expression levels of FSHβ and LHβ mRNA were significantly higher (p < 0.01) in pituitary of LB than in TB does, but the expression levels of FSHR and LHR mRNA in follicle of TB were greater (p < 0.05). Expression level of follicular ERβ was not different between the two breeds. Data provide evidence that the greater ovulation rate in the LB goat as compared to the TB breed is associated with a greater gonadotropin expression during follicular phase.  相似文献

Twenty-one street dust samples were collected from the urban sites in Baoding, China, to investigate the species and bioavailabilty of arsenic in them. The ecological risk and bioavailability of arsenic were evaluated using three models including Bioavailability Factor (BF), Contamination Factors (Cf)and Geoaccumulation Index model (Igeo). The species of arsenic in the dust samples were analyzed using an optimized BCR sequential extraction method. The total concentrations of As in the street dust samples ranged from 13.16 mg kg?1 to 67.26 mg kg?1. Geoaccumulation index (Igeo) of As ranged from 0.28 to 1.99. The speciation analysis indicated that As in the street dust samples were mainly in the residual fraction (F4), and the proportion ranged from 84.35% to 87.07%. Moreover, the ranges of the BF and Cf were 0.650–0.129 and 0.119–0.186, respectively. The results indicated that arsenic contained in the street dust samples was with low bioavailability.  相似文献

The in vivo and in vitro effects of 4-amino-3-(D-glucopentitol-l-yl)-5-mercapto-1,2,4-triazole and its 3-methyl analogue on α- and β-glucosidases, β-glucuronidase as well as α-amylase have been investigated. α-Glucosidase is the enzyme that is markedly affected in vivo and in vitro in a dose-dependent manner. The compounds showed a reversible inhibition of a competitive type for α-glucosidase. Moreover, they exert a relatively potent inhibition on α-glucosidase with a Ki magnitude of 3.6×10?4, 9.5×10? 5 M.  相似文献

Two genes of Pseudomonas putida (IFO 12996) which code for enzymes participating in amino acid metabolism, were cloned in Escherichia coli C600 using pBR322 as a vector. pST7549 is a 7.9 kb hybrid plasmid DNA which is composed of four SalI fragments (0.3, 1.4, 1.9 and 4.3 kb), and codes for β-isopropylmalate dehydrogenase (EC 1.1.1.85) in l-leucine biosynthesis. The enzyme activity in the crude extract from E. coli C600 bearing pST7549 was 80 ~ 90% lower than that of E. coli K12 or P. putida. When the foreign SalI fragments derived from P. putida were subcloned, a 1.9 kb SalI fragment was found to encode β-isopropylmalate dehydrogenase and it did not contain the promoter of P. putida DNA. Plasmid pST6961 has a 1.8 kb insert derived from the P. putida DNA in the SalI site of pBR322. E. coli cells carrying this recombinant plasmid show no leucine racemase activity and no d-leucine transaminase activity, but five-times higher d-leucine oxidation activity than the host strain, E. coli. Enzymological studies have suggested that plasmid pST6961 codes for d-amino acid dehydrogenase, a key enzyme in d-amino acid metabolism.  相似文献

3-Chloro-d-alanine chloride-lyase, which occurs in the cells of Pseudomonas putida CR 1-1, catalyzes not only the α,β-elimination reaction of 3-chloro-d-alanine to form pyruvate, but also its β-replacement reaction in the presence of a high concentration of sodium hydrosulfide to form d-cysteine. Using the β-replacement reaction, the enzymatic synthesis of d-cysteine by resting cells was investigated. The culture conditions for cell production of the bacterium with high d-cysteine-producing activity and the reaction conditions for d-cysteine production were optimized. Under these optimal reaction conditions, 100% of the added 3-chloro-d-alanine could be converted to d-cysteine and, as the highest yield, 20.6 mg of d-cysteine per 1.0 ml of reaction mixture could be synthesized.  相似文献

N-Benzoylgiycine amidohydrolase (hippurate hydrolase EC 3.5.1.32), which catalyzes the hydrolysis of hippuric acid to benzoic acid and glycine, was found in a cell-free extract of Pseudomonas putida C692-3 grown on a medium containing hippuric acid. The enzyme was purified from the extract by ammonium sulfate fractionation and column chromatographies on DEAE-cellulose, DEAE-Sephadex A-50, hydroxyapatite, and Sepharose CL-6B. The enzyme was finally crystallized. The crystalline enzyme was almost homogeneous on electrophoresis. The enzyme had a molecular weight of about 170,000 and consisted of four subunits identical in molecular weight (approximately 42,000). The enzyme hydrolyzed N-benzoylglycine most rapidly, and N-benzoyl-l-alanine and N-benzoyl-l-aminobutyric acid. The Km value for these substrates were 0.72 mm, 0.87 mm, and 0.87mm, respectively. The optimum pH of the enzyme reaction was 7.0 to 8.0 and the enzyme was stable from pH 6.0 to 8.0.  相似文献

Abstract

The new monomer 1 seems to be an excellent mimic of nucleosides with different sugar conformations (north, south, and envelope), because of the relatively free rotation around γ, δ, and χ. The rotation around χ is primarily controlled by the repulsion between H6 and the two hydrogen atoms on C4′ and not pi conjugation between the double bond and the nucleobase.
