The aims of this study were to evaluate the antifungal properties of Baccharis glutinosa and Ambrosia confertiflora extracts against Aspergillus flavus, A. parasiticus and Fusarium verticillioides, and to isolate the group of compounds that are responsible for the antifungal activity. Samples of aerial parts from each plant were extracted with 70% methanol and sequentially partitioned with hexane, ethyl acetate, and n-butanol. The partitioned fractions were evaluated in their capacity to inhibit the radial growth of the three species of fungi. The active fraction was used for an assay-guided chromatography of antifungal extracts. The results showed that the extract from B. glutinosa partitioned in ethyl acetate (Bea) showed the highest antifungal activity against the three fungi. Bea completely inhibited the growth of F. verticillioides at 0.8 mg/ml, whereas the radial growth of A. flavus and A. parasiticus was inhibited 70% at 1.5 mg/ml. The purified antifungal fraction from Bea showed 72, 54, and 52% of antifungal activity, respectively.  相似文献

The leaves of Cyclosorus extensa are used in the preparation of rice beer in Assam, India. The optimal conditions of time and temperature of fermentation for extraction of bioactive compounds from the dried leaves were obtained using response surface methodology. The central composite rotatable design was used and 13 experimental runs based on two-factor-five-level design were generated and performed for each of the solvents. The independent variables were extraction time (12 and 48?h) and temperature (25 and 55°C). The responses studied were total polyphenol content, radical scavenging activity, antibacterial activity, and antifungal activity. The analysis of variance of the test data was performed and the sequential sum of squares, F-value, R2, and adjusted R2 were deduced. The predicted models for all the response variables were adequately fitted to the observed experimental data (p?≤?0.001). The maximum extraction of bioactive compounds under the optimum conditions of extraction temperature and time for hexane, ethyl acetate, methanol, and distilled water were found to be 25°C for 29.43?h, 28.28°C for 41.27?h, 43.95°C for 29.61?h, and 55.00°C for 48.00?h, respectively. It was also observed that the solubility of the polyphenols was higher in methanol, followed by ethyl acetate, and the highest antibacterial activity against Escherichia coli was shown by the ethyl acetate extracts.  相似文献

Eumycetoma is a debilitating chronic inflammatory fungal infection that exists worldwide but it is endemic in many tropical and subtropical regions. The major causative organism is the fungus Madurella mycetomatis. The current treatment of eumycetoma is suboptimal and characterized by low cure rate and high recurrence rates. Hence, an alternative therapy is needed to address this. Here we determined the antifungal activity of seven Sudanese medicinal plant species against Madurella mycetomatis. Of these, only three species; Boswellia papyrifera, Acacia nubica and Nigella sativa, showed some antifungal activity against M. mycetomatis and were further studied. Crude methanol, hexane and defatted methanol extracts of these species were tested for their antifungal activity. B. papyrifera had the highest antifungal activity (MIC50 of 1 ug/ml) and it was further fractionated. The crude methanol and the soluble ethyl acetate fractions of B. papyrifera showed some antifungal activity. The Gas-Liquid-Chromatography hybrid Mass-Spectrophotometer analysis of these two fractions showed the existence of beta-amyrin, beta-amyrone, beta-Sitosterol and stigmatriene. Stigmatriene had the best antifungal activity, compared to other three phytoconstituents, with an MIC-50 of 32 μg/ml. Although the antifungal activity of the identified phytoconstituents was only limited, the antifungal activity of the complete extracts is more promising, indicating synergism. Furthermore these plant extracts are also known to have anti-inflammatory activity and can stimulate wound-healing; characteristics which might also be of great value in the development of novel therapeutic drugs for this chronic inflammatory disease. Therefore further exploration of these plant species in the treatment of mycetoma is encouraging.  相似文献

Moringa oleifera is a miracle plant rich in nutrients, antioxidants, and antibiotic properties. Present study was designed to evaluate various biochemical attributes of leaves and flowers of M. oleifera. Plant parts (leaves, flowers) of M. oleifera, collected from different roadsides of Multan district, Punjab, Pakistan, were used as experimental material. Result indicates that alkaloids, saponin, carbohydrates, fats, and protein had a high value in the aqueous extract of both leaves and flowers of M. oleifera. Whereas phenol content was high in methanolic leaves extract and the phenol contents were high in aqueous extract of flowers. The extract yield of M. oleifera leaves and flowers both showed a higher percentage in aqueous extract (57.5%), followed by methanol extract and lowest in ethyl acetate extract. Flavonoids contents were higher in ethyl acetate extract of leaves (33.67%) and aqueous extract of flowers (53.71%). While crude fiber was high in methanolic extract of leaves (12.40%) and in flowers crude fiber was high in ethyl acetate extract (15.86%). The moisture contents were higher in leaves (8.87%) than flowers (7.3%) and similarly, ash percentage in flowers (52.60%) than leaves (41.84%). Ethyl acetate extracts of M. oleifera leaves show antibacterial activity against Pseudomonas aeruginosa while methanolic extract of M. oleifera flowers shows antibacterial activity against Xanthomonas sp. Maximum growth inhibits show in all extracts of leaves against Aspergillus flavus, F. oxysporum, and P. glabrum except for the concentrated aqueous extract of leaves. While in flowers maximum growth inhibits all extracts against P. glabrum, A. niger, and A. flavus except the diluted ethyl acetate extract. Phytochemicals present in different parts of moringa have significant edible and commercial potential. Moringa extracts exhibited significant antimicrobial activity, therefore have applications in pharmaceuticals.  相似文献

In folk medicine, Adhatoda vasica Ness. (Acanthaceae) is used to treat asthma and cough. The leaves of A.vasica were powdered and extracted with hexane, ethyl acetate and methanol. The hexane extract showed 97% reduction in colony-forming units (CFU) at 100 μg/ml. The hexane extract was subjected to column chromatography. Two natural compounds, vasicine acetate and 2-acetyl benzylamine, were isolated from it. They were bioassayed against Mycobacterium tuberculosis. The two compounds showed strong antimycobacterial activity. Vasicine acetate and 2-acetyl benzylamine isolated from hexane extract of A.vasica leaves, significantly inhibited M. tuberculosis and one multi-drug-resistant (MDR) strain and one sensitive strain at 200 and 50 μg/ml, respectively. Our study demonstrated that both the compounds, vasicine acetate and 2-acetyl benzylamine, could be evaluated further for developing a drug to control M. tuberculosis.  相似文献

In the present study, in vitro anticariogenic potential of ethyl acetate, hexane and methanol and aqueous extracts of plant leaves of Eucalyptus globules Labill. were evaluated by using four cariogenic bacteria, Lactobacillus acidophilus, Lactobacillus casei, Staphylococcus aureus and Streptococcus mutans. Agar well diffusion method and minimum inhibitory concentration (MIC) were used for this purpose. The ethyl acetate extracted fraction of plant leaves showed good inhibitory effects against all selected bacteria. In Eucalyptus globules, hexane and ethyl acetate extracts found highly effective against, Lactobacillus acidophilus with MIC value of 0.031 and 0.062 mg/mL, respectively. Qualitative phytochemical investigation of above extracts showed the presence of alkaloids, phenolic compounds, steroids, cardiac glycosides and terpenes. Based on the MIC value and bioautography, ethyl acetate of plant leaf was selected for further study. Further investigation on the structure elucidation of the bioactive compound using IR, GC-MS and NMR techniques revealed the presence of alpha-farnesene, a sesquiterpene. Eucalyptus globules plant leaf extracts have great potential as anticariogenic agents that may be useful in the treatment of oral disease.  相似文献

The aim of the present study was to examine the efficacy of various seed extracts of Terminalia chebula as an antifungal potential against certain important plant pathogenic fungi. The organic extracts of methanol, ethyl acetate and chloroform at the used concentration of 1500 ppm/disc revealed remarkable antifungal effect as a fungal mycelial growth inhibitor against Fusarium oxysporum, Fusarium solani, Phytophthora capsici and Botrytis cinerea, in the range of 41.6–61.3%, along with MIC values ranging from 62.5 to 500 μg/ml. Also, the extracts had a strong detrimental effect on spore germination of all the tested plant pathogens along with concentration as well as time-dependent kinetic inhibition of B. cinerea. The results obtained from this study suggest that the natural products derived from Terminalia chebula could become an alternative to synthetic fungicides for controlling such important plant pathogenic fungi.  相似文献

This study reports the isolation of 63 endophytic fungal isolates from two traditional medicinal plants, Ocimum sanctum and Sapindus detergens from different locations of Amritsar, India. The functional characterization of the fungi for their ability to produce anti bacterial and anti cancer agent was carried out. Sixteen strains were characterized at molecular level by sequencing the amplified ITSI-5.8-ITSII region of rDNA. The phylogenetic tree resolved the endophytic fungi into different clades. The fungal endophytes belonging to order Pleosporales (Alternaria sp., Phoma sojicola and Exserohilum sp.) were functionally versatile as they produced diverse biomolecules including antibacterial agent active against Mycobacterium smegmatis, as well as cytotoxic activity against different human cancer cell lines of lung, ovary, breast, prostrate, neuroblastoma and colon.  相似文献

A systemic study of fungal endophytes associated with different plant parts of Cannabis sativa and their antifungal activity was investigated in the present study. A total of 281 plant segments, including 91 leaves, 93 stem and 97 petioles samples, were screened for the isolation of endophytic fungi. Totally, 212 (77.65%) segments were found colonised by different fungi. Highest colonisation frequency were observed in stem parts (84.94%), then leaves (82.41%) and lowest 59.79% in petiole. Total eight fungal genera belonging to 12 species were isolated. Aspergillus is recorded as the most frequently occurring genera with three species Aspergillus niger, Aspergillus flavus and Aspergillus nidulans followed by Penicillium with two species Penicillium chrysogenum and Penicillium citrinum, while Phoma, Rhizopus, Colletotrichum, Cladosporium and Curvularia with single species. The antifungal potential of A. niger and A. flavus – two most frequently isolated endophytic fungi – was evaluated against two common plant pathogen, Colletotrichum gloeosporioides and Curvularia lunata. Different plant and fungal extracts individually and in combinations showed variations in antifungal activity against both the pathogens. The primary results obtained on antifungal activity of endophytes show their possible role in plant defence mechanism but it is a preliminary approach and more extensive research is still required.  相似文献

Antimicrobial activities of rhizome extracts of Cyperus rotundus were investigated on selected plant pathogenic bacteria and fungi. Ethyl acetate and hexane extracts showed antibacterial activity against three isolates of Clavibacter michiganensis subsp. michiganensis at concentrations of 900 and 1000 μg/ml. However, Gram-negative bacterial pathogens of tomato; Pseudomonas syringae pv. tomato and Ralstonia solanacearum were not inhibited from the extracts. Ethyl acetate extracts at 100 μg/ml inhibited mycelial growth and spore germination of the two strawberry isolates of Botrytis cinerea; however, no significant inhibition was found in tomato fungal pathogen, Fusarium oxysporum f. sp. radicis lycopersici. Minimum inhibitory concentrations were determined as 0.0625 and 0.125 mg/ml against C. michiganensis subsp. michiganensis for ethyl acetate and hexane extracts of rhizomes, respectively. This study shows the potentials of extracts of C. rotundus rhizomes as antimicrobial agents that are effective against the tested plant pathogenic bacteria and fungi.  相似文献

Aims: To investigate the antifungal activity of aqueous and organic extracts of four weedy plant species viz. Tagetes minuta, Lippia javanica, Amaranthus spinosus and Vigna unguiculata against isolates of four agriculturally important fungi, i.e. Fusarium verticillioides, F. proliferatum, Aspergillus flavus and A. parasiticus. Methods and Results: Dried powdered aerial parts of the plants were extracted sequentially with hexane, dichloromethane, methanol and water and tested for activity using a serial microdilution assay. Results were read every day over 120 h. All extracts except for the water extracts showed growth inhibitory activity against most isolates of the Fusarium spp. The most active were the methanol and hexane extracts of V. unguiculata and A. spinosus with minimum inhibitory concentration (MIC) values of <0·5 mg ml?1 after 48 h against Fusarium spp. No inhibition of the Aspergillus spp. tested was observed, but conidium formation was stimulated on plates treated with plant extracts when visually compared to the growth controls. Conclusions: The results obtained from this study indicated that chemical constituents from these plant species may be developed as potential agrochemical fungicides. Significance and Impact of the Research: Food and feed are subject to infection by a variety of micro‐organisms that can induce spoilage and/or produce metabolites that are toxic to humans and animals. Extracts of V. unguiculata and A. spinosus were most active and maybe developed into environmentally friendly fungicides, which are affordable to rural farmers in developing countries.  相似文献

This study is to evaluate the potential of endophytic fungi of Salvadora persica for the production of bioactive compounds against pathogenic bacteria and fungi. Forty-two fungal isolates were obtained from 135 young and old stem and 125 root segments. Those 42 isolates representing ten fungi include: Trichoderma sp. (the most common), two species of Alternaria, Rhizopus arrhizus and 6 sterile mycelia. The ten fungi were grown in liquid culture and their crude extracts were tested against pathogenic bacteria and fungi. Nine crude extracts gave positive reactions against pathogenic bacteria of which Alternaria sp. (A8) was chosen further study. The fungal isolate was growing as sterile mycelium and was identified by phylogenetic analyses based on LSU rDNA sequence data and it might represent undescribed species of Alternaria. Sixty-two bioactive chemical compounds were identified from the ethyl acetate crude extracts of Alternaria sp., of which the following were recorded as major compounds in the active sub-fractions. On screening the leaf extracts of some higher plants for their volatile antifungal activity against the test organism Aspergillus flavus, the extract of Ocimum adscendens exhibited the strongest fungitoxicity. The leaves showed maximum, fungitoxicity as compared with other plant parts. The volatile fungitoxic fraction obtained as an essential oil was standardized by various physico-chemical properties. The oil showed its fungicidal nature and broad range of activity at its minimum inhibitory concentration. The oil was thermostable and the toxicity remained unchanged even on autoclaving and on storage for up to 360 days. Moreover, the oil proved more active than some prevalent synthetic fungicides and exhibited no phytotoxic effect onVigna radiata.  相似文献

Crude aqueous methanol extracts obtained from 31 species of various marine organisms (including flora land faunal), were screened for their antifungal activity against food poisoning strains of Aspergillus. Seventeen species exhibited mild (+ =zone of inhibition 1–2 mm) to significant (+++ =zone of inhibition 3–5 mm) activity against one or the other strain under experiment. However, extracts of 12 species were active against all the three strains. Organisms like Salicornia brachiata(obligate halophyte), Sinularia leptocladus(Soft coral), Elysia grandifolia (Mollusks),Gorgonian sp. 2 and Haliclona sp. exhibited significant (inhibition zone of 3–5 mm) antifungal activity against one or the other strains. However,extracts of A. ilicifolius, Amphiroa sp.,Poryphyra sp., Unidentified sponge, Suberites vestigium, Sinularia compressa,Sunularia sp., Sinularia maxima, Subergorgia suberosa, Echinogorgia pseudorassopo and Sabellaria cementifera were mild (inhibition zone of 1–2 mm) to moderate(inhibition zone of 2–3 mm) active against the respective strains. The growth of A. japonicuswas significantly inhibited by the extracts ofS. leptocladus (r = 0.992, p < 0.0001)and E. grandifolia (r = 0.989, p < 0.0001).This revised version was published online in October 2005 with corrections to the Cover Date.  相似文献

The aim of the present study is to assess the antimicrobial activities of various leaf extracts of Ocimum americanum were tested against pathogenic microorganisms. Preparation of different extracts viz., aqueous, acetone, ethyl acetate and methanol through soxhlet extraction method. Various extracts were investigated against MTCC strains of Bacillus cereus, Clostridium penfrigens, Klebsilla pnemoniae, Salmonella paratyphi, Candida albicans and Aspergillus niger by agar well diffusion and disc diffusion methods. Minimum inhibitory concentration (MIC), Minimum Bactericidal/Fungicindal Concentration (MBC/MFC) were determined through micro dilution method. Elucidation of phytochemicals and functional groups were observed by HPLC and FT-IR respectively. Ethyl acetate leaf extract of O.americanum showed significant antimicrobial activity against the all tested pathogens in agar well diffusion method in which B.cereus (17 mm) was observed high zone of inhibition. Whereas lowest inhibition was observed in aqueous extract against C.pentrigens (7 mm). The ranges of MIC values from 0.78 μg/ml to 50 μg/ml and MBC/MFC 1.56 μg/ml to 50 μg/ml were observed. Phytochemicals such as alkaloids, steroids, saponins, flavonoids, tannins, terepenes, phenolic compounds cardiac glycosides were detected. Saponinns, flavonoids, tannins, phenolic compounds were observed in only ethyl acetate leaf extracts. Functional group of the leaf extracts was exhibited by FTIR and HPLC analysis of the ethyl acetate leaf extract was elutated at six peaks. Based on the results we concluded that ethyl acetate leaf extract of O.americanum has proved to be potentially effective than the other extracts. Therefore, ethyl acetate leaf extract of O.americanum could act as antimicrobial agent and further studies are recommended for isolation of compounds and toxicological studies.  相似文献

Purpose of the present study was to evaluate antioxidant, antibacterial, antifungal, and antiviral activities of the petroleum ether, chloroform, ethyl acetate and methanol extracts as well as the alkaloid fraction of Lycopodium clavatum L. (LC) from Lycopodiaceae growing in Turkey. Antioxidant activity of the LC extracts was evaluated by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging method at 0.2 mg/ml using microplate-reader assay. Antiviral assessment of LC extracts was evaluated towards the DNA virus Herpes simplex (HSV) and the RNA virus Parainfluenza (PI-3) using Madin-Darby Bovine Kidney (MDBK) and Vero cell lines. Antibacterial and antifungal activities of the extracts were tested against standard and isolated strains of the following bacteria; Escherichia coli, Pseudomonas aeruginosa, Proteus mirabilis, Acinobacter baumannii, Klebsiella pneumoniae, Staphylococcus aureus, Bacillus subtilis as well as the fungi; Candida albicans and C. parapsilosis. All of the extracts possessed noteworthy activity against ATCC strain of S. aureus (4 μg/ml), while the LC extracts showed reasonable antifungal effect. On the other hand, we found that only the chloroform extract was active against HSV (16–8 μg/ml), while petroleum ether and alkaloid extracts inhibited potently PI-3 (16–4 μg/ml and 32–4 μg/ml, respectively). However, all of the extracts had insignificant antiradical effect on DPPH. In addition, we also analyzed the content of the alkaloid fraction of the plant by capillary gas chromatography-mass spectrometry (GC-MS) and identified lycopodine as the major alkaloid.  相似文献

In this study, strain Streptomyces sp. Act4Zk was isolated based on a method developed for the isolation of myxobacteria. Due to the low efficiency of the majority of conventional DNA extraction techniques, for molecular identification of the strain Streptomyces sp. Act4Zk, a new technique for DNA extraction of Actinobacteria was developed. In order to explore potential bioactivities of the strain, extracts of the fermented broth culture were prepared by an organic solvent (i.e. ethyl acetate) extraction method using. These ethyl acetate extracts were subjected to HPLC fractionation against standard micro-organisms, followed by LC/MS analysis. Based on morphological, physiological, biochemical and 16S rRNA gene sequence data, strain Streptomyces sp. Act4Zk is likely to be a new species of Streptomyces, close to Streptomyces genecies and Streptomyces roseolilacinus. Antimicrobial assay indicated high antifungal activity as well as antibacterial activity against Mycobacterium smegmatis and Gram-positive bacteria for the new strain. HPLC and LC/MS analyses of the extracts led to the identification of three different compounds and confirmed our hypothesis that the interesting species of the genus Streptomyces being a good producer of staurosporine and some derivatives.  相似文献

Amniotic membrane (AM) is frequently used in ophthalmologic surgery for rapid ocular surface reconstruction. Sometimes it may create a major problem with associated infections after biofilm formation over the membrane. To overcome this problem, AM was coated with the antimicrobial peptide clavanin A. The antifungal activity of clavanin A in the native and self-assembled form was determined against the common ocular surface pathogens Candida albicans, Aspergillus fumigatus, Alternaria sp. and Fusarium sp. Biofilm formation over the coated surface was significantly reduced in comparison with the uncoated membrane. The coated membrane revealed effectiveness in terms of biocompatibility, cell attachment colonization when tested in non-cancerous 3T3 and human embryonic kidney (HEK)-293 cell lines. Clavanin A-coated AM also exhibited excellent physical, morphological and antifungal characteristics, indicating potential applicability for ocular surface infection control.  相似文献

The use of plant‐based compounds to control insect pests is an alternative to the use of synthetic pesticides. We evaluated the repellent and antifeedant effects of Cordeauxia edulis Hemsley (Fabaceae) and Rhododendron tomentosum Harmaja (Ericaceae) extracts against Hylobius abietis L. (Coleoptera: Curculionidae) and Phyllodecta laticollis Suffrian (Coleoptera: Chrysomelidae). Repellent properties were evaluated by monitoring responses of adult insects toward the odor from extracts or extraction solvents (controls) in a Y‐tube olfactometer, and choice or no‐choice feeding tests were conducted by applying extracts or extraction solvents on stems of Scots pine [Pinus sylvestris L. (Pinaceae)] and European aspen [Populus tremula L. (Salicaceae)], host plants of H. abietis and P. laticollis, respectively. Extracts of R. tomentosum repelled adults of both insect species effectively. However, extracts of C. edulis did not repel H. abietis although its ethyl acetate extract showed repellence against the adults of P. laticollis. Feeding by H. abietis was significantly reduced by a methanol extract of C. edulis, and methanol and hexane extracts of R. tomentosum. Feeding by the adults and larvae of P. laticollis was significantly reduced by extracts from both plant species. Concomitant with less feeding, larval growth was retarded by ethyl acetate extracts of both plant species. Gas chromatography‐mass spectrometry analyses of the volatile components of the extracts showed that extracts from both plant species were mixtures of various terpene and non‐terpene compounds, which showed quantitative and/or qualitative variations between plant species and extraction solvents. This experiment showed that extracts from both plant species effectively manipulated the orientation and/or feeding behavior of the two beetle species. Hence, they may be considered as potential alternatives to synthetic chemical pesticides.  相似文献

Caesalpinia crista seed extracts were investigated in the laboratory against Helicoverpa armigera (Hubner). The extracts exhibited powerful antifeedant and growth disruption activity. The toxic symptoms of the poisoned larvae included reduction in weight gain and mortality of larvae and pupae, larval-pupal intermediates and malformed adults. Maximum anti-feedance is caused by methanol extract (AI50=0.0186%), followed by hexane extract (0.0212%), ethyl acetate extract (0.0416%), butanol extract (0.0767%) and aqueous extract (0.2199%). The larval mortality ranged from 10.00 to 70.00% in different extracts. The 50% larval growth inhibition recorded at 3DAT was statistically at par by methanol and hexane extract. The percent 150 values for inhibiting normal adult emergence were 0.0287, 0.0325, 0.0485, 0.0977 and 0.0547% for methanol, hexane, ethyl acetate, aqueous and butanol extract. The biosafety evaluation of these extracts carried out against the predator, Coccinella septumpunctata showed no mortality of the adults till nine days after treatment. Though the observation taken at 10 DAT, showed slight mortality of adults by methanol extract at both 5.0 and 1.0% concentration.  相似文献
