The process of strain selection is an important step in the development of insect pathogens for biological control. Bioassays were conducted in the laboratory to evaluate the efficacy of different methods of inoculation using Rhipicephalus pulchellus Gerstäcker (Acari: Ixodidae) as a model. Initially, an oil-based formulation of Metarhizium anisopliae (Metsch.) Sorok. (Ascomycota: Hypocreales) titred at 109 conidia ml?1 was applied to R. pulchellus adults using a Burgerjon spray tower or a microapplicator. Inoculation by microapplicator yielded poor results (25.0% tick mortality) compared to Burgerjon’s spray tower (52.3% tick mortality), although the mean number of fungal conidia on R. pulchellus adults was lower (1.5 × 104 ± 1.1 × 103 conidia ml?1) after spraying by Burgerjon’s spray tower compared to 1 × 106 conidia ml?1 obtained with the microapplicator. Thus, inoculation by Burgerjon’s spray tower was selected for further investigations. Different modes of inoculation were tested and included direct spray of inoculum on the tick and substrate (SS), direct spray on the substrate and tick followed by transfer of the tick to clean uncontaminated Petri dish (SP) or indirect inoculation of ticks through substrate (SW). The LC50 values following contamination of nymphs (LC50 = 1.4 × 107 conidia ml?1) and adults (LC50 = 6.7 × 107 conidia ml?1) in SS were significantly lower compared to SP; nymphs (LC50 = 5.7 × 108 conidia ml?1) and adults (LC50 = 5.3 × 109 conidia ml?1) and SW; nymphs (LC50 = 5 × 108 conidia ml?1). Although the LC50 value in SS was the lowest, it recorded the highest tick mortality among control ticks (24.2% at 2 weeks post-treatment) and (23.3% at 3 weeks post-treatment) in nymphs and adults respectively compared to SP (2.5 and 5.8%, respectively) and SW (0.0 and 0.0). Results show that among the modes of inoculation tested, SP was the most appropriate for inoculating R. pulchellus adults. SW and SP were identified as appropriate techniques for infecting the R. pulchellus nymphs with conidia formulated in oil.  相似文献

A preliminary virulence test of four fungal isolates, Beauveria bassiana IMI 382302, Beauveria bassiana IMI 386701, Trichoderma harzianum T24 and Aspergillus flavus Link against larvae of Spodoptera littoralis was performed. The most effective isolates against larvae of S. littoralis were B. bassiana 302 and T. harzianum T24, which also showed the lower percentage of pupation compared with the other two isolates under the same conditions of treatments. Three concentrations (1 × 106, 1 × 107 and 1 × 108 ml?1) of the aqueous conidial suspension of the four tested isolates were carried out against both larval and pupal stages of S. littoralis within five days post-treatment. T. harzianum T24 showed 80% larval mortality only when applied at the highest conidial concentration, while A. flavus showed 100% pupal mortality only, at all of its conidial concentrations. However, B. bassiana IMI 382302 showed relatively high dose-dependant larval and pupal mortalities, while strain IMI 386701 of B. bassiana showed a very weak mortality against pupae at higher concentrations, and no virulence against larvae was recorded. Enzymatic and antibiosis bioassays of the four fungal isolates showed relatively high activities against Fusarium spp. for most of the tested isolates. Clear zone of enzyme activity on agar plates proportionally increased with increasing the concentration of enzyme substrate and prolongation of the incubation period. Mtabolites produced in the agar culture inhibited the growth of Fusarium spp. and the productivity differed greatly among isolates or strains of the same isolate. Volatile and non-volatile compounds produced by A. flavus Link showed a higher inhibition activity against Fusarium spp. compared with the other fungal isolates. The humoral antifungal response of insect host is relatively high compared to the anti-bacterial one. Injection of larvae with the immune sensitive bacteria Micrococcus luteus (5 × 103 bacteria/larva) showed a detectable humoral response by 2 h, peaked around 12 h and became hardly detectable by 24 h post-injection. Injection of larvae with conidial suspension (5 × 103 conidia/larva) from each of the fungal isolates showed humoral antifungal activity against B. bassiana IMI 386701 and A. flavus only. This activity was detectable by 12 h, peaked around 36 h and became hardly detectable by 48 h post-injection. Although the humoral antifungal response was started slowly compared to the antibacterial one, it lasted for longer and enabled larvae to withstand the infection with these immune-sensitive fungal strains. No humoral activity was detected against B. bassiana IMI 382302, although however, weak activity was detected against T. harzianum T24 only at the low conidial concentration but not at the higher one (1 × 108 ml?1). Thus, this study concludes that larvae of S. littoralis showed immune-dependant sensitivity to T. harzianum T24 and B. bassiana IMI 382302. Therefore, this study may recommend these two fungal isolates as mycoinsecticides in the battle against cotton leaf worm in Egypt. Hence, they have been selected for future comprehensive bioassays in the laboratory under conditions similar to that in the field. This, in fact, may help for developing effective mycoinsecticides against this pest. Penetration mechanims of insect cuticle by entomopathogenic fungi will be discussed.  相似文献

In order to replace the conventional chemical pesticides, extensive researches have been done on entomopathogenic fungi. Entomopathogenic fungus Beauveria bassiana is an important biocontrol agent against major economic pests and is being employed in Integrated pest management (IPM) along with synthetic pesticides. Cabbage aphid Brevicoryne brassicae L. is one of the important pests of Brassicaceae family. Therefore, in this research, the virulence isolate of B. brassicae (IRAN 429C) was investigated on adults of cabbage aphid under laboratory conditions. The experiments were conducted at 25 ± 2 °C, 60 ± 10 R. H. and a photoperiod of 16:8 (L: D). After preliminary experiments, the adult aphids were treated with fungal concentrations of 1 × 103 to 1 × 107 spores/ml. Probit analysis was conducted to calculate LC50 and LC95 values for the isolate. Positive correlation was observed between concentrations and pest mortality. LC50 and LC95 values calculated for IRAN 429C isolate are 2.04 × 105 and 1.82 × 108, respectively. The mortality was counted one day after the treatment and then continued for 14 days. Cumulative mortality for 14 days after treatment varied from 54% for IRAN 429C at low concentration (103 conidia/ml) to 83% at high concentration (107 conidia/ml). The lowest LT50 was obtained at 7.67 days for IRAN 429C isolate at concentration 1 × 107 spore/ml. According to the insecticidal activity of mentioned fungi on cabbage aphid, it can be used in biocontrol programmes of B. brassicae.  相似文献

The pathogenicity of four isolates each of the entomopathogenic fungi, Beauveria bassiana (Bals.) Vuill. and Metarhizium anisopliae (Metsch.) Sorok. to apterous adult Aphis craccivora Koch was evaluated in the laboratory at 4 concentrations of conidia. All fungi isolates tested were found to be pathogenic to the insect but their virulence varied among species and isolates within species. Three isolates, B. bassiana CPD 11 and M. anisopliae CPD 4 and 5 caused significantly higher mortality than the other isolates at the various concentrations tested causing mortality of between 58–91%, 64 to 93% and 66–100%, respectively, at 7 days post treatment. At the highest concentration of 1 × 108conidiaml‐1, these isolates produced the shortest LT50s of 3.5, 3.6 and 3.4 days, respectively. Their LC50s were 6.8 × 105, 3.1 × 105 and 2.7 × 105 conidia ml‐1, respectively. The results indicate that these isolates are promising candidates for the control of the cowpea aphid but their pathogenicity to various aphid non‐target beneficial organisms within the cowpea agroecosystem warrant further investigation before initiating field control.  相似文献

Plectosphaerella cucumerina (Lindf.) W. Gams was evaluated as a bioherbicide for Cirsium arvense L. (Scop.) using a Canadian and a New Zealand isolate. Both isolates defoliated C. arvense when applied at 1013 conidia ha?1 in water volumes ranging from 250 to 6400 l ha?1 with a rapid decline in effect with declining conidial dose. Repeat application and the addition of the adjuvant Pulse® penetrant to the conidial suspension increased the disease severity in C. arvense. Maximum disease occurred at 20 °C with a 48 h post-application dew period. The experiments demonstrate that P. cucumerina can defoliate C. arvense under the environmental conditions of temperate pastures where the weed is problematic. The results also show that modifications to formulation and strategic application may reduce the 48 h dew period requirement and risk to non-target species respectively, supporting the conclusion that the fungus has potential as a bioherbicide for C. arvense.  相似文献

Aims: Larval stages of Frankliniella occidentalis are known to be refractory to fungal infection compared with the adult stage. The objective of this study was to identify promising fungal isolate(s) for the control of larval stages of F. occidentalis. Methods and Results: Ten isolates of Metarhizium anisopliae and eight of Beauveria bassiana were screened for virulence against second‐instar larvae of F. occidentalis. Conidial production and genetic polymorphism were also investigated. Metarhizium anisopliae isolates ICIPE 7, ICIPE 20, ICIPE 69 and ICIPE 665 had the shortest LT50 values of 8·0–8·9 days. ICIPE 69, ICIPE 7 and ICIPE 20 had the lowest LC50 values of 1·1 × 107, 2·0 × 107 and 3·0 × 107 conidia ml?1, respectively. Metarhizium anisopliae isolate ICIPE 69 produced significantly more conidia than M. anisopliae isolates ICIPE 7 and ICIPE 20. Internally transcribed spacers sequences alignment showed differences in nucleotides composition, which can partly explain differences in virulence. Conclusion: These results coupled with the previous ones on virulence and field efficacy against other species of thrips make M. anisopliae isolate ICIPE 69 a good candidate. Significance and Impact of the Study: Metarhizium anisopliae isolate ICIPE 69 can be suggested for development as fungus‐based biopesticide for thrips management.  相似文献

Management of the banana root borer (BRB), Cosmopolites sordidus (Germar; Coleoptera: Curculionidae), remains a challenge in banana and plantain production worldwide. Synthetic pesticides remain the most widely used solution while mycoinsecticides are increasingly being recommended. In this study, we selected indigenous isolates of Beauveria bassiana and Metarhizium anisopliae collected from plantain fields in Cameroon, and tested them in the laboratory for their viability, pathogenicity and virulence against all C. sordidus life stages. Of 13 isolates initially screened for spore germination and pathogenicity to adult weevils in conidial suspension of 3.2 × 108 conidia/ml, eight isolates with high to moderate germination and highest weevil mortality were selected for dose–response bioassays with four concentrations per isolate: 3.2 × 102, 3.2 × 104, 3.2 × 106 and 3.2 × 108 conidia/ml. The virulent isolates from adult bioassays were tested with eggs, larva and pupae in conidial suspension of 3.2 × 108 conidia/ml. Isolates performance depended on insect life stage with significantly high pathogenicity and virulence against larval, pupa and adult stages. The Beauveria isolate BIITAC6.2.2 caused the highest mortality rates followed by MIITAC1.1.5. Lethal times and lethal concentrations were relatively low for the three M. anisopliae isolates and three B. bassiana isolates which were the best isolates in almost all insect life stages. Apart from being effective in multiple life stages, these isolates were transmitted horizontally from one stage to another when eggs and pupae were treated. The implication of these findings for integrated management of the BRB, and potential biopesticides development and commercialization are discussed.  相似文献

The sensitivity of Alternaria solani isolates to the fungicides mancozeb and chlorothalonil was evaluated, to determine if inadequate disease management by these fungicides could be attributed to reduced sensitivity of A. solani isolates to these fungicides. The sensitivity of 60 isolates of A. solani was assessed using the inhibition of radial mycelial growth (RG) method, using fungicide concentrations of 0, 1.0, 10, 100, 500 and 1000 μg a.i ml?1 medium. EC50 was calculated for each isolate and fungicide combination. The EC50 values of different A. solani isolates to mancozeb ranged from 9.05 to 712.65 μg ml?1. EC50 values of different isolates to chlorothalonil ranged from 4.25 to 849.4 μg ml?1. The percentage of isolates with reduced sensitivity was 46.7 and 53.3% for mancozeb and chlorothalonil, respectively. Results of the in vivo tests demonstrated decline in disease control by the two fungicides with the reduced-sensitivity isolates compared to the sensitive ones.  相似文献

Twenty local isolates of entomopathogenic fungi were determined for control of the larvae and adults of Culex quinquefasciatus. In a laboratory experiment, a Penicillium sp. CM-010 caused 100 % mortality of third-instar larvae within 2 h using a conidial suspension of 1 × 106 conidia ml?1. Its LC50 was 3 × 105 conidia ml?1, and the lethal time (LT50) was 1.06 h. Cloning and sequencing of its internal transcribed spacer region indicated that this Penicillium species is Penicillium citrinum (100 % identity in 434 bp). Mortality of the adult was highest with Aspergillus flavus CM-011 followed with Metarhizium anisopliae CKM-048 from 1 × 109 conidia ml?1. P. citrinum CM-010 at 1 × 106 conidia ml?1 killed 100 % larvae within 2 h while Bacillus thuringiensis var. israelensis at 5 ITU ml?1 required 24 h. This P. citrinum CM-010 also greatly reduced survival of C. quinquefasciatus larvae in an unreplicated field test. Light and transmission electron micrographs showed that the fungal conidia were ingested by the larvae and deposited in the gut. The metabolite patulin was produced by P. citrinum CM-010 instead of citrinin.  相似文献

Microsclerotia of the entomopathogenic fungus (Metarhizium brunneum Petch strain F52; Hypocreales: Clavicipitaceae) were sprayed in a hydromulch formulation onto wood samples to test persistence and biological control efficacy when exposed on forest and orchard trees. The density of viable conidia and retention of hydromulch were not significantly affected by the tackifier in the formulation (psyllium, xanthan, or a mixture of both). Forest samples had a maximum conidial density and more viable conidia (7.2 × 105 conidia cm?2) than orchard samples (1.4 × 105 conidia cm?2). Increased rainfall, temperature and the time that samples were outdoors were critical factors for conidial density. Asian longhorned beetles [Anoplophora glabripennis (Motschulsky); Coleoptera: Cerambycidae] exposed to orchard samples in laboratory bioassays did not have significantly less mortality compared to forest samples, indicating that hydromulch may be feasible in open sunny conditions, if sufficient moisture is present.  相似文献

The ability of isolates of Paenibacillus spp. to protect Brassica oleracea var. capitata (cabbage) against the black rot pathogen, Xanthomonas campestris pv. campestris (Xcc),was evaluated. Twenty-four isolates of Paenibacillus spp., isolated from New Zealand-grown brassica hosts or soil, were evaluated for in vitro antagonism towards six Xcc isolates. Seven Paenibacillus spp. isolates with different levels of in vitro suppressive activity against Xcc were screened in pot experiments for their capacity to reduce black rot symptoms on cabbage. Two Paenibacillus isolates (P10 and P16) exhibited biocontrol activity against Xcc, and four isolates (P1, P6, P9, and P24) reduced cabbage seed germination and seedling emergence. The dependence of bioactivity on inoculum rate was investigated with three Paenibacillus isolates (P6, P10, and P16) at three different concentrations (5?×?108, 5?×?109, and 5?×?1010?CFU?ml?1). Negative effects on seedling emergence were detected with isolate P6 at concentrations?≥5?×?109?CFU?ml?1. All three isolates applied at the three concentrations reduced black rot symptoms on the cotyledons and true leaves. There was poor or no relationship between the inhibitory effect of Paenibacillus spp. isolates on the growth of Xcc in vitro, and their biocontrol activity in vivo. Paenibacillus isolate P16 was identified as a potential biological control of black rot in cabbage.  相似文献

Lifetable demography and reproductive traits of a Kenyan strain of the rotifer Brachionus angularis were investigated using individual and small batch culture approaches. The rotifer was identified morphologically before conducting studies at 20, 25 and 30 °C, using Chlorella vulgaris at 2.5 × 105 to 2.5 × 107 cells ml–1. The rotifers were highly fecund, producing 2.11 ± 0.07 offspring female–1 day–1 and reproductive, producing 8.43 ± 0.24 offspring female–1 at 25 °C with 2.5 × 106 algal cells ml–1. The highest intrinsic rate of natural increase (0.74 ± 0.02 d–1), specific population growth rate (0.49 ± 0.01), longest life expectancy at hatching (12.41 ± 0.28 d) and shortest generation time (2.87 ± 0.03 d) also occurred at 25 °C with 2.5 × 106 algal cells ml–1. The duration of hatching to first spawning was shortest (2.86 ± 0.21 h) at 30 °C with 2.5 × 107 algal cells ml–1 and longest (8.83 ± 0.39 h) at 20 °C with 2.5 × 105 algal cells ml–1. The highest population density (255.7 ± 12.6 ind. ml–1) was realised at 25 °C with 2.5 × 106 cells ml–1 on Day 8, whereas the lowest population density (122.0 ± 3.6 ind. ml–1) was realised at 20 °C with 2.5 × 105 cells ml–1 on Day 8. The lorica length and width of the Kenyan strain of B. angularis are 85.6 ± 3.1 µm and 75.4 ± 3.6 µm, respectively. The rotifer optimally reproduces at 25 °C when fed with 2.5 × 106 algal cells ml–1.  相似文献

Abstract

Microsclerotia (MS) production by two isolates of Lecanicillium lecanii on various culture media is described, and the efficacy of MS against western flower thrips is evaluated. High concentrations of MS (2.9–3.1×105·mL?1) were produced in media with C:N ratios of 7.4:1 and 10.3:1 by isolate SN21. Bioassays using soil-incorporated MS resulted in significant infection and mortality of thrips.  相似文献

The cypress aphid (Cinara cupressi) is listed among the hundred most important invasive pests in the world. In Chile, it was first detected in 2003 and currently is present throughout the country. In the course of a survey of their natural enemies in Chile, three strains of entomopathogenic fungi were isolated. The isolates were identified and tested against the aphid in laboratory experiments. Two further entomopathogenic fungi (ARSEF 5126 and 5128), formulated in the mycoinsecticides Vertalec® and Mycotal®, were used as reference strains. The three Chilean isolates were identified genomically as Lecanicillium attenuatum and were pathogenic to third-instar nymphs. The isolate ARSEF 13279 yielded the lowest overall lethal concentration (LC50), 3 × 105 conidia ml?1 at four days post-inoculation, and the shortest lethal time (LT50), 3.7 days after inoculation with 106 conidia ml?1. The results indicate that the isolates have considerable potential as microbial control agents of the invasive cypress aphid.  相似文献

The Mediterranean fruit fly, Ceratitis capitata Wiedemann (Diptera: Tephritidae), is the major tephritid pest in Morocco. This pest survives in Moroccan forests Argania spinosa and continually invades the nearest agricultural areas. Entomopathogenic fungi are an interesting tool for fruit fly control and hold a useful alternative to conventional insecticides. However, primary selection of effective pathogens should be taken in laboratory condition prior to applying them in the field. Here, we used third late instar larvae of C. capitata to investigate the effectiveness of 15 local Beauveria bassiana isolates. Results showed that all isolates were able to infect the larval stage, producing a large mortality rate in puparia ranging from 65 to 95 % and caused significant reduction in adult emergence. The fungal treatments revealed that the mycosis occurred also in adults escaping infection as pupariating larvae. The percentage of mycosed puparia was highest in strain TAM6.2 (95 %) followed by ERS4.16 (90 %), therefore they were the most virulent. Median lethal concentration (LC50) was studied for five isolates at four concentrations ranging from 105 to 108 conidia ml?1. The results showed that the slopes of regression lines for B. bassiana ERS4.16 (slope = 0.386) and TAM6.2 (slope = 0.41) were the most important and had the lowest LC50 values (2.85 × 103 and 3.16 × 103 conidia ml?1 respectively). This investigation suggests that the soil of Argan forests contains pathogenic B. bassiana isolates and highlights for the first time their potential as biological control toward C. capitata larval stage in Morocco.  相似文献

A total of 10 endophytic actinomycete strains were successfully isolated from healthy shoots and roots of Aquilaria crassna Pierre ex Lec (eaglewood). Analysis of 16S rDNA sequencing of those isolates showed that they belong to members of the genera Streptomyces (2 isolates), Nonomuraea (1 isolate), Actinomadura (1 isolate), Pseudonocardia (1 isolate) and Nocardia (3 isolates). The remaining 2 isolates were unidentified. All of isolates produced the amount of indole-3-acetic acid (IAA) and ammonia ranging between 9.85 ± 0.31 to 15.14 ± 0.22 μg ml?1 and 2 to 60 mg ml?1, respectively. Among 10 isolates tested, the amount of hydroxamate-type siderophore produced by 2 isolates was undetectable. While the remaining 8 isolates produced the amount of hydroxamate-type ranging between 3.21 ± 0.12 and 39.30 ± 0.40 μg ml?1. Also, catechols-type siderophore produced by 9 isolates was undetectable. Actinomadura glauciflava is only one isolate that produced catechols-type 4.12 ± 0.90 μg ml?1. In addition, 10 endophytic actinomycetes showed protease activity ranging from undetectable to 8.16 ± 0.15 unit ml?1. Genetic relatedness amongst these isolates was determined base on Random amplified polymorphic DNA (RAPD) and Enterobacterial repetitive intergenic consensus polymerase chain reaction (ERIC PCR). Both methodologies generated specific patterns corresponding to particular genotypes. RAPD fingerprinting proved to be slightly more discriminatory than ERIC PCR. This study is the first published report that actinomycetes can be isolated as endophytes within this plant. It is also the first published report that endophytic actinomycetes are capable of producing IAA and siderophores.  相似文献

Some morphological and physiological characteristics of an Isaria fumosorosea isolate with diminished virulence, IFCF01-D, and its parent isolate, IFCF01, were evaluated and laboratory bioassays were performed to assess their virulence against Plutella xylostella. The relationship among these traits and virulence against P. xylostella is discussed. There were no significant differences in conidial viability, spore production and the time required for 50% germination (GT50). Spore viability after incubation for 24 h at 25°C was greater than 98% for both isolates tested. Spore production on potato dextrose agar after 14 days incubation at 25°C was 4.68 × 108 and 4.59 × 108 conidia/mL for IFCF01 and IFCF01-D, respectively. When exposed to high temperatures (40, 45, 50 or 55°C) through a water bath for 10 min, conidial germination ranged from 0.83% to 84.0% for IFCF01 and 0% to 86% for IFCF01-D. Germination rate showed a negative relationship with the exposure temperature for both isolates. The per cent germination of isolate IFCF01 24 h after ultraviolet (UV) radiation (18 W, 240–260 nm) varied from 0% to 92% and 0% to 81% for IFCF01-D. Germination rate and the exposure time exhibited a negative correlation for both isolates tested. Conidial surface hydrophobicity of IFCF01 (60%) was significantly higher than that of isolate IFCF01-D (53%). Subsequently, using the cicada exuviae as the substrate for enzymatic analysis, Pr1 and chitinase activity demonstrated the contrasting virulence traits: higher specific activities for the more virulent IFCF01 and lower enzymatic levels for isolate IFCF01-D.  相似文献

The pathogenicity of 15 isolates of Beauveria bassiana (Balsamo) Vuillemin, five isolates of Metarhizium anisopliae (Metschnikoff) Sorokin and one isolate of M. flavoviride (Gams and Rozsypal) were tested under laboratory conditions against the subterranean life stages of the citrus pests, Ceratitis rosa Karsch, C. capitata Wiedemann (Diptera: Tephritidae) and Thaumatotibia leucotreta Meyrick. (Lepidoptera: Tortricidae). When these citrus pests were treated with a concentration of 1×107 conidia mL?1, fungal isolates had a significantly greater effect on the adults of C. rosa and C. capitata than they did on the puparia of these two fruit fly species. Further, C. rosa and C. capitata did not differ significantly in their response to entomopathogenic fungi when adult and pupal mycosis were considered. Depending on fungal isolate, the percentage of T. leucotreta adults which emerged from fungal treated sand ranged from 5 to 60% and the percentage of pupae with visible signs of mycosis ranged from 21 to 93%. The relative virulence of the four most promising fungal isolates, as well as the commercially available B. bassiana product, BroadBand® (Biological Control Products, South Africa), were compared against one another as log-probit regressions of mortality against T. leucotreta which exhibited a dose-dependent response. The estimated LC50 values of the three most virulent B. bassiana isolates ranged from 6.8×105 to 2.1×106 conidia mL?1, while those of the least pathogenic ranged from 1.6×107 to 3.7×107 conidia mL?1.  相似文献

Methods and Results: Kocuria SM1, isolated from the digestive tract of rainbow trout, was administered orally to rainbow trout (Oncorhynchus mykiss) for 2 weeks at a dose equivalent to c. 108 cells per g of feed and then challenged intraperitoneally with V. anguillarum and V. ordalii. Use of SM1 led to a reduction in mortalities to 15–20% compared to 74–80% mortalities in the controls. SM1 stimulated both cellular and humoral immune responses in rainbow trout, by elevation of leucocytes (5·5 ± 0·8 × 106 ml?1 from 3·7 ± 0·8 × 106 ml?1), erythrocytes (1·2 ± 0·1 × 108 ml?1 from 0·8 ± 0·1 × 108 ml?1), protein (23 ± 4·4 mg ml?1 from 16 ± 1·3 mg ml?1), globulin (15·7 ± 0·2 mg ml?1 from 9·9 ± 0·1 mg ml?1) and albumin (7·3 ± 0·2 mg ml?1 from 6·1 ± 0·1 mg ml?1) levels, upregulation of respiratory burst (0·05 ± 0·01 from 0·02 ± 0·01), complement (56 ± 7·2 units ml?1 from 40 ± 8·0 units ml?1), lysozyme (920 ± 128·8 units ml?1 from 760 ± 115·3 units ml?1) and bacterial killing activities. Conclusions: Kocuria SM1 successfully controlled vibriosis in rainbow trout, and the mode of action reflected stimulation of the host innate immune system. Significance and Impact of the Study: Probiotics can contribute a significant role in fish disease control strategies, and their use may replace some of the inhibitory chemicals currently used in fish farms.  相似文献

Abstract The red turpentine beetle (RTB), Dendroctonus valens LeConte, as a destructive invasive pest, has become one of the most economically important forest pest in China. Effective control measures are desperately needed. Entomopathogenic fungi, such as Beauveria bassiana, have shown great potential for the management of some bark beetle species. In this study, 12 isolates of B. bassiana from bark beetle were examined for biological characteristics and virulence, to assess their potential as biocontrol agents for RTB. There were significant differences (at P= 0.05) in colony growth rate, conidial yield, conidial germination, tolerance to UV light and extracellular proteases activity among the tested B. bassiana isolates. Isolates, including Bb1801, Bb1906, Bb789 and Bb773, exhibited the best characteristics, because they have faster hyphal growth rate, higher spore production and faster spore germination, higher UV tolerance and protease (Pr1) production. The results of a pathogenicity test of B. bassiana on RTB larvae showed that most isolates of B. bassiana have demonstrated high efficacy and the highest virulent isolate was Bb1801, which killed 100% of the treated insects and had a median lethal time (LT50) of 4.60 days at a concentration of 1×107 conidia/mL. Therefore, isolate Bb1801 has a great potential for sustainable control of RTB in the forest. The correlation between biological characteristics and virulence of the fungal isolates is discussed and the possibility of combination of entomopathogenic fungi with semiochemicals, as one of the promising strategy for RTB control, is considered.  相似文献
