To evolve eco-friendly management of fenugreek root rot caused by Rhizoctonia solani, a field trial was conducted during Kharif 2002 and Rabi seasons of 2002–2003 and 2003–2004. Experiments were conducted with eight treatments and three replications in RBD using the variety CO-2. The pooled analysis of the three season data showed that seed treatment with Trichoderma viride at 4g/kg of seed + soil application of Trichoderma viride at 5 kg/ha + soil application of neem cake at 150 kg/ha (T3) recorded a percent disease index (PDI) of 23.1 versus 65.5 PDI in the control which accounted for a disease reduction of 64.7%. It was on par with seed treatment with Trichoderma viride at 4g/kg of seed + soil application of T. viride at 5 kg/ha (T2) which reduced the disease incidence by 62.3% (24.7 PDI). The chemical treatment used for comparison, i.e. seed treatment with carbendazim + soil drenching at 0.1% + soil application of neem cake at 150 kg/ha recorded the lowest PDI of 16.8 with 74.4% disease reduction. Among the various treatments T3 gave a seed yield of 572.7 kg/ha followed by T2 (555.7 kg/ha). Treatment T7 recorded the highest yield of 578.7 kg/ha. In the control plot the recorded yield was only 359.3 kg/ha. Though T3 was more effective at reducing the disease incidence than T2, the C:B ratio was higher (1:9.1) in respect of T2 than T3, which gave a C:B ratio of only 1:3.9. Hence, seed treatment with T. viride at 4g/kg + soil application of T. viride at 5kg/ha is a cost effective, eco-friendly management strategy for fenugreek root rot. 相似文献

Biological control of fungi causing root rot on sugar beet by native Streptomyces isolates (C and S2) was evaluated in this study. The dry weight and colony forming unit (CFU) of S2 and C increased when 300 mM NaCl was added to medium. The in vitro antagonism assays showed that both isolates had inhibitory effect against Rhizoctonia solani AG-2, Fusarium solani and Phytophthora drechsleri. In dual culture, Streptomyces isolate C inhibited mycelial growth of R. solani, F. solani and P. drechsleri 45%, 53% and 26%, respectively. NaCl treatment of medium increased biocontrol activity of soluble and volatile compounds of isolate C and S2. After salt treatment, growth inhibition of R. solani, F. solani and P. drechsleri by isolate C increased up to 59%, 70% and 79%, respectively. To elucidate the mode of antagonism, protease, chitinase, beta glucanase, cellulase, lipase and α-amylase activity and siderophore and salicylic acid (SA) production were evaluated. Both isolates showed protease, chitinase and α-amylase activity. Also, biosynthesis of siderophore was detectable for both isolates. Production of siderophore and activity of protease and α-amylase increased after adding salt for both isolates. In contrast, chitinase activity decreased significantly. Production of SA, beta glucanase and lipase by isolate S2 and biosynthesis of cellulase by isolate C were observed in presence and absence of NaCl. Soil treatment with Streptomyces isolate C inhibited root rot of sugar beet caused by P. drechsleri, R. solani and F. solani. Results of this study showed that these two Streptomyces isolates had potential to be utilized as biocontrol agent against fungal diseases especially in saline soils. 相似文献

Abstract The efficiency of two biocontrol agents (Trichoderma harzianum NB and Bacillus subtilis NB) and two commercial biocides (Plant Guard and Rhizo-N) in controlling Fusarium root rot disease on some citrus rootstocks was evaluated under artificially infested soil in green house. Fusrium root rot on citrus rootstocks seedlings i.e. sour orange (SO), volkamer lime (VL), rangpur lime (RP) and cleopatra mandarin (CL) was successfully controlled by dipping the root system of such seedlings in water suspensions of each biological treatment i.e. Trichoderma harzianum (spore suspension 5×106 spore/ml), Bacillus subtilis (cell suspension 8×107 cell/ml), Plant Guard (3 g/l) and Rhizo-N (4 g/l), then transplanted into artificially infested soil with Fusarium solani and drenched with enough water suspension of such biological treatments. Plant Guard (3 g/l) and Rhizo-N (4 g/l) were highly effective treatments in decreasing infection and severity of the disease, Fusarium density in rhizosphere soil and colonization of Fusarium solani in the roots of all tested seedlings. Meanwhile, root dipping or soil drenching with the same treatments individually gave the least effect in reducing root rot incidenceon all tested rootstocks compared with application of the two methods together. It should be noted that using biocontrol agents and commercial biocides could be successfully used in controlling root rot pathogens on citrus in commercial greenhouses or under field conditions before transplanting in new reclaimed lands in the desert. 相似文献

The effect of four bio-inoculants namely, Glomus aggregatum, Streptomyces sp., Bacillus subtilis and Trichoderma harzianum and a fungicide, Ridomil-mancozeb was evaluated on the biomass production and control of root rot and wilt disease under glasshouse conditions. Results showed that 21-day- prior inoculation with G. aggregatum was most effective where none of the treated plants produced disease symptoms and interestingly their growth was increased by 39.4%. The colonisation by G. aggregatum (>80%) also increased P concentration in shoot. While, similar treatment with Streptomyces sp., B. subtilis, T. harzianum and Ridomil-mancozeb individually failed to produce any significant effect over Rhizoctonia solani inoculated control, where all inoculated plant died, prematurely. The simultaneous and 3-day-post treatments of G. aggregatum were non-effective but simultaneous treatment with Streptomyces sp. produced 70% disease control, while B. subtilis and T. harzianum individually provided 50% control. Their effects were either better or at par when compared with the simultaneous treatment of Ridomil-mancozeb. In 3-day-post treatment, Streptomyces, B. subtilis and Ridomil-mancozeb individually provided 50% disease control, whereas T. harzianum was least effective as it could protect only 25% plants against infection. The results reveals that 15 days prior treatment of G. aggregatum can significantly controls the root rot and wilt disease of pyrethrum. Further, treatment of Streptomyces can also serve the next effective post infection control method. 相似文献

Rhizoctonia solani isolates used in this investigation were identified as anastomosis-4 (AG-40), collected from different localities from Assiut governorate in Egypt. Pathogenicity test of seven isolates of R. solani was evaluated on soybean Giza 111 cultivar under greenhouse conditions. All tested isolates were able to infect soybean plants causing root rot with different degrees of severities, isolate No. 1, 2 and 3 showed significantly highest root rot severity, while isolate No. 5 gave the lowest percentage of root rot rating. The sodium dodecyl sulphate polyacrylamide gel electrophoresis patterns were used to compare three isolates of R. solani. There are no variations among R. solani isolates except a few exceptions according to their protein patterns. DNA markers obtained from all isolates showed genetic similarity among different isolates obtained from different geographical regions barring few exceptions. Correlation between DNA patterns of R. solani isolates and their virulence was detected, but no correlation with anastomosis groups (AG). 相似文献

Sesame (Sesamum indicum L.) is one of the most important oilseed crops in Egypt and worldwide. It is being infected with many pathogens, among these pathogens Fusarium oxysporum f.sp. sesami (Zap.) Cast is causing severe economic losses on sesame. In this study, antagonistic capability of 24 isolates of Trichoderma spp. was assessed in vitro against F. oxysporum f.sp. sesami. Two strains; T. harzianum (T9) and T. viride (T21) were revealed to have high antagonistic effect against F. oxysporum f.sp. sesami in vitro with inhibition percentage about 70 and 67%, respectively. These two isolates proved to have high ability to control Fusarium wilt disease under greenhouse conditions. The highest reduction in disease severity was achieved with T. viride followed by T. harzianum with reduction in disease severity about 77 and 74%, respectively. This study revealed that the time of application of bioagents is a decisive factor in determining the efficacy of Trichoderma isolates to control Fusarium wilt of sesame. It was revealed that the highest reduction in the disease severity was achieved when either Trichoderma viride or T. harzianum were applied 7 days before challenging with the F. oxysporum f.sp. sesami. 相似文献

Fourteen citrus species were screened for their resistance against dry root rot under artificial inoculation conditions and classified as resistant (RHRL-122, RHRL-124, Australian sour, Sour dig, Balaji, Rangpurlime), moderately resistant (PKM-1, AL-Srirampur, Rough lemon), Susceptible (TAL 95/1, TAL 95/2, TAL 95/3, Nalgonda selection) and highly susceptible (TAL 94/13). The higher polyphenol oxidase (PPO) activity was observed in all infected leaves and roots of citrus species when compared to healthy leaves and roots at 15 and 30 days after inoculation (DAI). Higher PPO activity was observed in Balaji, Australian sour and Rangpurlime whereas lowest PPO activity was observed in TAL 94/13. In the case of leaf Peroxidase (PEO) isozyme profile an additional band which was darker and thicker was observed at an Em value of 0.24 in the case of Australia sour, Balaji and AL-Srirampur. In roots the PEO isozyme profile has the induction of single thick and darker additional band with an Em value of 0.47 was observed in Australian sour and Balaji. The banding profiles of estarase in leaves showed the induction of an extra band in the ase of Australian sour at Em 0.1, and at Em 0.53 in the case of Rangpurlime and Sourdig when compared to other species. The banding profile of esterase in roots was well expressed in Australian sour, PKM-1, Rough lemon, TAL-95/3, Rangpurlime and Sour dig. However, a minor band at Em 0.27 in Australian sour, TAL 95/1 and at Em 0.33 in Balaji was observed. 相似文献

利用徒手切片、石蜡切片和超薄切片及显微摄像的方法,对当归根的显微结构及根腐病致病真菌的分布进行了研究。结果表明:当归的根由周皮和次生维管组织两部分组成,周皮由外向内依次分为木栓层、木栓形成层、栓内层;次生韧皮部占根径的比例在60%以上,主要成分包括筛胞、韧皮薄壁细胞、韧皮纤维和分泌道,薄壁细胞富含淀粉粒等营养物质;次生木质部由导管、木薄壁细胞和木射线组成,木质部呈多元形,木射线和韧皮射线明显。在根的周皮细胞和中柱中均有真菌分布,说明真菌由木栓层、木栓形成层、栓内层依次向里侵入到韧皮薄壁细胞,在薄壁细胞内定殖并形成菌丝结或团块状结构,进而扩展成一定的侵染区域;真菌不仅侵染周皮和韧皮部,而且还进一步侵染木质部并破坏导管。此外,研究还发现,淀粉粒是真菌定殖的主要场所,真菌穿透或缠绕在淀粉粒上,并利用其营养不断地生长与繁殖。 相似文献

Novel strains of rhizobacteria, Pseudomonas fluorescens Pf 9A‐14, Pseudomonas sp. Psp. 8D‐45 and Bacillus subtilis Bs 8B‐1, showed broad‐spectrum antagonistic activity and provided suppression of Pythium damping‐off and root rot of cucumber. Their biocontrol potential was further investigated for suppression of additional seedling diseases of cucumber (Phytophthora capsici) and radish (Rhizoctonia solani). Bacterial strains were also characterised for production of antibiotics, metabolites, volatiles, phytohormones and lytic enzymes. Seed and pre‐plant applications of all three antagonistic bacteria as cell suspension and talc or irradiated peat formulations to the infested potting mix provided overall high level of suppression of Phytophthora damping‐off and root rot of cucumber (66–85% healthy seedlings) and relatively low level of suppression of Rhizoctonia damping‐off of radish (18–38% healthy seedlings). Bacterial treatments also resulted in higher plant fresh masses. Seed coating with irradiated peat formulation of a mixture of three bacteria resulted in superior control of Phytophthora damping‐off and root rot of cucumber and much higher plant fresh masses. The presence of genes for biosynthesis of phenazine‐1‐carboxylic acid, 2,4‐diacetylphloroglucinol, pyrrolnitrin and pyoluteorin was confirmed in Pseudomonas strains, and that of fengycin, bacillomycin, bacilysin, surfactin and iturin A in B. subtilis Bs 8B‐1. All three strains produced HCN, salicylic acid, indole‐3‐acetic acid, protease and β‐1,3‐glucanase. Both Pseudomonas strains produced siderophores and only P. fluorescens Pf 9A‐14 showed phosphate solubilisation and chitinase activity. All three strains inhibited pathogen growth by producing volatiles, and gas chromatography–mass spectrometry analysis revealed eight compounds in Pf 9A‐14, 10 in Bs 8B‐1 and 4 in Psp 8D‐45, some with known antifungal activity. The antagonistic and plant‐growth promotion activities of these strains might be due to production of antibiotics, metabolites, lytic enzymes or phytohormones. 相似文献

Aims: To evaluate the potentiality of three rhizosphere microorganisms in suppression of Sclerotinia rot in pea in consortia mode and their impact on host defence responses. Methods and Results: Pseudomonas aeruginosa PJHU15, Trichoderma harzianum TNHU27 and Bacillus subtilis BHHU100 from rhizospheric soils were selected based on compatibility, antagonistic and plant growth promotion activities. The microbes were used as consortia to assess their ability to trigger the phenylpropanoid and antioxidant activities and accumulation of proline, total phenol and pathogenesis‐related (PR) proteins in pea under the challenge of the soft‐rot pathogen Sclerotinia sclerotiorum. The triple‐microbe consortium and single‐microbe treatments showed 1·4–2·3 and 1·1–1·7‐fold increment in defence parameters, respectively, when compared to untreated challenged control. Activation of the phenylpropanoid pathway and accumulation of total phenolics were highest at 48 h, whereas accumulation of proline and PR proteins along with activities of the antioxidant enzymes was highest at 72 h. Conclusions: The compatible microbial consortia triggered defence responses in an enhanced level in pea than the microbes alone and provided better protection against Sclerotinia rot. Significance and Impact of the Study: Rhizosphere microbes in consortium can enhance protection in pea against the soft‐rot pathogen through augmented elicitation of host defence responses. 相似文献

Abstract A study was conducted in the greenhouse to examine the resistance of three soybean cultivars against root-infecting fungi, and to determine the role of five strains of Pseudomonas aeruginosa in protecting the roots from these fungal pathogens. In this study soybean cv RAWAL was found to be less susceptible against charcoal rot fungus Macrophomina phaseolina than cvs PARC and BRAGG. Most of the strains of P. aeruginosa used as seed dressing significantly reduced M. phaseolina and Rhizoctonia solani infection on all three cvs PARC, BRAGG and RAWAL (p < 0.05). Most of the strains of P. aeruginosa were effective on cv PARC against Fusarium solani infection, while on cv BRAGG P. aeruginosa strain Pa3, and on cv RAWAL strain Pa5 were effective. Both strains Pa3 and Pa22 gave maximum plant height and fresh weight of shoots, respectively on cvs PARC and BRAGG than other strains. These characteristics make these P. aeruginosa strains good candidates for use as biocontrol agents against soil-borne plant pathogens. 相似文献

We have investigated in vitro antifungal efficiency of nitrogen-doped carbon nanohorn (NCNH) against Rhizoctonia solani (R. solani) plant pathogenic fungi. NCNH with size of 50–60 nm and concentrations of 10, 50, 100, and 150 μg mL?1 were used. The results showed that growth of fungi in the presence of NCNH was significantly (p > .05) inhibited at 150 μg mL?1 (85.13 ± .97) after 72 h. The results were validated through computational approaches. Molecular docking analysis of NCNH with endochitinase protein of R. solani was performed to validate the potential of antifungal activity of NCNH. Docking results showed different conformations of interaction of NCNH with endochitinase enzyme. The conformation with least binding energy ?13.54 kcal/mol was considered further. It is likely that NCNH interacts with the pathogens by mechanically wrapping, which may be one of the major toxicity actions of NCNH against R. solani. The analysis showed that NCNH might interwinds to endochitinase of R. solani leading to the deactivation of the enzyme. To best of our knowledge, this is the first report of antifungal efficacy of NCNH against R. solani and provides useful information about the application of NCNH in resisting crop disease. 相似文献

Eight fungicides (benodanil, carboxin, cyproconazole, fenpropimorph, fur-mecyclox, iprodione, pencycuron and tolclofos-methyl) were evaluated, under growth chamber conditions, as seed treatments against pre-emergence damping-off and post-emergence seedling root rot in six Brassica species. Five cultivars of B. rapa, four cultivars of B. juncea, four cultivars of B. napus and one cultivar/ strain from each of B. carinata, B. nigra and B. oleracea were grown in soilless mix infested with an isolate of Rhizoctonia solani AG-2-1. B. nigra and B. juncea were considerably less susceptible to R. solani than the four other species. Cyproconazole at 0.05-0.1 g a.i./kg seed and the other fungicides at 2–4 g a.i./ kg seed provided almost complete control of pre-emergence damping-off in most Brassica species and their cultivars. Their efficacy varied against the post-emergence seedling root rot. Furmecyclox, iprodione, tolclofos-methyl and pencycuron consistently gave good control of seedling root rot in all Brassica species and their cultivars. Benodanil and fenpropimorph provided moderate control, and carboxin and cyproconazole gave poor control against root rot. Efficacy of carboxin, cyproconazole, benodanil and fenopropimorph against seedling root rot varied significantly (P ≤ 0.05) among cultivars within a Brassica species. 相似文献

Induction of defense-related enzymes, such as phenylalanine ammonia lyase (PAL), peroxidase (PO), polyphenol oxidase (PPO), superoxide dismutase (SOD) and catalase (CAT) due to bacterial antagonists viz., Pseudomonas fluorescens (Pf1) and Bacillus subtilis (EPCO16) and plant-derived lipoxygenase volatile compound hexanal, were studied in mango fruits against Lasiodiplodia theobromae causing stem-end rot disease. The results showed increased induction of all the defense-related enzymes in mango fruits 3–5 days after dipping treatment with combination of bacterial antagonists and hexanal when compared to untreated control treatment and treatment with fungicide carbendazim in storage condition. The increased activity was observed up to 3 days after treatment and thereafter declined. Further, increased expression of specific isoforms of PO, PPO, SOD and CAT were also observed in the treatment effect of P. fluorescens (0.5%)?+?hexanal (0.02%) treated fruits against L. theobromae. From the results obtained, it is inferred that due to the enhancement of defense-related enzymes via the phenylpropanoid pathway and due to secretion of secondary metabolites that would play significant role in hindering the pathogen quiescence and further invasion in mango fruits and thereby prevent the fruit rot. 相似文献

Fusarium root rot in row crops is typically managed by cultural practices and fungicide seed treatments. Biological control using microbial agents is another option but needs further development for improved disease management. Screening to identify biocontrol agents are crucial. However, relationships among the steps and how to improve the screening process are unresolved questions. Strains of Burkholderia (4), Bacillus (5) and Trichoderma (26) were studied in vitro against six Fusarium pathogens. All the bacteria and five selected Trichoderma strains were tested in planta in the greenhouse against diseases of Fusarium graminearum and Fusarium oxysporum. Burkholderia ambifaria C628, Bacillus simplex R180, and all Trichoderma isolates showed high reduction in disease levels in corn, soybean and wheat, ranging from 16 to 63%. Responses of the biocontrol agents during in vitro and in planta screening did not always correlate. In vitro and in planta tests should be considered independently in selecting biocontrol candidates. 相似文献

This study investigated the antagonistic effects of Trichoderma harzianum isolate (TRIC8) on mycelial growth, hyphal alteration, conidial germination, germ tube length and seed colonization by the seedborne fungal pathogens Alternaria alternata, Bipolaris cynodontis, Fusarium culmorum and F. oxysporum, the causes of seedling rot in over 30% of sunflowers. The antagonistic effect of TRIC8 on mycelial growth of pathogens was evaluated on dual culture that included two inoculation assays: inoculation of antagonist at 48 h before pathogen (deferred inoculation) and inoculation at the same time with pathogen (simultaneous inoculation). TRIC8 inhibited mycelial growth of the fungal pathogens between 70·67 and 76·87% with the strongest inhibition seen with deferred inoculation. Alterations in hyphae were observed in all pathogens. Conidial germination of F. culmorum was inhibited by most of the fungal pathogens (38·28%) by TRIC8. Inhibition of germ tube length by the antagonist varied from 31·83 to 37·67%. In seed colonization experiments, TRIC8 was applied in combination with each pathogen to seeds of a sunflower genotype that is highly tolerant to downy mildew. Seed death was inhibited by TRIC8 and the antagonist did not allow growth of A. alternata, B. cynodontis and F. culmorum on seeds and inhibited the growth of F. oxysporum at the rate of 58·32%. 相似文献

从大豆根围筛选到1株对尖孢镰刀菌和立枯丝核菌都具有很好拮抗作用的菌株KJB04-11,经形态观察、生理生化特征和16SrDNA序列分析,属于枯草芽孢杆菌(Bacillussubtilis)。具有抗菌活性的KJB04-11发酵液无菌滤液对热和酸碱具有较强的稳定性。采用SephadexG-25柱层析、反相HPLC和冷冻干燥从KJB04-11发酵液中分离纯化了抗菌活性成分。由红外光谱、MALDI-TOF-MS、氨基酸组成及脂肽合成酶基因扩增结果推测该菌株产生的抗菌物质为C16、C17的mycosubtilin和C15的surfactin。田间试验表明,大豆种子经KJB04-11发酵液包衣处理对大豆根腐病防效为53.6%,大豆产量提高12.5%。 相似文献

Endocrine-disrupting genistein was treated with the white rot fungus Phanerochaete sordida YK-624 under ligninolytic condition with low-nitrogen and high-carbon culture medium. Genistein decreased by 93% after 4 days of treatment and the activities of ligninolytic enzymes, manganese peroxidase (MnP) and laccase, were detected during treatment, thus suggesting that the disappearance of genistein is related to ligninolytic enzymes produced extracellularly by white rot fungi. Therefore, genistein was treated with MnP, laccase, and the laccase-mediator system with 1-hydroxybenzotriazole (HBT) as a mediator. HPLC analysis demonstrated that genistein disappeared almost completely in the reaction mixture after 4 h of treatment with either MnP, laccase, or the laccase-HBT system. Using the yeast two-hybrid assay system, it was also confirmed that three enzymatic treatments completely removed the estrogenic activity of genistein after 4h. These results strongly suggest that ligninolytic enzymes are effective in removing the estrogenic activity of genistein. 相似文献
