We previously reported that the dried peel powder of Citrus kawachiensis exerted anti-inflammatory effects in the brain in several animal models. Hyperglycemia induces inflammation and oxidative stress and causes massive damage in the brain; therefore, we herein examined the anti-inflammatory and other effects of the dried peel powder of C. kawachiensis in the streptozotocin-induced hyperglycemia mice model and in the type 2 diabetic db/db mice model. The C. kawachiensis administration inhibited microglial activation in the hippocampus in the streptozotocin-injected mice. Moreover, The C. kawachiensis treatment inhibited astroglial activation in the hippocampus and the hyperphosphorylation of tau at 231 of threonine and 396 of serine in hippocampal neurons, and also relieved the suppression of neurogenesis in the dentate gyrus of the hippocampus in the db/db mice. It was suggested that the dried peel powder of C. kawachiensis exerts anti-inflammatory and neuroprotective effects in the brain.  相似文献

Cerebral ischemia/reperfusion is known to induce the generation of reactive oxygen species and inflammatory responses. Numerous studies have demonstrated that naringin (NGIN) has anti-oxidant and anti-inflammatory properties. We previously reported that Citrus kawachiensis contains a large quantity of NGIN in its peel. In the present study, we orally (p.o.) administered dried peel powder of C. kawachiensis to mice of a transient global ischemia model and found in the hippocampus region that it 1) suppressed neuronal cell death, 2) reversed the reduction in the level of phosphorylated calcium-calmodulin-dependent protein kinase II, 3) had the tendency to reverse the reduction in the level of glutathione, and 4) blocked excessive activation of microglia and astrocytes. These results suggested that the dried peel powder of C. kawachiensis had a neuroprotective effect against ischemic brain via anti-oxidative and anti-inflammatory effects. We also showed that these effects of the dried peel powder were more powerful than those obtained with a comparable amount of NGIN alone.  相似文献

Summary As the hydrolysis of mandarin orange peel with macerating enzyme (40°C, 24 h) produced 0.59 g g–1 reducing sugar per dry peel compared to 0.36 by acid-hydrolysis (15 min at 120°C with 0.8 N H2SO4), the production of single cell protein (SCP) from orange peel was studied mostly using enzymatically hydrolyzed orange peel. When the enzymatically hydrolyzed peel media were used, the utilization efficiency of reducing sugars (%) and the growth yield from reducing sugars (g g–1) were: 63 and 0.51 for Saccharomyces cerevisiae; 56 and 0.48 for Candida utilis; 74 and 0.69 for Debaryomyces hansenii and 64 and 0.70 for Rhodotorula glutinis. SCP production from orange peel by D. hansenii and R. glutinis were further studied. Batch cultures for 24 h at 30°C using 100 g dried orange peel produced 45 g of dried cultivated peel (protein content, 33%) with D. hansenii and 34 g (protein content, 50%) with R. glutinis, and 38 g (protein content, 44%) with a mixture of both yeasts.  相似文献

Mango peel is one of the major wastes from fruit processing industries, which poses considerable disposal problems and ultimately leads to environmental pollution. The objective of the current research was to determine the significant parameters on the production of polygalacturonase from mango peel which is a major industrial waste. Solid state culture conditions for polygalacturonase production by Fusarium moniliforme from dried mango peel powder were optimized by Taguchi’s L-18 orthogonal array experimental design methodology. Eight fungal metabolic influencing variables, viz. temperature, mango peel, inoculum, peptone, ammonium nitrate (NH4NO3), magnesium sulphate (MgSO4), zinc sulphate (ZnSO4) and potassium dihydrogen phosphate (KH2PO4) were selected to optimize polygalacturonase production. The optimized parameters composed of temperature (30°C), mango peel (6.5%, g, w/v), inoculum (8%, ml, v/v), peptone (1%, g, w/v), NH4NO3 (0.60%, g, w/v), MgSO4 (0.05%, g, w/v), ZnSO4 (0.06%, g, w/v) and KH2PO4 (0.4%, g, w/v). Based on the influence of interaction of fermentation components of fermentation, these could be classified as the least significant and the most significant at individual and interaction levels. The temperature, inoculum level, mango peel substrate and KH2PO4 showed maximum production impact at optimized conditions. From the optimized conditions the polygalacturonase activity was maximized to 43.2 U g−1.  相似文献

Extract from yuzu fruit peel (Citrus junos Sieb. ex Tanaka) strongly suppressed the germination of lettuce seeds while that from the peel of other citrus fruits such as navel orange (C. sinensis) and lemon (C. limon Burm. f.) had very little or no effect. The highest inhibitory activity was located in the peel followed by the segment but no significant activity was found in the seed extract of yuzu fruit. The effect of yuzu peel extract was tested on a variety of major crops including 38 crop species. Germination of seeds of tomato (Lycopersicon esculentum Mill.), celery (Apium graveolens L. var. dulce) and watercress (Nasturtium officinale R. Br.) was completely blocked by 27.8 mg dry peel equivalent ml–1 of yuzu peel extract, while that of cucumber (Cucumis sativus L.) and pumpkin (Cucurbita moschata [Duch. ex Lam.] Duch. ex Poir.) seeds was not affected at this concentration, showing a broad variation in the sensitivity of crop seeds to the extract. The effectiveness of yuzu peel was also tested on some harmful weeds. The extract inhibited the elongation of both radicle and hypocotyl in summer weed quinoa (Chenopodium quinoa) grown on agar plates. Yuzu peel powder added to the soil was also effective in suppressing the growth of C. quinoa, Sonchus oleraceus L. and Digitaria ciliaris, while it had little effect on the spring weed Alopecurus aequalis Sobol. A strong growth inhibitory activity of yuzu peel was found in the aqueous phase after solvent extraction and most of the activity was recovered in the neutral fraction that passed through cation and anion exchange resins.  相似文献

The hippocampus has become one of the most extensively studied areas of the mammalian brain, and its proper function is of utmost importance, particularly for learning and memory. The hippocampus is the most susceptible brain region for damage, and its impaired function has been documented in many human brain diseases, e.g. hypoxia, ischemia, and epilepsy regardless of the age of the affected patients. In addition to experimental in vivo models of these disorders, the investigation of basic anatomical, physiological, and molecular aspects requires an adequate experimental in vitro model, which should meet the requirements for well-preserved representation of various cell types, and functional information processing properties in the hippocampus. In this review, the characteristics of organotypic hippocampal slice cultures (OHCs) together with the main differences between the in vivo and in vitro preparations are first briefly outlined. Thereafter, the use of OHCs in studies focusing on neuron cell death and synaptic plasticity is discussed. Special issue dedicated to Dr. Simo S. Oja  相似文献

Hemp (Cannabis sativa) produces phloem fibres, which are formed in bundles towards the periphery of the stem. This study investigated one aspect of the retting process, the dissociation of these fibres from the core of the stem, which is influenced by the action of micro-organisms. Peel tests were used to investigate the work required to remove a tissue peel, rich in fibres, from the core of the stem. Two separate experiments studied (1) the effects of tissue dehydration as stems were dried out in the oven; and (2) the progress of retting in stems that were dew-retted in the field. A comparison between two different types of peel tests, where the stems were clamped either vertically or horizontally in the jaws of a tensile testing machine and the load applied vertically, showed that the work required to peel stems in the vertical position was over 20% greater than that for the horizontal position. Freshly harvested stems were dehydrated in an oven at 40°C over a period of 42 h and as the moisture content decreased there was a significant increase in the work to peel. In contrast, stems that had been dew-retted in the field showed a significant decrease in work to peel. When the moisture content of stems dried in the oven decreased below 12%, the work to peel rapidly decreased. As the plant tissues dehydrated their fracture characteristics changed and this was reflected in the increase in the work to peel seen in the oven-dried samples. This, however, was not evident in the field-retted samples, in which there was a decrease in work to peel. The field-retted samples were exposed to the activity of micro-organisms for a prolonged period of time compared with the oven-dried samples; this increased the potential degradation caused by microbial enzymes. Thus, the reduced work to peel in dew-retted stems is thought to be due to the colonisation of the stem by micro-organisms and the subsequent retting process. The dramatic decrease in work to peel at low moisture content in the oven-dried stems is thought to result from mechanical changes leading to brittle fracture characteristics. This study indicates that peel tests can be used to objectively monitor the reduction in work to peel for dew-retted stems and relate this reduction to the progress of retting. Thus, peel tests may be used to monitor the effects of different factors on the process of retting in hemp.  相似文献

ABSTRACT

Depressive disorders are partly caused by chronic inflammation through the kynurenine (KYN) pathway. Preventive intervention using anti-inflammatory reagents may be beneficial for alleviating the risk of depression. In this study, we focused on the Japanese local citrus plant, Citrus tumida hort. ex Tanaka (C. tumida; CT), which contains flavonoids such as hesperidin that have anti-inflammatory actions. The dietary intake of 5% immature peels of CT fruits slightly increased stress resilience in a subchronic and mild social defeat (sCSDS) model in mice. Moreover, the dietary intake of 0.1% hesperidin significantly increased stress resilience and suppressed KYN levels in the hippocampus and prefrontal cortex in these mice. In addition, KYN levels in the hippocampus and prefrontal cortex were significantly correlated with the susceptibility to stress. In conclusion, these results suggest that dietary hesperidin increases stress resilience by suppressing the augmentation of KYN signaling under sCSDS.  相似文献

The influence of dried cell powder and culture filtrates of endophytic fungi on production of inophyllum in cell suspension cultures of leaf- and stem-derived callus of Calophyllum inophyllum was investigated. Two fungi, Nigrospora sphaerica and Phoma spp., endophytic to C. inophyllum, were isolated from leaf tissues, and were identified by both 18S rRNA gene amplification and sequencing. Elicitation of suspension cultures of both callus types of C. inophyllum with dried cell powder and culture filtrates of both fungi consistently elicited production of inophyllum A, B, C, and P. In comparison to stem-derived callus, suspension cultures of leaf-derived callus enhanced production of most inophyllum. Of the four inophyllum studied, the highest production of inophyllum A, C, and P was achieved in elicited suspension cultures of leaf-derived callus. Suspension cultures of stem-derived callus enhanced production only of inophyllum B. When suspension cultures of leaf-derived callus were elicited with 40 mg dried cell powder of Phoma spp., a level of 751-fold (6.84 mg/100 g elicited biomass) of inophyllum A was produced, compared to control. Whereas, a level of 414-fold (6.22 mg/100 g elicited biomass) of inophyllum B was produced when suspension cultures of stem-derived callus were elicited with 20 mg dried cell powder of N. sphaerica. When compared to control, a 10% culture filtrate of N. sphaerica in suspension cultures of leaf-derived callus elicited inophyllum C and P production by 928-fold (7.43 mg/100 g elicited biomass) and 750-fold (1.5 mg/100 g elicited biomass), respectively.  相似文献

Ginger (the rhizome of Zingiber officinale Roscoe) has been used worldwide for many centuries in cooking and for treatment of several diseases. The main pharmacological properties of ginger include anti-inflammatory, antihyperglycemic, antiarthritic, antiemetic and neuroprotective actions. Recent studies demonstrated that ginger significantly enhances cognitive function in various cognitive disorders as well as in healthy brain. However, the biochemical mechanisms underlying the ginger-mediated enhancement of cognition have not yet been studied in normal or diseased brain. In the present study, we assessed the memory-enhancing effects of dried ginger extract (GE) in a model of scopolamine-induced memory deficits and in normal animals by performing a novel object recognition test. We found that GE administration significantly improved the ability of mice to recognize novel objects, indicating improvements in learning and memory. Furthermore, to elucidate the mechanisms of GE-mediated cognitive enhancement, we focused on nerve growth factor (NGF)-induced signaling pathways. NGF enzyme-linked immunosorbent assay analysis revealed that GE administration led to elevated NGF levels in both the mouse hippocampus and rat glioma C6 cells. GE administration also resulted in phosphorylation of extracellular-signal-regulated kinase (ERK) and cyclic AMP response element-binding protein (CREB), as revealed by Western blotting analysis. Neutralization of NGF with a specific NGF antibody inhibited GE-triggered activation of ERK and CREB in the hippocampus. Also, GE treatment significantly increased pre- and postsynaptic markers, synaptophysin and PSD-95, which are related to synapse formation in the brain. These data suggest that GE has a synaptogenic effect via NGF-induced ERK/CREB activation, resulting in memory enhancement.  相似文献

Casearia sylvestris is an endemic tree of the Latin America that the essential oil (EO) has anti-inflammatory and gastroprotective actions. This study evaluates the chemical composition of the EO from the volatile fractions of in natura, fresh, and dried C. sylvestris var. sylvestris and var. lingua leaves. For both varieties, the dried leaves presented higher EO yield as compared to fresh leaves. The major EO chemical components were (E)-caryophyllene, α-humulene, germacrene D, bicyclogermacrene, spathulenol, caryophyllene oxide, and humulene epoxide II. In both varieties, the content of sesquiterpene hydrocarbons decreased and oxygenated sesquiterpenes increased on going from in natura to fresh and dried leaves, which indicated that leaf drying and hydrodistillation modified the volatile composition. The results also suggested that bicyclogermacrene and (E)-caryophyllene were oxidized during processing, to generate spathulenol and caryophyllene oxide, respectively. C. sylvestris varieties and in natura, fresh, and dried leaves differed in terms of the chemical composition of volatiles, which could affect the EO biological activities.  相似文献

The binding of tritiated 8-hydroxy-2-(di-n-propyl-amino)tetralin, or [3H]8-OH-DPAT, to membranes from rat cerebral cortex and hippocampus could be inhibited by serotonin (5-HT) and buspirone, and by the 5-HT antagonists propranolol, NAN-190, pindolol, pindobind-5-HT1A, WAY100135, spiperone and ritanserin. All competition curves, except for ritanserin, best fitted a two-site model. In vitro treatment of the membranes withN-ethylmaleimide (NEM), to alkylate sulfhydryl groups, caused dose-dependent decreases of binding; the inhibition curves were biphasic, and the effects irreversible. Reduction of disulfide bonds withl-dithiothreitol (L-DTT) also decreased binding, but in a monophasic way; these effects were fully reversible in cortex, but only partially reversible in hippocampus. In the latter region, but not in cerebral cortex, previous occupancy by [3H]8-OH-DPAT partially protected binding from the effects of bothL-DTT and NEM, suggesting that the thiol groups in the receptor recognition site(s) of this brain region are readily accessible. The binding characteristics were examined with the aid of saturation curves, carried out with increasing concentrations, up to 140 nM, of [3H]8-OH-DPAT. The saturation data were suggestive of a two-site receptor model incorporating a high-affinity site (Kh of 0.3–0.5 nM) corresponding to the 5-HT1A receptor, and a low-affinity site (Kl ofca 25 nM). After in vivo alkylations, carried out by treating rats withN-ethoxycarbonyl-2-ethoxy-1,2-dihydro-quinoline (EEDQ), the saturation curves from both control and EEDQ-treated rats were again best fitted to a two-site model. For EEDQ-treated animals, a drastic decrease of 5-HT1A receptor activity was noted; this loss was greater in hippocampus than in cerebral cortex. Since the decrease in 5-HT1A receptors was not associated with changes in low-affinity binding, the results suggest independent regulations of the two [3H]8-OH-DPAT binding proteins. Altogether, the present data further supports the notion that [3H]8-OH-DPAT, besides labelling 5-HT1A receptors, also binds to other structures in rat cerebral cortex and hippocampus. Plasmalogens are ether-linked phospholipids highly abundant in nervous tissue. Previously we demonstrated that acute administration of myo-inositol (myo-Ins) + [2-13C] ethanolamine ([2-13C]Etn) significantly elevated phosphatidylethanolamine plasmalogen (PlsEtn) in rat whole brain. Current experiments investigated the effects of acute myo-Ins+[2-13C]Etn administration on [PlsEtn] and the biosynthesis of new Etn lipids using NMR spectroscopy in rat cerebral cortex, hippocampus, brainstem, midbrain and cerebellum. Treated rats received a single dose of myo-Ins+[2-13C]Etn and controls received saline rather than myo-Ins. Data reveal that the cerebellum is the brain region most affected by treatment, which resulted in a 22% increase in [PlsEtn] and 89% increase in newly synthesized Etn lipids relative to controls (P 0.05). Furthermore, the cerebellar PlsEtn/phosphatidylethanolamine ratio and molar percentage of PlsEtn were significantly elevated by 12% and 8%, respectively (P 0.05). These data suggest that myo-Ins influences Etn lipid metabolism in brain, particularly in the cerebellum where there is a stimulation in the biosynthesis of new Etn lipids with a preference towards PlsEtn.  相似文献

Aqueousmethanol extracts of Cit rus junos, C. unshiu and C. sudachi fruit peel in hib ited the growth of the roots and hypocotyls of alfalfa (Medicago sativa L.), cress (Lepidiumsativum L.) and lettuce (Lactuca sativa L.) seedlings. Signifi cantreductions in the root and hypocotyl growth were observed as the extract concentration in creased in all bioassays. The in hibitory activity of C. junos ex tract on the growth of test plants was 3.3-to 17.9-fold and 3.6- to 20.6-fold greater than that of C. unshiu and C. sudachi extracts, respectively. Theconcent rationin C. junos was 3.5- and 4.9-fold greater than that in C. unshiu and C. sudachi, re spectively. Thus, there was a good correlation-between abscisic acid-b-D-glucopyranosylester (ABA-GE) concentrationsin C. junos, C. unshiu and C. sudachi fruit peel and theinhibito ryactivities of the irextracts. The concentration of ABA-GE in C. junos fruit peel was in creased with fruit maturityasgrowthinhibit oryactivity of C. junos fruit peel was reported to be in creased with fruit maturity, in dicating that the concentrations of ABA-GEin C. junos fruit peel was correlated withgrowthinhibit oryactivity of C. junos fruit peel in time course of fruit maturation. These findings suggest that ABA-GE may be involved in the growth inhibitory effect of C. junos, C. unshiu and C. sudachi fruit peel.  相似文献

An undescribed xanthone dimer, 1,3,5,8-tetrahydroxy-7-(1′,5′,8′-trihydroxy-3′-methoxy-2′-xanthonyl)xanthone ( 1 ) was separated together with eleven known compounds ( 2 – 12 ) from the dried whole herb of Swertia pseudochinensis. It was the first time that the compounds 8 – 12 were isolated from the Swertia genus. The structure of compound 1 was illuminated based on chemical evidence and spectral data analysis (UV, 1D and 2D-NMR, HR-ESI-MS). Moreover, the inhibitory effects of all compounds on NO production in LPS-induced RAW 264.7 cells were tested, compounds 8 , 9 , 10 , 11 and 12 showing significant inhibition. The IC50 value of compound 12 was 3.05±1.10 μM. Using target screening and molecular docking, we hypothesized that compound 12 may bind neutrophil elastase to exert its anti-inflammatory effects.  相似文献

A distributional survey of the large branchiopod crustaceans of the rice paddies of Shiga Prefecture, Japan, was conducted by the Lake Biwa Museum in May and June of 1999 and 2000. Collecting kits were issued to over 100 lay Field Reporters, who returned all samples, usually dried, to the museum for processing. Their 242 species-lots were augmented by 146 from Grygier, 46 from Ida, and 41 from other museum staff. Apart from this survey, 50 additional records in 1997–2000 were also taken into account. Seven species were recorded in the prefecture's southern half: Branchinella kugenumaensis (Ishikawa), Caenestheriella gifuensis (Ishikawa), and Lynceus biformis (Ishikawa) with wide distributions; Triops longicaudatus (LeConte) and Leptestheria kawachiensis Uéno limited to paddy areas around the south end of Lake Biwa; and Eulimnadia sp. mostly in remnant paddy areas threatened with urban development in Otsu city, with Triops granarius (Lucas) at a single such site in Otsu. In the prefecture's northern half, only B. kugenumaensis and C. gifuensis occurred to a limited degree. Possible reasons for these differences are discussed. Data pertaining to species co-occurrence and occurrence relative to the ongoing program of paddy field consolidation in the prefecture are presented and discussed. Success of the survey was due to the use of Field Reporters, who were able to provide a high degree of coverage of the study area, as well as the clearly defined habitat, the low number and easy recognizability of species, and the collection of voucher material from all sites. Conservation-related results included the listing of Eulimnadia sp. as a `rare' species by Shiga Prefecture.  相似文献

Summary Thirty-nine fungal cultures belonging to the genera of Aspergillus, Podospora, Sordaria, Cbaetomium, Iodophanus, Scleotinia, Coniella, Pellicularia and others, were examined for the production of enzymes which macerate the mandarin orange peel using a wheat bran as substrate. An isolated strain of Aspergillus niger was an excellent producer of macerating enzymes compared to other organisms tested. The peel of the mandarin orange could be macerated by the crude enzymes produced by isolated A. niger. The maceration of 1 g of peel/24 h yielded 0.57 g of reducing sugars. Expressed differently, 83% of solid peel materials were released from the peel into the water/24 h under the following conditions: a peel concentration of 8 g peel/l, a crude enzyme concentration of 1 g protein/l, a temperature of 40°C, a pH of 5, a 24 h incubation time and 120 rpm reciprocal shaking. The test of the macerating activity of commercially available hydrolases on the orange peel showed that the two samples of pectinase originating from A. niger had about the same activity as isolated A. niger whereas the two samples of cellulase originating from Trichoderma viride had remarkably lower activities than A. niger.  相似文献

Aims: Resuscitation of dried cultures represents a critical control point in obtaining active and effective probiotic strains. This study examined the effects of various rehydration conditions on the viability of Bifidobacterium longum NCC3001 and Lactobacillus johnsonii La1. Methods and Results: Reconstitution conditions for these strains were optimized using a multivariate experimental design approach. Furthermore, using flow cytometry, the cell integrity was followed during reconstitution. By adjusting the pH, availability of a metabolizable sugar, reconstitution duration, powder matrix and ratio of powder to reconstitution solution, the recovery of Bif. longum NCC3001 and Lact. johnsonii La1 following reconstitution was increased eight‐ and two‐fold, respectively, over standard reconstitution in maximum recovery diluent. It was shown that pH had a significant effect on the recovery of Bif. longum NCC3001 and Lact. johnsonii La1. Conclusions: The recovery of dried probiotic cultures is greatly dependent on the reconstitution conditions. The maximum recovery of 11·7 10log CFU g?1Bif. longum NCC3001 was achieved at 30‐min reconstitution at pH 8, in the presence of 2%l ‐arabinose and a ratio of 1 : 100 of powder to diluent. Lact. johnsonii La1 showed highest recovery (9·3 10log CFU g?1) after reconstitution, when mixed with maltodextrin at pH 4. Significance and Impact of the Study: To achieve accurate viable probiotic numbers from dried probiotic cultures, the reconstitution conditions should be optimized for the strain used.  相似文献

Controlling the snail intermediate host of schistosomiasis is one of the strategies used to reduce disease incidence. In this study screening tests were carried out on the action of dried powder of eight algal species against Biomphalaria alexandrina snails. The most effective powders were from Dictyota dichotoma and Cystoseira barbata with LC50 values of 560 and 2200?ppm, respectively. Prolonged exposure to sublethal concentrations of these two powders caused adverse effects on snails' reproduction especially at LC25. Hatchability of exposed eggs was lowest at LC25 of C. barbata, and LC15 of D. dichotoma. The most pronounced effects on the hermaphrodite gland of snails were caused by high concentrations of both algal suspensions, where ova were either degenerated or deformed and there were detrimental effects on all spermatogenesis stages. The electrophoretic separation of haemolymph proteins showed that the concentration with most effect was the LC15 for both tested algae as they resulted in more bands than the control and also an elevation in total protein content.  相似文献
