Abstract  An overview is presented on recent (1989–19911 investigations on insecticidal plants in China, specially in South China. The main emphasis was laid on studies of Meliaceae including Azadirachta indica, Melia azedarach and M. toosendan , as well as on Rhododendron molle, Derris elliptica, Tephrosia voglii, Ajuga nipponensis, Plumbago idea and Celastrus anplatus . Research work on the bioactivities and application of essential oils from plants for the control of stored-grain insects is also discussed. The results of these investigations reveal a great potential of the active ingredients of a number of Chinese plants for integrated pest management, as well as for using these natural product models as guides for the synthesis of a novel class of insecticides.  相似文献

The need for peroxidase purification is stressed as a requirement for comparative studies on isoenzyme structure as well as for detailed investigations on biosynthesis. A single cationic protein possessing the major peroxidase activity was isolated from the medium in which peanut cells had grown. The antibodies raised against this pure protein were employed as a probe to study the site of synthesis of peroxidase in the cell as well as the proportion of total synthesized protein which was peroxidase. Structural studies on the purified isoenzymes suggest the presence of three gene loci for peroxidase in cultured peanut cells. The results are discussed together with potential assays for induction of this enzyme and the relationship to cell development.  相似文献

This paper defines the truncated normalized max product operation for the transformation of states of a network and provides a method for solving a set of equations based on this operation. The operation serves as the transformation for the set of fully connected units in a recurrent network that otherwise might consist of linear threshold units. Component values of the state vector and outputs of the units take on the values in the set [0, 0.1,..., 0.9, 1]. The result is a much larger state space given a particular number of units and size of connection matrix than for a network based on threshold units. Since the operation defined here can form the basis of transformations in a recurrent network with a finite number of states, fixed points or cycles are possible and the network based on this operation for transformations can be used as an associative memory or pattern classifier with fixed points taking on the role of prototypes. Discrete fully recurrent networks have proven themselves to be very useful as associative memories and as classifiers. However they are often based on units that have binary states. The effect of this is that the data to be processed consisting of vectors in R(n) have to be converted to vectors in [0, 1]m with m much larger than n since binary encoding based on positional notation is not feasible. This implies a large increase in the number of components. The effect can be lessened by allowing more states for each unit in our network. The network proposed demonstrates those properties that are desirable in an associative memory very well as the simulations show.  相似文献

Silicon-based microfabricated microbial fuel cell toxicity sensor   总被引:1,自引:0,他引:1  
Microbial fuel cells (MFCs) have been used for several years as biosensors for measuring environmental parameters such as biochemical oxygen demand and water toxicity. The present study is focused on the detection of toxic matter using a novel silicon-based MFC. Like other existing toxicity sensors based on MFCs, this device is capable of detecting the variation on the current produced by the cell when toxic compounds are present in the medium. The MFC approach presented in this work aims to obtain a simple, compact and planar device for its further application as a biosensor in the design and fabrication of equipment for toxicity monitoring. It consists on a proton exchange membrane placed between two microfabricated silicon plates that act as current collectors. An array of square 80 μm × 80 μm vertical channels, 300 μm deep, have been defined trough the plates over an area of 6 mm × 6 mm. The final testing assembly incorporates two perspex pieces positioned onto the plates as reservoirs with a working volume of 144 μL per compartment. The operation of the microdevice as a direct electron transfer MFC has been validated by comparing its performance against a larger scale MFC, run under the same conditions. The device has been tested as a toxicity sensor by setting it at a fixed current while monitoring changes in the output power. A drop in the power production is observed when a toxic compound is added to the anode compartment. The compact design of the device makes it suitable for its incorporation into measurement equipment either as an individual device or as an array of sensors for high throughput processing.  相似文献

Risk-based concentrations (RBCs) for lead (Pb) in soil were estimated using equations for development of Canadian soil quality guidelines. Based on the latest toxicological assessments by various health agencies, risk specific doses for Pb were defined for children (for impacts on intelligence quotient [IQ]) and adults (for impacts on systolic blood pressure [SBP] as well as protection of fetal effects in the case of pregnant women). The analysis suggests that a RBC in soil of 180 μg/g (dry weight) for residential and other areas where children routinely play is protective of a 1 IQ point decrement on a population basis and may actually be associated with decrements of less than 0.2 to 0.35 IQ points when the weight of evidence is considered. For soils that children do not contact on a frequent basis, RBCs as great as 8800 μg/g are considered to be protective of a 1 mmHg SBP increase in adults (as well as IQ effects to the fetus). It is stressed that non-soil sources of Pb may be even more important than soil. The approach may also be useful in jurisdictions outside of Canada as the importance of considering IQ decrements on a population basis rather than an individual basis and uncertainties in soil ingestion rates are considered.  相似文献

Generally accepted methods of single-cell microsurgery in terms of their compliance with the task of preserving cell integrity and minimizing possible damage in various microsurgery manipulations on embryos and somatic cells, namely, during microsurgical transfer of nuclei for cloning, production of monozygotic twins, and microinjections into the cell, are analyzed in detail. The problems regarding the cell contacts with microtools, perforation as a factor of cell damage (when a microtool penetrates a cell), the influence of light on the cell, and retention of the cell during microsurgery (and the resulting effect of negative pressure on the cell) are discussed. The question of how long a single cell can persist in a microchamber in vitro is considered, as well as the impact of mineral oil used as a barrier against solvent evaporation. The effect of the physicochemical composition of the glass that is used for microtools is addressed for the first time, as well as the impact of thermal shock and fresh culture medium on a living cell.  相似文献

The construction of a modified perfusion chamber is presented, which can be used for a prolonged cultivation of mammalian cells and tissues, for observation of the behavior of living cells as well as for the study of different effects on these cells. The chamber is made as non-demountable of optical glass, with a diffusive barrier separating the pericellular zone from that with a perfusion medium. The scheme of the equipment for cultivation of cells and tissues in this diffusion chamber on controlling the composition of nutrient medium and gas phase is given.  相似文献

Three-dimensional cellular structures formed by MCF-7 human mammary carcinoma cells within collagen gels were isolated with collagenase and cultivated on plastic substratum to examine whether the cytoskeleton specific for cells forming cellular structures (S-type) changes to that specific for cells grown as monolayers (M-type). The cytoskeleton isolated as 0.05% Triton-insoluble fraction from the cellular structures after culture for 1 day on plastic was exclusively S-type. However, both types of cytoskeletons were observed in the cellular structures cultivated for 7 days on plastic as well as in the cells grown as monolayers for 2 days after dissociation of the cellular structures with trypsin. By use of an antibody raised against a 65-kD polypeptide that was specific for the M-type cytoskeleton, the presence of the polypeptide was found to be restricted to the cells grown out as monolayers from the edge of the cellular structures. In the cells grown for 2 days as monolayers, a mixture of cells both having and lacking the polypeptide was observed. After a 7-day culture of the dissociated cells as monolayers on plastic, however, most of the cells had M-type cytoskeletons. The present results show that the apparent change in the cytoskeleton of MCF-7 cells from S-type to M-type does not occur in cells involved in the three-dimensional cellular structures even in the absence of collagen gels, but that it occurs in cells which are grown as monolayers for at least 7 days on plastic substratum.  相似文献

The influence of biomembrane lipids on the catalytic activity of a peripheral membrane enzyme, acid phosphatase (AP), was studied in a reverse micellar system. It was found that the interaction of AP with lipids led to a number of kinetic effects depending on lipid nature on enzyme function. The observed effects might be caused by the formation of lipoprotein complexes as well as by the influence of lipids on structure and properties of the micellar matrix. The results are important for clear understanding of molecular mechanisms of regulation of the catalytic activity of the membrane-associated enzyme in vivo. These data can also be used as a physicochemical basis for application of AP in medical fields as a diagnostic tool for diseases caused by changes in lipid metabolism, e.g. urinary, orthopedic, and allergic diseases.  相似文献

The Corynebacterium glutamicum ATCC 13032 lysC(fbr) strain was engineered to grow fast on racemic mixtures of lactate and to secrete lysine during growth on lactate as well as on mixtures of lactate and glucose. The wild-type C. glutamicum only grows well on L-lactate. Overexpression of D-lactate dehydrogenase (dld) achieved by exchanging the native promoter of the dld gene for the stronger promoter of the sod gene encoding superoxide dismutase in C. glutamicum resulted in a duplication of biomass yield and faster growth without any secretion of lysine. Elementary mode analysis was applied to identify potential targets for lysine production from lactate as well as from mixtures of lactate and glucose. Two targets for overexpression were pyruvate carboxylase and malic enzyme. The overexpression of these genes using again the sod promoter resulted in growth-associated production of lysine with lactate as sole carbon source with a carbon yield of 9% and a yield of 15% during growth on a lactate-glucose mixture. Both substrates were taken up simultaneously with a slight preference for lactate. As surmised from the elementary mode analysis, deletion of glucose-6-phosphate isomerase resulted in a decreased production of lysine on the mixed substrate. Elementary mode analysis together with suitable objective functions has been found a very useful tool guiding the design of strains producing lysine on mixed substrates.  相似文献

Abstract— The activity profiles of the solubilized protein kinases from the microsomal and myelin fractions of bovine brain were examined by column chromatography and sucrose density gradient centrifugation. The main peak of adenosine 3',5'-monophosphate (cyclic AMP)-dependent activity with histone as substrate for each membrane enzyme was eluted with about 0.2 m -NaCl on a DEAE-cellulose column. A peak of activity stimulated with cyclic AMP was also eluted with about 0.1 m -NaCl for the microsomal enzyme. A peak with protamine and casein as substrate for the microsomal or myelin enzyme, respectively, was larger than that with histone as substrate for each enzyme. The first peak with histone as substrate on a DEAE–cellulose column appeared as two peaks on the Sepharose 6B column. The second peak with histone as substrate on DEAE–cellulose column was shown to be a holoenzyme consisting of regulatory and catalytic subunits. The holoenzyme and subunits were eluted at similar positions to each other between both membrane enzymes on Sepharose 6B column. The holoenzyme sedimented as two peaks of activity on sucrose density gradient centrifugation, both of which were stimulated with cyclic AMP. The preincubation of the holoenzyme with cyclic AMP resulted in shifting to a position of a smaller molecular size.
The results indicate the occurrence of multiple forms of protein kinases in membrane fractions of brain with respect to substrate specificity and physical property.  相似文献

The RecA protein of E coli promotes a strand exchange reaction in vitro which appears to be similar to homologous genetic recombination in vivo. A model for the mechanism of strand transfer reaction by RecA protein has been proposed by Howard-Flanders et al based on the assumption that the RecA monomer has two distinctive DNA binding sites both of which can bind to ssDNA as well as dsDNA. Here, I propose an alternative model based on the assumption that RecA monomer has a single domain for binding to a polynucleotide chain with a unique polarity. In addition, the model is based on a few mechanical assumptions that, in the presence of ATP, two RecA molecules form a head to head dimer as the basic binding unit to DNA, and that the binding of RecA protein to a polynucleotide chain induces a structural change of RecA protein that causes a higher state of affinity for another RecA molecule that is expressed as cooperativy. The model explains many of the biochemical capabilities of RecA protein including the polar polymerization of RecA protein on single stranded DNA and polar strand transfer of DNA by the protein as well as the formation of a joint DNA molecule in a paranemic configuration. The model also presents the energetics in the strand transfer reaction.  相似文献

There is a strong need for methods within life cycle assessment (LCA) that enable the inclusion of all complex aspects related to land use and land use change (LULUC). This article presents a case study of the use of one hectare (ha) of forest managed for the production of wood for bioenergy production. Both permanent and temporary changes in above‐ground biomass are assessed together with the impact on biodiversity caused by LULUC as a result of forestry activities. The impact is measured as a product of time and area requirements, as well as by changes in carbon pools and impacts on biodiversity as a consequence of different management options. To elaborate the usefulness of the method as well as its dependency on assumptions, a range of scenarios are introduced in the study. The results show that the impact on climate change from LULUC dominates the results, compared to the impact from forestry operations. This clearly demonstrates the need to include LULUC in an LCA of forestry products. For impacts both on climate change and biodiversity, the results show large variability based on what assumptions are made; and impacts can be either positive or negative. Consequently, a mere measure of land used does not provide any meaning in LCA, as it is not possible to know whether this contributes a positive or negative impact.  相似文献

Little is known about techniques for applying untreated microbial cells containing enzymes directly to industrial processes as a biocatalyst. The kinetic behavior of alpha-galactosidase-containing spherical pellets which are formed naturally under given conditions in a submerged culture of Mortierella vinacea was studied on the hydrolysis of PNPG (p-nitrophenyl-alpha-D-galactopyranoside). The effect on intraparticle diffusion on the overall reaction rate was assessed by the use of an effectiveness factor, which was calculated by the approximate solution to the equation derived from the mass balance within a pellet. The experimental effectiveness factors were found to be represented as a single function of the modified Thiele modulus, including such parameters as pellet size, enzyme concentration in the pellet, and substrate concentration. As the diffusional effect became more significant, the marked substrate inhibition as seen for a free enzyme disappeared gradually. The effect of product inhibition on the pellets was much weaker than that for a free enzyme at a given substrate concentration. In the region of diffusion controlled reaction, it was found that the rate is proportional to the square root of the enzyme concentration in the pellet. In addition, similarly to what was reported previously for a free enzyme,the reaction in a batch system was found to be approximately representable as simple first-order kinetics in which the rate constant was dependent on the initial substrate concentration.  相似文献

This review presents the current knowledge on the interaction of lipophilic, poorly water soluble drugs with liposomal and biological membranes. The center of attention will be on drugs having the potential to dissolve in a lipid membrane without perturbing them too much. The degree of interaction is described as solubility of a drug in phospholipid membranes and the kinetics of transfer of a lipophilic drug between membranes. Finally, the consequences of these two factors on the design of lipid-based carriers for oral, as well as parenteral use, for lipophilic drugs and lead selection of oral lipophilic drugs is described. Since liposomes serve as model-membranes for natural membranes, the assessment of lipid solubility and transfer kinetics of lipophilic drug using liposome formulations may additionally have predictive value for bioavailability and biodistribution and the pharmacokinetics of lipophilic drugs after parenteral as well as oral administration.  相似文献

Further to the discovery of bicyclic furanopyrimidine nucleoside analogues (BCNAs) as potent anti-VZV agents, a branched series of this family of compounds was synthesised. The aim was to study the impact of the geometry and steric hindrance in the side chain as well as lipophilic role of this moiety on biological activity. The results showed a detrimental effect of branching on antiviral activity, with a different magnitude depending on the position of branching in the side chain. This study again showed the importance of this moiety for biological activity, as well as the limited efficacy of the ClogP value as a tool for predicting the potency of BCNAs, while suggesting an alternative rationale behind the design of future series.  相似文献

The desire to reduce dependence on the ever diminishing fossil fuel reserves coupled with the impetus towards green energy has seen increased research in biofuels as alternative sources of energy. Lignocellulose materials are one of the most promising feedstocks for advanced biofuels production. However, their utilisation is dependent on the efficient hydrolysis of polysaccharides, which in part is dependent on cost-effective and benign pretreatment of biomass to remove or modify lignin and release or expose sugars to hydrolytic enzymes. Laccase is one of the enzymes that are being investigated not only for potential use as pretreatment agents in biofuel production, mainly as a delignifying enzyme, but also as a biotechnological tool for removal of inhibitors (mainly phenolic) of subsequent enzymatic processes. The current review discusses the major advances in the application of laccase as a potential pretreatment strategy, the underlying principles as well as directions for future research in the search for better enzyme-based technologies for biofuel production. Future perspectives could include synergy between enzymes that may be required for optimal results and the adoption of the biorefinery concept in line with the move towards the global implementation of the bioeconomy strategy.  相似文献

In this study, we propose substrate-independent modification for creating a protein-repellent surface based on dopamine-melanin anchoring layer used for subsequent binding of poly(ethylene oxide) (PEO) from melt. We verified that the dopamine-melanin layer can be formed on literally any substrate and could serve as the anchoring layer for subsequent grafting of PEO chains. Grafting of PEO from melt in a temperature range 70-110 °C produces densely packed PEO layers showing exceptionally low protein adsorption when exposed to the whole blood serum or plasma. The PEO layers prepared from melt at 110 °C retained the protein repellent properties for as long as 10 days after their exposure to physiological-like conditions. The PEO-dopamine-melanin modification represents a simple and universal surface modification method for the preparation of protein repellent surfaces that could serve as a nonfouling background in various applications, such as optical biosensors and tissue engineering.  相似文献
