Highly basophilic plate-shaped regions from oocytes of the surf clam have been examined with the electron microscope. The regions are composed of flat, hollow vesicles perforated by pores arranged, in surface view, in a hexagonal pattern. Cross-sections of this structure show a periodicity consisting of loops (cross-sections of the continuous space within the vesicle) alternating with spaces partly filled with dense material (pores). These structures are shown to resemble closely, the nuclear envelope. Similarities to and differences from basophilic regions of other cells are discussed and it is suggested that the small granules of Palade (38) are represented by granules composing the walls of the annuli of the nuclear envelope and assumed to be present in the annuli of the vesicles. Because of differences in the structure of these regions from basophilic regions of other cells, the name periodic lamellae is suggested since the structures show periodically repeating substructures (annuli) in both cross-sections and surface views.  相似文献

Electron micrograph evidence is presented that the nuclear envelope of the mature ovum of Dendraster excentricus is implicated in a proliferation of what appear as nuclear envelope replicas in the cytoplasm. The proliferation is associated with intranuclear vesicles which apparently coalesce to form comparatively simple replicas of the nuclear envelope closely applied to the inside of the nuclear envelope. The envelope itself may become disorganized at the time when fully formed annulate lamellae appear on the cytoplasmic side and parallel with it. The concept of interconvertibility of general cytoplasmic vesicles with most of the membrane systems of the cytoplasm is presented. The structure of the annuli in the annulate lamellae is shown to include small spheres or vesicles of variable size embedded in a dense matrix. Dense particles which are about 150 A in diameter are often found closely associated with annulate lamellae in the cytoplasm. Similar structures in other echinoderm eggs are basophilic. In this species, unlike other published examples, the association apparently takes place in the cytoplasm only after the lamellae have separated from the nucleus. If 150 A particles are synthesized by annulate lamellae, as their close physical relationship suggests, then in this species at least the necessary synthetic mechanisms and specificity must reside in the structure of annulate lamellae.  相似文献

Semi-isolated annulate lamellae were prepared from single newt oocytes (Triturus alpestris) by a modified Callan-Tomlin technique. Such preparations were examined with the electron microscope, and the negative staining appearance of the annulate lamellae is described. The annulate lamellae can be detected either adhering to the nuclear envelope or being detached from it. Sometimes they are observed to be connected with slender tubular-like structures interpreted as parts of the endoplasmic reticulum. The results obtained from negative staining are combined with those from sections. Especially, the structural data on the annulate lamellae and the nuclear envelope of the very same cell were compared. Evidence is presented that in the oocytes studied the two kinds of porous cisternae, namely annulate lamellae and nuclear envelope, are markedly distinguished in that the annulate lamellae exhibit a much higher pore frequency (generally about twice that found for the corresponding nuclear envelope) and have also a relative pore area occupying as much as 32% to 55% of the cisternal surface (compared with 13% to 22% in the nuclear envelopes). The pore diameter and all other ultrastructural details of the pore complexes, however, are equivalent in both kinds of porous cisternae. Like the annuli of the nuclear pore complexes of various animal and plant cells, the annuli of the annulate lamellae pores reveal also an eightfold symmetry of their subunits in negatively stained as well as in sectioned material. Furthermore, the annulate lamellae are shown to be a site of activity of the Mg-Na-K-stimulated ATPase.  相似文献

The germ cells, and germ cell precursors, in the nematode Caenorhabditis elegans contain distinctive granules called P granules. During early embryogenesis, P granules are segregated asymmetrically into those blastomeres that eventually produce the germ line. Because of the correlation between P granule distribution and the development of the germ line, P granules are widely thought to function in some aspect of germ line specification or differentiation. Most of the analysis of P granule structure and localization has focused on the early embryo, when P granules are located in the cytoplasm. However, during most of development P granules are associated with germ cell nuclei. We report here an ultrastructural analysis of the nuclear-associated P granules in the germ cells of the adult hermaphrodite gonad. We show that P granules are tightly associated with nuclear pores and that the positions of certain structures within the P granules correspond to the positions of pores on the nuclear envelope. We present immunocytochemical and ultrastructural data suggesting that P granules can associate, or remain associated, with pore-like structures even after they detach from the nuclear envelope during oogenesis. Finally, we show that nuclear-associated P granules in the gonad contain RNA, complementing previous studies showing that cytoplasmic P granules in embryos contain RNA.  相似文献

Nuclei of frog oocytes were isolated, fixed in OsO4 or KMnO4, and washed. Nuclear envelopes were then dissected off, placed on grids, and air-dried for electron microscopy. Envelopes from immature oocytes at the stage of beginning yolk deposition were compared with those from mature oocytes. Envelopes from the immature stage had "pores" whose annuli contained more material and showed central globules in the center much more frequently than envelopes from mature eggs. Annuli and central globules had similar appearance and fixation properties, suggesting similar chemical composition. After fixation with KMnO4, residual densities suggested that "pore" diaphragms are much more variable in thickness or composition in the younger stages. Envelopes of the immature oocytes had about 40 per cent more "pores" per unit area than mature envelopes. In crowding together, the "pores" tended to assume geometrical packing arrays in the young envelope, showing minimum center-to-center spacings of about 1530 A. Since the actual discontinuities in the membranes of the envelope are only about 950 A in diameter, this minimum distance of approach suggests that adjacent formations of the nuclear surface are associated with "pore" structure and perhaps set their limiting spacing distances. If this is true, then it can be deduced that "pore"-associated structures of the nuclear surface are probably circular in outline and about 1500 A in diameter. Isotopically labeled lysine was administered to intact, growing oocytes for 1 to 4 hours and the envelopes were subsequently isolated and fixed. Autoradiography of entire envelopes showed little or no incorporation of lysine into proteins, as compared with small fragments from other parts of the cell of roughly comparable mass. It was concluded that the isolated envelope, as seen in the electron micrographs, does not synthesize or turn over lysine-containing protein at a high rate.  相似文献

The fine structure and cytochemistry of the extremely large RNA puffs, or Balbiani rings, in salivary gland nuclei of midge, Chironomus thummi, larvae have been investigated. The Balbiani rings are composed of a diffuse mass of electron-opaque 400 to 500 A granules, short threads about 180 to 220 A in diameter and associated fine chromatin fibrils. These components appear to be organized into brushlike elements which form the ring. Electron microscope cytochemistry has shown that the granules and short threads contain RNA. After ribonuclease digestion, only 50 to 100 A chromatin fibrils were apparent in the Balbiani ring, and the granules were no longer demonstrable. Deoxyribonuclease digestion had no apparent effect on these structures. Observations indicate that the granules are formed from the short threads and released into the nucleoplasm in which they are evenly distributed. At the nuclear envelope, many granules have been observed partially or completely within the nuclear pores. These granules become elongated and are shown to penetrate the center of the pore in a rodlike form, about 200 A in diameter. The Balbiani ring granules are not normally visible within the cytoplasm adjacent to the nuclear envelope, but have been rarely found in this region. It is suggested that the granules represent the product of the Balbiani ring, possibly a messenger RNA bound to protein, and that they regularly pass into the cytoplasm through a narrow central channel in the pores of the nuclear envelope.  相似文献

Nuclei from nearly ripe eggs of Rana pipiens were isolated and cleaned in 0.1 M KCl. The whole nucleus was then digested to various degrees with ribonuclease or trypsin, followed by washing and fixation in either osmium tetroxide or potassium permanganate. The nuclear envelope was dissected off, placed on a grid, air dried, and compared with undigested controls in the electron microscope. Some envelopes were dehydrated, embedded in methacrylate, and sectioned. Annuli around "pores" are composed of a substance or substances, at least partially fibrillar, which is preserved by osmium but lost during permanganate fixation. Material within the "pores" is also preserved by osmium but partially lost after permanganate. No evidence of granules or tubules in the annuli was found in air dried mounts although a granular appearance could be seen in tangentially oriented thin sections. Thin sections of isolated envelopes give evidence of diffuse material within the "pores" as well as a more condensed diaphragm across their waists. In whole mounts of the envelope the total density within "pores" is relatively constant from "pore" to "pore." All material within "pores," including the condensed diaphragm, is removable by trypsin digestion. Wispy material from the "pore" structure projects into the nucleus and annular material extends into the cytoplasm. Both annular and diaphragm materials remain with the envelope when it is isolated and are thus considered a part of its structure, not merely evidences of material passing through. There is no evidence of ribonuclease-removable material in any part of the "pore" complex.  相似文献

To investigate the extent to which the electron-opaque pore material can regulate nucleocytoplasmic exchanges which occur through the nuclear annuli, experiments were performed in which polyvinylpyrrolidone (PVP)-coated colloidal gold particles (25 to 170 A in diameter) were microinjected into the cytoplasm of amebas (Amoeba proteus). The cells were fixed at various times after injection and examined with the electron microscope in order to determine the location of the gold particles. High concentrations of gold were found associated with the pore material at specific points adjacent to and within the pores. It is tentatively suggested that such specific accumulations could be a means of selecting substances from the cytoplasm for transport through the pores. Particles were also scattered throughout the ground cytoplasm and nucleoplasm. A comparison of the diameters of particles located in these two regions showed that the ability of materials to penetrate the nuclear envelope is a function of their size. It was estimated that the maximum size of the particles able to enter the nucleus is approximately 125 to 145 A indiameter. The regulation of exchanges with regard to particle size is thought to be dependent on the specific organization of the electron-opaque pore material.  相似文献

The endomembranous system of Serrasalmus spilopleura oocyte secondary growth was analysed using structural and ultrastructural cytochemical techniques. In vitellogenic oocytes, the endoplasmic reticulum components, the nuclear envelope intermembranous space, some Golgi dictiossomes, lysosomes, yolk granules, regions of the egg envelope and sites of the follicle cells react to acid phosphatase detection (AcPase). The cortical alveoli, some heterogeneous cytoplasmic structures, regions of the egg envelope, and sites of the follicle cells are strongly contrasted by osmium tetroxide and zinc iodide impregnation (ZIO). The endoplasmic reticulum components, some vesicles, and sites of the follicle cells also react to osmium tetroxide and potassium iodide impregnation (KI). The biosynthetic pathway of lysosomal proteins, such as acid phosphatase, required for vitellogenesis, involves the endoplasmic reticulum, Golgi complex, vesicles with inactive hydrolytic enzymes, and, finally, lysosomes. In S. spilopleura oocytes at secondary growth, the endomembranous system takes part in the production of the enzymes needed for vitellogenesis, and in the metabolism of yolk exogenous components (AcPase detection). The endomembranous system compartments also show reduction capacity (KI reaction) and are involved in the metabolism of proteins rich in SH‐groups (ZIO reaction).  相似文献

In ultrathin sections and freeze-fractures in hexagonal nuclear pore arrangement is described in midgut epithelial cells of Dytiscus marginalis. The majority of the pores is concentrated in several cup-like indentations of the nuclear envelope and the pore center-to-center distance is about 130 to 140 nm. In addition, in the regions of the nuclear pore clusters, patches of cytoplasm containing paracrystalline structures of closely packed hexagons of 18 to 20 nm width are found sandwiched between the nuclear envelope and a "secondary" envelope. In such areas, structures are commonly observed that suggest transfer of material from the nucleus to the cytoplasm, sometimes in a dumb-bell-shaped state and more often as slender filaments which migrate across the nuclear pores.  相似文献

Electron microscopical observations on the relationship of the Golgi region to other intracellular organelles in certain protein-secreting cells have substantiated and extended existing hypotheses. In micrographs of several cell types, the juxtanuclear Golgi regions were observed to be closely associated with nuclear "pores." The "transition elements" of the ergastoplasmic membranes possess "blebs" which may represent a transport process facilitating the movement of intracisternal contents into the Golgi zone. A "blebbing" process of this nature may be one source of the small variety of Golgi vesicles. Zymogen granules of different densities were observed and their significance was postulated. Light Golgi vacuoles were observed. It is suggested that these vacuoles represent accumulations of relatively fluid material segregated from the secretory product in these cell types. These hypotheses from inferential evidence are discussed and extended.  相似文献

Summary The ovary of the liver fluke has been studied with light and electron microscopy. The organ consisted of germ cells and a layer of peripheral cells suggested to be nurse cells, and was surrounded by a capsule containing muscular tissue. The peripheral cells rested on a thick basement membrane and were irregular in outline. Their nuclei were of irregular shape, the mitochondria were dark with few cristae and the endoplasmic reticulum was tubular or vesicular and partly studded with ribosomes. The germ cells were rounded or polyhedral except in the outer part of the ovary where some of them showed irregular processes. The germ cells of the outer region (oogonia) were relatively small and in close contact with the cells suggested to be nurse cells. The inner germ cells (oocytes) were large and loosely packed. Their nuclei were irregular and contained round distinct nucleoli. The nuclear envelopes showed numerous pores. The endoplasmic reticulum was very sparse, but free ribosomes were abundant in the cytoplasm. This corresponded with a strong basophilia removable with RNase. In addition round basophilic bodies formed by densely packed ribosomes and membraneous material occurred in close spatial relation to mitochondria. The latter contained dense granules and few cristae. Groups of vesicles and membraneous lamellae were found in the cytoplasm, but they were considerably smaller than vertebrate Golgi complexes. Numerous dense spherical granules were found mainly in the periphery of the large germ cells. The granules were strongly osmiophilic except in the terminal part of the ovary. Oogenesis in fetal pig ovaries comprises the successive changes from the primordial germ cells to the dictyotene oocytes in primordial ovarian follicles. In this study the observations were carried out with an electron microscope and stereological analysis was performed. At the ultrastructural level there are no differences between the primordial germ cells and oogonia, but oogonia are connected with the intercellular bridges. The onset of the dictyotene phase was accompanied by the changes in the cytoplasm of oocytes. Near the nucleus, the yolk nucleus is formed containing numerous Golgi bodies, endoplasmic reticulum (ER), mitochondria and granules. ER proliferates in contact with the external leaflet of the nuclear envelope forming the narrow ER cisterns. Between the nuclear envelope and ER cisterns, the vesicles with grey content are visible. The proliferating ER forms numerous concentric cisterns around the nucleus. Next, the most external cisterns fragment, detach, and then form the cup-like structures. These structures separate the distinct areas of cytoplasm-compartments, which contain mitochondria, ribosomes and lipid droplets. The cells of cortical sex cords of the ovary, which encloses the oocyte, form the follicles. The volume of oocytes in forming follicle increases due to the increase in the number of the cell inclusions: lipid droplets, vacuoles and yolk globules. In the oocytes of primordial ovarian follicles, the compartments are transformed into the yolk globules, which are encountered by a sheath of ER cisterns and the grey vesicles; they contain the mitochondria, lipid droplets and light vacuoles. The role of the compartments and yolk globules as metabolic units is discussed in comparison with similar structures of the mature eggs of pigs and other mammal species.  相似文献

The nuclear envelope consists of two parallel membranes enclosing an aqueous lumen. In places there are pores in both membranes at which the two membranes are joined. Within these pores reside the nuclear pore complexes. The current structural models of the nuclear pore complex have been derived from a number of studies using different electron microscopical techniques. Recently, using surface imaging techniques such as field emission in-lens scanning electron microscopy, novel structures have been identified, particularly at the periphery of the structure, most notably the nucleoplasmic basket. One limitation of the current models is that they are based almost entirely on nuclear envelopes isolated from amphibian oocytes and a pressing question is whether this structure is the same in other organisms and tissues. Here we have studied the structure of nuclear envelopes isolated from bird oocytes. We show that the overall structure is remarkably conserved. In particular, recently discovered peripheral structures appear very similar. We see variations in basket conformation but believe that this is related to the functional states of individual pore complexes.  相似文献

Elaborate surface ornamentation of the secondary egg envelope of the annual fishes, Cynolebias melanotaenia and C. ladigesi, is comprised of 250-Å components which are synthesized in the follicle cells during Stage 5 and are secreted during Stage 6. Tubules, due to their structure and electron density, act as naturally occurring tracers which can be used to localize their site of synthesis and path of intracellular transport. At Stage 5, follicle cells acquire the morphological features of a protein synthesizing-secreting cell. Appearance of a well-developed granular endoplasmic reticulum coincides with the initial appearance of tubular material. Tubular precursors first appear in the cisternae of the RER. These regions become dilated and seem to bud off to form vesicles. Vesicle enlargement occurs through continued synthesis or by fusion with other vesicles. Vesicles contain partially assembled tubules in a loose aggregation of disks and short rods. The limiting membrane of vesicles, even up to half the size of the nucleus, is uniformly studded with ribosomes. Late in Stage 5, the contents of the vesicles coalesce into compact granules in which 250-Å tubules have assembled. During Stage 6, shortly before fusing with the plasma membrane, the RER-derived vesicles appear to shed their ribosomes. Upon fusion, the vesicle discharges a discrete granule. The RER-derived vesicles transport tubular components directly from the site of synthesis to the exterior of the cell. Morphological evidence indicates that the pathway of intracellular transport and secretion bypasses the Golgi complex. Hexagonal close packing of the follicle cells seems to determine the spatial distribution of pattern in the secondary envelope. Generation of elaborate structures within the overall pattern results from the morphogenetic activity of individual cells. On the basis of structure, the secondary envelope constitutes a chorionic respiratory system similar to insect eggs. It probably functions as a “plastron” or physical gill.  相似文献

The nuclear envelope is the hallmark of all eukaryotic cells, separating the nucleoplasm from the cytoplasm. At the same time, the nuclear envelope allows for the controlled exchange of macromolecules between the two compartments through nuclear pores and presents a surface for anchoring and organizing cytoskeletal components and chromatin. Although our molecular understanding of the nuclear envelope in higher plants is only just beginning, fundamental differences from the animal nuclear envelope have already been found. This review provides an updated investigation of these differences with respect to nuclear pore complexes, targeting of Ran signalling to the nuclear envelope, inner nuclear envelope proteins, and the role and fate of the nuclear envelope during mitosis.  相似文献
