Modulations in the lipid metabolism in the gill, liver, muscle and brain of freshwater teleost Labeo rohita exposed to 1, 2 and 3 days to lethal (1.2 mg/l) and 1, 15 and 30 days to sublethal (0.24 mg/l) concentrations of copper were studied. The total lipids decreased and there was an increase in the free fatty acids, glycerol and lipase activity in the organs studied at lethal concentration of copper. The degree in these shifts increased over time of exposure (1 less than 2 less than 3 days). In sublethal concentration, the levels of total lipids, free fatty acids, glycerol and lipase activity increased in all the four organs and the shifts followed two different trends during the exposure periods, 1 less than 15 less than 30 days in total lipids and 1 greater than 15 greater than 30 days in the other parameters. Among the organs, in both concentration media the changes in the lipid metabolism were in the order liver greater than gill greater than muscle greater than brain.  相似文献

Pathological alterations in various organs of rohu (L. rohita) fingerlings following acute (0, 7.50, 11.25 and 13.75 mg/kg body weight) and subchronic (0, 1.25 and 2.50 mg/kg body weight) single i.p. aflatoxin B1 exposure for 10 and 90 days, respectively, were investigated. Mortality (dose-dependent) was marked only during acute toxicosis. The changes observed in various organs were dose and time dependent. The acute dose groups revealed toxic changes viz., necrotic and vascular changes in liver and gill lamellae; meningitis, congestion in brain, degeneration and inflammatory reaction in heart along with degenerative to necrotic changes in kidney tubules and sloughing of the intestinal mucosa. During subchronic exposure to this toxin, preneoplastic lesions in liver along with changes in spleen, intestine, gill and pancreas were recorded. With low doses of aflatoxin, the fish did not reveal any mortality or external signs other than catchexia and increased pigmentation on scales. In composite culture practice of Indian major carps, this could be of economic significance.  相似文献

Different male germ cells identified on the basis of histological and cytological characteristics in the testicular lobules of Labeo rohita have been grouped into spermatogonia, primary spermatocytes, secondary spermatocytes, spermatids and spermatozoa. The seasonal changes of the testis in L. rohita have been described according to its morphological peculiarities as well as to its variations in gonadal volumes, GSI values and frequency percentages of the different male germ cells occurring in the testicular lobules. Consequently, the entire testicular cycle in L. rohita may be categorised into 4 distinct phases viz., growth, maturation of pre-spawning, spawning, and resting or post-spawning phases.  相似文献

Chromatophores show significant changes during healing of skin wounds in Labeo rohita (Common Name - Rohu). Wound area can be divided into regions I, II and III. After infliction of wound, skin colour becomes significantly dark by 2 h that is gradually restored by 2 d. In regions II and III at 5 min, epidermal melanophores appear with beaded dendrites. In these regions at 2 h and in region I at 6 h, epidermal melanophores appear small, rounded or irregular shaped having dendritic processes with aggregated melanosomes. Subsequently, melanophores appear having elongated dendrites with dispersed or aggregated melanosomes. At 24 h, clusters of pigmented bodies appear in regions I and II. These bodies increase up to 2 d, and then diminish gradually and disappear by 8 d. Changes in dermal melanophores in region II at 5 min indicate the onset of degeneration. Degenerating melanophores increase up to 12 h, then gradually decline, and disappear by 4 d. Simultaneously, stellate melanophore reappear, gradually increase and appear like control by 8 d. Dermal melanophores in region III at different intervals appear stellate. In region I stellate dermal melanophores appear at 4 d. Stellate melanophores in all regions show different distribution of dispersed or aggregated melanosomes. With the appearance of dermal melanophores, highly refractive, crystalline structures, possibly the refractive platelets of the iridophores, are visualized around them. At subsequent intervals, these are frequently observed. This study provides interesting insights in injury induced changes in chromatophores in fish. The findings could be considered useful in perception of intriguing features in the development of pigment research in future.  相似文献

Dietary valine requirement of Indian major carp, Labeo rohita Hamilton, fry (3.0 ± 0.02 cm, 0.16 ± 0.03 g) was determined using dose‐response method. Fishes were fed six isonitrogenous [40% crude protein (CP)] and isocaloric (4.28 kcal g?1, Gross Energy (GE)) amino acid test diets containing casein, gelatin, and l ‐crystalline amino acids with graded levels of valine (0.75, 1.00, 1.25, 1.50, 1.75, and 2.00% dry diet) at 5% body weight for 6 weeks in triplicate groups twice a day at 07.00 and 17.30 hours. Live weight gain (158.52%), feed conversion ratio (FCR, 1.70), specific growth rate (SGR, 2.25), and protein efficiency ratio (PER, 1.46) were significantly (P < 0.05) higher in fish fed a diet containing 1.5% of the dietary valine (diet IV). Second‐degree polynomial regression analysis of the live weight gain and FCR data indicated the dietary valine requirement at 1.63 and 1.5% of the dry diet, corresponding to 4.0 and 3.75% of dietary protein. Maximum carcass protein, minimum moisture, and fat were recorded at 1.5% of the dietary valine level, except carcass ash, which remained constant throughout the treatments. No mortality was observed during the entire length of the feeding trial. On the basis of FCR and protein deposition data, it is recommended that dietary valine inclusion at 1.5% of dry diet, corresponding to 3.75% of dietary protein, is optimal for the growth of L. rohita fry.  相似文献

Alterations in the liver histology of Labeo rohita were examined after exposure to different concentrations of carbofuran (0.06 and 0.15 mg L?1) and cypermethrin (0.16 and 0.40 μl L?1) for 28 days. Histological recovery was also studied by maintaining the intoxicated fish in a freshwater system for an additional 28 days. Major damages caused by carbofuran toxicity were diffuse necrosis, cordal disarrangement, individualization of hepatocytes, etc.; significant changes induced by cypermethrin were hyperplasia, disintegration of hepatic mass, focal coagulative necrosis, etc. In both cases, damages were dose‐dependent, with cypermethrin exhibiting more sensitivity than carbofuran. In all cases, recovery was prominent and rate of recovery was faster with carbofuran than when using cypermethrin.  相似文献

Hematological and plasma chemistry indices are simple and essential diagnostic tools for monitoring the physiological and health status of fish. Aim of the present study was to obtain reference values for the hematological and plasma chemistry of wild populations of Labeo rohita captured in a freshwater pond between July 2008 and June 2010. These reference values and the mean were evaluated according to sex and season. In summer, the red blood cells (1.84 × 106 38 per cubic mm), haemoglobin (8.52 gm dl?1) and haematocrit (31.49%) were highest in males, whereas the maximum values for white blood cells (5.635 × 103 40 per cubic mm) were found in females, however, no significant variation of mean corpuscular volume (MCV) or mean corpuscular haemoglobin (MCH) was observed between sexes. Various blood parameter levels between the sexes in summer were notably different from those measured in other seasons except for MCH and MCHC values (p < 0.05). Compared to most teleosts, the L. rohita has similar mean values for PCV and Hb. Throughout summer the glucose (76.0 mg dl?1), lipid (3.41 gm dl?1) and cholesterol (145.0 mg dl?1) levels were highest. In spring the plasma protein levels were higher in males, but higher in winter for females. Consequently, the seasons are key factors when using blood parameters as biomarkers for environmental alterations.  相似文献

Labeo rohita, popularly known as rohu is a widely cultured species in the whole Indian subcontinent. Knowledge of the genetic diversity of this species is important to support management and conservation programs which will subsequently help in sustainable production of this species. DNA markers, mostly microsatellite markers are excellent tool to evaluate genetic variation of populations. Genetic variation of three wild and one farm population was assessed using eleven microsatellite loci. In analyzing 192 samples, the number of alleles ranged from 4 to 23; observed heterozygosity 0.500 to 0.870 and expected heterozygosity from 0.389 to 0.878. Exact test for Hardy Weinberg disequilibrium revealed that each riverine sample had at least one locus not in equilibrium except one river. Negative inbreeding coefficients (FIS) were observed across populations indicating very high level of genetic diversity but little genetic differentiation among populations.  相似文献

The biochemical and stress responses of two Indian major carps, rohu Labeo rohita and mrigal Cirrhinus mrigala were studied after acclimating them to four preset temperatures (26, 31, 33 and 36° C) for 30 days. The blood glucose and liver glycogen levels showed an inverse trend in both the species and were significantly different in L. rohita at higher temperatures. The decrease in the liver glycogen level of C. mrigala , however, was not significant. Plasma cortisol levels increased significantly whereas the ascorbic acid content in the brain and kidney of both the species decreased significantly with increasing temperatures. Total lipid content in the liver of both the species decreased significantly with increasing acclimation temperatures. The phospholipid concentration decreased in L. rohita with increasing acclimation temperatures, and in C. mrigala the values decreased up to 33° C and increased at 36° C. In C. mrigala , the cholesterol level decreased up to 33° C and then increased at 36° C, but the absolute value was lower in comparison to L. rohita . The cholesterol levels, however, were not significantly different in L. rohita . Triglycerides and free fatty acids concentrations decreased significantly with increasing acclimation temperatures in both the species. The present study indicates species-specific metabolic responses of L. rohita and C. mrigala to thermal acclimation.  相似文献

The present study was carried out to investigate the carcass traits of farmed freshwater Labeo rohita in relation to body size as important information to calculate yield and/or support for machine/manual processing. For this purpose ninety specimens were collected for nine weight groups from 600 to 1100 g, each group with a 100 g increment. Specimens were collected from grow‐out culture ponds of the Central Institute of Freshwater Aquaculture (CIFA), Bhubaneswar. The carcass evaluation technique was followed to evaluate carcass yield, offal yield and carcass cutability. Head size percentage of rohu increased as the body mass increased, which in turn increased the processing yield. Dry matter and ether extract percentage also increased with the body mass; however, moisture percentage showed a decreasing trend. The fore cut showed a distinctly higher ether extract percentage followed by the middle and posterior cuts. There was a good correlation development to describe the relationship between yield traits and body weight.  相似文献

Impact of three different types of seaweed diets on growth, feed utilization and nutrient digestibility of L. rohita was studied for 120 days. The seaweed diet fed fishes, especially Ulva based diet showed comparatively higher growth and weight increment. Good food conversion ratio, food assimilation efficiency, protein efficiency ratio and better nutrient digestibility were recorded for seaweed diet fed fishes. The results suggests the suitability of utilizing seaweeds, Ulva fasciata, Spyridia insignis and Sargassum wightii as partial substitute for fishmeal in formulated diets of L. rohita.  相似文献

A 30 day feeding trial was conducted using a freshwater fish, Labeo rohita (rohu), to determine their thermal tolerance, oxygen consumption and optimum temperature for growth. Four hundred and sixteen L. rohita fry (10 days old, 0.385±0.003 g) were equally distributed between four treatments (26, 31, 33 and 36 °C) each with four replicates for 30 days. Highest body weight gain and lowest feed conversion ratio (FCR) was recorded between 31 and 33 °C. The highest specific growth rate was recorded at 31 °C followed by 33 and 26 °C and the lowest was at 36 °C. Thermal tolerance and oxygen consumption studies were carried out after completion of growth study to determine tolerance level and metabolic activity at four different acclimation temperatures. Oxygen consumption rate increased significantly with increasing acclimation temperature. Preferred temperature decided from relationship between acclimation temperature and Q10 values were between 33 and 36 °C, which gives a better understanding of optimum temperature for growth of L. rohita. Critical thermal maxima (CTMax) and critical thermal minima (CTMin) were 42.33±0.07, 44.81±0.07, 45.35±0.06, 45.60±0.03 and 12.00±0.08, 12.46±0.04, 13.80±0.10, 14.43±0.06, respectively, and increased significantly with increasing acclimation temperatures (26, 31, 33 and 36 °C). Survival (%) was similar in all groups indicating that temperature range of 26–36 °C is not fatal to L. rohita fry. The optimum temperature range for growth was 31–33 °C and for Q10 values was 33–36 °C.  相似文献

A 122‐day growth trial was conducted to observe the impact of polyhouse in winter on growth, conversion efficiencies and body composition of fingerling Labeo rohita (7.9 ± 1.1 cm; 4.60 ± 0.18 g). Fish were stocked (1.5 m?2) in polyhouse and outdoor concrete tanks (250 m2) in duplicate and fed a compound diet [35% crude protein (CP)] to apparent satiation twice daily, at 09.00 and 17.00 hours. A natural photoperiod was maintained during the trials. At the end of the trials, polyhouse‐reared fish produced significantly (P < 0.01) higher values for weight increment, specific growth rate (SGR %), protein efficiency rate (PER), protein productive value (PPV) and better feed conversion ratio (FCR). Polyhouse‐reared fish showed higher (P < 0.01) CP and fat, and lower (P < 0.01) moisture and ash contents in the muscle. In the second part of the study, which also lasted 122 days, polyhouse‐reared young L. rohita (21.7 ± 1.3 cm; 127.8 ± 0.69 g) were raised to marketable size in outdoor earthen ponds (500 m2) at a stocking density of 0.37 m?2 to examine the effects of feeding traditional feed mixture and compound diet (30% CP) on growth and body composition of fish. Labeo rohita fed the compound diet showed higher (P < 0.01) values for weight increment, SGR (%), PER and PPV, and better (P < 0.01) FCR than those fed traditional feed. Proximate composition of fish muscle showed higher (P < 0.05) CP and fat, and lower (P < 0.05) moisture and ash contents in fish fed the compound diet. The results suggest that polyhouse may be used successfully to culture L. rohita during winter, and that thereafter the fish may be reared in earthen ponds using the compound diet to obtain better production.  相似文献

A 60 days experiment was conducted to study the effect of dietary gelatinized (G) and non-gelatinized (NG) starch on immunomodulation of Labeo rohita juveniles. Two hundred and thirty four juveniles (av. wt. 2.53+/-0.04) were randomly distributed in six treatment groups with each of three replicates. Six semi-purified diets containing NG and G corn starch, each at six levels of inclusion (0, 20, 40, 60, 80, 100) were prepared viz., T(1) (100% NG, 0% G starch), T(2) (80% NG, 20% G starch), T(3) (60% NG, 40% G starch), T(4) (40% NG, 60% G starch), T(5) (20% NG, 80% G starch) and T(6) (0% NG, 100% G starch). After a feeding period of 60 days, the juveniles were challenged with Aeromonas hydrophila to study their immunomodulation due to feeding of G and NG starch. RBC and haemoglobin content were significantly (P<0.05) reduced due to bacterial challenge, but dietary starch (G/NG starch) had no effect on it. G:NG starch ratio in the feed had significant effect on total leukocyte count during pre- and post-challenge periods. The leukocyte count concomitantly increased with the increased level of G starch in the diet. Highest albumin/globulin (A/G) ratio was recorded in T6 group (100% G starch) and lowest in T1 group (100% NG starch) group followed by T2 group both in pre- and post-challenge periods. NBT, lysozyme activity, total protein and globulin content were highest in T2 group (80% NG, 20% G starch) both in pre- and post-challenge periods. After challenge with A. hydrophila, the highest survival was recorded in T2 group, whereas lowest survival was recorded in T6 group. Conclusively high level of G starch was found to be immunosuppressive in Labeo rohita juveniles and NG:G starch ratio of 80:20 seems to be optimum for promoting growth and protecting immunity in L. rohita juveniles.  相似文献

The role of microRNA in gene regulation during developmental biology has been well depicted in several organisms. The present study was performed to investigate miRNAs role in the liver tissues during carbohydrate metabolism and their targets in the farmed carp rohu, Labeo rohita, which is economically important species in aquaculture. Using Illumina-HiSeq technology, a total of 22,612,316; 44,316,046 and 13,338,434 clean reads were obtained from three small-RNA libraries. We have identified 138 conserved and 161 novel miRNAs and studies revealed that miR-22, miR-122, miR-365, miR-200, and miR-146 are involved in carbohydrate metabolism. Further analysis depicted mature miRNA and their predicted target sites in genes that were involved in developmental biology, cellular activities, transportation, etc. This is the first report of the presence of miRNAs in liver tissue of rohu and their comparative profile linked with metabolism serves as a vital resource as a biomarker.  相似文献

A continuous leukocyte cell line with phagocytic activity was established from peritoneal macrophages of rohu, Labeo rohita (LRPM). LRPM was initiated from adherent mononuclear leukocytes isolated from peritoneal cavity of rohu, without use of any growth factors or feeder cells. These cells exhibited maximum growth at 30 °C in L-15 medium containing 20 % foetal bovine serum, and has been subcultured for more than 60 passages till date. The cells showed 85 % viability after 6 months of storage in liquid nitrogen. The species of origin of the LRPM was confirmed by the amplification and sequencing of 655 bp fragment of cytochrome oxidase subunit I of mitochondrial DNA. Functionally, LRPM showed phagocytic activity of yeast cells and fluorescent latex beads as evaluated by phase contrast and scanning electron microscopy, respectively. Immuno-modulators such as bacterial lipopolysaccharide and phorbol myristate acetate resulted in functional activation of LRPM; and enhanced their microbicidal activity through release of reactive oxygen species and nitric oxide. Culture supernatant from activated cells also revealed lysozyme activity. Cells of LRPM were positive for alpha-naphthyl acetate esterase enzyme indicating macrophage lineage. Our results indicate that this cell line can be a useful in vitro tool to study the role of macrophages in teleost immune system and to evaluate the effects of new aquaculture drugs. The LRPM cell line represents the first reported leukocyte cell line of peritoneal origin from any freshwater species of fish.  相似文献

The level of genetic variation provides the raw material for selective improvement of a stock. Random amplified polymorphic DNA (RAPD) assay was used to assess the genetic variation in three rivers: the Halda, the Jamuna and the Padma as well as in one hatchery population of the commercially important Indian major carp, Labeo rohita. RAPD markers were amplified from DNA samples of 35 fish from each of the four populations using six decamer random primers. The polymorphic loci proportions were 0.33, 0.28, 0.28 and 0.26 and Nei's gene diversity values were 0.06, 0.07, 0.06 and 0.05 for the Halda, the Jamuna, the Padma and the hatchery populations, respectively. The pairwise population differentiation (FST) values indicated a low level of genetic differentiation between the population pairs. From the unweighted pair group method of arithmetic mean (UPGMA) dendrogram based on Nei's genetic distances a correlation between genetic affinities and geographical area was found. The populations were segregated into two groups: the Halda in one group and the Jamuna, the Padma and the hatchery in another group. Overall, the RAPD technique can be introduced as a tool in the population genetics of the rohu fish to provide information on their genetic stock structure.  相似文献
