Experimentally determined ranges of thermal tolerance and requirements for completion of the life history of some 60 seaweed species from the North Atlantic Ocean were compared with annual temperature regimes at their geographic boundaries. In all but a few species, thermal responses accounted for the location of boundaries. Distribution was restricted by: (a) lethal effects of high or low temperatures preventing survival of the hardiest life history stage (often microthalli), (b) temperature requirements for completion of the life history operating on any one process (i.e. [sexual] reproduction, formation of macrothalli or blades), (c) temperature requirements for the increase of population size (through growth or the formation of asexual propagules). Optimum growth/reproduction temperatures or lethal limits of the non-hardiest stage (often macrothalli) were irrelevant in explaining distribution. In some species, ecotypic differentiation in thermal responses over the distribution range influenced the location of geographic boundaries, but in many other species no such ecotypic differences were evident. Specific daylength requirements affected the location of boundaries only when interacting with temperature. The following types of thermal responses could be recognised, resulting in characteristic distribution patterns: (A) Species endemic to the (warm) temperate eastern Atlantic had narrow survival ranges (between ca 5 and ca 25°C) preventing occurrence in NE America. In species with isomorphic life histories without very specific temperature requirements for reproduction, northern and southern boundaries in Eur/Africa are set by lethal limits. Species with heteromorphic life histories often required high and/or low temperatures to induce reproduction in one or both life history phases which further restricted distribution. (B) Species endemic to the tropical western Atlantic also had narrow survival ranges (between ca 10 and ca 35°C). Northern boundaries are set by low, lethal winter temperatures. Thermal properties would potentially allow occurrence in the (sub) tropical eastern Atlantic, but the ocean must have formed a barrier to dispersal. No experimental evidence is so far available for tropical species with an amphi-Atlantic distribution. (C) Tropical to temperate species endemic to the western Atlantic had broad survival ranges (<0 to ca 35°C). Northern boundaries are set by low summer temperatures preventing (growth and) reproduction. Thermal properties would permit occurrence in the (sub)tropical eastern Atlantic, but along potential “stepping stones” for dispersal in the northern Atlantic (Greenland, Iceland, NW Europe) summer temperatures would be too low for growth. (D) In most amphi-Atlantic (tropical-) temperate species, northern boundaries are set by low summer temperatures preventing reproduction or the increase of population size. On European shores, species generally extended into regions with slightly lower summer temperatures than in America, probably because milder winters allow survival of a larger part of the population. (E) Amphi-Atlantic (Arctic-) temperate species survived at subzero temperatures. In species with isomorphic life histories not specifically requiring low temperatures for reproduction, southern boundaries are set by lethally high summer temperatures on both sides of the Atlantic. None of the species survived temperatures over 30°C which prevents tropical occurrence. Species with these thermal responses are characterized by distribution patterns in which southern boundaries in Eur/Africa lie further south than those in eastern N America because of cooler summers. In most species with heteromorphic life histories (or crustose and erect growth forms), low temperatures were required for formation of the macrothalli (either directly or through the induction of sexual reproduction). These species have composite southern boundaries in the north Atlantic Ocean. On American coasts, boundaries are set by lethally high summer temperatures, on European coasts by winter temperatures too high for the induction of macrothalli. Species with this type of thermal responses are characterized by distribution patterns in which the boundaries in Eur/Africa lie further north than those in eastern N America because of warmer winters. Three membrane thermotropic transitions at 8, 20, and 40 degrees C have been detected in human red blood cells (RBC) by using spin-labeled stearic acids. Red blood cells infected in vitro by Plasmodium falciparum showed the disappearance of the 8 degrees C transition and a lowering of the 40 degrees C transition to 32 degrees C. The disappearance of the 8 degrees C transition was observed in synchronized cultures of P. falciparum trophozoites as well as in mouse RBC infected in vivo by an asynchronous population of P. berghei. Furthermore, erythrocytes infected by P. falciparum showed an increase in the phosphorylation of protein 4.1. This protein was shown previously to be involved in the 8 degrees C transition, (T. Forte, T. L. Leto, M. Minetti, and V. T. Marchesi, Biochemistry 24, 7876-7880 (1985). Our results suggest that the malaria parasite invasion produces a disorganization of the protein 4.1-membrane interaction.  相似文献

Over 2,000 cases of imported malaria have been confirmed by blood examination. Ninety percent. of cases from tropical Africa were infected with P. falciparum. Most of the patients were Caucasians and had primary infections. All developed fever within a month after arrival and most of them within two weeks of arrival. In some patients malaria parasites were seen in routine blood films.Developing forms of P. falciparum were always present in the peripheral blood of patients suffering from a primary attack which was not diagnosed or treated until a week or more after the onset of fever.All deaths investigated were caused by P. falciparum and were primary infections.In not one of the P. falciparum infections did the victim continue taking prophylactic drugs for more than a few days after leaving the endemic area. Had drugs been continued for one month probably not a single overt case of P. falciparum would have occurred.A primary attack of P. falciparum malaria is seldom, if ever, classical in that the fever is never tertian and may resemble clinically many other diseases.Children in boarding-schools returning from the tropics should be supplied with prophylactic tablets and instructions to the matron. If there is an epidemic of a fever any students who have recently returned from the tropics should have a blood film examined for malaria.The risk of contracting malaria among drug addicts is considerable, especially with P. falciparum.  相似文献

Sera were obtained in 415 known cases of malaria (88 residents, 327 immigrants) at different times after diagnosis. Three antigens were used in the indirect fluorscence antibody test to detect antibodies to either Plasmodium falciparum or P vivax. Results in residents and immigrants were analysed separately. Most residents had detectable antibodies within one week after an attack, which began to wane after a month. The strongest reactions were obtained in cases of falciparum malaria with the homologous antigen and in cases of vivax malaria with P fieldi. The overall pattern of results was the same in the immigrants but the proportions positive for malaria antibodies, mean titres, persistence of antibodies, and the cross-reaction were usually greater. Testing for malaria antibodies is probably of value in the retrospective differential diagnosis of malaria in patients who have not been exposed to malaria before but must be interpreted with caution in others.  相似文献

ABSTRACT: BACKGROUND: Malaria and HIV co-infection adversely impact the outcome of both diseases and previous studies have mostly focused on falciparum malaria. Plasmodium vivax contributes to almost half of the malaria cases in India, but the disease burden of HIV and P. vivax co-infection is unclear. METHODS: HIV-infected subjects (n=460) were randomly selected from the 4,611 individuals seen at a Voluntary Counseling and Testing Center in Chennai, India between Jan 2 to Dec 31 2008. Malaria testing was performed on stored plasma samples by both nested PCR using both genus-specific and species-specific primers and immunochromatography-based rapid diagnostic test for detecting antibodies against both Plasmodium falciparum and P. vivax. RESULTS: Recent malaria co-infection, defined by the presence of antibodies, was detected in 9.8% (45/460) participants. Plasmodium vivax accounted for majority of the infections (60%) followed by P. falciparum (27%) and mixed infections (13%). Individuals with HIV and malaria co-infection were more likely to be men (p=0.01). Between those with and without malaria, there was no difference in age (p=0.14), CD4+ T-cell counts (p=0.19) or proportion CD4+ T-cell below 200/mL (p=0.51). CONCLUSIONS: Retrospective testing of stored plasma samples for malaria antibodies can facilitate identification of populations with high rates of co-infection, and in this southern India HIVinfected cohort there was a considerable burden of malaria co-infection, predominantly due to P. vivax. However, the rate of P. falciparum infection was more than 6-fold higher among HIV-infected individuals than what would be expected in the general population in the region. Interestingly, individuals co-infected with malaria and HIV were not more likely to be immunosuppressed than individuals with HIV infection alone.  相似文献

Genetic diversity provides Plasmodium falciparum with the potential capacity of avoiding the immune response, and possibly supporting the selection of drug or vaccine resistant parasites. These genetic characters play key roles in the selection of appropriate malaria control measures. Diverse clones of Plasmodium falciparum, often denoted as strains, has been documented, and the degree of genetic diversity supported by several kinds of PCR (polymerase chain reaction) assays. Many studies in different endemic regions with differences in their level of disease transmission have clarified the interactions between the parasite populations and malaria epidemiology. This paper describes recombination events of the malaria parasite life cycle that originate such genetic diversity in P. falciparum, reviewing different studies on this aspect and its implications in the immunity and development of control measures in regions with different degrees of endemicity.  相似文献

About 43Cladophora species inhabit the coasts of the northern Atlantic Ocean. These can be subdivided into seven distribution groups: (a) the tropical western Atlantic group (16 species); (b) the warm temperate Mediterranean-Atlantic group (9 species); (c) the warm temperate North American group (1 species); (d) the Arctic group (1 species); (e) the amphiatlantic tropical to warm temperature group (7 species); (f) the amphiatlantic tropical to temperate group (4 species), and (g) the amphiatlantic temperate group (5 species). These groups agree with general phytogeographic patterns. Thus, the high numbers of species restricted to the tropical western Atlantic region and the warm temperate Mediteranean-Atlantic region are in agreement with the richness and high degree of endemism of these regions. The fact that all species occurring in northeast America also occur in Europe agrees with the high floristic similarity of the boreal areas in America and Europe. The sediment coasts of the Carolinas are an efficient barrier to the south-north dispersal of benthic algae. The temperature bound phytogeographic limits are set in most cases by the species ability to survive adverse temperatures; for “northern” species to survive a high summer temperature in the south, and for “southern” species to survive a low winter temperature in the north. The limits in the Arctic region are all set by the species ability for sufficient growth and reproduction in summer. Conversely, only few northern species have a southern limit which is set by a winter temperature that is not too high for sufficient growth and reproduction. Most species of this group are winter-annuals at their southern limit, and summer-annuals at their northern limit. A comparatively small number of species with a tropical-to-warm temperate distribution have a northern limit at temperate latitudes which is set by a sufficiently high summer temperature for growth and reproduction. A high proportion of this group are lagoonal or quiet water species, which profit by higher summer temperatures in sheltered waters.C. vagabunda is an example.C. rupestris andC. sericea have an amphiboreal distribution and also occur in the southern temperate belt. They probably used a Pleistocene temperature drop to disperse, through the Atlantic along the African coast, from one hemisphere to the other. In the Pacific temperatures were not sufficiently low for this dispersal; and hence these two species reached the Pacific probably by way of the Bering Strait.  相似文献

The resurgence of malaria in Venezuela as an epidemic has caused an increase in the number of cases of this disease. 898 cases were registered in 1962 in contrast to 46,279 cases in 1988. This reality implies that all hospitals will have to face the increase of patients with this disease. Ten cases of malaria have been described and analyzed. These have been chosen for their medical and surgical complications, for which they were admitted to the "Hospital Universitario de Caracas" and "Hospital de Clínicas Caracas". Of these ten cases, eight were acquired in a natural way and two in an induced form. In six cases, the agent was P. falciparum; in three, P. vivax; in one, P. ovale and P.vivax combined. Five of the cases by P.falciparum were resistant to chloroquine, being necessary to treat them with a combination of quinine and other antimalaric drugs. Among the 10 described patients, there were: one case of splenic rupture in malaria by P. vivax, a severe case of anemia in a child with malaria by this same species and a case of relapse produced by P.vivax in another child with an insufficient radical treatment. The usual mistakes are observed in the management of malaria, such as a delay in the diagnosis and inappropriate radical treatment. As conclusion, publication of epidemiological, clinical and parasitological problems which are causing the actual epidemic, is recommended. This will be a guide in the fight against malaria, that must be considered again a national emergency.  相似文献

The influence of environmental factors on the timing of parturition has not been investigated in viviparous squamates. We investigated the interaction between temperature and parturition in two viviparous skink species, the southern snow skink (Niveoscincus microlepidotus) and the metallic skink (N. metallicus). In these species, the timing of parturition is separated from the completion of embryonic development; the delay is attributed to their cool and variable habitats. We examined whether the neurohypophyseal hormone arginine vasotocin (AVT) stimulated parturition in southern snow skinks with late stage embryos in autumn (approximately 6-7 months prior to parturition) and in metallic skinks with late stage embryos in summer (approximately 2-3 weeks prior to parturition). The experiments were conducted at a range of environmentally relevant temperatures (6 degrees C, 15 degrees C, 22 degrees C, and 28 degrees C). AVT induced parturition in both species at all temperatures; time until birth, however, occurred more quickly at warmer temperatures (22 degrees and 28 degrees C), whereas cooler temperatures delayed parturition. We hypothesize that if cool temperatures are preventing parturition, then temperature must act at some level within the brain to prevent or slow the secretion of AVT. Future experiments will need to determine how temperature influences AVT production. Further research is also required to determine how the timing of parturition is influenced by interactions between temperature, photoperiod, and seasonal hormone patterns.  相似文献

Plasmodium falciparum malaria is a major cause of morbidity and mortality in many developing countries especially in sub-Saharan Africa. A susceptibility locus for mild malaria has been mapped to the MHC region, and TNF polymorphisms have been associated with mild malaria. The Natural Cytotoxicity-triggering Receptor 3 (NCR3) gene is located in the peak region of linkage, and is 15kb distal to TNF. In this study, we considered NCR3 as a candidate gene, and we genotyped ten NCR3 single nucleotide polymorphisms (SNPs). Here, we report evidence of an association between mild malaria and NCR3 -412G>C polymorphism located within the promoter. Population-based association analysis showed that NCR3 -412C carriers had more frequent mild malaria attacks than NCR3 -412GG individuals (P=0.001). Using the family-based association test (FBAT) program and its phenotype (PBAT) option, we further found that NCR3 -412C (P=0.0009) and a haplotype containing NCR3 -412C (P=0.008) were significantly associated with increased risk of mild malaria, and that the association was not due to the association of TNF with mild malaria. These observations suggest that there are at least two genes located on the central region of MHC involved in genetic control of human malaria. The association of NCR3 with malaria should provide new insights into the role of Natural Killer cells in this common disease.  相似文献

There is evidence that pregnancy enhances the clinical severity of malaria, especially of P. falciparum infections. In pregnant women with little or no prior experience of the disease, P. falciparum causes severe clinical illness, substantial malaria mortality, increased rates of abortion and stillbirth and low birthweight of offspring; moreover, in such women, the clinical consequences seen unmodified by maternal parity. However, in pregnant women resident in highly endemic areas who have acquired considerable immunity through prolonged prior contact with malaria, parity appears to influence susceptibility to an important degree. Women who are pregnant for the first time are most affected, showing increased prevalence and density of parasitaemia, increased frequency of clinical illness (but not mortality) and significantly increased frequency of delivery of low birthweight children. In contrast, in multigravid women these clinical features are much less obvious and rarely attain statistical significance. The differences in susceptibility to malaria of pregnant women associated with parity and previous immunological experience require that protective strategies must be planned with full knowledge of the local epidemiology of malaria and be specifically targeted to the women who require them. Furthermore, the effectiveness of each strategy requires careful monitoring to permit such modifications as may be required by change in the immune status of the resident population.  相似文献

Plasmodium vivax is the most prevalent malaria parasite on the American continent. It generates a global burden of 80-100 million cases annually and represents a tremendous public health problem, particularly in the American and Asian continents. A malaria vaccine would be considered the most cost-effective measure against this vector-borne disease and it would contribute to a reduction in malaria cases and to eventual eradication. Although significant progress has been achieved in the search for Plasmodium falciparum antigens that could be used in a vaccine, limited progress has been made in the search for P. vivax components that might be eligible for vaccine development. This is primarily due to the lack of in vitro cultures to serve as an antigen source and to inadequate funding. While the most advanced P. falciparum vaccine candidate is currently being tested in Phase III trials in Africa, the most advanced P. vivax candidates have only advanced to Phase I trials. Herein, we describe the overall strategy and progress in P. vivax vaccine research, from antigen discovery to preclinical and clinical development and we discuss the regional potential of Latin America to develop a comprehensive platform for vaccine development.  相似文献

Geographic overlap between malaria and the occurrence of mutant hemoglobin and erythrocyte surface proteins has indicated that polymorphisms in human genes have been selected by severe malaria. Deletion of exon 3 in the glycophorin C gene (called GYPCDeltaex3 here) has been found in Melanesians; this alteration changes the serologic phenotype of the Gerbich (Ge) blood group system, resulting in Ge negativity. The GYPCDeltaex3 allele reaches a high frequency (46.5%) in coastal areas of Papua New Guinea where malaria is hyperendemic. The Plasmodium falciparum erythrocyte-binding antigen 140 (EBA140, also known as BAEBL) binds with high affinity to the surface of human erythrocytes. Here we show that the receptor for EBA140 is glycophorin C (GYPC) and that this interaction mediates a principal P. falciparum invasion pathway into human erythrocytes. EBA140 does not bind to GYPC in Ge-negative erythrocytes, nor can P. falciparum invade such cells using this invasion pathway. This provides compelling evidence that Ge negativity has arisen in Melanesian populations through natural selection by severe malaria.  相似文献

Apical membrane antigen-1 (AMA-1) is a target of antibodies that inhibit invasion of Plasmodium falciparum into human erythrocytes and is a candidate for inclusion in a malaria vaccine. We have identified a line of P. falciparum (W2mef) less susceptible to anti-AMA1 antibodies raised to the protein from a heterologous parasite line (3D7). We have constructed transgenic P. falciparum expressing heterologous AMA-1 alleles. In vitro invasion assays show that these transgenic parasites differ from parental lines in susceptibility to inhibitory antibodies, providing direct evidence that sequence polymorphisms within AMA-1 are responsible for evasion of immune responses that inhibit parasite invasion. We also generated a parasite line that would express a chimeric AMA-1 protein, in which highly polymorphic residues within domain 1 were exchanged. Inhibition assays suggest that these residues are not sufficient for inhibition by invasion-blocking antibodies. This study is the first to use P. falciparum allelic exchange to examine the relationship between genetic diversity and susceptibility to protective antibodies. The findings have important implications for the development of an AMA-1-based malaria vaccine.  相似文献

In recent years, associations of particular factors of the human leukocyte antigen (HLA) system with two major infectious diseases of tropical countries have been recognized: common West African HLA antigens are associated with protection from severe Plasmodium falciparum malaria, and HLA-D alleles are associated with generalized disease, localized disease and putative immunity in Onchocerca volvulus infection. Here, Christian Meyer and Peter Kremsner summarize current information on the involvement of HLA factors in P. falciparum malaria and O. volvulus infection, and briefly report on HLA-related immunological characteristics of various conditions in these infectious diseases.  相似文献

Summary Male and femalePsammodromus hispanicus from southern Europe were acclimated to four seasonal conditions of photoperiod and night time temperature. During the dark period, the lizards' body temperatures fell to ambient air temperature but during the light period the lizards were allowed to thermoregulate behaviourally and at such times the lizards' mean body temperature varied from 29.0°C to 32.6°C. The resting metabolic rate of these lizards was measured in 5°C steps from 5°C to 30°C or 35°C. Sexual condition had little effect on resting metabolic rate, but at low temperatures lizards acclimated to winter or spring seasonal conditions had lower resting metabolic rates than those acclimated to summer or autumn conditions. At temperatures above 20°C seasonal acclimation had no effect on resting metabolic rate. It is considered that the reduction in low temperature metabolic rate in spring and winter is induced by low night time temperatures and serves to conserve energy during those seasons when lizards must spend long periods at low temperature without being able to feed.  相似文献

ABSTRACT: BACKGROUND: The Lao Government changed the national policy for uncomplicated Plasmodium falciparum malaria from chloroquine to artemether-lumefantrine (AL) in 2005. Since then, no information on AL efficacy has been reported. With evidence of resistance to artemisinin derivatives in adjacent Cambodia, there has been a concern as to AL efficacy. Monitoring of AL efficacy would help the Lao Government to make decisions on appropriate malaria treatment. METHODS: The efficacy of a three-day, twice daily oral artemether-lumefantrine for the treatment of uncomplicated falciparum malaria in Xepon District, Savannakhet Province, southern Laos was studied over 42 days follow-up. This was part of a trial of thiamin supplementation in falciparum malaria. RESULTS: Of 630 patients with P. falciparum enrolled in the trial of thiamin treatment, 549 (87%, 357 children [less than or equal to]15 years and 192 adults) were included in this study. The per protocol 42-day cure rates were 97% (524/541) [96% (337/352) for children and 99% (187/189) for adults, p = 0.042]. By conventional intention-to-treat analysis, the 42-day cure rates adjusted for re-infection, were 97% (532/549) [96% (342/357) in children and 99% (190/192) in adults, p = 0.042]. The proportion of patients who remained parasitaemic at day 1 after treatment was significantly higher in children [33% (116/356)] compared to adults [15% (28/192)] (p < 0.001) and only one adult patient had detectable parasitaemia on day 2. There were no serious adverse events. Potential side effects after treatment were reported more commonly in adults (32%) compared to children (15%) (p < 0.001). Patients with recrudescent infections were significantly younger, had longer mean time to fever clearance, and had longer median time to parasite clearance compared to those who were cured. CONCLUSIONS: The current nationally-recommended anti-malarial treatment (artemether-lumefantrine) remains highly efficacious for the treatment of uncomplicated falciparum malaria five years after introduction in Laos. Regular monitoring is required in case artemisinin-resistant P. falciparum parasites should appear. Trial registration ISRCTN85411059.  相似文献

Genes encoding proteins homologous to the catalytic subunits of DNA polymerase alpha and delta have been cloned from the human malaria parasite Plasmodium falciparum. These are among the first cellular replicative DNA polymerase genes to be cloned and their sequences allow us to make new statements about the relative degrees of conservation of these two enzymes. The most important finding was that P. falciparum Pol delta showed considerable homology to the only other Pol delta enzyme for which published sequence is available, that of S. cerevisiae, displaying an overall amino acid identity of 45% and identity over a highly conserved central region of 59%. In contrast, the level of identity shown over the equivalent central region of Pol alpha between the P. falciparum and S. cerevisiae sequences is only 32%. The sequence data also allowed us to examine the degree of conservation in putative exonuclease domains of Pol delta. The Pol delta gene of P. falciparum maps to chromosome 10 and evidence is presented for the presence of different sized Pol delta mRNA's in the asexual and sexual erythrocytic stages of parasite development.  相似文献

Pieris brassicae L. is adapted to a wide variety of local climates throughout the Palaeartic by means of geographic variation in daylengths and temperatures triggering winter diapause. We report for the first time a population from Sotogrande in southern Spain (strain S) that additionally exhibits a pronounced summer diapause in the pupal stage as a response to long daylengths. As well as having diapause triggered by long daylengths, this population develops directly at short, ecologically relevant daylengths. This unusual short-day type response is expressed at temperatures of 21 degrees C and above. The absence of cryptic summer diapause in a population from southern France indicates that this summer diapause evolved locally in Spain or Northern Africa in a population of P. brassicae which lacked this trait. The PPRs of strains S, H (Denmark) and B (France) and crosses between them suggest that summer diapause is inherited as a recessive allele which has a clinal decrease of frequency in populations northwards. The population from southern France shows that this simple pattern of inheritance is further modified by an unknown genetic mechanism controlling the expression of summer diapause near the border between aestivating and non-aestivating populations. On the basis of the photoperiodic response and climatic data we predict P. brassicae to have five or more generations each year in southern Spain.  相似文献
