This study examines the genetic variations and mechanisms involved in the development of permethrin resistance in individual mosquitoes from a field population of Culex quinquefasciatus, HAmCq^G0, and characterizes susceptible reference lines of mosquitoes with a similar genetic background to the field HAmCq^G0 strain. Six upregulated cytochrome P450 genes, CYP9M10, CYP9J34, CYP6P14, CYP9J40, CYP6AA7, and CYP4C52v1, previously identified as being upregulated in the larvae of resistant HAmCq68 mosquitoes were examined in the larvae of 3 strains (susceptible S-Lab, parental HAmCq^G0 and permethrin-selected highly resistant HAmCq68) and 8 HAmCq^G0 single- egg raft colonies, covering a range of levels of susceptibility/resistance to perrnethrin and exhibiting different variations in the expression of A and/or T alleles at the L-to-F kdr locus of the sodium channel. The 2 lines with the lowest tolerance to permethrin and bearing solely the susceptible A allele at the L-to-F kdr locus of the sodium channels, from colonies Cx_SERC5 and Cx_SERC8, showed lower or similar levels of all 6 of the P450 genes tested compared with the S-Lab strain, suggesting that these 2 lines could be used as the reference mosquitoes in future studies characterizing insecticide resistance in HAmCq mosquitoes. This study also provides a detailed investigation of the mechanisms involved in insecticide resistance in individuals within a population: individuals with elevated levels of resistance to permethrin all displayed one or more potential resistance mechanisms-either elevated levels of P450 gene expression, or L-to-F mutations in the sodium channel, or both.  相似文献

Nothofagus pumilio is the dominant and almost ubiquitous tree species in mountainous environments of temperate South America. We used two types of molecular markers (cpDNA and isozymes) to evaluate the effects of the Paleogene paleogeography of Patagonia and more recent climatic oscillations of the Neogene on such cold‐tolerant species’ genetic makeup. Phylogeographic analysis on sequences of three cpDNA non‐coding regions at 85 populations yielded two latitudinally disjunct monophyletic clades north and south of c. 42°S containing 11 and three haplotypes, respectively. This indicates a long‐lasting vicariant event due to the presence of an extended open paleobasin at mid latitudes of Patagonia. Also distribution patterns of cpDNA haplotypes suggest regional spread following stepping‐stone models using pre‐Cenozoic mountains as corridors. Comparable genetic diversity measured along 41 sampled populations using seven polymorphic isozyme loci provides evidence of local persistence and spread from multiple ice‐free locations. In addition, significantly higher heterozygosity and allelic richness at high latitudes, i.e. in areas of larger glacial extent, suggest survival in large and isolated refugia. While, higher cpDNA diversity in lower latitudes reflects the complex orogeny that historically isolated northern populations, lower isozyme diversity and reduced FST values provide evidence of local glacial survival in numerous small locales. Therefore, current genetic structure of N. pumilio is the result of regional processes which took place during the Tertiary that were enhanced by contemporary local effects of drift and isolation in response to Quaternary climatic cycles.  相似文献

Background and Aims

The Hawaiian silversword alliance (Asteraceae) is one the best examples of a plant adaptive radiation, exhibiting extensive morphological and ecological diversity. No research within this group has addressed the role of geographical isolation, independent of ecological adaptation, in contributing to taxonomic diversity. The aims of this study were to examine genetic differentiation among subspecies of Dubautia laxa (Asteraceae) to determine if allopatric or sympatric populations and subspecies form distinct genetic clusters to understand better the role of geography in diversification within the alliance.

Methods

Dubautia laxa is a widespread member of the Hawaiian silversword alliance, occurring on four of the five major islands of the Hawaiian archipelago, with four subspecies recognized on the basis of morphological, ecological and geographical variation. Nuclear microsatellites and plastid DNA sequence data were examined. Data were analysed using maximum-likelihood and Bayesian phylogenetic methodologies to identify unique evolutionary lineages.

Key Results

Plastid DNA sequence data resolved two highly divergent lineages, recognized as the Laxa and Hirsuta groups, that are more similar to other members of the Hawaiian silversword alliance than they are to each other. The Laxa group is basal to the young island species of Dubautia, whereas the Hirsuta group forms a clade with the old island lineages of Dubautia and with Argyroxiphium. The divergence between the plastid groups is supported by Bayesian microsatellite clustering analyses, but the degree of nuclear differentiation is not as great. Clear genetic differentiation is only observed between allopatric populations, both within and among islands.

Conclusions

These results indicate that geographical separation has aided diversification in D. laxa, whereas ecologically associated morphological differences are not associated with neutral genetic differentiation. This suggests that, despite the stunning ecological adaptation observed, geography has also played an important role in the Hawaiian silversword alliance plant adaptive radiation.  相似文献

Two Santacrucian borhyaenoids, Borhyaena tuberata and Prothylacinus patagonicus , are analyzed from a functional-adaptive perspective. Seven extant placental and marsupial models are examined in order to interpret the locomotor adaptations of the two fossils. These carnivorous models are characterized by various hunting types and locomotor habits, and the association of their skeletal adaptive features with diet, substrate preference, and locomotor performance permits a functional interpretation of the postcranium of Borhyaena and Prothylacinus . The analysis shows that the forelimb of Prothylacinus is modified to provide strength and flexibility for controlled climbing. This taxon exhibits semiplantigrade fore- and hind feet. Its vertebral column was flexible, and the hindlimb suggests an active predatory mode of hunting. The tail was muscular, heavy, and was probably used as a balancing organ. By comparison, the forelimb of Borhyaena indicates a more terrestrial mode of life, with a digitigrade forefoot, and more parasagittal movements. The tail was lighter and less muscular than in Prothylacinus . Both fossils are characterized by a powerful neck musculature related to predatory habits.  相似文献

The Uruguay River starts in Serra do Mar in Brazil runs through the Paranense forest and flows southward through grassland and savannas. It has a continuous gallery forest of 750 km from the southern border of the Paranense forest to the river mouth. The gallery forest extends for 100 km more along the Río de la Plata. 125 (68.7%) of the 182 species of forest birds recorded in the southern Paranense forest penetrate into the gallery forest of the Uruguay River and only 13 (7.1%) reach the end of the gallery forest (Punta Lara). The number of bird species is inversely correlated (r2 = 0.942) with distance and the slope of the regression is 58.10. This means a decline in diversity with 32% of species lost per unit distance. A hundred and eighty forest tree species were recorded in the southern Paranense forest, of which 113 (62.8%) penetrate into the gallery forest of the Uruguay River, and 28 (15.6%) reach Punta Lara. The number of tree species is inversely correlated (r2 = 0.976) with distance and the slope of the regression is ?45.62. This means a decline in diversity with 25% of species lost per unit distance. The Uruguay River enables the dispersal of many species of forest birds and trees from the rain forest, but species richness tends to decrease with increased distance from the source area. A clear association pattern was found for birds between size, diet, habitat use and distance reached into the gallery forest. Species of smaller body size, granivores, insectivores and those that use both the interior and exterior parts of the gallery forest advanced noticeably further along the river than larger species, carnivores, nectarivores or frugivores, and those that frequent only a part of the forest. Similarly, a clear association between dispersal mechanism, water dependence and distance reached into the gallery forest was found for trees. Species with vegetative reproduction, zoochorous species and riparian species advanced markedly longer distances along the river than, anemochorous species and non‐riparian species.  相似文献

The Atlantic coast of South America is characterized by significant variation in environmental attributes, such as the salinity and transparency of the water, substrate, temperatures, and oceanic currents, which contribute to the existence of heterogeneous environments with distinct biotic and abiotic conditions. The present study shows that the Aspistor quadriscutis from the equatorial Amazon–Orinoco plume and Aspistor luniscutis from the northeast to southeast coast of Brazil represent different morphological lineages, with distinct life habits and behavior, belonging to a single genetic lineage. This condition is related to the distinct selective pressures (ecological conditions) found in the geographic regions in which the two species occur. Intermediate morphological patterns correspond to the ancestral traits, present in fossil species, which, together with the absence of differentiation in mitochondrial markers, characterize a process of incipient speciation.  相似文献

Abstract

Actualistic studies on mammalian carnivore taphonomy in southern South America are reviewed here, including pumas, small cats, foxes, and other, smaller carnivores. Patterns for different carnivore taxa and their variation are elicited. Also temporal and spatial variability is analysed, and comparisons are made to other carnivores and regions. While generally these Neotropical carnivores produce low damage intensity, stronger modifications and their implications are described as well. These patterns and their variation are put in context by taking into account the particular physical and biotic conditions in the southern Neotropics.  相似文献

Contemporary systematic treatments of the Central and South American bracken ferns in the genus Pteridium Gled. ex Scop. recognize morphotype caudatum as either a full species or a variety of P. aquilinum (L.) Kuhn. Geographically representative sporophytes of morphotype caudatum , including the type in the Linnaean Herbarium, are shown using spore size, guard-cell length and morphology of the cells of the false indusium to be tetraploid (based on 4 n  = 208). DNA fingerprinting of field-collected Venezuelan samples supports the generalization that morphotype caudatum is a fertile allotetraploid containing genomic elements otherwise distinctive of the southern hemisphere diploid P. arachnoideum (Kaulf.) Maxon, together with elements characteristic of northern hemisphere diploids including the North American P. aquilinum var. pubescens Underw. and P. aquilinum var. pseudocaudatum (Clute) A. Heller. Evidence of genetic isolation from taxa with overlapping distributions, as well as morphological, biochemical and ecological data, validate recognition of P. caudatum (L.) Maxon at species level. Heterogeneity observed within P. caudatum is consistent with multiple origins through independent hybridization events. Pteridium caudatum is strikingly analogous to the tropical Asian/Australasian allotetraploid P. semihastatum (N. Wallich ex J. G. Agardh) S. B. Andrews [= P. yarrabense (Domin) N. A.
