水稻白叶枯菌(Xanthomonas oryzae Dawson)重复侵染时,往往先侵害叶片,其侵染途径,一般都由叶片前端的水孔和气孔器侵入。水稻叶片顶端没有明显的排水器,但是分布一些水孔。这些水孔较大,排列较不规则。用人工喷雾接种的白叶枯细菌,由水孔和其他一些气孔器侵入后,通过靠近水孔的管胞,进入大小叶脉的导管。叶片上的许多横向小脉,对病菌的扩散也有很大关系。有时在尚未完全分化的、生活的管胞或导管细胞内也可看到病菌。用针刺接种的,发病较快,而扩散方式基本上与喷雾接种的一样。  相似文献

用一个具链霉素(Sm)抗性,含无启动子CAT基因的广宿主启动子探针质粒pIJ3100.用鸟枪法克隆水稻白叶枯病菌染色体DNA的BamH1酶切片段,重组质粒转化大肠杆菌ED_(8767),在含链霉素和氯霉素的LB平板上筛选含有水稻白叶枯病菌启动子活性片段的转化子。在帮助质粒pRK2013的帮助下,通过三亲交配,将转化子中重组质粒转移进野生型的水稻白叶枯病菌中去,在分别含有链霉素和氯霉素的PSA对照平板上筛选对热氯霉素敏感的接合子。随机挑取200个该接合子,接种用氯霉素处理的水稻感病品种金南风,得到15个比对  相似文献

在已明确我国存在甲霜灵抗性菌株的基础上,通过单游动孢子分离技术获得马铃薯晚疫病菌DZO1和HZO1无性后代菌系。研究了无性单游动孢子后代菌系的生物学特性及甲霜灵抗性,主要结果如下:对无性后代菌系的菌落形态、生长量和产孢能力进行测定,发现被测的DZO1和HZO1无性后代菌系大部分菌株的生物学性状与亲本菌株相比没有发生明显的变异,个别菌株的生物学性状与亲本菌株有显著差异,说明晚疫病菌的无性繁殖未引起较大的变异;对后代菌系的甲霜灵抗性水平进行测定,结果表明DZO1和HZO1无性后代菌系的甲霜灵抗性水平与其对应的亲本菌株基本相同。  相似文献

从多种植物叶片内分离、纯化出31株植物内生真菌,活性筛选表明分离自健康水稻叶片内生真菌Bo-1菌的乙酸乙酯粗提物对水稻白叶枯病菌具有较强的抑菌活性。当供试浓度为100μg/mL时,Bo-1菌乙酸乙酯粗提物对水稻白叶枯病菌抑菌圈为22.4 mm,与同浓度的氯霉素对水稻白叶枯病菌的抑菌效果相当。通过形态学特征观察和ITS rDNA测序分析,Bo-1菌被鉴定为淡色生赤壳菌(Bionectria ochroleuca)。抗菌谱实验表明,Bo-1菌对多种细菌类病原体具有较强的拮抗能力。  相似文献

以转拟南芥AtNPR1基因的恢复系品种桂99T3代纯合株系为材料,考查其农艺性状及其抗病性,并比较转基因植株与桂99侵染水稻白叶枯病菌后的农艺性状。结果表明:转基因植株表现出对水稻白叶枯病的抗性显著增强77%以上;穗长、剑叶长、有效穗数、一次枝梗数、每穗实粒数、单株产量和谷粒宽等农艺性状与未转基因桂99无显著差别。在受到水稻白叶枯病菌侵染后,转基因植株的一次枝梗数、每穗粒数、每穗实粒数和单株产量等方面均比对照桂99高出13%~78%。说明AtNPR1基因增强了水稻的抗病能力,从而降低了病害引起的产量损失。转基因植株的恢复力不受影响,稻米品质比桂99更加优良。本工作为转基因水稻抗病育种的研究奠定了基础。  相似文献

为降低水稻品种中花8号,10号的株高,同时又能保持这两个品种原有的丰产特性.我们从1986年起,用它们的幼穗进行组织培养,对体细胞突变体后代的植株高度等性状进行了观察,并对选择的矮杆突变体的其它农艺性状和产量等作了调查,从中选择矮杆、高产的突变细胞系用于生产。水稻幼穗经体细胞培养后,株高的变异明显,矮杆突变体出现的频率高。原品种中花8号、10号的植株偏高,平均株高分别是:113.6厘米和114.5厘米。中花8号、10号的株高突  相似文献

经实验我们成功地建立了在细胞水平上筛选烟草抗黑胫病突变体的筛选体系。该体系的主要内容为:γ-射线500—2000拉德诱变高度感病品种的花药后用50—80%的黑胫病菌粗毒素为选择压力,筛选出抗毒素花粉植株,用离体叶片法测定选出抗病植株,再从后代鉴定中选出抗病性能够稳定遗传的突变系。γ-射线及高浓度毒素处理均能得到抗病植株。选自感病品种的花粉植株中约有9—50%是真正抗病的。这些抗病植株中有一部分的抗病性能够稳定遗传。用该法已从感病优质品种小黄金1025及乔庄黑苗中选出6个突变系。并自N.C.628(抗)×小黄金1025(感)及N.C.628(抗)×庆胜2号(感)的F_1花粉植株中选出4个抗病系。所有的抗病系经3—4代后均表现出稳定抗性。其中一个突变体(R400)的抗性似由不完全显性多基因控制。  相似文献

Fifty pineapple buds (cv. Red Spanish Pinar, donor) were collected from field-grown plants and cultured in vitro. Forty-three young pineapple shoots were obtained after 42 d of implantation. Shoots were micropropagated for 168 d to produce 24,768 shoots. Three hundred young leaves were randomly selected as explants for callus formation. Calli proliferated for 4 months. Five hundred calli were randomly selected and transferred to the plantlet regeneration medium. Four hundred twenty-seven in vitro-plantlets were obtained and later hardened ex vitro. Then, 387 plantlets were transferred to the field environment and asexually propagated for two generations (30 months). Only two phenotype variants were identified: P3R5 and Dwarf. A more detailed study was carried out to compare these two variants with the donor plant. The variant P3R5 showed differences in the number of slips and suckers, and in the presence of thorns in the leaves and in the fruit crowns. The somaclonal variant Dwarf, was different from the donor plant in regard with the plant height; the peduncle diameter; the number of shoots, slips and suckers; the fruit mass with crown; the number of eyes in the fruit; the fruit height and diameter; the leaf color; the plant architecture; the length of plant generation cycle; and the fruit color and shape. Both somaclonal variants showed different AFLP banding patterns in comparison with the donor cultivar.  相似文献

Sulfate is important for mammalian development but is not routinely measured in clinical settings. The renal NaS1 sulfate transporter maintains circulating sulfate levels and is linked to renal sulfate wasting in mice. Some autistic individuals exhibit renal sulfate wasting, but the etiology is yet unknown. We measured plasma and urinary sulfate levels, calculated the fractional excretion index (FEI) of sulfate, and screened for two loss-of-function NaS1 sequence variants (R12X and N174S) in 23 autistic individuals. The FEI sulfate values ranged from 0.13 to 0.50. NaS1 variants were detected in 18 of the 23 individuals (11 heterozygous N174S, four homozygous N174S, two heterozygous R12X, and one individual carried both R12X and N174S). Those individuals with neither sequence variant had FEI sulfate ≤ 0.34, whereas FEI sulfate ≥ 0.35 was found in about 60 % (11 of 18) of individuals that had R12X and/or N174S. This study links renal sulfate wasting with loss-of-function NaS1 sequence variants in humans.  相似文献

The occurrence of somaclonal variation among regenerants derived through indirect shoot organogenesis from leaf explants of three Dieffenbachia cultivars Camouflage, Camille and Star Bright was evaluated. Three types of somaclonal variants (SV1, SV2, and SV3) were identified from regenerated plants of cv. Camouflage, one type from cv. Camille, but none from cv. Star Bright. The three variants had novel and distinct foliar variegation patterns compared to cv. Camouflage parental plants. Additionally, SV1 was taller with a larger canopy and longer leaves than parental plants and SV2. SV2 and SV3 did not produce basal shoots (single stem) but basal shoot numbers between SV1 and parental plants were similar ranging from three to four. The variant type identified from regenerated cv. Camille had lanceolate leaves compared to the oblong leaves of the parent. This variant type also grew taller and had a larger canopy than parental plants. The rates of somaclonal variation were up to 40.4% among regenerated cv. Camouflage plants and 2.6% for regenerated cv. Camille. The duration of callus culture had no effect on somaclonal variation rates of cv. Camouflage as the rates between plants regenerated from 8 months to 16 months of callus culture were similar. The phenotypes of the identified variants were stable as verified by their progenies after cutting propagation. This study demonstrated the potential for new cultivar development by selecting callus-derived somaclonal variants of Dieffenbachia.  相似文献

We previously reported a partial agricultural and amplified fragment length polymorphism (AFLP) characterization of two new pineapple somaclones (P3R5 and Dwarf) derived from in vitro culture of the donor cv. Red Spanish Pinar. Both somaclonal variants showed different AFLP banding patterns compared to the donor cultivar, although they were separated by less than 0.09 U of genetic distance. The present report shows data of various indicators of morphology and physiology of P3R5 and Dwarf D leaves. The stoma diameter, number of stomata per square millimiter, diameter of leaf vascular tissue, thickness of the leaf aquiferous parenchyma, and thickness of the leaf photosynthetic parenchyma were measured. The photosynthetic rate, the transpiration rate, the water use efficiency, the internal leaf CO2 concentration, and the chlorophyll pigment contents were recorded as well. Between the somaclonal variant P3R5 and the donor genotype, statistically significant differences were recorded in all indicators with the exception of the stoma diameter and the photosynthetic rate. Comparing the somaclonal variant Dwarf and the cv. Red Spanish Pinar (donor), statistically significant differences were also recorded in all parameters except in the stoma diameter and in the transpiration rate. This investigation was performed to demonstrate that small changes in the pineapple DNA may result in relevant phenotypic modifications.  相似文献

We report a high frequency regeneration protocol in cowpea (Vigna unguiculata Walp. var. C 152) via somatic embryogenesis from 10-d-old primary leaf explants. A study was conducted to examine the effect of somaclonal variations in in vitro derived cowpea plants under field conditions. The regenerated plantlets were successfully transferred to field after hardening in vitro and grown for collecting R0, R1 and R2 seeds. The seeds of R1 and R2 generations were subsequently, grown under field conditions and their various biometrical traits were compared and evaluated with non-tissue cultured cowpea plants as check. There was no detectable somaclonal variation induced in R0-R2 in any of the biometrical traits. The results indicate that the inclusion of different plant growth promoters at specified concentrations and duration in our earlier tissue culture work did not induce any detectable mutation. The RAPD analysis also shows that there is no genetic variation among R2 cowpea plants. The somatic embryogenesis protocol we report could thus be safely applied for high frequency true-to-type regeneration and transformations protocols without any somaclonal variation.  相似文献

CXCR4-using (X4) human immunodeficiency virus type 1 (HIV-1) variants evolve from CCR5-using (R5) variants relatively late in the natural course of infection in 50% of HIV-1 subtype B-infected individuals and subsequently coexist with R5 HIV-1 variants. This relatively late appearance of X4 HIV-1 variants is poorly understood. Here we tested the neutralization sensitivity for soluble CD4 (sCD4) and the broadly neutralizing antibodies IgG1b12, 2F5, 4E10, and 2G12 of multiple coexisting clonal R5 and (R5)X4 (combined term for monotropic X4 and dualtropic R5X4 viruses) HIV-1 variants that were obtained at two time points after the first appearance of X4 variants in five participants of the Amsterdam Cohort Studies on HIV-1 infection and AIDS. Recently emerged (R5)X4 viruses were significantly more sensitive to neutralization by the CD4-binding-site-directed agents sCD4 and IgG1b12 than their coexisting R5 viruses. This difference was less pronounced at the later time point. Early (R5)X4 variants from two out of four patients were also highly sensitive to neutralization by autologous serum (50% inhibition at serum dilutions of >200). Late (R5)X4 viruses from these two patients were neutralized at lower serum dilutions, which suggested escape of X4 variants from humoral immunity. Autologous neutralization of coexisting R5 and (R5)X4 variants was not observed in the other patients. In conclusion, the increased neutralization sensitivity of HIV-1 variants during the transition from CCR5 usage to CXCR4 usage may imply an inhibitory role for humoral immunity in HIV-1 phenotype evolution in some patients, thus potentially contributing to the late emergence of X4 variants.  相似文献

Macrophagetropic R5 human immunodeficiency virus type 1 (HIV-1) isolates often evolve into dualtropic R5X4 variants during disease progression. The structural basis for CCR5 coreceptor function has been studied in a limited number of prototype strains and suggests that R5 and R5X4 Envs interact differently with CCR5. However, differences between unrelated viruses may reflect strain-specific factors and do not necessarily represent changes resulting from R5 to R5X4 evolution of a virus in vivo. Here we addressed CCR5 domains involved in fusion for a large set of closely related yet functionally distinct variants within a primary isolate swarm, employing R5 and R5X4 Envs derived from the HIV-1 89.6(PI) quasispecies. R5 variants of 89.6(PI) could fuse using either N-terminal or extracellular loop CCR5 sequences in the context of CCR5/CXCR2 chimeras, similar to the unrelated R5 strain JRFL, but R5X4 variants of 89.6(PI) were highly dependent on the CCR5 N terminus. Similarly, R5 89.6(PI) variants and isolate JRFL tolerated N-terminal CCR5 deletions, but fusion by most R5X4 variants was markedly impaired. R5 89.6(PI) Envs also tolerated multiple extracellular domain substitutions, while R5X4 variants did not. In contrast to CCR5 use, fusion by R5X4 variants of 89.6(PI) was largely independent of the CXCR4 N-terminal region. Thus, R5 and R5X4 species from a single swarm differ in how they interact with CCR5. These results suggest that R5 Envs possess a highly plastic capacity to interact with multiple CCR5 regions and support the concept that viral evolution in vivo results from the emergence of R5X4 variants with the capacity to use the CXCR4 extracellular loops but demonstrate less-flexible interactions with CCR5 that are strongly dependent on the N-terminal region.  相似文献

The aim of this research was to characterize somaclonal variation in populations derived from embryos cultured on two types of induction medium (supplemented with either 2,4-D or dicamba), as well as to select and characterize several somaclonal lines. The sexual progenies of 40 R(0)regenerants - A somaclones (derived on the medium with 2,4-D) and B somaclones (derived on the medium with dicamba) - were analysed according to the following traits: plant height, total number of tillers, number of productive tillers, spike length, number of spikelets per spike, spike compactness, number of normally developed grains per spike, weight of grains per spike, and the weight of 1000 grains. The results for twenty-two R(1)plants surpassed the variability range for the control. The transmission of positive changes to the next generation was proved in the case of 8 originally chosen R(1) plants: 7 plants selected from the A somaclones and one plant from the B somaclones. Five out of the eight created somaclonal lines proved to be stable somaclonal variants. The absolute rate of the efficiency of positive somaclonal changes was calculated as 0.64%.  相似文献

Using the fluorescence in situ hybridization (FISH) technique, we conducted karyotype analyses to identify the lost chromosomes in three somaclonal variants obtained from tissue culture of wildAllium tuberosum (2n = 4X = 32). The three lost chromosomes of the At29 variant (2n = 29) were all chromosome 2, the two for At30 (2n = 30) were chromosomes 7 and 8, and At31 was missing chromosome 2. Chromosome compositions of these variants were confirmed as being fixed lines during two years of greenhouse cultivation. The bicolor FISH technique, involving both 5S and 18S–5.8S–26S ribosomal RNA genes as probes, was used to assign chromosomal locations and to confirm whether the lost chromosomes contained any rRNA markers. The 5S rRNA gene signals in all variants as well as the wild type were detected as two sets, one on the intercalary region of the short arm of chromosome 3, the other on the intercalary region of the long arm of chromosome 6. One 18S–5.8S–26S rRNA gene site on the secondary constriction included a flanking satellite and terminal region on the short arm of chromosome 8. Signals of the 18S–5.8S–26S rRNA gene in At30 showpd in only three chromosomes, indicating that one of the lost chromosomes was chromosome 8. Overall, three marker chromosomes were established by FISH, using rRNA multigene families.  相似文献

The magnitude of the effect of salt stress on proline content, pyrroline-5-carboxylate (P5C) reductase activity and water relations was found to be leaf position dependent in an advance generation (R4) of twoBrassica juncea L. somaclones (SR-2 and SR-3) selected in vitro for NaCl-tolerance and the parent cv. Prakash. Free proline content and P5C reductase activity increased with increase in salt stress in all the lines but at different rates; the maximum increase being in the SR-3 derived somaclonal line. At 100 mM NaCl, SR-3 showed a nearly 19 fold increase in proline content compared to a 4–5 fold increase in the other two genotypes. The proline level and P5C reductase activity of the first (youngest) leaf was higher than in the other leaves and decreased linearly with increase in age of the leaf in all the lines. The relationship between relative water content and osmotic potential of the leaves at different positions also varied. The results indicate that a significant effect of salt may appear non-significant if the position of the leaves is not taken into account while sampling.  相似文献
