  • 1.1. An endoxylanase (EC 3.2.1.8) was purified from an Escherichia coli strain carrying a xylanase gene from the extreme thermophile “Caldocellum saccharolyticum strain Tp8T6.3.3.1. It was found to have an Mr of 42,000 and an isoelectric point of approx. 5.0.
  • 2.2. The enzyme showed optimum activity at pH 5.0–7.7 and had an activation energy of 44 kJ mol−1. It was stable at room temperature at pH 4.5–11.5 in the presence of 0.5 mg ml−1 bovine serum albumin. The half-life of the enzyme at 75°C was 20 min at pH 6.0 in the presence of 0.5 mg ml−1 bovine serum albumin.
  • 3.3. The xylanase had highest activity on oat spelts xylan, releasing xylobiose and some xylotriose. The Km for oat spelts xylan was 0.021% (w/v) at pH6.0.
  • 4.4. The enzyme had high activity on sugar cane bagasse hemicelluloses A and B, lower activity on larchwood xylan and also hydrolysed carboxymethylcellulose, 4-methylumbelliferyl β-D-cellobioside and p-nitrophenyl β-D-cellobioside, but could not hydrolyse xylobiose.
  • 5.5. It showed transferase activity on p-nitrophenyl β-D-xylopyranoside. Xylose did not inhibit the enzyme.

  1. The main pathway of the anaerobic metabolism of l-malate in Saccharomyces bailii is catalyzed by a l-malic enzyme.
  2. The enzyme was purified more than 300-fold. During the purification procedure fumarase and pyruvate decarboxylase were removed completely, and malate dehydrogenase and oxalacetate decarboxylase were removed to a very large extent.
  3. Manganese ions are not required for the reaction of malic enzyme of Saccharomyces bailii, but the activity of the enzyme is increased by manganese.
  4. The reaction of l-malic enzyme proceeds with the coenzymes NAD and (to a lesser extent) NADP.
  5. The K m-values of the malic enzyme of Saccharomyces bailii were 10 mM for l-malate and 0.1 mM for NAD.
  6. A model based on the activity and substrate affinity of malic enzyme, the intracellular concentration of malate and phosphate, and its action on fumarase, is proposed to explain the complete anaerobic degradation of malate in Saccharomyces bailii as compared with the partial decomposition of malate in Saccharomyces cerevisiae.

EPR spectroscopy is a powerful tool to identify at a molecular level, the different steps of catalyst preparation, and of catalytic reactions:
  1. Deposition of paramagnetic transition metal ions onto a support is monitored, and the coordination sphere of the metallic center is characterized by EPR.
  2. The catalyst is also characterized after activation (thermal oxidation or reduction):
  • - the distribution among the different sites in zeolites can be determined;
  • - the dispersion of the active phase may be appreciated;
  • - the unsaturation degree of the active site may be evaluated using probe molecules such as water or13C enriched carbon monoxide.
    1. The catalytic mechanisms can be investigated by studying the elementary steps of the catalytic reaction, as illustrated for methanol oxidation over Mo/SiO2 catalysts whose EPR results have extended the reaction mechanism proposed on the basis of kinetic data. In addition, reaction intermediates may be isolated inquasi-in situ conditions as in the case of olefin oligomerization catalyzed by Ni/SiO2 systems.

    • 1.1. The larvae of mayflies (Baetis thermicus) that inhabited a metal-contaminated river (the river Mazawa in Yamagata, Japan) were collected and the concentrations of several heavy metals in mayflies were compared with those inhabiting a river not contaminated with metals.
    • 2.2. Cadmium, copper and zinc in mayflies of the contaminated river were 13.4, 18.4 and 15.6 times higher in concentration than those in the non-contaminated river.
    • 3.3. Distributions of the three metals in the supernatants of the larvae collected from the contaminated and non-contaminated rivers were determined by HPLC-atomic emission spectrometry with inductively coupled argon plasma.
    • 4.4. Cadmium and copper in the supernatant of the larvae collected from the contaminated river were sequestered by metal-binding components induced for the respective metals, while zinc seemed to be bound loosely to native components.

    Galanthamine (GAL) is increasingly used in the treatment of Alzheimer's disease. We have attempted to develop a method of producing this alkaloid using in vitro cultures of Narcissus confusus plants. The “shoot-clump” culture in liquid medium was shown to be an appropriate method for the micropropagation of this bulbous plant. The complete process included three steps:
    1. culture of “twin-scales” starting from the bulbs;
    2. culture of the newly formed shoots in a medium for bud proliferation (Murashige Skoog+1 mg l-1 of 2,4-dichlorophenoxyacetic acid+5 mg l-1 of benzyladenine), and
    3. culture of “shoot-clumps” in a liquid-shake medium. Here we describe the effect of the addition of trans-cinnamic acid, a precursor in the biosynthesis of the Amaryllidaceae alkaloids, on the production of galanthamine and related alkaloids, and also on the growth of the “shoot-clump” culture. The production of galanthamine was found to be inhibited by the addition of the precursor, which promoted the production of the other alkaloid in the same biosynthetic pathway, N-formyl-norgalanthamine. The total production of galanthamine in the control cultures in day-long photoperiod was 2.50 mg per culture, of which 1.97 mg per culture were released into the medium.

    1. The aim of this investigation was to elucidate how four acquatic insect larvae, from different habitats and having different respiratory organs or types of respiratory regulation, react to a lowered oxygen concentration, and how their oxygen consumption is affected. The species investigated were the stoneflies Taeniopteryx nebulosa, Diura nanseni and Nemoura cincerea and the mayfly Cloëon dipterum.
    2. The measurements were performed in a respiratory apparatus of open, flowing-water type. Its design is shown in Fig. 1. Water of known oxygen concentration was allowed to flow past the experimental larvae. The oxygen consumption of the larvae was calculated from the lowering of the oxygen concentration in which ensued.
    3. The water used in the experiments was standardized, so that the electrode had the necessary stability (conductivity 470 micromhos/cm). The calcium ion was excluded in order to prevent the precipitation of CaCO3 in the electrode capillary.
    4. A large variation in the values of oxygen consumption was found as seen in Fig. 2–5. The reason for that is a corresponding variation in the motor activity of the experimental animals.
    5. The physiological reasons for the general form of the curves A and C in Fig. 2–5 are discussed. The curves A and C represent oxygen consumption of the larvae at different degrees of stimulation, entailing different levels of motor activity. Curve A represents intentinally activated animals, curve C non-activated, motionless animals. The curves A and C are boundary curves corresponding to a sort of scope for activity of the animals. Over this scope area a series of curves of the same form could in principal be construed, representing different degrees of stimulation.
    6. Within a certain oxygen concentration interval a motor activation was observed caused by a reduced oxygen concentration. The result of that activation can be seen in Fig. 2–5 as a zone with no or very few oxygen consumption values between curve C and D. The more easily activated the species is, the broader the zone will be. Cloën has the most narrow zone and was observed to be less activated than the other species.
    7. Small larvae of Cloën (2–4 mm and 42–6 mm) and Nemoura (2–4 mm) showed clearly a greater ability to take up oxygen at low oxygen concentrations than full-grown larvae (see Fig. 8 and 9).
    8. The critical point on the curve representing mean oxygen consumption as a function of oxygen concentrations was found to be at 2–5 mg O2/1 for Taeniopteryx and Diura, at 2.2–2.5 mg O2/1 for Cloëon, and at about 2–7 mg O2/1 for Nemoura. The values refer to 8°. Cloëon is the species which is exposed to the greatest variations in oxygen concentration in its natural environment.
    9. No influence on the oxygen consumption of starvation for 4 to 5 days was found. No difference between the oxygen consumption values obtained in the presence or in the absence of calcium ions could be observed during the experiments (Fig. 10, 11).
    10. The basic picture obtained in this investigation is a set of oxygen consumption values scattered between a curve connecting highest values obtained and a curve of the standard metabolism together with a zone in which the larvae are activated by reduced oxygen concentrations. This picture is presumed to be general in aquatic animals with a well developed motor activity.

    1. Polyhedral particles were isolated from cells of Nitrobacter winogradskyi and of Nitrobacter strains K1, K4 and α1. Their physical and biological properties are characterized.
    2. The investigated strains contain polyhedral particles, 1000–1200 Å in size. With increasing age of the culture more particles are found in cells of Nitrobacter. Simultaneously the number of colony producing nitritoxidants decreases.
    3. In strain α1 the loss of the capability to form colonies is connected with partial lysis of the cell and release of particles.
    4. A homogeneous fraction of particles was obtained by zone density gradient centrifugation in Tris-Mg-SH-buffer.
    5. The polyhedral particles have a sedimentation coefficient of s w,20 0 =825S and a CsCl-buoyant density of ?25 g/cm3.
    6. Based on the determined properties the particles are classified as phage-like Nitrobacter particles Nb1.

    • 1.1. The binding of O2 to goldfish haemoglobin showed a strong pH dependence P50=5.5 mmHg; n = 2.4 at pH 8.0 and P50 = 170 mmHg; n = 1.0 at pH 5.5 such that the protein is only 50% saturated in a solution of air equilibrated buffer at pH 5.5.
    • 2.2. The binding of CO is cooperative at high pH (n = 2.8; L = 1000; KR = 0.1 μM; KT = 4 μM) and non-cooperative (n = 1) at pH 5.5.
    • 3.3. The rate of O2 dissociation is extremely fast and pH dependent; being 30 sec−1 at pH 8.0 and 400 sec−1 at pH 6.0 at 1°C. At 23°C the rate of this process is too fast to obtain accurate data using stopped-flow techniques.
    • 4.4. Partial photolysis of the oxyhaemoglobin species leads to homogeneous recombination kinetics at pH 8.0 with an associated rate constant of 4.7 × 107 M−1 sec−1. At pH < 7.5 the recombination process occurs in two steps. One rate is equal to that observed at pH 8.0. The slower process is favoured at low pH.
    • 5.5. Photolysis of the CO haemoglobin complex indicates that, at high pH, combination of CO with deoxyhaemoglobin is cooperative, whilst recombination with Hb(CO)3 is non-cooperative and occurs at a rate of 1.2 × 106 M−1 sec−1.
    • 6.6. At neutral pH recombination of CO with partially linganded haemoglobin occurs in a two-step process. The proportion contributed by each of these two steps in pH dependent.
    • 7.7. The functioning of this Root effect haemoglobin is discussed in terms of the two state-model of cooperativity in which the αβ chain heterogeneity is minimal

    • 1.1. The temperature and water relations of Centruroides hentzi females were investigated. At 12 and 72% relative humidity (RH), the lower and upper Lt50 were -4.5 and 43.7°C, and -4.7 and 45.1°C, respectively. When exposed to high temperature stress, survivorship was significantly greater under mesic conditions.
    • 2.2. Cuticular water loss was higher under xeric conditions (12% RH), ranging from 0.061 mg/cm2/hr at 30°C to 0.211 at 41°C.
    • 3.3. Exposure to dry air (0–5% RH) resulted in a significant increase in hemolymph osmolality: from 441 to 688 mOsm over a 5 day period.
    • 4.4. Mean oxygen consumption rates increased from 161.7 mm3/g/hr at 34°C to 541.6 at 44°C. ATPase activity was significantly higher in animals acclimated and tested at 35°C.

    1. The 1-P-fructokinase (1-PFK) and 6-P-fructokinase (6-PFK) from Pseudomonas doudoroffii were partially purified by a combination of (NH4)2SO4 fractionation and DEAE-Sephadex column chromatography. The pH optima of these enzymes were 9.0 and 8.5, respectively.
    2. When the concentrations of the substrates of the 1-PFK reaction were varied, Michaelis-Menten kinetics were observed. The Kms for d-fructose-1-P (F-1-P) and ATP were 3.03×10-4 M and 3.39×10-4 M, respectively. Variation of MgCl2 at fixed concentrations of F-1-P and ATP resulted in sigmoidal kinetics; about 10 mM MgCl2 was necessary for maximal activity. Activity of 1-PFK was inhibited when the ratio of ATP: Mg++ was higher than 0.5, suggesting that ATP: 2Mg++ was the substrate and that free ATP was inhibitory. Although an absolute requirement for K+ or NH + 4 could not be demonstrated, these cations stimulated the rate of the reaction. Activity of 1-PFK was not significantly affected by 3 mM AMP, cyclic-AMP, Pi, d-fructose-6-P (F-6-P), ADP, P-enolpyruvate (PEP), pyruvate, citrate, or l-glutamate.
    3. Sigmoidal kinetics were observed for 6-PFK when the concentration of F-6-P was increased and the level of ATP was kept constant. Activity of 6-PFK was increased by ADP, inhibited by PEP, and unaffected by 3 mM AMP, cyclic-AMP, Pi, F-1-P, pyruvate, or citrate.

    The present study was carried out to find the effects of Pb acetate (10-50 mg/kg body wt) after oral administration on:
    1. The distribution of elements, such as Fe, Cu, Zn, and Mn;
    2. The activity of 6-amino levulenic acid dehydratase (δ-ALAD) and alkaline phosphatase (PAP); and
    3. On the level of reduced glutathione (GSH) in murine placenta.
    Pb toxicity expressed on a dry-wt basis was reflected in terms of deficiency of δ-ALAD and PAP and enhanced content of GSH. Analysis of trace elements following Pb exposure showed low levels of Mn and Cu. Although Fe composition of placenta remained within normal range with increasing load of endogeneous Pb, Zn decline was not consistent after oral feeding of Pb acetate. Deficiency of PAP after Pb exposure did not correlate with the endogeneous levels of Pb or Zn therein, but correlated with endogeneous levels of Mn. Placental deficiencies of Cu and Mn have been related to the disturbed placental functions by Pb accumulation.  相似文献

    1. Cell-free extracts from vegetative cells and developing myxospores of Myxococcus xanthus were found to contain similar amounts of proteolytic activity, approximately 80% of which was due to one or more neutral metal proteases.
    2. Sixty per cent of the proteolytic activity was particulate.
    3. The specific activity of the proteases was high throughout all stages of myxospore formation and displayed small increases in activity at two stages of development: (1) during cell shortening and (2) immediately following the conversion to spheres. The first peak in activity was apparent in assays conducted at pH 8 or 10 whereas the second peak was obvious only at pH 6.
    4. A mutant which develops into myxospores only after a lag of approximately 7–8 h possessed levels of proteases similar to the wild type and displayed a peak in proteolytic activity after a delay of 7–8 h.
    5. Low levels of serine protease activity were occasionally detected in both vegetative cells and myxospores; no sulfhydryl proteases were detectable in either cell type.
    6. Extracellular proteases accumulated in the medium throughout myxospore development but differed from the intracellular proteases in pH optima and sensitivity to inhibitors.

    1. The affinity of ATP-supported Ca2+ accumulation for both Ca2+ and ATP was determined from initial rate studies employing isolated rat liver mitochondria. TheK m values for “free” Ca2+ and ATP were calculated to be of the order of 2 μM and 100 μM, respectively. TheK m for ATP decreased as the Ca2+ concentration was increased.
    2. The curve relating initial rates of Ca2+ accumulation to Ca2+ concentration was singmoidal in shape; values obtained for the Hill coefficient were in the range 1.5–1.9.
    3. Concomitant with the ATP-stimulated accumulation of Ca2+, ATP translocation was itselt increased in the presence of Ca2+. This stimulation took place independently of Ca2+ accumulation.
    4. Decreasing the pH of the incubation medium decreased the rate of Ca2+ accumulation. This inhibition was competitive in that the affinity of mitochondrial for Ca2+ could be altered. The maximal rate of accumulation did not change with change in pH.
    5. The permeant anions inorganic phosphate and acetate stimulated the accumulation of Ca2+ in a non-competitive manner. Both theV max and Km varied when either of the anions were present.
    6. The data are discussed in relation to the role that mitochondria play in controlling the cellular ionic environment.

    • 2.2. Reductions in bathing solution pH inhibited Jmsna (mucosal-to-serosal flux) and Jnetna (net flux) (r = 0.90) and JnetCl (r = 0.92) [due to an increase in JsmCl, (serosal-to-mucosal)] and decreased short circuit current (Isc).
    • 3.3. Luminal bumetanide (0.1 mM) and amiloride (1 mM) inhibited Na+ and Cl absorption by reducing Jms.
    • 4.4. Luminal barium (5mM) and luminal copper (100 μM) decreased JmsCl and increased JsmCl.
    • 5.5. We conclude that reductions in extracellular pH inhibit a luminal membrane NaCl absorptive process (Na+-K+-2Cl) and stimulate an electrogenic Cl secretory process.

    Mild (not harmful) stress may initiate anadaptive mechanism, protecting the heart from harmful consequences of a more severe stress. There are at least three known types of cardiac adaptation to stress such as:
    1. the gradually developing long lasting adaptation to chronic mechanical overload, leading to cardiac hypertrophy, later to cardiomyopathy and heart failure,
    2. the rapidly developing adaptation to moderate stress initiated by ‘preconditioning’ brief coronary occlusion(s) or brief periods of rapid cardiac pacing, protecting for less than 1 h against consequences of a subsequent, severe stress,
    3. the later appearing, more prolonged cardio-protective adaptation, described by us in 1983, induced by various forms of more severe but not injurious stimuli, such as an optimal dose of prostacyclin or its stable analogues; or a series of brief periods of rapid pacings.
    This form of cardiac adaptation to stress protects for 24–48 h against consequences of a more severe stress such as:
    1. myocardial ischaemia;
    2. early and late postocclusion and reperfusion arrhythmias;
    3. early morphologic changes secondary to ischaemia and reperfusion;
    4. ischaemia induced myocadial loss of K+ and accumulation of Na+ and Ca++;
    5. it may increase the tolerance to the toxic effects of cardiac glycosides.
    A reduced response to beta-adrenergic stimuli and a concomitant increase in activity and amount of PDE I and IV was shown by us earlier. The hypothesis that these factors may play a role in the mechanism of delayed protection was confirmed by our present findings according to which 7-oxo-PgI2-treatment greatly attenuated the dose dependent isoprenaline-induced increase in contractility, relaxation and myocardial cAMP level in rat hearts isolated 48h after 7-oxo-PgI2. In addition all these values are in close correlation with each other. The endogenous ‘self-defence’ of the heart based on adaptation represents anew therapeutic concept, different from the classical drug-receptor interaction produced protection. Its possible exploitation to therapeutic use requires that the adaptation inducing stress should beapplicable topatients, furthermore prolongation of duration of protection should be possible. As a first step in testing applicability to therapy we had to show that drug induced adaptive protection is existing in the conscious animal. In our present study we found an attenuation of rapid pacing induced elevation of the ST-segment in the endocardial electrogram and in the left ventricular end diastolic pressure in conscious rabbits 24–48 h after treatment with Iloprost. Besides we found an attenuation of tachycardia and arrhythmias due to two stage coronary artery ligation in conscious dog 48 h after pretreatment with 7-oxo-PgI2. Finally we were able to demonstrate that protection against coronary artery occlusion-induced ST segment elevation and arrhythmias can be prolonged at will by periodically repeated maintenance doses.  相似文献
