The site of anaerobic respiration in the latex is the serum. The main respiratory substrate is fructose. The CO2 formation in serum is increased by additional fructose on the average about 2.5–3 times. Glucose does not influence CO2 evolution by serum but slightly increases O2 consumption. With respect to sugars, latex serum contains essentially only sucrose and a low amount of raffinose. During the incubation of serum sucrose is hydrolysed, the fructose component is immediately utilized in respiration and glucose accumulates. The rate of CO2 formation in latex as influenced by fructose is negatively related to the rubber content of the latex. Latex with a high rubber content reacts only slightly or not at all on additional fructose. The main limiting factors of latex respiration and sugar utilization are the following:
  1. The deficiency of substrate, due to low activity of β-fructofuranosidase.
  2. The rate of glucose phosphorylation (D'Auzac, Jacob 1967).
  3. Presumably the low activity of phosphoglucoisomerase.
  4. The rubber content of the latex.
  5. The concentration of CO2 in latex; this factor may be important in vivo, in the laticiferous system.

Isolated microspores of Hordeum vulgare L. cv. Igri were incubated in the presence of different sugars. In the presence of maltose, the optimum concentration for the development of embryoids or calluses from the microspores was 175 mM. At this concentration 0.2% of the cells developed into embryoids or calluses. Microspores cultured without a carbohydrate source died after three days' incubation. In contrast microspores incubated in the presence of sucrose, glucose or fructose died within three days. Moreover, microspores also died when incubated in the presence of a combination of 175 mM maltose with varying concentrations of either sucrose, glucose or fructose. It is concluded that incubation of microspores in the presence of sucrose, glucose or fructose results in the death of the cells via some unknown mechanism. In contrast to this, maltose can sustain development of embryoids and calluses from cultured microspores.  相似文献

Androgenic hormones seem to be of beneficial effects on sports performance:
  • - they increase motivation, will, aggressiveness, resistance to the stress and to the fatigue, leading to an increase of the training quantity,
  • - they increase bone mineralization and probably mechanical resistance,
  • - they stimulate the bone marrow and so, with the erythropoietin, the erythropoiesis,
  • - they increase the tendancy to hyperglycemia, but with a decrease of the tolerance to the glucose,
  • - they stimulate the fatty acids mobilization from the adipose tissue, for their utilization in the muscle during the exercise,
  • - they participate, for the trained sportmen, to a better gestion of the muscle glycogen storage: their utilization during exercise is decreased,
  • - they increase the lean body mass, with an increase of the protein synthesis and a decrease of the protein catabolism, leading also to an increase of the muscle force under training. There is no beneficial effect upon the tendons,
  • - they have an immunomodulation action.

    1. The cellular content of galactolipids in Chlorobium and Chloroflexus is not related to bacteriochlorophyll content nor to the total amount of chlorosome material in the cells.
    2. Chlorosomes of both bacteria were agglutinated by Ricinus lectin and the agglutination was increased after treatment of the chlorosomes with trypsin.
    3. When cell free preparations of both bacteria were treated with trypsin prior to centrifugation on sucrose gradients, the resulting chlorosome fractions were less contaminated with material derived from the cytoplasmic membrane than when trypsin was not employed.

    1. The neutral red technique was employed to study the rate of filtration in Katelysia opima.
    2. The weight specific water filtration was found to be greater for younger clams compared to the older ones.

    4. Water filtration was found to increase as temperature increased, reaching a maximum at 35°C. but then sharply decreasing at 39°C.
    5. Light had no significant effect on the rate of filtration.

    8. The rate of water filtration was found to be faster during high tide than during low tide.
    9. The presence of the parasitic crab, Pennotheris sp., in the mantle cavity of clams had a marked effect on the particle filtration.
    10. Accidental cut of the siphon tips had no effect on the rate of filtration.

    • 1.1. The addition of cAMP to stimulating solutions of NaCl, fructose (furanose sugar), sucrose, or glucose (pyranose sugars) decreases the responsiveness of labellar chemosensilla in Phormia.
    • 2.2. The addition of ATP, while decreasing the responsiveness to NaCl or fructose enhances the responsiveness to glucose and sucrose.
    • 3.3. The inhibiting effect of ATP on NaCl or fructose responses is suppressed by GDPßS, an inhibitor of adenylate cyclase (and thus of cAMP synthesis); moreover GDPßS further enhances the increase in response due to ATP when added to the sucrose or glucose solutions.
    • 4.4. Results suggest a possible involvement of cAMP and ATP in the taste reception mechanism in the blowfly.

    Acetoin dehydrogenase can be catabolite repressed by numerous sources of carbon. The following results point out that the catabolite repression of this enzyme and the inhibition of sporulation are mediated by the same mechanism:
    1. Mutants, able to synthesize acetoin dehydrogenase in the presence of glucose, sporulate in glucose medium at a higher rate than the standard strain.
    2. The catabolite repressing effect of a compound and its ability to inhibit sporulation are in a direct relation to each other.
    3. The limitation of inorganic phosphate in the growth medium, which is known to favour sporulation, counteracts the catabolite repressing effect of glucose.

    1. Thioglycolic acid, a Cu-chelating agent, totally inhibited extracellular laccase activity without affecting growth and morphology of Fomes annosus.
    2. In the presence of thioglycolic acid Fomes annosus cleaved high molecular weight lignosulfonate with a molecular weight range of 2×106 to 1000. In the absence of thioglycolic acid the polymerizing activity of laccase prevented the detection of lignosulfonate breakdown products.
    3. Oxidative polymerization of a lignin monomer, coniferyl alcohol, occurred in the presence but not in the absence of laccase activity.
    4. Catechol and guaiacol added to the medium at a concentration of 2 mmol, are normally oxidized by fungal laccase and strongly inhibit growth. Presence of thioglycolic acid prevented the oxidation of these phenols and simultaneously permitted normal growth.

    1. The transfer of immature embryos from maternal plants to artificial media influenced the radial arrangement of vascular bundles in developing root primordia. The variability in the number of poles of the prospective protoxylem and protophloem, observed as a rule during embryogenesis under natural conditions, could not be suppressed even under the conditions ofin vitro cultivation. The possibility is admitted that when using agar medium the nutrient supply need not necessarily be equivalent for all embryos.
    2. Using excised embryos of various ages the period of delimination of the vascular system in the root primordium was determined. It is relatively short and occurs in the first half of embryogenesis. The results obtained revealed no relationship between vascular system arrangement in root primordium and mature grain and mature embryo size.
    3. Maize ear represents a type of inflorescence of which the apical part is delayed in development. Histogenically this uneven development becomes evident with the formation of a significantly lower mean number of poles in root primordia from the grains originating from the apical region of the cob. This is further evidence of the adaptibility of the vascular system development to environmental conditions.
    4. As further causes of the variability in pole number those differences are considered which occur during sex cell formation, pollination and fertilization.

    1. Comparisons were made of the effects of salt on the exponential growth rates of two unicellular algae,Dunaliella tertiolecta (marine) andDunaliella viridis (halophilic).
    2. The algae contained glycerol in amounts which varied directly with the salt concentration of the growth media. The highest measured glycerol content ofD. tertiolecta was approximately equivalent to 1.4 molal and occurred in algae grown in 1.36 M sodium chloride. The highest glycerol content measured inD. viridis was approximately equivalent to 4.4 molal and occurred in algae grown in 4.25 M sodium chloride. Lower concentrations of free glucose, which varied inversely with extracellular salt concentration, were also detected.
    3. It is inferred that Na+ is effectively excluded from the two algae. There was some evidence of a moderate uptake of K+.
    4. Comparisons were made of erude preparations of the glucose-6-phosphate dehydrogenase and an NADP-specific glycerol dehydrogenase from each species and of the effects of salt and glycerol on the activities of these enzymes. It is concluded that the different salt tolerances of the two algae cannot be explained by generalized differences between their enzyme proteins.
    5. Although intracellular glycerol must necessarily contribute to the osmotic status of the algae, its primary function in influencing their salt relations is considered to be that of a compatible solute, whereby glycerol maintains enzyme activity under conditions of high extracellular salt concentration and hence low (thermodynamic) water activity.

Malic enzyme was induced by malic acid and malo-lactic enzyme was induced by malic acid and glucose in cells of three strains ofLactobacillus casei that were able to grow on malate as carbon source. Two strains ofStreptococcus faecalis formed malic enzyme only, whereas only malo-lactic enzyme was formed by a glucose requiring strain ofStreptococcus lactis.
    2. Given sequential induction, cells ofLactobacillus casei M40 were found to contain malic enzyme and malo-lactic enzyme simultaneously.
    3. Malic enzyme and malo-lactic enzyme have been separated by chromatography on Sephadex G-200. These two enzymes have a different pH optimum, different affinities for substrates, form different end products from malate, and have molecular weights of 120000 and 150000 daltons respectively.

    The present paper deals with the coordination of energy metabolism, glucose consumption rate, glycolytic and TCA cycle enzyme activities in the lysine-producing bacterium Brevibacterium flavum. It is shown, that inhibition of the elctron transport chain causes changes of the following sequence:
  • at first, TCA cycle enzymes are activated;
  • secondly, TCA cycle enzyme activity decreases, and glycolytic enzyme activities as well as glucose transport rate increase; there is a slight increase in Qo2 and a considerable one of O2 consumption in cyanide-resistant respiration pathway;
  • thirdly, TCA cycle enzyme activities and glucose transport rate decrease.
  • It is supposed, that coordination of carbon and energy metabolism in B. flavum depends on intracellular ATP concentration or energy charge value.  相似文献

    • 1.1. The amount of sugar required for growth of Heliothis zea larvae on a chemically defined diet was determined.
    • 2.2. Larvae grew well on fructose, galactose, sucrose, trehalose and raffinose diets but not on diets containing more than 0.5% glucose.
    • 3.3. A starch diet did not promote rapid larval growth.
    • 4.4. Hemolymph trehalose levels in 12-day-old larvae ranged from none to 45μmoles/ml.
    • 5.5. A method for analysis of hemolymph trehalose by gas chromatography is described.

    • 1.1. A maximum rate of dolichyl phosphate [14C]glucose synthesis from 55-day embryos was achieved at 16nM concentration of exogenous dolichyl phosphate and exceeded about 3 times that without addition of dolichyl phosphate.
    • 2.2. The highest values of [14C]glucose incorporation from UDP-[14C]glucose into dolichyl phosphate [14C]glucose, dolichyl diphosphate [14C]Glc-oligosaccharides and proteins were reached at 5 min time point of incubation of liver microsomes both from embryos and sows.
    • 3.3. The radioactive incorporation into proteins was about 7-fold higher in liver microsomes from sows compared to that from embryos, probably due to the greater content of acceptor proteins in microsomes from sows.
    • 4.4. The enzymatic transfer of Glc3-oligosaccharide from a lipid carrier to endogenous protein acceptor in microsomes from pig embryonic and adult livers was considerably faster than the removal of glucose residues during the initial stages of processing of protein-bound oligosaccharides.
    • 5.5. One labelled compound was discovered in the Chcl3-Ch3Oh-H2O (1:1:0.3, by vol) extract after incubation of liver microsomes from embryos and sows with UDP-[14C]glucose. On the basis of its mobility on the chromatogram it appears to be GlcNAc2Man9Glc3.

    1. The lipid composition of a mutant ofSaccharomyces cerevisiae which cannot synthesize unsaturated fatty acid (UFA) can be extensively manipulated by growing the organism in the presence of added fatty acids.
    2. Growth of the mutant is supported by a wide range of unsaturated fatty acids including oleic, palmitoleic, petroselenic, 11-eicosaenoic, ricinoleic, arachidonic, clupanodonic, linoleic and linolenic acids; 9- and 10-hydroxystearic acids support growth less effectively, but erucic, nervonic, elaidic and saturated fatty acids (C8∶0?C20∶0)* are ineffective. All the fatty acids which support growth are incorporated into cell lipids, apparently without further metabolism.
    3. The effects of altered lipid composition on the energy metabolism of yeast cells were investigated. Cells containing less than approximately 20% of their fatty acids as UFA cannot grow on non-fermentable substrates, and their growth on glucose is restricted to that which can be supported by fermentation alone.
    4. UFA-depleted cells contain mitochondria which are apparently normal in morphology, furthermore they have normal levels of cytochromesa+a 3,b,c 1 andc and respire at normal rates. This suggests that the lesion in energy metabolism produced by UFA-depletion may be the loss of the ability of the mitochondria to couple respiration to phosphorylation.
    5. UFA-depleted cells incorporate added UFA into their cell lipids and subsequently regain the ability to grow on non-fermentable substrates, showing that the lesion in energy metabolism is fully reversible.

    1. Growth of the floating aquatic weed, Salvinia, in sterile culture was exponential for at least 2 weeks under standardized conditions.
    2. Increase in light intensity or in CO2 resulted in increases in growth rate, but did not extend the exponential period of growth.
    3. This aquatic plant, like many others, discriminates against calcium relative to strontium.
    4. In culture Salvinia exhibited luxury consumption of N and P.
    5. Because of high C/N ratios, Salvinia may not be a favorable source of animal food, but might be useful in nutrient removal schemes.
    6. In sterile culture, S. molesta produced fewer leaves than S. minima, but maintained a significant increase in leaf area and dry weight. This may be correlated with the ability of the first species to rapidly spread over tropical waterways.

    • 1.1. Electrophysiological responses to NaCl, sucrose, and a complex mixture containing NaCl, glucose, fructose, phenylalanine and valine were obtained from large, medium, and small hair sensilla on the proboscis of the fleshfly Sarcophaga bullata.

    1. Succinic acid is formed in amounts of 0.2–1.7 g/l by fermenting yeasts of the genusSaccharomyces during the exponential growth phase. No differences were observed between the various species, respiratory deficient mutants and wild type strains.
    2. At low glucose concentrations the formation of succinic acid depended on the amount of sugar fermented. However, the nitrogen source was found to be of greater importance than the carbon source.
    3. Of all nitrogen sources, glutamate yielded the highest amounts of succinic acid. Glutamate led to an oxidative and aspartate to a reductive formation of succinic acid.
    4. A reductive formation of succinic acid by the citric acid cycle enzymes was observed with malate. This was partially inhibited by malonate. No evidence was obtained that the glyoxylate cycle is involved in succinic acid formation by yeasts.
    5. Anaerobically grown cells ofSaccharomyces cerevisiae contained α-ketoglutarate dehydrogenase. Its activity was found in the 175000 x g sediment after fractionated centrifugation. The specific activity increased 6-fold after growth on glutamate as compared with cells grown on ammonium sulfate.
    6. The specific activities of malate dehydrogenase, fumarase, succinate dehydrogenase, succinylcoenzymeA synthetase, α-ketoglutarate dehydrogenase and glutamate dehydrogenase (nicotinamide adenine dinucleotide dependent) were determined in yeast cells grown on glutamate or ammonium sulfate. Similar results were obtained with a wild type strain and a respiratory deficient mutant. The latter did not contain succinate dehydrogenase.
    7. In fermenting yeasts succinic acid is mainly formed from glutamate by oxidation.

    1. The affinity of ATP-supported Ca2+ accumulation for both Ca2+ and ATP was determined from initial rate studies employing isolated rat liver mitochondria. TheK m values for “free” Ca2+ and ATP were calculated to be of the order of 2 μM and 100 μM, respectively. TheK m for ATP decreased as the Ca2+ concentration was increased.
    2. The curve relating initial rates of Ca2+ accumulation to Ca2+ concentration was singmoidal in shape; values obtained for the Hill coefficient were in the range 1.5–1.9.
    3. Concomitant with the ATP-stimulated accumulation of Ca2+, ATP translocation was itselt increased in the presence of Ca2+. This stimulation took place independently of Ca2+ accumulation.
    4. Decreasing the pH of the incubation medium decreased the rate of Ca2+ accumulation. This inhibition was competitive in that the affinity of mitochondrial for Ca2+ could be altered. The maximal rate of accumulation did not change with change in pH.
    5. The permeant anions inorganic phosphate and acetate stimulated the accumulation of Ca2+ in a non-competitive manner. Both theV max and Km varied when either of the anions were present.
    6. The data are discussed in relation to the role that mitochondria play in controlling the cellular ionic environment.
