Synchronized activity (spindles, augmentation response) evoked by stimulation of thalamic nonspecific, association, and specific nuclei was investigated in chronic experiments on 11 cats before and after successive destruction of the caudate nuclei. After destruction of the caudate nuclei the duration of spindle activity in the frontal cortex and subcortical formations (thalamic nuclei, globus pallidus, putamen) was reduced to only three or four oscillations. In the subcortical nuclei its amplitude fell significantly (by 50±10%); in the cortex the decrease in amplitude was smaller and in some cases was not significant. Different changes were observed in the amplitude of the augmentation response, depending on where it was recorded. In the subcortical formations it was considerably and persistently reduced (by 50±10%); in the cortex these changes were unstable in character. Unilateral destruction of the caudate nucleus inhibited synchronized activity evoked by stimulation of the thalamic nuclei on the side of the operation only. Destruction of the basal ganglia (caudate nucleus, globus pallidus, entopeduncular nucleus, and putamen) did not prevent the appearance of synchronized activity; just as after isolated destruction of the caudate nucleus, after this operation synchronized activity was simply reduced in duration and amplitude. It is suggested that the caudate nucleus exerts an ipsilateral facilitatory influence on the nonspecific system of the thalamus during the development of evoked synchronized activity.A. A. Evoked potentials arising in the motor cortex in response to its direct stimulation (dendritic and slow negative potentials), to stimulation of the ventrolateral (primary response) and intralaminar (nonspecific response) thalamic nuclei, and to stimulation of the pyramidal tracts (antidromic response), and also postsynaptic responses of neurons corresponding to them were studied in acute experiments on curarized cats. Evoked potentials arising in response to direct cortical stimulation and also to stimulation of the specific and nonspecific thalamic nuclei and pyramidal tracts were recorded from the same point of the motor cortex, and the corresponding intracellular responses were recorded from the same neuron. Slow negative potentials arising under these conditions of stimulation and the IPSPs corresponding to them were shown to have an identical time course. The results show that slow negative potentials are a reflection of hyperpolarization of pyramidal neurons. It is suggested that the individual components of responses evoked by direct stimulation of the cortex and thalamic nuclei have a common genesis.I. S. Responses of 137 neurons of the rostral pole of the reticular and anterior ventral thalamic nuclei to electrical stimulation of the ventrolateral nucleus and motor cortex were studied in 17 cats immobilized with D-tubocurarine. The number of neurons responding antidromically to stimulation of the ventrolateral nucleus was 10.5% of all cells tested (latent period of response 0.7–3.0 msec), whereas to stimulation of the motor cortex it was 11.0% (latent period of response 0.4–4.0 msec). Neurons with a dividing axon, one branch of which terminated in the thalamic ventrolateral nuclei, the other in the motor cortex, were found. Orthodromic excitation was observed in 78.9% of neurons tested during stimulation of the ventrolateral nucleus and in 52.5% of neurons during stimulation of the motor cortex. Altogether 55.6% of cells responded to stimulation of the ventrolateral nucleus with a discharge of 3 to 20 action potentials with a frequency of 130–350 Hz. Similar discharges in response to stimulation of the motor cortex were observed in 30.5% of neurons tested. An inhibitory response was recorded in only 6.8% of cells. Convergence of influences from the thalamic ventrolateral nucleus and motor cortex was observed in 55.7% of neurons. The corticofugal influence of the motor cortex on responses arising in these cells to testing stimulation of the ventrolateral nucleus could be either inhibitory or facilitatory.A. A. In acute experiments on cats anesthetized with pentobarbital and chloralose, single-unit and focal responses of the medial group of thalamic nuclei (mediodorsal, central lateral, paracentral, central medianum, parafascicular) were studied to stimulation of the frontobasal regions of the cortex (proreal, posterior orbital, basal temporal regions). Depending on the number of neurons responding to cortical stimulation and on the length of the latent period of the responses three functionally heterogeneous subdividions of the medial nuclei were distinguished; the parvocellular and magnocellular portions of the mediodorsal nucleus and the intralaminar nuclei with the parafascicular complex. On the basis of responses of neurons activated antidromically by stimulation of the same cortical region and synaptically by stimulation of another region, the concept of the integrative function of nuclei of the medial group, integrating the frontobasal zones of the neocortex with the aid of neuron circuits in which the medial nuclei are included, is argued.M. Stimulation of the head of the caudate nucleus in cats anesthetized with chloralose and pentobarbital evoked spike responses of the Purkinje cells and other cerebellar cortical neurons in the paramedian lobes, lobulus simplex, and the tuber of the vermis. Phasic responses in the form of simple discharges (on account of activation of the neurons through mossy fibers) appeared mainly after a latent period of 5–12 and 14–20 msec; the latent period of responses consisting of complex discharges (on account of activation of Purkinje cells through climbing fibers) was 5–6, 9–22 msec, or more. Depending on the latent period, the spike responses differed in their rhythm of generation. In response to stimulation of the caudate nucleus with a frequency of 4–6/sec recruiting responses were found. An inhibitory pause was an invariable component of the tonic responses. During stimulation of the globus pallidus responses of the same types (phasic and tonic) appeared as during stimulation of the caudate nucleus, but they differed in the distribution of the neurons by latent period of spike responses. The minimal latent period was 4 msec. Recruiting also was observed during repetitive stimulation of the globus pallidus. During stimulation of the substantia nigra Pukinje cells activated by climbing fibers responded. Evoked complex discharges appeared after a stable latent period of 8.5±0.3 msec. Arguments are put forward regarding the role of the substantia nigra, the globus pallidus, nuclei of the inferior olive, and also the thalamic nuclei in the mechanism of caudato-cerebellar oligosynaptic and polysynaptic connections.N. The reactions of neurons of the isolated cortex of one hemisphere to direct cortical stimulation were investigated in cats under Nembutal anesthesia. Isolation of the cortex was carried out by Khananashvili's method [10]. It is shown that phasic reactions develop in the isolated cortex in response to such stimulation: initial discharge, initial pause, first after-discharge, first after-pause, late after-discharges and pauses, as well as reactions of presumably inhibitory neurons. A majority of the cells (85%) which manifest background activity respond to direct electrical stimulation, and the frequency of the late after-reactions is twice as great as in the intact cortex. In acute experiments on cats evoked potentials (EP) of the orbital cortex were recorded and the electrogenesis and functional purpose of individual components of associative responses (AR) were investigated. It was concluded that the initial negative fluctuation of the AR arises as a consequence of the physical propagation of potentials from the projection somatosensory cortex and the second, positive, component and the following negative component are the result of arrival of an afferent volley into the orbital cortex via specific thalamic nuclei. These two components are due to activation of neurons of the orbital cortex. The afterdischarge, which appears sometimes, develops under the effect of impulses arriving from nonspecific thalamic nuclei. It is shown that during the second, positive, phase of the AR, primarily afferent neurons are activated, and during the negative phase, efferent neurons of the orbital cortex. The thalamic relays for the conduction of impulses arising during photic stimulation of the eyes and electrical stimulation of the tectum in the general cortex, hyperstriatum (the dorsal ventricular ridge), and the striatum proper were studied in the turtleEmys orbicularis. Acute experiments on immobilized animals showed that anodal polarization temporarily and destruction of n. rotundus irreversibly suppress the main negative wave of the responses to tectal stimulation and to flashes in the hyperstriatum, whereas the corresponding responses in the general cortex still persist. Polarization and destruction of the lateral thalamic region, including the lateral geniculate body, have the opposite effect: responses in the hyperstriatum to photic and tectal stimulation are virtually unchanged whereas those in the general cortex disappear, except their late components. Preceding single stimulation of the tectum or n. rotundus depresses responses in the hyperstriatum evoked by flashes. However, during stimulation of the lateral thalamic region, combined potentials and single unit responses appear in the hyperstriatum and interact with responses evoked by tectal stimulation. It is concluded that the main pathways in turtles which supply visual information to the general cortex and hyperstriatum differ: the former relay in the lateral thalamic region, the latter in n. rotundus, although some overlapping of their projections in the hyperstriatum and striatum is possible.I. Responses of 189 neurons of the somatosensory cortex to stimulation of the nonspecific reticular (R) and anteroventral (AV) nuclei of the thalamus were studied in cats anesthetized with thiopental and immobilized with tubocurarine. In the series of experiments with stimulation of R and, for comparison, of the specific ventral posterolateral nucleus (VPL), 132 neurons were recorded, of which 22 (16.7%) did not respond to stimulation of these nuclei, 77 (58.3%) responded only to stimulation of VPL, and 33 (25%) responded to stimulation of both VPL and R. In the series of experiments in which AV was stimulated, 57 neurons were recorded. Eight (14.8%) responded to neither stimulus and 25 (43.1%) responded only to stimulation of VPL; 24 responded to stimulation of AV (42.1%), and of these, 10 also responded to stimulation of VPL. A characteristic feature of unit responses in the somatosensory cortex to stimulation of the nonspecific nuclei was the irregularity of the responses and their longer latent period. Only five cells responded sooner to stimulation of the nonspecific nuclei than to stimulation of VPL. Responses of the nonspecific nuclei to stimulation appeared clearly only if the stimulation was repetitive. Neuronal responses of the dorsomedial thalamic nucleus of anesthetized rats to stimulation of the anterior periamygdalar cortex were compared before and after destruction of the basolateral nuclei of the ipsilateral amygdala. After destruction of the afferent inflow from the nuclei of the amygdala a new type of response appeared, in the form of prolonged generation of bursts of spikes, and also a significant decrease in the number of cells with a phasic activation response and an increase in the number of tonically activated neurons. Destruction of the amygdala was shown to lead to an increase in the number of cells of the dorsomedial nucleus with spontaneous activity. The duration of the tonic spike response in animals undergoing the operation was found to be significantly increased from 142.5±42.4 to 375.4±53.8 msec, whereas the parameters of other types of neuronal responses did not differ statistically significantly.Institute of Higher Nervous Activity and Neurophysiology, Academy of Sciences of the USSR, Moscow. Reactions of neurons of area 7 in the parietal association cortex to paired stimulation of the dorsolateral and posteriolateral thalamic nuclei were investigated using methods of extra- and intracellular recording. It was found that conditioning stimulation of association nuclei caused inhibition or facilitation of the test response; in some cases, the effect was indeterminant. Inhibition frequently led to other types of reactions. Inhibition of response to the test stimulus was pronounced during the initial period of IPSP, but facilitation of the response was possible during the repolarization phase. There was no inhibition of test responses when the chloride component of the IPSP was reversed. It is suggested that inhibition of responses to the test stimulus takes place in the presence of an inward chloride current and that facilitation of responses to such a stimulus may be due to increased effectiveness of late EPSPS.I. Structural and ultrastructural changes in the frontal areas of the cortex and in the region of the globus pallidus were investigated after local and extensive destruction of the caudate nucleus. It was shown by the Fink-Heimer method that after local injury to the caudate nucleus by means of electrodes implanted 2–16 months before electrolytic destruction, only a few degenerating fibers of medium and thin caliber were present. Extensive destruction of the caudate nucleus (without preimplantation of electrodes) was followed by massive degeneration of fibers of different caliber in the frontal area of the cortex. After local injury to the caudate nucleus numerous thin degenerating axons 0.5–0.6 µ in diameter and degenerating terminals were found in the region of the globus pallidus. Degenerative changes in the axo-dendritic and axo-somatic terminals followed the "dark" type of course. It is concluded that no considerable direct projections of neurons of the caudate nucleus are present in the cortex. Degenerating fibers of average caliber in frontal areas of the cortex after destruction of the caudate nucleus are evidently axons of thalamic neurons and not from cells of the damaged nucleus.A. A. Unit responses of the first (SI) somatosensory area of the cortex to stimulation of the second somatosensory area (SII), the ventral posterior thalamic nucleus, and the contralateral forelimb, and also unit responses in SII evoked by stimulation of SI, the ventral posterior thalamic nucleus, and the contralateral forelimb were investigated in experiments on cats immobilized with D-tubocurarine or Myo-Relaxin (succinylcholine). The results showed a substantially higher percentage of neurons in SII than in SI which responded to an afferent stimulus by excitation brought about through two or more synaptic relays in the cortex. In response to cortical stimulation antidromic and orthodromic responses appeared in SI and SII neurons, confirming the presence of two-way cortico-cortical connections. In both SI and SII intracellular recording revealed in most cases PSPs of similar character and intensity, evoked by stimulation of the cortex and nucleus in the same neuron. Latent periods of orthodromic spike responses to stimulation of nucleus and cortex in 50.5% of SI neurons and 37.1% of SII neurons differed by less than 1.0 msec. In 19.6% of SI and 41.4% of SII neurons the latent period of response to cortical stimulation was 1.6–4.7 msec shorter than the latent period of the response evoked in the same neuron by stimulation of the nucleus. It is concluded from these results that impulses from SI play an important role in the afferent activation of SII neurons.A. A. Thalamic afferent inputs of the motor cortex (area 4) were studied in cats by retrograde axonal transport of horseradish peroxidase (HRP). The main concentration of HRP-labeled neurons was found in rostral zones of the relay nuclei (of the ventrolateral and ventrobasal complex). A few labeled neurons were found in the mediodorsal association nucleus, where their distribution is quite local. HRP-labeled neurons of nonspecific intralaminar nuclei, projecting into the motor cortex, are present only in single numbers and show no tendency toward grouping in any parts of these nuclei. The results are evidence that the motor cortex receives heterogeneous afferentation from various thalamic nuclei, and it is evidently this which guarantees the reliability of transmission of incoming information.L. The reactions of 288 neurons of the orbitofrontal cortex (OFC) to stimulation of the posteroventral (VP), ventral anterior (VA), and reticular (R) nuclei, as well as the median center (CM) of the thalamus, were investigated in acute experiments on cats. OFC neurons can be divided into four groups by their reactions to stimulation of thalamic nuclei: 1) those which respond with an increase in the frequency of the discharges to single and serial stimuli with a frequency of up to 20/sec; 2) those which respond doubtfully to single stimuli with a frequency of 4–12/sec; 3) those which respond with inhibition of the background impulses; 4) those which do not respond to stimulation of the nuclei. Stimulation of the thalamic nuclei evoked responses of OFC neurons with a large scatter of the latent period duration. The responses of neurons to stimulation of the VP (mean latent period 19.1±6.1 msec) had the shortest latent period (sometimes less than 3–4 msec). Reactions with a longer latent period developed upon stimulation of the VA (23.8±7.4 msec) and CM (42.8±12.8 msec). The uniqueness of the links of the OFC with the various optic thalamic nuclei is shown in an analysis of the material obtained and possible methods of the activation of the neurons of this region from thalamic structures are discussed.State Medical Institute, Kemerovo. It was shown that the rabbit sensorimotor cortex received afferent fibers from neurons located in the specific, nonspecific, and association thalamic nuclei using the retrograde axonal transport technique. The distribution, dimensions, and shape of the somata of relay neurons spread through the thalamic nuclei were analyzed. The total number of neurons sending out thalamo-sensorimotor-cortical fibers was calculated and the coordinates of loci with the highest density of these cells in each thalamic nucleus were identified. Multipolar and stellate cells with somata measuring 12–20 µm and 10–15 µm in diameter, respectively, prevailed amongst relay neurons. Amongst the specific nuclei, the majority of afferent fibers are sent out by the ventrolateral, ventral anterior, and anterior ventral nuclei. A comparable number of afferent fibers are sent out by the mediodorsal and paracentral nuclei; these split up among the association nuclei and paracentral nuclei, respectively. A comparative analysis of monosynaptic afferent and efferent connections of callosal neurons and target neurons of transcallosal fibers with neurons of the specific ipsilateral thalamic nuclei (ventral posterolateral, ventral posteromedial, ventral lateral, and anteroventral) was undertaken on the sensomotor cortex of unanesthetized rabbits, using an electrophysiological method. Differences were demonstrated between callosal neurons and target neurons of transcallosal fibers with respect to monosynaptic inputs from the thalamic nuclei and pathways proceeding toward these structures and (or) entering the pyramidal tract. Among target neurons, compared with callosal neurons, more cells had descending projections (54 and 14%, respectively). Monosynaptic action potentials arose in 22% of target neurons in response to stimulation of specific thalamic nuclei, whereas no such responses occurred in callosal neurons. Projections of target neurons into thalamic nuclei were shown to be formed both by independent fibers and by axon collaterals of the pyramidal tract. It is postulated that the distinctive properties thus discovered indicate significantly greater convergence of influence of thalamic relay neurons on the target neurons; this determines differences known to exist in characteristics of receptive fields and spontaneous and evoked activity of callosal neurons, on the one hand, and of neurons excited synaptically by transcallosal stimulation, on the other hand.Institute of Higher Nervous Activity and Neurophysiology, Academy of Sciences of the USSR, Moscow. Responses of caudate neurons to electrical stimulation of the afferent input from thepulvinar thalamic nucleus and to visual stimuli of various orientations were studied extracellularly in awake chronic cats. Activation responses dominated among reactions of these neurons. The response latencies have ranged from 4 to 85 msec for units with primary activation and from 20 to 150 msec for inhibited ones. The values are indicative of both rapidly and slowly conducting afferent pathways. A possibility of monosynaptic transmission in thepulvinarcaudate projections is also revealed.Pulvinar stimulation is found to be efficient for a significant (more than 50 percent) number of caudate neurons responding to visual stimuli, including orientation-selective cells. The mode of influences from other structures of the visual system (optic tract, area 17, the Clare-Bishop area) on caudate neurons responding topulvinar stimulation is described. The data are discussed with respect to the possible role of cortical and subcortical projections of the visual system in the creation of sensory specific responses of the caudate nucleus.A. A. The dynamics of evoked potentials during blocking of cholinergic cortical structures was investigated in unanesthetized cats. Application of the anticholinergic drug benactyzine inhibits the negative phases of cortical responses to stimulation of the reticular formation and non-specific thalamic nuclei and also of responses to direct cortical stimulation. Direct cortical responses (DCRs), inverted by -aminobutyric acid, are also depressed, indicating the role of cholinergic mechanisms in the genesis of these responses. During blocking of cholinergic synapses, negative phases of the primary response (PR) and response to stimulation of the specific thalamic nucleus are facilitated. A tendency is then observed toward grouping of spontaneous unit discharges and abolition of inhibition of cortical neurons produced by high-frequency stimulation of the reticular formation. Extracellular and intracellular responses of 183 neurons in the primary projection area of the somatosensory cortex to electrical and tactile stimulation of the skin on the contralateral fore limb and to stimulation of the ventro-posterolateral thalamic nucleus of the ipsilateral hemisphere were studied in chronic experiments on cats. Spike responses to afferent stimuli are subdivided into three types: initial with a latent period of under 60 msec; initial followed by late responses with a latent period of over 60 msec; late with a latent period of over 60 msec. In addition another group of neurons responding to peripheral stimuli in the interval between the initial and the late response was identified. In nearly all cases the initial responses to peripheral stimulation had the form of a series of spikes, unlike responses to thalamic stimulation. It is concluded from the durations of the latent periods of these responses that about 70% of neurons in the primary projection area are activated mono- and disynaptically in response to peripheral stimulation; consequently, the intracortical spread of excitation in this zone is restricted.  相似文献
