[背景]海科贝特氏菌(Cobetia marina)可产生大量具有絮凝活性的胞外产物,可视为一种新型的生物絮凝剂。高温栅藻(Desmodesmus sp.F51)是一种具有较高叶黄素含量的微藻,被认为是一种新兴的叶黄素来源,但利用该生物絮凝剂高效富集高温栅藻的相关研究迄今尚未见报道。[目的]以高温栅藻为对象,研究该新型生物絮凝剂的絮凝效率,并对絮凝机理进行初步探讨。[方法]探索在不同生长阶段微藻培养液添加生物絮凝剂、添加量、絮凝时间、pH对絮凝效率的影响,分析生物絮凝剂的功能基团,并测定在不同pH条件下添加生物絮凝剂前后高温栅藻的Zeta电位变化,以及在显微镜下分析藻细胞在添加生物絮凝剂前后的形态。[结果]在高温栅藻生长至稳定期(pH 8.0)添加2 mL生物絮凝剂,絮凝15 min絮凝效果最佳,达82.1%。傅里叶红外光谱(fourier transform infrared spectroscopy,FTIR)显示了多糖及酰胺结构的特征吸收峰,由此推测生物絮凝剂主要是多糖的混合物,含有少量蛋白质。根据Bradford法测定絮凝剂中蛋白含量约为0.4%(质量比),通过苯酚-硫酸法测定总糖质量分数约为34.5%(质量比),与FTIR分析结果基本相符。生物絮凝剂在pH 4.0-11.0保持60%以上的絮凝效率,说明无论是酸性或是碱性条件下絮凝效率都较高,结合Zeta电位的分析表明,推测生物絮凝剂对高温栅藻的絮凝机理中占主导地位的可能是吸附架桥作用。[结论]该研究对微藻生物絮凝具有重要的理论和实践意义。  相似文献

Microalgae harvesting via pH induced flocculation along with utilization of recovered medium after flocculation is one of the most economical methods for separating the microalgal biomass in order to reduce the dewatering cost. In this study, optimization of marine and freshwater microalgae flocculation by pH adjustment was investigated via central composite design methodology. One molar of KOH and NaOH solutions were used to increase the pH level of the microalgal culture. Increasing pH value of the medium provided the highest flocculation efficiency up to 92.63 and 86.18% with pH adjusted to 10.5 with KOH and NaOH solutions for marine microalgae Nannochloropsis oculata and freshwater microalgae Chlorella minutissima, respectively. Also, it was revealed that microalgae cells were still alive after flocculation process and their biochemical composition was not changed, and flocculated medium can be used again for the next microalgal production. According to the results, it can be said that this method is cheap and effective, simple to operate and provides the utilization of flocculated medium again.  相似文献

Microalgae are being considered as a promising raw material for biofuel production. However, rapid, efficient, and economic technologies for harvesting microalgae are essential for successful applications. In this study, the high–pH-induced flocculation method was applied to harvest marine Chlorella sp. strains. These algae could be concentrated up to approximately 20-fold by increasing pH using NaOH, with a flocculation efficiency of 90 %. When NaOH dosage was low (1 or 3 mM), the flocculation efficiency decreased considerably with the increase of biomass concentration. At higher NaOH dosage tested (5 or 7 mM), flocculation occurred quickly and efficiently, which tended to be independent of biomass concentration. In larger volumes, all strains were flocculated with similar efficiencies (approximately 90 %) after adding 5 mM NaOH. After flocculation, the flocculated algae cells could be re-cultured as inoculum, and the growth yields in flocculated medium were slightly higher than those from fresh medium. Additionally, for each strain, there were no significant differences in lipid extraction yield and fatty acid composition according to different harvesting methods. These results showed that the high–pH-induced flocculation method could be used to harvest marine Chlorella sp. for biofuel production successfully.  相似文献

This study explored efficient methods of harvesting the Tetraselmis sp. KCTC12236BP using flocculation and dissolved air flotation. Concentration ranges of flocculation agents were optimized using jar tests (batch flocculation experiments) using inorganic (aluminum sulfate, ferric sulfate) and organic (chitosan) flocculants in a pH range of 4 ~ 10. The optimal dosage and pH level were 1.2 g/L and pH 5 ~ 6 for aluminum sulfate, 0.7 g/L and pH 4 ~ 8 for ferric sulfate, and 5.0 mg/mL and pH 7 ~ 8 for chitosan. The highest harvesting efficiency achieved with each of the four compounds was 85.6, 92.6, 93, and 91.3%, respectively.  相似文献

Many researchers consider efficient harvesting is the major challenge of commercialising micro-algal biofuel. Although micro-algal biomass can be ‘energy rich’, the growth of algae in dilute suspension at around 0.02–0.05 % dry solids poses considerable challenges in achieving a viable energy balance in micro-algal biofuel process operations. Additional challenges of micro-algae harvesting come from the small size of micro-algal cells, the similarity of density of the algal cells to the growth medium, the negative surface charge on the algae and the algal growth rates which require frequent harvesting compared to terrestrial plants. Algae can be harvested by a number of methods; sedimentation, flocculation, flotation, centrifugation and filtration or a combination of any of these. This paper reviews the various methods of harvesting and dewatering micro-algae for the production of biofuel. There appears to be no one method or combination of harvesting methods suited to all micro-algae and harvesting method will have a considerable influence on the design and operation of both upstream and downstream processes in an overall micro-algal biofuel production process.  相似文献

Microalgae are a promising new source of biomass; however, large-scale economical harvesting of microalgal biomass is a major technological and economic challenge, limiting the commercial production of microalgal biomass for high-value compounds. In this study, the cationic polymer chitosan was used for the harvesting of the marine diatom Chaetoceros muelleri. Natural flocculation, and pH and chitosan-induced flocculation were studied in detail. The effects of flocculant dosage, culture pH, initial biomass concentration, and sedimentation time were investigated on biomass recovery. The results showed that flocculation efficiency can reach > 99% with an optimum dosage of chitosan (80 mg L?1) at pH 9.6 and settling time of 40 minutes for biomass concentration from 0.2 to 1.2 g L?1. The reusability of the recycled water, preservation of biomass after harvesting, and cost of the harvesting process were evaluated. The results showed that the chitosan-induced flocculation offers an efficient, cost-effective, rapid, and sustainable harvesting method for C. muelleri biomass for food and feed applications in aquaculture.

Microbial flocculants for harvesting mass cultured Chlorella vulgaris were screened and that from Paenibacillussp. AM49 was identified as the best. The flocculation efficiency of this bioflocculant increased with the pH within a range of pH 5–11 and was 83%, which was higher than the 72% and 78% produced by aluminum sulfate and polyacrylamide, respectively. The highest flocculation efficiency was with 6.8 mm CaCl2 as co-flocculant. The bioflocculant from Paenibacillussp. AM49 can be used effectively to harvest C. vulgaris from large-scale cultures.  相似文献

A bioflocculant-producing bacterium isolated from soil was identified as Bacillus sp. and the bioflocculant produced was named MBFF19. Effects of physico-chemical conditions including pH, carbon sources and nitrogen sources on MBFF19 production were studied. Chemical analyses of the purified bioflocculant MBFF19 indicated that it was a sugar-protein derivative, composed of neutral sugar (3.6%, w/w), uronic acid (37.0%, w/w), amino sugars (0.5%, w/w) and protein (16.4%, w/w). The two neutral sugar components were mannose and glucose and the molar ratio was 1.2:1. Infrared spectrophotometry analysis revealed that MBFF19 contained carboxyl, hydroxyl and methoxyl groups in its structural. Flocculating properties of bioflocculant MBFF19 was examined using kaolin, activated carbon and fly coal suspension. Cation supplement had no positive effects on the flocculating activity whereas the presence of Fe3+ inhibited flocculation. Influences of pH and bioflocculant dosage on the flocculation were also examined.  相似文献

Biodiesel production from microalgae feedstock should be performed after growth and harvesting of the cells, and the most feasible method for harvesting and dewatering of microalgae is flocculation. Flocculation modeling can be used for evaluation and prediction of its performance under different affective parameters. However, the modeling of flocculation in microalgae is not simple and has not performed yet, under all experimental conditions, mostly due to different behaviors of microalgae cells during the process under different flocculation conditions. In the current study, the modeling of microalgae flocculation is studied with different neural network architectures. Microalgae species, Chlorella sp., was flocculated with ferric chloride under different conditions and then the experimental data modeled using artificial neural network. Neural network architectures of multilayer perceptron (MLP) and radial basis function architectures, failed to predict the targets successfully, though, modeling was effective with ensemble architecture of MLP networks. Comparison between the performances of the ensemble and each individual network explains the ability of the ensemble architecture in microalgae flocculation modeling.  相似文献

The optimal flocculating conditions for harvesting high-density cultures of Scenedesmus sp. were investigated using inorganic coagulants and the bioflocculant produced by Paenibacillus polymyxa AM49. The flocculated medium as nutrients for subsequent algal cultivation was also tested. Consecutive treatment with 8.5 mM CaCl(2) and 0.2 mM FeCl(3) as coagulants and 1% bioflocculant from the culture broth of P. polymyxa AM49 showed the highest flocculating activity of up to 95% for high density algal cultures. The medium flocculated with the coagulants and bioflocculant showed less than 8% decrease in the growth yield in the subsequent algal cultivation. Furthermore, a 20% or 50% fresh BG11 medium supplement allowed the flocculated medium to maintain a high growth yield in subsequent algal cultivation. These results suggest that the flocculation method presented here is efficient and bio-friendly, and allows the reuse of the flocculated medium, thereby contributing to the economic cultivation and harvest of microalgae.  相似文献

Flocculation is an essential step in the concentration and harvesting of microalgae from aquatic media. Salinity of brackish water and sea water requires high flocculant dosages and renders flocculation less effective than in freshwater algal media. Experiments with the marine microalgae Isochrysis galbana and Chlorella stigmatophora showed that effective alum or ferric chloride flocculation was obtained only with dosages which are 5 to 10 times higher than the dosages required for the flocculation of freshwater microalgae. The flocculant dosages required for removing over 90% of the algae from suspensions were found to increase linearly with salinity as expressed in ionic strength. High salinity was found to inhibit flocculation with polyelectrolytes which are quite effective in freshwater algae flocculation. This inhibition was diminished at reduced salinity levels and effective flocculation was attained at salinity levels of 5 g/liter and below, which is typical of desert brackish water. Two methods were found to induce flocculation in sea water: (a) combining polyelectrolytes with inorganic flocculants such as ferric chloride or alum, and (b) ozone oxidation pretreatment followed by flocculation with inorganic flocculants.  相似文献

Flocculation harvesting of the fucoxanthin-rich marine microalga Isochrysis galbana has received little attention. Therefore, we attempted to screen for an optimal chemical flocculant and optimize flocculation conditions from five chemical flocculants—ferric chloride (FC), aluminum sulfate (AS), polyaluminum chloride (PAC), aluminum potassium sulfate (APS), and zinc sulfate (ZS)—for effective flocculation of I. galbana. The growth rate, photosynthetic performance, and fucoxanthin content were determined in re-suspended flocculated algal cells and in the flocculation supernatant cultured algal cells. The results showed that high growth rate and fucoxanthin accumulation were observed when FC was used as the flocculant in I. galbana cultures, which indicated that FC may cause less harm to I. galbana than the other aluminum-based flocculants. Furthermore, satisfactory flocculation efficiency was also observed when FC was used to flocculate I. galbana, and the FC dosage was less than that required for flocculation of I. galbana using PAC, APS, and AS. Thus, we selected FC as the optimal flocculant for harvesting I. galbana based on its flocculation efficiency together with algal physiological performance, growth rate, and fucoxanthin content.  相似文献

This research assessed the efficacy of three harvesting methods on a strain of Dunaliella viridis. While there is strong potential to use lipids from microalgae as a feedstock for biofuels to replace petroleum-based fuel, at present microalgal harvesting for biofuel production is not yet economically feasible or energy efficient. pH-induced flocculation (by adjusting the pH of exponentially growing cells), indirect electrocoagulation (applying aluminum hydroxide coagulant to culture), and hollow fiber filtration (separating biomass from medium using tangential flow) were compared as potential harvesting mechanisms for small-scale (3–10 L) and large-scale (30–150 L) volumes of D. viridis. Both pH-induced flocculation and electrocoagulation yielded significant biomass recovery (>95 %), but both methods required removal of added chemicals and/or coagulant before the medium could be reused. In contrast, hollow fiber filtration did not require added chemicals or coagulant, and as another advantage, the filtrate was successfully reused as culture medium without apparent detrimental effects on cell size, cell shape, or cell production. When high salinity stress was imposed on the concentrate produced from the filtration method, total fatty acids (FAs) did not increase. However, total FAs did significantly increase after hollow fiber filtration (49 %) in comparison to FA content before filtration (36 %). This research indicates that hollow fiber filtration as a commercial harvesting mechanism offers attractive advantages as a harvesting mechanism for microalgae such as Dunaliella, relative to pH-induced flocculation and indirect electrocoagulation.  相似文献

The purpose of this study was to explore efficient methods of harvesting the microalga Phaeodactylum tricornutum. Natural sedimentation experiments, performed at different light and temperature conditions, did not yield significant improvements in efficiency even after 1?week. When alkalinity-induced flocculation was performed, both the flocculation efficiency and the concentration factor dramatically improved at pH?=?9.75 (0.5–0.7 units over the original pH of the culture) after 10?min settling time. Sedimentation rates are documented at pH ranging between pH?9.75 and 11.0. The results of the application of two conventional flocculants used in wastewater treatment, polyaluminium chloride and aluminium sulphate, are also presented. Chitosan was also used as a natural flocculating agent to improve possible contamination problems in the downstream process. pH was adjusted in order to determine optimum flocculation efficiency of chitosan in combination with a high concentration factor. Satisfactory results were found with chitosan at an adjusted pH of 9.9 using concentrations as low as 20?mg?L?1, after testing a flocculant range of 5–200?mg?L?1.  相似文献

Several protocols for isolation of mycobacteria from water exist, but there is no established standard method. This study compared methods of processing potable water samples for the isolation of Mycobacterium avium and Mycobacterium intracellulare using spiked sterilized water and tap water decontaminated using 0.005% cetylpyridinium chloride (CPC). Samples were concentrated by centrifugation or filtration and inoculated onto Middlebrook 7H10 and 7H11 plates and Lowenstein-Jensen slants and into mycobacterial growth indicator tubes with or without polymyxin, azlocillin, nalidixic acid, trimethoprim, and amphotericin B. The solid media were incubated at 32 degrees C, at 35 degrees C, and at 35 degrees C with CO(2) and read weekly. The results suggest that filtration of water for the isolation of mycobacteria is a more sensitive method for concentration than centrifugation. The addition of sodium thiosulfate may not be necessary and may reduce the yield. Middlebrook M7H10 and 7H11 were equally sensitive culture media. CPC decontamination, while effective for reducing growth of contaminants, also significantly reduces mycobacterial numbers. There was no difference at 3 weeks between the different incubation temperatures.  相似文献

Addition of starch to suspensions of Escherichia coli K-12 resulted in the formation of bacterial flocs. The flocculation was dependent on the high expression of a receptor for starch (maltoporin) on the surface of the bacterium. Factors influencing floc formation were investigated and optimal conditions for flocculation based on cell density, starch concentration, time, and pH established. As quantitated by a sedimentation assay, over 80% of bacteria in a culture could be removed by settling without centrifugation in 3 h under optimal conditions. Floc formation was evident with bacteria containing wild-type maltoporin but was faster and occurred to a greater extent with strains expressing a high-affinity allele (lamB1400) of the starch receptor. Bacteria could be harvested by floc formation directly in growth medium under defined conditions of maltoporin expression and medium composition. These results demonstrate the effectiveness of starch-dependent aggregation in the harvesting of cells, using an inexpensive, biologically acceptable agent to induce flocculation.  相似文献

Gas vesicles are hollow, proteinaceous structures found in some strains of cyanobacteria. They have been used to increase the oxygen supply and improve the cultivation of shear-sensitive mammalian cells. However, the production and, especially, collection of cells and gas vesicles were laborious and ineffective. In this study we examined the use of the cationic polymer, polyethyleneimine (PEI), for improving the cell harvesting by flocculation and flotation. PEI was examined to determine the appropriate molecular weight, pH range, and dosage. The dose of 20–30 mg/l of PEI with molecular weight of 25,000 in the pH range of 6.0–8.5 was found to provide effective and efficient cell flocculation and flotation. As the PEI dose increased, the rate of flotation increased but the clearance (collection) efficacy declined slightly. The culture samples used in this study were taken from light-limiting continuous culture systems at different dilution rates (0.05–0.24 h−1). Without PEI addition, the cells at dilution rates lower than 0.1 h did not float while those at higher dilution rates would float slowly. With PEI addition, the flocculated cells at the dilution rate of 0.05 h−1 sank and those of higher dilution rates would float and the flotation rate increased with increasing specific growth rate. Nonetheless, PEI flocculation and flotation (or sedimentation) could be used to harvest cells at a wide range of growth states.  相似文献

An antimicrobial agent is produced by the cyanobacterium Synechococcus leopoliensis which was found to be active against the Gram-positive bacterium Staphylococcus aureus. The effects of temperature, pH, incubation period, some media and different nitrogen and carbon sources on both growth and antimicrobial activity were investigated. Temperature 35 °C and pH 8 were the best for growth and antimicrobial agent production and 14 and 15 days of incubation were found to be the best for maximum growth and antimicrobial activity, respectively, in the medium BG-11.

No antimicrobial activity could be detected by the use of G medium, moderate activity was recorded with Chu 10 medium, while high activity was reported in BG-11 medium. Leucine was the best nitrogen source for antimicrobial activity, while maximum antimicrobial activity was introduced by using the carbon sources, citrate and acetate. Very high antimicrobial activity could be detected by using the carbon source galactose in combination with the nitrogen source alanine or by using arabinose with methionine.  相似文献

AIM: To investigate an effective harvesting method for Spirulina platensis. METHODS AND RESULTS: Eighty per cent of S. platensis cells in the logarithmic growth phase were harvested by flotation when the cells were set in a static condition for 2 h. The optimum harvesting time was about day 6 of cultivation. The flotation activity of S. platensis cells was enhanced by the addition of NaCl. CONCLUSIONS: The harvesting of S. platensis by flotation is a cost-effective and straightforward method that can retain the algal quality. The optimum harvesting time of S. platensis can be predicted by the cellular protein to carbon ratio. SIGNIFICANCE AND IMPACT OF THE STUDY: Flotation harvesting is also applicable to other cyanobacteria with gas vesicles.  相似文献
