The heavier analogs of C2H2 have been studied at the B3LYP level for their μ and μ42 coordination properties with the transition metals. Based on known alkyne compounds, transition metal fragments [W2(μ-NH)(Cp)2(Cl)2] and [Fe4(CO)12] have been chosen. The SBKJC relativistic effective core potentials and their associated basis sets were used on W, Fe, Sn and Pb, and the 6-31G(d) basis set was used on all other elements. All the complexes of Si2H2, Ge2H2, Sn2H2 and Pb2H2 are found to be local minima. The trans-twist nature of the ligand A2H2 (A = Si-Pb) is large in μ-coordinated complexes of W, and it is very small in μ42 coordinated complexes of Fe. The electronic structure of these complexes was investigated using fragment molecular orbital method (FMO).  相似文献

The potential for C4 photosynthesis was investigated in five C3-C4 intermediate species, one C3 species, and one C4 species in the genus Flaveria, using 14CO2 pulse-12CO2 chase techniques and quantum-yield measurements. All five intermediate species were capable of incorporating 14CO2 into the C4 acids malate and aspartate, following an 8-s pulse. The proportion of 14C label in these C4 products ranged from 50–55% to 20–26% in the C3-C4 intermediates F. floridana Johnston and F. linearis Lag. respectively. All of the intermediate species incorporated as much, or more, 14CO2 into aspartate as into malate. Generally, about 5–15% of the initial label in these species appeared as other organic acids. There was variation in the capacity for C4 photosynthesis among the intermediate species based on the apparent rate of conversion of 14C label from the C4 cycle to the C3 cycle. In intermediate species such as F. pubescens Rydb., F. ramosissima Klatt., and F. floridana we observed a substantial decrease in label of C4-cycle products and an increase in percentage label in C3-cycle products during chase periods with 12CO2, although the rate of change was slower than in the C4 species, F. palmeri. In these C3-C4 intermediates both sucrose and fumarate were predominant products after a 20-min chase period. In the C3-C4 intermediates, F. anomala Robinson and f. linearis we observed no significant decrease in the label of C4-cycle products during a 3-min chase period and a slow turnover during a 20-min chase, indicating a lower level of functional integration between the C4 and C3 cycles in these species, relative to the other intermediates. Although F. cronquistii Powell was previously identified as a C3 species, 7–18% of the initial label was in malate+aspartate. However, only 40–50% of this label was in the C-4 position, indicating C4-acid formation as secondary products of photosynthesis in F. cronquistii. In 21% O2, the absorbed quantum yields for CO2 uptake (in mol CO2·[mol quanta]-1) averaged 0.053 in F. cronquistii (C3), 0.051 in F. trinervia (Spreng.) Mohr (C4), 0.052 in F. ramosissima (C3-C4), 0.051 in F. anomala (C3-C4), 0.050 in F. linearis (C3-C4), 0.046 in F. floridana (C3-C4), and 0.044 in F. pubescens (C3-C4). In 2% O2 an enhancement of the quantum yield was observed in all of the C3-C4 intermediate species, ranging from 21% in F. ramosissima to 43% in F. pubescens. In all intermediates the quantum yields in 2% O2 were intermediate in value to the C3 and C4 species, indicating a co-function of the C3 and C4 cycles in CO2 assimilation. The low quantum-yield values for F. pubescens and F. floridana in 21% O2 presumably reflect an ineffcient transfer of carbon from the C4 to the C3 cycle. The response of the quantum yield to four increasing O2 concentrations (2–35%) showed lower levels of O2 inhibition in the C3-C4 intermediate F. ramosissima, relative to the C3 species. This indicates that the co-function of the C3 and C4 cycles in this intermediate species leads to an increased CO2 concentration at the site of ribulose-1,5-bisphosphate carboxylase/oxygenase and a concomitant decrease in the competitive inhibition by O2.Abbreviations PEP phosphoenolpyruvate - PGA 3-phosphoglycerate - RuBP ribulose-1,5-bisphosphate  相似文献

[3H]gibberellin A9 was applied to shoots or seed parts of G2 pea to produce radiolabeled metabolites. These were used as markers during purification for the recovery of endogenous GA9 and its naturally occurring metabolites. GA9 and its metabolites were purified by HPLC, derivatized and examined by GC-MS. Endogenous GA9, GA20, GA29 and GA51 were identified in pea shoots and seed coats. GA51-catabolite and GA29-catabolite were also detected in seed coats. GA70 was detected in seed coats following the application of 1 g of GA9. Applied [3H]GA9 was metabolized through both the 13-hydroxylation and 2-hydroxylation pathways. Labeled metabolites were tentatively identified on the basis of co-chromatography on HPLC with endogenous compounds identified by GC-MS. In shoots [3H]GA51 and [3H]GA51-catabolite were the predominant metabolites after 6 hrs, but by 24 hrs there was little of these metabolites remaining, while [3H]GA29-catabolite and an unidentified metabolite predominated. In seed coats [3H]GA51 was the initial product, later followed by [3H]GA51-catabolite and an unidentified metabolite (different from that in shoots), with lesser amounts of [3H]GA20, [3H]GA29 and [3H]GA29-catabolite. Large quantities of African dust are carried into the southeastern United States each summer with concentrations typically in the range of 10 to 100?µg m?3. Because approximately one-third to one-half of the dust mass is in the size range under 2.5?µm diameter, the advection of African dust has implications for the EPA's newly implemented standard for PM2.5 particulate matter and for the assessment of human health effects. It will be difficult to assess the impact of African dust events on air quality because they occur during the summer (maximum in July) when photochemical pollution events are frequent and intense in the eastern United States. Indeed, the presence of dust could affect the evolution of photochemical dust events. In order to assess the role of African dust in air quality in the United States, it will be necessary to develop a set of diagnostic indicators; it appears that dust mineralogy and elemental composition might be useful for this purpose. Various satellite products can be used to characterize the spatial coverage of dust events and, when coupled with meteorological tools, to predict impact regions.  相似文献

Species in the Laxa and Grandia groups of the genus Panicum are adapted to low, wet areas of tropical and subtropical America. Panicum milioides is a species with C3 photosynthesis and low apparent photorespiration and has been classified as a C3/C4 intermediate. Other species in the Laxa group are C3 with normal photorespiration. Panicum prionitis is a C4 species in the Grandia group. Since P. milioides has some leaf characteristics intermediate to C3 and C4 species, its photosynthetic response to irradiance and temperature was compared to the closely related C3 species, P. laxum and P. boliviense and to P. prionitis. The response of apparent photosynthesis to irradiance and temperature was similar to that of P. laxum and P. boliviense, with saturation at a photosynthetic photo flux density of about 1 mmol m-2 s-1 at 30°C and temperature optimum near 30°C. In contrast, P. prionitis showed no light saturation up to 2 mmol m-2 s-1 and an optimum temperature near 40°C. P. milioides exhibited low CO2 loss into CO2-free air in the light and this loss was nearly insensitive to temperature. Loss of CO2 in the light in the C3 species, P. laxum and P. boliviense, was several-fold higher than in P. milioides and increased 2- to 5-fold with increases in temperature from 10 to 40°C. The level of dark respiration and its response to temperature were similar in all four Panicum species examined. A biocatalatic pathway involving chromogenic probe has been proposed for the determination of catalase activity by means of iso-nicotinicacidhydrazide (INH) and pyrocatechol (PC). The assay is based on the enzymatic consumption of hydrogen peroxide using INH-PC system. The response of the catalase activity was ascertained by the rate of the reaction involving 14.10 mM H2O2. On addition of H2O2, INH-PC indicator system formed a chromogenic product with absorbance maxima at 490 nm. Hence the activity of catalase was directly measured by the chromogenic response in the formation of the coupled product. The catalase assay was elaborated by the kinetic response of the INH-PC system. The linearity of the catalase activity and H2O2 was in the range 0.2-7.0 units and 1.76-7.0 mM, respectively in 3 ml solution. The catalytic efficiency and catalytic power were calculated. The Michaelis-Menten constant of INH, PC and H2O2 were found to be 0.344, 0.176 and 8.82 mM, respectively. The indicator reaction was applied in the determination of catalase activity in mycelia mats and culture media.  相似文献

Possibilities of the linear-polarized infrared (IR-LD) spectroscopy of oriented colloid suspensions in nematic liquid crystals, for structural and local structural elucidation for first time are demonstrated of inorganic compounds and glasses. The advantages of the method for tellurite and borate glasses are shown. The IR-band assignment of the typical local structural units in the glasses are proposed by a comparison with the IR-characteristics of appropriate crystalline analogues as α-TeO2, V2O5, MoO3 · H2O and its high temperature form. The IR-spectroscopic characteristics of BO3, BO4 and boroxol ring are elucidated, using crystalline β-BaB2O4, SrB4O7, H3BO3 and B2O3 as model systems, where the structural moieties have been refined by single crystal X-ray diffraction.  相似文献

Integrins not only mediate cell adhesion but also contribute to a variety of other cellular processes including proliferation, cytokine production, cytotoxicity and apoptosis. They act as bi-directional signal transducers, mediating signaling from inside-to-outside the cell and from outside-to-inside the cell. Evidence is emerging that signaling through leukocyte integrins (β2 and β7) is distinct from signaling by the more widely distributed β1 integrins. Here we discuss the role of the cytoplasmic domains of leukocyte integrins and that of cytosolic proteins that bind integrins in mediating signal transduction. Distinct sites in the alpha as well as the common beta chain contribute to this process. We also show that β2 integrin distribution on the cell surface is of particular relevance for leukocytes to rapidly alter their adhesive state and display their highly dynamic adhesive behavior. From these and recently published findings the picture is arising that particular cell functions may be supported by integrin-specific signaling pathways.  相似文献

Chloroplasts with high rates of photosynthetic O2 evolution (up to 120 mol O2· (mg Chl)-1·h-1 compared with 130 mol O2· (mg Chl)-1·h-1 of whole cells) were isolated from Chlamydomonas reinhardtii cells grown in high and low CO2 concentrations using autolysine-digitonin treatment. At 25° C and pH=7.8, no O2 uptake could be observed in the dark by high- and low-CO2 adapted chloroplasts. Light saturation of photosynthetic net oxygen evolution was reached at 800 mol photons·m-2·s-1 for high- and low-CO2 adapted chloroplasts, a value which was almost identical to that observed for whole cells. Dissolved inorganic carbon (DIC) saturation of photosynthesis was reached between 200–300 M for low-CO2 adapted chloroplasts, whereas high-CO2 adapted chloroplasts were not saturated even at 700 M DIC. The concentrations of DIC required to reach half-saturated rates of net O2 evolution (Km(DIC)) was 31.1 and 156 M DIC for low- and high-CO2 adapted chloroplasts, respectively. Mitochondria are central to heart function and dysfunction, and the pathways activated by different cardioprotective interventions mostly converge on mitochondria. In a context of perspectives in innate and acquired cardioprotection, we review some recent advances in F0F1ATPsynthase structure/function and regulation in cardiac cells. We focus on three topics regarding the mitochondrial F0F1ATPsynthase and the plasma membrane enzyme, i.e.: i) the crucial role of cardiac mitochondrial F0F1ATPsynthase regulation by the inhibitory protein IF1 in heart preconditioning strategies; ii) the structure and function of mitochondrial F0F1ATPsynthase oligomers in mammalian myocardium as possible endogenous factors of mitochondria resistance to ischemic insult; iii) the external location and characterization of plasma membrane F0F1 ATP synthase in search for possible actors of its regulation, such as IF1 and calmodulin, at cell surface.  相似文献

Cations such as Mg2+ regulate spillover of absorbed excitation energy mainly in favour of photosystem (PS) 2. Effect of low concentration (<10 mM) of the monovalent cation Na+ on chlorophyll (Chl) a fluorescence was completely overridden by divalent cation Mg2+ (5 mM). Based on Chl a fluorescence yield and 77 K emission measurements, we revealed the role and effectiveness of anions (Cl-, SO4 2-, PO4 3-) in lowering the Mg2+-induced PS2 fluorescence. The higher the valency of the anion, the lesser was the expression of Mg2+ effect. Anions may thus overcome Mg2+ effects up to certain extent in a valency dependent manner, thereby diverting more energy to PS1 even in the presence of MgCl2. The effect of inorganic phosphate (Pi) on uni-site ATP binding and hydrolysis by the nucleotide-depleted F1-ATPase from beef heart mitochondria (ndMF1) has been investigated. It is shown for the first time that Pi decreases the apparent rate constant of uni-site ATP binding by ndMF1 3-fold with the Kd of 0.38 ± 0.14 mM. During uni-site ATP hydrolysis, Pi also shifts equilibrium between bound ATP and ADP + Pi in the direction of ATP synthesis with the Kd of 0.17 ± 0.03 mM. However, 10 mM Pi does not significantly affect ATP binding during multi-site catalysis.  相似文献

Modulatory effects of auxiliary alpha(2)delta(2) and gamma(5) subunits on intramembrane charge movement originating from the expressed Ca(v)3.1 calcium channel were investigated. Inward current was blocked by 1mM La(3+). Voltage dependences of Q(on) and Q(off), kinetics of ON- and OFF-charge movement, and I(max)/Q(max) ratio were measured in the absence and the presence of an auxiliary subunit. The alpha(2)delta(2) subunit accelerated significantly both ON- and OFF-charge movement. I(max)/Q(max) ratio and Q(on)-V, Q(off)-V relations were not affected. Coexpression of the alpha(2)delta(2) subunit may accelerate channel transitions between individual closed states, but not the transition from the last closed channel state into an open state. Coexpression of the gamma(5) subunit accelerated the decay of the ON-charge transient and enhanced I(max)/Q(max) ratio. These effects suggest improvement of the coupling between the charge movement and the channel opening due to facilitation of transitions between individual closed states and the transition between the last closed state and an open state.  相似文献

Human non-pancreatic secretory phospholipase A2 (hnpsPLA2) is a group IIA phospholipase A2 which plays an important role in the innate immune response. This enzyme was found to exhibit bactericidal activity toward Gram-positive bacteria, but not Gram-negative ones. Though native hnpsPLA2 is active over a broad pH range, it is only highly active at alkaline conditions with the optimum activity pH of about 8.5. In order to make it highly active at neutral pH, we have obtained two hnpsPLA2 mutants, Glu89Lys and Arg100Glu that work better at neutral pH in a previous study. In the present study, we tested the bactericidal effects of the native hnpsPLA2 and the two mutants. Both native hnpsPLA2 and the two mutants exhibit bactericidal activity toward Gram-positive bacteria. Furthermore, they can also kill Escherichia coli, a Gram-negative bacterium. The two mutants showed better bactericidal activity for E. coli at neutral pH than the native enzyme, which is consistent with the enzyme activities. As hnpsPLA2 is highly stable and biocompatible, it may provide a promising therapy for bacteria infection treatment or other bactericidal applications.  相似文献

Gibberellin A4 (GA4) was identified for the first time in the garden pea (Pisum sativum) L.), by gas chromatography-mass spectrometry. However, in wild-type shoots the level of GA4 was only about 6% of the level of GA1, and it is therefore unlikely that GA4 plays a major role per se in the control of pea stem elongation. In shoots of the le mutant, GA4 was not detected, while the level of GA9 was approximately twice that found in the wild-type. The le mutation also markedly reduced the elongation response to applied GA9. It appears, therefore, that in Pisum the le mutation blocks the 3-hydroxylation of GA9 to GA4, in addition to the 3-hydroxylation of GA20 to GA1. In contrast, the le mutation did not reduce the response to applied GA5, suggesting the step GA5 to GA3 is not catalysed by the enzyme controlled by the Le gene. The step GA5 to GA3 was confirmed in peas by metabolite analysis after treatment with deuterated GA5.  相似文献

The vacuolar ATPases (V-type ATPases) are a family of ATP-dependent ion pumps and found in two principal locations, in endomembranes and in plasma membranes. This family of ATPases is responsible for acidification of intracellulare compartments and, in certain cases, ion transport across the plasma membrane of eucaryotic cells. V-ATPases are composed of two distinct domains: a catalytic V1 sector, in which ATP hydrolysis takes place, and the membrane-embedded sector, V0, which functions in ion conduction. In the past decade impressive progress has been made in elucidating the properties structure, function and moleculare biology. These knowledge sheds light also on the evolution of V-ATPases and their related families of A-(A1A0-ATPase) and F-type (F1F0-ATPases)ATPases.  相似文献

The human multidrug resistance-associated protein 1 (hMRP1/ABCC1) belongs to the ATP-binding cassette transporter superfamily. Together with P-glycoprotein (ABCB1) and the breast cancer resistance protein (BCRP/ABCG2), hMRP1 confers resistance to a large number of structurally diverse drugs. The current topological model of hMRP1 includes two cytosolic nucleotide-binding domains and 17 putative transmembrane (TM) helices forming three membrane-spanning domains. Mutagenesis and labeling studies have shown TM16 and TM17 to be important for function. We characterized the insertion of the TM16 fragment into dodecylphosphocholine (DPC) or n-dodecyl-β-d-maltoside (DM) micelles as membrane mimics and extended our previous work on TM17 (Vincent et al., 2007, Biochim. Biophys. Acta 1768, 538). We synthesized TM16 and TM17, with the Trp residues, W1198 in TM16 and W1246 in TM17, acting as an intrinsic fluorescent probe, and TM16 and TM17 Trp variants, to probe different positions in the peptide sequence. We assessed the interaction of peptides with membrane mimics by evaluating the increase in fluorescence intensity resulting from such interactions. In all micelle-bound peptides, the tryptophan residue appeared to be located, on average, in the head group micelle region, as shown by its fluorescence spectrum. Each tryptophan residue was partially accessible to both acrylamide and the brominated acyl chains of two DM analogs, as shown by fluorescence quenching. Tryptophan fluorescence lifetimes were found to depend on the position of the tryptophan residue in the various peptides, probably reflecting differences in local structures. Far UV CD spectra showed that TM16 contained significant β-strand structures. Together with the high Trp correlation times, the presence of these structures suggests that TM16 self-association may occur at the interface. In conclusion, this experimental study suggests an interfacial location for both TM16 and TM17 in membrane mimics. In terms of overall hMRP1 structure, the experimentally demonstrated amphipathic properties of these TM are consistent with a role in the lining of an at least partly hydrophilic transport pore, as suggested by the currently accepted structural model, the final structure being modified by interaction with other TM helices.  相似文献

Procedures are described for the isolation of the individual components A1, A2, and A3 of native R-ovalbumin from freshly laid domestic hen eggs. Because heavy metal ion contaminants result in spurious irreproducible kinetics, particularly at high pH, considerable care is taken to avoid their presence. Kinetics studies are made of the behavior of whole R-ovalbumin and its individual components in urea solution over the pH range 3.7–9.6 following the reaction by determining absorbance differences at 233, 287, and 293 nm and ORD and CD changes at 350 and 221 nm, respectively. Reaction is rapid at low pH, slowing with increasing pH. Except under limited conditions, the reaction is not simple first order. Equations are presented for describing the reactions, and the nature of the reaction products is considered. Unfolding equilibrium profiles were also determined by ORD at several wavelengths and were not stigmoidal in shape and the normalized curves were not superimposed.Deceased December 8, 2001  相似文献
