A comparative study was carried out to determine some valuable phytochemical components, macro- and microelement and redox parameters in leaves of male and female Ginkgo biloba trees and in extracts made from them. G. biloba extracts have become more popular as a therapeutic agent in the modern pharmacology in neurodegenerative diseases, in which increased brain metal levels can be observed and free radical reactions are involved. Macro- and microelement components, total phenol content, H-donating activity and reducing power as well as total scavenger capacity were determined in the samples. Well detectable differences were obtained for micro- and macroelement contents between male and female samples, but no toxic elements could be detected in the extracts. Male extracts contained more hazardous metals (e.g. Fe) compared to the female ones, while extracts from female leaves had higher levels of ions, which are known to have beneficial effects in neurodegenerative diseases. The ethanolic extracts of male leaves showed the highest H-donating activity, reducing power and total phenol content, as well as the best total scavenger activity. Ginkgo extracts due to the antioxidant properties may have favourable effects as dietary supplements in several neurodegenerative diseases, but this study draws the attention that critical evaluation is required in view of the potential hazard induced by their metal ion constitution. Our results lead us to the conclusion that although the aqueous extracts of female leaves are characterized by relatively lower antioxidant properties, they may be more eligible for these purposes due to their favourable metal ion constitution.  相似文献

Minocycline is neuroprotective in animal models of a number of acute CNS injuries and neurodegenerative diseases. While anti-inflammatory and anti-apoptotic effects of minocycline have been characterized, the molecular basis for the neuroprotective effects of minocycline remains unclear. We report here that minocycline and a number of antioxidant compounds protect mixed neuronal cultures in an oxidative stress assay. To evaluate the role of minocycline's direct antioxidant properties in neuroprotection, we determined potencies for minocycline, other tetracycline antibiotics, and reference antioxidant compounds using a panel of in vitro radical scavenging assays. Data from in vitro rat brain homogenate lipid peroxidation and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assays show that minocycline, in contrast to tetracycline, is an effective antioxidant with radical scavenging potency similar to vitamin E. Our findings suggest that the direct antioxidant activity of minocycline may contribute to its neuroprotective effects in some cell-based assays and animal models of neuronal injury.  相似文献

The antioxidant activities of extracts from leaves of the medicinal plants growing in Siberia were examined. Total antioxidant activity was determined using in vitro methods including DPPH (2,2-diphenyl-1-picrylhydrazyl radical) free radical scavenging assay, chelating capacity assay with ferrozine, evaluation of capacity to protect plasmid DNA against oxidative damage, measurement of H2O2 production, and measurement of total flavonoid and tannin content as well. Using in vivo experiments, we also evaluated capacities of the plant extracts to protect bacteria Escherichia coli against bacteriostatic and bactericidal effects of H2O2, and influence of the plant extracts on expression of antioxidant gene katG, encoding catalase. The extracts from Chamerion angustifolium, Filipendula vulgaris and Pyrola rotundifolia indicated the highest levels of antioxidant activity both in vivo and in vitro. Our data suggest that the extracts of the tested plants may provide antioxidant effects on bacteria simultaneously through different pathways, including direct radical scavenging, iron chelation and induction of genes encoding antioxidant enzymes.  相似文献

Thermopsis turcica, endemic to Turkey, is in danger of extinction. Studies on this species are very few due to the fact that it was only discovered in 1983 and grows in a small circumscribed area in Turkey. In this study, free radical scavenging activity, total phenolic content, total oxidant status (TOS), and total antioxidant status (TAS) of methanol (TTM) and acetone (TTA) extracts of T. turcica were measured spectroscopically. Free radical scavenging activity was determined according to the elimination of DPPH radicals and total phenol content was determined by the Folin–Ciocalteu reaction. Total oxidant status (TOS) and total antioxidant status (TAS) were measured with commercially available kits. Methanol and acetone extracts of T. turcica were found to have a specific radical scavenging effect. This effect was found to be related to the total phenolic content of the extracts. Since the TTA had a higher phenolic content than the methanol extract, it had a stronger radical scavenging effect. In addition, the total antioxidant capacity of the methanol extract was observed to be higher than that of its acetone counterpart. As a result, due to its antioxidative properties, T. turcica is thought to be a natural source of antioxidants.  相似文献

This study evaluated the antioxidant potential of extracts from Cornus walteri W. (CW) using various assays for natural antioxidants. We determined the polyphenol and flavonoid contents, as well as the antioxidant properties of water and ethanol extracts from the CW compared with those of other natural and synthetic antioxidants. CW extracts had high total phenolic and flavonoid contents in both the water and ethanol extracts. Various radical (DPPH, hydroxyl, and alkyl) scavenging activities of extracts from CW were higher than that of vitamin C. In addition, the antioxidant capacity measured as ferric reducing antioxidant power (FRAP), and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) as diammonium salt (ABTS) radical scavenging were higher than that of 2,6-di-tert-butyl-4-methylphenol (BHT), used as a positive control. The cytoprotective effect of CW extracts was also observed in tert-butyl hydroperoxide (t-BHP)-induced oxidative damage in Chang cells. These results showed that CW extracts have antioxidant properties through their ability to enhance cell viability, prevent reactive oxygen species (ROS) generation, and inhibit mitochondrial membrane depolarization. The antioxidant potency of the CW extracts could be exploited for their health promoting potential.  相似文献

Among health-promoting phytochemicals in whole grains, phenolic compounds have gained attention as they have strong antioxidant properties and can protect against many degenerative diseases. Aim of this study was to profile grain phenolic extracts of one modern and five old common wheat (Triticum aestivum L.) varieties and to evaluate their potential antiproliferative or cytoprotective effect in different cell culture systems.Wheat extracts were characterized in terms of antioxidant activity and phenolic composition (HPLC/ESI-TOF-MS profile, polyphenol and flavonoid contents). Results showed that antioxidant activity (FRAP and DPPH) is mostly influenced by flavonoid (both bound and free) content and by the ratio flavonoids/polyphenols. Using a leukemic cell line, HL60, and primary cultures of neonatal rat cardiomyocytes, the potential antiproliferative or cytoprotective effects of different wheat genotypes were evaluated in terms of intracellular reactive oxygen species levels and cell viability. All tested wheat phenolic extracts exerted dose-dependent cytoprotective and antiproliferative effects on cardiomyocytes and HL60 cells, respectively. Due to the peculiar phenolic pattern of each wheat variety, a significant genotype effect was highlighted. On the whole, the most relevant scavenging effect was found for the old variety Verna. No significant differences in terms of anti-proliferative activities among wheat genotypes was observed. Results reported in this study evidenced a correspondence between the in vitro antioxidant activity and potential healthy properties of different extracts. This suggests that an increased intake of wheat grain derived products could represent an effective strategy to achieve both chemoprevention and protection against oxidative stress related diseases.  相似文献

Fermented food is a rich source of antioxidants and micronutrients with the potential to prevent various human diseases. The increasing evidence indicates that in addition to its direct action, radical-scavenging antioxidants may modulate the cellular antioxidant system such as glutathione. In the present study, we investigated the antioxidant activity of Antioxidant Biofactor (AOB) extracts, a mixture of commercially available fermented grain food by using chemical and cellular experimental systems. In the former system, the total radical scavenging capacity was assessed from the bleaching of pyranine and pyrogallol red that is induced by free radicals generated from an azo initiator. In this assay system, the radical scavenging capacity per gram of AOB was estimated to be 95 micromol. On the other hand, the cytoprotective effect of AOB was also investigated on the basis of PC12 cell death induced by 6-hydroxydopamine. In this cellular system, AOB extract exhibited a cytoprotective effect only when the cells were pretreated with AOB. This pretreatment resulted in a significant increase in the levels of cellular glutathione as well as regulator of glutathione synthesis, such as the cystine/glutamate exchange transport system (xCT). This evidence suggests that AOB possesses both direct and indirect antioxidant activities to cope with oxidative insults.  相似文献

Cellular damage caused by reactive oxygen species has been implicated in several diseases and hence antioxidants have significant importance in human health. Cold water, hot water and methanolic extract of Pleurotus squarrosulus were evaluated for antioxidant activity against hydroxyl radical, DPPH (1,1-Diphenyl-2-picrylhydrazyl) radical, superoxide radical, nitric oxide (NO) scavenging, reducing power, ferrous ion chelating ability and beta-carotenellinoleic acid assay. Total phenol, flavonoid, beta-carotene and lycopene content were also determined. Hot water extract showed significant antioxidant activity in all the test systems. Hydroxyl radical scavenging activity of all the extracts has been significant compared to positive control. Hot water extract has been found to have higher phenolic, total flavonoid, beta-carotene and lycopene content than cold water and methanolic extract of the mushroom. Results of this study showed that, hot water extract has maximum antioxidant property and may be utilized as a promising source of therapeutics.  相似文献

The methanolic extract of Aloe vera L. gel was subjected to antioxidant guided fractionation with silica gel column chromatography to screen the potent fraction. The antioxidant capacities of different fractions were evaluated using DPPH radical scavenging assay and correlated with total phenol content. Total phenolic contents of different fractions were determined according to the Folin-Ciocalteu spectrophotometric method. The positive correlation was observed between DPPH radical scavenging assay and total phenolic contents indicating that phenolics in Aloe vera L. gel were fundamental contributor of antioxidant activity. Third pooled fraction was identified as potential fraction with highest antioxidant potential. This fraction indicated the presence of well resolved fluorescent components. Characteristic UV–vis absorption and HPLC analysis indicates that aloin take part in antioxidant potential attributed to aloe gel.  相似文献

Fifty-four different extracts of nine Bolivian plants belonging to the family Asteraceae were evaluated for their radical scavenging activity by the DPPH*, NBT/hypoxanthine superoxide, and (*)OH/luminol chemiluminescence methods, and for their antioxidant activity by the beta-carotene bleaching test. The total phenolic content was also determined by the Folin-Ciocalteu method, and the oxidative stability by the Rancimat test. Both remarkably high phenolic content and radical scavenging and antioxidant activities were found mainly in the ethyl acetate fractions among the different plant extracts. Some ethyl acetate and even some defatted crude extracts exhibited activities comparable to those of commercial extracts/compounds, thus making it possible to consider some of the studied plants as a potential source of antioxidants of natural origin.  相似文献

研究了蓝莓果渣提取物总酚含量及其抗氧化活性。分别采用水、40%乙醇及纤维素酶辅助乙醇超声提取蓝莓果渣,并用Folin-Ciocalteu试剂对3种提取物的总酚含量进行评估;并采用DPPH清除实验及O2—.清除实验对3种提取物的抗氧化活性进行研究。实验结果表明,纤维素酶辅助超声提取蓝莓果渣的总酚含量最高,可达425.36±15.21 mg GAE.100 g-1DW,远远高于水提物(169.46±9.75 mg GAE.100 g-1DW)及醇提物(218.39±12.54mg GAE.100 g-1DW)中的总酚含量。且纤维素酶辅助乙醇超声提取物对DPPH的清除能力为2.67±0.13 gVc.100 g-1DW,对O—.2的清除能力2.48±0.14 g Vc.100 g-1DW,明显好于醇提物及水提物抗氧化活性。  相似文献

Uncaria rhynchophylla (UR) is widely used as a traditional Chinese herbal medicine. However, there are few studies on antioxidant activity of UR extracts. The study was aimed at determining the antioxidant activity, the contents of polyphenol and flavonoid of UR extracts. Various assays were employed to evaluate the antioxidant property of water and ethanol extracts from the UR, compared to those of the other natural and synthetic antioxidants. UR extracts had high total phenolic contents in both the water (160 ± 2.32 mg GAE/g extracts) and ethanol extracts (190.2 ± 3.16 mg GAE/g extracts). In addition, total flavonoid contents were high in both the water extracts (154 ± 1.47 mg CE/g extracts) and ethanol extracts (184.2 ± 2.41 CE/g extracts). Specially, DPPH radical scavenging activity of ethanol extracts from UR were similar with vitamin C as a positive control. In addition, antioxidant capacity in ferric reducing antioxidant power (FRAP) were higher than that for BHT, which was used as a positive control. The antioxidant activity of extracts from UR showed stronger activity than those of vitamin C and α-tocopherol in ferric thiocyanate (FTC) and thiobarbituric acid (TBA) methods. The ethanol extracts of UR protected on H2O2-induced DNA damage. In addition, cytoprotective and anti-apoptotic effect of ethanol extracts from UR was also investigated in t-BHP-induced hepatotoxicity. Therefore, these results indicate that UR extracts have antioxidant properties through its ability to enhance the cell viability, mediation of production of ROS. The UR extracts could be suitable as an antioxidant in the food industry.  相似文献

Free radicals, the key mediators of a range of oxidative reactions involved in lipid oxidation are responsible for food quality deterioration leading to several health hazards. Antioxidants synthesized naturally or synthetically are capable of preventing oxidation of lipids and other related compounds. However, natural antioxidants have many benefits over synthetic ones. Sesame seeds contain large amount of natural bioactive components with high antioxidant potential. In the present study, 14 accessions of sesame containing wild species and cultivars were investigated. The antioxidant potential of sesame seed meal extract was evaluated by total phenolic content (TPC) method using Folin–Ciocalteu reagent, linoleic acid peroxidation by Ferric thiocyanate method, and free radical scavenging assay with 2,2-diphenyl-1-picryl hydrazyl radical. S. laciniatum showed highest mean values for total polyphenol content with maximum % inhibition of linoleic acid peroxidation on 10th day of course of the reaction span and highest antioxidant scavenging power. S. indicum subsp. malabaricum and S. radiatum also showed high total phenol content and radical scavenging capacity. Among the Sesamum indicum cultivars, Gujarat til 2 showed high TPC and high radical scavenging activity. Higher antioxidant property of Sesamum species in comparison to sesame cultivars highlights the need to utilize the wild genepool for the improvement of cultigens for enhanced nutraceutical value.  相似文献

Oxidative stress is the cause of neurodegenerative disorders such as Lou Gehrig’s disease, Parkinson’s disease, and Huntington’s disease; one practical way to prevent and manage neurodegenerative diseases is through the eating of food rich in antioxidants (dietary means). This present study sought to compare the ability of aqueous extract of sour tea (Hibiscus sabdariffa, calyx) and green tea (Camellia sinensis) to prevent some pro-oxidant [Fe (II), sodium nitroprusside, quinolinic acid]-induced lipid peroxidation in rat’s brain in vitro. Aqueous extracts of both teas were prepared (1 g tea in 100 ml of hot water). Thereafter, the ability of the extracts to prevent 25 μM FeSO4, 7 μM sodium nitroprusside, and 1 mM quinolinic acid-induced lipid peroxidation in isolated rat’s brain tissue preparation was determined in vitro. Subsequently, the total phenol content, reducing power, Fe (II) chelating and OH radical scavenging ability were determined. The results of the study revealed that both teas significantly (P < 0.05) inhibit lipid peroxidation in basal and pro-oxidant-induced lipid peroxidation in the rat’s brain homogenates in a dose-dependent manner. Also, the teas had high total phenol content [sour (13.3 mg/g); green (24.5 mg/g)], reducing power, and Fe (II) chelating and OH radical scavenging ability (except sour tea). However, green tea had a significantly higher (P < 0.05) ability to inhibit lipid peroxidation in both the basal and pro-oxidant-induced lipid peroxidation in rat’s brain homogenates in vitro. Therefore, it is obvious from the study that both teas had high antioxidant properties and could inhibit Fe2+, sodium nitroprusside, and quinolinic acid-induced lipid peroxidation in brain. However, green tea had a higher inhibitory effect, which may probably be due to its high total phenol content, reducing power, Fe (II) chelating ability, and OH radical scavenging ability.  相似文献

The aim of this investigation was to determine the content of total phenolics, antioxidant activity and cytotoxicity of methanol extracts from the aerial parts of 11 Korean medicinal salad plants. The highest total phenolic content of the methanol extracts was found in Aster scaber (17.1 mg 100 g(-1)), followed by Ixeris dentate (16.4 mg 100 g(-1)), Aster yomena (12.0 mg 100 g(-1)) and Sedum sarmentosum (9.1 mg 100 g(-1)) of FW. Methanol extracts of Ixeris dentate and Aster scaber at 50 microg mL(-1) exhibited the highest DPPH radical scavenging activity by 86.4 and 83.3%, respectively. It was registered a dose-dependent increase of DPPH free radical scavenging activity. Total phenolic content of the studied plant extracts was correlated with the DPPH radical scavenging activity. It was found by means of MTT assay, that cytotoxicity of the methanol extracts was the highest against HCT-116. Methanol extracts from Petasites japonicus (IC(50)<25.0 microg mL(-1)) showed the highest activity against HCT-116, following by Angelica gigas (34.75 microg mL(-1)), Erythronium japonicum (44.06 microg mL(-1)), and Aster scaber (54.87 microg mL(-1)). In conclusion, the studied salad plants have high total phenolics content and high antioxidant activity. These plants dose-dependently increased DPPH free radical scavenging activity. The total phenolics level was highly correlated with the free radical scavenging activity. Most of the studied salad plants have potent cytotoxicity activity. The results of this investigation suggest that the extracts of studied salad plants could be an addition to basic medicine for some diseases.  相似文献

Proteins are major target for radicals and other oxidants when these are formed in both intra- and extracellular environments in vivo. Formation of lesions on proteins may be highly sensitive protein-based biomarkers for oxidative damage in mammalian systems. Oxidized proteins are often functionally inactive and their unfolding is associated with enhanced susceptibility to proteinases. ROS scavenging activities of intact proteins are weaker than those of misfolded proteins or equivalent concentrations of their constituent amino acids. Protein oxidation and enhanced proteolytic degradation, therefore, have been suggested to cause a net increase in ROS scavenging capacity. However, certain oxidized proteins are poorly handled by cells, and together with possible alterations in the rate of production of oxidized proteins, may contribute to the observed accumulation and damaging actions of oxidized proteins during ageing and in pathologies such as diabetes, arteriosclerosis and neurodegenerative diseases. Protein oxidation may play a controlling role in cellular remodelling and cell growth. There is some evidence that antioxidant supplementation may protect against protein oxidation, but additional controlled studies of antioxidant intake to evaluate the significance of dietary/pharmacological antioxidants in preventing physiological/pathological oxidative changes are needed.  相似文献

为有效开发利用沙苑子,探究其抗氧化特性为深加工提供理论依据和经验参数,本文主要从沙苑子酚类提取物的总还原能力,对超氧阴离子自由基和羟基自由基的清除能力,及对Fe2+诱导的脂质过氧化的抑制作用等四个方面进行了研究并比较了三种不同沙苑子酚类提取物的抗氧化性能。结果表明沙苑子酚类提取物均具有一定的抗氧化能力,在实验浓度范围内呈现一定的量效关系。  相似文献

对印度块菌Tuber indicum子实体的提取物,包括55%乙醇粗提物(ECE)、石油醚提取物(PEF)和乙酸乙酯提取物(EAF)的清除DPPH自由基和羟基自由基能力、铁离子鳌合能力以及各提取物的总酚含量等进行了研究和测定,结果显示,3种提取物的清除自由基能力和铁离子鳌合能力具有显著差异(P0.05);ECE对DPPH自由基的清除活性最高,其EC50值为1.61g/L;EAF对羟基自由基及铁离子表现出较强的清除或螯合的能力,其EC50值分别为3.31g/L和0.70g/L;EAF的总酚含量(2.964mg GAE/g提取物)最高,其次是ECE,总酚含量为(2.618mg GAE/g提取物);PEF的清除自由基和铁离子鳌合能力较差,其总酚含量也最低(1.124mg GAE/g提取物);总酚含量与印度块菌提取物清除自由基以及鳌合铁离子的能力密切相关。  相似文献

This study aimed to evaluate the antioxidant activities of a cultured medicinal fungus--Armillariella mellea (Vahl. ex Fr.) Karst. (AM). Three antioxidant assay systems, namely cytochrome c, xanthine oxidase inhibition and FeCl2-ascorbic acid stimulated lipid peroxidation in rat tissue homogenate tests, were used. Total flavonoid and phenol contents of AM extracts were also analyzed. Results showed that both aqueous (AM-H2O) and ethanolic (AM-EtOH) extracts of solid state cultured AM showed antioxidant activities in a concentration-dependent manner. At concentrations 1-100 microg/ml, the free radical scavenging activity was 73.7-92.1% for AM-H2O, and 60.0-90.8% for AM-EtOH. These extracts also showed an inhibitory effect on xanthine oxidase activity, but with a lesser potency (IC50 - 9.17 microg/ml for AM-H2O and 7.48 microg/ml for AM-EtOH). In general, AM-H2O showed a stronger anti-lipid peroxidation activity on different rat's tissues than AM-EtOH. However, both AM extracts displayed a weak inhibitory effect on lipid peroxidation in plasma. Interestingly, the anti-lipid peroxidation activity of AM-H2O (IC50 - 6.66 microg/ml) in brain homogenate was as good as alpha-tocopherol (IC50 - 5.42 microg/ml). AM-H2O (80.0 mg/g) possessed a significant higher concentration of total flavonoids than AM-EtOH (30.0 mg/g), whereas no difference was noted in the total phenol content between these two extracts. These results conclude that AM extracts possess potent free radical scavenging and anti-lipid peroxidation activities, especially the AM-H20 in the brain homogenate.  相似文献

The antioxidant potential of crude extracts and fractions from leaves of Ouratea parviflora, a Brazilian medicinal plant used for the treatment of inflammatory diseases, was investigated in vitro through the scavenging of radicals 2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH), hydroxyl radical (HO*), superoxide anion (O2*-), and lipid peroxidation in rat liver homogenate. The crude extract (CEOP) and hydro-alcoholic fraction (OP4) showed strong inhibitory activity toward lipid peroxidation induced by tert-butyl peroxide (IC50 = 2.3 +/- 0.2 and 1.9 +/- 0.1 microg/ml, respectively). The same products exhibited a strong concentration-dependent inhibition of deoxyribose oxidation (14.9 +/- 0.2 and 0.2 +/- 0.1 microg/ml, respectively), and also showed a considerable antioxidant activity against O2*- (87.3 +/- 0.1 and 73.1 +/- 0.4 microg/ml, respectively) and DPPH radicals (55.4 +/- 0.3 and 38.3 +/- 0.4 microg/ml, respectively). The protective effects of CEOP and OP4 were also studied in mouse liver. CCl4 significantly increased (by 90%) levels of lipid hydroperoxides, carbonyl protein content (64%), DNA damage index (133%), aspartate aminotransferase (261%), alanine aminotransferase (212%), catalase activity (23%), and also caused a decrease of 60% in GSH content. The results showed that CEOP and OP4 exerted cytoprotective effects against oxidative injury caused by CCl4 in rat liver, probably related to the antioxidant activity showed by the in vitro free radical scavenging property.  相似文献
