We have recently reported that cannabinoid agonists can up-regulate and enhance the activity of serotonin 2A (5-HT2A) receptors in the prefrontal cortex (PFCx). Increased expression and activity of cortical 5-HT2A receptors has been associated with neuropsychiatric disorders, such as anxiety and schizophrenia. Here we report that repeated CP55940 exposure selectively up-regulates GRK5 proteins in rat PFCx and in a neuronal cell culture model. We sought to examine the mechanism underlying the regulation of GRK5 and to identify the role of GRK5 in the cannabinoid agonist-induced up-regulation and enhanced activity of 5-HT2A receptors. Interestingly, we found that cannabinoid agonist-induced up-regulation of GRK5 involves CB2 receptors, β-arrestin 2, and ERK1/2 signaling because treatment with CB2 shRNA lentiviral particles, β-arrestin 2 shRNA lentiviral particles, or ERK1/2 inhibitor prevented the cannabinoid agonist-induced up-regulation of GRK5. Most importantly, we found that GRK5 shRNA lentiviral particle treatment prevented the cannabinoid agonist-induced up-regulation and enhanced 5-HT2A receptor-mediated calcium release. Repeated cannabinoid exposure was also associated with enhanced phosphorylation of CB2 receptors and increased interaction between β-arrestin 2 and ERK1/2. These latter phenomena were also significantly inhibited by GRK5 shRNA lentiviral treatment. Our results suggest that sustained activation of CB2 receptors, which up-regulates 5-HT2A receptor signaling, enhances GRK5 expression; the phosphorylation of CB2 receptors; and the β-arrestin 2/ERK interactions. These data could provide a rationale for some of the adverse effects associated with repeated cannabinoid agonist exposure.  相似文献

Abstract: Interleukin (IL)-2, initially discovered for its mitogenic activity on T cells, also acts on monocytes, resulting in the activation of cytokine production, superoxide production, and tumoricidal activity. Because severe brain damage was observed in IL-2-transgenic mice, this cytokine may have some influence(s) on the cells of the CNS. We investigated IL-2 receptor-bearing cells in the CNS and found that activated microglia expressed α-chain mRNA and immunoreactive IL-2 receptor β-chain protein in culture. Although microglia did not express IL-2 receptors under normal culture conditions, they were induced to express these receptors by lipopolysaccharide (LPS) in a time-dependent manner. The IL-2 receptors were found to be functional because the viability and growth activity of LPS-treated microglia, but not untreated controls, increased in response to recombinant mouse IL-2 as determined by a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide colorimetric assay and bromodeoxyuridine uptake experiment, respectively. These effects of recombinant IL-2 were blocked by pretreatment with anti-mouse IL-2 receptor β-chain antibody. Our findings suggest that activated microglia in the CNS can respond to this T cell-derived factor regulating their growth, which may be an important mechanism of communication between nervous and immune systems in physiological and pathological conditions.  相似文献

The glucagon and glucagon-like peptide-1 (GLP-1) receptors play important, opposing roles in regulating blood glucose levels. Consequently, these receptors have been identified as targets for novel diabetes treatments. However, drugs acting at the GLP-1 receptor, although having clinical efficacy, have been associated with severe adverse side-effects, and targeting of the glucagon receptor has yet to be successful. Here we use a combination of yeast reporter assays and mammalian systems to provide a more complete understanding of glucagon receptor signaling, considering the effect of multiple ligands, association with the receptor-interacting protein receptor activity-modifying protein-2 (RAMP2), and the role of individual G protein α-subunits. We demonstrate that RAMP2 alters both ligand selectivity and G protein preference of the glucagon receptor. Importantly, we also uncover novel cross-reactivity of therapeutically used GLP-1 receptor ligands at the glucagon receptor that is abolished by RAMP2 interaction. This study reveals the glucagon receptor as a previously unidentified target for GLP-1 receptor agonists and highlights a role for RAMP2 in regulating its pharmacology. Such previously unrecognized functions of RAMPs highlight the need to consider all receptor-interacting proteins in future drug development.  相似文献

前列腺素D2(PGD2)是前列腺素(PGs)家族成员之一,广泛分布于各种哺乳动物组织中,并发挥多种生理功能。PGD,可以促进睡眠、诱导过敏反应、抑制血小板凝集及松弛平滑肌等,并且在生殖系统中起重要作用。机体中存在生化和免疫功能截然不同的两类前列腺素D合成酶(PGDs):脑型PGDS(L-PGDS)和生血型PGDS(hPGDS)。生殖系统中,L-PGDS主要存在于雄性生殖道,可能在睾丸发育、精子发生、精子成熟以及血一睾和血一附睾屏障等方面发挥重要作用。hPGDS在妊娠时期的子宫内膜和胚胎滋养层中表达,由其产生的PGD2可能通过DP和CRTH2两种受体来维持妊娠。此外,PGD2还可能与女性不孕以及精子在雌性生殖道内的运输等有关。  相似文献

用定点突变法分别得到了两个人白细胞介素-2(IL-2)的部分拮抗剂15Val-IL-2和126Asp-IL-2以及一个为IL-2受体α亚基结合缺陷型的突变体62Leu-IL-2,当将15Val-IL-2或126Asp-IL-2与62Leu-IL-2共同保温时,62Leu-IL-2的活性受到明显抑制,对此现象机理的分析表明15Val-IL-2或126Asp-IL-2可用于IL-2受体亚基结合缺陷型突变体的初步鉴定.同时,这一思路在其它受体-配基系统中具有一定的适用性.  相似文献

The Ca2+ sensor calmodulin (CaM) regulates numerous proteins involved in G protein-coupled receptor (GPCR) signaling. CaM binds directly to some GPCRs, including the dopamine D2 receptor. We confirmed that the third intracellular loop of the D2 receptor is a direct contact point for CaM binding using coimmunoprecipitation and a polyHis pull-down assay, and we determined that the D2-like receptor agonist 7-OH-DPAT increased the colocalization of the D2 receptor and endogenous CaM in both 293 cells and in primary neostriatal cultures. The N-terminal three or four residues of D2-IC3 were required for the binding of CaM; mutation of three of these residues in the full-length receptor (I210C/K211C/I212C) decreased the coprecipitation of the D2 receptor and CaM and also significantly decreased D2 receptor signaling, without altering the coupling of the receptor to G proteins. Taken together, these findings suggest that binding of CaM to the dopamine D2 receptor enhances D2 receptor signaling.  相似文献

In-Cell Western技术是基于近红外激光成像系统而开发的检测技术,可应用于大分子ryanodine受体2(RyR2)蛋白的检测。本研究通过对In-Cell Western固定剂的选择、细胞通透化、封闭液的选择、抗体工作浓度和心肌细胞密度等方面进行优化,建立心肌细胞RyR2蛋白检测的In-Cell Western方法,并分别用SGF和Verapamil以及免疫细胞化学技术对研究结果及方法进行了验证。优化后的In-Cell Western参数:H9C2细胞的种板密度选择1×104个/孔,选择甲醛作为固定剂,细胞经过Triton X-100洗脱液通透化处理,选择酪蛋白作为封闭液,选择1∶500稀释的RyR2一抗工作浓度。应用优化的参数检测心肌细胞RyR2蛋白,并用前期已被证明能显著提高心肌细胞RyR2蛋白荧光值的土茯苓黄酮和Verapamil进行验证,同时与免疫组化方法检测心肌细胞RyR2蛋白相比,检测结果一致。表明本文建立的In Cell Western方法检测大分子功能蛋白RyR2是可行的。  相似文献

吗啡在疼痛治疗中广泛应用,但其长期使用可以导致耐受,这大大影响了其临床应用价值,吗啡耐受是临床亟待解决的问题。研究发现大麻素受体2(cannabinoid receptor 2,CB2受体)参与吗啡耐受的发生与发展。CB2受体选择性激活剂与吗啡联合使用,可以减弱吗啡诱导产生的痛觉过敏和异常疼痛,抑制吗啡耐受的发生与发展。激活CB2受体抑制吗啡耐受的机制尚未明确,本文将就CB2受体在吗啡耐受中作用的研究现状作一综述。  相似文献

1.5-Hydroxytryptamine (10 M) enhanced ionic current mediated through the wild-type P2X2 receptor/channel expressed in Xenopus oocytes.2.5-Hydroxytryptamine (10 M) inhibited a current mediated through P2X2 receptor/channel mutants when Thr330 or Asn333 was replaced with Ile (T330I and N333I).3.Our results suggest that neutralization of Thr330 or Asn333 exposes a high-affinity, inhibitory binding site for 5-hydroxytryptamine. 目的:通过研究COX-2、PGE2、EP2受体及小胶质细胞在甲基苯丙胺中毒大鼠纹状体内的表达变化探讨甲基苯丙胺中毒大鼠纹状体中COX-2/PGE2系统与小胶质细胞活化之间的关系。方法:将40只健康成年雄性SD大鼠,随机分成对照组10只和实验组30只(实验组分成三个亚组,分为末次给药后1天组、2天组和3天组,n=10)。实验组给予10mg/kg的MA腹腔注射,对照组给予同样剂量的生理盐水,每天注射两次,注射时间为8:00、20:00,连续注射4天。分别于末次给药后的第1天、第2天、第3天处杀。用免疫组化技术对中毒大鼠纹状体(CPU)中COX-2、EP2受体及Iba1(钙离子接头蛋白,小胶质细胞内一种特异性标记物)的表达进行检测,并进行图像分析。另外,取大鼠的纹状体运用酶联免疫法检测PGE2的含量。结果:COX-2、PGE2、EP2受体及小胶质细胞在各组均有表达。与对照组相比,实验组中:COX-2、PGE2、EP2受体的1天组表达均不同程度下降;2天组中COX-2表达水平大幅度上升,PGE2、EP2受体表达仍低于正常水平;3天组COX-2表达水平继续升高,而PGE2、EP2受体表达趋于正常组水平。而小胶质细胞表达水平则是三个实验组均高于正常组,且3天组高于2天组,2天组高于1天组。对照组与实验组有显著性差异(P<0.05)。结论:COX-2/PGE2系统与甲基苯丙胺中毒大鼠纹状体内小胶质细胞活化无明显相关性;COX-2与甲基苯丙胺的神经毒性有关。  相似文献

Chemoattractant receptors regulate leukocyte accumulation at sites of inflammation. In allergic airway inflammation, although a chemokine receptor CCR2 was implicated in mediating monocyte-derived dendritic cell (DC) recruitment into the lung, we previously also discovered reduced accumulation of DCs in the inflamed lung in mice deficient in formylpeptide receptor Fpr2 (Fpr2−/−). We therefore investigated the role of Fpr2 in the trafficking of monocyte-derived DCs in allergic airway inflammation in cooperation with CCR2. We report that in allergic airway inflammation, CCR2 mediated the recruitment of monocyte-derived DCs to the perivascular region, and Fpr2 was required for further migration of the cells into the bronchiolar area. We additionally found that the bronchoalveolar lavage liquid from mice with airway inflammation contained both the CCR2 ligand CCL2 and an Fpr2 agonist CRAMP. Furthermore, similar to Fpr2−/− mice, in the inflamed airway of CRAMP−/− mice, DC trafficking into the peribronchiolar areas was diminished. Our study demonstrates that the interaction of CCR2 and Fpr2 with their endogenous ligands sequentially mediates the trafficking of DCs within the inflamed lung.  相似文献

Transferrin receptor 2 (TfR2) is a homologue of transferrin receptor 1 (TfR1) but has distinct functions from TfR1 in iron homeostasis. In keeping with its proposed role in iron sensing, previous studies showed that TfR2 has a short half-life and that holo-Tf stabilizes TfR2 by redirecting it from a degradative pathway to a recycling pathway. In this study, we characterized how the endocytosis, recycling and degradation of TfR2 relates to its function and differs from TfR1. TfR2 endocytosis was adaptor protein-2 (AP-2) dependent. Flow cytometry analysis showed that TfR1 and TfR2 utilized the same endocytic pathway only in the presence of holo-Tf, indicating that holo-Tf alters the interaction of TfR2 with the endocytic machinery. Unlike TfR1, phosphofurin acidic cluster sorting protein 1 (PACS-1) binds to the cytoplasmic domain of TfR2 and data suggest that PACS-1 is involved in the TfR2 recycling. Depletion of TSG101 by siRNA or expression of a dominant negative Vps4 inhibited TfR2 degradation, indicating that TfR2 degradation occurs through a multivesicular body (MVB) pathway. TfR2 degradation is not mediated through ubiquitination on the single lysine (K31) in the cytoplasmic domain or on the amino terminal residue. No ubiquitination of TfR2 by HA-ubiquitin was detected, indicating a lack of direct TfR2 ubiquitination involvement in its degradation.  相似文献

瞬时受体电位通道M2(transient receptor potential channel melastatin 2, TRPM2)是人体中一个重要的Ca2+通透性非选择性阳离子通道,通常表达于正常细胞胞膜和溶酶体膜上,并在氧化应激中发挥重要的离子调节作用。但近年发现,TRPM2也在多种恶性肿瘤(神经母细胞瘤,舌/喉鳞状细胞癌,肺癌,乳腺癌,胃癌,胰腺癌,膀胱癌,前列腺癌和T细胞白血病)中高表达,能通过调节细胞线粒体功能和自噬促进肿瘤细胞的生物学能量而促进其生存能力,通过调节抗氧化物水平增强细胞对氧化刺激的耐受力而表现出化疗抵抗作用。同时,在肿瘤细胞膜上该通道大量激活又对化疗药物联合使用发挥协同作用。此外,TRPM2能通过激活多种不同的分子的信号通路,促进细胞增殖、侵袭和转移能力。总之,根据肿瘤的不同,TRPM2对肿瘤细胞生物学行为的调控机制也不同,甚至具有复杂的双重作用。所以,对TRPM2的生化及分子机制的研究必将使我们对肿瘤的发生发展的认识更加全面。本文将从TRPM2蛋白质的结构,生理功能及肿瘤机制等不同角度系统阐述TRPM2的研究现状和进展。  相似文献

Expression of the receptor tyrosine kinase-like orphan receptor 2 (Ror2) has been identified in an increasing array of tumor types and is known to play a role as an important mediator of Wnt signaling cascades. In this study, we aimed to clarify Ror2 interactions with the Wnt pathways within the context of renal cell carcinoma (RCC). An examination of Ror2 expression in primary human RCC tumors showed a significant correlation with several Wnt signaling genes, including the classical feedback target gene Axin2. We provide evidence that Ror2 expression results in a partially activated state for canonical Wnt signaling through an increased signaling pool of β-catenin, leading to an enhancement of downstream target genes following Wnt3a stimulation in both renal and renal carcinoma-derived cells. Additionally, inhibition of low-density lipoprotein receptor-related protein 6 (LRP6) with either siRNA or dickkopf decreased the response to Wnt3a stimulation, but no change was seen in the increased β-catenin pool associated with Ror2 expression, suggesting that LRP6 cofactor recruitment is necessary for a Wnt3a-induced signal but that it does not participate in the Ror2 effect on β-catenin signaling. These results highlight a new role for Ror2 in conveying a tonic signal to stabilize soluble β-catenin and create a poised state of enhanced responsiveness to Wnt3a exogenous signals in RCC.  相似文献
