Domozych, D. S. and Korbusieski, T. J. 1985. The disruptionof dictyosome structure, polarity and secretory activity inthe scale-producing green alga Pyramimonas inconstans.—J.exp.Bot.36: 1304–1312. Dictyosome morphology and scale morphogenesis in the green alga,Pyramimonas inconstans, are well-defined ultrastructurally.By applying the sodium-specific ionophore, monensin, to actively-growingcell cultures, both dictyosome micro-architecture and the scale-basedsecretory pathway can be significantly altered. Applicationof 0.1 mol m3 and 0.01 mol m –3monensin for short periods(1 -3 h) results in cisternal curling and the disorientationof dictyosome polarity. Prolonged exposure to the ionophore(4-24 h) results in the complete disruption of dictyosome morphologyand scale morphogenesis with the emergence of irregular vesiclesfrom the maturing face. Removal of monensin by repeated washingreturns dictyosome structure and secretory activity to normal.These results indicate that Golgi apparatus integrity and secretoryactivity in Pyramimonas may be controlled by a localized, cytoplasmicproton gradient Key words: Monensin, Pyramimonas, dictyosome  相似文献

Maize (Zea mays) seedlings were grown under standard conditionsand the time for reformation of a slime droplet of standardsize was determined. The structure of the secretory cells wasexamined by light and electron microscopy. Osmium-zinc iodide(OZI) impregnation was used to provide contrast enhancementof the dictyosome forming-face cisternae and the endoplasmicreticulum (ER). The three-dimensional structure and relationshipsof these membrane systems were established from examinationof 300 nm thick sections. The presence of a marginal cisternalnetwork at the forming-face was confirmed, but no specific orientationof the ER to this face, or connection of ER to any part of thedictyosome could be demonstrated. Neither observation of reforming slime droplets, nor quantificationof secretory vesicle numbers gave any support to the suggestionthat slime formation is a phasic, or cyclic, process. Dilute solutions of solvents (1% ethanol, 1% and 0.1% DMSO)commonly used to solubilize potential inhibitors of Golgi activitywere found to delay significantly the time taken for reformationof a slime droplet of standard size. These solutions also hadslight effects on the numerical density of secretory vesicles. Cytochalasin D-induced accumulation of secretory vesicles wasused to determine rates of vesicle formation (0.39 dictyosome–1min–1) and hence turnover times for cisternal (6.5 min)and total dictyosome (26–39 min) membranes and the plasmamembrane (9.7 min). The volume contribution of the secretory vesicles to the slimedroplet was shown to be exceedingly small (0–1%), throwingdoubt on the value of using droplet reformation rates as anindicator of secretory activity. This reports the first sequence for a small subunit (185) ribosomal RNA gene (SSU rDNA) fromTetraselmis convolutae.Tetraselmis (=Platymonas) species are important feed stocks for many organisms important in mariculture. This sequence is an essential molecular marker to distinguish the DNAs ofTetraselmis from the DNAs of other organisms in the culture. In addition thisTetraselmis SSU rDNA sequence provides an important entry for the gradually expanding database of symbiotic microalgae.  相似文献

The fungal-derived derivative, brefeldin A, was used to disruptthe Golgi apparatus (GA) of the green alga, Gloeomonas kupfferi.Upon short treatments (10 µg ml–1 for 10 min orless), the Golgi bodies maintain their perinuclear positioning.However, the medial locus transforms from a tight stack of elongatecisternae to a network of swollen tubules. Upon longer treatments(60 min), swelling and vesiculation of cis face cisternae becomeapparent. Likewise, the edges of several trans face cisternaemay fuse with those of adjacent Golgi bodies leading to theformation of multiGolgi complexes. Metapenaeus ensis was spawned and laboratory cultured untilthe post-larval stage to illustrate and describe its embryonicand larval development. Fecundities ranged between 454 000 and689 000 eggs, and {small tilde}99.2% of spawned eggs hatchedafter 12 h and 40 min at 33.5 p.p.t. and 29.0-29.1°C. Sixnaupliar, three protozoeal, three mysis and one post-larvalstages were noted. A minimum of {small tilde}10 days (249 h20 min) from the time of hatching were required to completethe larval stages by subsisting only on Chaeioceros gracilisand Tetraselmis tetrathele at varying temperature and salinityof 28.1-29.8°C and 33.5—34.5 p.p.t., respectively.The embryonic and larval developmental stages of M.ensis aregenerally similar to those of other known penaeid species, exceptfor the size and structure of diagnostic characters, setationof appendages and duration of metamorphoses.  相似文献

The effect of abscisic acid (ABA), non-permeating osmoticumand desiccation treatment on storage protein synthesis duringmaturation of somatic embryos of Picea glauca (Moench) Voss.was examined. Sodium dodecyl sulphate-polyacrylamide gel electrophoresis(SDS-PAGE) and Western blot analysis demonstrated that someof the major crystalloid and matrix polypeptides were absentfrom somatic embryos maturing on medium containing ABA and lowosmoticum. However, treatment with polyethylene glycol-4000(PEG) in combination with ABA resulted in the synthesis of aspectrum of storage polypeptides resembling that of mature zygoticembryos. These storage proteins accumulated throughout an 8-weekculture period, resulting in a threefold higher protein contentthan somatic embryos maturing for the same time in the absenceof PEG. The structure and distribution of protein bodies incells of these osmotically treated somatic embryos was similarto that in cells of mature zygotic embryos. Treatment with 5·0-7·5%PEG prevented catabolism of the accumulated storage polypeptidesduring desiccation. The optimal culture conditions for somaticembryo maturation and storage protein deposition was 16 µMABA and 7·5% PEG for 8 weeks followed by desiccation.Analysis of mRNAs by in vitro translation and immunoprecipitationof translated products showed that the crystalloid protein mRNAprofiles of zygotic and those of somatic embryos maturing on16 µM ABA in the absence of PEG were similar. The differencesobserved in the pattern of accumulated polypeptides in thesesomatic embryos and those of mature zygotic embryos, therefore,indicates that storage-protein synthesis in response to osmoticumis in part regulated at the translational level. A freeze fracture analysis of the cell wall of Tetraselmis convolutae (Parke et Manton) revealed the existence of a crystalline median layer consisting of regular repeating subunits of 27 nm. These circular subunits lie in curved, interlocking, longitudinal rows with some irregular discontinuities appearing in the subunit pattern of the crystalline lattice. A comparison with the cell walls of other green algal flagellates is presented, revealing similarities and suggesting evolutionary patterns.  相似文献

The concentrations of various peel pigments of Cox’s OrangePippin apples have been measured during ripening on the treeand during storage at 12 °C. Total chlorophyll decreased and total carotenoid increased atthe time of the respiration climacteric. These changes weremore pronounced in fruit maturing on the tree where a significantincrease of anthocyanin occurred; it did not occur in storedfruit. There was no consistent or marked difference in the ratesof destruction of chlorophylls a and b. The carotenoids found in the unripe fruit were those characteristicof photosynthetic tissue, ß-carotene, lutein, violaxanthin,and neoxanthin. These decreased to a greater or lesser extent,and at different rates, on and off the tree. Other carotenoidswhich increased greatly during ripening were identified as esters,mainly of violaxanthin. During the climacteric there is a transition from an assemblageof pigments associated with the chloroplast to that typicalof a chromoplast.  相似文献

Abstract

Some ultrastructural features of cells of the marine haptophycean alga, Ochrosphaera neapolitana Schussnig in the palmelloid stage were examined. Chloroplasts which are contained in a compartment isolated from the cytoplasm by ER profiles and nuclear envelope, display trilamellated thylakoids running along the major axis. The stalked pyrenoid with inner bilamellated thylakoids, protrudes in a large membrane-bounded vacuole. Other structures, as the haptonematic and flagellar bases, autophagic vacuoles and mitochondria, are typical of the chrysophycean and haptophycean genera so far investigated.

The Golgi apparatus is represented by a single dictyosome composed of stacked cisternae fonctioning in a way that they form organic scales which constitute the main part of the cell covering. The scales, build up of microfibrils disposed parallel each to other, lie in cisternal lumina of the dictyosomal maturing face; scaly cisternae are numerous in the peripheral cytoplasm and are observed merging in the plasma membrane and discharging the content outside the protoplast.

Dictyosomal activity is evidenced morphologically by massive vesicle production. Three kinds of membrane-bounded vesicles were identified in the present material: i) inner-granulated vesicles, arising from the maturation face; ii) coated vesicles, scattered in the cytoplasm or at the periphery of the golgi body, and iii) dense-cored vesicles, present in the proximity of the maturation face. The possible functional relationships related to scale production and assembly outside the protoplast, and between the nucleus and dictyosome are discussed.  相似文献

Gastroclonium clavatum tetrasporogenesis have been studied usinglight and electron microscopy. They are spherical bodies whichdifferentiate early and 50–70 per branch segment havebeen recorded. The developing sporangia expand into the largethallus cavity. The plastids become dispersed evenly throughthe cytoplasm at this stage. Thylakoids become arranged as parallelarrays and sporadically a single peripheral thylakoid enclosesall the others; the matrices contain globules reaching a maximumdiameter of 270 nm. Plastid multiplication occurs through binaryor multiple fission. The starch occurs in bowl-shaped grains.The mitochondria are always rich in cristae and often providedwith small opaque inclusions. The Golgi apparatus is markedby pleomorphic dictyosome features. Dictyosome-mitochondrionassociations are frequent. The different kinds of cytoplasmicinclusion are described. The biosynthesis of complex asparagine (N)-linked oligosaccharidesin vertebrates proceeds with the linkage of N-acetylglucosamine(GlcNAc) to the core mannose residues. UDP-N-acetylglucosamine:ß-D-mannosideß1–4 N-acetylglucosaminyltransferase III (GlcNAc-TIII,EC2.4.1.144) catalyzes the addition of GlcNAc to the mannosethat is itself ß1–4 linked to underlying N-acetylglucosamine.GlcNAc-TIII thereby produces what is known as a ‘bisecting’GlcNAc linkage which is found on various hybrid and complexN-glycans. Background and Aims: The branch-base xylem structure of the endangered Wollemia nobiliswas anatomically investigated. Wollemia nobilis is probablythe only extant tree species that produces only first-orderbranches and where all branches are cleanly abscised. An investigationwas carried out to see if these unusual features might influencebranch-base xylem structure and water supply to the foliage. Methods: The xylem was sectioned at various distances along the branchbases of 6-year-old saplings. Huber values and relative theoreticalhydraulic conductivities were calculated for various regionsof the branch base. Key Results: The most proximal branch base featured a pronounced xylem constriction.The constriction had only 14–31 % (average 21 %)of the cross-sectional area and 20–42 % (average28 %) of the theoretical hydraulic conductivity of themore distal branch xylem. Wollemia nobilis had extremely lowHuber values for a conifer. Conclusions: The branch-base xylem constriction would appear to facilitatebranch abscission, while the associated Huber values show thatW. nobilis supplies a relatively large leaf area through a relativelysmall diameter ‘pipe’. It is tempting to suggestthat the pronounced decline of W. nobilis in the Tertiary isrelated to its unusual branch-base structure but physiologicalstudies of whole plant conductance are still needed.  相似文献

The influence of various treatments and temperature regimeson total chlorophylls and on the chlorophyll a:b ratio of westernwheatgrass and blue grama plants was investigated at differenttime intervals during the 120-day growth period. Western wheatgrass,a C3 species, accumulated greater amounts of chlorophyll thandid blue grama plants, a C4 species. Maximum concentrations(mg gd wt–1) of chlorophylls in western wheatgrass andin blue grama were recorded at the lower (13/7°C) and higher(30/18°C) temperature regimes. Nitrogen fertilizer alonedecreased the chlorophyll content in both species. The chlorophylla:b ratio in blue grama ranged from an average of 2·00under irrigated plus fertilized conditions to 3·00 undercontrol and fertilized conditions. On the other hand, the chlorophylla:b ratio in western wheatgrass remained constant at 3·00throughout the growing season under various treatments and temperatureregimes.  相似文献

The lobate ctenophore, Mnemiopsis leidyi, consumed eggs andlarvae of the bay anchovy, Anchoa mitchilli, in laboratory experiments.This ctenophore exhibited a type I functional response to increasesin egg densities without reaching saturation at high prey densities.Clearance rate increased with increasing experimental containervolume. There was a 3-fold increase in the volume of water clearedby 2.0–2.5-cm ctenophores and a >5-fold increase for4.5–5.0-cm ctenophores in 15–1 versus 100–200–1containers. Clearance rate was dependent on tenophore lengthbut was probably underestimated for the larger animals due tocontainer effects. The presence of various densities of alternateprey, Acartia hudsonica or Anemia sp. nauplii, in addition toAnchoa mitchilli eggs did not affect the clearance rates onthe eggs alone. Comparison of clearance rates of 2.0–2.5-cmctenophores on various ages of starved and fed bay anchovy larvaeindicated that predation may be higher on yolk-sac larvae thanon eggs but decreases as the larvae grow. After 3 days posthatch starved anchovy larvae become more vulnerable to predationthan fed larvae. Comparative ultrastructural investigations on many isolates ofTetraselmis from Japan and the Pacific coast of North America, and on cultures from the Culture Centre of Algae and Protozoa, Cambridge, England, have revealed that the species have characteristic fine structural features of the pyrenoid. Using the pyrenoid structure as a basic character it is proposed that the genus be subdivided into four subgenera,Tetraselmis, Prasinocladia, Tetrathele andParviselmis. In the present paper, species of the subgenusTetraselmis, includingT. cordiformis, T. ascus, T. convolutae andT. astigmatica sp. nov., are described in detail.  相似文献

Three marine phytoplankton species (Skeletonema costatum, Olisthodiscusluteus andGonyaulax tamarensis) were grown in batch culturesat 15°C and a 14:10 L:D cycle at irradiance levels rangingfrom 5 to 450 µEinst m–2 s–1. At each irradiance,during exponential growth, concurrent measurements were madeof cell division, carbon-specific growth rate, photosyntheticperformance (both O2 and POC production), dark respiration,and cellular composition in terms of C, N and chlorophyll a.The results indicate that the three species were similar withrespect to chemical composition, C:N (atomic) = 6.9 ±0.4, photo-synthetic quotient, 1.43 ± 0.09, and photosyntheticefficiency, 2.3 ±0.1 x 10–3 µmol O2 (µgChl a)–1 h–1 (µEinst m–2 s–1)–1.Differences in maximum growth rate varied as the –0.24power of cell carbon. Differences in growth efficiency, werebest explained by a power function of Chl a:C at µ = 0.Compensation intensities, ranged from 1.1 µEinst m–2s–1 for S. costatum to 35 forG. tamarensis and were foundto be a linear function of the maintenance respiration rate.The results indicate that interspecific differences in the µ–Irelationship can be adequately explained in terms of just threeparameters: cell carbon at maximum growth rate, the C:Chl aratio (at the limit as growth approaches zero) and the respirationrate at zero growth rate. The causes of interspecific differences in the µ-l relationshipare examined in the context of a mechanistic model which relatesµ to irradiance in terms of six factors:, kc photosyntheticquotient (PQ), Chl a:C, respiration and excretion. The effectof cell size on the light saturated growth rate is also considered.It is shown that photosynthetic efficiency and PQ exhibit remarkablylittle interspecific variability, and average 0.024 ±0.005 µg C(µg Chl a)–1 h–1 (µEm–2 s–1)–1 and 1.5 ± 0.2 mol 02 molC–1 (when NO3 is the nitrogen source) respectively.Two useful relationships were derived: (i) between growth efficiency,g and Chl a:C at µ. = 0; (ii) between the compensationintensity, Ic and the Chl a-specific maintenance respirationrate. Both relationships were independent of temperature anddaylength. Species best adapted to growth at low light werefound to exhibit high Chl a:C ratios and low maintenance respirationrates. As a group, diatoms were consistently the best adaptedfor growth at low irradiance. Chiorophytes, haptophytes, chrysophytesand cryptophytes were intermediate in their performance at lowirradiance. Dinoflagellates exhibited extreme diversity, withspecies spanning the spectrum from very good performance atlow irradiance to very poor. A new µmax-cell carbon relationshipis given based on growth rates normalized to 15°C. Evidenceis presented to show that noise in this relationship can besignificantly reduced by using only carbon-specific growth ratesand using only data for species grown at the same daylength.  相似文献

The objective of this study was to quantify the functional responsein feeding rate in the various developmental stages of Calanusfinmarchicus to different concentrations of the diatoms Thalassiosiranordenskioeldii and Porosira glacialis, and the haptophyseanPhaeocystis pouchetii. Grazing of copepodite stage I–VC.finmarchicus was measured using two different approaches.Feeding rates were obtained from either incubation experiments,estimating the rate of removal of particles from suspension,or by quantifying the turnover rate of the plant pigments inthe gut. Clearance as a function of algal concentration (1–30µg plant pigment 1–1) was described in juvenilestages of C.finmarchicus fed the diatoms T.nordenskioeldii [20µm equivalent spherical diameter (ESD)], P.glacialis (40µm ESD), and two size categories (30–100 µmand >100 µm ESD) of the gelatinous alga P.pouchetii.When the copepodite stages were fed T.nordenskioeldii, the gutcontent of plant pigments was in general higher than when fedP.glacialis. Rates obtained were variable when the same copepoditestages were offered the two size categories of P.pouchetii,but within the same order of magnitude as those obtained forthe larger diatom. At unialgal diets, diatoms were more readilyconsumed than the larger size fraction among colonies of P.pouchetiiby copepodite stage I–III C.finmarchicus. But given anappropriate prey size, C.finmarchicus grazed both diatoms andcolonies of gelatinous algae at equal rates. A linear relationshipbetween gut content and food concentrations <10 µgchlorophyll 1–1 was found. Before they emerged from the fascicular sheath, the tissuesof young needles of Pinus ponderosa P. et C. Lawson alreadyshowed some characteristics typical of mature needles. The organelles,particularly the plastids, had undergone different development.The plastids in different types of cell varied in their ultrastructureand in association with the endoplasmic reticulum (ER). A sheathof ER was observed around the amoeboid plastids in epidermalcells, epithelial cells of resin ducts and maturing transfusiontracheids whereas there was no ER sheath around the young mesophyllchloroplasts, the fusiform chloroplasts in some transfusiontracheids and the proplastids in xylem and phloem cells. Thecontent of chlorophyll (a+b) was 0·85 g kg-1 dry matterand chlorophyll a/b ratio was 2·70. Precise measurements of the net flux of protons in Chara internodalcells were made with a recently designed high-resolution pH-meter.Survival of intact Chara internodal cells in artificial pondwater (APW) that contained HC1 at various concentrations wasalso examined. The apparent net flux of H+ was inward and muchsmaller than that reported so far. In APW at pH 4.005, a valuehigher than the extracellular pH expected from the values ofH+ efflux reported to date, all of the intact Chara internodalcells died within a day. With reference to the data on the circadianflow of ions in the pulvinus of Phaseolus [Kiyosawa (1979) PlantCell Physiol. 20: 1621–1634, Hosokawa and Kiyosawa (1983)Plant Cell Physiol. 24: 1065–1072] and ionic regulationin Chara L-cells [Kiyosawa and Okihara (1988) Plant Cell Physiol.29: 9–19], a discussion is presented of the prossiblyminor contribution of the net flux of H+ in the generation ofthe electrical membrane potential.
