The production of a microfluidic device for microbial culture has necessitated the development of techniques for the prevention of bacterial adhesion to a range of polymeric substrates including fluoropolymers such as fluorinated ethylene polypropylene, and polyolefins such as low-density polyethylene. Treatment of such materials to increase hydrophilicity reduces the incidence of attachment of Escherichia coli during the first 4 h of cultivation, although no decrease in the number of biofilm initiation sites was detected after 16 h. The incorporation of a mannose analogue to block binding proteins on the F1 binding fimbriae was also investigated. The possibility of ensuring suspension culture of bacterial cells in high surface area to volume ratio nano-vessels is thus facilitated by the correct choice and pre-treatment of materials used in their construction.  相似文献

Combination drug therapies exist that combat HIV replication and the production of virions. But just as the easiest way to deal with an interloper is to keep him from entering your home rather than removing him after the fact, most studies directed at reducing HIV infection are designed to keep HIV at bay, outside the host cell. Work on keeping HIV out has progressed from early work on CD4 to the chemokine receptors CCR5 and CXCR4 found on the surface of host cells. More recently, the depletion of cholesterol, the presence of which is essential for HIV entry, has been studied as a means to subvert HIV entry, and new work by Finnegan and others suggest that another useful strategy may involve increasing the amount of the sphingolipid ceramide found in lipid rafts on the surface of host cells. Increased ceramide might inhibit HIV entry by a number of means, including the displacement of cholesterol and modifying the overall organization and structure of the lipid rafts.  相似文献

Summary Formulations which are economical and which can deliver a viable organism are critical to developing successful biocontrol products for plant pathogens. In the present study, alginates derived from commercial kelp and produced byAzotobacter vinelandii isolates ATCC 9104 and 12 837 were compared in their ability to form stable, biodegradable granular formulations of the biocontrol fungiTalaromyces flavus andGliocladium virens. Bacteria were grown in shake flask cultures (180 rpm) at 32°C for 104 h. The cultures were monitored for pH, dissolved oxygen, glucose concentration, dry cell weight, and alginate dry weight. Aqueous solutions of the bacterial alginates, as well as the kelp-derived alginate products, gelled readily in 0.25 M calcium chloride. Mannuronate (M) and guluronate (G) compositions of the alginate samples were determined by circular dichroism. M/G ratios for cultures of isolate 12837 averaged 0.98±0.18; for isolate 9104, 1.59±0.12; and for kelp, 1.54±0.39. The viability ofT. flavus in the kelp and bacterial alginate formulations were similar over 84 days. An exploratory experiment indicated good viability ofG. virens using the same bacterial alginates. This study demonstrated a practical use for bacterial alginate as a potentially less costly substitute for kelp alginate in the preparation of biocontrol agent formulations.  相似文献

Persistent virus infections are increasingly being recognized as a significant cause of human morbidity and mortality. To establish persistence, a virus must establish infection and evade eradication by the host immune response, in particular by cytotoxic T lymphocytes (CTL). We have studied a virus that establishes persistence in part by suppressing the CTL response of the infected host. The virus persists in many cell types, including lymphocytes and macrophages. We show that prior immunization with a vaccine designed to induce CTL (in the absence of antiviral antibody) confers complete protection against subsequent establishment of persistence in all tissues analyzed. The vaccine can be designed to express as few as 10 amino acids of a viral protein that comprise the CTL epitope. Further, two CTL epitopes for two discrete MHC haplotypes can be successfully used in a single vaccine that protects both strains of mice. Hence, a "string of CTL epitopes" (beads) concept for vaccination is feasible. Finally, the CTL vaccine provided protection against the establishment of persistence by an immunosuppressive virus.  相似文献

以细菌为基础的生物技术在蓬勃发展的同时也不断受到噬菌体感染的威胁,噬菌体感染已成为微生物发酵过程中的一个顽疾,其实质是噬菌体与细菌之间复杂的共进化关系。在漫长的进化过程中,噬菌体已经形成了多种针对细菌抗性系统的逃逸机制。合理的工厂设计、菌株的轮换策略和传统的基因工程方法能在一定程度上降低噬菌体感染的风险,但仍然无法避免。基于CRISPR-Cas系统的防治策略仅需噬菌体的序列信息就可以理性设计噬菌体抗性菌株,且可以通过叠加效应不断增强菌种抗性,从而避免噬菌体的逃逸;群体感应信号分子则可以从整体水平上调节细菌的噬菌体抗性。这些新发现为噬菌体感染问题的解决带了新的希望,而噬菌体基因组编辑技术和合成生物学的快速发展则将进一步加深人们对噬菌体感染防治领域的认识。  相似文献

The delivery of DNA to mammalian cells is of critical importance to the development of genetic vaccines, gene replacement therapies and gene silencing. For these applications, targeting, effective DNA transfer and vector safety are the major roadblocks in furthering development. In this report, we present a novel DNA delivery vehicle that makes use of protoplasted, achromosomal bacterial minicells. Transfer of plasmid DNA as measured by green fluorescent protein expression was found to occur in as high as 25% of cultured Cos-7 cells when a novel chimeric protein containing the D2-D5 region of invasin was expressed and displayed on the surface of protoplasted minicells. Based on endoplasmic reticulum stress and other responses, protoplasted minicells were non-toxic to recipient eukaryotic cells as a consequence of the transfection process. Taken together, these results suggest that bacterial minicells may represent a novel and promising gene delivery vehicle.  相似文献

Use of Lactobacillus to prevent infection by pathogenic bacteria   总被引:8,自引:0,他引:8  
This review focuses on the use and potential of Lactobacillus to prevent infections of the urogenital and intestinal tracts. The presence and dominance of Lactobacillus in the vagina is associated with a reduced risk of bacterial vaginosis and urinary tract infections. The mechanisms appear to involve anti-adhesion factors, by-products such as hydrogen peroxide and bacteriocins lethal to pathogens, and perhaps immune modulation or signaling effects. The instillation of Lactobacillus GR-1 and B-54 or RC-14 strains into the vagina has been shown to reduce the risk of urinary tract infections, and improve the maintenance of a normal flora. Ingestion of these strains into the gut has also been shown to modify the vaginal flora to a more healthy state. In addition, these strains inhibit the growth of intestinal, as well as urogenital pathogens, colonize the gut and protect against infections as shown in mice. Other probiotic strains, such as Lactobacillus GG, have been shown to prevent and treat gastroenteritis caused by rotavirus and bacteria. Given that lactobacilli are not the dominant commensals in a gut which comprises around 10(10) organisms, much work is still needed to define the mechanisms whereby GR-1, RC-14, GG and other strains contribute to health restoration and maintenance. Such critically important studies will require the medical science community to show a willingness to turn away from pharmaceutical remedies as the only solution to health and disease.  相似文献

The number of users of intravenous drugs (UID), altogether from 35,000 to 25,000 persons, was determined by the method of coefficients. The analysis of 130 anonymous questionnaires, including 75 street questionnaires (61%) and 51 questionnaires (39%) distributed among UID in narcological and infectious hospitals showed that out of 130 respondents 60% underwent hospital treatment. The study revealed that most of the questioned users used crudely made opiates, 48 (37%) out of 130 persons made injections with used syringes and needles (in younger age groups the proportion of such UID amounted to 63%). 70% of the respondents used individual injection instruments (in a group of persons older than 25 years they amounted to 90%). The conclusion was made on the necessity of introducing a school program on the healthy mode of life and information programs on the prevention of the spread of infection among UID with due regard to different age groups, as well as training outreach workers for disseminating information on the prophylaxis of HIV infection among UID.  相似文献

Additive manufacturing or 3D printing has spearheaded a revolution in the biomedical sector allowing the rapid prototyping of medical devices. The recent advancements in bioprinting technology are enabling the development of potential new therapeutic options with respect to tissue engineering and regenerative medicines. Bacterial polysaccharides have been shown to be a central component of the inks used in a variety of bioprinting processes influencing their key features such as the mechanical and thermal properties, printability, biocompatibility, and biodegradability. However, the implantation of any foreign structure in the body comes with an increased risk of bacterial infection and immunogenicity. In recent years, this risk is being potentiated by the rise in nosocomial multidrug-resistant bacterial infections. Inks used in bioprinting are being augmented with antimicrobials to mitigate this risk. The applications of bacterial polysaccharide-based bioinks have the potential to act as a key battlefront in the war against antibiotic resistance. This paper reviews the range of bacterial polysaccharides used in bioprinting and discusses the potential of various bioactive polysaccharides to be integrated into these inks.  相似文献

A rapid, simple and sensitive technique was developed for the immunodetection and immunopurification of enterotoxins from contaminated materials. This technique, which avoids interference by protein A, involves the application of either staphylo-coccal culture supernatant fluids or contaminated materials to an immunoabsorbent of rabbit immunoglobulins coupled to a solid matrix followed by passage through an anti-enterotoxin antibody affinity column. The total elimination of the cross-reactivity caused by protein A was monitored by immunoblotting with monoclonal anti-enterotoxin antibodies.  相似文献

Biofouling is a major problem faced by marine industries. Physical and chemical treatments are available to control fouling, but most are costly, time consuming or negatively affect the environment. The use of aeration (ie continuous streams of air bubbles) to prevent fouling was examined. Experiments were conducted at three sites with different benthic communities. Experimental panels (10 cm × 10 cm; PVC and concrete) were deployed with or without aeration. Aeration flowed continuously from spigots 0.5 m below the panels at a rate of ~3.3 to 5.0 l min?1. After 1 and 4 weeks, aerated PVC panels from all sites had significantly less fouling than non-aerated controls. Aeration reduced fouling on both the PVC and concrete surfaces. Fouling was reduced on panels directly in bubble streams while panels 30 cm and 5 m away had significantly more fouling. Thus, under the conditions used in this study, aeration appears to be an effective and simple way to prevent fouling.  相似文献
