We analysed the endogenous cytokinin levels of Dendrobium Madame Thong-In seedlings grown in vitro during vegetative and flowering-inductive periods. HPLC was used to fractionate the extracts and radioimmunoassay (RIA) was used for assay of zeatin (Z), dihydrozeatin (DZ), N(6)-(Delta(2)-isopentenyl)-adenine (iP) and their derivatives. Coconut water used in experiments was found to contain high level (>136 pmol ml(-1)) of zeatin riboside (ZR). Protocorms and seedlings cultured in medium with coconut water were found to contain 0.5-3.9 pmol g(-1) FW of the cytokinins analysed. Seedlings (1.0-1.5 cm) cultured in flowering-inductive liquid medium containing 6-benzyladenine (BA, 4.4 muM) and coconut water (CW, 15%) contained up to 200 and 133 pmol g(-1) FW of iP and iPA, respectively. These levels were significantly higher than all other cytokinins analysed in seedlings of the same stage and were about 80- to 150-folds higher than seedlings cultured in non-inductive medium. During the transitional (vegetative to reproductive) stage, the endogenous levels of iP (178 pmol g(-1) FW) and iPA (63 pmol g(-1) FW) were also significantly higher than cytokinins in the zeatine (Z) and dihydrozeatin (DZ) families in the same seedlings. Seedlings that grew on inductive medium but remained vegetative contained lower levels of iPA. The importance of the profiles of iP and its derivatives in induction of in vitro flowering of D. Intrinsically photosensitive retinal ganglion cells (ipRGCs) express the photopigment melanopsin and serve as the primary retinal drivers of non-image-forming visual functions such as circadian photoentrainment, the pupillary light reflex, and suppression of melatonin production in the pineal. Past electrophysiological studies of these cells have focused on their intrinsic photosensitivity and synaptic inputs. Much less is known about their voltage-gated channels and how these might shape their output to non-image-forming visual centers. Here, we show that rat ipRGCs retrolabeled from the suprachiasmatic nucleus (SCN) express a hyperpolarization-activated inwardly-rectifying current (I(h)). This current is blocked by the known I(h) blockers ZD7288 and extracellular cesium. As in other systems, including other retinal ganglion cells, I(h) in ipRGCs is characterized by slow kinetics and a slightly greater permeability for K(+) than for Na(+). Unlike in other systems, however, I(h) in ipRGCs apparently does not actively contribute to resting membrane potential. We also explore non-specific effects of the common I(h) blocker ZD7288 on rebound depolarization and evoked spiking and discuss possible functional roles of I(h) in non-image-forming vision. This study is the first to characterize I(h) in a well-defined population of retinal ganglion cells, namely SCN-projecting ipRGCs.  相似文献

Actinobacterial diversity was investigated in three hot springs [Tengchong Frog Mouth (TFM) hot spring (in Tengchong, China), Kamchatka Robb Flag hot spring (in Kamchatka, Russia), and Nevada Boiling Spring (in Nevada, USA)], a Tengchong Frog Mouth hot spring-derived pond and a soil sample near the TFM hot spring. Culture-independent techniques were employed including denaturing gradient gel electrophoresis and actinobacterial 16S rRNA gene phylogenetic analysis. The three hot springs and the hot spring-derived pond had pH of 6.8 to 7.9 and temperature of 50 to 81°C. Actinobacteria were for the first time found to be very diverse in hot springs with temperature up to 81°C, indicating that they have an unexpected capability of adapting to hot spring environments. A total of 346 actinobacterial clones were obtained from the three hot springs, and these clones were identified as 31 RFLP types, falling into the Actinomycetales (only limit to the Frankineae suborder), the Rubrobacterales, uncultured Candidatus Microthrix, unclassified Actinobacteria, and uncultured Candidate Division OP10. Our data have implications for a better understanding of the distribution of Actinobacteria in hot springs.  相似文献

The proventriculi of 11 Eurasian cranes (Grus grus) from central Iran were examined for the existence of parasitic helminths. Preliminary reports suggested that the death of these birds was related to untimely cold weather. Nine proventriculi (82%) were heavily infected by the nematode Tetrameres grusi. Glandular structure of the infected proventriculi was replaced by epithelial atrophy but significant inflammatory reactions were not observed in any of the infected organs. In serious infections, the nematode produced vast structural and functional changes, causing organ dysfunction and glandular necrosis. The coincidence of heavy helminth infection at times of environmental stress may lead to debilitation, wasting, and perhaps mortality in migratory cranes.  相似文献

It is well known that no chlorophyll synthesis and photosystem biogenesis have been detected in dark-grown angiosperm seedlings. However, in this report, we showed that both PSⅡ and PSⅠ could be formed in dark-grown lotus ( Nelumbo nucifera Gaertn.) seedlings. Lots of evidence were given: First, during the dark-grown period, the single fluorescence emission peak at 679 nm in lotus embryo red-shifted and transformed into the normal PSⅡ fluorescence emission; Simultaneously, PSⅠ fluorescence emission at 730 nm appeared and increased obviously; Second, with partial denaturing SDS-PAGE method, PSⅠ chlorophyll-protein complex could be clearly separated from 10 days dark-grown lotus seedlings; Third, the existence of Lhca1 was also proved by Western blots. Moreover, measurements of electron transfer rate demonstrated that both PSⅡand PSⅠ core in dark-grown lotus seedlings were photochemically active.  相似文献

Seed cone in Platycladus orientalis (L.) France consists of four or five pairs of decussate bracts. Usually, two pairs of the fertile bracts in the middle of the cone subtend six ovules, which initiate in an acropetal manner. Only one ovule presents on each of the upper fertile bract, while two ovules initiate from a common primordium in the axil of lower bracts. In Beijing, most female cones initiated in July. All parts of the cone formed before dormancy, which occurred during November to the next January. After pollination in March, bract morphology changed dramatically; intercalary growth of the bract base formed a conspicuous protuberance, in which inverted vascular system developed. Furthermore, ovules on different pairs of bracts initiated in an acropetal manner and two ovules in each lower fertile bract initiated from a common primordium, which was different from the basipetal initiation of ovules and independently formed single ovule as reported by Takaso in Calltris.  相似文献

Nucella lapillus is a common and widely distributed carnivorous gastropod of North Atlantic rocky sea shores. Populations of this animal usually show variation in shell shape according to the exposure of their habitat to wave action, with individuals from exposed sites having a short squat shell and a wide aperture as compared with a more elongated form seen in shelter. The same pattern of variation is seen over most of the species European range, but there are some exceptions. One of these has been described in Sullom Voe (Shetland) and this prompted an investigation of Faroese populations to ascertain whether they behaved in a similar manner. It is shown in this paper that Faroese populations vary in shell shape with exposure in the usual way (and not like those in Shetland) and moreover show such a fine and precise reflection of minor alterations in environmental conditions that measurement of dog-whelk shells may prove to be the best way of 'measuring' exposure in Faroe.  相似文献

Cowdria ruminantium was isolated from a natural infection of heartwater in a captive sitatunga (Tragelaphus spekei) which died after a short illness in Jos Zoo, Nigeria. The isolate killed laboratory mice in a shorter period of time than did isolates of C. ruminantium recovered from natural cases in cattle. This is the first confirmed case of heartwater in a captive wild ruminant in Nigeria, where the disease is endemic.  相似文献

Abstract

Background: Accumulating evidence, mainly from small-scale experiments, suggests that species diversity helps stabilise ecological communities; however, this relationship needs testing at larger scales in a wider range of natural communities.

Aims: In experimentally burnt ericaceous shrub stands, we aimed to determine whether more diverse stands had more stable vegetation cover.

Methods: Using two prescribed fire management experiments – one on heathland and one in forest – at scales of 100–700 m2, we compared pre-disturbance vegetation characteristics (measures of diversity, traits and composition) with resistance (degree of perturbation on disturbance), resilience (here defined as post-disturbance recovery rate) and stability (the inverse of temporal variability).

Results: Responses to disturbance were usually best explained by a measure of dominance: Simpson's index. High dominance was associated with high resilience, but low resistance and low stability. Within the forest, the shrub community had lower dominance and higher stability than it did on heathland. Dominance and diversity were strongly linked as opposite poles of a principal axis of vegetation variation; therefore, more diverse vegetation was more stable.

Conclusions: Ericaceous shrub cover is known to support important ecosystem services, such as soil protection, carbon sequestration, flood prevention and livestock grazing. Our results suggest that high shrub diversity would support stable provision of these services. Land managers who prioritise this aim in British heathlands and pinewoods should consider a wider range of management approaches than those of typical current practice.  相似文献

Mapping of catecholaminergic (CAergic) neurons in the nervous system has been performed in 5 species of turbellarians of the order Neorhabdocoela: in three species of typhloplanides, Mesostoma lingua, Bothromesostoma essenii, and Rhynchomesostoma rostratum, and in two species of Dalyellioida, Castrella truncata and Gieysztoria cuspidata. In spite of an essential diversity of their orthogones, the number of cerebral neurons varies insignificantly: from 4 to 5 pairs at 3 variants of geometry of their arrangement. In the presence of the medial paddle-shaped outgrowth of neuropil, two pairs of anterior cerebral neurons are located on its both sides, while 2 pairs of posterior neurons, in the inferior lateral regions of neuropil. The absence of the medial outgrowth and a stretching of neuropil provided a fan-shaped arrangement of cerebral neurons behind the eyes and on the sides of the pharynx. The three-store arrangement of cerebral neurons is revealed in the presence of a cone-shaped trunk at the anterior end of the body, when all 5 pairs of neurons are connected to each other with longitudinal and transverse processes. The rosette-shaped pharynx is innervated with five pharyngeal nerves, while the barrel-shaped pharynx, with six nerves, each of them with one bipolar neuron. From 2 to 5 pairs of neurons of the L-group were revealed. Homology of the L-group neurons has been confirmed in the order Neorhabdocoela. The total number of CAergic neurons varies from 24 to 29.  相似文献

The presence of brodifacoum residues in possum (Trichosurus vulpecula) livers following routine possum control was investigated. Possums were poisoned in six nature reserves in the Wellington Region, New Zealand, using cereal baits containing 20 mg kg(-1) brodifacoum dispensed from bait stations. Thirty-five surviving possums, and five dead possums were sampled from the reserves following poisoning, and their livers analysed for the presence of brodifacoum. The majority (83%) of samples contained brodifacoum at concentrations ranging from 0.007 mg kg(-1) to 6.2 mg kg(-1). The presence of significant quantities of brodifacoum in possum carcasses following poisoning operations creates potential for secondary and tertiary poisoning of non-target species.  相似文献

In Rhodobacter capsulatus, cobalamin biosynthesis has been shown to occur when the bacteria are grown either aerobically or anaerobically. However, a comparison of the main cobalamin biosynthetic operon found within R. capsulatus would suggest that the encoded proteins belong to the oxygen-dependent pathway for cobalamin biosynthesis, although, significantly, no homologue of the essential mono-oxygenase CobG has yet been detected. Nonetheless, within this main cob operon is found a large open reading frame termed orf663 that is not found in any other cobalamin biosynthetic operon. When overproduced in Escherichia coli, orf663 was found to encode a 90 kDa integral membrane protein. Some of this protein is cleaved within E. coli to give a soluble N-terminal region that can easily be purified and yields a 50 kDa flavoprotein. When expressed in harness with the genes for precorrin-3a synthesis, ORF663 appears to mediate the transformation of precorrin-3a into a new chromophoric compound. Another open reading frame in close proximity to orf663 is termed orf647, and was found to encode a 2Fe-2S ferredoxin-like protein. We suggest that these two proteins may provide an alternative oxygen-independent mechanism for ring contraction within R. capsulatus.  相似文献

Male Codoma ornata produce sounds associated with aggression and spawning activities during the breeding season. Females do not produce sounds. Males most often produced sounds associated with escalated displays of aggression, courtship and the spawning act. C. ornata spawn in crevices, but previously were reported to spawn as egg-clusterers in cavities. Structurally, sounds are low frequency, vary in duration according to context and are not harmonic. Conformational studies on three DNA-oligomers (d(CGCGCGTTAATT), d(CGCGTTAA) and d(CGCGCGTT) in solution by circular dichroism spectroscopy are reported. In low salt solution, all three DNA oligomers exhibit a characteristic B-conformation. However, under the influence of high salt concentration i.e. 5M NaCl, the octamer d(CGCGCGTT) exhibits 'A' conformation whereas the decamer and dodecamer retain B-conformation. On addition of millimolar amount of NiCl2 to the 5M NaCl, solution of oligodeoxynucleotides a B-Z transition is observed in octamer, decamer and dodecamer. However, NiCl2 titrations show that mid point of transition for dodecamer is at 2.25 mM, for decamer is at 13 mM NiCl2 and for octamer is 17 mM at NiCl2. In 60% alcohol all three oligonucleotides remain in the B-conformation. The melting temperatures of oligonucleotides at various salt concentration are also reported. Thermodynamic parameters calculated by melting profile using a two state model show that dodecamer and decamer are most stable in their 5M NaCl, B-form. However, octamer is more stable in its Z form than that of its 'A' form.  相似文献

Summary The pattern of diversity in forty genotypes of clusterbean (Cyamopsis tetragonoloba (L.) Taub) was analysed using the centroid method of factor analysis based on nine characters. Three factors were found to adequately account for most of the intercorrelations in the four environments. Branches, clusters, pods and seed yield/plant could be grouped as productivity factors; days to flowering, maturity and plant height as growth factors; seeds/pod and gum content as economic factor. Interestingly, the constitution of the factors were the same in all the environments. The loadings on the economic factors were relatively unstable, whereas the loadings on productivity and growth factors were stable. It is concluded that productivity and growth factors play a pivotal role towards diversity in clusterbean. The results of factor analysis provides confirmatory evidence of diversity in clusterbean, which was earlier studied by using generalized distance, and thus proves the adequacy of the centroid method in biological investigations.  相似文献

在标本观察和野外调查的基础上,对西藏的嵩草属Kobresia植物进行了分类学修订,共确认了36种和1亚种。更正了6个类群的学名,它们正确的名称是K. esenbeckii,K. fissiglumis,K. gammiei,K. littledalei,K. myosuroides ssp. bistaminata和K. vaginosa。有13个名称,即K. angusta,K. cercostachys var. capillacea,K. clarkeana,K. curticeps var. gyirongensis,K. deasyi,K. glaucifolia,K. hookeri,K. nudicarpa,K. prainii var. elliptica,K. seticulmis,K. stenocarpa,K. stenocarpa var. simplex和K. williamsii处理为异名,其中K. prainii var. elliptica,K. glaucifolia和K. stenocarpa var. simplex为新异名。发现了西藏分布的一个新记录种密穗嵩草K. handel-mazzettii。还收录了最近发表的假钩状嵩草K. pseuduncinoides和阔鳞嵩草K. woodii。此外,重新编制了西藏嵩草属分种和亚种检索表,并较为详细地登记了各类群在西藏的分布地点。  相似文献

Because resistance to parasites usually has a cost for host species, it is theoretically expected that, in case of multi-infection, host immune responses should vary according to the levels of parasite pathogenicity. The crustacean gammarid Gammarus aequicauda is the second intermediate host of 4 trematode species. Three of these parasites always encyst in the abdomen of gammarids and have no particular effect on the host. However, 1 of these species is sometimes able to encyst in the cerebroid ganglia of the gammarid and strongly alter its behavior in a way that increases its predation risk by aquatic birds, the definitive hosts. In accordance with the hypothesis that the level of parasite pathogenicity influences the likelihood and the degree of host reaction, cases of melanization in our gammarid collection almost exclusively concern the cerebral metacercariae. Our results also indicate that this melanization is able to cancel the behavioral alterations induced by the parasite, suggesting that the cause of the manipulation is not the physical presence of metacercariae in the brain.  相似文献
