Cryopreservation of turbot (Scophthalmus maximus) spermatozoa   总被引:4,自引:0,他引:4  
The aim of this study was to develop a method for cryopreserving turbot semen and to compare sperm motility characteristics, metabolic status and fertilization capacity of frozenthawed and fresh semen. The best results were obtained when spermatozoa were diluted at a 1:2 ratio with a modified Mounib extender, supplemented with 10% BSA and 10% DMSO. For freezing sperm samples, straws were placed at 6.5 cm above the surface of liquid nitrogen (LN) and plunged in LN. The straws were thawed in water bath at 30 degrees C for 5 sec. Use of this simple method resulted in a 60 to 80% reactivation rate of the thawed spermatozoa. Although the percentage of motile spermatozoa in the frozen-thawed semen samples was significantly lower than in fresh semen, spermatozoa velocity and respiratory rate remained unchanged. The process of cryopreservation significantly decreased intracellular ATP content. The fertilization rate of frozen-thawed spermatozoa was significantly lower than that of fresh spermatozoa, but it increased with sperm concentration.  相似文献

A consensus microsatellite-based linkage map of the turbot (Scophthalmus maximus) was constructed from two unrelated families. The mapping panel was derived from a gynogenetic family of 96 haploid embryos and a biparental diploid family of 85 full-sib progeny with known linkage phase. A total of 242 microsatellites were mapped in 26 linkage groups, six markers remaining unlinked. The consensus map length was 1343.2 cM, with an average distance between markers of 6.5 +/- 0.5 cM. Similar length of female and male maps was evidenced. However, the mean recombination at common intervals throughout the genome revealed significant differences between sexes, approximately 1.6 times higher in the female than in the male. The comparison of turbot microsatellite flanking sequences against the Tetraodon nigroviridis genome revealed 55 significant matches, with a mean length of 102 bp and high sequence similarity (81-100%). The comparative mapping revealed significant syntenic regions among fish species. This study represents the first linkage map in the turbot, one of the most important flatfish in European aquaculture. This map will be suitable for QTL identification of productive traits in this species and for further evolutionary studies in fish and vertebrate species.  相似文献

A surface-spreading synaptonemal complex (SC) technique was used to analyse spermatocytes and oocytes of triploid turbot (Scophthalmus maximus) in order to visualise the process of chromosome synapsis. The most conspicuous characteristic of triploid oocytes is that, in the trivalents, the lateral elements of the SC were frequently associated in threes, either completely along the length of the trivalent, or partially, forming a variety of forked structures. In these nuclei, synapsis usually occurred among homologous chromosomes and the number of bivalents observed was significantly higher than that expected under the assumption of random chromosome association among all partners. However, the frequency of trivalents was very low in triploid spermatocytes, triple synapsis being also scarce. In these nuclei chromosomes that were excluded from homologous synapsis become engaged in random SC formation, and, therefore a considerable number of non-homologous associations are produced. The causes of the synaptic differences observed in triploid males and females of turbot and their possible relation to the sterility displayed by these animals are discussed.  相似文献

The histopathology of biliary hyperplasia and renal calcinosis, associated with infections of two parasites, Myxidium incurvatum and Rhabdospora thelohani , is described in turbot, Scophthalmus maximus (L.), from Scottish marine fish farms and wild populations. Liver and kidney lesions were divided into four, and parasite infections into five grades of severity. A few of the fish examined were not parasitized, but every one showed some evidence of pathological change, even the apparently normal individuals from all populations. Although there is a positive association between the parasitic and pathological data, parasitism is not considered to be the principal cause of either the hepatic or renal lesion; the evidence suggests rather a secondary opportunist role for the parasites. The general severity of the condition in captive stocks suggests that the cause may lie in the fundamentals of present techniques of turbot husbandry, but no specific agent has been identified. A case is put forward for considering Rhabdospora to be a parasite rather than a specialized host cell.  相似文献

Unravelling the factors shaping the genetic structure of mobile marine species is challenging due to the high potential for gene flow. However, genetic inference can be greatly enhanced by increasing the genomic, geographical or environmental resolution of population genetic studies. Here, we investigated the population structure of turbot (Scophthalmus maximus) by screening 17 random and gene‐linked markers in 999 individuals at 290 geographical locations throughout the northeast Atlantic Ocean. A seascape genetics approach with the inclusion of high‐resolution oceanographical data was used to quantify the association of genetic variation with spatial, temporal and environmental parameters. Neutral loci identified three subgroups: an Atlantic group, a Baltic Sea group and one on the Irish Shelf. The inclusion of loci putatively under selection suggested an additional break in the North Sea, subdividing southern from northern Atlantic individuals. Environmental and spatial seascape variables correlated marginally with neutral genetic variation, but explained significant proportions (respectively, 8.7% and 10.3%) of adaptive genetic variation. Environmental variables associated with outlier allele frequencies included salinity, temperature, bottom shear stress, dissolved oxygen concentration and depth of the pycnocline. Furthermore, levels of explained adaptive genetic variation differed markedly between basins (3% vs. 12% in the North and Baltic Sea, respectively). We suggest that stable environmental selection pressure contributes to relatively strong local adaptation in the Baltic Sea. Our seascape genetic approach using a large number of sampling locations and associated oceanographical data proved useful for the identification of population units as the basis of management decisions.  相似文献

The preparation of fish oil concentrates containing only ( n -3) polyunsaturated fatty acids (PUFA) with different ratios of 20:5 ( n -3)/22:6 ( n -3) is described. Three groups of turbot were maintained on different diets containing: 1, 10% of the dry weight of the diet as natural fish oil, equivalent to 2.5% ( n -3) PUFA and 0–23% ( n -6) PUFA; 2, 10% of the dry weight of the diet as palmitic acid, i.e. no PUFA; 3, 8–7% palmitic acid and 1–3% of the dry weight as ( n -3 PUFA and negligible ( n -6) PUFA. Only the fish on the diet containing natural fish oil showed significant growth over a 15-week period. In addition there were high mortalities on the two experimental diets (2 and 3). Changing the ratio of 20:5 ( n -3)/22:6 ( n -3) from 13–8 to 2–2 in the diet containing 1 3% (n-3) PUFA and negligible ( n -6) PUFA markedly decreased the mortalities. Fish fed the two experimental diets (2 and 3) developed gross changes in gill structure involving the disappearance of chloride cells, a 'sloughing off' of the epithelium along the primary and secondary filaments and an accumulation of cellular material in the inter-lamellar spaces. The tissue ultimately disintegrated to leave a skeleton of connective tissue and a mass of cellular material in the inter-lamellar spaces. It is concluded that 22:6 (n-3) is an essential fatty acid for turbot and that the gill epithelium is a sensitive indicator of this deficiency in this species.  相似文献

Ferritin is an evolutionarily conserved protein that plays an important role in iron storage and detoxification. In this study, the gene encoding a ferritin H subunit homologue (SmFer1) was cloned from turbot (Scophthalmus maximus) and analyzed at the expression and functional levels. The open reading frame of SmFer1 is 534 bp and preceded by a 5′-untranslated region that contains a putative Iron Regulatory Element (IRE). The deduced amino acid sequence of SmFer1 shares extensive sequence identities with the H ferritins of a number of fish species and contains the ferroxidase center that is preserved in ferritin H subunits. Examination of tissue specific expression indicated that SmFer1 expression was most abundant in muscle, liver, and blood. Experimental infection with bacterial pathogens induced significant induction of SmFer1; however, the magnitudes of induction effected by Gram-negative pathogens were much higher than that induced by Gram-positive pathogen. Consistently, lipopolysaccharide (LPS) challenge drastically augmented SmFer1 expression. In addition to bacterial pathogens and LPS, poly(I:C) also induced a strong but transient induction of SmFer1 which differs in profile from those induced by bacterial pathogens. In vitro iron-chelating analysis showed that recombinant SmFer1 purified from Escherichia coli was able to bind ferrous iron in a concentration-dependent manner. To examine whether SmFer1, with its iron-chelating capacity, could have any effect on the infection of bacterial pathogens, purified recombinant SmFer1 was subjected to bacteriostatic analysis and proved to be able to inhibit the growth of the fish pathogen Listonella anguillarum which enhanced SmFer1 expression upon infection. Taken together, these results suggest that SmFer1 is likely to play a role in both iron storage and immune defense against microbial infections.  相似文献

In order to extend our knowledge of the presence of bacteria in hatcheries and their influence on rearing performance, the aerobic and facultative bacterial flora associated with farmed turbot larvae were studied in relation to the microflora of the water and diets. A settlement of specific groups of bacterial populations was found in the gut of the larvae. A clear succession of bacterial phenotypes was also observed from day 1 to day 90 post-hatching. Oxidative Gram-negative rods were predominant at the initial stages, whereas some phenotypes of Vibrio were frequent at the final stages. A high heterogeneity of Vibrio species was observed in the intermediate period when the highest mortalities of turbot larvae occur.  相似文献

In this study, autometallography and immunohistochemistry were used to localize and quantify cadmium and metallothionein (MT) levels, respectively, in cellular compartments of turbot liver on exposure to cadmium for 7 days and further depuration treatment for 14 days. Metals weakly bound to proteins (i.e. MTs) in hepatocyte lysosomes were visualized as black silver deposits (BSDs) using a light microscope. With the aid of a newly developed immunohistochemical procedure, MTs were localized and semi-quantified in both the cytosolic and the lysosomal compartments of hepatocytes. The BSD extent in the lysosomes of hepatocytes increased significantly as a result of cadmium exposure. This response was evidenced after 1h. Further, a progressive increase in the volume density of BSDs occurred up to the seventh day. Total MT immunohistochemical levels increased at a lower rate, starting after 1 day of cadmium exposure. BSD extent values recovered after depuration, whilst MT levels remain unchanged. It is possible that the detoxification rate of metals via lysosomes was diminished, whilst MT levels remained unchanged, at least after 14 days of depuration. It can be concluded that autometallography and MT immunohistochemistry are good tools for clarifying metal and metal-MT trafficking routes in hepatocytes, and also that BSD extent and MT immunohistochemical levels in the lysosomes and cytosol of fish hepatocytes can be considered to be useful biomarkers of metal exposure.  相似文献

Regulatory volume decrease (RVD) following hyposmotic stimulation was studied in isolated turbot, Scophthalmus maximus, hepatocytes. Exposed to a reduced osmolality (from 320 to 240 mosm kg−1), cells first swelled and then exhibited a RVD. Volume regulation was significantly inhibited in presence of NPPB, 9-AC, acetazolamide, DIDS and barium. Taken together, these results could suggest that RVD operated via separate K+ and Cl- channels and probably Cl-/HCO 3 exchanger in turbot hepatocytes. The K+/Cl- cotransporter could also be involved as furosemide and DIOA strongly inhibited the process whereas NEM, a K+/Cl- cotransporter activator, added under isosmotic conditions, led to cell shrinkage. RVD in turbot hepatocytes appeared also to depend on proteins p38 MAP kinase and tyrosine kinase but not on proteins ERK 1/2. Arachidonic acid and leukotrienes could also be involved since inhibition of synthesis of both these compounds by quinacrine and NDGA, respectively, inhibited the volume regulation. Likewise, Ca2+ has been proved to be an essential messenger as RVD was prevented in absence of Ca2+. Finally, this work provides bases for novel studies on cell volume regulation in marine teleosteans.  相似文献

Immunomodulatory effects of nisin in turbot (Scophthalmus maximus L.)   总被引:3,自引:0,他引:3  
In the present work, the effect of nisin on the non-specific immune response of turbot (Scophthalmus maximus L.) leukocytes has been studied both in vitro and in vivo. The head kidney macrophage chemiluminescent (CL) response was significantly increased with intermediate doses of nisin (2.5 and 0.025 micro g ml(-1)) whilst the higher dose (25 micro g ml(-1)) significantly decreased the response after 24h incubation. When the incubation time was extended to 72 h, significant differences between doses were observed and the lower nisin concentration (0.025 micro g ml(-1)) appeared to be the optimum dose for increasing the CL response. The phagocytic activity of HK macrophages was also affected by in vitro nisin treatments. Nisin at 0.25 micro g ml(-1) and 0.025 micro g ml(-1) significantly stimulated the response after 24 and 72 h incubation respectively. Nitric oxide (NO) production by HK macrophages was not influenced by any nisin concentration employed for 24 or 72 h incubationsIn vivo, one week post injection, a slightly but non-significant stimulation of the CL response was observed with the lowest nisin concentration (0.0025 micro g fish(-1)). NO in serum and serum antibacterial index were not significantly affected by nisin treatments. On the other hand, lysozyme concentration in serum was significantly augmented with the lowest nisin dose (0.0025 micro g fish(-1)).The antibacterial effect of nisin against the fish pathogenic bacteria Carnobacterium piscicola (CECT 4020) was also demonstrated in vitro.  相似文献

BACKGROUND INFORMATION: ATP is released from many cell types exposed to hypo-osmotic shock and is involved in RVD (regulatory volume decrease). Purinergic signalling events have been extensively investigated in mammals, but not in marine teleosteans. RESULTS: The effect of hypo-osmotic shock on ATP release was examined in isolated hepatocytes from turbot (Scophthalmus maximus), a marine flatfish. Hypo-osmotic stress (240 mOsm x kg(-1)) induced a significant increase in ATP efflux, and was inhibited by a potential CFTR (cystic fibrosis transmembrane conductance regulator) inhibitor, glibenclamide, but not by the MDR1 (multidrug resistance 1) P-glycoprotein inhibitor, verapamil. ATP efflux could be a cAMP-dependent process, as IBMX (isobutylmethylxanthine) and forskolin triggered the process under iso-osmotic conditions. Protein kinases, including protein kinase C, could also be involved, as staurosporine and chelerythrine inhibited the mechanism. Calcium could contribute to ATP efflux as ionomycin, a calcium ionophore, elicited a rapid release under iso-osmotic conditions, and chelation using EGTA abolished ATP release under hypo-osmotic conditions. RVD was partially abolished by apyrase, an ATP scavenger, and suramin, a purinoceptor antagonist. Moreover, hypo-osmotic shock induced a rise in intracellular calcium which could be involved in RVD. Since extracellular ATP triggered an increase in cellular free-calcium content under iso-osmotic conditions, our results could indicate that hypo-osmotic-induced ATP efflux contributes to RVD in turbot hepatocytes by stimulating purinergic receptors, which may lead to activation of a calcium signalling pathway. CONCLUSIONS: These data provide the first evidence of volume-sensitive ATP signalling for volume maintenance in a marine teleost fish cell type.  相似文献

The purpose of this study was to investigate whether meat from blue mussels (Mytilus edulis) harvested from offshore structures could be used as an alternative protein source in aquaculture fish feed for turbot (Scophthalmus maximus). Two feeding experiments with turbot were conducted in an initial experiment to test the applicability of mussel meal as a fishmeal replacement and/or supplement in turbot feed. In experiment A, the welfare, acceptance, and digestibility of feedstuff with 100%, 50% and 0% of mussel meal replacement for the fishmeal were tested for 8 weeks. In experiment B that followed, the convenience ratio of mussel meal protein in turbot feed for optimum growth was examined. Feedstuff with 25%, 10% and 0% of mussel meal were also fed for 8 (+6) weeks. Growth rates of fish were measured every 2 weeks and their health condition (liver somatic index) determined at the beginning and end of the experiments. Experiment A revealed that a replacement of fishmeal with either 100% or 50% mussel meal resulted in depressed growth (SGR: 1.31 ± 0.04 with 100% mussel meal; 1.54 ± 0.04 with 50% mussel meal; 1.71 ± 0.07 with 0% mussel meal). Experiment B revealed that a fishmeal replacement with 10% or 25% of mussel meal did not reduce growth nor did it have a negative effect on the health of the tested turbot. The study revealed that mussel meal has a high potential to serve as a supplement or as a fishmeal replacement in feed for turbot raised in aquaculture systems. Thus, its use in commercial aquaculture should be given future consideration.  相似文献

Juvenile turbot,Scophthalmus maximus (L.), were exposed to 0.58 μg 1−1 Aroclor 1254 in seawater, to sediments containing 100, 60 and 1 ppm or fed with cockle containing 20 ppm PCB (polychlorinated biphenyls). Concentration factors for liver and muscle were 104 and 103, respectively, for uptake of PCB from seawater. Contamination of muscle was similar to that of sediments containing 1 and 60 ppm PCB to which turbot were exposed, but less than the 20 ppm in their experimental diet. Contamination of flatfish in the North Sea area is compared with the levels of PCB in the flounder,Platichthys flesus (L.), in the River Thames and predictable values for uptake of PCB from different pathways discussed.  相似文献

Pleiotropic resistance driven by transport proteins constitutes a very ubiquitous protection mechanism against natural or synthetic toxic compounds. The multidrug (MDR) or multixenobiotic (MXR) system has been identified in many different species, and may be used as a biomarker for pollution assessment. Here we report the existence of a gene encoding a MXR-related protein in a benthic fish species, the turbot Scophthalmus maximus, and its constitutive expression in several tissues. A 433bp cDNA fragment has been cloned by RT-PCR. The deduced amino-acid sequence shares close to 80% homology with class I or class II mammalian MDR proteins. This cDNA corresponds to a major mRNA of 5.6 kb and encodes a protein having an apparent molecular weight of 83 kDa. Constitutive expression levels assessed by semi-quantitative RT-PCR and Western blot, revealed that the kidney and the brain, and to a lesser extent, the heart, gills and intestine, are the organs which contain the highest amount of both MXR mRNAs or proteins. This tissue specific expression suggests a role for the identified mechanism in protection against endogenous or exogenous toxic compounds.  相似文献

The present study was to investigate the interactive effect of ammonia and crowding stress on ion-regulation and expression of immune-related genes in juvenile turbot (Scophthalmus maximus). The fish were exposed to four total ammonia nitrogen (0, 5, 20, 40 mg/L TAN) and two stocking density. After 96 h of exposure, blood, gill, and liver samples were collected to measure biochemical parameters and mRNA levels of immune-related genes. The results showed that co-exposure to high TAN (20 and 40 mg/L) and high density significantly increased plasma sodium (Na+), gill Na+/K+-ATPase activity and mRNA levels. Following individual and combined exposure to high TAN and high density, the heat shock protein 70 (HSP 70), HSP 90, tumor necrosis factor-α (TNF-α), and interleukin-1β (IL-1β) genes expression were obviously higher than their control. Conversely, the lysozyme (LZM) and hepcidin mRNA levels were down-regulated in liver of fish exposed to high TAN alone and combination of high TAN-density. Moreover, glutathione S-transferase (GST) mRNA levels significantly increased in treatments with individual high TAN and high density, but decreased in high TAN-density co-exposed fish. Overall, ion homeostasis and immune status were adversely influenced in high exposed turbot under high density.  相似文献

Mean values of the composition of the seminal fluid of 30 turbots were: osmotic pressure =306 mosml−1, pH =7–31, total protein =8.8mg ml1, Na+=133.0 mmoll1, K+=3.80mmoll−1, Cl=129mmol l−1. The rounded nucleus, the reduced middle piece and the typical '9 + 2' structure of the flagellum mean that the spermatozoon of turbot can be considered to be of a primitive type.  相似文献

Young turbot (1-20 g) were maintained for not less than 14 weeks on three diets: (1) a control diet containing normal amounts of polyunsaturated fatty acids (PUFA); (2) a diet totally deficient in PUFA; (3) a diet deficient in the (n-6) series of PUFA but containing (n-3) PUFA. At 14 weeks the fatty acid compositions of the phospholipids from liver, gut, gills and muscle were analysed. Large changes in the amounts of PUFA in the phospholipids were found. Fish maintained on the totally PUFA deficient diet 2 had retained arachidonic acid, 20:4(n-6), and docosahexaenoic acid, 22:6(n-3), at the expense of eicosapentaenoic acid, 20:5(n-3). Fish maintained on the (n-6) PUFA-deficient diet (3) contained decreased amounts of 20:4(n-6) and 22:6(n-3) while retaining 20:5(n-3). In all cases phosphatidylinositol had the lowest n-3/n-6 ratios. These results are discussed in terms of PUFA function.  相似文献
