Possible candidates for hemic stem cells were studied in a quantitative investigation of the bone marrow of growing guinea pigs. The stem cell candidates were classified as lymphoid cells of types I-IV, roughly corresponding to small lymphocytes, large lymphocytes, micromyeloblasts and myeloblasts of other authors. The cellular content of the bone marrow of guinea pigs of three different ages was determined. All types of nucleated cells per femur were found to increase with age. When the cellular content was related to the body weight, a peak number of lymphoid cells was found in the intermediate age group with the exception of the lymphoid cells of type III, which reached its peak in the oldest animals. This cell type has been suggested by Dicke et al. as the probable stem cell responsible for colony formation.  相似文献

Abstract The blood cells of a solitary ascidian, Halocynthia roretzi, were examined by electron microscopy (EM) with reference to their appearance by light microscopy (LM). In addition, their movement and stainability by vital dyes was observed by phase-contrast microscopy, and their stainability by Giemsa was also examined. Nine cell types were recognized: vacuolated cells, hyaline amoebocytes, small amoebocytes, granular amoebocytes, macrogranular cells, globular cells, lymphocyte-like cells, large basophilic cells and large granular cells. Vacuolated cells were found to possess various numbers of vacuoles containing strongly electron-dense materials and could be divided into at least three subgroups. Granular amoebocytes contained microfilaments and many granules of uniform size. Hyaline amoebocytes and small amoebocytes seemed to be specialized as phagocytes. Macrogranular cells and globular cells were not well characterized. In the blood of adult individuals, hemoblasts were rarely found, although lymphocyte-like cells were present. Each of two large cells, large basophilic cells and large granular cells, possessed novel granules or vacuoles, whose functions remain to be elucidated. The possible functions and relationships of these cells among various ascidian species are discussed.  相似文献

The cortex of enlarging thymic lobes from adult haemorrhaged Quelea quelea were found to be similar to those of wild birds where the thymic enlargement was occurring naturally. A detailed stereological analysis of cells broadly designated as lymphoid, and the construction of models to account for the results, indicates that the enlarging thymic lobe contains both large and small blast cells, a heterogenous group of medium lymphocytes, erythroid cells, and two types of very small lymphocytes. The distinction between early erythroid cells and some lymphocytes, despite this detailed analysis is very difficult, but it is possible in enlarging thymic lobes that up to 42% of the lymphoid cells may have erythroid characteristics.  相似文献

A radioautographic immunolabeling technique has been developed to detect pre-B cells bearing cytoplasmic mu chains among populations of bone marrow lymphoid cells identified by conventional hematologic stains. 125I-Anti-mu antibody was applied either to fixed marrow smears, labeling total mu chains both in the cytoplasm (c mu) and at the cell surface (s mu), or to cell suspensions, labeling s mu alone. In stained radioautographs the incidence of c mu+ s mu- pre-B cells was derived both indirectly by subtracting values for s mu+ cells from those for total mu+ cells of various sizes in normal mice and directly by the total mu chain labeling in mice depleted of s mu+ cells by anti-IgM treatment in vivo. Binding specificity was demonstrated by the displacement of labeling by nonradioactive anti-mu antibody. The c mu+ s mu- cells showed a bimodal size distribution. They accounted for 40% of the large lymphoid cells and 30% of the small lymphocytes in the marrow. A further 50% of the small lymphocytes were B lymphocytes (s mu+) and 8% were T lymphocytes (Thy 1.2+). Thus, the technique demonstrates the presence of c mu+ s mu- pre-B cells among both proliferating large lymphoid cells and nondividing small lymphocytes, as classically defined in marrow smears. In addition, the results reveal a broad size distribution of mu- lymphoid cells, including a subset of small lymphocytes which lack c mu, s mu, and Thy 1.2 and thus cannot be assigned to either B or T lineage by these criteria. The findings suggest that in addition to B cells the marrow may produce other types of lymphoid cells, yet to be defined.  相似文献

This paper describes the fine structure of epithelial vesicles as observed in the thymus of the chicken. Three types of vesicles are found. The largest is made up of a stratified columnar epithelium containing lymphoid cells. The lumen contains a fairly light material and cell debris. The lumen-bordering cells are columnar and their microvilli are sometimes developed to such an extent as to appear as a brush border. The wall of the vesicles contains lymphocytes and granular cells with dense granules 100-500 nm in diameter. Smaller vesicles are found which contain no lymphoid cells. They are made of a simple or stratified epithelium containing typical mucous cells, and columnar cells similar to the bordering cells of the large vesicles. The third type is an intracytoplasmic vesicle in epithelial reticular cells, which differs from usual vacuoles by the presence of microvilli and its often unusually large size. The morphology of the first 2 types of vesicles indicates a secretory capability, the function of which is unknown. Data from the literature suggest that the third type produces the factor(s) conferring cellular immunological competence.  相似文献

Lymphokine activated killer (LAK) cells have been utilized as a useful tool in cancer adoptive immunotherapy. The lineage origin of this population has always been controversial since it shares phenotypic markers with both myelomonocytic cells and T lymphocytes. Recently we described a new monoclonal antibody (MoAb), termed LAK1, which recognizes a 120 Kd surface molecule expressed on human large granular lymphocytes (LGL) and LAK precursors and effectors. LAK1 MoAb defines two different populations of positive cells amongst peripheral lymphocytes: the first subset (20%), represented by brightly stained cells, belongs to the non T-LGL population, whereas the second subset (30%) displays low fluorescence intensity and was partially composed of T lymphocytes. More interestingly, LAK1 is shared by some other cell types, such as monocytes and vascular endothelial cells. Immunohistochemical staining performed on muscle, endometrium and lymphoid or other non lymphoid tissues shows that LAK1 antigen is selectively expressed by the reticuloendothelial system.  相似文献

The interaction between human natural killer (NK) cells and NK-susceptible target cells, as well as the mechanism involved in target cell lysis, were studied with scanning electron microscopy (SEM). Low density human peripheral blood lymphocytes, highly enriched with large granular lymphocytes (LGL), were used as effector cells, and K562-cells were used as NK-susceptible target cells. The surface features of LGL/NK cells were examined under SEM. In the area of interaction, NK/target-cell conjugates showed microvilli and/or filipodia, and extensive areas of intercellular contact. In addition, the effector cells in some NK/target-cell conjugates were polarized toward the target cell. Changes in target cell surface features included loss of microvilli, large surface blebs and the appearance of small pore-like lesions on the cell membrane. Our findings show that target cell lysis occurred by apoptosis and plasma membrane lesions analogous to those seen during complement-mediated cytotoxicity.  相似文献

Summary Lymphocyte and macrophage subpopulations and the stroma of mucosa-associated lymphoid tissue in the nasal cavity of the rat were examined by application of immunohistochemical and enzyme histochemical methods to cryostat sections. Nasal-associated lymphoid tissue was composed of a loose reticular network with lymphocytes and macrophages, covered by epithelium. The epithelium was infiltrated with B cells, T helper (W3/13-positive) and T suppressor/cytotoxic or large granular cells (OX8-positive), ED1-positive macrophages and Ia-positive cells. The B cell areas were populated by B cells, immunopositive for surface IgM or IgG. B cells with surface IgA or IgE were rare. Germinal centres were found infrequently. T helper cells were scattered throughout the B cell area. A few ED1-positive macrophages and ED5-positive follicular dendritic cells were observed. Strong Ia staining (mostly of B cells) was found in this area. The T cell areas contained T helper and T suppressor/cytotoxic cells in about equal amounts, and numerous ED1-positive macrophages. ED1 staining was also found in the subepithelial area. Numerous ED1-, ED2- and ED3-positive macrophages were found in the border between the lymphoid mass and the surrounding connective tissue. A few non-lymphoid cells showed weak acid phosphatase or non-specific esterase activity. The morphological observations suggest that nasal-associated lymphoid tissue plays an important role in the first contact with inhaled antigens.  相似文献

A monoclonal antibody specific for rat intestinal lymphocytes   总被引:4,自引:0,他引:4  
A monoclonal antibody, RGL-1, was produced by fusion of NSI myeloma cells with spleen cells of a mouse immunized with isolated rat intraepithelial lymphocytes (IEL). SDS-PAGE analysis revealed that RGL-1 precipitated two major noncovalently bound chains of about m.w. 100,000 and 125,000, and a minor component of m.w. 200,000. Examination of both tissue sections and isolated cells indicated that RGL-1 stained the majority of the lamina propria lymphocytes and IEL but only very few cells (less than 2%) in the lymphoid organs and small numbers of lymphocytes in other mucosae. In the small intestine, RGL-1 stained lymphocytes with the helper (W3/25) as well as the cytotoxic/suppressor (OX8) phenotype. The antibody reacted with 95% of the granular IEL but with less than 0.1% of the blood large granular lymphocytes. Although mature IgA plasma cells in the lamina propria were RGL-1-, some large IgA-containing cells were weakly positive. In the gut-associated lymphoid tissues (GALT), studies combining immunofluorescence and autoradiography indicated that 56 and 73% of rapidly dividing cells of mesenteric lymph nodes and of thoracic duct lymph (TDL) stained with RGL-1, respectively. In addition, 90 to 100% of the IgA-containing blasts of MLN and 75% of those of TDL were labeled by RGL-1. In contrast, rapidly dividing cells of spleen and of peripheral lymph nodes did not stain with RGL-1. Because RGL-1 can be demonstrated on both intestinal lymphocytes and their immediate precursors in the GALT, its expression may be related to the homing of lymphocytes into the gut mucosa.  相似文献

The endogenous inhibitors of cysteine proteinases, cystatin A and cystatin B, have been localized in sections of human liver by indirect immunofluorescence microscopy. Cystatin A was localized in neutrophils and cystatin B was found in lymphocytes. Both types of cell were present in the blood vessels and within the sinusoids between the parenchymal cells. Cystatin A was also found in some of the parenchymal cells surrounding the blood vessels but, in contrast to the uniform fluorescence of the leucocytes, the staining in the parenchymal cells was granular and rather irregular.  相似文献

Monoclonal antibodies reactive with T cells, T cell subsets, B cells, monocytes, and natural killer cells were used to characterize the nature of mucosal lymphocytes in the human small intestine by application of the immunoperoxidase technique to tissue sections for light and electron microscopic examination. In addition, for comparison, peripheral blood mononuclear cells (PBL) were studied by immunoelectron microscopy. Most of the intraepithelial lymphocytes (IEL) were T cells (Leu-1+, T3+) and expressed the phenotype associated with cytotoxic/suppressor T cells (Leu-2a+, T8+). In contrast, a majority of T lymphocytes in the lamina propria expressed the phenotype associated with helper/inducer T cells (Leu-3a+, T4+). These observations confirm and extend the findings previously reported. In addition, a small number of cells in the lamina propria with the ultrastructural features of macrophages were found to react with anti-Leu-3a and anti-T4 antibodies. Although many IEL contained cytoplasmic granules and had ultrastructural features similar to those of circulating granular lymphocytes, none of these cells reacted with anti-Leu-7 (HNK-1), anti-T10, or anti-M1 antibodies. This suggests that IEL may not be related to circulating large granular lymphocytes, which are Leu-7+, T10+, M1+ and are associated with natural killer activity. Not only Leu-7+ PBL, but T8+, T4+, or T3+ mucosal lymphocytes or PBL also may contain cytoplasmic granules. Therefore, the cytoplasmic granules are not restricted to one cell type, in particular, to Leu-7+ cells.  相似文献

Hemocytes of Mytilus edulis were examined cytologically and cytochemically. On the basis of structure, staining reactions, and phagocytic behavior, they were divided into two main groups: basophilic hemocytes and eosinophilic granular hemocytes (granulocytes). The basophilic cells were further divided into small lymphocytes and larger phagocytic macrophages reactive for lysosomal hydrolases. Mitosis was observed in granulocytes and in small lymphoid cells, believed to be the stem cells for the basophilic cell line. A few cells appeared to be intermediate between lymphocytes and small granulocytes. Macrophages were the main cell type involved in the clearance of injected carbon particles. However, granulocytes did show some phagocytic activity. Brown cells displaying apparent amoebocytic behavior were found to contain Fe3+ and Pb2+ in cytoplasmic inclusions, some of which were also reactive for β-glucuronidase and glucosaminidase. These cells appear to have a separate origin from the hemocytes.  相似文献

Abstract. DNA labelling by [3H]thymidine and the sandwich radioimmunolabelling method were used to characterize marrow lymphoid cells and to study the kinetics of production and maturation of small lymphocytes in the bone marrow of adult mice. Marrow lymphoid cells consisted of non-proliferating small lymphocytes, 30–40% of which had detectable surface immunoglobulin (SmIg), and proliferating large lymphoid cells lacking SmIg. Double-labelling experiments employing [3H]thymidine in vivo followed by sandwich radioimmunolabelling in vitro indicated that marrow small lymophocytes lack detectable SmIg when they are formed but develop SmIg within the first few days after production. Marrow lymphocytopoiesis includes; (1) praliferation of large lymphoid cells, which are presumptive small lymphocyte progenitors, which have a cell cycle time of 14–15 hr, and (2) a 3–5-day intramyeloid stage when many newly formed small lymphocytes undergo maturational changes towards the B cell lineage.  相似文献

通过相差显微镜和电镜观察,根据中华绒螯蟹血细胞胞质内有无颗粒以及颗粒大小、染色反应、折光性和形成方式的特点,血细胞分为胞质内无颗粒的无颗粒细胞、胞质内只有具折光性和呈淡红色反应大颗粒的大颗粒细胞、胞质内只有无折光性和呈淡蓝色染色反应小颗粒的小颗粒细胞以及胞质内同时具有大颗粒和小颗粒二种颗粒特性的大小颗粒中间型细胞.小颗粒的形成方式是高尔基体成熟面小泡脱离后直接成为小颗粒,而大颗粒的形成方式是高尔基体成熟面小泡脱离后,数个小泡逐渐聚集成蜂窝状大颗粒,进一步发育成熟为均质大颗粒.实验结果表明:三种有颗粒的细胞是互相独立的,可能分别由无颗粒细胞分化而成.    相似文献

A morphologic study of bronchus-associated lymphoid tissue in turkeys   总被引:1,自引:0,他引:1  
Bronchus-associated lymphoid tissue (BALT) in normal turkeys of ages 1 day and 1, 2, 3, 4, 8, and 18 weeks was examined by light microscopy and by scanning and transmission electron microscopy. Turkey BALT resembled other mucosa-associated lymphoid tissues; it was made up of a population of lymphocytes covered by a specialized epithelium different from typical pseudostratified ciliated columnar bronchial epithelium. There were distinct age-related differences in BALT structure. Bronchus-associated lymphoid nodules were larger and more numerous in older turkeys. In 1-day- to 2-week-old turkeys, the primary cell type of BALT epithelium was nonciliated cuboidal; in 2-week old turkeys it was squamous; and in turkeys older than 4-weeks of age, the epithelium was primarily ciliated columnar. In 1- to 4-week old turkeys, large numbers of intraepithelial lymphocytes disrupted the normal organization of the epithelium. In older turkeys, epithelial and lymphoid cells were in discrete compartments separated by connective tissue. Lymphocytes in 1-day-old turkeys were found in loose aggregates around venules and within the epithelium. In 1-week old turkeys, lymphocytes were organized into compartments of morphologically similar cells. By 3-weeks of age, lymphocytes were present in distinct germinal centers. Epithelial cells of BALT did not have large numbers of apical vesicles and thus were not structurally specialized for antigen uptake by endocytosis. However, the epithelial barrier appeared to be disrupted over lymphoid nodules, suggesting that antigen would be readily available to lymphocytes and phagocytes in BALT. Age-related differences in turkey BALT structure may have functional consequences with respect to the respiratory immune response.  相似文献

Summary In the present study attention was focussed on several lymphoid subpopulations and specific stationary cells of the human tonsilla palatina. They were labeled at the light- and electron-microscopic levels by means of monoclonal antibodies to cell surface antigens. Cells resembling interdigitating cells (IDC-like cells) within the crypt epithelium and the interdigitating cells in the parafollicular T-cell region express the HLA-DR antigen. This fact suggests a relationship between these two populations of cells. Both cell types were frequently found in close contact to T-helper cells labeled with Anti-Leu 3a. This fact is discussed as a confirmation of earlier suggestions that the tonsillar crypt epithelium serves as T-cell region. Cytotoxic/ suppressor-T cells (OKT 8 +) and Leu 7-positive cells do not appear to contact interdigitating cells. Anti-Leu 7 is a monoclonal antibody, that defines a differentiation antigen shown to be selectively expressed on human natural killer cells (NK-cells). With the use of the immuno-electron-microscopic labeling method it was possible to analyze the ultrastructure of this lymphoid subpopulation. Two morphologically distinguishable subtypes of Leu 7-positive cells populate different microenvironments: The Leu 7-positive large-granular lymphocyte was predominantly found in the crypt epithelium, while numerous Leu 7-positive cells located in the germinal centers had the appearance of small lymphocytes. This finding is discussed in favour of distinct phenotypes representing different stages in a differentiation pathway of the maturing NK-cell: Small Leu 7-positive lymphocytes in the germinal centers are supposed to be functionally inactive precursors, and only the Leu 7-positive large granulated lymphocytes in the crypt epithelium may represent differentiated active NK-cells. Immune lymphocytes sorbed on the surface of the target cells were characterized during the period of the first three hours of combined incubation by the presence of the electron-dense matrix, abundance of mitochondria and lipids; small lymphocytes had disseminated ribosome organized into polysomes in the medium lymphocytes forming individual cysterns of the granular endoplasmic reticulum in the large lymphocytes, this indicating active protein synthesis by these cells. There were also revealed cells of plasmatic type. Cells incubated with the PHA for one hour represented a homogenous population of small lymphocytes of the same size as the clear cytoplasm containing free ribosomes and individual mitochondria. The proportion of the medium lymphocytes and the blasts increased with increase of the incubation period. These are cells with the clear cytoplasm freely disseminated polyribosomes in which no developed granular endoplasmic reticulum was sometimes revealed. The presence of two types of cells whose ultrastructure reflected their functional characteristics is discussed.  相似文献

Forward angle light scattering of two different wavelengths by cells in a flow cytometer was used to investigate physical differences between lymphocytes of different lineage, functional subclass and developmental stage. Correlation of the ultraviolet (UV: 351 nm and 364 nm) and 488 nm light scattering signals produced by lymphoid cells demonstrated that the two signals were not equivalent and that they placed different emphasis on the physical parameters characterizing lymphocytes. Both small T and B lymphocytes from peripheral lymphoid tissues and mitogenically activated large T and B lymphocyte blasts were discriminated by both wavelengths. Differences between the Lyt-2 negative and Lyt-2 positive T lymphocyte subsets were also apparent. Two color light scattering could also discriminate between immature thymocytes and mature peripheral T cells and between small bone marrow cells and mature peripheral B cells. In bone marrow an increase in UV light scattering coincided with the appearance of cell surface immunoglobulin on small cells. These data establish that two color light scattering is a sensitive probe for distinguishing cells of apparently similar morphology and that it can be used to study the physical changes that occur during lymphoid cell differentiation.  相似文献

BACKGROUND: Cytologic examination of cerebrospinal fluid (CSF) continues to be important in the diagnosis of malignancies involving the leptomeninges. A well-recognized pitfall is overinterpretation of the presence of atypical lymphocytes that resemble malignant lymphoid cells in the CSF. A definite diagnosis is often difficult because of limited viability of cells and small sample size of CSF. CASE: A 25-year-old patient with a past history of treated large granular lymphocytic leukemia and presence of a predominant population of large, atypical lymphoid cells in the CSF, giving us the impression of involvement with large cell lymphoma. However, a timely call to the hematologist revealed that the serology was positive for acute Epstein-Barr virus infection. Flow cytometry of CSF confirmed polyclonal population of B-cells and T-cells. CONCLUSION: The presence of atypical cells in the CSF certainly warrants a detailed look at the patient's laboratory investigations and communication with the hematologist, because it may be the only specimen available for diagnosis on which therapy and prognosis is based.  相似文献
