The ultrastructure of the endolymphatic sac (ES) of the late stage larva of the Japanese red-bellied newt, Cynops pyrrhogaster (stage 57), was examined by light and transmission electron microscopy. The two endolymphatic sacs are located at the dorsal-medial side of the otic vesicle on the dorsal-lateral side of the midbrain in the cranial cavity. The wall of the sac is composed of a layer of cubical epithelial cells with loose, interposed intercellular spaces. The sac contains a large luminal cavity, in which endolymph and numerous otoconia are present. The epithelial cells of different portions of the sac have a similar structure. These cells contain an abundance of cytoplasmic organelles, including ribosomes, Golgi complexes, and numerous vesicles. Two types of vesicles are found in the epithelial cells: the “floccular” vesicle and the “granular” vesicle. The floccular vesicles are located in the supra- and lateral-nuclear cytoplasm and contain flocccular material. The granular vesicles have a fine granular substance and are usually situated apposed to the apical cell membrane. The granular vesicles are suggested to be secreted into the lumen, while the floccular vesicles are thought to be absorbed from the lumen and conveyed to the intercellular spaces by the epithelial cells. The apical surfaces of the epithelial cells bear numerous microvilli. Apparently floating cells, which bear long microvilli on the free surfaces, are observed in the lumen of the ES. Based on the fine structure, the function of the endolymphatic sac of the newt Cynops pyrrhogaster is discussed. 相似文献

The transepithelial shunt pathway of newt proximal tubule was examined with glass micro-electrode and electron microscopic methods. The input resistance of the peritubular (basal) membrane and tubular wall were found to be () and , respectively. The input resistance of the peritubular membrane was approximately 40-times larger than that of the tubular wall. When the kidneys were perfused in a lanthanum solution, the lanthanum ions were then observed in the junctional complexes and in the intercellular spaces on both the basal and apical sides. The results indicate that the electrical shunt pathway corresponds to the apical junctional complexes and the intercellular spaces, and that the tight junctions are not truly ‘tight’ for the transepithelial movement of small ions in the proximal tubule of the newt kidney. 相似文献

In quinoline treated blastomeres of Triturus helveticus Raz. and Pleurodeles waltlii Michah., microtubules disappear and around the centrioles markedly enlarged dense bodies accompanied by striated bodies are accumulated, from prophase beyond metaphase. It is concluded that these bodies in untreated cells play a role in the formation of the spindle microtubules. — During metaphase the chromosome are smooth-surfaced and show a pronounced tendency to stickiness. During ana-telophase they become surrounded by nuclear membranes and form caryomeres. — Quinoline does not interfere with centriole replication, but prevents the separation if diplosomes that have been already formed, if it acts before that separation. — Some centrioles exhibit different degrees of ultrastructural disarrangement. 相似文献

1. The effect of external Na+ concentration on p-aminohippurate uptake by isolated kidneys of newt (Triturus pyrrhogaster) was studied kinetically and electrophysiologically. 2. p-Aminohippurate uptake conformed to Michaelis-Menten type kinetics in regard to both p-aminohippurate and Na+ concentrations in the incubation medium. Kinetic studies revealed that reduction of Na+ concentration increased the values of Kt without altering the maximal rate (V) of p-aminohippurate uptake. The values of Kt were a linear function of the reciprocal of Na+ concentration. These results suggest the presence of interaction between p-aminohippurate and Na+ at the carrier level, i.e. Na+-coupled cotransport. 3. p-Aminohippurate had no effect on the electrical potential difference across the peritubular membrane in both 10 and 100 mM Na+ solutions, suggesting that p-aminohippurate is transported across the peritubular membrane in a form of electrically neutral carrier complex. This is consistent with the results of the kinetic studies. 4. p-Aminohippurate uptake was proportional to the electrochemical potential gradient of Na+ (delta mu Na) across the peritubular membrane. This result indicates that the maintenance of sufficient delta mu Na appears to be necessary for the accumulation of p-aminohippurate against its electrochemical potential gradient, supporting Na+ gradient hypothesis. 相似文献

The investigation of the degree of interpenetration between the two component phases of whewellite kidney stones, the protein matrix and calcium oxalate monohydrate crystallites, is extended by a technique of microchemical analysis, employing X-ray photoelectron spectroscopy (XPS). The technique involves determining the surface elemental (hence chemical) composition of fractured whewellite stones. Comparison with the bulk composition of about 95% COM and 5% matrix yielded information on the fracture path, hence on the spatial distribution of the two phases. The results showed large increases in matrix signal upon fracture, confirming that the minor phase, matrix, which is expected to provide an easy path for fracture, pervades the structure down to the level of individual crystallites. 相似文献

The thick ascending limb of Henle (TAL) in the rat until recently has been considered a morphologically homogeneous structure despite physiologic and biochemical evidence to the contrary. The present study was designed to examine the ultrastructural characteristics of the TAL in the inner cortex and the outer and inner stripes of the outer medulla using qualitative and quantitative transmission electron microscopy. Kidneys of male Sprague-Dawley rats were preserved by in vivo perfusion with glutaraldehyde for light and electron microscopy. The peritubular diameter and cell height were determined by direct measurements on tubule cross sections. Morphometric analyses were performed on montages of tubule cross sections. The peritubular diameter of the TAL was similar in the three regions under investigation, but the TAL cells were taller in the inner stripe than in the inner cortex and outer stripe. Morphometry revealed significant differences between the three regions with respect to the mean tubular cross-sectional area (AT), the surface density (SV), and the surface area per mm of tubule (ST) of apical and basolateral plasma membranes, and the volume density (VV) of mitochondria. The major morphologic division appeared to be between the inner stripe segment and the remainder of the TAL. Summary Male rats were divided in two experimental groups. In group I two partes distales of the hypophysis were grafted under the kidney capsule and in group II two complete hypophyses were transplanted. Animals were killed 5 to 22 months after the operation. The grafted tissue was excised and processed for light and electron microscopy.The transplanted pars distalis tissue showed a well developed vascularisation in contrast to the pars intermedia which appeared poorly vascularised. Six different cell types were observed in grafted pars distalis. They correspond to the different types of cells found in the rat pars distalis in situ. The predominant cell type in the graft displayed all the morphological characteristics of stimulated prolactotrophs. Pars intermedia cells appeared hypertrophied resembling the MSH cells under stimulation. Two types of syncytial formations were frequently seen. One of them appeared to originate from prolactotrophs and the other from MSH cells. Bodian impregnated fibres and structures resembling either growth cones of axons or typical nerve endings were observed in the pars intermedia of long-term grafted hypophyses. Pituicytes remained as isolated clusters of cells. Canaliculi lined by two or more pituicytes were observed. Saccular formations resembling the hypophyseal cleft appeared in all grafts studied. The present findings suggest that in the male rat the chronically grafted pituitary gland is capable of synthesising most or all the hormones which are known to be produced by the gland in situ. Furthermore, prolactin and MSH seem to be the predominant secretion of the transplanted pituitary.Supported by the National Research Council of Argentina. Summary The mesangial and juxtaglomerular cells were studied in kidneys of hibernating and non-hibernating ground squirrels, Citellus tridecemlineatus. In the hibernating animal, as compared with the non-hibernating, the mesangial cells show signs of increased activity. The cells are relatively larger, and numerous vacuoles appear in the cytoplasm. The juxtaglomerular cells also show signs of hyperactivity. Secretion droplets, mitochondria and free ribosomes increase in number and the endoplasmic reticulum becomes dilated. It is postulated that during hibernation, increased activity of mesangial cells slows glomerular filtration by absorption of filtrate into the mesangial matrix, and increased activity of juxtaglomerular cells results in increased renin secretion which in turn may stimulate hypersecretion of aldosterone to conserve sodium for water balance mechanisms necessary at the time.Supported by Research Grant A-2027 from the National Institute of Arthritis and Metabolic Diseases, National Institutes of Health.We wish to gratefully acknowledge the technical assistance of Mary Gandia. 相似文献

1. The relation between p-aminohippurate uptake and the electrochemical potential gradient of Na+ (delta muNa+) across the peritubular membrane was examined in newt (Triturus pyrrhogaster) kidney. The delta muNa+ was modified by changing cellular Na+ concentration and/or lowering the electrical potential difference across the peritubular membrane (peritubular membrane potential) 2. Elevation of external K+ concentration or addition of alanine at 40 mM to the medium decreased the delta muNa+ mainly through the depolarization of the cells. Addition of 1 mM ouabain resulted in a decrease in the peritubular membrane potential and increase in cellular Na+ concentration, thus decrease in the delta muNa+. 3. p-Aminohippurate uptake decreased in proportion to the decrease in the delta muNa+ under all experimental conditions, indicating that the maintenance of the delta muNa+ is required for p-aminohippurate transport. 4. All three different experimental conditions, high medium K+ concentration, 40 mM alanine or 1 mM ouabain, increased the apparent Michaelis constant, Kt, without affecting the maximal uptake rate, V, for p-aminohippurate. These results suggests that the delta muNa+, largely the peritubular membrane potential, may affect the association and/or dissociation of p-aminohippurate and Na+ at both interfaces of the peritubular membrane of the proximal tubular cells. 相似文献

Peculiar ultrastructural features of the kidney internal medullary substance have been studied in some typical representatives of the Middle Asia desert and semi-desert fauna (Spermophilopsis leptodactylus and Meriones erythrourus). Papillary organization in the species studied has a specific high differentiation of the fine nephron loop (Henle's) parts; flattening (especially in Meriones erythrourus) of epithelium in the collecting tubes while its cytoplasm is rather poor in organelle; high contents of the interstitial cells and their tight junction with canaliculi, capillaries, floccular material of the interstitial tissue; thinning of the interstitial sheets, especially between the collecting tubes and vessels, presence of collagene fibres in the interstitium, that is especially, specific for all the species of the Spermophilopsis leptodactylus studied. It is possible to suppose that development of similar features in the structure of the kidney internal medullary substance, where the main water resorption takes place in the species investigated, is connected with their life in similar environmental conditions with low hydratation level and is aimed to increase efficiency of the concentrating apparatus work. 相似文献

The localization of Na(+) , K(+) -ATPase (NKA) and the ultrastructural features of kidney were examined in larvae of the Persian sturgeon Acipenser persicus (L 31-41 mm total length and 182·3-417·3 mg). Investigations were conducted through light and electron microscopy and through immunofluorescence for NKA detection. The kidney nephrons consisted of a large glomerulus and tubules (neck, proximal, distal and collecting), which connected to the ureters. Posteriorly, ureters extended and joined together into a thin-walled ureter terminal sac. Ultrastructurally, the glomerular cells (podocytes) possessed distinctive pedicels that extended to the basal membrane. The proximal tubule (PT) showed two different cells. The cells lining the anterior part of PT possessed apical tall microvilli (c. 2·7 μm), a sub-apical tubular system, a basal nucleus and dense granules. Posteriorly in the cells, the sub-apical tubular system and granules were absent and round mitochondria associated with basolateral infoldings were found; the apical microvilli were reduced. Distal and collecting tubular cells showed the typical features of osmoregulatory cells, i.e. well-developed basolateral infoldings associated with numerous mitochondria. No immunofluorescence of NKA was detected in the glomeruli. A weak immunostaining was observed at the basolateral side of the cells lining the neck and PT. A strong immunostaining of NKA was observed in the entire cells of the distal tubules, collecting tubules and in some isolated cells of the ureters. In all immunostained cells, the basolateral region showed a much higher fluorescence and nuclei were immunonegative. In conclusion, the epithelial cells of kidney tubules had morphological and enzymatic features of ionocytes, particularly in the distal and collecting tubules. Thus, the kidney of A. persicus larvae possesses active ion exchange capabilities and, beside its implication in excretion, participates in osmoregulation. 相似文献

The pronephric kidneys were examined in upstream migrant sea lampreys, Petromyzon marinus L., by transmission and scanning electron microscopy. Each pronephros consists of an enlarged renal corpuscle (glomus) and ciliated nephrostomes, but there are no renal tubules. The renal corpuscle contains an extensive mesangium, which consists of a highly fibrous extracellular matrix, numerous mesangial cells, granulocytes, and macrophages. The extracellular matrix contains microfibrils with a morphology similar to amyloid P microfibrils, fibrils with a periodicity similar to fibrin, and abundant collagen. Often these fibrillar components are aggregated in the region of the basement membrane, giving it a thickened appearance. Some podocytes of the visceral epithelium appear swollen, and their cytoplasm contains numerous vacuolar inclusions, and many have only primary major processes with only a few or no foot processes. The morphological features of the pronephric kidney of the lamprey at this time in the life cycle reflect the regression of this organ, but some features also resemble those seen in renal pathologies of higher vertebrates. 相似文献

Summary In the median eminence of the newt a medial region and two lateral regions are described.In cross section, the medial region appears to be made up of 1) an outer or glandular zone (Zone I) containing aldehyde-thionine-positive and negative nerve fibres and blood capillaries. Nerve fibres appear aligned in palisade array along the capillaries. 2) An inner zone (Zone II) made up of a) a layer of aldehyde-thionine-positive nerve fibres (fibrous layer) belonging to the preoptic hypophyseal tract and b) a layer of ependymal cells lining the infundibular lumen and reaching the blood vessels with their long processes.The lateral regions display a less pronounced stratification and aldehyde-thionine positive nerve fibres are nearly absent.A slender lamina (ependymal border) containing mainly aldehyde-thionine-positive nerve fibres and ependymal cells connects the median eminence to the pars nervosa.At the ultrastructural level, in the outer zone of the medial region at least 4 types of nerve fibres and nerve endings are identified:Type I nerve fibres containing granular vesicles of 700–1000 Å and clear vesicles (250–400 Å).Type II nerve fibres containing granular vesicles and polymorphous granules of 900–1300 Å and clear vesicles (250–400 Å).Type III nerve fibres containing dense granules of 1200–2000 Å and clear vesicles of 250–400 Å.Type IV nerve fibres containing only clear vesicles of 250–400 Å. In the inner zone too, all these nerve fiber types are found among ependymal cells, while the fibrous layer consists of nerve fibres containing granules of 1200–2000 Å in diameter.In the lateral regions Type I, Type II and Type IV nerve fibres and their respective perivascular terminals are found; axons containing dense granules (1200–2000 Å) are scanty. In these regions typical synapses between Type I nerve fibres and processes rich in microtubules are visible.The classification and functional significance of nerve fibres in the median eminence are still unsolved, but it may be assumed that nerve fibres of the medial region belong to both the preoptic hypophyseal and tubero hypophyseal tract, while the lateral regions are characterized by nerve fibres of the tubero hypophyseal tract.
