Summary Changes in epicuticular wax morphology of Pseudotsuga menziesii needles were studied with scanning electron microscopy throughout the growing season in current-year and older needles in 20 trees from two sites, Kootwijk and Garderen (The Netherlands). Fusion of crystalline wax rods leading to a reticulate structure and ultimately to degradation of the regular three-dimensional porous structure started several weeks after bud break and reached a high level at the end of the first growing season. The increase in amorphous (solid) wax showed a similar, but slightly slower development. In 1- and 2-year-old needles the degradation of crystalline wax and increase in amorphous wax had progressed only slightly further. The rates of change in crystalline wax morphology were very similar for sun-exposed and shaded needles, for the two sites, and for the two tree vitality classes included in the samples. The development of epiphyllous fungi also appeared to be largely needle age dependent. The effects of NH3 on needle wax morphology were studied in young trees used in fumigation experiments. Short-term fumigation (approx. 5 weeks) did not affect wax morphology in current-year needles, but 1-year-old needles which had been exposed to different concentrations from bud break onwards showed a severe degradation of the crystalline wax, regardless of the NH3 concentration used. In 2-yearold needles the effect of NH3 could not be traced and was overshadowed by the natural ageing process. Ambient O3, SO2 and NOx levels did not effect epicuticular wax morphology. It is suggested that the variation recorded for the two forest stands does not show effects of local pollution levels.  相似文献

A cold-stage SEM was used to document the morphology of ice in apple tissue and a sucrose solution frozen at rates ranging from 450 to 0.03 degrees K/min. Freezing rates of 3-mm-thick apple discs were measured with a differential thermocouple technique, which gave measurement of the growth velocity and the temperature gradient through the solidified specimen as well as the cooling rate during solidification. Cold-stage SEM micrographs were used to measure the dendritic spacing of the ice structures, and these data were found to correlate linearly with the square root of the cooling rate during solidification as would be predicted by a theoretical analysis of mass transfer in the formation of dendrites. Comparison of freeze-substituted and freeze-dried apple-tissue micrographs with those from a cold-stage SEM showed that the cold-stage SEM technique was the only one which correctly represented ice morphology in apple tissue.  相似文献

The aim of this study was to reveal the causes and consequences of individual differences in foraging behaviour of coal tits, Parus ater, on the island of Gotland in the Baltic. On Gotland the willow tit, P. montanus, and the crested tit, P. cristatus, are absent, while these larger species are abundant competitors on the mainland. The Gotland population of coal tits exploited parts of the foraging niches of their absent competitors. Gotland coal tits are bigger than those on the mainland; they have thus evolved towards the size of the missing competitors. Individual differences in niche use were related to their morphology. Large birds more often used the inner parts of trees whereas small individuals foraged more on needles in the outer parts of the trees. Also, individuals with wing shapes more suitable for hovering and slow flight foraged more on needles. It is argued that morphology affects the profitability of different foraging sites. However, for foraging site selection, age was more important than morphology. Older, and probably more dominant, individuals occupied the most profitable foraging sites.  相似文献

For several decades, southern California experienced the worst ozone pollution ever reported. Peak ozone concentrations have, however, declined steadily since 1980. In this study, the structural injuries underlying ozone symptoms in needles of ponderosa pine (Pinus ponderosa Dougl. ex Laws.) collected in summer 2006 from one of the most polluted sites in the San Bernardino Mountains were investigated using serial sections examined by light and electron microscopy. Ozone-specific light-green diffuse mottling was observed in the current-year needles, whereas older foliage showed brownish mottling similar to winter fleck injury. Especially, within the outer layers of mesophyll, many markers of oxidative stress, typical for ozone, were observed in both apoplast and symplast. Altogether within cells of mottles, these markers were indicative of hypersensitive-like response, whereas degenerative structural changes were diagnosed in the surrounding mesophyll. Evidence of drought stress and frost injury to older needles was also detected. Hence, mottling injury appeared to be primarily caused by ozone stress, however, other environmental stressors also determined the symptom morphology and distribution, especially within the older foliage.  相似文献

To understand the corrosion properties of surgical scissors, 416 stainless steel disks and custom electrodes were used as simulated surfaces under various conditions. These simulated surfaces were exposed to tap water and 400-ppm synthetic hard water as Ca2CO3 under different conditions. The samples were evaluated by various techniques for corrosion potential and the impact of environmental conditions on the integrity of the passive film. The electrodes were used to monitor the corrosion behavior by potentiodynamic polarization technique in water both in the presence and absence of a cleaning product. The surface topography of the 416 stainless steel disks was characterized by visual observations and scanning electron microscopy (SEM), and the surface chemistry of the passive film on the surface of the scissors was characterized by x-ray photoelectron spectroscopy (XPS). The results suggest that surgical instruments made from 416 stainless steel are not susceptible to uniform corrosion; however, they do undergo localized corrosion. The use of suitable cleaning products can offer protection against localized corrosion during the cleaning step. More importantly, the use of potentiodynamic polarization techniques allowed for a quick and convenient approach to evaluate the corrosion properties of surgical instruments under a variety of simulated-use environmental conditions.  相似文献

BACKGROUND AND AIMS: The morphology of the epicuticular leaf waxes of Wollemia nobilis (Araucariaceae) was studied with special emphasis on the relationship between the microstructure of epicuticular wax crystals and their chemical composition. Wollemia nobilis is a unique coniferous tree of the family Araucariaceae and is of very high scientific value as it is the sole living representative of an ancient genus, which until 1994 was known only from fossils. METHODS: Scanning electron microscopy (SEM), gas chromatography (GC) combined with mass spectrometry (GC-MS) and nuclear magnetic resonance spectroscopy (NMR) were used for characterizing the morphology and the chemical structure of the epicuticular wax layer of W. nobilis needles. KEY RESULTS: The main component of the leaf epicuticular wax of W. nobilis is nonacosan-10-ol. This secondary alcohol together with nonacosane diols is responsible for the tubular habit of the epicuticular wax crystals. Scanning electron micrographs revealed differences in the fine structure of adaxial and abaxial leaf surfaces that could be explained by gas chromatographic studies after selective mechanical removal of the waxes. CONCLUSIONS: SEM investigations established the tubular crystalline microstructure of the epicuticular wax of W. nobilis leaves. GC-MS and NMR experiments showed that nonacosan-10-ol is the major constituent of the epicuticular wax of W. nobilis leaves.  相似文献

The aim of the present study was to develop surgical instruments necessary to achieve a precisely reamed surface and stable initial fixation. The instruments used to prepare the socket were a gauge-drill guide, a liner for the spigot hole and two spigotted reamers of different design and indentation intended to achieve a precisely reamed surface while preserving subchondral bone. For each reamer we implanted in a synthetic hip model 10 uncemented cups with 2 mm press-fit and loaded at 2.4 kN in the physiological axis (Mod. 8501, Instron, Canton, MA, USA). The micromotion between implant and bone socket was measured using an inductive micrometric measuring system (MultiNCDT-500, Micro-Epsilon, Ortenburg, Germany) and compared with that seen after using conventional instruments. The use of the new reamer of elliptical design significantly reduced the standard deviations of the measured values (p < 0.01 at the ischium and pubis) and also reduced maximum movement (p < 0.01 at the ischium); at the same time, all the components showed overall limited movement (< 150 microns at the ischium, pubis and ilium) under maximum loading (2.4 kN). Manufacturing tolerances, the quality and wear of the instruments, acetabular bone stock and surgical technique all impact on the degree of press-fit obtainable at surgery. The results of our study show that press-fit and initial stability can be optimized by using adequate instruments to prepare the socket.  相似文献

Summary In order to study physiological strain caused by release cutting, suppressed Norway spruce on mesic and moist sites was completely released from overstory birch, or 500 birches per hectare were left as a shelter. The treatments were conducted in late June in 1988 and in 1989. The spruce's reaction to the environmental change was monitored by measurements of fast chlorophyll fluorescence kinetics and analysis of chlorophyll content. This was done before treatment, 1 week after treatment, 2 months after treatment, and twice during the following growth period. Complete release resulted in a more pronounced decrease in the ratio of variable fluorescence to maximal fluorescence (Fv/Fm) than partial release. There was also a tendency for the build-up of chlorophyll content in needles to be more affected when the spruce was completely released. Released spruces on moist sites tended to be more affected by the release than released spruces on mesic sites. The results suggest that in this kind of stand the risk of damage to the spruces is greatest when the spruces are completely released on moist sites. Furthermore, it is shown that the weather conditions prevailing shortly before and after the release have a large influence on the spruce's reaction to the release. The results also indicate some adjustment to the new environment in mature needles.  相似文献

The potential of Univariate and Multivariate Analysis and specifically Principal Components Analysis (PCA), has been employed to assess the performance of pine needles as bioindicators of polycyclic aromatic hydrocarbons (PAHs) and particularly emphasize the crucial importance of making a distinction between different pine species if more than one is sampled. Four sampling sessions were done in 29 sites and needles of two common pine species (Pinus pinaster Ait. in 19 sites and Pinus pinea L. in 12) were collected and analysed using gas chromatography-mass spectrometry (GC-MS) and PCA. The results obtained indicated significant differences between species, attributed to their different morphology. The mean total PAH concentration of the P. pinaster needles are over two times higher than P. pinea's. This difference is lower when the results are presented in lipid weight, but still statistically significant. Samples from the two sites with adjacent trees reinforce these conclusions, showing significant differences in terms of PAH mean concentration and aromatic ring patterns.  相似文献

The aim of the research was to evaluate the content of amino acids in the needles of Pinus sylvestris growing in the area affected by a nitrogen fertilizer factory and to compare them with other parameters of needles, trees, and sites. Three young-age stands of Scots pine were selected at a distance of 0.5 km, 5 km, and 17 km from the factory. Examination of the current-year needles in winter of the year 2000 revealed significant (p < 0.05) differences between the site at a 0.5-km distance from the factory and the site at a 17-km distance from the factory--with the site closest to the factory showing the highest concentrations of protein (119%), total arginine (166%), total other amino acids (depending on amino acid, the effect ranged between 119 and 149%), free arginine (771%), other free amino acids (glutamic acid, threonine, serine, lysine--depending on amino acid, the effect ranged between 162 and 234%), also the longest needles, widest diameter, largest surface area, and heaviest dry weight (respectively, 133, 110, 136, and 169%). The gradient of nitrogen concentration in the needles was assessed on the selected plots over the period of 1995-2000, with the highest concentration (depending on year, 119 to 153%) documented in the site located 0.5 km from the factory. Significant correlations were determined between the total amino acid contents (r = 0.448 -0.939, p < 0.05), some free amino acid (arginine, aspartic acid, glutamic acid, lysine, threonine, and serine) contents (r = 0.418 - 0.975, p < 0.05), and air pollutant concentration at the sites, the distance between the sites and the factory, and characteristics of the needles. No correlation was found between free or total arginine content and defoliation or retention of the needles. In conclusion, it was revealed that elevated mean monthly concentration of ammonia (26 microg m(-3)) near the nitrogen fertilizer factory caused changes in nitrogen metabolism, especially increasing (nearly eight times) concentration of free arginine in the needles of Scots pine.  相似文献

There is increasing evidence that central noradrenaline (NA) transport mechanisms are implicated in the central nervous system complications of acute liver failure. In order to assess this possibility, binding sites for the high affinity NA transporter ligand [3H]-nisoxetine were measured by quantitative receptor autoradiography in the brains of rats with acute liver failure resulting from hepatic devascularization and in appropriate controls. In vivo microdialysis was used to measure extracellular brain concentrations of NA. Severe encephalopathy resulted in a significant loss of [3H]-nisoxetine sites in frontal cortex and a concomitant increase in extracellular brain concentrations of NA in rats with acute liver failure. A loss of transporter sites was also observed in thalamus of rats with acute liver failure. This loss of NA transporter sites could result from depletion of central NA stores due to a reserpine-like effect of ammonia which is known to accumulate to millimolar concentrations in brain in ischemic liver failure. Impaired NA transport and the consequent increase in synaptic concentrations and increased stimulation of neuronal and astrocytic noradrenergic receptors could be implicated in the pathogenesis of the encephalopathy and brain edema characteristic of acute liver failure.  相似文献

Fertilized stands of Pseudotsuga menziesii were found to have glaucous needles. We investigated the morphological and quantitative characteristics of the epicuticular waxes of needles of fertilized and control trees. Glaucousness was caused by ornate tubular epicuticular wax. Dipping needles in chloroform, which dissolves waxes, eliminated the glaucous appearance. Based on cryostage scanning electron microscopic observations, the epicuticular waxes in the nonstomatal region were much more ornate on the needles of the fertilized trees (experimental needles) than in unfertilized trees (control needles). The stomatal region in both experimental and control needles showed similarly ornate waxes. Quantities of waxes were similar in experimental and control needles. The glaucousness was not the result of greater quantities of wax; rather, fertilization altered wax morphology in the nonstomatal regions.  相似文献

Speciation involves the evolution of traits and genetic differences that contribute to reproductive isolation and the cessation of gene flow, and studying closely related species and divergent populations gives insight into how these phenomena proceed. Here, we document patterns of gene flow within and between two members of a rapid Neotropical species radiation, Costus pulverulentus and Costus scaber (Costaceae). These species co‐occur in the tropical rainforest and share pollinators, but are reproductively isolated by a series of prezygotic barriers, some of which show evidence of reinforcement at sympatric sites. Here, we genotype microsatellite markers in plants from eight sites that span the geographical range of both species, including four sympatric sites. We also genotype putative hybrids found at two sympatric sites. We find high levels of genetic isolation among populations within each species and low but detectable levels of introgression between species at sympatric sites. Putative hybrids identified by morphology are consistent with F1 or more advanced hybrids. Our results highlight the effectiveness of prezygotic isolating mechanisms at maintaining species boundaries in young radiations and provide empirical data on levels of gene flow consistent with reinforcement.  相似文献

The objective of this study was to determine how different techniques used during the freezing step of lyophilization affect morphology of the dried protein solids. Aqueous solutions containing recombinant human albumin, trehalose, and sodium phosphate buffer were dried after their freezing by shelf-ramp cooling, immersion in liquid nitrogen, or controlled ice nucleation. Some shelf-frozen solutions were heat treated (annealed) before the vacuum drying. We used three-dimensional (3D) X-ray micro-computed tomography (micro-CT) and scanning electron microscopy (SEM) to study the morphology of solids. The X-ray micro-CT images of the lyophilized microporous solids showed traces of varied size and structure ice crystals that were comparable to corresponding SEM images. A post-freeze heat treatment and a controlled nucleation both induced larger ice crystal ghosts in the solids. The variations in the structure of walls surrounding ice crystals, formed by the different freezing procedures, should affect the water vapor transition during the primary and secondary drying. Some solids also showed higher-density layer in the upper surface. Overall, the simple sample preparation procedures and the ample morphological information make the X-ray micro-CT appropriate for analyzing lyophilized pharmaceuticals.  相似文献

The quantification of focal adhesion sites offers an assessable method of measuring cell-substrate adhesion. Such measurement can be hindered by intra-sample variation that may be cell cycle derived. A combination of autoradiography and immunolabelling techniques, for scanning electron microscopy (SEM), were utilised simultaneously to identify both S-phase cells and their focal adhesion sites. Electron-energy 'sectioning' of the sample, by varying the accelerating voltage of the electron beam, combined with backscattered electron (BSE) imaging, allowed for S-phase cell identification in one energy 'plane' image and quantitation of immunogold label in another. As a result, it was possible simultaneously to identify S-phase cells and their immunogold-labelled focal adhesions sites on the same cell. The focal adhesion densities were calculated both for identified S-phase cells and the remaining non-S-phase cells present. The results indicated that the cell cycle phase was a significant factor in determining the density of focal adhesions, with non-S-phase cells showing a larger adhesion density than S-phase cells. Focal adhesion morphology was also seen to correspond to cell cycle phase; with 'dot' adhesions being more prevalent on smaller non-S-phase and the mature 'dash' type on larger S-phase cells. This study demonstrated that when quantitation of focal adhesion sites is required, it is necessary to consider the influence of cell cycle phases on any data collected.  相似文献

Rates of hybridization vary among angiosperm taxa. Among-taxon variation in hybridization rate has been used to compare the importance of pre- and post-zygotic reproductive isolating mechanisms. Variation in rates of hybridization within a single-species pair would suggest that local conditions also affect reproductive isolation within a single taxonomic context. In this study, contact sites of Ipomopsis aggregata-Ipomopsis tenuituba were surveyed for variation in frequency of hybrids, and spatial structure. Floral morphology was used to identify parent species and hybrids in seven contact sites in the western Rocky Mountains, USA. Contact sites varied widely in elevational range, the degree to which morphological variation was clinal rather than mosaic and the frequency of hybrids. Two sites provided a strong contrast between a clinal, unimodal site and a mosaic, bimodal site. This natural variation among contact sites of the same species pair provides an opportunity to assess the effect of local ecological conditions and spatial structure of parent populations on reproductive isolation, while controlling for between-taxon variation.  相似文献

Human amniotic membrane that has been processed and sterilised by gamma irradiation is widely used as a biological dressing in surgical applications. The morphological structure of human amniotic membrane was studied under scanning electron microscopy (SEM) to assess effects of gamma radiation on human amniotic membrane following different preservation methods. The amniotic membrane was preserved by either air drying or submerged in glycerol before gamma irradiated at 15, 25 and 35 kGy. Fresh human amniotic membrane, neither preserved nor irradiated was used as the control. The surface morphology of glycerol preserved amnion was found comparable to the fresh amniotic membrane. The cells of the glycerol preserved was beautifully arranged, homogonous in size and tended to round up. The cell structure in the air dried preserved amnion seemed to be flattened and dehydrated. The effects of dehydration on intercellular channels and the microvilli on the cell surface were clearly seen at higher magnifications (10,000×). SEM revealed that the changes of the cell morphology of the glycerol preserved amnion were visible at 35 kGy while the air dried already changed at 25 kGy. Glycerol preservation method is recommended for human amniotic membrane as the cell morphological structure is maintained and radiation doses lower than 25 kGy for sterilization did not affect the appearance of the preserved amnion.  相似文献
