The electrophoretic mobility of L5178Y cells in 0.0145 M NaCl, 4.5% sorbitol, 0.6 mM NaHCO3, pH 7.2, at 25°C was — 1.78 μ·s?1·V?1·cm?1 while that of an L-asparaginase resistant subline, L5178Y/ASN, was — 1.11 μm·s?1·V?1·cm?1. Both cell lines were characterized by terminal sialic acid residues on their surfaces. Treatment of L5178Y cells for 90 min with 10 units of L-asparaginase per ml in saline decreased the electrophoretic mobility of the cells to — 1.65 μm·s?1·V?1·cm?1 while treatment in Fischer's medium decreased the mobility to — 1.25 μm·s?1·V?1·cm?1; neither treatment had a significant effect on the L5178Y/ASN electrophoretic mobility. The results suggest that L-asparaginase has an immediate and specific effect on synthesis of cell surface asparaginyl glycoproteins.  相似文献

A dense community of shade adapted microalgae dominated by the diatom Trachyneis aspera is associated with a siliceous sponge spicule mat in McMurdo Sound, Antarctica. Diatoms at a depth of 20 to 30 m were found attached to spicule surfaces and in the interstitial water between spicules. Ambient irradiance was less than 0.6 μE · m?2· s?1 due to light attenuation by surface snow, sea ice, ice algae, and the water column. Photosynthesis-irradiance relationships determined by the uptake of NaH14CO3 revealed that benthic diatoms beneath annual sea ice were light-saturated at only 11 μE·m?2·s?1, putting them among the most shade adapted microalgae reported. Unlike most shade adapted microalgae, however, they were not photoinhibited even at irradiances of 300 μE·m?2·s?1. Although in situ primary production by benthic diatoms was low, it may provide a source of fixed carbon to the abundant benthic invertebrates when phytoplankton or ice algal carbon is unavailable.  相似文献

Sea ice microalgae in McMurdo Sound, Antarctica were examined for photosynthesis-irradiance relationships and for the extent and time course of their photoadaptation to a reduction in in situ irradiance. Algae were collected from the bottom centimeter of coarse-grained congelation ice in an area free of natural snow cover. Photosynthetic rate was determined in short term (1 h) incubations at ?2° C over a range of irradiance from 0 to 286 μE·m?2·s?1. Assimilation numbers were consistently below 0.1 mg C·mg chl a?1·h?1. The Ik's3 averaged only 7 μE·m?2·s?1, and photosynthesis was inhibited at irradiances above 25 μE·m?2·s?1. Photosynthetic parameters of the ice algal community were examined over a nine day period following the addition of 4 cm of surface snow while a control area remained snow-free. A reduction of 40% in PmB relative to the control occurred after two days of snow cover; α, β, Ik, and Im were not significantly altered. Low assimilation numbers and constant standing crop size, however, suggested that the algal bloom may have already reached stationary growth phase, possibly minimizing their photoadaptive response.  相似文献

Microalgae growing within brine channels (85 psu salinity) of the surface ice layers of Antarctic pack ice showed considerable photosynthetic tolerance to the extreme environmental condition. Brine microalgae exposed to temperatures above ?5°C and at irradiances up to 350 μmol photons·m?2·s?1 showed no photosynthetic damage or limitations. Photosynthesis was limited (but not photoinhibited) when brine microalgae were exposed to ?10°C, provided the irradiance remained under 50 μmol photons·m?2·s?1. The highest level of photosynthetic activity (maximum relative electron transport rate [rETRmax]) in brine microalgae growing within the surface layer of sea ice was at approximately 18 μmol electrons·m?2·s?1, which occurred at ?1.8°C. Effective quantum yield of PSII and rETRmax of the halotolerant brine microalgae exhibited a temperature‐dependent pattern, where both parameters were higher at ?1.8°C and lower at ?10°C. Relative ETRmax at temperatures above ?5°C were stable across a wide range of irradiance.  相似文献

Mangroves on Pacific high islands offer a number of important ecosystem services to both natural ecological communities and human societies. High islands are subjected to constant erosion over geologic time, which establishes an important source of terrigeneous sediment for nearby marine communities. Many of these sediments are deposited in mangrove forests and offer mangroves a potentially important means for adjusting surface elevation with rising sea level. In this study, we investigated sedimentation and elevation dynamics of mangrove forests in three hydrogeomorphic settings on the islands of Kosrae and Pohnpei, Federated States of Micronesia (FSM). Surface accretion rates ranged from 2.9 to 20.8 mm y?1, and are high for naturally occurring mangroves. Although mangrove forests in Micronesian high islands appear to have a strong capacity to offset elevation losses by way of sedimentation, elevation change over 6½ years ranged from ?3.2 to 4.1 mm y?1, depending on the location. Mangrove surface elevation change also varied by hydrogeomorphic setting and river, and suggested differential, and not uniformly bleak, susceptibilities among Pacific high island mangroves to sea-level rise. Fringe, riverine, and interior settings registered elevation changes of ?1.30, 0.46, and 1.56 mm y?1, respectively, with the greatest elevation deficit (?3.2 mm y?1) from a fringe zone on Pohnpei and the highest rate of elevation gain (4.1 mm y?1) from an interior zone on Kosrae. Relative to sea-level rise estimates for FSM (0.8–1.8 mm y?1) and assuming a consistent linear trend in these estimates, soil elevations in mangroves on Kosrae and Pohnpei are experiencing between an annual deficit of 4.95 mm and an annual surplus of 3.28 mm. Although natural disturbances are important in mediating elevation gain in some situations, constant allochthonous sediment deposition probably matters most on these Pacific high islands, and is especially helpful in certain hydrogeomorphic zones. Fringe mangrove forests are most susceptible to sea-level rise, such that protection of these outer zones from anthropogenic disturbances (for example, harvesting) may slow the rate at which these zones convert to open water.  相似文献

Rats were fed a diet containing 1% β-guanidinopropionic acid (GPA) for 6–10 weeks to deplete their skeletal muscle of creatine. 31P-NMR was used to monitor metabolic changes in the gastrocnemius muscle at rest, during stimulated steady-state isometric contraction at 4 Hz and during recovery from stimulation. In resting muscles, the [creatine phosphate] was reduced to 10% (2.8 μmol·g?1) and the [ATP] to 50% (3.3 μmol·g?1) of those found in rats fed a control diet. The concentration of the phosphorylated form of the analogue (PGPA) was 23 μmol·g?1. There was no significant difference in muscle performance or in the relative changes in the [ATP] during stimulation. Intracellular pH decreased rapidly on stimulation and recovered during the stimulation period to near resting values in both groups. In control rats, the initial decrease in pH was greater and the time to recovery was longer than in GPA-fed rats. The rate at which PGPA supplied energy to the contracting muscle (0.027 mM·s?1) was insignificant relative to the minimum estimated rate of ATP turnover (1 mM·s?1). The rate of PGPA resynthesis during recovery (0.018 mM·s?1) is enzyme-limited and provides an independent estimate of creatine kinase flux during this period (18.9 mM·s?1). The creatine kinase flux (creatine phosphate → ATP) in the resting muscle of GPA-fed rats was 12-fold less than in control animals, 1.3 vs. 15.7 mM·s?1. These results demonstrate that neither the [creatine phosphate] nor the activity of creatine kinase is critical for aerobic metabolism. Skeletal muscle appears to adapt to a diminished creatine pool by enhancing its aerobic capacity.  相似文献

Aims: To investigate the relationships between sulfate‐reducing bacteria (SRB), growth conditions, bentonite densities and copper sulfide generation under circumstances relevant to underground, high‐level radioactive waste repositories. Methods and Results: Experiments took place 450 m underground, connected under in situ pressure to groundwater containing SRB. The microbial reduction of sulfate to sulfide and subsequent corrosion of copper test plates buried in compacted bentonite were analysed using radioactive sulfur (35SO42?) as tracer. Mass distribution of copper sulfide on the plates indicated a diffusive process. The relationship between average diffusion coefficients (Ds) and tested density (ρ) was linear. Ds (m2 s?1) = ?0·004 × ρ (kg m?3) + 8·2, decreasing by 0·2 Ds units per 50 kg m?3 increase in density, from 1·2 × 10?11 m2 s?1 at 1750 kg m?3 to 0·2 × 10?11 m2 s?1 at 2000 kg m?3. Conclusions: It is possible that sulfide corrosion of waste canisters in future radioactive waste repositories depends mainly on sulfide concentration at the boundary between groundwater and the buffer, which in turn depends on SRB growth conditions (e.g., sulfate accessibility, carbon availability and electron donors) and geochemical parameters (e.g., presence of ferrous iron, which immobilizes sulfide). Maintaining high bentonite density is also important in mitigating canister corrosion. Significance and Impact of the Study: The sulfide diffusion coefficients can be used in safety calculations regarding waste canister corrosion. The work supports findings that microbial activity in compacted bentonite will be restricted. The study emphasizes the importance of growth conditions for sulfate reduction at the groundwater boundary of the bentonite buffer and linked sulfide production.  相似文献

Microsupercapacitors (MSCs) with high energy densities offer viable miniaturized alternatives to bulky electrolytic capacitors if the former can respond at the kilo Hertz (kHz) or higher frequencies. Moreover, MSCs fabricated on a chip can be integrated into thin‐film electronics in a compatible manner, serving the function of ripple filtering units or harvesters of energy from high‐frequency sources. In this work, wafer‐scale fabrication is demonstrated of MXene microsupercapacitors with controlled flake sizes and engineered device designs to achieve excellent frequency filtering performance. Specifically, the devices (100 nm thick electrodes and 10 µm interspace) deliver high volumetric capacitance (30 F cm?3 at 120 Hz), high rate capability (300 V s?1), and a very short relaxation time constant (τ0 = 0.45 ms), surpassing conventional electrolytic capacitors (τ0 = 0.8 ms). As a result, the devices are capable of filtering 120 Hz ripples produced by AC line power at a frequency of 60 Hz. This study opens new avenues for exploring miniaturized MXene MSCs as replacements for bulky electrolytic capacitors.  相似文献

Light intensity and temperature interactions have a complex effect on the physiological process rates of the filamentous bluegreen alga Anabaena variabilis Kütz. The optimum temperature for photosynthesis increased with increasing light intensity from 10°C at 42 μE·m?2·s?1 to 35°C at 562 μE·m?2·s?1. The light saturation parameter, IK, increased with increasing temperatures. The maximum photosynthetic rate (2.0 g C·g dry wt.?1·d?1) occurred at 35°C and 564 μE·m?2·s?1. At 15°C, the maximum rate was 1.25 g C·g dry wt.?1·d?1 at 332 μE·m?2·s?1. The dark respiration rate increased exponentially with temperature. Under favorable conditions of light intensity and temperature the percent of extracellular release of dissolved organic carbon was less than 5% of the total C fixed. This release increased to nearly 40% under combinations of low light intensity and high temperature. A mathematical model was developed to simulate the interaction of light intensity and temperature on photosynthetic rate. The interactive effects were represented by making the light-saturation parameters a function of temperature.  相似文献

In slow mainstream flows (<4–6 cm · s?1), the transport of dissolved nutrients to seaweed blade surfaces is reduced due to the formation of thicker diffusion boundary layers (DBLs). The blade morphology of Macrocystis pyrifera (L.) C. Agardh varies with the hydrodynamic environment in which it grows; wave‐exposed blades are narrow and thick with small surface corrugations (1 mm tall), whereas wave‐sheltered blades are wider and thinner with large (2–5 cm) edge undulations. Within the surface corrugations of wave‐exposed blades, the DBL thickness, measured using an O2 micro‐optode, ranged from 0.67 to 0.80 mm and did not vary with mainstream velocities between 0.8 and 4.5 cm · s?1. At the corrugation apex, DBL thickness decreased with increasing seawater velocity, from 0.4 mm at 0.8 cm · s?1 to being undetectable at 4.5 cm · s?1. Results show how the wave‐exposed blades trap fluid within the corrugations at their surface. For wave‐sheltered blades at 0.8 cm · s?1, a DBL thickness of 0.73 ± 0.31 mm within the edge undulation was 10‐fold greater than at the undulation apex, while at 2.1 cm · s?1, DBL thicknesses were similar at <0.07 mm. Relative turbulence intensity was measured using an acoustic Doppler velocimeter (ADV), and overall, there was little evidence to support our hypothesis that the edge undulations of wave‐sheltered blades increased turbulence intensity compared to wave‐exposed blades. We discuss the positive and negative effects of thick DBLs at seaweed surfaces.  相似文献

The catalase from Proteus mirabilis peroxide-resistant bacteria is one of the most efficient heme-containing catalases. It forms a relatively stable compound II. We were able to prepare samples of compound II from P. mirabilis catalase enriched in 57Fe and to study them by spectroscopic methods. Two different forms of compound II, namely, low-pH compound II (LpH II) and high-pH compound II (HpH II), have been characterized by Mössbauer, extended X-ray absorption fine structure (EXAFS) and UV-vis absorption spectroscopies. The proportions of the two forms are pH-dependent and the pH conversion between HpH II and LpH II is irreversible. Considering (1) the Mössbauer parameters evaluated for four related models by density functional theory methods, (2) the existence of two different Fe–Oferryl bond lengths (1.80 and 1.66 Å) compatible with our EXAFS data and (3) the pH dependence of the α band to β band intensity ratio in the absorption spectra, we attribute the LpH II compound to a protonated ferryl FeIV–OH complex (Fe–O approximately 1.80 Å), whereas the HpH II compound corresponds to the classic ferryl FeIV=O complex (Fe=O approximately 1.66 Å). The large quadrupole splitting value of LpH II (measured 2.29 mm s?1 vs. computed 2.15 mm s?1) compared with that of HpH II (measured 1.47 mm s?1 vs. computed 1.46 mm s?1) reflects the protonation of the ferryl group. The relevancy and involvement of such (FeIV=O/FeIV–OH) species in the reactivity of catalase, peroxidase and chloroperoxidase are discussed.  相似文献

Meiospores of Lessonia nigrescens Bory collected in Central Chile, were cultivated under different light and nutrient conditions to test for their influence on the development of gametophytes and gametogenesis. The microscopic stages of the life cycle are described. Under 45 μE·m?2 ·s?1 (12 h daily) and sea water enriched with nitrate and phosphate, development of few-celled gametophytes, gametogenesis and sporophyte differentiation occur within 35 days. At 10 μE ·m?2 · s?1, most gametophytes are multicellular and remain vegetative up to 65 days. Only enriched sea water or enriched sea water with double concentrations of phosphate lead to fertile few-celled gametophytes under 45 μE · m?2 · s?1. The addition of double concentrations of nitrate allows for the development of few-celled gametophytes but inhibits gametogenesis. The average gametophyte size progressively decreases when increasing amounts of nitrate and phosphate are added to the media.  相似文献

We present a new version of a compact insect-mimicking flapping-wing system driven by a small motor, and suggest two testing approaches to measure the thrust or lift generated by a flapping-wing system. Flapping performance tests show the proposed flapping-wing system, which is powered by an onboard battery (lithium, 3.7 V, 180 mAh), could flap at flapping frequency of 25 Hz, and produce an average thrust or lift of about 3 g. In a wired-flight test under constrained conditions, the flapping-wing system could fly at an average forward velocity of 700 mm·s?1. For measuring the average thrust or lift produced by the flapping-wing system, we propose two testing approaches of wired-flight test and swing test with the aid of a high-speed camera and they are compared with a load cell measurement. The average thrust or lift values from the two proposed approaches agree well with the average thrust or lift values measured by a load cell.  相似文献

With the aid of measurements of the fluorescence yield, the efficiency of the various deexcitation mechanisms of an exciton in the light-harvesting system has been determined. For this purpose, the fluorescence of dark-adapted as well as of 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU)-treated and preilluminated leaves of Zea mays L. was excited by single ultrashort laser pulses of different energies. The experimental results have served for the fitting of solutions of rate equations, which describe the deexcitation by linear relaxation processes like fluorescence and radiationless transitions, by annihiation of excitons, and by traps both in the ground state and in an excited state. We have obtained the following results: a ratio of antenna chlorophyll molecules to Photosystem II traps of 600:1, an annihilation constant γ = 2·10?8 cm3·s?1, a mean trapping time of t?=0.5 ns, a trapping probability for traps in the ground state of 2·10?8 cm3·s?1, and 6·10?9 cm3·s?1 for traps in an excited state.  相似文献

The rates of electron transfer to P700 from plastocyanin and cytochrome f have been compared with those from three other c-type cytochromes and azurin, a copper protein resembling plastocyanin. Three different disruptive techniques were used to expose P700; digitonin, Triton X-100 and sonication. The following rate constants were measured at 25 °C, pH 7.0, with digitonin-treated chloroplasts: plastocyanin, 8 · 107 M?1 · s?1; red-algal cytochrome c-553, 1.9 · 107 M?1 · s?1; Pseudomonas cytochrome c-551, 8 · 106 M?1 · s?1; azurin, ? 3 · 105 M?1 · s?1; cytochrome f, ? 2 · 104 M?1 · s?1; mammalian cytochrome c, ? 2 · 104 M?1 · s?1. For electron transfer from plastocyanin, the effects of ionic strength, pH and temperature were also studied, and saturation effects found in earlier work were avoided by a full consideration of the various secondary reactions and inclusion of superoxide dismutase. The binding of chlorpromazine · HCl at the human erythrocyte surface has been detected through its effect on cellular electrophoretic mobility. Incubation of erythrocytes (approx. 5 · 106/ml) in 23 μM chlorpromazine · HCl resulted in a reduction of negative electrophoretic mobility from the control value of ?1.11 ± 0.01 (μm · s?1)/(V · cm?1) to ?1.00 ± 0.02 (μm · s?1)/(V · cm?1) (pH 7.2, ionic strength 0.155). This mobility change was completely reversed when chlorpromazine · HCl was removed by centrifugal washing. Increasing the drug concentration to 70μM did not affect the mobility change, indicating saturation of the electrophoretically detectable drug binding sites over chlorpromazine · HCl concentration range studied here. The effect of the 23 μM chlorpromazine · HCl on electrophoretic mobility was also measured in isotonic media of reduced ionic strength. The drug-induced reduction in negative surface charge density was found to be independent of ionic strength over the range 0.155 (Debye length, 0.8 nm) to 0.00310 (Debye length, 5.7 nm).Fixation of erythrocytes with glutaraldehyde affected neither the normal electrophoretic mobility of discocytes nor the reduced electrophoretic mobility of chlorpromazine · HCl-induced stomatocytes. When these stomatocytes were first fixed with glutaraldehyde, then washed free of chlorpromazine · HCl, they retained the stomatocyte form while regaining a normal control electrophoretic mobility. Conversely, when discocytes fixed in that form were treated with chlorpromazine · HCl, they showed the same mobility change as did fixed or unfixed stomatocytes. The drug-induced mobility change is therefore independent of the shape change, but reflects a contribution to cellular surface charge density from the membrane-bound chlorpromazine · HCl molecules. From the charge reduction, it is estimated that about 106 chlorpromazine · HCl molecules are bound at the electrokinetic cell surface and occupy approximately 0.4% of the total surface area.  相似文献

Laser multiple processing, i.e. laser surface texturing and then Laser Shock Processing (LSP), is a new surface processingtechnology for the preparation of bionic non-smooth surfaces. Based on engineering bionics, samples of bionic non-smoothsurfaces of stainless steel 0Crl 8Ni9 were manufactured in the form of reseau structure by laser multiple processing. The mechanicalproperties (including microhardness, residual stress, surface roughness) and microstructure of the samples treated bylaser multiple processing were compared with those of the samples without LSP The results show that the mechanical propertiesof these samples by laser multiple processing were clearly improved in comparison with those of the samples without LSP Themechanisms underlying the improved surface microhardness and surface residual stress were analyzed, and the relations betweenhardness, comnressive residual stress and roughness were also presented.  相似文献

Colonisation of soft lining materials by micro‐organisms Objective: This study evaluated the in vitro adherence of pathogenic micro‐organisms, Candida albicans, Staphylococcus aureus and Pseudomonas aeruginosa, to soft lining materials and their inhibitory effect on these micro‐organisms. Materials and Methods: To measure adherence, specimens of Molloplast B and Ufi Gel P were inoculated [107 colony‐forming units per millimetre (cfu/ml)] with TSB media containing the micro‐organisms. To determine the number of micro‐organisms in the 10?2–10?5 dilutions, 25 μl of the suspension were transferred to plates of selective media. Colony counts of each specimen were quantified (cfu/ml). The surface roughness was measured with a perfilometer to assess the relationship between the adherence of micro‐organisms and surface roughness of each material. For the inhibition test, specimens of materials were placed in agar plates inoculated individually with the micro‐organisms. After 48 h, the inhibition zones around the specimens were measured. Results: None of the materials exhibited inhibition zones. The number of cfu/ml of S. aureus and P. aeruginosa were significantly greater than C. albicans for both materials. The Ufi Gel P exhibited greater adherence of C. albicans than Molloplast B. No correlation was observed between the adherence of micro‐organisms and surface roughness. Conclusion: The surface roughness of the materials is not the only factor governing micro‐organism adherence.  相似文献

Mechanical damage to seeds occurring during harvesting and threshing, especially in naked cultivars, is one of the main factors decreasing the size and quality of yield. The objective of this study was to assess the susceptibility of naked oat cultivars to mechanical damage, considering the biometric parameters of seeds determined on the basis of computer image analysis and weight tests. The testing was carried out on eight cultivars harvested between 2008 and 2010 at 15% moisture and threshed at either 1.6 or 2.4 m·s?1 threshing drum speed. A 50% increase in the threshing speed caused an average 19% increase in the frequency and a 29% increase in the area of microdamage to seeds. There was a corresponding 4.1 mm2 change in the microdamage area when using the threshing speed of 1.6 m·s?1, and a 6.1 mm2 change when using the threshing speed of 2.4 m·s?1 was 53% and 68%, respectively, determined by a decreasing seed shape coefficient indicating seed elongation. The greatest resistance to mechanical damage was found on the Bullion cultivar, which was also characterized by the largest total projected area of seeds (7.14 mm2), as well as the greatest seed density (63.8 kg·hL?1) and thousand kernel weight (TKW) (28.2 g).  相似文献

The small intestine is known to possess mechanisms for intact transport and membrane hydrolysis of oligopeptides. To determine the relative role of these processes in peptide assimilation the fate of two model peptides known to be high-affinity substrates for the brush border aminooligopeptidase were studied in rat small intestine in vivo. Both 20 mM Gly-L-Pro, a potent inhibitor of peptide transport, and specific inhibitors of the aminopeptidase, 10 mM L-Ala-β-naphthylamide or the phthalimido derivative of 0.1 mM L-leucine bromomethyl ketone, reduced assimilation of L-Leu-Gly-Gly and L-Leu-L-Leu. Further inhibition was found when both transport and peptidase inhibitors were included in the intestinal perfusate suggesting that the model di- and tripeptides utilize both intact transport and surface hydrolysis for their assimilation. Although comparative kinetic parameters of intact transport (Km = 22 mM; V = 1.9 · 10?3μmol · s?1 · cm?2) and surface hydrolysis (Km = 8.7; V = 1.1 · 10?3) for l-Leu-l-Leu differed markedly, the relationship of peptide concentration to assimilation rate was nearly identical for intact transport and surface hydrolysis in the physiological range of 1–10 mM substrate. Both intact peptide transport and surface hydrolysis appear to be efficient and complementary processes that promote efficient assimilation of dipeptides and tripeptides. The relative importance of each assimilation process appears to depend upon the amino acid composition of the peptide nutrient.  相似文献
