SARS病毒的种型变异之比较及其PCR的分子诊断技术应用

概述了SARS-CoV冠状病毒的基因组结构与功能,比较了它与其它冠状病毒的种型变异,并以生物信息学的方法 探讨PCR技术在进行SARS病毒早期诊断上的应用。     

Polymorphism of microsatellite DNA and study of maize genetic resources

Microsatellite loci PCR-analysis of 80 maize lines and hybrids from two regions of Ukraine has been carried out. Allelic sets individual for each maize genotype have been revealed and discrimination potential of applied marker system has been determined. Comparative analysis of frequencies of alleles and genotypes occurrence in both samples has been carried out and possible participation of the investigated SSR-loci in formation of maize adaptability system has been shown.     

Characteristics of Pseudomonas aeruginosa pili and an elucidation of their relation to the virulence of the bacteria

The electron microscopic study of 175 P. aeruginosa museum and newly isolated strains has been made and their virulence has been determined. In 32% of these strains the presence of pili has been established. However, among the museum strains those with pili have been found to constitute only 19.5%, while among newly isolated strains pili have been detected in 39.7% of cases (the difference is statistically significant at p less than 0.01). No correlation between the formation of pili and the virulence of P. aeruginosa has been established.     

Purification and substrate specificity of BcmI restriction endonuclease

A strain producing the new specific restriction endonuclease BcmI has been found in the Bacillus generum. The enzyme has been purified by chromatography on the blue sepharose, phosphocellulose PII, heparin sepharose. The analogous purification has been obtained when the blue sepharose has been substituted for the orange sepharose, the home produced sorbent. The BcmI enzyme has been shown by the substrate specificity definition to be an isoschizomer of the restriction endonuclease ClaI.     

Construction of recombinant plasmids encoding the biosynthesis of the beta-subunit of cholera toxin

The structure of the cholera toxin operon and the location of A and B toxin subunits have been studied by the Southern blot hybridization on filters. The gene coding for the synthesis of the cholera toxin B-subunit has been cloned in the vector plasmid pBR322. The structural gene of A-subunit has been partially deleted by the restriction endonuclease Bal31 digestion. The size of the 250 b. p. deletion has been defined by electron microscopy. The production of the cholera toxin B-subunit in Escherichia coli K12 cells has been studied.     

Surgical management of lymphedema of the extremity.

Lymphedema of the extremity has been managed in 14 patients by staged excisions of the edematous subcutaneous tissue beneath skin flaps. Six patients have been followed for 4 to 6 years. This method has been successful in dealing with the symptoms of this condition. No episodes of postoperative celluitis have occurred in these patients. Normal function of the extremity has been achieved, and the contour has been preserved. It appears at this time that excision of the subcutaneous tissue is the most important aspect of the surgical management of lymphedema.     

Inhibitors of the nitrite oxidation of hemoglobin

Different types of active inhibitors of the reaction of nitrite hemoglobin oxidation have been revealed and studied. The dependence of inhibition of methemoglobin formation, on concentration of inhibitors at pH 5.9 and 7.17 has been determined. Differential absorption spectra of the inhibitors in the presence sodium nitrite in UV and visual light has been studied. The values of oxidation-reduction potentials have been estimated. Possible mechanism of action of the inhibitors has been discussed.     

Mode of action of intercalators: a theoretical study

Mode of action of some intercalators has been theoretically investigated on the basis of quantum mechanical perturbation method. Energies of H-bond interaction between drug chromophore and base pairs have been calculated in all possible orientations. The stacking energy has also been calculated with the base pairs. The effect of these interactions on specific recognition has also been discussed. On the basis of these studies, a model for the interaction of these drugs has been proposed. This explains the relative activities of acridine intercalators and satisfies the experimental observations.     

Sensitivity of the rabbit embryonal Leydig cells to the action of different pituitary hormones

Leydig cells reaction of rabbits testis to choriogonin action has been investigated during the prenatal period of ontogenesis. It has been found, that these cells sensitivity to the hormones studied has been detected at the early stages of embryogenesis. The specialization of Leydig cells response to choriogonin, thyrotrophin and prolactin in the process of prenatal period has been shown.     

Morphometric analysis of the state of the intramural nervous apparatus of rat duodenum]

Structural organization of the intramural apparatus of the duodenum has been studied in normal white rats. By means of a planimeter, the area of the nerve cells and their nuclei has been estimated in serial sections of the cranial and caudal parts of the duodenum; a definite regularity on the nerve cell types distribution has been stated in different parts of the duodenum. Concentration of afferent nerve elements has been noted in the cranial part of the duodenum.     

Epidemiological assessment of the microbial contamination of the breast milk of healthy women

The qualitative and quantitative composition of the microflora of human milk has been studied. The dynamics of the occurrence of different microorganisms constituting the microflora at different periods of lactation has been established. The connection between the appearance of septic purulent infections in infants and the microbial contamination of milk has been revealed. The presence of Staphylococcus aureus in human milk in an amount of 500 microbial cells/ml has been found to constitute no menace to healthy infants.

从断奶大鼠的肝脏中纯化得到了肝刺激物质（ＨＳＳ）的有效成份,即肝再生增强因子（ＡＬＲ）的蛋白质,酶解并对其多肽末端测序,据此推导出简并核苷酸序列,合成探针,对大鼠肝脏来源的ｃＤＮＡ文库进行筛选,首选获得了大鼠ＡＬＲ的ｃＤＮＡ克隆,随后又分别克隆了人和小鼠的ＡＬＲ的ｃＤＮＡ。与此同时,从酵母细胞中克隆了与线粒体氧化－－磷酸化功能密切相关的ＥＲＶ１基因,然后克隆了人的ＥＲＶ１同源基因,从功能上证实人     

Structural and functional aspects of the heart ventricle myoglobin of bluefin tuna

The heart ventricle myoglobin of bluefin tuna has been purified to an apparent homogeneity. The amino acid analysis has revealed only a limited number of substitutions between the myoglobins of yellowfin and bluefin tuna. The alpha-helix content of tuna myoglobin has been found considerably lower than that of mammalian myoglobin. No correlation has been discovered between the conformational stability and alpha-helix content. Denaturation experiments have shown that the whole structure of tuna myoglobin results from the interaction of two structural units which represent the product of independent folding processes. The structure of tuna myoglobin has been found more open and disorganized than that of sperm whale. This result has been related to the low content of electrostatic interactions and explained in terms of evolutive adaptations.     

Study of the interaction of main potato glycoalkaloids in inhibition of immobilized butyryl cholinesterase

The interaction of main potato glycoalkaloids alpha-solanine and alpha-chaconine in inhibition of horse serum butyryl cholinesterases immobilized on the pH-sensitive field-effect transistors has been investigated. The method of isobol diagram of Loewe and Muishnek has been used for interpretation of results. It has been shown the alpha-chaconine inhibits the immobilized bytyryl cholinesterases more strongly than alpha-solanine, and their mixture has the addition effect.     

Linked Cross Sectional Schemes for Estimating Growth of Children

For estimation of growth, the efficiency of linked cross sectional scheme has been compared with pure longitudinal and cross sectional schemes. The relevant estimation theory has been developed and the expressions for the optimum estimators alongwith their variances have been derived. It has been observed that for estimation of growth, the linked cross-sectional scheme has been observed to be less efficient as compared to pure longitudinal scheme but more efficient than pure cross-sectional scheme.     

Sampling Schemes for Studying Growth of Children

Efficiency of linked cross-sectional scheme has been compared with pure longitudinal schemes for generalised estimation of norm relaxing the condition of constancy of variances. The relevant theory of estimation has been developed and the expression for the optimum estimators alongwith their variances have been derived. The percentage gain in efficiency has been calculated. It has been observed that for higher values of Q, percentage gain in efficiency of proposed scheme over cross-sectional is very high.     

Identification of cell structure antigens of the causative agent of melioidosis by a 2-dimensional immunoelectrophoresis method

The antigenic composition of some cell structures of P. pseudomallei has been studied and the chemical nature of the antigens has been determined by the method of two-dimensional electrophoresis. In some cell components common antigens have been detected; at the same time these components have been found to possess their own characteristic antigenic complexes. The place of the cell structure antigens in the total antigenic structure of P. pseudomallei has been determined.     

Effect of intraparticle diffusion resistance on apparent stability of immobilized enzymes

The effect of the intraparticle diffusion resistance on the apparent stability of the immobilized enzyme suffering from the first-order deactivation has been studied quantitatively. A general expression for the relationship between the decreasing observed enzymatic reaction rate and the intrinsic enzyme deactivation rate has been introduced. The method to estimate the intrinsic deactivation rate constant also has been proposed. Using the invertases immobilized on a anion-exchange resin, the theory proposed in this work has been verified experimentally.     

A study of the mineral composition and the composition of volatiles of marsh cinquefoil

The mineral composition of different vegetative parts of marsh cinquefoil has been studied by inductively coupled plasma atomic emission spectroscopy and ion chromatography. The volatile oil has been obtained from leaves of marsh cinquefoil by hydro and steam distillation, and its composition has been determined by chromatography-mass spectrometry.
