During the neonatal period, high protein breakdown rate is a metabolic process inherent to elevated rates of protein accretion in skeletal muscle. To determine the relationship between hindlimb net movements of essential and nonessential amino acids in the regulation of hindlimb protein breakdown during an overnight fasting-feeding cycle, we infused overnight-food-deprived 10- and 28-day-old piglets with [1-(13)C]phenylalanine and [ring-(2)H(4)]tyrosine over 7 h (during 3 h of fasting and then during 4 h of feeding). Extraction rates for aspartate and glutamate after an overnight fast were 15% and 51% in the 10-day-old compared with 6% and 25% in the 28-day-old (P < 0.05) piglets, suggesting an altered requirement for precursors of amino acids to shuttle nitrogen to the liver as early life progresses. This occurred simultaneously with marginal positive hindlimb net balance of essential amino acids after an overnight fast, with negative net release of many nonessential amino acids, such as alanine, asparagine, glutamine, glycine, and proline. This suggests that newborn muscle does not undergo significant protein mobilization after a short period of fasting in support of an elevated rate of protein accretion. Furthermore, tyrosine efflux from hindlimb breakdown between overnight fasting and feeding periods was not different in the 10-day-old piglets, for which tyrosine was limiting, but when tyrosine supply balanced requirements in the 28-day-old piglet, hindlimb efflux was increased (P = 0.01). The results of the present study indicate that proteolysis and net movements of amino acids are coordinated mechanisms that sustain the elevated rate of net protein accretion during overnight feeding-fasting cycles in the neonate.  相似文献

To assess the reference dietary amino acid profiles for juvenile kuruma prawn Marsupenaeus japonicus a feeding trial was conducted using six semi-purified diets containing casein-gelatin and pre-coated supplemental crystalline amino acids (CAA) and a control diet containing intact protein (casein-gelatin). Pre-coated CAA were supplemented to the diets to simulate dietary amino acid profiles to those of the prawn egg protein (PEP), prawn larvae whole body protein (PLP), prawn juvenile whole body protein (PJP), squid meal protein (SMP), short-necked clam protein (SNP) and brown fish meal protein (BFP). The result showed that kuruma prawn juveniles are capable of utilizing the pre-coated CAA and higher growth performances were observed in the groups fed the PJP, SMP and the control diets than those fed the PLP, SNP, BFP and PEP diets. The essential amino acid proportions (A/E ratios) of the whole body of kuruma prawn differ slightly when compared with the other penaeids or freshwater prawn. The results suggest that PJP and SMP would be suitable as a reference dietary amino acid profile for juvenile prawn.  相似文献

Besides amino acid composition of a protein, their bioavailability is an important determinant of the protein quality. In view of the observations over the last decade or two, implicating the small peptide uptake by the mammalian intestine as a major route of protein absorption, a few animal and plant proteins were subjected to sequential enzymatic digestionin vitro with pepsin, pancreatin + trypsin and erepsin and the release of amino acids as small (including dipeptides) and large peptides and free amino acids, was determined. The relative protein values of α-lactalbumin, egg whites, casein, gluten, zein and protein isolates of soyabeans and groundnuts was determined using rat growth method. It was observed that relative protein value were positively correlated with the essential amino acid index of protein, quantity of essential amino acids released as small peptides and the dipeptide content of enzymatic digests, while there was a negative correlation between relative protein value and essential amino acid content of large peptide fraction.  相似文献

本次实验选取了圣景园、天嘉湖、今珠、富兴四种不同品牌的柴鸡蛋,对其蛋清和蛋黄中的粗蛋白含量以及氨基酸组成和含量进行了测定和分析,通过氨基酸比值系数法对其营养价值进行了评定。结果表明,这四种不同品牌的柴鸡蛋氨基酸含量丰富,种类齐全,组成比例合理,其中,今珠品牌的柴鸡蛋不论是粗蛋白含量,还是氨基酸比值系数分均高于其他几种柴鸡蛋,评分达到了82.34,说明其所含的必需氨基酸的含量及相互间的组成比例与人体的需求更加接近,营养价值更高。同时也说明柴鸡蛋营养价值较高,具有广阔的市场开发和利用前景。  相似文献

Sulfated glycopeptides were isolated from pronaisc and tryptic digests of egg shell membranes and hen oviduct. They were precipitated by cationic detergents and separated by preparative electrophoresis, after removal of small quantities of glucuronoglycosaminoglycans detected only in the oviduct (isthmus and magnum). The principal isolated sulfated glycopeptides were divided according to increasing electrophoretic mobilities into two groups A and B. The homogeneity of the purified glycopeptides was confirmed by gel filtration and polyacrylamide gel electrophoresis.Glycopeptides from pool preparation of tissue are analysed and carbohydrate and amino acids average values are estimated. Hexosamines (mainly N-acetylglucosamine), hexoses (galactose, glucose, mannose) and fucose were found in Glycopeptides A. The molar ratio of hexose/hexosamine was 0.4. N-Acetylneuraminic acid and sulfate were also present in Glycopeptides A. The molar ratio of sulfate/hexosamine ranged from 0.1 to 0.25. The Glycopeptides A composition indicated the presence of chains with many glycosyl groups and a few of amino acids residues. The carbohydrate components of Glycopeptides B from egg shell membranes and magnum were found to be hexosamines (N-acetylgalactosamine and N-acetylglucosamine in equimolar proportions), hexoses (galactose mainly and glucose), N-acetylneuraminic acid, and fucose. The molar ratio of hexose/hexosamine was 1. Sulfate was also present and the molar ratio of N-acetylneuraminic acid and sulfate to hexosamine was ranged from 0.8 to 1. The main amino acid residues in these glycopeptides were serine and threonine with destruction of these hydroxyamino acids during alkali treatment. Glycopeptides B probably consist of short carbohydrate chains, linked to the polypeptide through O-glycosidic bonds involving N-acetylgalactosamine and serine and threonine. Approximately 40% of the amino acid residues were linked to carbohydrate chains.Glycopeptides B from egg shell membranes magnum and egg white were very similar in their carbohydrate and amino acid composition and in their properties.Gylcopeptides A from egg shell membranes, isthmus and magnum showed similarities and divergences especially in the amino acid composition. These results suggest that magnum and isthmus in oviduct are both concerned with the synthesis of egg shell membrane glycoproteins.  相似文献

目的为明确人工和野生冬虫夏草在氨基酸方面的差异,并为进一步开发利用人工冬虫夏草提供理论依据,对室内全人工培殖冬虫夏草和产自四川康定的野生冬虫夏草的氨基酸营养价值进行分析评价。方法用日立L-8800型全自动分析仪检测氨基酸含量并根据FAO/WHO的氨基酸评分标准模式和鸡蛋蛋白模式进行氨基酸营养价值评价。结果人工冬虫夏草氨基酸种类齐全,其全草氨基酸总量高于产自康定的野生冬虫夏草;其全草的鲜味氨基酸高于产自康定的野生冬虫夏草;其全草必需氨基酸总量低于产自康定的野生冬虫夏草。人工冬虫夏草必需氨基酸与氨基酸总量的比值高于WHO/FAO评分模式而低于鸡蛋蛋白模式。结论人工冬虫夏草氨基酸不仅种类组成合理,而且具有较高的营养价值。这一结果为人工冬虫夏草的进一步开发利用提供了理论基础。  相似文献

The utilization of the blood meal by mosquitoes was investigated by first feeding females quantities of blood ranging from 1 to 5 mg, and then analyzing the faeces for the various by-products of protein catabolism that were subsequently eliminated. The nitrogeneous waste products in order of importance were uric acid, histidine, ammonia and arginine. Only traces of the other amino acids were excreted.The total amount of each faecal substance varied linearly with the quantity of blood ingested, however their relative proportions did not change. Regardless of blood meal size the quantily of uric acid and ammonia produced indicates that about 80% of the non-histidine and arginine amino acids are deaminated and utilized for metabolic purposes other than egg protein synthesis.Most of the histidine and about one half of the arginine content of the blood were excreted as free amino acids, but the other amino acids were lost in trace amounts.Nineteen per cent of the total ingested amino acids was incorporated into soluble yolk proteins and this proportion was constant even for small blood meals that result in a reduction in the numbers of eggs produced.The comparative aspects of nitrogen partitioning and blood meal utilization by haematophagous insects, as well as the factors that affect blood meal utilization and fecundity in A. aegypti are discussed.  相似文献

The influence of first-pass splanchnic metabolism was investigated by comparing the response of 5 lactating cows to an infusion of an amino acid mixture into the abomasum or a jugular vein over 5 d according to a complete block design. The basal diet and the amino acid infusion provided 71% and 14% of crude protein requirements, respectively. The jugular infusion increased (P = 0.01) milk yield by 0.80 kg in comparison to the abomasal infusion, but milk protein yield was not altered. The jugular infusion tended to increase (P = 0.06) the arterial concentration of total essential amino acids by 11% relatively to the abomasal infusion. Mammary plasma flow and net fluxes of amino acids and glucose were not affected by the infusion sites. Variations in essential amino acid concentrations suggest that splanchnic metabolism alters peripheral delivery of amino acids but the recirculation of amino acids within the animal decreased the impact of the first-pass splanchnic metabolism on lactational performances.  相似文献

A new method for the preparation of whole egg protein was presented and its qualification as a standard reference for evaluating dietary protein was made in the course of this experiment: e.g., Purity, 96.5% and Biological value, 99.9. The amino acid composition was almost identical with that shown in the table of FAO (1970).

Using this whole egg protein, casein and yeast, the nutritive values of dietary proteins in the adult rats at the normal level of body protein and in those after 8 day’s protein depletion were compared. It was found that the evaluation after protein depletion had a tendency to overestimate the nutritive value of proteins rather making the difference little between each other. The significance of the relative evaluation by use of the purified whole egg protein was discussed with special regard to the actual retention of body nitrogen, i.e., the net nitrogen retention.  相似文献

Identifying the subcellular localization of proteins is particularly helpful in the functional annotation of gene products. In this study, we use Machine Learning and Exploratory Data Analysis (EDA) techniques to examine and characterize amino acid sequences of human proteins localized in nine cellular compartments. A dataset of 3,749 protein sequences representing human proteins was extracted from the SWISS-PROT database. Feature vectors were created to capture specific amino acid sequence characteristics. Relative to a Support Vector Machine, a Multi-layer Perceptron, and a Naive Bayes classifier, the C4.5 Decision Tree algorithm was the most consistent performer across all nine compartments in reliably predicting the subcellular localization of proteins based on their amino acid sequences (average Precision=0.88; average Sensitivity=0.86). Furthermore, EDA graphics characterized essential features of proteins in each compartment. As examples, proteins localized to the plasma membrane had higher proportions of hydrophobic amino acids; cytoplasmic proteins had higher proportions of neutral amino acids; and mitochondrial proteins had higher proportions of neutral amino acids and lower proportions of polar amino acids. These data showed that the C4.5 classifier and EDA tools can be effective for characterizing and predicting the subcellular localization of human proteins based on their amino acid sequences.  相似文献

Fecundity, measured by utilization of dietary protein for the production ofyolk protein was examined in several mosquito species (A. aegypti, A. simpsoni, A. atropalpus, A. triseriatus) using blood from eight vertebrate hosts over a whole range of biological blood mean volumes. All experimental blood meals were injected by enema in measured amounts. The total ovarian and excretory nitrogen produced during each gonotrophic cycle was quantified.Blood proteins are utilized for oögenesis only to a limited extent: two thirds, at the most, are detectable in the mature ovaries. This efficiency decreases with increasing blood meal size to about one fifth. The nitrogen content of each mature egg is constant and species-specific, irrespective of blood meal size or source.There are no significant differences in the efficiency of conversion of protein from different types of vertebrate blood into oöcyte protein except for human blood which led to a drastically reduced efficiency. This is shown to be due to a lack of isoleucine which is essential for reproduction. Hominid primates lack this amino acid in their haemoglobin molecule and it exists at a low titre in human plasma. Apparently, in other vertebrates the amino acids are better balanced in the haemoglobin or, can be mobilized in sufficient amounts from various non-haemoglobin protein fractions.All blood protein is catabolized and the excess nitrogen excreted quantitatively; excretory nitrogen therefore reflects the quantitative demands of a gonotrophic cycle. Pectoral muscle can be an important source of protein for birds. During egg formation Zebra Finches Taeniopygia guttata are able to compensate for nutritional inadequacies in their diet by utilization of the protein in their flight muscles. This analysis of flight muscle sarcoplasm supported earlier observations of protein depletion during egg production. However, SDS gel electrophoresis of the sarcoplasm produced no evidence to support a previous suggestion of the existence of a high molecular weight storage protein, and it is thought that the original observation may have arisen as an artefact of experimental methodology. During laying, protein removal from the sarcoplasm occurred over a range of different proteins and was not confined to any one specific protein band. Additionally, the protein band most reduced over the course of laying did not contain elevated levels of the amino acids most limiting to egg production. These results indicate that during laying, flight muscle sarcoplasm contributes towards the nutrient requirements of egg production from general protein reserves, rather than from a specific storage protein containing elevated levels of limiting amino acids.  相似文献

The effects of arsenate on the levels of free amino acids in rat plasma and red blood cells have been investigated. The biggest changes occur in the branched-chain amino acids valine, isoleucine and leucine which are markedly decreased. The levels of several other amino-acids also change. The effects on the amino acids are not due to a change in the food intake caused by dosing with arsenate. This has been shown by comparing the results with those on fasting rats (40 h). Branched-chain amino acids in particular are markedly increased in plasma of fasting rats as opposed to a decrease in arsenate-treated rats.  相似文献

Experiments were conducted on growing male SPF-rats to study the optimum supplementation with essential amino acids of diets containing wheat and barley (1.4:1) as the only source of nitrogen. In growth and balance experiments comprising 30 and 17 combinations of L-lysine and L-threonine, respectively, the concentrations of the two amino acids corresponding to maximum body weight gain, feed conversion efficiency, biological value of protein and net protein utilization were calculated using multiple regression analysis. Addition of lysine and threonine to the basal diet markedly improved the nutritional value of cereal proteins. The optimum concentrations found were 0.69–0.70% total dietary lysine and 0.52–0.55% total dietary threonine. Statistical analysis showed that the number of observations used for the estimation of regression functions in the growth experiment could be reduced to ca. 100 without substantially affecting precision.  相似文献

The influence of protein quality on the growth-depressing effect of excessive amount of 12 individual essential and semiessential amino acids was examined. Growing rats were fed for 3 weeks diets containing either 10.5% egg albumin or 11.6% wheat gluten (equivalent to the protein content of a 10% casein diet) supplemented with 5% of each of the l-amino acids. In general, the pattern of growth depression produced by the addition of excess amino acids to the egg albumin or the wheat gluten diet was similar to that of the case of casein diet obtained previously under the same experimental conditions. However, the extent of these effects was dependent not only upon the kind of amino acid supplemented with but also upon the source of protein used, and the depressing effect of each of excess amino acids added to the wheat gluten diet was usually severer than those added to casein and egg albumin diets. No evidence was noted of any striking changes in the liver protein and nucleic acid concentrations by either diets, but total liver protein, RNA and DNA contents were decreased in some amino acid groups of the egg albumin diet and in all amino acid groups of the wheat gluten diet except the lysine addition. The free amino acid level in plasma generally showed extreme elevation for the amino acid supplemented in excess in the diet, and in most cases the extent of the elevation was correlated with the growth depression.  相似文献

Nitrogen flux across the hindquarters of fetal and maternal sheep (n = 15) was determined during normal feeding and following 5 days of maternal fasting. Arterial and venous whole blood concentrations of free amino acids, ammonia and oxygen were measured entering and exiting the hindquarters. Further, the DNA, protein and nitrogen contents of the hindlimb skeletal muscle of the fetus were determined in the fed state and following the 5-day fast. Results of these studies indicate that maternal and fetal hindlimb metabolism differ during fasting. There is a net efflux of alanine, glutamine and total nitrogen from the maternal hindquarters following 5 days of fasting. The fetus also releases glutamine and alanine from the hindquarters during the fast, presumably as potential energy substrate. However, nitrogen balance across the fetal hindquarter remains positive as a result of increased positive arteriovenous differences for other amino acids (particularly leucine and isoleucine). The concentrations of DNA, protein and nitrogen in fetal skeletal muscle remain unchanged during fasting. These data indicate that, whereas the mother undergoes protein catabolism and net nitrogen loss from the hindquarter during fasting, the fetus maintains a positive nitrogen balance across the hindquarter.  相似文献

Male rats aged 45, 85, 145 and 270 days (daily body mass increments on an optimal diet containing casein were 6.73, 2.88, 0.53 and 0.31 g respectively) were fed 15 days ad libitum on a diet with a nutrient content physiological for their age, in which the protein source was milk casein (ratio of essential to nonessential amino acids E/N = 0.79, compensation coefficient K = 14) or wheat gluten (E/N = 0.30, K = -8). In the case of gluten, net protein utilization (NPU) fell markedly in rapidly growing animals aged 45 and 85 days (33 and 30% more than with casein), indicating that without essential amino acid compensation, gluten is inadequate for animals of this age, whose organism requires fully ensured proteosynthesis for growth and development. In adolescence and adulthood (145 and 270 days), the utilization of proteins is not dependent on their quality (the decrease in NPU 13 and 12%--is nonsignificant). That means that a smaller amount of essential amino acids, including the limiting amino acid in uncompensated protein, is sufficient for the maintenance and renewal of organs and tissues, i.e. for proteosynthesis. The activation of gluconeogenesis (phosphoenolpyruvate carboxykinase activity in the liver) after the intake of plant protein confirms the effect of proteins on catabolic processes.  相似文献

Abstract— —The concentration of free amino acids has been determined in lumbar CSF in 37 fasting normal subjects. The values obtained have been compared with the concentration of the same amino acids measured in venous plasma collected simultaneously and with ventricular CSF amino acid concentrations. Twenty-three amino acids have been identified and quantitated in CSF and plasma. Trace quantities of eight other amino acids have been also detected.
The concentration of 13 amino acids in CSF has been shown to be directly related to the plasma concentration. No such relationship was noted for the other 7 amino acids. Significant variations in the concentration of individual amino acids relating to both age and sex have been noted. A large number of unidentified ninhydrin positive compounds have been found in CSF. Preliminary studies have identified one of these as ɛ-aminobutyric acid (GABA).  相似文献
