The arginase activity of the liver of rats was measured after they had been maintained for 11 or 12 days on diets containing natural proteins (casein or gluten) or amino acid mixtures of various tryptophan levels.

Specific activity or total arginase activity increased with the increasing protein quality. Liver arginase activity of the casein group was significantly higher than that of the gluten group in every case for 27-, 61-, and 158-day-old rats. In the case of amino acid diets, the arginase increased with the increments of tryptophan levels up to the “tentative” minimum requirement in the diet. Moreover, these alterations in arginase activity varied inversely with the urinary urea excretion.

From the results, it was assumed that the total activity of liver arginase is not necessarily determined only by the metabolic needs for urea biosynthesis.  相似文献

Relationship between serine dehydrase activity in rat liver and dietary protein and carbohydrate levels, using casein and wheat starch, was investigated. Between 10% to 50% of dietary protein levels the enzyme activity sharply increased with increase of protein level. The enzyme activity was also increased by the restriction of carbohydrate intake, even when protein intake was equal in amount. Apparently enzyme activity is controlled by dietary protein level and also by carbohydrate level. In the case of high protein or low carbohydrate diet these changes may be mainly acting for utilization of protein as caloric source.  相似文献

Effects of the change of dietary protein on serine dehydrase activity in rat liver have been studied, using egg albumin, casein, rice protein, and wheat gluten as protein source. At 35% of dietary protein level, the activity induced by egg albumin and casein diets were higher than those by rice protein and wheat gluten diets. Parallel relation was observed between the enzyme activity and the protein intake. These results suggest that the dietary induction of this enzyme are based on the protein intake, which reflects the nutritional quality of dietary protein, rather than merely on the dietary protein level.

The contribution of individual amino acid for the enzyme induction by the egg albumin diet at 35% level was investigated, and it was concluded that this enzyme induction is dependent not on a specific amino acid but on the combined effect of each amino acid.  相似文献

高脂喂养大鼠肝脏的NF-κBp65表达与胰岛素抵抗的相关性   总被引:1,自引:0,他引:1  
目的探讨高脂饲料喂养大鼠肝脏NF-κBp65蛋白的表达与胰岛素抵抗的关系。方法采用高脂饲料喂养建立胰岛素抵抗大鼠模型,并用正常血糖-高血浆胰岛素钳夹技术评估。应用Western blotting方法检测大鼠肝脏中NF-κBp65蛋白的表达。结果①高脂饲料组大鼠的葡萄糖输注率明显低于基础饲料组[GIR60~120(0.76±0.28vs4.26±0.70)mg/(kg.min),P〈0.01]。②高脂饲料组大鼠肝脏NF-κBp65蛋白的表达明显高于基础饲料组(A值118.48±1.45vs68.13±4.84,P〈0.01)。③高脂胰岛素抵抗大鼠肝脏NF-κBp65蛋白表达与GIR60-120(r=-0.993,P=0.000)和ISI(r=-0.773,P=0.009)负相关。结论高脂诱导的胰岛素抵抗大鼠肝脏NF-κB的激活可能是产生肝脏和全身胰岛素抵抗的根源。  相似文献

Recent results from our laboratory have indicated that during aging, the activity of Cu/Zn-SOD decreases in rat liver and that there is an accumulation of altered protein. It was also shown that the old Cu/Zn-SOD had one histidine fewer than the young one. In the present study, the immunoprecipitation experiments showed that the amount of immunoprecipitable Mn-SOD from liver of old rats was greater than from young ones, but when amino acid residues were measured in purified young and old Mn-SOD from liver, no change was observed. In lung, no significant age-related differences in total SOD, Cu/Zn-SOD and Mn-SOD activity were found, nor was there accumulation of altered protein during aging.  相似文献

Renal reabsorption of urea was studied in 8 9-month old reindeer calves fed low protein (19–34 g crude protein, mostly lichens) and high protein (68–69 g crude protein, lichens + soybean meal) diets. Low protein diets fed for a 3-month period resulted in an average renal reabsorption of 93% of the filtered urea, while only 50 % was reabsorbed on the high protein ration. It was calculated that if the reabsorbed urea was used completely for protein synthesis in the rumen, 4 g crude protein could be made daily in the lichen fed animals. This amount would be a very significant contribution to the nitrogen economy of animals which are usually in a negative nitrogen balance when lichens are the main food consumed.  相似文献

The role of auxin and protons in the gravitropic response of the sunflower (Helianthus annuus L. cv Sungold) hypocotyl has been investigated. No physiological asymmetry in acid-growth capacity could be detected between the upper and lower surfaces of gravistimulated hypocotyls. These data imply that neutral buffers inhibit shoot gravitropism by preventing the establishment of a lateral proton gradient along gravitropically stimulated hypocotyls. Indirect evidence that auxin is involved in the establishment and/or maintenance of such a gradient derives from the quantitative assessment of the effects of exogenous auxin, anti-auxins, and vanadate on gravicurvature. At low concentrations, exogenous auxin accelerated curvature; at high concentrations, curvature was prevented. Vanadate, an inhibitor of auxin-enhanced H+ secretion, α-(p-chlorophenoxy)isobutyric acid (PCIB), an anti-auxin, and 2,3,5-triiodobenzoic acid (TIBA), an auxin-transport inhibitor, prevented observable asymmetric proton excretion using a brom cresol purple agar technique and also inhibited gravicurvature. Vanadate, PCIB, and TIBA inhibition of gravicurvature could be reversed with acid treatment to the lower surface of a gravistimulated hypocotyl. Auxin treatment to the lower surface of a gravistimulated hypocotyl did not reverse vanadate-induced inhibition, but it did partially reverse PCIB- and TIBA-induced inhibition. These results indicate a close relationship between the acid-growth theory and the differential growth responses of the sunflower hypocotyl during gravitropism.  相似文献

To understand the nature and amount of excretory products of Isoparorchis hypselobagri, a digenetic trematode inhabiting the swim bladder of Wallago attu the parasites were kept in PBS media without glucose (control) and with glucose (treated) successfully up to 300 h and estimated different excretory products in the media after every alternate 12 h. The quantitative estimation of urea excreted by I. hypselobagri is done. It is lower when the flukes were kept in vitro in the control incubation media than in the treated incubation media. The highest amount of urea excreted in control media was 18.563 mg % at 276 h, and in treated condition 29.759 mg % after 216 h of incubation, while the lowest amount of urea excreted, 6.08 mg %, in control media and 10.343 mg % in treated media respectively after 12 h of incubation. The results were highly significant at 5 % level. The rate of change of excretion of urea in treated condition after every 12 h of incubation time interval was also studied. In the swim bladder washings of host urea is also present in significant amount and the amount excreted depends on the number of parasites harbor. From the results presence of both ammonotelic and ureotelic conditions are suggested.  相似文献

Circadian changes in renal hemodynamics and urinary glycosaminogly-can (GAG) excretion were studied in normal Sprague-Dawley rats to further investigate rhythms in kidney function. Urinary water, protein, and GAG excretion, as well as glomerular filtration rate (GFR) and renal plasma flow (RPF), were determined every 4h over the 24h cycle in an attempt to characterize any temporal changes. Urinary flow rate and proteinuria peaked during the dark activity period of the animals, consistently at the same hour, whereas the lowest values were detected during the resting phase. GAG are mucopolysaccharides entering the constitution of the glomerular basement membrane (GBM), which is the key component in the process of glomerular filtration. Similarly, the urinary excretion rate of GAG showed a circadian rhythmicity in phase with urinary water and protein excretion, with markedly increased values observed during the nocturnal phase of the animals. Moreover, GFR and RPF were demonstrated to exhibit large circadian variations in phase with renal excretory rhythmicity, showing nighttime values significantly greater compared to daytime ones. Strong correlations were found between GFR and RPF rhythms, as well as between GAG and GFR, and GAG and RPF rhythms, although the latter were not statistically significant. This pattern suggests that the circadian rhythmicity in urinary excretion rate of GAG in physiological conditions could presumably be secondary to the temporal changes in renal hemodynamics. In this respect, knowledge of renal chronobiology helpfully contributes to increase our understanding of renal physiology.  相似文献

The effects of chronic ethanol or sucrose administration to rats on acetylcholinesterase from brain and liver were investigated. Membrane-bound and soluble acetylcholinesterase activities were determined in fractions prepared by centrifugation. The thermal stability and the effects of temperature and different types of alcohols on acetylcholinesterase activity were also studied. Membrane-bound acetylcholinesterase activity increased (p < 0.01) in the liver after chronic ethanol administration, whereas no differences among groups in the encephalic areas, except in the brain stem soluble form, were found. Membrane-bound acetylcholinesterase from the ethanol- and sucrose-treated groups was more stable at the different temperatures assayed between 10 and 50°C than that corresponding to the control group. Non-linear Arrhenius plots were obtained with preparations of membrane-bound acetylcholinesterase from rat liver, with discontinuities at 30°C (control or sucrose groups) or 34–35°C (alcohol group). Assays made with membrane-bound or soluble enzyme from brain showed linear Arrhenius plots in all groups studied. The inhibitory effects of increasing concentrations of ethanol, n-propanol and n-butanol on acetylcholinesterase preparations from forebrain, cerebellum, brain stem and liver of the three experimental groups (control, sucrose-fed and ethanol-fed) were very similar. However, n-butanol displayed a biphasic action on particulate or soluble preparations of rat forebrain. n-butanol inhibited (competitive inhibition) at higher concentrations (250–500 mM), while at lower concentrations (10–25 mM), the alcohol inhibited at low substrate concentrations but activated at high substrate concentration. These results suggest that the liver is more affected by ethanol than the brain. Moreover, the lipid composition of membranes is probably modified by ethanol or sucrose ingestion and this would affect membrane fluidity and consecuently the behaviour of acetylcholinesterase.  相似文献

A new binary vector, pZT4B, containing the UDP-N-acetylglucosamine: dolichol phosphate N-acetylglucosamine-1-P transferase (GPT) gene as a selection marker, was constructed. The green fluorescent protein (GFP) gene was inserted into pZT4B, and the resulting plasmid was used in the transformation of Arabidopsis. Superoxide dismutase (SOD) activity was measured in the brain and liver of 24–26- and 3-month-old rats. No significant age-related differences in Cu/Zn-SOD activity were found in any of the tissues studied. A small but significant increase in total SOD activity was observed in the whole brain (10-20%), cerebral cortex (11%), and hypothalamus (18%) of old rats, whereas a much more important increase in Mn-SOD activity was found in the whole brain (48%), cerebral cortex (70%), striatum (60%), and hypothalamus (30%). The increase of Mn-SOD activity in the brain of old rats suggests the enzyme may play an important role in the process of aging. Mn-SOD is found only in the mitochondrion, which could be an important site of oxygen free radical production, and a significant increase in the enzyme activity was also found in the lung of hypoxic rats. A significant decrease in total SOD and Mn-SOD activity was observed in the liver of old rats. Preliminary experiments in 23–24-month-old mice similarly showed an increase and a decrease in total SOD and Mn-SOD activity, respectively, in the whole brain and liver. These results suggest that the regulatory mechanisms of Mn-SOD in the brain and liver vary differentially with age.  相似文献

The activities of all urea cycle enzymes (carbamyl phosphate synthetase, ornithine trans- carbamylase, argininosuccinate synthetase, argininosuccinase and arginase) have been determined in the liver of rats forcibly fed diets lacking in individual essential amino acids from amino acid mixture simulating to a casein. In general, these enzyme activities (units/g liver and total units/body wt) in rats fed the single essential amino acid-devoid diet decreased as compared with those activities in animals fed complete diet, but their decreases were not as large as those observed in group of all amino acid-devoid diet. The degree of decrease in these enzyme activities differed somewhat from each other in individual enzymes and each essential amino acie-devoid groups. In contrast, in rats fed the arginine devoid diet, the activities (total units/body wt) of all enzymes expect the case of arginase increased more than those in the group of complete diet.  相似文献

It is well known that even under identical task conditions, there is a tremendous amount of trial-to-trial variability in both brain activity and behavioral output. Thus far the vast majority of event-related potential (ERP) studies investigating the relationship between trial-to-trial fluctuations in brain activity and behavioral performance have only tested a monotonic relationship between them. However, it was recently found that across-trial variability can correlate with behavioral performance independent of trial-averaged activity. This finding predicts a U- or inverted-U- shaped relationship between trial-to-trial brain activity and behavioral output, depending on whether larger brain variability is associated with better or worse behavior, respectively. Using a visual stimulus detection task, we provide evidence from human electrocorticography (ECoG) for an inverted-U brain-behavior relationship: When the raw fluctuation in broadband ECoG activity is closer to the across-trial mean, hit rate is higher and reaction times faster. Importantly, we show that this relationship is present not only in the post-stimulus task-evoked brain activity, but also in the pre-stimulus spontaneous brain activity, suggesting anticipatory brain dynamics. Our findings are consistent with the presence of stochastic noise in the brain. They further support attractor network theories, which postulate that the brain settles into a more confined state space under task performance, and proximity to the targeted trajectory is associated with better performance.  相似文献
