稻草秸秆纤维素分解菌的分离筛选   总被引:7,自引:0,他引:7  
本研究基于获得高效木质纤维素分解菌的目的,以刚果红纤维素琼脂和滤纸条培养基为初筛培养基,从分离获得的124株真菌中筛选出透明圈与菌落直径比值较大、滤纸条分解能力较强的11个菌株.经液体发酵,测定其酶活力,复筛得到羧甲基纤维素酶活和滤纸酶活均较高的4个菌株;并进行了不同碳源和不同pH对筛选菌株产酶能力的影响试验,发现不同菌株对不同纤维素物质的分解能力不一样,同一菌株对不同纤维素碳源的利用能力也不相同.  相似文献

校园空气污染微生物的检测与评价   总被引:10,自引:0,他引:10  
采用沉降平板法 ,选用牛肉膏培养基与察贝克培养基 (czpekmedium) ,分别在夏季与冬季检测了校园不同环境中空气污染细菌与霉菌的含量。结果表明 :校园不同环境的空气污染微生物类群与数量随季节的变化而变化。学生寝室、学生食堂、校区餐厅以及网吧的空气中污染微生物以细菌为主。其中 ,在冬季的细菌平均含菌量为 91 4 7.6 7个 /m3 空气 ,而以女生寝室的含菌量最高 ,达到 2 1 5 4 6个 /m3 空气 ,超出卫生标准的 3.1倍 ;在夏季时 ,细菌的平均含菌量为 1 0 6 6 1 .75个 /m3 空气 ,而以校园餐厅的含菌量最高 ,达到 4 35 1 1个 /m3 ,超出卫生标准的 6 .9倍。校图书馆、电影院、学生寝室及餐厅中的霉菌含量较多。其中 ,在冬季的霉菌平均含菌量为 5 6 8.4 4个 /m3 空气 ,以女生寝室的含菌量最高 ,84 6个 /m3 ;在夏季的霉菌含菌量为 85 5 .88个 /m3 ,最高时为 1 6 6 5个 /m3 ,这是由于卫生条件差 ,居室狭小 ,人口拥挤 ,空气潮湿 ,通风不良等因素造成的  相似文献

高效降解生活污水中COD的根际微生物的分离筛选   总被引:2,自引:0,他引:2  
采用平板划线法从人工湿地的芦苇、美人蕉的根际土壤中分离出若干细菌、真菌、放线菌菌株,在实验室条件下检测了这些菌株对灭菌生活污水和自然生活污水COD的去除效果,结果表明4株细菌、1株放线菌、1株真菌对灭菌生活污水和自然生活污水COD均具有较高的去除率。4株细菌在降解灭菌生活污水COD的去除率在48 h后依次为75.4%、78.7%、83.5%、69.8%;其在降解未灭菌生活污水COD的去除率在48 h后依次为43.4%、47.8%、50.7%、36.8%;真菌对灭菌和未灭菌生活污水COD的去除率在48 h后分别为60.2%、41.3%;放线菌对灭菌和自然生活污水COD 48 h后的去除率分别为57.8%、46.4%。这几株高效降解COD的湿地根际微生物具有潜在的应用价值。  相似文献

以透明圈筛选法和滤纸崩解法相结合,建立初筛方法;模拟田间覆土菌种筛选方法,以秸秆腐解率为指标,建立了复筛方法,并进行了模拟田间应用效果分析。Y13、774、HX及J1 4株菌的腐解率较高,比对照分别提高27.04%、29.65%、35.38%和27.86%;其产CO2能力皆高于空白对照,分别提高41.67%、51.85%、30.56%和57.41%;其可溶性糖含量均提高,比对照平均提高15.40%。  相似文献

Production of titanium-dioxide nanomaterials (nano-TiO2) is increasing, leading to potential risks associated with unintended release of these materials into aquatic ecosystems. We investigated the acute effects of nano-TiO2 on metabolic activity and viability of algae and cyanobacteria using high-throughput screening. The responses of three diatoms (Surirella angusta, Cocconeis placentula, Achnanthidium lanceolatum), one green alga (Scenedesmus quadricauda), and three cyanobacteria (Microcystis aeruginosa, Gloeocapsa sp., Synechococcus cedrorum) to short-term exposure (15 to 60 min) to a common nano-TiO2 pigment (PW6; average crystallite size 81.5 nm) with simulated solar illumination were assessed. Five concentrations of nano-TiO2 (0.5, 2.5, 5, 10, and 25 mg L-1) were tested and a fluorescent reporter (fluorescein diacetate) was used to assess metabolic activity. Algae were sensitive to nano-TiO2, with all showing decreased metabolic activity after 30-min exposure to the lowest tested concentration. Microscopic observation of algae revealed increased abundance of dead cells with nano-TiO2 exposure. Cyanobacteria were less sensitive to nano-TiO2 than algae, with Gloeocapsa showing no significant decrease in activity with nano-TiO2 exposure and Synechococcus showing an increase in activity. These results suggest that nanomaterial contamination has the potential to alter the distribution of phototrophic microbial taxa within freshwater ecosystems. The higher resistance of cyanobacteria could have significant implications as cyanobacteria represent a less nutritious food source for higher trophic levels and some cyanobacteria can produce toxins and contribute to harmful algal blooms.  相似文献

Evidence suggests that gut microbes colonize the mammalian intestine through propagation as an adhesive microbial community. A bacterial artificial chromosome (BAC) library of murine bowel microbiota DNA in the surrogate host Escherichia coli DH10B was screened for enhanced adherence capability. Two out of 5,472 DH10B clones, 10G6 and 25G1, exhibited enhanced capabilities to adhere to inanimate surfaces in functional screens. DNA segments inserted into the 10G6 and 25G1 clones were 52 and 41 kb and included 47 and 41 protein-coding open reading frames (ORFs), respectively. DNA sequence alignments, tetranucleotide frequency, and codon usage analysis strongly suggest that these two DNA fragments are derived from species belonging to the genus Bacteroides. Consistent with this finding, a large portion of the predicted gene products were highly homologous to those of Bacteroides spp. Transposon mutagenesis and subsequent experiments that involved heterologous expression identified two operons associated with enhanced adherence. E. coli strains transformed with the 10a or 25b operon adhered to the surface of intestinal epithelium and colonized the mouse intestine more vigorously than did the control strain. This study has revealed the genetic determinants of unknown commensals (probably resembling Bacteroides species) that enhance the ability of the bacteria to colonize the murine bowel.  相似文献

This report describes a comparison of the Behringwerke antistreptolysin O (ASO) latex screening test with the ASO hemolytic test. Agreement between the two tests was poor when Difco streptolysin O (SLO) reagent was employed in the hemolytic test; approximately 34% of the sera with ASO titers in the normal range of the hemolytic test gave false-positive latex test reactions. However, the percentage of false-positive latex test reactions was only 5% when Behringwerke SLO reagent was used in the hemolytic test. An assay of the Difco and Behringwerke SLO reagents against an ASO standard indicated that the Difco SLO reagent was more potent than the Behringwerke SLO reagent. The lack of agreement between the Behringwerke latex test and the hemolytic test using Difco SLO reagent is attributed to the potency of the SLO reagents.  相似文献

A potent fungus for blood-coagulating enzyme was isolated from soil during screening tests of about 1500 strains of microorganisms. The fungus was identified as a strain belonging to Cephalosporium. This enzyme could be produced effectively by solid culture on wheat bran and partially purified to about 800 fold. This enzyme was one of the alkaline proteases. It was stable over a pH range of 7~10 and was activated by Ca2+ The enzyme could not coagulate 1% fibrinogen solution. The enzyme mixed with diluted blood plasma coagulated the fibrinogen solution. The enzyme also coagulated the citrated plasma from humans, bovines, mice, goats and hens.  相似文献

作物全生育期抗旱性对发掘和利用抗旱品种和抗旱基因至关重要,但谷子全生育期抗旱鉴定至今未有报道,抗旱鉴定缺乏鉴定指标。本试验在干旱池模拟干旱条件下,对苗期抗旱性表现不同的谷子品种进行了全生育期抗旱性研究,调查分析了根干重等形态和生理性状在干旱胁迫条件下的变化;采用相关和灰色关联度等方法,分析这些指标与抗旱性(DRI)的关系。结果表明,相对根冠比、相对单穗粒重和灌浆期光合速率、蒸腾速率同抗旱性表现极显著相关,可以作为谷子全生育期抗旱性鉴定的指标;而相对根干重、相对单穗重、相对株高和气孔导度则可以作为谷子全生育期抗旱性鉴定的参考指标;在供试的品种中,红根谷和大齐头白表现了良好的综合抗旱性。本文还讨论了抗旱性的复杂性以及不同性状对干旱胁迫的反应,所建立的指标对谷子全生育期抗旱鉴定具有指导意义。  相似文献

Model organisms are central to contemporary biology and studies of embryogenesis in particular. Biologists utilize only a small number of species to experimentally elucidate the phenomena and mechanisms of development. Critics have questioned whether these experimental models are good representatives of their targets because of the inherent biases involved in their selection (e.g., rapid development and short generation time). A standard response is that the manipulative molecular techniques available for experimental analysis mitigate, if not counterbalance, this concern. But the most powerful investigative techniques and molecular methods are applicable to single-celled organisms (‘microbes’). Why not use unicellular rather than multicellular model organisms, which are the standard for developmental biology? To claim that microbes are not good representatives takes us back to the original criticism leveled against model organisms. Using empirical case studies of microbes modeling ontogeny, we break out of this circle of reasoning by showing: (a) that the criterion of representation is more complex than earlier discussions have emphasized; and, (b) that different aspects of manipulability are comparable in importance to representation when deciding if a model organism is a good model. These aspects of manipulability harbor the prospect of enhancing representation. The result is a better understanding of how developmental biologists conceptualize research using experimental models and suggestions for underappreciated avenues of inquiry using microbes. More generally, it demonstrates how the practical aspects of experimental biology must be scrutinized in order to understand the associated scientific reasoning.  相似文献
